In this study, we highlight the effects of methyl jasmonate (MeJa) on cystocarp development in the red macroscopic alga Grateloupia imbricata. In G. imbricata, jasmonate release is related to the reproductive state, as fertile thalli (i.e., those that have cystocarps) released significant amounts of this volatile compound (1.27 ± 0.20 mM · mg fw?1 · h?1) compared with infertile thalli (0.95 ± 0.12 mM · mg fw?1 · h?1). Treating G. imbricata thalli with MeJa revealed a significant increase in cystocarp number (1.5 ± 0.27 cystocarps · mm?2), which was ~7.5‐fold greater than in untreated thalli (0.2 ± 0.07 cystocarps · mm?2). Maturation was completed within 48 h with MeJa treatment, a shortening of the typical >3‐week maturation period, and included the opening of cystocarps and the presence of dehiscent cavities. Release rates of jasmonates after exogenous MeJa treatment were also modified based on the cystocarp maturation level. All of these effects were reduced in the presence of phenidone, which blocks MeJa production, indicating that the MeJa action is genuine. The effects of MeJa during cystocarp maturation were not replicated by derivatives of reactive oxygen species from the same jasmonic acid biosynthetic pathway, as the activities of scavenger enzymes and lipid peroxidation were unchanged between infertile and fertile thalli. Therefore, a reactive oxygen species–based mechanism is not involved during cystocarp development. We conclude that MeJa has an independent function as a growth regulator during G. imbricata reproduction.  相似文献

Dissolved inorganic phosphorus (DIP ) is an essential macronutrient for maintaining metabolism and growth in autotrophs. Little is known about DIP uptake kinetics and internal P‐storage capacity in seaweeds, such as Ulva lactuca (Chlorophyta). Ulva lactuca is a promising candidate for biofiltration purposes and mass commercial cultivation. We exposed U. lactuca to a wide range of DIP concentrations (1–50 μmol · L?1) and a nonlimiting concentration of dissolved inorganic nitrogen (DIN ; 5,000 μmol · L?1) under fully controlled laboratory conditions in a “pulse‐and‐chase” assay over 10 d. Uptake kinetics were standardized per surface area of U. lactuca fronds. Two phases of responses to DIP ‐pulses were measured: (i) a surge uptake (VS ) of 0.67 ± 0.10 μmol · cm?2 · d?1 and (ii) a steady state uptake (VM ) of 0.07 ± 0.03 μmol · cm?2 · d?1. Mean internal storage capacity (ISCP ) of 0.73 ± 0.13 μmol · cm?2 was calculated for DIP . DIP uptake did not affect DIN uptake. Parameters of DIN uptake were also calculated: VS  = 12.54 ± 1.90 μmol · cm?2 · d?1, VM  = 2.26 ± 0.86 μmol · cm?2 · d?1, and ISCN  = 22.90 ± 6.99 μmol · cm?2. Combining ISC and VM values of P and N, nutrient storage capacity of U. lactuca was estimated to be sufficient for ~10 d. Both P and N storage capacities were filled within 2 d when exposed to saturating nutrient concentrations, and uptake rates declined thereafter at 90% for DIP and at 80% for DIN . Our results contribute to understanding the ecological aspects of nutrient uptake kinetics in U. lactuca and quantitatively evaluating its potential for bioremediation and/or biomass production for food, feed, and energy.  相似文献

This study examined the effects of anesthesia on the hematological and biochemical parameters as well as the reproductive performance of wild female Persian sturgeon, Acipenser persicus, during controlled spawning. Fourteen mature females were divided into two groups: ‘anesthetized’ and ‘non‐anesthetized’. All activities including transportation, catheterizing and handling were performed with both groups: (i) under anesthesia (150 ppm clove oil), and (ii) without anesthesia. After 10 days storage and handling, blood samples were taken from all fish after anesthesia. No significant differences were found in the reproductive performance of either group. However, differences were found in the hematological parameters, with values being significantly higher in the non‐anesthetized group, including neutrophils (34.36 ± 6.33% vs 23.63 ± 5.22%), monocytes (2.84 ± 1.70% vs 1.27 ± 0.64%), mean corpuscular volume (321.3 ± 39.40 pg vs 228.0 ± 24.46 pg) and mean corpuscular hemoglobin (106.9 ± 15.70 fl vs 76.50 ± 7.50 fl). Significantly lower values were found in the non‐anesthetized group for lymphocytes (60.68 ± 7.25% vs 73.54 ± 4.80%), Hb (4.62 ± 0.74 mg dl?1 vs 6.28 ± 1.21 mg dl?1), Hct (13.86 ± 1.76% vs 18.84 ± 3.85%), red blood cell (0.43 ± 0.05 cell mm?3 vs 0.85 ± 0.13 × 106 cell mm?3) and white blood cell (22 403 ± 2240 cell mm?3 vs 35 318 ± 3084 cell mm?3). The non‐anesthetized fish had significantly higher cortisol levels compared to the anesthetized group (62.33 ± 8.85 ng ml?1 vs 46.12 ± 8.07 ng ml?1). There was no difference in plasma glucose levels between groups. It is concluded that the use of clove oil as an anesthetic is suitable for handling of wild female Persian sturgeon in controlled propagation programmes. However, further research is needed to determine a standardized protocol for the safe application of anesthesia for use in sturgeons in general.  相似文献

  • Oxalic acid is widely distributed in biological systems and known to play functional roles in plants. The gene AAE3 was recently identified to encode an oxalyl‐CoA synthetase (OCS) in Arabidopsis that catalyses the conversion of oxalate and CoA into oxalyl‐CoA. It will be particularly important to characterise the homologous gene in rice since rice is not only a monocotyledonous model plant, but also a staple food crop.
  • Various enzymatic and biological methods have been used to characterise the homologous gene.
  • We first defined that AAE3 in the rice genome (OsAAE3) also encodes an OCS enzyme. Its Km for oxalate is 1.73 ± 0.12 mm , and Vm is 6824.9 ± 410.29 U·min?1·mg protein?1. Chemical modification and site‐directed mutagenesis analyses identified thiols as the active site residues for rice OCS catalysis, suggesting that the enzyme might be regulated by redox state. Subcellular localisation assay showed that the enzyme is located in the cytosol and predominantly distributed in leaf epidermal cells. As expected, oxalate levels increased when OCS was suppressed in RNAi transgenic plants. More interestingly, OCS‐suppressed plants were more susceptible to bacterial blight but more resistant to Al toxicity.
  • The results demonstrate that the OsAAE3‐encoded protein also acts as an OCS in rice, and may play different roles in coping with stresses. These molecular, enzymatic and functional data provide first‐hand information to further clarify the function and mechanism of OCS in rice plants.

The incipient levels of lipid hydroperoxides (LHPOs) were determined in selected green, brown, and red macroalgae by the FOX assay using hydroperoxy HPLC mix. The LHPOs contents varied between the investigated species and showed relatively low values in this study. Among the greens, it varied from 12 ± 6.2 μg · g?1 (Codium sursum) to 31.5 ± 2.8 μg · g?1 (Ulva lactuca), whereas in reds, from 5.7 ± 1.6 μg · g?1 (Gracilaria corticata) to 46.2 ± 6 μg · g?1 (Sarconema filiforme), and in browns, from 4.6 ± 4.4 μg · g?1 (Dictyota bartayresiana) to 79 ± 5.0 μg · g?1 (Sargassum tenerrimum), on fresh weight basis. These hydroperoxides represented a minor fraction of total lipids and ranged from 0.04% (S. swartzii) to 1.1% (Stenerrimum) despite being a rich source of highly unsaturated fatty acids. The susceptibility of peroxidation was assessed by specific lipid peroxidazibility (SLP) values for macroalgal tissues. The LHPO values were found to be independent of both the PUFAs contents and their degree of unsaturation (DBI), as evident from the PCA analysis. SLP values were positively correlated with the LHPOs and negatively with DBI. The FOX assay gave ≥20‐fold higher values for LHPOs as compared to the TBARS method for all the samples investigated in this study. Furthermore, Ulactuca cultured in artificial seawater (ASW) enriched with nutrients (N, P, and NP) showed a sharp decline in LHPOs contents relative to those cultured in ASW alone ≤ 0.05. It is inferred from this study that the FOX assay is an efficient, rapid, sensitive, and inexpensive technique for detecting the incipient lipid peroxidation in macroalgal tissues.  相似文献

Isoflavones genistein and daidzein are nonsteroidal phytoestrogens occurring mainly in soybean foods. These phytoestrogens possess estrogenic properties and show a variety of health benefits as anti‐inflammatory agents. However, the mechanism of their action has not been identified in detail. The aim of this study is to characterize the antioxidant powers of genistein, daidzein and daidzein metabolite–equol through their activities to scavenge superoxide anion radical (O?2?), hydroxyl radical (HO?), 2,2–diphenyl–1‐picrylhydrazyl radical (DPPH?) and hydrogen peroxide (H2O2) using chemiluminescence and spectrophotometry techniques. Potassium superoxide in dimethyl sulphoxide (DMSO) and 18‐crown‐6 ether were used as a source of O?2?. Hydroxyl radicals were produced using the Fenton reaction. In free radical assays, genistein had the IC50 values (an amount of antioxidant concentration required to decrease the initial radical concentration by 50%) 0.391 ± 0.012 mM for O?2?, 0.621 ± 0.028 mM for HO? and 1.89 ± 0.16 mM for DPPH?. The IC50 values for daidzein for these free radicals were 1.924 ± 0.011 mM, 0.702 ± 0.012 mM and 2.81 ± 0.03 mM, respectively. Equol was the most active the free radical scavenger with IC50 = 0.451 ± 0.018 mM for HO? and IC50 = 1.36 ± 0.11 mM for DPPH?. All tested compounds exerted a significant effect on the H2O2: IC50 = 18.1 ± 1.1 μM for genistein, IC50 = 2.1 ± 0.5 μM for daidzein, and IC50 = 1.06 ± 0.2 μM for equol. 利用徒手切片,在光学显微镜下对芋(Colocasia esculenta(L.)Schott)营养器官中晶体的类型和分布进行了观察和研究,并用化学方法对晶体的化学成分进行了鉴定。结果表明,芋营养器官中的晶体为草酸钙结晶体,形态上可以分为针晶和簇晶两大类。含针晶束的异细胞有3种类型:含发射型草酸钙针晶束异细胞(存在于叶片、叶柄、块茎中),含大型草酸钙针晶束异细胞(存在于叶片、叶柄、块茎、块茎皮中),含大量草酸钙针晶的管状异细胞(仅存在于不定根中)。草酸钙针晶也有散乱分布于块茎和不定根中的。草酸钙簇晶在叶片、叶柄、块茎、块茎皮、不定根中均有分布,且叶片、叶柄、块茎皮中的簇晶比块茎和不定根中的尖锐。芋营养器官中的草酸钙晶体很可能是作为一种防御机制,防止动物的取食。  相似文献

The net carbon uptake rate and net production rate of mycosporine‐like amino acids (MAAs) were measured in phytoplankton from 2 different melt ponds (MPs; closed and open type pond) in the western Arctic Ocean using a 13C stable isotope tracer technique. The Research Vessel Araon visited ice‐covered western‐central basins situated at 82°N and 173°E in the summer of 2012, when Arctic sea ice declined to a record minimum. The average net carbon uptake rate of the phytoplankton in polycarbonate (PC) bottles in the closed MP was 3.24 mg C · m?3 · h?1 (SD = ±1.12 mg C · m?3 · h?1), while that in the open MP was 1.3 mg C · m?3 · h?1 (SD = ±0.05 mg C · m?3 · h?1). The net production rate of total MAAs in incubated PC bottles was highest (1.44 (SD = ±0.24) ng C · L?1 · h?1) in the open MP and lowest (0.05 (SD = ±0.003) ng C · L?1 · h?1) in the closed MP. The net production rate of shinorine and palythine in incubated PC bottles at the open MP presented significantly high values 0.76 (SD = ±0.12) ng C · L?1 · h?1and 0.53 (SD = ±0.06) ng C · L?1 · h?1. Our results showed that high net production rate of MAAs in the open MP was enhanced by a combination of osmotic and UVR stress and that in situ net production rates of individual MAA can be determined using 13C tracer in MPs in Arctic sea ice.  相似文献

In Greenland, free‐living red coralline algae contribute to and dominate marine habitats along the coastline. Lithothamnion glaciale dominates coralline algae beds in many regions of the Arctic, but never in Godthåbsfjord, Greenland, where Clathromorphum sp. is dominant. To investigate environmental impacts on coralline algae distribution, calcification and primary productivity were measured in situ during summers of 2015 and 2016, and annual patterns of productivity in L. glaciale were monitored in laboratory‐based mesocosm experiments where temperature and salinity were manipulated to mimic high glacial melt. The results of field and cold‐room measurements indicate that both L. glaciale and Clathromorphum sp. had low calcification and photosynthetic rates during the Greenland summer (2015 and 2016), with maximum of 1.225 ± 0.17 or 0.002 ± 0.023 μmol CaCO 3 · g?1 · h?1 and ?0.007 ±0.003 or ?0.004 ± 0.001 mg O2 · L?1 · h?1 in each species respectively. Mesocosm experiments indicate L. glaciale is a seasonal responder; photosynthetic and calcification rates increase with annual light cycles. Furthermore, metabolic processes in L. glaciale were negatively influenced by low salinity; positive growth rates only occurred in marine treatments where individuals accumulated an average of 1.85 ± 1.73 mg · d?1 of biomass through summer. These results indicate high freshwater input to the Godthåbsfjord region may drive the low abundance of L glaciale , and could decrease species distribution as climate change increases freshwater input to the Arctic marine system via enhanced ice sheet runoff and glacier calving.  相似文献

The rate of emergence of micropredatory gnathiid isopods from the benthos, the proportion of emerging gnathiids potentially eaten by Labroides dimidiatus, and the volume of blood that gnathiids potentially remove from fishes (using gnathiid gut volume) were determined. The abundance (mean ±s.e .) of emerging gnathiids was 41·7 ± 6·9 m?2 day?1 and 4552 ± 2632 reef?1 day?1 (reefs 91–125 m2). The abundance of emerging gnathiids per fish on the reef was 4·9 ± 0·8 day?1; but excluding the rarely infested pomacentrid fishes, it was 20·9 ± 3·8 day?1. The abundance of emerging gnathiids per patch reef was 66 ± 17% of the number of gnathiids that all adult L. dimidiatus per reef eat daily while engaged in cleaning behaviour. If all infesting gnathiids subsequently fed on fish blood, their total gut volume per reef area would be 17·4 ± 5·6 mm3 m?2 day?1; and per fish on the reefs, it would be 2·3 ± 0·5 mm?3 fish?1 day?1 and 10·3 ± 3·1 mm3 fish?1 day?1 (excluding pomacentrids). The total gut volume of gnathiids infesting caged (137 mm standard length, LS) and removed from wild (100–150 mm LS) Hemigymnus melapterus by L. dimidiatus was 26·4 ± 24·6 mm3 day?1 and 53·0 ± 9·6 mm3 day?1, respectively. Using H. melapterus (137 mm LS, 83 g) as a model, gnathiids had the potential to remove, 0·07, 0·32, 0·82 and 1·63% of the total blood volume per day of each fish, excluding pomacentrids, caged H. melapterus and wild H. melapterus, respectively. In contrast, emerging gnathiids had the potential of removing 155% of the total blood volume of Acanthochromis polyacanthus (10·7 mm LS, 0·038 g) juveniles. That L. dimidiatus eat more gnathiids per reef daily than were sampled with emergence traps suggests that cleaner fishes are an important source of mortality for gnathiids. Although the proportion of the total blood volume of fishes potentially removed by blood‐feeding gnathiids on a daily basis appeared to be low for fishes weighing 83 g, the cumulative effects of repeated infections on the health of such fish remains unknown; attacks on small juvenile fishes, may result in possibly lethal levels of blood loss.  相似文献

  • Plant growth at both growth stages was notably reduced in response to eUV‐B, except for the number of nodes and tillers at vegetative and reproductive stages. At anthesis, tillering increased due to the perennial habit of this plant, but leaf senescence reduced the number of leaves per tiller. Unlike ambient UV‐B, eUV‐B at the vegetative stage resulted in diversion of photosynthate for the formation of secondary metabolites (tannins and phenolics), providing dual protection from photooxidative damage and from herbivory.
  • The forage biomass as well as quality showed a marked decline under eUV‐B and relative nutritive value was reduced at both growth stages.

Ventilation was measured directly in the hagfish, Myxine glutinosa L., by means of an electro-magnetic blood flowmeter. Ventilatory flow and frequency increased from 0.86 ± 0.27 ml·min?, and 18.2 ± 5.1·min?, respectively, at 7°C to 1.70 ± 0.20 ml·min?, and 70.1 ± 9.5·min? at 15 ·C.Standard oxygen consumption,V?O2, was measured in non-buried hagfish. The red seaweed Gracilariopsis is an important crop extensively cultivated in China for high‐quality raw agar. In the cultivation site at Nanao Island, Shantou, China, G. lemaneiformis experiences high variability in environmental conditions like seawater temperature. In this study, G. lemaneiformis was cultured at 12, 19, or 26°C for 3 weeks, to examine its photosynthetic acclimation to changing temperature. Growth rates were highest in G. lemaneiformis thalli grown at 19°C, and were reduced with either decreased or increased temperature. The irradiance‐saturated rate of photosynthesis (Pmax) decreased with decreasing temperature, but increased significantly with prolonged cultivation at lower temperatures, indicating the potential for photosynthesis acclimation to lower temperature. Moreover, Pmax increased with increasing temperature (~30 μmol O2 · g?1FW · h?1 at 12°C to 70 μmol O2 · g?1FW · h?1 at 26°C). The irradiance compensation point for photosynthesis (Ic) decreased significantly with increasing temperature (28 μmol photons · m?2 · s?1 at high temperature vs. 38 μmol photons · m?2 · s?1 at low temperature). Both the photosynthetic light‐ and carbon‐use efficiencies increased with increasing growth or temperatures (from 12°C to 26°C). The results suggested that the thermal acclimation of photosynthetic performance of G. lemaneiformis would have important ecophysiological implications in sea cultivation for improving photosynthesis at low temperature and maintaining high standing biomass during summer. Ongoing climate change (increasing atmospheric CO2 and global warming) may enhance biomass production in G. lemaneiformis mariculture through the improved photosynthetic performances in response to increasing temperature.  相似文献

The toxigenic diatom Pseudo‐nitzschia cuspidata, isolated from the U.S. Pacific Northwest, was examined in unialgal batch cultures to evaluate domoic acid (DA) toxicity and growth as a function of light, N substrate, and growth phase. Experiments conducted at saturating (120 μmol photons · m?2 · s?1) and subsaturating (40 μmol photons · m?2 · s?1) photosynthetic photon flux density (PPFD), demonstrate that P. cuspidata grows significantly faster at the higher PPFD on all three N substrates tested [nitrate (NO3?), ammonium (NH4+), and urea], but neither cellular toxicity nor exponential growth rates were strongly associated with one N source over the other at high PPFD. However, at the lower PPFD, the exponential growth rates were approximately halved, and the cells were significantly more toxic regardless of N substrate. Urea supported significantly faster growth rates, and cellular toxicity varied as a function of N substrate with NO3?‐supported cells being significantly more toxic than both NH4+‐ and urea‐supported cells at the low PPFD. Kinetic uptake parameters were determined for another member of the P. pseudodelicatissima complex, P. fryxelliana. After growth of these cells on NO3? they exhibited maximum specific uptake rates (Vmax) of 22.7, 29.9, 8.98 × 10?3 · h?1, half‐saturation constants (Ks) of 1.34, 2.14, 0.28 μg‐at N · L?1, and affinity values (α) of 17.0, 14.7, 32.5 × 10?3 · h?1/(μg‐at N · L?1) for NO3?, NH4+ and urea, respectively. These labo‐ratory results demonstrate the capability of P. cuspidata to grow and produce DA on both oxidized and reduced N substrates during both exponential and stationary growth phases, and the uptake kinetic results for the pseudo‐cryptic species, P. fryxelliana suggest that reduced N sources from coastal runoff could be important for maintenance of these small pennate diatoms in U.S. west coast blooms, especially during times of low ambient N concentrations.  相似文献

Populations of Cladophora with two different levels of ploidy, n = 18/2n = 36 (18/36) and n = 24/2n = 48 (24/48), are present in creeks in the southern part of the province of Buenos Aires, Argentina. The goals of our study were to 1) relate the number of apical branches · mm?2 in 18/36 and 24/48 populations with the water velocity at the collection site; 2) correlate the number of apical ramifications · mm?2 in plants of the same population (24/48) growing in sectors with distinct water velocities; 3) compare cell length among populations with different ploidy levels, analyzing the sources of variation in different sectors of the same creek and in different plants of the same sector; and 4) analyze the sources of variation in cell length in 24/48 populations, including variations among different creeks. Our results suggest that 1) the number of branches · mm?2 tends to increase with higher water velocity; 2) the 24/48 populations have more ramifications · mm?2 than the 18/36 ones; 3) the length of vegetative cells is not an adequate criterion for differentiating between 18/36 and 24/48 populations; and 4) variations in vegetative cell length in 24/48 populations are highly significant among plants from different sectors of the same creek.  相似文献
