Thirty Chlorella and 30 Scenedesmus strains grown in nitrogen-stressed conditions (70 mg L?1 N) were analyzed for biomass accumulation, lipid productivity, protein, and fatty acid (FA) composition. Scenedesmus strains produced more biomass (4.02?±?0.73 g L?1) after 14 days in culture compared to Chlorella strains (2.57?±?0.12 g L?1). Protein content decreased and lipid content increased from days 8 to 14 with an increase in triacylglycerol (TAG) accumulation in most strains. By day 14, Scenedesmus strains generally had higher lipid productivity (53.5?±?3.7 mg lipid L?1 day?1) than Chlorella strains (35.1?±?2.8 mg lipid L?1 day?1) with the lipids consisting mainly of C16–18 TAGs. Scenedesmus strains generally had a more suitable FA profile with higher amounts of saturated fatty acids and monounsaturated fatty acids (MUFAs) and a smaller polyunsaturated fatty acid (PUFA) component. Chlorella strains had a larger PUFA component and smaller MUFA component. The general trend in the FA composition of Chlorella strains was oleic > palmitic > α-linolenic = linoleic > eicosenoic > heptadecenoic > stearic acid. For Scenedesmus strains, the general trend was oleic > palmitic > linoleic > α-linolenic > stearic > eicosenoic > palmitoleic > heptadecenoic acid. The most promising strains with the highest lipid productivity and most suitable FA profiles were Scenedesmus sp. MACC 401, Scenedesmus soli MACC 721, and Scenedesmus ecornis MACC 714. Although Chlorella sp. MACC 519 had lower lipid productivity, the FA profile was good with a lower PUFA component compared to the other Chlorella strains analyzed and a low linolenic acid concentration.  相似文献

There has been considerable interest on cultivation of green microalgae (Chlorophyta) as a source of lipid that can alternatively be converted to biodiesel. The ideal microalga characteristics are that it must grow well even under high cell density and under varying outdoor environmental conditions and be able to have a high biomass productivity and contain a high oil content (~25–30 %). The main advantage of Chlorophyta is that their fatty acid profile is suitable for biodiesel conversion. Tetraselmis suecica CS-187 and Chlorella sp. were grown semi-continuously in bag photobioreactors (120 L, W?×?L?=?40?×?380 cm) over a period of 11 months in Melbourne, Victoria, Australia. Monthly biomass productivity of T. suecica CS-187 and Chlorella sp. was strongly correlated to available solar irradiance. The total dry weight productivity of T. suecica and Chlorella sp. was 110 and 140 mg L?1 d?1, respectively, with minimum 25 % lipid content for both strains. Both strains were able to tolerate a wide range of shear produced by mixing. Operating cultures at lower cell density resulted in increasing specific growth rates of T. suecica and Chlorella sp. but did not affect their overall biomass productivity. On the other hand, self shading sets the upper limit of operational maximum cell density. Several attempts in cultivating Dunaliella tertiolecta CS-175 under the same climatic conditions were unsuccessful.  相似文献

Northern regions are generally viewed as unsuitable for microalgal biofuel production due to unfavorable climate and solar insolation levels. However, these conditions can potentially be mitigated by coupling microalgal cultivation to industrial processes such as wastewater treatment. In this study, we have examined the biomass and lipid productivity characteristics of 14 microalgae isolates (Chlorophyta) from the Canadian province of Saskatchewan. Under both photoautotrophic and mixotrophic cultivation, a distinct linear trend was observed between biomass and lipid productivities in the 14 SK isolates. The most productive strain under cultivation in TAP media was Scenedesmus sp.-AMDD which displayed rates of biomass and fatty acid productivities of 80 and 30.7?mg?L?1?day?1, respectively. The most productive strain in B3NV media was Chlamydomonas debaryana-AMLs1b which displayed rates of biomass and fatty acid productivities of 51.7 and 5.9?mg?L?1?day?1, respectively. In 11 of the isolates tested, secondary municipal wastewater (MCWW) supported rates of biomass productivity between 21 and 33?mg?L?1?day?1 with Scenedesmus sp.-AMDD being the most productive. Three strains, Chlamydomonas debaryana-AMB1, Chlorella sorokiniana-RBD8 and Micractinium sp.-RB1b, showed large increases in biomass productivity when cultivated mixotrophically in MCWW supplemented with glycerol. High relative oleic acid content was detected in 10 of the 14 isolates when grown mixotrophically in media supplemented with acetate. There was no detectable effect on the fatty acid profiles in cells cultivated mixotrophically in glycerol-supplemented MCWW. These data indicate that biomass and lipid productivities are boosted by mixotrophic cultivation. Exploiting this response in municipal wastewater is a promising strategy for the production of environmentally sustainable biofuels.  相似文献

Prolific algal growth in sewage ponds with high organic loads in the tropical regions can provide cost-effective and efficient wastewater treatment and biofuel production. This work examines the ability of Euglena sp. growing in wastewater ponds for biofuel production and treatment of wastewater. The algae were isolated from the sewage treatment plants and were tested for their nutrient removal capability. Compared to other algae, Euglena sp. showed faster growth rates with high biomass density at elevated concentrations of ammonium nitrogen (NH4-N) and organic carbon (C). Profuse growth of these species was observed in untreated wastewaters with a mean specific growth rate (μ) of 0.28 day?1 and biomass productivities of 132 mg ?L?1? day?1. The algae cultured within a short period of 8 days resulted in the 98 % removal of NH4-N, 93 % of total nitrogen 85 % of ortho-phosphate, 66 % of total phosphate and 92 % total organic carbon. Euglenoids achieved a maximum lipid content of 24.6 % (w/w) with a biomass density of 1.24 g ?L?1 (dry wt.). Fourier transform infrared spectra showed clear transitions in biochemical compositions with increased lipid/protein ratio at the end of the culture. Gas chromatography and mass spectrometry indicated the presence of high contents of palmitic, linolenic and linoleic acids (46, 23 and 22 %, respectively), adding to the biodiesel quality. Good lipid content (comprised quality fatty acids), efficient nutrient uptake and profuse biomass productivity make the Euglena sp. as a viable source for biofuel production in wastewaters.  相似文献

The accumulation of atmospheric CO2, primarily due to combustion of fossil fuels, has been implicated in potential global climate change. The high rate of CO2 bioremediation by microalgae has emerged as a favourable method for reducing coal-fired power plant emissions. However, coal-fired power station flue gas contains other chemicals such as SOx which can inhibit microalgal growth. In the current study, the effect of untreated flue gas as a source of inorganic carbon on the growth of Tetraselmis in a 1000 L industrial-scale split-cylinder internal-loop airlift photobioreactor was examined. The culture medium was recycled after each harvest. Tetraselmis suecica grew very well in this airlift photobioreactor during the 7-month experiment using recycled medium from an electroflocculation harvesting unit. Increased medium SO4 2? concentration as high as 870 mg SO4 2??L?1 due to flue gas addition and media recycling had no negative effect on the overall growth and productivity of this alga. The potential organic biomass productivity and carbon sequestration using an industrial-scale airlift PBR at International Power Hazelwood, Gippsland, Victoria, Australia, are 178.9?±?30 mg L?1 day?1 and 89.15?±?20 mg?‘C’?L?1 day?1, respectively. This study clearly indicates the potential of growing Tetraselmis on untreated flue gas and using recycled medium for the purpose of biofuel and CO2 bioremediation.  相似文献

The fatty acid composition, the effect of different concentrations of nitrogen (16.5-344 mg ?L?1), phosphorus (9–45 mg? L?1), iron (9–45 mg? L?1) and salinity levels (0–20 psu) on lipid production in the green microalga Scenedesmus dimorphus KMITL, a new strain isolated from a tropical country, Thailand, were studied. The alga was isolated from a freshwater fish pond, and cultured in Chlorella medium by varying one parameter at a time. The main fatty acid composition of this strain was C16–C18 (97.52 %) fatty acids. A high lipid content was observed in conditions of 16.5 mg? L?1-N, or 22 mg ?L?1-P, or 45 mg ?L?1-Fe, or 5 psu salinity, which accumulated lipids to 20.3?±?0.4, 19.4?±?0.2, 24.7?±?0.5, and 14.3?±?0.2 % of algal biomass, respectively. Increasing lipid content and lipid productivity was noted when the alga was cultured under high iron concentration and high salinity, as well as under reduced phosphorus conditions, whereas nitrogen limitation only resulted in an increased lipid content.  相似文献

Microalgae are a promising feedstock for biofuel production. Lipid content in microalgae could be enhanced under nutrient depletion. This work investigated the effect of the nutrient on lipid accumulation in Ankistrodesmus sp. culture. Batch cultures were carried out using fresh BG11 medium, and after the harvest, the medium was reused for the next culture; this method was repeated two times. The maximum lipid productivity of 29.75 mg L?1 day?1 was obtained from the culture with the second reuse medium. In continuous cultures, Ankistrodesmus sp. was cultured in both fresh and modified BG11 mediums. The modified BG11 medium was adjusted to resemble the content of the first reuse medium. As a comparison between batch and continuous cultures, it was proven that the productivity in the continuous culture was better than in the batch, where the achievable maximum biomass and lipid were 188.30 and 38.32 mg L?1 day?1. The maximum lipid content of 34.22% was obtained from the continuous culture at a dilution rate of 0.08 day?1, whereas the maximum saturated and unsaturated fatty acid productivities of 79.96 and 104.54 mg L?1 day?1 were obtained at a dilution rate of 0.16 day?1.  相似文献

Cultivation temperature is one of the major factors affecting the growth and lipid accumulation of microalgae. In this study, the effects of temperature on the growth, lipid content, fatty acid composition and biodiesel properties of the marine microalgae Chaetoceros sp. FIKU035, Tetraselmis suecica FIKU032 and Nannochloropsis sp. FIKU036 were investigated. These species were cultured at different temperatures (25, 30, 35 and 40 °C). The results showed that the specific growth rate, biomass and lipid content of all microalgae decreased with increasing temperature. With regards to fatty acids, the presence of saturated fatty acids (SFAs) in T. suecica FIKU032 and Nannochloropsis sp. FIKU036 decreased with increasing temperature, in contrast with polyunsaturated fatty acids (PUFAs). Moreover, Chaetoceros sp. FIKU035 was the only species that could grow at 40 °C. The highest lipid productivity was observed in Chaetoceros sp. FIKU035 when cultivated at 25 °C (66.73 ± 1.34 mg L?1 day?1) and 30 °C (61.35 ± 2.89 mg L?1 day?1). Moreover, the biodiesel properties (cetane number, cold filter plugging point, kinematic viscosity and density) of the lipids obtained from this species were in accordance with biodiesel standards. This study indicated that Chaetoceros sp. FIKU035 can be considered as a suitable species for biodiesel production in outdoor cultivation.  相似文献

Microalgae cultivation systems can be divided broadly into open ponds and closed photobioreactors. This study investigated the growth and biomass productivity of the halophilic green alga Tetraselmis sp. MUR-233, grown outdoors in paddle wheel-driven open raceway ponds and in a tubular closed photobioreactor (Biocoil) at a salinity of 7 % NaCl (w/v) between mid-March and June 2010 (austral autumn/winter). Volumetric productivity in the Biocoil averaged 67 mg ash-free dry weight (AFDW) L?1 day?1 when the culture was grown without CO2 addition. This productivity was 86 % greater, although less stable, than that achieved in the open raceway pond (36 mg L?1 day?1) grown at the same time in the autumn period. The Tetraselmis culture in the open raceway pond could be maintained in semi-continuous culture for the whole experimental period of 3 months without an additional CO2 supply, whereas in the Biocoil, under the same conditions, reliable semi-continuous culture was only achievable for a period of 38 days. However, stable semi-continuous culture was achieved in the Biocoil by the addition of CO2 at a controlled pH of ~7.5. With CO2 addition, the volumetric biomass productivity in the Biocoil was 85 mg AFDW L?1 day?1 which was 5.5 times higher than the productivity achieved in the open raceway pond (15 mg AFDW L?1 day?1) with CO2 addition and 8 times higher compared to the productivity in the open raceway pond without CO2 addition (11 mg AFDW L?1 day?1), when cultures were grown in winter. The illuminated area productivities highlight an alternative story and showed that the open raceway pond had a three times higher productivity (3,000 mg AFDW m?2 day?1) compared to the Biocoil (850 mg AFDW m?2 day?1). Although significant differences were found between treatments and cultivation systems, the overall average lipid content for Tetraselmis sp. MUR-233 was 50 % in exponential phase during semi-continuous cultivation.  相似文献

The need to develop biomass-based domestic production of high-energy liquid fuels (biodiesel) for transportation can potentially be addressed by exploring microalgae with high lipid content. Selecting the strains with adequate oil yield and quality is of fundamental importance for a cost-efficient biofuel feedstock production based on microalgae. This work evaluated 29 strains of Chlorella isolated from Malaysia as feedstock for biodiesel based on volumetric lipid productivity and fatty acid profiles. Phylogenetic studies based on 18S rRNA gene revealed that majority of the strains belong to true Chlorella followed by Parachlorella. The strains were similarly separated into two groups based on fatty acid composition. Of the 18 true Chlorella strains, Chlorella UMACC187 had the highest palmitic acid (C16:0) content (71.3?±?4.2 % total fatty acids, TFA) followed by UMACC84 (70.1?±?0.7 %TFA), UMACC283 (63.8?±?0.7 %TFA), and UMACC001 (60.3?±?4.0 %TFA). Lipid productivity of the strains at exponential phase ranged from 34.53 to 230.38 mg L?1 day?1, with Chlorella UMACC050 attaining the highest lipid productivity. This study demonstrated that Chlorella UMACC050 is a promising candidate for biodiesel feedstock production.  相似文献

Evaluation of antioxidant capacities of green microalgae   总被引:2,自引:0,他引:2  
Three strains of green microalgae, Chlorococcum sp.C53, Chlorella sp. E53, and Chlorella sp.ED53 were studied for their antioxidant activities. Crude extracts of these microalgae in hot water and in ethanol were examined for their total phenolic contents and for their antioxidant capacities. In order to determine their phenolic contents, the Folin–Ciocalteu method was used. As for the determination of their antioxidant capacities, four different assays were used: (1) total antioxidant capacity determination; (2) DPPH radical scavenging assay; (3) ferrous ion chelating ability assay; and (4) inhibition of lipid peroxidation (using thiobarbituric acid reactive substance). For all the strains we have studied, their ethanolic extract showed more antioxidant activities than their hot water extract. Categorically, the ethanolic extract of Chlorella sp.E53 exhibited both the highest total phenolic content of 35.5?±?0.14 mg gallic acid equivalent (GAE) g?1 dry weight and the highest DPPH radical scavenging of 68.18?±?0.38 % at 1.4 mg mL?1 (IC50 0.81 mg mL?1), whereas Chlorella sp.ED53 showed both the highest ferrous ion chelation activity of 42.78?±?1.48 % at 1 mg mL?1 (IC50 1.23 mg mL?1) and the highest inhibition of lipid peroxidation of 87.96?±?0.59 % at 4 mg mL?1. This high level of inhibition is comparable to 94.42?±?1.39 % of butylated hydroxytoluene, a commercial synthetic antioxidant, at the same concentration.  相似文献

In the present study, process engineering strategy was applied to achieve lipid-rich biomass with high density of Chlorella sp. FC2 IITG under photoautotrophic condition. The strategy involved medium optimization, intermittent feeding of limiting nutrients, dynamic change in light intensity, and decoupling growth and lipid induction phases. Medium optimization was performed using combinations of artificial neural network or response surface methodology with genetic algorithm (ANN-GA and RSM-GA). Further, a fed-batch operation was employed to achieve high cell density with intermittent feeding of nitrate and phosphate along with stepwise increase in light intensity. Finally, mutually exclusive biomass and lipid production phases were decoupled into two-stage cultivation process: biomass generation in first stage under nutrient sufficient condition followed by lipid enrichment through nitrogen starvation. The key findings were as follows: (i) ANN-GA resulted in an increase in biomass titer of 157 % (0.95 g L?1) in shake flask and 42.8 % (1.0 g L?1) in bioreactor against unoptimized medium at light intensity of 20 μE m?2 s?1; (ii) further optimization of light intensity in bioreactor gave significantly improved biomass titer of 5.6 g L?1 at light intensity of 250 μE m?2 s?1; (iii) high cell density of 13.5 g L?1 with biomass productivity of 675 mg L?1 day?1 was achieved with dynamic increase in light intensity and intermittent feeding of limiting nutrients; (iv) finally, two-phase cultivation resulted in biomass titer of 17.7 g L?1 and total lipid productivity of 313 mg L?1 day?1 which was highest among Chlorella sp. under photoautotrophic condition.  相似文献

Phycobiliproteins, light-harvesting pigments found in cyanobacteria and in some eukaryotic algae, have numerous commercial applications in food, cosmetic, and pharmaceutical industries. Colorant production from cyanobacteria offers advantages over their production from higher plants, as cyanobacteria have fast growth rate and high photosynthetic efficiency and require less space. In this study, three cyanobacteria strains were studied for phycobiliprotein production and the influence of sodium nitrate, potassium nitrate and ammonium chloride on the growth and phycobiliprotein composition of the strains were evaluated. In the batch culture period of 12 days, Phormidium sp. and Pseudoscillatoria sp. were able to utilize all tested nitrogen sources; however, ammonium chloride was the best nitrogen source for both strains to achieve maximum growth rate μ?=?0.284?±?0.03 and μ?=?0.274?±?0.13 day?1, chlorophyll a 16.2?±? 0.5 and 12.2?±? 0.2 mg L?1, and phycobiliprotein contents 19.38?±?0.09 and 19.99?±?0.14% of dry weight, whereas, for Arthrospira platensis, the highest growth rate of μ?=?0.304?±?0.0 day?1, chlorophyll a 19.1?±?0.5 mg L?1, and phycobiliprotein content of 22.27?±?0.21% of dry weight were achieved with sodium nitrate. The phycocyanin from the lyophilized cyanobacterial biomass was extracted using calcium chloride and food grade purity (A620/A280 ratio >?0.7) was achieved. Furthermore, phycocyanin was purified using two-step chromatographic method and the analytical grade purity (A620/A280 ratio >?4) was attained. SDS-PAGE demonstrated the purity and presence of two bands corresponding to α- and β-subunits of the C-phycocyanin. The results showed that Phormidium sp. and Pseudoscillatoria sp. could be good candidates for phycocyanin production.  相似文献

The membraneless bioelectrochemical reactor (Ml-BER) is useful for dark hydrogen fermentation. The effect of the electrochemical reaction on microorganisms in the Ml-BER was investigated using glucose as the substrate and compared with organisms in a membraneless non-bioelectrochemical reactor (Ml-NBER) and bioelectrochemical reactor (BER) with a proton exchange membrane. The potentials on the working electrode of the Ml-BER and BER with membrane were regulated to ?0.9 V (versus Ag/AgCl) to avoid water electrolysis with a carbon electrode. The Ml-BER showed suppressed methane production (19.8?±?9.1 mg-C·L?1·day?1) and increased hydrogen production (12.6?±?3.1 mg-H·L?1·day?1) at pHout 6.2?±?0.1, and the major intermediate was butyrate (24.9?±?2.4 mM), suggesting efficient hydrogen fermentation. In contrast, the Ml-NBER showed high methane production (239.3?±?17.9 mg-C·L?1·day?1) and low hydrogen production (0.2?±?0.0 mg-H·L?1·day?1) at pHout 6.3?±?0.1. In the cathodic chamber of the BER with membrane, methane production was high (276.3?±?20.4 mg-C·L?1·day?1) (pHout, 7.2?±?0.1). In the anodic chamber of the BER with membrane (anode-BER), gas production was low because of high lactate production (43.6?±?1.7 mM) at pHout 5.0?±?0.1. Methanogenic archaea were not detected in the Ml-BER and anode-BER. However, Methanosarcina sp. and Methanobacterium sp. were found in Ml-NBER. Prokaryotic copy numbers in the Ml-BER and Ml-NBER were similar, as were the bacterial community structures. Thus, the electrochemical reaction in the Ml-BER affected hydrogenotrophic and acetoclastic methanogens, but not the bacterial community.  相似文献

the research aim of this study was to characterize an isolated native strain of Chlorella sp. ACUF_802, well adapted to a high nitrate concentration environment and to investigate its potential to nitrate and phosphate removal from industrial wastewaters with the minimal addition of chemical reagents and energy. The isolated strain was identified and evaluated for its capability to support biomass growth and nutrient removal from synthetic wastewater in batch tests using different concentrations of carbon and nitrogen, different carbon sources and N:P ratios. The strain was isolated via the plating method from the settler of a pilot scale moving bed biofilm reactor performing a nitrification process. The strain was identified using molecular analysis with rDNA primers. Using sodium bicarbonate as carbon source, the batch productivity (71.43 mg L?1 day?1) of the strain Chlorella sp. ACUF_802 was calculated with a logistic model and compared to the values reported in the literature. Assays on the effect of the N:P ratio indicated that the productivity was increased 36% when the N:P ratio was close to 1 (111.96 mg L?1 day?1), but for a complete phosphorus removal a 5:1 N:P ratio with nitrate concentrations ≤125 mg?L?1 is recommended. The isolated microalgae strain Chlorella sp. ACUF_802 showed versatility to grow in the synthetic industrial wastewaters tested and can be considered as an appropriate organism for nitrogen removal from industrial wastewaters in the presence of an organic or inorganic carbon source.  相似文献

The biomass productivity of Scenedesmus obliquus was investigated outdoors during all seasons in solar tracked flat panel photobioreactors (PBR) to evaluate key parameters for process optimization. CO2 was supplied by flue gas from an attached combined block heat and power plant. Waste heat from the power plant was used to heat the culture during winter. The parameters pH, CO2, and inorganic salt concentrations were automatically adjusted to nonlimiting levels. The optimum biomass concentration increased directly with the photosynthetic active radiation (PAR) from 3 to 5 g dry weight (DW)?L?1 for a low PAR of 10 mol photons m?2 day?1 and high PAR of 40–60 mol photons m?2 day?1, respectively. The annual average biomass yield (photosynthetic efficiency) was 0.4?±?0.5 g DW mol?1 photons. However, biomass yields of 1.5 g DW mol?1 photons close to the theoretical maximum were obtained at low PAR. The productivity (including the night biomass losses) ranged during all seasons from ?5 up to 30 g DW m?2 day?1 with a mean productivity of 9?±?7 g DW m?2 day?1. Low night temperatures of the culture medium and elevated day temperatures to the species-specific optimum increased the productivity. Thus, continuous regulation of the biomass concentration and the culture temperature with regard to the fluctuating weather conditions is essential for process optimization of outdoor microalgal production systems in temperate climates.  相似文献

The addition of bicarbonate (NaHCO3; 0, 1, or 2 g L?1) to microalgal cultures has been evaluated for two species (Tetraselmis suecica and Nannochloropsis salina) in respect of growth and biochemical composition. In batch cultures, addition of bicarbonate (1 g L?1) resulted in significantly (P?<?0.05) higher final mean cell abundances for both species. No differences in specific growth rates (SGRs) were recorded for T. suecica between treatments; however, increasing bicarbonate addition decreased SGR values in N. salina cultures. Bicarbonate addition (1 g L?1) significantly improved nitrate utilisation from the external media and photosynthetic efficiency (F v /F m ) in both species. For both T. suecica and N. salina, bicarbonate addition significantly increased the cellular concentrations of total pigments (3,432–3,587 and 19–37 fg cell?1, respectively) compared to cultures with no additional bicarbonate (1,727 and 11 fg cell?1, respectively). Moreover, final concentrations of total cellular fatty acids in T. suecica and N. salina cultures supplemented with 2 g L?1 bicarbonate (7.6?±?1.2 and 1.8?±?0.1 pg cell?1, respectively) were significantly higher than those cells supplemented with 0 or 1 g L?1 bicarbonate (3.2–3.5 and 0.9–1.0 pg cell?1, respectively). In nitrate-deplete cultures, bicarbonate addition caused species-specific differences in the rate of cellular lipid production, rates of change in fatty acid composition and final lipid levels. In summary, the addition of sodium bicarbonate is a viable strategy to increase cellular abundance and concentrations of pigments and lipids in some microalgae as well as the rate of lipid accumulation in nitrate-deplete cultures.  相似文献

HQ was tested in five different synthetic water, and it was found to achieve the maximum biomass (0.27 g L?1) and lipid yield (41.3 mg L?1) in the synthetic secondary effluent. Next, the effects of the stationary phase elongation and initial nitrogen (N) and phosphorus (P) concentrations were investigated. The results show that the algal characteristics were affected apparently under different N concentrations but not P, which were verified by Logistic and Monod models. At the early stationary phase, the algal biomass, lipid and triacylglycerols (TAGs) yields, and P removal efficiency increased and reached up to 0.19 g L?1, 46.7 mg L?1, 14.3 mg L?1, and 94.3 %, respectively, but N removal efficiency decreased from 86.2 to 26.8 % under different N concentrations. And the largest TAGs yield was only 6.4 mg L?1 and N removal efficiency was above 71.1 % under different P concentrations. At the late stationary phase, the maximal biomass, lipid and TAGs yields, and P removal efficiencies primarily increased as the initial N and P concentrations increase and climbed up to 0.49 g L?1, 99.2 mg L?1, 54.0 mg L?1, and 100.0 %, respectively. It is concluded that stationary phase elongation is of great importance and the optimal initial N/P ratio should be controlled between 8/1 and 20/1 to serve Chlorella sp. HQ for better biodiesel production and secondary effluent purification.  相似文献

Semi-continuous algal cultivation was completed in outdoor flat-panel photobioreactors (panels) and open raceway ponds (raceways) from February 17 to May 7, 2015 for side-by-side comparison of areal productivities at the Arizona Center for Algae Technology and Innovation in Mesa, AZ, USA. Experiments used two strains of Scenedesmus acutus (strains LB 0414 and LB 0424) to assess productivity, areal density, nutrient removal, and harvest volume across cultivation systems and algal strains. Panels showed an average biomass productivity of 19.0?±?0.6 g m?2 day?1 compared to 6.62?±?2.3 g m?2 day?1 for raceways. Photosynthetic efficiency ranged between 1.32 and 2.24 % for panels and between 0.30 and 0.68 % for raceways. Panels showed an average nitrogen consumption rate of 38.4?±?8.6 mg N L?1 day?1. Cultivation in raceways showed a consumption rate of 3.8?±?2.5 and 7.1?±?4.2 mg N L?1 day?1 for February/March and April/May, respectively, due to increase in biomass productivity. Excess nutrients were required to prevent a decrease in productivity. Daily biomass harvest volumes between 18 and 36 % from panels did not affect culture productivity, but density decreased with increased harvest volume. High cultivation temperatures above 30 °C caused strain LB 0414 to lyse and crash. Strain LB 0424 did not show any difference in biomass productivity when peak temperatures reached 34, 38, or 42 °C, but showed decreased productivity when the peak temperature during cultivation was 30 °C. Using algal strains with different temperature tolerances can generate increased annual biomass productivity.  相似文献

The aim of the present study was to survey the growth and astaxanthin production of E17, an astaxanthin-rich mutant of Chlorella zofingiensis, through feeding the low-cost carbon source cane molasses. In heterotrophic batch cultivation, E17 fed with pretreated molasses achieved biomass (1.79 g L?1 day?1) and astaxanthin (1.99 mg L?1 day?1) productivities comparable to those with glucose, which were about 2- and 2.8-fold of those fed with untreated molasses, respectively. Molasses-induced astaxanthin accumulation may be attributed to the elicited expression of carotenogenic genes, in particular the genes specifically responsible for the ketolation and hydroxylation of β-carotene to form astaxanthin. A two-stage fed-batch strategy was employed to grow E17 and induce astaxathin accumulation, resulting in 45.6 g L?1 biomass and 56.1 mg L?1 astaxanthin, the highest volumetric astaxanthin yield ever reported for this alga. In addition, the astaxanthin production by E17 was tested with a semi-continuous culture method, where the directly diluted raw molasses (giving 5 g L?1 sugar) was used as the carbon source. Little growth inhibition of E17 was observed in the semi-continuous culture with a biomass productivity of 1.33 g L?1 day?1 and an astaxanthin productivity of 0.83 mg L?1 day?1. The mixotrophic semi-continuous cultures enhanced the biomass and astaxanthin productivities by 29.3 % and 42.2 %, respectively. This study highlights the potential of using the industrially cheap cane molasses towards large-scale cost-saving production of the high-value ketocarotenoid astaxanthin.  相似文献
