A human colon kininogenase (kallikrein) was isolated by gel filtration on Sephacryl S-200 and affinity chromatography on Trasylolbound Sepharose, yielding a material with a specific activity of 1.3 U/mg (substrate: AcPheArgOEt). The molecular weight of the enzyme as estimated by gel filtration is approximately 70 000. After reduction with mercaptoethanol two bands were obtained in dodecyl sulfate eletrophoresis with molecular weights of 27 000 and 70 000. The bimolecular velocity constant for the inhibition by diisopropyl fluorophosphate was determined as 4 l x mol-1 x min-1. The preparation was characterized by immunological and enzymatic methods. Using the radioimmumoassay for human urinary kallikrein cross-reactivity and parallel binding curves were obtained. Kinin liberation from human high Mr-kininogen was totally inhibited by antibodies directed against human urinary kallikrein. Trasylol and diisopropyl fluorophosphate, but not by antibodies directed against human trypsin and plasma kallikrein. The effect on dog blood pressure was comparable to that obtained with human urinary kallikrein. The amino acid composition of human large intestine kallikrein is very similar to that of human urinary kallikrein. 相似文献

The immunoblotting technique was used to analyse polypeptides of purified Epstein-Barr virus reacting with antibodies present in sera from clinically healthy individuals, from patients with infectious mononucleosis (IM) or AIDS, and from renal transplant recipients with molecular sizes in the range of 40–290 kDa were detected.The 47- and 160-kDa nucleocapsid polypeptides, as well as the 72-, 74-, 140-, 220- and 290-kDa membrane polypeptides were the major viral proteins detected in the sera. Sera from clinically healthy individuals contained antibodies directed against all EBV membrane and nucleocapsid antigens. Sera from renal transplant recipients, from patients with IM and from patients with AIDS failed to react with certain nucleocapsid and membrane antigens; in particular, sera from AIDS patients and renal transplant recipients did not react with the 220-kDa polypeptide, one of the major membrane antigens, while sera from subjects with IM and from healthy individuals did.A high proportion of sera from patients with IM (38% vs 5% of clinically healthy individuals and 0–5% of the AIDS patients and renal transplant recipients) reacted with a 42-kDa polypeptide, suggesting its possible role in acute EBV infection. 相似文献

Human urinary kallikrein was purified by gel filtration on Sephacryl S-200 and affinity chromatography on aprotinin-Sepharose, followed by ion exchange chromatography on DEAE-Sepharose. Thus an enzyme preparation with a specific activity (using AcPheArgOEt as substrate) of 1 100 U/mg protein was obtained. A specific bioligical activity of 2 300 KE/mg was measured in the dog blood pressure assay and of 0.742 HMW kininogen-U/mg, corresponding to the liberation of 787 micrograms bradykinin per mg enzyme per min from HMW-kininogen, in the rat uterus assay. In dodecyl sulfate electrophoresis two protein bands with apparent molecular weights of 41 000 and 34 000 were separated. The amino acid composition was determined and isoleucine was identified as the only aminoterminal amino acid. On isoelectric focusing six protein bands with isoelectric points of 3.75, 3.80, 3.90, 4.00, 4.05 and 4.25 were separated. The kinetic constants for the kallikrein-catalyzed hyrdolysis of AcPheArgOEt and D ValLeuArgNHNp were determined. The bimolecular velocity constant for the inhibition by diisopropyl fluorophosphate was determined as 9 +/- 2l x mol-1 x min-1. Immunological studies showed that a close relationship exists between the urinary enzyme and other human glandular kallikreins. Deoxycholate, lysolecithin and other amphiphiles activated human urinary kallikrein. 相似文献

This is the first report to demonstrate that chloro(N alpha-p-tosyllysyl)methane (TosLys-CH2Cl) inhibits mammalian glandular kallikrein activities. The inhibitory effect of TosLysCH2Cl on purified rat urinary kallikrein was carried out with three assay methods: 1) Tos-Arg-OMe hydrolysis activity measured by a radiochemical method; 2) kininogenase activity using purified bovine low molecular weight kininogen as substrate and the released kinins subsequently measured by radioimmunoassay; 3) bioassay using isolated rat uterus preparation. Purified rat urinary kallikrein was inhibited by TolLysCH2Cl in a dose and time-dependent manner with all three methods used. The inhibition of purified human urinary kallikrein esterase and kinin-releasing activities were also demonstrated. The results indicate that TosLysCH2Cl inactivates kallikrein activity and support the notion that reactive histidine residue(s) participates in the active center of Kallikrein for catalysis. 相似文献

Urinary kallikrein excretion is reduced in patients with hypertension of unknown etiology. In addition, the excretion of this renal, kinin-forming enzyme was found to be elevated in hypertensive patients with primary aldosteronism. Aldosterone regulates kallikrein excretion, as normal subjects show increased kallikrein excretion in response to a low sodium intake, high potassium intake, or the synthetic mineralocorticoid, fludrocortisone, whereas kallikrein excretion falls during treatment with spironolactone. The relationship between kallikrein excretion and aldosterone activity may directly reflect the intrarenal activity of the kallikrein-kinin system, as determined by studies of kallikrein levels from isolated renal cells or of plasma kinin levels in man in response to postural changes or saline loads. Some patients with essential hypertension do not show a normal increase in kallikrein excretion in response to low dietary sodium intake despite an apparently normal aldosterone response, suggesting that there may be a defect in the renal kallikrein-kinin system in these patients. Whether these findings are of pathogenetic significance in human hypertensive disease remains to be determined. 相似文献

The localization of kallikrein in human exocrine organs was studied with a direct immunofluorescence method. In the submandibular and parotid salivary glands, kallikrein was found apically in the striated duct cells whereas it was absent from the main excretory ducts or present only as a weak luminal rim. Kallikrein was not found in the acinar cells or in cells of the intercalated ducts. In the pancreas, kallikrein-specific fluorescence was seen in the granular portion of the acinar cells, whereas the islets of Langerhans and ductal cells were unstained. 相似文献

Human hexosaminidase B has been purified to virtually homogeneous state from placenta. An anti-serum has been prepared in rabbits against the purified preparation. An antiserum was prepared against the recently purified bullfrog (bf) growth hormone (GH); it was applied to sections of brain and pituitary of three urodele (Ambystoma, Pleurodeles and Cynops) and three anuran (Xenopus, Bufo vulgaris and B. japonicus) species. No immunostaining was obtained in the urodele pituitary, being consistent with the results of immunoblot analysis of the pituitary homogenate. In the three anuran species, strong immunoreactivity was observed in GH cells that were concentrated in the posterodorsal region of the pars distalis. No GH-like immunoreactivity was detectable in the brain of any of the species. A comparison using adjacent sections stained with anti-bf prolactin (PRL) confirmed the anteroventral localization of PRL cells. Colocalization of GH and PRL was not apparent. These data suggest that the molecular structure of amphibian GHs is considerably different between anurans and urodeles. The antiserum used in the present work shows a high species specificity, recognizing only anuran GHs. In contrast anti-bfPRLlabeled PRL cells in all the amphibian species studied in the present work, suggesting that PRLs possess common amino acid sequences recognized by the anti-bfPRL. 相似文献

Herpes simplex virus type 1 ribonucleotide reductase is associated with two polypeptides of apparent molecular weights 136,000 and 38,000. The two polypeptides form a tight complex and, therefore, are often coprecipitated by monoclonal antibodies. We report here that immunoglobulins G purified from polyclonal rabbit antisera (P9) raised against a nonapeptide corresponding to the carboxy terminus of the 38,000-dalton polypeptide specifically neutralize the herpes simplex virus ribonucleotide reductase activity. We suggest that the P9 immunoglobulin G neutralizes the reductase activity by impairing the association of the two subunits (H1 and H2) of the enzyme. 相似文献

A rabbit antiserum against a purified embryonal carcinoma (EC) cell surface glycoprotein interferes with cell-cell interaction in mouse preimplantation embryos. The 123 kD glycoprotein seems not to be an integral membrane component. The reactivity pattern of the antiserum was studied by immunofluorescence on cryostat sections of post-implantation embryos and of adult tissues. During embryonic development positive reactions were found on all epithelial cells, irrespective of their germ layer origin. Epithelial cells of adult tissues—tongue, uterus, gut, kidney, trachea and liver—react with the antibodies. The results are compared with cell-adhesive molecules previously described on EC cells and preimplantation embryos. 相似文献
