The temporal changes of potassium (K+) and sodium (Na+) contentsin the growing front of Neurospora crassa (al-2, bd strain)grown on solid medium showed circadian rhythms which persistedfor at least 45 h in the dark. The K+ content reached a maximumat about 10 and 30 h after the transfer from light to darkness,while the Na+ content was at a minimum at these times. Boththe rhythms were set off by the light to dark transition andwere not observed in constant light. The phase of the circadianrhythm of conidiation of this strain was delayed by 5 h by exposureto 50 min of white light (photon fluence rate 20.7 W/m2) 7 hafter the light to dark transition. The effect of riboflavin analogs on blue light responses in a riboflavin mutant of Neurospora crassa was studied. The analogs 1-deazariboflavin and roseoflavin, which have red-shifted absorption, acted as photoreceptors for the photosuppression and phase shifting of circadian conidiation by 540 nm light, but were ineffective as photoreceptors for the induction of carotenoid synthesis. These results provide addtional evidence implicating a flavin photoreceptor for at least two blue light responses of Neurospora.  相似文献

A method has been devised for observing both circadian and noncircadian rhythms in a single wild type strain of Neurospora crassa. This method allows a direct comparison of the properties of the two types of rhythm. The circadian rhythm of conidiation always entrains to a light-dark cycle, damps out in constant light, and has a temperature-compensated period length. The noncircadian rhythm of hyphal branching, expressed by the same strain under different environmental conditions, does not entrain to a light-dark cycle, persists in constant light, and its period length is temperature-dependent. These results suggest that the two rhythms have different underlying mechanisms and demonstrate that the differences in the rhythms previously observed in different strains (patch, band, and clock) are not due to genetic differences between these strains but rather are inherent properties of the rhythms themselves.  相似文献

Mycclia of Neurospora crassa wild type (FE SC no. 853), harvestedduring the exponential phase of growth on defined minimal mediaincorporated glycine-2-14C, serine-3-14C and formate-14C intoproteins, DNA and RNA. Supplementing the growth medium with1 mM glycine increased the flow of glycine and formate carboninto these products. In contrast, this supplement decreasedthe incorporation of serine-14C. When such cultures were preincubatedfor 30 min with adenine, formaldehyde, formate or L-methionine,labelling of the nucleic acids and protein fractions by glycine-2-14Cwas altered. It is concluded that glycine increases the turnoverof C1 units in Neurospora, resulting in greater contributionsof the C-2 in nucleic acid and protein synthesis. (Received May 14, 1977; )  相似文献

The upr-I gene controlling UV sensitivity in Neurospora crassa has been transferred from a microconidial strain into a macroconidial strain. The properties of the strain with respect to UV sensitivity and photoreactivation (PR) have been modified in the macroconidial background leading to an enhancement of resistance to UV by a factor of about 2 and some suggestion of enzymatic PR. The upr-I gene confers sensitivity to nitrous acid to about the same degree as that for UV. However, inactivation of strains carrying the upr-I gene by white light in the presence of photosensitizing dyes is equivalent to that of strains exhibiting normal sensitivity to UV.  相似文献

Phosphatidylcholine is one of the most frequent phospholipid components of the inner mitochondrial membrane ofNeurospora crassa. Quantitative analysis of phospholipids of the wild strain ofNeurospora crassa and of its twocho mutants showed that these strains did not significantly differ in the content of phosphatidylcholine. Mutants cultivated in a medium without choline contained, as compared with the wild strain, an increased amount of phosphatidylserine and a decreased quantity of phosphatidic acid. Respiratory activity increased and sensitivity to inhibitors of respiration changed. It is likely that the presence of choline in the growth medium exerts a regulatory effect on the cell metabolism of these mutants.  相似文献

Two mutants of Neurospora crassa that grow well in a mediumin which the concentration of calcium ions is lower than thatin the medium used for the culture of the wild-type strain havebeen isolated. One mutant strain, ca-101, must be allelic ortightly linked to co-1(19) which was isolated previously. Recombinationfrequencies for ca-1 and cys-9 and for ca-1 and un-1 in linkagegroup I were about 10%, in each case. The other strain, ca-3(105),carried a mutation in another gene, ca-3, which appeared tobe closely linked to ca-1 since the frequency of recombinationbetween these two genes was about 5%. All strains were comparedin terms of period length and phase of the circadian conidiationrhythm with the wild-type strain. The flower-inducing activities of benzoic and salicylic acidsadded to the medium differ with the species (Lemna paucicostataand L. minor), and even with the strains used. The type andpH of the medium used, full or 1/10 strength M medium at pH3.8, 4.4 or 5.1, or 1/2 or 1/20 strength NH4+-free Hutner'smedium at pH 5.0, 6.0 or 7.0, also modify their activity. L.paucicostata, strain 151 is the most sensitive of the strainsused to both benzoic and salicylic acids followed by strain381. Such dramatic flowering responses were not obtained withthe other strains, but even strain 321, reportedly insensitiveto benzoic acid, could be induced to flower by adding benzoicacid to a modification of the medium. Benzoic acid is more effectivethan salicylic acid for all strains of L. paucicostata, butthe contrary is true for two L. minor strains tested. A higherpercentage of flowering is obtained in L. paucicostata in 1/2strength NH4+-free Huter'sn medium than in M medium, exceptfor strain 151. When diluted, both media enhance flowering inall L. paucicostata strains. The parent wild strainNeurospora crassa Em 5297a and three Ni2+ resistantNeurospora crassa mutants have been shown to excrete pyruvate into the culture medium in Ni2+ and Co2+ toxicities. Ni2+ has a more pronounced effect in this regard. The excretion is progressive with growth inhibition and is abolished by Mg2+ in all strains and by Fe3+ partially in the Em strain but not inNeurospora crassa NiR1. Pyruvate, citrate and malate supplementation reverse growth inhibition caused by excess Ni2+, but with concomitant suppression of Ni2+ accumulation. It is suggested that one of the features of Ni2+ toxicity inNeurospora crassa is a derangement in carbohydrate metabolism at step(s) beyond pyruvate and that this is possibly due to decreased invivo activity of Mg2+ dependent processes  相似文献

The met(S2706) mutant (FGSC 4248) of Neurospora crassa was culturedin Vogel's minimal medium with and without L-methionine supplementation.Methionine Stimulated growth but significant mycelial productionalso occurred in minimal medium. The mutant had ability to generatefree and protein methionine from [35S]-cysteine in vivo butthe rate of this synthesis was below that shown by a wild typestrain (Lindegren A, FGSC 853). Cytosolic and mitochondrialfractions of met(S2706) also generated methionine from [3-14C]-serine.Following dialysis, this reaction was stimulated by additionsof tetrahydrofolate and its diglutamate derivative. Folate analysesshowed that the mutant had a folylpolyglutamate concentrationthat was only 10% of that detected in the wild type. Despitethis, mitochondria of met(S2706) contained folates that werelargely polyglutamates and the total mitochondrial folate concentration(ng/mg protein) was comparable to that of the wild type. Assays of folylpolyglutamate synthetase showed that met(S2706)had a lesion affecting a cytosolic, tetrahydrofolate diglutamate-formingactivity. Cytosolic protein had ability to catalyze a diglutamate hexaglutamate reaction. Mitochondria of the mutant catalyzedthe formation of methylenetetrahydrofolate triglutamate fromthe corresponding methylene monoglutamate. It is suggested thatthe limited folylpolyglutamate synthesis of met(S2706) may involvecytosolic and mitochondrial folylpolyglutamate synthetase activities. Conidial germination in liquid shake culture of Neurospora crassawas affected by the number of conidia in the medium (conidialdensity effect) with the optimum density being around 2?106conidia/ml. The conidial density effect at 2?104 and 2?105 conidia/mlwas eliminated by the addition of auxin, IAA or 2,4-D with theoptimum concentrations of IAA and 2,4-D being around 10–6M. IAA and 2,4-D had no effect on the effect at 2?107 conidia/ml.An active substance(s) for conidial germination which was relatedto the conidial density effect was detected in the cell-freefiltrate of the germination medium, and was found to be non-dialyzableand thermolabile. At the early stage of germination, the concentrationof active substance(s) in the medium increased in proportionto the conidial density and reached a supraoptimum amount forgermination at 2?107 conidia/ml. IAA (10–6M) enhancedthe concentration. Endogenous auxin concentrations in filtratesof the germination media containing 2?105, 2?106 and 2?107 conidia/mlwere 5.8?10–12, 4.6?10–11 and 1.7?10–10M IAAequivalent, respectively. A colorless mutant of Chlorella vulgaris (Mutant #125) starvedin darkness, showed suppressed rates of respiration and darkCO2 fixation, which were significantly recovered by illuminationwith blue light. The main CO2 fixation product under blue lightwas aspartate. Such enhancements did not take place in cellsactively growing in the glucose medium. Both enhancing effectsof blue light (456 nm) were saturated at light intensities aslow as 400–800 erg.cm-2.sec-1. The action spectra forthese enhancing effects were similar to each other; both showedpeaks at 460 nm and 380 nm, which correspond to the absorptionmaxima of flavin. All these findings indicate that the samemechanism underlies the observed effects of blue light on CO2fixation and respiration. Period lengths at different temperatures and phase responsecurves at a high temperature (35°C) of circadian conidiationrhythms were examined in 13 temperature-sensitive (un) strainsof Neurospora crassa. Two strains, un-16 and un-18, had longerperiod lengths than the wild-type strain even at permissivetemperatures. Period lengths of six strains, un-4, un-11, un-16,un-18, un-19 and un-22, changed differently from that of thewild-type strain at restrictive temperatures. However, the shapeof phase response curves for high temperature (35°C) for3 h was almost the same for all un strains and the wild-typestrain. We isolated 97 temperature-sensitive mutants with periodlengths from 19.2 to 24.8 h and determined the dependence ontemperature of the period length of the conidiation rhythm foreach mutant. Two mutant strains (denoted as DCMUr-I and DCMUr-II) of theblue-green alga Aphanocapsa 6714, capable of growing in thepresence of 10–5 M DCMU (an inhibitor of electron transporton the acceptor side of photosystem II), were isolated. The DCMU sensitivity of growth rates and of two photosystemII activities, O2 emission and fluorescence, was investigatedin the mutant cells and compared with that of the wild type.The DCMU sensitivity of isolated thylakoids was also studied.The sensitivity of the mutant cells to other DCMU-type inhibitors(o-phenanthroline and atrazine) was tested. The results suggest that strain DCMUr-I resistance could bedue to the acquisition of a cellular permeability barrier toDCMU, expressed only after an adaptation phase in the presenceof the inhibitor. Blue light was specifically required for the induction of carbonicanhydrase (CA) activity in Chlamydomonas reinhardtii. The enhancingeffect of blue light (460 nm) was saturated at energy fluencerate as low as 0.6-0.8 W/m2. The wavelength dependency curvehad a peak at 460 nm with no effect at wavelengths above 510nm, thus showing the strong similarities to other blue lightresponses in microalgae. CA induction was strongly inhibitedby UV irradiation at 280 nm. The light-harvesting system of photosynthesis was studied infour strains of Porphyra yezoensis differing in their phycoerythrin(PE) content; the red strain, richer in PE than the wild strain,and the green and the yellow strains, poorer in PE. Specialattention was given to possible alteration of pigment systemin response to PE content, especially in the green and the yellowstrains. The relative quantum yields of Chi a fluorescence at–196°C and O2 evolution were compared. Four strains commonly showed a low yield in pigment system II(PS II) fluorescence on Chl a excitation. The yield was as lowas in those algae in which PS II has only a small portion ofChl a as the light harvester. Measurement of O2 evolution gavethe same results. Results indicate that the functional compositionof Chl a system remains unaltered in four strains with differentPE content. PS II in the green and the yellow strains reflectsa reduction in the size of light-harvesting components, suggestingthat pigmentation in these strains is fixed genetically as asun-type. 4 On leave from University of Washington, Seatle, Washington,U.S.A.
