We investigated the feasibility of producing bioethanol from mixed softwood pretreated with the ionic liquid 1-butyl-3-methylimidazolium acetate ([Bmim]Ac). The optimal pretreatment conditions were determined by response surface methodology to be 100°C for 15 h, and the fermentable sugar yield was estimated to be 92.5%. Efficient pretreatment of softwood was maintained even after reutilizing [Bmim]Ac up to four times. Through the enzymatic saccharification and subsequent fermentation, bioethanol was produced with 0.42 g/g of yield and 0.24 g/L/h of productivity, which clearly suggests that efficient and economical bioethanol production can be achieved by optimizing pretreatment processes and reutilizing ionic liquid.  相似文献

Lignocellulosic feedstocks are utilized for the production of fuel ethanol and butanol through dilute acid/enzymatic hydrolysis and fermentation. Hydrolysis residue, a major by-product of biomass hydrolysis, is rich in recalcitrant carbon as majority of cellulosic and hemicellulosic components are released during pretreatment. With the intention of their effective utilization, hydrolysis residues from forestry (pinewood), energy crop system (timothy grass), and agriculture (wheat straw) were pyrolysed in a fixed-bed reactor at 600 °C with slow heating rate of 5 °C/min for 4 h. In order to understand the product (biochar, bio-oil, and gases) properties and advocate their energy and environmental values, chemical characterizations such as carbon–hydrogen–nitrogen–sulfur analysis, inductively coupled plasma-mass spectrometry, pH, electrical conductivity, scanning electron microscopy, porosity analysis, thermogravimetric analysis, X-ray diffraction, Fourier transform infrared (FTIR) and Raman spectroscopy, nuclear magnetic resonance (NMR) and gas chromatography–mass spectrometry (GC-MS) were employed. The yield of biochar, bio-oil and gases was 38.9–41.7, 18.6–22.3, and 24.9–28.8 wt%, respectively. The high pH and electrical conductivity of biochars with substantial amounts of Na, Mg, K, and Ca indicated their alkaline and saline nature, which would necessitate proper agronomical soil applications. Variable intensities of C–C, C–H, C–O, O–H, and C–N functional groups were detected in the FTIR spectra of residues, biochars, and bio-oils. Raman spectroscopy showed the development of graphite (1,580–1,610 cm?1) and defect (1,325–1,380 cm?1) carbon structures in biochars. 1H NMR of bio-oils indicated aromatics, olefinics, and aliphatics, whereas 13C NMR indicated carbonyls, aromatics, carbohydrates, alkyls, methoxy, and hydroxy carbon. GC studies of pyrolysis gases identified chiefly H2 and CO with traces of CH4, CO2, and C2+ components.  相似文献

One molecular species of prothoracicotropic hormone with a molecular weight of about 22, 000 (22K-PTTH) of the silkworm, Bombyx mori, was isolated from 5 × 105 adult heads. The purification procedure consisted of 16 steps including defatting, salt-extraction, fractional precipitations, conventional column chromatographies, and high performance liquid chromatographies. An approximately 5 × 106-fold purification was attained to yield 5.4 μg (0.25 nmol) of the pure hormone with a recovery of 3.3%. Injection of 0.11 ng of the purified 22K-PTTH could elicit adult development in a brainless Bombyx pupa. 22K-PTTH is a basic protein (pI 7.7 ~ 8.7) containing disulfide bond(s). The amino-terminal amino acid sequence of 22K-PTTH was determined to be Gly-Asn-Ile-Gln-Val-Glu-Asn-Gln-Ile-Pro-Asp-Pro-.  相似文献

The release kinetics of four model aroma compounds from coarse (d 32 = 1.0 μm) and fine (d 32 = 0.25 μm) eicosane and hydrogenated palm stearin (HPS) emulsions prepared with either solid or liquid lipid droplets were measured using a model mouth instrument. For both lipids, the release of aroma compounds from emulsions with solid droplets was higher than from emulsions with liquid droplets. This difference was greater for less polar aroma compounds. The release from solid eicosane droplets increased with particle size but no such effect was observed for HPS emulsions, however, the release from solid eicosane was higher than solid HPS. The initial aroma release profile of the solid droplet emulsion matches that of a similar liquid oil emulsion but requires much less added aroma. Abstract

Criteria are presented which enable the differentiation between the extent of solid-like and liquid-like character in a heterogeneous system on a per atom basis. Such criteria are developed for two different interatomic potentials, the Stillinger-Weber model for group IV semiconductors and the Lennard-Jones model for insulators. For the Stillinger-Weber potential model, three criteria are presented: one based on the coordination number of nearest neighbors, one based on the three-body energy, and one based on the angular positions (spatial arrangements) of neighbors. For the Lennard-Jones model an angular criterion is used. The difficulties associated with the assignation of interfacial atoms (those with partial solid and partial liquid character) are discussed. The effectiveness of these criteria for both models is tested by application to the identification of solid-like nuclei in the melt.  相似文献

Bacteria in oil-contaminated and oil-free estuarine water and sediment were isolated on solid and in liquid petroleum media. A greater variety of bacteria was noted when direct plating techniques were employed, compared with liquid enrichment. In addition, strains isolated by liquid enrichment were found more frequently to be capable of utilizing model petroleum in vitro , compared with isolates obtained by direct plating.  相似文献

在常温下,分离水葫芦为水葫芦渣和水葫芦汁,水葫芦渣与猪粪结合进行批量发酵试验,研究了产气量及原料半纤维素、纤维素、木质素的降解,结果表明原料的产气率为504.04 L/g TS,甲烷含量65%,半纤维素、纤维素降低,木质素升高.对水葫芦汁厌氧发酵的产气及COD降解情况进行了研究,结果表明水葫芦汁的产气潜力为2.192 mL/g,COD降解率达到91.27%.  相似文献

This paper presents a novel one-step process for converting a liquid stabilized nano-suspension into a solid formulation via hot-melt extrusion combined with an internal devolatilization process (nano-extrusion, NANEX). A polymer (Soluplus®) was fed into the extruder and molten, after which a stable nano-suspension was added via side-feeding devices. The solvent (water) was removed by devolatilization and the polymer solidified at the outlet. The solid material can be tableted or filled in a capsule directly. The results showed that the obtained extrudates comprised nanocrystals in the de-aggregated form, confirming that a solid nano-formulation was prepared. This method is capable of overcoming many of the problems associated with other processes involving solid nano-dosage forms and poses a straightforward approach towards manufacturing such products.  相似文献

From view point of the study on LET effects on bacteria, log- and stationary-phase cells of M. radiodurans, an extremely radioresistant vegetative bacterium, suspended in the phosphate buffer were exposed to 26 MeV α-particles from the IPCR cyclotron. The results obtained were compared with those on γ-irradiation. On such liquid-phase irradiation, survival curves of M. radiodurans for α-particles were found exponential while they were of sigmoidal or cumulative type for γ-rays. In addition, the dose-rate effect was observed with α-bombardment, although it was not found with γ-irradiation. For a given dose and in a system where the effective volume was sufficiently small as compared with the total sample volume, the relation of the surviving fraction S to the beam intensity I was represented by an empirical equation, where t is irradiation time, q0 is D37, and k and p are constant. By means of sedimentation analysis, double-strand scissions in DNA caused by α-particles were found repairable during the postirradiation incubation similarly to the case of γ-irradiation. The findings observed here with M. radiodurans do not support the view that DNA is an only primary target for cell killing by α-particles.  相似文献

Microalgal biomass was hydrolyzed using a solid acid catalyst with the aid of liquid acid. The use of solid acid as the main catalyst instead of liquid acid was to omit subsequent neutralization and/or desalination steps, which are commonly required in using the resulting hydrolysates for microbial fermentation. The hydrolysis of 10 g/L of lipid-extracted Chlorella vulgaris containing 12.2% carbohydrates using 7.6 g/L Amberlyst 36 and 0.0075 N nitric acid at 150°C resulted in 1.08 g/L of mono-sugars with a yield of 88.5%. For hydrolysis of higher concentrations of the biomass over 10 g/L, the amount of Amberlyst 36 needed to be increased in proportion to the biomass concentration to maintain similar levels of hydrolysis performance. Increasing the solid acid concentration protected the surface of the solid acid from being severely covered by cell debris during the reaction. A hydrolysate of lipid-extracted C. vulgaris 50 g/L was used, with no post-treatment of desalination, for the cultivation of Klebsiella oxytoca producing 2,3-butanediol. Cell growth in the hydrolysate was found to be almost the same as in the conventional medium with the same monosaccharide composition, confirming its fermentation compatibility. Spirillum volutans grows only under microaerobic conditions in a peptone-succinate-salts broth, but can grow aerobically when the peptone is replaced by vitamin-free acid-hydrolyzed casein broth. The addition of potassium metabisulfite, norepinephrine, catalase or superoxide dismutase (SOD) permitted aerobic growth in peptone-succinate-salts broth. A combination of catalase and SOD had a synergistic effect. S. volutans lacked catalase and had only a low level of peroxidase activity, but did possess SOD activity (12 to 14 U/mg of protein). The organism was found to be extraordinarily sensitive to exogenous hydrogen peroxide. Illumination of peptone-succinate-salts broth generated hydrogen peroxide and rendered the medium inhibitory to growth. A combination of catalase and SOD prevented this inhibition. Growth of S. volutans on solid media, not previously possible, was accomplished by the use of vitamin-free acid-hydrolyzed casein and peptone-succinate-salts agar media; maximum growth responses were dependent on the following combination of factors: addition of bisulfite, catalase, or SOD, protection of the media from illumination, incubation in a highly humid atmosphere, and incubation under atmospheres of 12% oxygen or less. The results indicate that the microaerophilic nature of S. volutans is attributable largely to the high sensitivity of the organism to exogenous hydrogen peroxide and, to a lesser extent, superoxide radicals occurring in the culture medium.  相似文献

In-depth analyses of cancer cell proteomes are needed to elucidate oncogenic pathomechanisms, as well as to identify potential drug targets and diagnostic biomarkers. However, methods for quantitative proteomic characterization of patient-derived tumors and in particular their cellular subpopulations are largely lacking. Here we describe an experimental set-up that allows quantitative analysis of proteomes of cancer cell subpopulations derived from either liquid or solid tumors. This is achieved by combining cellular enrichment strategies with quantitative Super-SILAC-based mass spectrometry followed by bioinformatic data analysis. To enrich specific cellular subsets, liquid tumors are first immunophenotyped by flow cytometry followed by FACS-sorting; for solid tumors, laser-capture microdissection is used to purify specific cellular subpopulations. In a second step, proteins are extracted from the purified cells and subsequently combined with a tumor-specific, SILAC-labeled spike-in standard that enables protein quantification. The resulting protein mixture is subjected to either gel electrophoresis or Filter Aided Sample Preparation (FASP) followed by tryptic digestion. Finally, tryptic peptides are analyzed using a hybrid quadrupole-orbitrap mass spectrometer, and the data obtained are processed with bioinformatic software suites including MaxQuant. By means of the workflow presented here, up to 8,000 proteins can be identified and quantified in patient-derived samples, and the resulting protein expression profiles can be compared among patients to identify diagnostic proteomic signatures or potential drug targets.  相似文献

Fast pyrolysis of lignocellulosic biomass is a facile method for producing high yields of liquid fuel intermediates. However, because most fast pyrolysis oils are highly oxygenated, acidic, and unstable, identification of feedstocks that produce higher quality pyrolysis liquids is desirable. Therefore, the effect of feedstock protein content was studied by performing fast pyrolysis experiments on biomass with varying protein content. The feedstocks ranged from low-protein content, ??5% up to feedstocks with >40 wt.% protein content. Protein content was not a major factor in the yield of pyrolysis oil or the distribution of biomass carbon into the pyrolysis products. However, elevated levels of protein did cause a deoxygenation effect in the pyrolysis process with more of the oxygen rejected from the biomass as water. The deoxygenation caused the pyrolysis oil from the higher protein containing biomass to have higher energy content. Furthermore, the concentration of basic nitrogen groups caused the pyrolysis oil from the higher protein biomass to shift to a more neutral pH and lower total acid number than has been measured typically for lignocelluloic biomass pyrolysis oils. Some of the pyrolysis oils, particularly those from the mustard seed family presscakes exhibited better thermal stability than low-protein pyrolysis oils.  相似文献

Bentazon removal by Ganoderma lucidum cultured in liquid and solid state conditions was compared in this work. In solid state cultures, the fungus produced both ligninolytic enzymes, namely laccase and Mn peroxidase. In liquid cultures, the main ligninolytic enzyme produced was laccase. In both types of cultures bentazon improved the production of laccase without significant alteration in the production of Mn peroxidase. In solid state cultures, where high levels of both laccase and Mn peroxidase activities were found, the fungus was more resistant to the action of the herbicide (50 mM in solid state cultures against 20 mM in liquid cultures) and more efficient in removing bentazon (90% removal against 55% in liquid cultures after 10 days of cultivation). Furthermore, the solid state culture filtrates were more efficient in the in vitro degradation of bentazon than the liquid culture filtrates. These observations suggest that both enzymes, laccase and Mn peroxidase, are involved in bentazon degradation. The results further suggest that solid state cultures of Ganoderma lucidum could be useful in strategies designed to reduce environmental contamination by bentazon.  相似文献

Conditions have been selected for reversed-phase high-performance liquid chromatographic assay of intermediate products formed in the course of utilization of toluene by Pseudomonas putida. The composition of products indicates that degradation of toluene by strain BS590-P proceeds primarily through the formation of benzoate and catechol. This is followed by degradation of catechol via ortho-cleavage. In strain BS3701-P, toluene oxidation involves both the side chain and the aromatic ring.  相似文献
