The effects of cadmium chloride concentration on root, bulb and shoot growth of garlic (Allium sativum L.), and the uptake and accumulation of Cd2+ by garlic roots, bulbs and shoots were investigated. The range of cadmium chloride (CdCl2 x 2.5H2O) concentrations was 10(-6) - 10(-2) M. Cadmium stimulated root length at lower concentrations (10(-6) - 10(-5) M) significantly (P < 0.005) during the entire treatment period. The seedlings exposed to 10(-3) - 10(-2) M Cd exhibited substantial growth reduction (P < 0.005), but did not develop chlorosis. Garlic has considerable ability to remove Cd from solutions and accumulate it. The Cd content in roots of garlic increased with increasing solution concentration of Cd2+. The roots in plants exposed to 10(-2) M Cd accumulated a large amount of Cd. approximately 1,826 times the control. The Cd contents in roots of plants treated with 10(-3), 10(-4), 10(-5) and 10(-6) M Cd were approximately 114, 59, 24 and 4 times the control, respectively. However, the plants transported only a small amount of Cd to their bulbs and shoots and concentrations in these tissues were low.  相似文献

The effect of excessive Cd on the growth and metal uptake by leafy vegetables Brassica chinensis L. (cv. Wuyueman) and Brassica pekinensis (Lour.) Rupr. (cv. Qingyan 87-114) were studied in hydroponic solution culture. The Cd concentration higher than 10 μM significantly decreased the root elongation and leaf chlorophyll contents of both plant species. The shoots of B. pekinensis had significantly higher concentrations of total and water-soluble Cd than B. chinensis. The roots of both species accumulated more Cd than the shoots in all the Cd treatments. Most of the Cd in the roots was found in the cell walls. The shoot/root ratio of Cd concentrations in B. pekinensis was always greater than that in B. chinensis. But, the concentration and proportion of Cd in the cell walls in B. chinensis were higher than that in B. pekinensis. Cadmium treatments also increased the concentrations of total non-protein thiols (NPT) in the shoots of the both species. A significant correlation was found between the concentrations of soluble Cd and NPT in plant shoots.  相似文献

A wide range of cadmium concentrations (from 4 to 200 μM for seedlings and up to 2 mM for germinating kernels) was used to assess Cd toxic effects on maize (Zea mays L.) plants at the different developmental stages: germinating kernels, seedlings (4–9 days), and juvenile plants (34 days). Cd accumulation in plant organs was followed, and its lethal concentration was elucidated. In maize, cadmium was accumulated predominantly in roots; in shoots it was mainly accumulated in the lower leaves, and the higher was leaf position the lower was Cd content in it. At high concentrations (80 and 200 μM), kernels became the substantial cadmium depot. Germinating kernels manifested the lowest sensitivity to cadmium; seedlings were more sensitive; the inhibition of juvenile plant growth attained 90% and more. In the tested range of concentrations, cadmium suppressed shoot mass accumulation harder than that of roots. In 34-day-old plants, water content in shoots was stronger reduced than in roots. Plant death was also manifested earlier in shoots. It was concluded that maize plant sensitivity to cadmium increases with plant growing and that, under conditions of normal mineral nutrition, cadmium inhibits shoot growth more severe than root growth.  相似文献

The effects of Cd have been investigated in tomato (Lycopersicon esculentum) plants grown in a controlled environment in hydroponics, using Cd concentrations of 10 and 100 μM. Cadmium treatment led to major effects in shoots and roots of tomato. Plant growth was reduced in both Cd treatments, leaves showed chlorosis symptoms when grown at 10 μM Cd and necrotic spots when grown at 100 μM Cd, and root browning was observed in both treatments. An increase in the activity of phosphoenolpyruvate carboxylase, involved in anaplerotic fixation of CO2 into organic acids, was measured in root extracts of Cd-exposed plants. Also, significant increases in the activities of several enzymes from the Krebs cycle were measured in root extracts of tomato plants grown with Cd. In leaf extracts, significant increases in citrate synthase, isocitrate dehydrogenase and malate dehydrogenase activities were also found at 100 μM Cd, whereas fumarase activity decreased. These data suggest that at low Cd supply (10 μM) tomato plants accumulate Cd in roots and this mechanism may be associated to an increased activity in the PEPC–MDH–CS metabolic pathway involved in citric acid synthesis in roots. Also, at low Cd supply some symptoms associated with a moderate Fe deficiency could be observed, whereas at high Cd supply (100 μM) effects on growth overrule any nutrient interaction caused by excess Cd. Cadmium excess also caused alterations on photosynthetic rates, photosynthetic pigment concentrations and chlorophyll fluorescence, as well as in nutrient homeostasis.  相似文献

The objective of this study was to assess the effect of different Cd and Si concentrations on the maize plants. The following Cd and/or Si treatments were used: 5 Cd; 10 Cd; 100 Cd; 5 Cd + 0.08 Si; 10 Cd + 0.08 Si; 100 Cd + 5 Si treatments (Cd concentration in μM, Si concentration in mM). The plant growth, photosynthetic pigments content, antioxidant enzymes activities (POX, SOD, CAT), Cd and Si accumulation, translocation and cell wall deposition of the maize plants was observed. Changes in the endodermal cell walls development and late metaxylem elements lignification due to Cd and/or Si treatment were also evaluated. The negative effect of Cd (5 and 10 μM) on the growth parameters was alleviated by Si at 0.08 mM. The positive effect of Si was not observed at higher Cd and Si concentrations. This indicates that the alleviating effect of Si on Cd toxicity depends on the Cd and Si concentrations. Plants responded to Cd toxicity by an increase of antioxidant enzyme activity. Silicon addition in Cd + Si treatment stimulated an increase in the activity of antioxidant enzymes in comparison with the Cd treatment. Chlorophyll and carotenoid content in the Cd treated plants was not significantly affected by Si. The young maize plants retained much more Cd in their roots as they translocated into the shoots. 5 Cd + 0.08 Si and 10 Cd + 0.08 Si treatments correlated with an increase in Cd concentration in the roots and shoots, and in the cell walls. Silicon caused a slight decrease of the Cd translocation into the shoots in 5 Cd + 0.08 Si and 10 Cd + 0.08 Si treatments. Negative correlation between the root Cd cell wall deposition and Cd translocation was observed. Cadmium and/or Si altered root anatomy. Cadmium enhanced suberin lamellae development and late metaxylem lignification; silicon in Cd + Si treatments accelerated suberin lamellae deposition and enhanced the tertiary endodermal cell walls formation in comparison with Cd treatments. Negative correlation between the endodermal cell walls development and Cd translocation was observed.  相似文献

Seedlings of wheat (Triticum aestivum L.) cultivars Jing 411, Jinmai 30 and Yangmai 10 were exposed to 0, 10, 20, 30, 40 or 50 μM of CdCl2 in a solution culture experiment. The effects of cadmium (Cd) stress on wheat growth, leaf photon energy conversion, gas exchange, and Cd accumulation in wheat seedlings were investigated. Gas exchange was monitored at 3, 9, 24 days after treatment (DAT). Growth parameters, chlorophyll content, leaf chlorophyll fluorescence, and Cd concentration in shoot and root were measured at 24 DAT. Seedling growth, gas exchange, chlorophyll content, chlorophyll fluorescence parameters were generally depressed by Cd stress, especially under the high Cd concentrations. Cd concentration and accumulation in both shoots and roots increased with increasing external Cd concentrations. Relationships between corrected parameters of growth, photosynthesis and fluorescence and corrected Cd concentrations in shoots and roots could be explained by the regression model Y = K/(1 + exp(a + bX)). Jing 411 was found to be Cd tolerant considering parameters of chlorophyll content, photosynthesis and chlorophyll fluorescence in which less Cd translocation was from roots into shoots. The high Cd concentrations were in shoots and roots in Yangmai 10 which has been found to be a relative Cd tolerant cultivar in terms of most growth parameters.  相似文献

Binding of Cd to non-specific metal-binding peptides (phytochelatins)in birch roots has been suggested as an explanation for toleranceto Cd toxicity in birch (Betula pendula). In the present study,the tolerance of birch roots to Cd was further investigatedby using buthionine sulphoximine (BSO) as an inhibitor of phytochelatinsynthesis. Birch seedlings, grown in nutrient solution at pH4.2, were exposed to 0 or 2 µM CdCl2 combined with 0 or0.1 mM BSO for 6 d. Plant growth (fresh weight increase andshoot to root dry weight ratio) and the nutrient compositionin fine roots, whole roots and shoots were determined. The effectsof Cd on growth confirms the results of earlier studies on birch,suggesting a reduced shoot growth, but preserved or stimulatedroot growth. When Cd and BSO were combined, overall plant growthwas severely reduced. BSO was also shown to aggravate Cd-inducedreductions of root and shoot concentrations of K, Ca and Mgbut to impede the accumulation of Cd. The results suggest that phytochelatins participate in protectingthe root against Cd interferences with growth, possibly by restrictingCd-induced changes in the nutrient composition of the plant. Soil pollution is a world-wide problem, with heavy metals being a major part of the concern. To investigate the effect of temperature on cadmium (Cd) uptake and translocation, as well as Cd tolerance in wild and cultivated species of safflower, a hydroponic experiment was conducted under controlled conditions. The responses of four wild genotypes (Isfahan, Arak, Azari, and Shiraz) and four cultivated genotypes (AC-Sterling, 2811, Saffire, and C111) of safflower to nine levels of CdCl2 (0, 0.5, 1, 5, 10, 20, 50, 100, and 500 μM) in solution were examined under two temperatures (18 and 23 °C). Cadmium sensitivity was determined using the Weibull model on the total dry weight of the plants. Cadmium uptake and translocation were analyzed on 1 μM Cd treated plants. Results revealed that safflower genotypes differed in terms of uptake, translocation, and tolerance to Cd, with AC-Sterling and Arak indicating the most and the least tolerance to Cd, respectively. Relative Cd uptake and Cd concentration in roots and shoots increased with an increase in temperature in all genotypes, with the exception of AC-Sterling. Net accumulation of Cd via root increased with an increase in temperature for the wild Azari and the cultivated 2811, Saffire, and C111, though it decreased for the rest of genotypes. Cadmium translocation to shoots significantly increased with increased temperature in all genotypes. Cadmium translocation from roots to shoots in cultivated genotypes was significantly greater than in wild genotypes. Root Cd concentration in wild genotypes was significantly greater than in cultivated genotypes. It seems that wild and cultivated species of safflower differ in their response to Cd. Furthermore, temperature may affect the plant's tolerance to Cd, probably through accompanying changes in Cd uptake and translocation from root to shoot.  相似文献

In order to analyze the adaptation potential of tomato shoots to a sudden increase in Cd concentration, tomato plants (Solanum lycopersicum L. var. Ailsa Craig) were exposed under controlled environmental conditions to a high dose of this heavy metal (250 μM CdCl2>) in nutrient solution for 7 and 14 d. Both root and shoot growth was completely inhibited but all plants remained alive until the end of the treatment. Cell viability remained unaffected but the activity of the mitochondrial alternative pathway was stimulated by Cd stress at the expense of the cytochrome pathway. Cadmium concentration was higher in roots than in shoots and a decrease In the rate of net Cd translocation was noticed during the second week of stress. Cadmium decreased both leaf conductance (g1>) and chlorophyll concentration. However, the effect on net CO2 assimilation remained limited and soluble sugars accumulated in leaves. Photochemical efficiency of PSll (FvlFm) was not affected despite a decrease in the number of reaction centers and an inhibition of electron transfer to acceptors of PSII. It is concluded that tomato shoot may sustain short term exposure to high doses of cadmium despite growth inhibition. This property implies several physiological strategies linked to both avoidance and tolerance mechanisms.  相似文献

Growth parameters and cadmium accumulation were investigated in alfalfa seedlings treated with 10 μM salicylic acid (SA) at the beginning of seed imbibition. Shoot and root growths were accelerated by SA treatment and suppressed by Cd both in presence and absence of SA. Cd accumulation was stimulated by SA in alfalfa seedlings in dependence of the treatment duration. K, Mg, Ca and Fe contents in roots are decreased in the presence of Cd alone, while SA induces a decrease of Mg, Ca and Fe. Shoot K, Mg and Ca concentrations are increased by Cd only in the absence of SA, while SA induces also an increase of these concentrations, but only in the absence of Cd. High negative correlation of Cd concentration with K and Ca concentrations in root indicates a competition for the same carrier not regulated by SA. Positive correlation between Cd and Mg concentrations in shoots, which is decreased by SA pre-treatment, together with the increase of positive correlation between Cd and Fe concentrations in shoots under the influence of SA, indicates a possible mechanism of SA action through maintenance of ionic homeostasis.  相似文献

The effect of cadmium on growth parameters of seedlings of maize, rye and wheat as well as the role of phytochelatins in Cd detoxication in these species were studied. Cadmium was found to inhibit root growth and decrease fresh weight and water content in roots and shoots of the studied plants. Although a considerably lower Cd accumulation was shown in maize seedlings than in other species, they were characterized by the highest sensitivity to cadmium. Among γ-Glu-Cys peptides synthetized by plant species, phytochelatins — glutathione derivatives predominated. In maize they were synthetized in amounts sufficient for binding the total pool of the metal taken up, and the detoxication mechanism was localized in their roots. Larger amounts of cadmium were accumulated in roots of wheat and rye, but the quantity of the formed γ-Glu-Cys peptides seems insufficient for detoxication of the metal.  相似文献

采用营养液培养法研究了Cd处理时间对有铁膜和无铁膜水稻根表吸附Cd及植株吸收Cd动态变化的影响.水稻根表铁膜由50 mg·L-1 Fe2+(Fe50)诱导形成.供试植株在含10 μmol·L-1Cd的营养液中生长不同时间后收获.结果表明, 随Cd处理时间的延长,无铁膜和有铁膜处理水稻根表DCB-Cd含量均为先增加后减少,Cd处理2 h达到最高,之后逐渐下降并趋于稳定. 根系和地上部Cd含量均持续上升,Cd处理8 h前增加缓慢,8 h后增加幅度加大.有铁膜水稻根系和地上部Cd含量增加幅度均低于无铁膜水稻.有铁膜处理DCB-Cd含量、根系和地上部Cd含量均低于无铁膜处理.表明铁膜不影响水稻各部分Cd含量随时间的变化趋势;不同Fe处理之间根系和地上部Cd含量的差异可能与根系含Fe量有关.  相似文献

Endophytic fungi are a group of fungi that live asymptomatically inside plant tissue. These fungi may increase host plant tolerance to biotic and abiotic stresses. The effect of Neotyphodium endophytes in two grass species (Festuca arundinacea and Festuca pratensis) on cadmium (Cd) tolerance, accumulation and translocation has been our main objective. The plants were grown in a hydroponic system under different Cd concentrations (0, 5, 10, and 20 mg L(-1)) for 6 weeks. They were also grown in soil spiked with different concentrations of Cd (0, 10, 20, and 40 mg kg(-1)) for 2 months. The results from all Cd treatments showed higher biomass production (12-24%) and higher potential to accumulate Cd in roots (6-16%) and shoots (6-20%) of endophyte-infected plants than endophyte-free plants. Cadmium accumulation by plants indicated that the grasses were capable of Cd hyperaccumulation, a property that was augmented after endophyte infection. Maximum photochemical efficiency of photosystem II (Fv/Fm) revealed that Cd stress was significantly reduced in endophyte-infected plants compared to non-infected ones.  相似文献

The effects of cadmium (Cd) administration on primary root growth, mitotic activity of apical meristems, mitotic aberrations and percentage of nucleus ploidy classes of differentiated roots were examined in Pisum sativum L. cv. Frisson. Cadmium caused a reduction of root length related to concentration, with an almost complete block of growth in plants treated with 250 μM Cd, from 24 h of treatment. Root lengthening is generally related to apical meristem activity, however, in the examined pea plants, mitotic activity was suppressed by 2.5 and 25 μM Cd treatment, while the highest Cd concentration, 250 μM, caused the occurrence of mitotic figures consisting almost exclusively of prophases. The lack of relation between root lengthening and mitotic activity was explained by the meristematic activity in the first period of treatment and by a different cell elongation. Lower (0.25, 0.5 and 1 μM), non-blocking Cd concentrations induced a number of mitotic aberrations, mainly consisting of sticky metaphases and anaphase bridges, whose frequency increased with Cd concentration. Besides, Cd induced variations of the percentages of nucleus populations in the differentiated roots, increasing the percentage of 4C nuclei and decreasing that of 2C. The mechanisms involved in the nuclear response to Cd, and the possible relations between Cd alteration of meristem cell activity and nuclear ploidy of differentiated cells are discussed.  相似文献

近年关于镉(Cd)单一污染对植株伤害的报道比较多,但是自然背景下,污染多呈复合型,而双酚A(BPA)是一种新兴的环境激激素,故研究二者复合污染具有一定理论价值和实践意义。本研究综述了Cd和BPA胁迫下植物根系的形态学变化及生理生化改变,以及镉和双酚A在植物体内的代谢过程和其对植物毒性机制的研究进展,并提出有待进一步研究的内容。研究表明Cd和BPA对根系生长的影响可能与其影响植物根系活力,矿质代谢和根系保护酶有关,低浓度的Cd和BPA对植株干质量、根系总长度、根系表面积、根体积和根系活力都有促进作用,高浓度则可能存在抑制效应。  相似文献

The possible interaction of two stresses, UV-B radiation and cadmium, applied simultaneously, was investigated in Brassica napus L. cv. Paroll with respect of chlorophyll fluorescence, growth and uptake of selected elements. Plants were grown in nutrient solution containing CdCl2, (0, 0.5, 2 or 5 M) and irradiated with photosynthetically active radiation (PAR, 400-700 nm, 800 mol m-2 s-1) with or without supplemental ultraviolet-B radiation (UV-B, 280-320 nm, 15 kJ m-2 d-1, weighted irradiance). After 14 d of treatment, the most pronounced effects were found at 2 and 5 M CdCl2 with and without supplemental UV-B radiation. Exposure to cadmium significantly increased the amount of Cd in both roots and shoots. In addition, increases occurred in the concentrations of Fe, Zn, Cu, and P in roots, while K was reduced. In shoots the S content rose significantly both in the presence and absence of UV-B radiation, while significant increases in Mg, Ca, P, Cu, and K occurred only in plants exposed to Cd and UV-B radiation. Manganese decreased significantly under the combined exposure treatment. The rise in S content may have been due to stimulated glutathione and phytochelatin synthesis. Cadmium exposure significantly decreased root dry weight, leaf area, total chlorophyll content, carotenoid content, and the photochemical quantum yield of photosynthesis. As an estimation of energy dissipation processes in photosynthesis, non-photochemical quenching (qNPQ) was measured using a pulse amplitude modulated fluorometer. The qNPQ increased with increasing Cd, while the combination of cadmium and UV-B reduced the qNPQ compared to that in plants exposed only to cadmium or UV-B radiation. The chlorophyll a:b ratio showed a reduction with UV-B at no or low Cd concentrations (0 M, 0.5 M CdCl2), but not at the higher Cd concentrations used (2 M, 5 M CdCl2). Thus in some instances there appeared to be a UV-B and Cd interaction, while in other plants response could be attributed to either treatment alone.Keywords: Brassica napus, cadmium, ultraviolet-B radiation.   相似文献

Tobacco (Nicotiana tabacum L.) is a tolerant species that accumulates cadmium. We studied the effect of Cd (0, 10, 25, 50, 100 μM) on growth parameters, chlorophyll and proline contents, enzymatic antioxidative response and lipid peroxidation of tobacco plants grown in hydroponic culture for 11 days to clarify the strategy of plant response against oxidative stress caused by this heavy metal. Cadmium accumulated more in roots than in shoots. Plant growth was not significantly affected by the cadmium concentrations used. Young leaves were more affected, showing visible chlorosis and a significant decrease in chlorophyll content at high Cd concentrations. Dry weight of both leaves and roots increased indicating a lower capacity for roots to absorb water. An increase in malondialdehyde levels was observed, indicating that lipid peroxidation occurred as a result of ROS formation. The activity of guaiacol peroxidase in leaves increased, indicating that it was very important in the scavenging of H2O2, while superoxide dismutase activity only increased in old leaves. Ascorbate peroxidase showed constant activity levels in tobacco leaves, suggesting that the ascorbate–glutathione pathway was less important as a defense mechanism.  相似文献

Biomass production, leaf number and area, photosynthetic and dark respiration rates, leaf concentration of photosynthetic pigments, nitrate reductase activity, as well as cadmium concentrations in leaves, stem, and roots were measured in poplar clones PE 4/68, B-229, 665, and 45/51. Plants were grown hydroponically under controlled conditions and treated with two different cadmium (Cd) concentrations (10(-5) and 10(-7) M) in the same background solution (Hoagland's solution). The presence of Cd did not cause serious disturbance of growth and physiological parameters in the studied poplar clones. Cd concentrations in plant tissues reflected external concentrations. In treated plants, root contents increased from 38.57 to 511.51 ppm, leaf contents from 0.91 to 7.50, while stem contents ranged from 1.37 to 9.50 ppm.  相似文献

Effects of zinc (12–180 μM) alone and in mixtures with 12 μM Cd on metal accumulation, dry masses of roots and shoots, root respiration rate, variable to maximum fluorescence ratio (FV/FM), and content of photosynthetic pigments were studied in hydroponically cultivated chamomile (Matricaria recutita) plants. The content of Zn in roots and shoots increased with the increasing external Zn concentration and its accumulation in the roots was higher than that in the shoots. While at lower Zn concentrations (12 and 60 μM) the presence of 12 μM Cd decreased Zn accumulation in the roots, treatment with 120 and 180 μM Zn together with 12 μM Cd caused enhancement of Zn content in the root. Presence of Zn (12–120 μM) decreased Cd accumulation in roots. On the other hand, Cd content in the shoots of plants treated with Zn + Cd exceeded that in the plants treated only with 12 μM Cd. Only higher Zn concentrations (120 and 180 μM) and Zn + Cd mixtures negatively influenced dry mass, chlorophyll (Chl) and carotenoid content, FV/FM and root respiration rate.
