In the study, two Lactobacillus cultures i.e. L. casei (NK9) and L. fermentum (LF) were studied for their proteolytic activity, di and tripeptidase activity, ACE-inhibitory activity and peptides production under optimized growth condition from fermented goat milk (Capra aegagrus hircus). NK9 and LF were found to be a strong proteolytic culture with 2.0% rate of inoculation after 48 h. LF (10 kDa retentate) produced maximum peptides among all the retentates of the fermented goat milk. Goat milk fermented with NK9 (10 kDa permeates) exhibited peptide sequence i.e. AFPEHK which had ACE inhibitory activity, matched with goat milk protein databases of AHTPDB. However, L. casei (NK9) and L. fermentum (LF) could be explored for the production of ACE inhibitory peptides from fermented goat milk. 相似文献

目的:研究生物活性肤SSDI的固相合成工艺,并为大规模合成目标肽提供理论依据.方法:采用固相合成法,原料氨基酸以Fmoe形式保护,用Wang树脂为载体,经1-氧3-双二甲胺羰基苯骈三氮唑四氟化硼盐(TBTU)\1-羟基-苯并-三氮唑(HOBt)\二-异丙基乙胺(DIEA)混合试剂缩合,20%哌啶的DMF溶液脱保护,用三氟乙酸\茴香硫醚\巯基乙醇\苯酚\水混合作为切割试剂将多肽从Wang树脂上切割下来.结果:多肽粗品的得率高达70%,经RP-HPLC纯化,可获得纯度在98%以上的目标肽,经MALDI-MS质谱鉴定其分子量与理论值一致.结论:此合成方法操作简单,产品得率高,适合大规模合成目标肽. 相似文献

Abstract: The subcellular distribution of cholecystokinin (CCK), especially its exact localization within the synaptosome, was studied in the rat cerebral cortex. Highest CCK-like bioactivity was measured in the synaptic membrane fractions, paralleling the distribution of (Na+ + K+ )-dependent ATPase. Banana (Musa spp.) is an ancient and popular fruit plant with highly
nutritious fruit. The pseudo-stem of banana represents on average 75% of the total
dry mass but its valorization as a nutritional and industrial by-product is limited.
Recent advances in metabolomics have paved the way to understand and evaluate
the presence of diverse sets of metabolites in different plant parts. This study
aimed at exploring the diversity of primary and secondary metabolites in the
banana pseudo-stem. Hereby, we identified and quantified 373 metabolites from
a diverse range of classes including, alkaloids, flavonoids, lipids, phenolic acids,
amino acids and its derivatives, nucleotide and its derivatives, organic acids, lignans and coumarins, tannins, and terpene using the widely-targeted metabolomics
approach. Banana pseudo-stem is enriched in metabolites for utilization in the
food industry (L-lysine and L-tryptophan, L-glutamic acid, Phenylalanine, Palmitoleic acid, α-Linolenic acid, and Lauric acid, and Adenine) and pharmaceutical
industry (Guanosine and Cimidahurinine, Bergapten, Coumarins, Procyanidin
A2, Procyanidin B1, Procyanidin B3, Procyanidin B2, and Procyanidin B4, Asiatic acid). The metabolome of banana pseudo-stem with integration across multiomics data may provide the opportunity to exploit the rich metabolome of banana

论述了运用生物信息学方法和计算机技术,快速从由蛋白酶模拟酶解蛋白质而产生的大量未知生物活性的系列肽中,预测有生物活性的肽,以实现生物活性肽功能的预测。主要建立了生物活性肽数据库,应用已有生物活性肽作为序列比对的标准,实行大量未知生物活性的系列肽自动无人值守的和已知生物活性的肽序列比对查询,以发现新物种中包括动物和植物的具有新的生物活性的功能肽。应用该软件系统AQS成功地预测并发现了造血细胞增殖肽、成骨细胞生长肽以及高血压押制肽。 相似文献

Background
Current Ebola virus (EBOV) detection methods are costly and impractical for epidemic scenarios. Different immune-based assays have been reported for the detection and quantification of Ebola virus (EBOV) proteins. In particular, several monoclonal antibodies (mAbs) have been described that bind the capsid glycoprotein (GP) of EBOV GP. However, the currently available platforms for the design and production of full-length mAbs are cumbersome and costly. The use of antibody fragments, rather than full-length antibodies, might represent a cost-effective alternative for the development of diagnostic and possibly even therapeutic alternatives for EBOV.Methods/Principal Findings
We report the design and expression of three recombinant anti-GP mAb fragments in Escherichia coli cultures. These fragments contained the heavy and light variable portions of the three well-studied anti-GP full-length mAbs 13C6, 13F6, and KZ52, and are consequently named scFv-13C6, scFv-13F6, and Fab-KZ52, respectively. All three fragments exhibited specific anti-GP binding activity in ELISA experiments comparable to that of full-length anti-GP antibodies (i.e., the same order of magnitude) and they are easily and economically produced in bacterial cultures.Conclusion/Significance
Antibody fragments might represent a useful, effective, and low cost alternative to full-length antibodies in Ebola related capture and diagnostics applications. 相似文献19.

The porcine cathelicidin PR-39 is a host defence peptide that plays a pivotal role in the innate immune defence of the pig against infections. Besides direct antimicrobial activity, it is involved in immunomodulation, wound healing and several other biological processes. In this study, the antimicrobial- and immunomodulatory activity of PR-39, and N- and C-terminal derivatives of PR-39 were tested. PR-39 exhibited an unexpected broad antimicrobial spectrum including several Gram positive strains such as Bacillus globigii and Enterococcus faecalis. Of organisms tested, only Staphylococcus aureus was insensitive to PR-39. Truncation of PR-39 down to 15 (N-terminal) amino acids did not lead to major loss of activity, while peptides corresponding to the C-terminal part of PR-39 were hampered in their antimicrobial activity. However, shorter peptides were all much more sensitive to inhibition by salt. Active peptides induced ATP leakage and loss of membrane potential in Bacillus globigii and Escherichia coli, indicating a lytic mechanism of action for these peptides. Finally, only the mature peptide was able to induce IL-8 production in porcine macrophages, but some shorter peptides also had an effect on TNF-α production showing differential regulation of cytokine induction by PR-39 derived peptides. None of the active peptides showed high cytotoxicity highlighting the potential of these peptides for use as an alternative to antibiotics. 相似文献
