Saline washed red blood cells of the toadfish convert [1-14C] arachidonic acid to products that cochromatograph with prostaglandin E2 and prostaglandin F. This synthesis is inhibited by indomethacin (10 μg/ml). Conversion of arachidonic acid to prostaglandin E2 was confirmed by mass spectrometry. When saline washed toadfish red blood cells were incubated with a mixture of [1-14C]-arachidonic acid and [5,6,8,9,11,12,14,15,-3H]-arachidonic acid, comparison of the isotope ratios of the radioactive products indicated that prostaglandin F was produced by reduction of prostaglandin E2. The capacity of toadfish red blood cells to reduce prostaglandin E2 to prostaglandin F was confirmed by incubation of the cells with [1-14C] prostaglandin E2.  相似文献

Prostaglandin F (PGF) is one of the most common metabolites of arachidonic acid (AA) in rat brain. When administered intracerebroventricularly (i.c.v.) to rats, both AA and PGF exert dose-related hypertensive, tachycardic and hyperthermic effects. Metabolic alterations in the endogenous formation of some prostaglandins in the brain-stem of spontaneously hypertensive rats (SHR) have been reported. Therefore the central effects of AA and PGF on blood pressure, heart rate and body temperature were studied both in SHR and normotensive Wistar rats (NR) under urethane-anaesthesia. The hypertensive effect of AA i.c.v. (0.01–100 μg/rat) was larger in magnitude in SHR than in NR, but there was no significant difference in the AA-induced changes of heart rate and body temperature between the groups. Pretreatment of NR with sodium meclofenamate (1 mg/rat i.c.v.) antagonised the central effects of AA indicating that these effects are not due to AA itself but to its conversion to prostaglandins. Unlike the effects of AA, the central hypertensive, tachycardic and hyperthermic responses to PGF (0.5–50 μg/rat i.c.v.) were significantly attenuated in SHR. The present results obtained with AA are compatible with the previous assumption that the synthesis of prostaglandins in the brain of SHR might differ from that in NR. The results also demonstrate that the central effects of PGF are reduced in SHR.  相似文献

Prostacyclin (PGI2), prostaglandin E2 (PGE2) and prostaglandin F2∝ (PGF2∝) were tested here in unanesthetized male Sprague-Dawley rats for their effects on the cardiovascular system as mediated by the Central nervous system. Cannulae were chronically implanted into the third cerebral ventricle, femoral arteries and femoral veins of rats. Both PGE2 and PGF2∝ induced increased arterial blood pressure and tachycardia by an action on the central nervous system. The changes seen with PGE2 were larger than those observed with PGF2∝. Only transient depressor effects were seen with PGI2 and these changes appeared to be due to the leakage of the substance into the peripheral vascular system.  相似文献

Prostaglandin F and E2 contents in human cerebrospinal fluid were determined by the radioisotope dilution method. The mean values of PGF and PGE2 of men were 9.8±0.87 ng/ml and 6.5±1.39 ng/ml, respectively. Those of women were 8.3±1.4 ng/ml and 6.9±1.72 ng/ml, respectively. The correlation between age and PG was significantly with PGE2 of men and with PGF of women.  相似文献

Proinflammatory cytokines may promote preterm labor in the setting of intrauterine infection. Tumor necrosis factor (TNF) and interleukin-1 (IL-1) synergistically stimulate the production of prostaglandin E2 (PGE2) by amnion cells. Transforming growth factor-β (TGF-β) inhibits the cytokine-stimulated PGE2 production. In the present study, we investigated the binding of IL-1β on human amnion cells in culture. Untreated amnion cells possessed 540±60 IL-1 receptors per cell, with a dissociation constant of 1.4±0.4 nM. Cells treated with TGF-β1 (10 ng/ml) had 570±110 receptors per cell. TNF-α (50 ng/ml) increased the number of IL-1 receptors to 2930±590. TGF-β1 inhibited the receptor upregulation by TNF-α. Cells treated with TGF-β1 and TNF-α expressed 1140±590 receptors per cell. The binding affinity was not changed by the cytokines. IL-1 receptor antagonist (IL-1ra) inhibited the stimulation of amnion cell PGE2 production by IL-1β, but not by TNF-α. Amnion cells secreted large amounts of IL-1ra (1.1±0.3 ng/105 cells). Treatment of the cells with TGF-β1 or TNF-α did not affect the release of IL-1ra. We conclude that IL-1 receptor expression is an important step in the regulation of the effects of cytokines on amnion cell PGE2 production.  相似文献

Prostaglandin F formation caused by pentametylenetetrazol convulsions was studied as a function of the duration, the doses of the convulsant and the intensity of the seizures. It was shown by the statistical analysis of the results in the case of clonic convulsions that the amount of synthetized PGF did not depend on the doses of convulsant, while close relation existed between the duration and the PGF production. At the same time, during tonic convulsions lasting longer than 50 sec, no more increase in the PGF content of the brain was observed. An experimental model is suggested to study in vivo the mechanisms regulating the brain's prostaglandin biosynthesis.Pretreatment of the animals with reserpine did not affect the rate of convulsion-induced PGF-formation.  相似文献

Background and purpose

Prostaglandin ethanolamides or prostamides are naturally occurring neutral lipid derivatives of prostaglandins that have been shown to be synthesised in vivo following COX-facilitated oxygenation of arachidonoyl ethanolamine (anandamide). Although the actions of prostaglandins have been extensively studied, little is known about the physiological or pathophysiological effects of prostamides. Since prostaglandin E2 has potent immunosuppressive/immunomodulating actions, the aim of the present study was to determine whether the derivative, prostaglandin E2 ethanolamide (PGE2-EA), could modulate the production of the pro-inflammatory cytokine tumour necrosis factor-α in human blood and human monocytic cells and indicate whether this action involved the same receptor systems/signals as PGE2.

Experimental approach

Whole human blood, monocytes isolated from the blood or the human monocytic cell line THP-1 was incubated with LPS and the level of TNF-α produced was measured by ELISA assay. The actions of PGE2-EA were assessed on the LPS-induced TNF-α release. In addition, in order to ascertain the receptors involved, the levels of cyclic AMP in cells were measured in monocytes and THP-1 cells in response to PGE2-EA and directly compared to those of PGE2. The effect of PGE2-EA on the binding of radiolabelled PGE2 to cells was also measured. Cells were incubated with radiolabelled arachidonic acid and ethanolamine to estimate the production of PGE2-EA.

Key results

PGE2-EA potently suppressed TNF-α production in blood, monocytes and the cell line THP-1 in a concentration-dependent manner. This occurred via cyclic AMP pathways as indicated by agents which interfere with these pathways and also direct ligand binding experiments. It was also shown that the cells were able to endogenously produce PGE2-EA.

Conclusions and implications

This study reports that PGE2-EA can downregulate the production of TNF-α by human mononuclear cells in response to an immune stimulus, i.e. LPS-activated TLR4, and that this appears to occur via a cAMP-dependent mechanism that most likely involves binding to the EP2 receptor.  相似文献

Levels of prostaglandin F (PGF) in the amniotic fluid were determined by radioimmunoassay. Concentrations of the prostaglandin were relatively constant between 15 and 35 weeks' gestation, but an increase was observed after 36 weeks. The rise was continued up to 44 weeks. A still greater elevation of PGF levels was recorded during labour, when the levels were related to the amount of cervical dilatation.Amniotic fluid PGF levels in toxaemia of pregnancy did not significantly differ from those found in normal pregnancy.  相似文献

It has been demonstrated that apolipoprotein M (APOM) is a vasculoprotective constituent of high density lipoprotein (HDL), which could be related to the anti-atherosclerotic property of HDL. Investigation of regulation of APOM expression is of important for further exploring its pathophysiological function in vivo. Our previous studies indicated that expression of APOM could be regulated by platelet activating factor (PAF), transforming growth factors (TGF), insulin-like growth factor (IGF), leptin, hyperglycemia and etc., in vivo and/or in vitro. In the present study, we demonstrated that palmitic acid could significantly inhibit APOM gene expression in HepG2 cells. Further study indicated neither PI-3 kinase (PI3K) inhibitor LY294002 nor protein kinase C (PKC) inhibitor GFX could abolish palmitic acid induced down-regulation of APOM expression. In contrast, the peroxisome proliferator-activated receptor beta/delta (PPARβ/δ) antagonist GSK3787 could totally reverse the palmitic acid-induced down-regulation of APOM expression, which clearly demonstrates that down-regulation of APOM expression induced by palmitic acid is mediated via the PPARβ/δ pathway.  相似文献

Analysis of prostaglandin F (PGF) in urine is a useful indicator of renal prostaglandin synthesis. A mass fragmentographic method for PGF analysis in human urine was developed using [3,3,4,4-2H4]PGF as an internal standard and carrier. PGF was extracted from urine (20 ml) with chloroform, purified by preparative thin-layer chromatography and converted to the methyl ester trimethylsilyl ether before analysis by gas chromatograph—mass spectrometry. The specificity of the urine analysis was demonstrated by retention time and the use of two pairs of fragments m/e 494/498 and 513/517 with the same results. The coefficient of variation for duplicate analysis averaged 12.6%, n = 17. Urine from recumbent women contained 4.9 ± 2.6 (S.D.) ng/ml or 4.1 ± 1.0 ng PGF per mg creatinine (n = 10) with little diurnal variation. Male urine contained 5.0 ± 2.7 (S.D.) ng/ml or 3.7 ± 2.1 ng/mg creatinine (n = 10). Similar concentrations were found in boys and in girls. These observations indicate that urinary PGF originates from the kidneys with little contribution from the male accessory sexual glands. This method can also be applied to analysis of PGF in rabbit urine.  相似文献

Calcium-independent phospholipase A2 group VIA (iPLA2β) releases docosahexaenoic acid (DHA) from phospholipids in vitro. Mutations in the iPLA2β gene, PLA2G6, are associated with dystonia-parkinsonism and infantile neuroaxonal dystrophy. To understand the role of iPLA2β in brain, we applied our in vivo kinetic method using radiolabeled DHA in 4 to 5-month-old wild type (iPLA2β+/+) and knockout (iPLA2β−/−) mice, and measured brain DHA kinetics, lipid concentrations, and expression of PLA2, cyclooxygenase (COX), and lipoxygenase (LOX) enzymes. Compared to iPLA2β+/+ mice, iPLA2β−/− mice showed decreased rates of incorporation of unesterified DHA from plasma into brain phospholipids, reduced concentrations of several fatty acids (including DHA) esterified in ethanolamine- and serine-glycerophospholipids, and increased lysophospholipid fatty acid concentrations. DHA turnover in brain phospholipids did not differ between genotypes. In iPLA2β−/− mice, brain levels of iPLA2β mRNA, protein, and activity were decreased, as was the iPLA2γ (Group VIB PLA2) mRNA level, while levels of secretory sPLA2-V mRNA, protein, and activity and cytosolic cPLA2-IVA mRNA were increased. Levels of COX-1 protein were decreased in brain, while COX-2 protein and mRNA were increased. Levels of 5-, 12-, and 15-LOX proteins did not differ significantly between genotypes. Thus, a genetic iPLA2β deficiency in mice is associated with reduced DHA metabolism, profound changes in lipid-metabolizing enzyme expression (demonstrating lack of redundancy) and of phospholipid fatty acid content of brain (particularly of DHA), which may be relevant to neurologic abnormalities in humans with PLA2G6 mutations.  相似文献

The following experiments were designed in order to examine the inter-relationships of various prostaglandins (PG's) and the adrenergic nervous system, in conjunction with blood pressure and heart rate responses, in vivo. Stimulation of the entire spinal cord (50v, 0.3–3 Hz, 1.0 msec) of the pithed rat increased blood pressure, heart rate and plasma epinephrine (EPI) and norepinephrine (NE) concentration (radioenzymatic-thin layer chromatographic assay). Infusion of PGE2(10–30 μg/kg. min, i.v.) suppressed blood pressure and heart rate responses to spinal cord stimulation while plasma EPI (but not NE) was augmented over levels found in control animals. PGI2 (0.03–3.0 μg/kg. min, i.v.) suppressed the blood pressure response to spinal cord stimulation without any effect on heart rate or the plasma catecholamine levels. PGE2 and PGF2α(10–30 μg/kg. min, i.v.) did not change the blood pressure, heart rate or plasma EPI and NE responses to the spinal cord stimulation although PGF2α disclosed an overall vasopressor effect during the pre-stimulation period. At the pre-stimulation period it was also observed that PGE2, PGF2α and PGI2, had a positive chronotropic effect on the heart rate, the cardiac accelerating effect of PGE2 was not abolished by propanolol. These in vivo studies suggest that in the rat, PGE2 and PGI2 modulate sympathetic responses, primarily by interaction with the post-synaptic elements — PGE2 on both blood vessels and the heart and PGI2 by acting principally on blood vessels.  相似文献

Recent evidence suggests a possible role for pyridoxal 5′-phosphate, the active physiological form of vitamin B6, in steroid hormone action (1–3). We now report that 5′-deoxypyridoxal, a synthetic vitamin B6 antagonist, causes a rapid and complete loss of dexamethasone receptor binding in cytosol preparations in whole HeLa S3 cells or in rat thymocytes. This effect is concentration and time dependent, and is specific for 5′-deoxypyridoxal. In whole cell incubations of either HeLa S3 cells or rat thymocytes at 37°C, a 50% reduction in [3H]dexamethasone binding was observed in the presence of 0.25 mM 5′-deoxypyridoxal. Cytotoxicity was not evident in rat thymocytes or HeLa S3 cells incubated with 3.0mM or 1.0mM 5′-deoxypyridoxal respectively. Pyridoxal 5′-phosphate, 5′-deoxypyridoxine and 5′-deoxypyridoxamine were ineffective in causing a loss of steroid receptor binding. These studies suggest that 5′-deoxypyridoxal may be an effective non-steroidal compound which can effect the binding of glucocorticoids to specific receptor proteins in whole cells.  相似文献

Thirty dairy cows serving as the treated group (Group A) were injected intramuscularly with 100 mcg gonadotropin-releasing hormone (GnRH) at 10 to 16 days postpartum followed by 25 mg prostaglandin F2α (PGF2α) 14 days later. Twenty-nine herdmate dairy cows (Group B) serving as controls were treated in a similar manner using saline injections rather than GnRH or PGF2α treatments. Only cows without obvious uterine infection were assigned to the experimental groups, and any uterine pathology that developed during the treatment interval was treated accordingly following the experimental period. Internal genitalia were evaluated via rectal palpation prior to each injection. Blood samples were collected for progesterone analysis before each injection and at 30 hours following the PGF2α or the second saline injection. Experimental animals were artificially inseminated at the first detected postpartum estrus starting 35 to 40 days following calving. Results indicated evidence of enhanced cyclicity when Group A cows were compared with those in Group B. However, there were no significant differences between the two groups for interval to first observed estrus, interval to first serive, first serive pregnancy rate, services per pregnancy and days open. Furthermore, no difference in the incidence of follicular or luteal cysts, incidence of repeat breeders or number of reproductive culls was observed. From observations in this study, the GnRH and PGF2α treatment scheudule might not be economically beneficial in lactating dairy cows as long as reproductive tract abnormalities are promptly diagnosed and subsequently treated by the attending practitioner.  相似文献
