The heart-forming fields: one or multiple?   总被引:4,自引:0,他引:4  
The recent identification of a second mesodermal region as a source of cardiomyocytes has challenged the views on the formation of the heart. This second source of cardiomyocytes is localized centrally on the embryonic disc relative to the remainder of the classic cardiac crescent, a region also called the pharyngeal mesoderm. In this review, we discuss the concept of the primary and secondary cardiogenic fields in the context of folding of the embryo, and the subsequent temporal events involved in formation of the heart. We suggest that, during evolution, the heart developed initially only with the components required for a systemic circulation, namely a sinus venosus, a common atrium, a 'left' ventricle and an arterial cone, the latter being the myocardial outflow tract as seen in the heart of primitive fishes. These components developed in their entirety from the classic cardiac crescent. Only later in the course of evolution did the appearance of novel signalling pathways permit the central part of the cardiac crescent, and possibly the contiguous pharyngeal mesoderm, to develop into the cardiac components required for the pulmonary circulation. These latter components comprise the right ventricle, and that part of the left atrium that derives from the mediastinal myocardium, namely the dorsal atrial wall and the atrial septum. It is these elements which are now recognized as developing from the second field of pharyngeal mesoderm. We suggest that, rather than representing development from separate fields, the cardiac components required for both the systemic and pulmonary circulations are derived by patterning from a single cardiac field, albeit with temporal delay in the process of formation.  相似文献

Cryptococcus neoformans, the agent of cryptococcosis, had been considered a homogeneous species until 1949 when the existence of four serotypes was revealed based on the antigenic properties of its polysaccharide capsule. Such heterogeneity of the species, however, remained obscure until the two morphologically distinct teleomorphs of C. neoformans were discovered during the mid 1970s. The teleomorph Filobasidiella neoformans was found to be produced by strains of serotype A and D, and Filobasidiella bacillispora was found to be produced by strains of serotype B and C. Ensuing studies revealed numerous differences between the anamorphs of the two Filobasidiella species with regard to their ecology, epidemiology, pathobiology, biochemistry and genetics. At present, the etiologic agent of cryptococcosis is classified into two species, C. neoformans (serotypes A and D) and Cryptococcus gattii (serotypes B and C). Intraspecific genetic diversity has also been revealed as more genotyping methods have been applied for each serotype. As a result, the number of scientifically valid species within C. neoformans has become a controversial issue because of the differing opinions among taxonomists as to the appropriate definition of a species. There are three major species concepts that govern classification of organisms: phenetic (morphologic, phenotypic), biologic (interbreeding) and cladistic (evolutionary, phylogenetic). Classification of the two C. neoformans species has been based on the phenetic as well as the biologic species concept, which is also supported by the cladistic species concept. In this paper, we review and attest to the validity of the current two-species system in light of the three major species concepts.  相似文献

The association between obesity and hypertension is well documented, although the exact nature of this relation remains unclear. Sympathetic nervous and renin-angiotensin-aldosterone system activation appear to play an important role in the sodium and water retention, rightward shift in the pressure-natriuresis, and blood pressure elevation observed in obese individuals. Visceral obesity and the ectopic deposition of adipose tissue may be important in the activation of these systems and in the target organ damage that ensues. Weight loss is critical in the effective management of obesity hypertension and the accompanying target organ damage, although recidivism rates are high. However, prevention of weight gain should be the major priority for combating hypertension and its consequences in the future. The present review will provide an overview of our understanding of the etiology, pathophysiology, and treatment of obesity hypertension. Our focus is on the state of knowledge in humans. The potential role of abdominal obesity is considered throughout our review. We refer to relevant animal literature for supportive evidence and where little or no data in humans are available.  相似文献

In multicellular organisms, cells are polarized in the plane of the epithelial sheet, revealed in some cell types by oriented hairs or cilia. Many of the underlying genes have been identified in Drosophila melanogaster and are conserved in vertebrates. Here we dissect the logic of planar cell polarity (PCP). We review studies of genetic mosaics in adult flies - marked cells of different genotypes help us to understand how polarizing information is generated and how it passes from one cell to another. We argue that the prevailing opinion that planar polarity depends on a single genetic pathway is wrong and conclude that there are (at least) two independently acting processes. This conclusion has major consequences for the PCP field.  相似文献

Two families of ATP-binding cassette (ABC) transporters in which one or two extracytoplasmic substrate-binding domains are fused to either the N- or C-terminus of the translocator protein have been detected. This suggests that two, or even four, substrate-binding sites may function in the ABC transporter complex. This domain organization in ABC transporters, widely represented among microorganisms, raises new possibilities for how the substrate-binding protein(s) (SBPs) might interact with the translocator. One appealing hypothesis is that multiple substrate-binding sites in proximity to the entry site of the translocation pore enhance the transport capacity. We also discuss the implications of multiple substrate-binding sites in close proximity to the translocator in terms of broadened substrate specificity and possible cooperative interactions between SBPs and the translocator.  相似文献

Many agricultural pest species occur in seasonal metapopulations with a period of asexual reproduction. We use evolutionary theory to predict timing of dispersal for such species, and identify four sequential phases: no dispersal, dispersal from initially occupied patches, dispersal from later colonized patches, and no dispersal. The third type of phase occurs only when reproductive rates are relatively high; we speculate that this could explain why among aphids there can be either one or two waves of dispersal during a season, depending on the species. Our model also explains other features of aphid biology, including a summer crash in colony size, and a decline in the number of colonies towards the end of each reproductive season. The presence of an additional surge of dispersal becomes more likely as season length increases, and does not require further evolution. This could have profound implications for pest management during future climatic warming.  相似文献

When, where and how is the head-tail axis of the embryo set up during development? These are such fundamental and intensely studied questions that one might expect them to have been answered long ago. Not so; we still understand very little about the cellular or molecular mechanisms that lead to the orderly arrangement of body elements along the head-tail axis in vertebrates. In this paper, we outline some of the major outstanding problems and controversies and try to identify some reasons why it has been so difficult to resolve this important issue.  相似文献

The power of genome-wide association studies (GWAS) rests on several foundations: (i) there is a significant amount of additive genetic variation, (ii) individual causal polymorphisms often have sizable effects and (iii) they segregate at moderate-to-intermediate frequencies, or will be effectively ‘tagged'' by polymorphisms that do. Each of these assumptions has recently been questioned. (i) Why should genetic variation appear additive given that the underlying molecular networks are highly nonlinear? (ii) A new generation of relatedness-based analyses directs us back to the nearly infinitesimal model for effect sizes that quantitative genetics was long based upon. (iii) Larger effect causal polymorphisms are often low frequency, as selection might lead us to expect. Here, we review these issues and other findings that appear to question many of the foundations of the optimism GWAS prompted. We then present a roadmap emerging as one possible future for quantitative genetics. We argue that in future GWAS should move beyond purely statistical grounds. One promising approach is to build upon the combination of population genetic models and molecular biological knowledge. This combined treatment, however, requires fitting experimental data to models that are very complex, as well as accurate capturing of the uncertainty of resulting inference. This problem can be resolved through Bayesian analysis and tools such as approximate Bayesian computation—a method growing in popularity in population genetic analysis. We show a case example of anterior–posterior segmentation in Drosophila, and argue that similar approaches will be helpful as a GWAS augmentation, in human and agricultural research.  相似文献

The primary objective of this study was to determine if the ventilatory (&OV0312;E) breakpoint during incremental exercise to fatigue is best represented by a single breakpoint (BP1), double breakpoint (BP2), or an exponential function (EXP) exhibiting no BP. A secondary objective was to determine if visual assessment of the BP was valid and reliable. Subjects (n = 49) were both men (n = 12) and women (n = 37) who identified either running (n = 31) or triathlon (n = 18) as their primary sport. Subjects completed a maximal oxygen test on a motor-driven treadmill according to a modified Bruce protocol. The difference in mean square error (MSE) for BP1 (4.91 ± 3.35 L) was slightly but significantly (p < 0.05) greater than in MSE for BP2 (2.75 ± 1.7 L) for VE. Mean square error for EXP (7.85 ± 5.1 L) was significantly greater (p < 0.05) than BP1 and BP2. Visual assessment of VE on scatterplots produced poor validity, with a mean of 71.7% being misclassified compared to the computer model. Both interevaluator reliability (30.6% agreement) and intraevaluator reliability (59.7% agreement) were low. From the results of this study, it is concluded that (a) VE exhibits a BP2 model rather than BP1 or EXP and (b) visual assessment of BP is both invalid and unreliable.  相似文献

We have determined histone stoichiometries in nuclei from several sources by a direct chemical method, with the particular aim of quantitating histone H1 and, in chicken erythrocytes, H5, and of distinguishing between one and two molecules per nucleosome. The four histones H3, H4, H2A and H2B are found in equimolar amounts, as expected for the core histone octamer. The molar ratio of H1 in lymphocyte and glial nuclei is 1.0 per octamer, and in liver nuclei from three species 0.8 per octamer. These results suggest that each nucleosome has one H1 molecule; nucleosomes could acquire two molecules of H1 only at the expense of others containing none. The stoichiometry of H5 in chicken erythrocyte nuclei is similar to that of H1 in other nuclei, being about 0.9 molecules per nucleosome; the H1 also present in these nuclei amounts to 0.4 molecules per nucleosome.  相似文献

Alkaline phosphatase and horseradish peroxidase are the most commonly used reporter systems in chemiluminescent enzyme immunoassay (CLEIA). Which one, therefore, would be better when establishing a CLEIA method for a new target substance? There was no standard answer. In this study, both reporters were compared systematically including luminescence kinetics, conjugation methods, optimal condition and detection performance, using two common drugs, SD‐methoxy‐pyrimidine and enrofloxacin, as determination objects. The results revealed that there was much difference between the luminescence kinetics of the two systems. However, there was little difference between these systems when detecting the same substance, including in optimal conditions and determination of performance. Both reporters were suitable for establishing chemiluminescent enzyme immunoassays. Therefore, the choice of alkaline phosphatase or horseradish peroxidase as the reporter system in chemiluminescent enzyme immunoassays depends on availability. Conversely, these two report systems could be applied in simultaneous analysis of multicomponents due to their different optical behaviors and similar performances. But attention should be paid to conjugation method and coating buffer, which affected the luminescent intensity of different determination targets. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

Oxysterols are potent signalling lipids that directly bind liver X receptors (LXRs) and a subset of oxysterol binding protein (OSBP) related proteins (ORPs). It is relatively well established that the oxysterol-regulated function of LXRs is to control the expression of genes involved in reverse cholesterol transport, catabolism of cholesterol, and lipogenesis. In contrast, the mechanisms by which oxysterols and ORPs affect cellular lipid metabolism have remained poorly understood. In this review, we summarize the information available on function of the ORPs and compare the two families of proteins binding oxysterol to demonstrate the different responses that similar lipids can elicit within cells. The other focus is on the membrane targeting determinants and the protein interaction partners of ORPs, which provide interesting clues to the mode(s) of ORP action. Specifically, we suggest a model in which a general property of ORPs is to function at membrane contact sites, specialized zones of communication between two different organelles.  相似文献

A computer simulation study has been made of the accuracy of estimates of Theta = 4Nemu from a sample from a single isolated population of finite size. The accuracies turn out to be well predicted by a formula developed by Fu and Li, who used optimistic assumptions. Their formulas are restated in terms of accuracy, defined here as the reciprocal of the squared coefficient of variation. This should be proportional to sample size when the entities sampled provide independent information. Using these formulas for accuracy, the sampling strategy for estimation of Theta can be investigated. Two models for cost have been used, a cost-per-base model and a cost-per-read model. The former would lead us to prefer to have a very large number of loci, each one base long. The latter, which is more realistic, causes us to prefer to have one read per locus and an optimum sample size which declines as costs of sampling organisms increase. For realistic values, the optimum sample size is 8 or fewer individuals. This is quite close to the results obtained by Pluzhnikov and Donnelly for a cost-per-base model, evaluating other estimators of Theta. It can be understood by considering that the resources spent collecting larger samples prevent us from considering more loci. An examination of the efficiency of Watterson's estimator of Theta was also made, and it was found to be reasonably efficient when the number of mutants per generation in the sequence in the whole population is less than 2.5.  相似文献
