Fatty acid methyl esters of Polytrichum commune spore triglyceride and mono- and diglycosyl diglyceride fractions were analysed by glass capillary column gas chromatography provided with a precolumn system. The composition of the fatty acids in the lipid fractions differed only quantitatively: the diglycosyl diglyceride fraction was characterized by a high content of C 18: 3ω3 (67.7%), and the triglyceride and monoglycosyl diglyceride fractions by about 35%. The monoglycosyl diglyceride fraction contained a high proportion of C 14: 0 (18.4%). In all fractions the content of polyunsaturated C 20 acids was low, ranging from trace amounts to 4.9%.  相似文献

Winter wheat (Triticum sativum L. cv. Nisu) was grown in sand which contained 0, 0.25, 0.5 and 1.0 mg S-ethyl dipropylthiocarbamate (EPTC) per kg air dry sand. The galactolipids of leaves of 21-day-old seedlings were isolated by preparative thin layer chromatography. The fatty acids of the mono- and digalactosyl diglycerides were analysed by gas liquid capillary chromatography. The major fatty acids of the wheat leaf galactolipids were palmitic, palmitoleic, stearic, oleic, vaccenic, linoleic and linolenic acids (in the monogalactosyl diglyceride fraction of untreated plants 22.5, 2.4, 3.1, 5.2, 2.5, 51.1 and 1526.6 μg and in the digalactosyl diglyceride fraction 108.8, 2.3, 10.4, 9.9, 8.2, 42.3 and 1120.7 μg/g leaf fresh weight, respectively). Total fatty acid content of the mono- and digalactosyl diglyceride fractions was decreased by 85 and 87%, respectively, at 1 mg EPTC/kg sand, while decrease in the fresh weight of the leaves was 79%. The content of linoleic and linolenic acids/g fresh weight of the leaves was decreased in the monogalactosyl diglyceride fraction by 27 and 43%, respectively, while the content of all other fatty acids was increased. In the digalactosyl diglyceride fraction the content of both linoleic and linolenic acids/g leaf fresh weight was decreased by 55%. The content of palmitic and vaccenic acids was also decreased, whereas the content of other fatty acids remained at the level of the untreated samples. The general quality of the fatty acids in the mono- and digalactosyl diglyceride fractions was altered slightly by EPTC.  相似文献

Dipolyunsaturated phosphatidylcholines from bovine retina contain a whole series of unusual fatty acids. Methyl esters from these acids are very strongly retained on polar and nonpolar gas-liquid chromatography stationary phases. On thin layers of silica-AgNO3, they separate as tetra-, penta-, and hexaenoic fatty acid methyl esters. After hydrogenation, the three polyunsaturated fractions give the same series of saturated methyl esters, having 20 (or 22)-36 carbon atoms. High pressure liquid chromatography, as well as gas-liquid chromatography, indicates that the new components of the three fractions are even-carbon homologs of well known polyenoic fatty acids of the n-6 and n-3 families, since they behave as series of 20-36-carbon tetraenoic (n-6), pentaenoic (n-3 and n-6), and hexaenoic (n-3) fatty acids. Their occurrence in phospholipid molecules also having docosahexaenoate (22:6) explains the separation of major dipolyunsaturated phosphatidylcholines from retina into dodecaenoic, undecaenoic, and decaenoic fractions after argentation thin layer chromatography. Using high pressure liquid chromatography, the latter are resolved into individual species having 10-12 double bonds and 42-58 carbon atoms. The unusual PCs are thus endowed not only with the highest degree of unsaturation, but with the longest hydrocarbon chains yet reported for vertebrate glycerophospholipids. It is shown that phosphatidylcholines containing the novel fatty acids are highly concentrated in photoreceptor membranes and that they occur in the retina of vertebrates so distant in evolution as fish, birds, and various mammals.  相似文献

1. Four Gastropoda species and two Bivalvia species from the Volga river basin were examined. 2. Distribution of phospholipids in the molluscs was studied by qualitative and quantitative micro thin-layer chromatography. 3. Major phospholipid classes, phosphatidylethanolamine and phosphatidylcholine, were found to contain plasmalogens. 4. One mollusc species notably contained 67 fatty acids including 25 saturated (both iso and anteiso), 24 monoenoic, five dienoic, four trienoic and eight polyenoic compounds identified by capillary gas chromatography; fatty acid contents in the other studied species were considerably lower. 5. Pure preparations of phosphatidylcholine were isolated from spinach leaf chloroplasts, spinach leaf microsomes, and cauliflower inflorescence. The isolated phosphatidylcholine was treated with snake venom phospholipase A, and the fatty acid distribution and composition of the fatty acid methyl esters prepared from the lysophosphatidylcholine and the freed fatty acid were determined by gas-liquid chromatography. The results showed that saturated fatty acids were preferentially esterified at position 1 and unsaturated fatty acids at position 2. The phosphatidylcholine from cauliflower was also treated with phospholipase C. The resulting diglycerides were fractionated on AgNO(3)-impregnated thin-layer plates. The diglyceride fractions were transesterified and the fatty acid composition of each was determined by gas-liquid chromatography. The predominant species contained linolenic acid only (22% of the total), linolenic and oleic acids (19%), and linolenic and palmitic acids (37%). These molecular species could not be accounted for by random distribution of the fatty acids.  相似文献

Rat brain has been shown to contain polyenoic very-long-chain fatty acids (VLCFA) belonging to the n-3 and n-6 series with four, five and six double bonds and even-carbon chain lengths from 24 to 38. These fatty acids are almost exclusively located in unusual molecular species of phosphatidylcholine at the sn-1 position of the glycerol backbone, whereas saturated, monoenoic and polyenoic fatty acids with less than 24 carbon atoms are present at the sn-2 position. Polyenoic VLCFA phosphatidylcholine in neonatal rat brain is enriched with n-6 pentaenoic and n-3 hexaenoic VLCFA with up to 36 carbon atoms, whereas the corresponding phospholipid in adult rat brain mainly contains n-6 tetraenoic and n-3 pentaenoic VLCFA with up to 38 carbon atoms. The total amount of polyenoic VLCFA associated with phosphatidylcholine is highest in the brain of immature animals. Polyenoic VLCFA phosphatidylcholine appears to be predominantly confined to nervous tissue in rats, and it is envisaged that this phospholipid is of physiological significance.  相似文献

About one-fourth the phosphatidylcholines (PCs) from bovine disk photoreceptor membranes contain very long chain (24-36 carbons) polyunsaturated (4, 5, and 6 double bonds) fatty acids of the n-3 and n-6 series (VLCPUFA). Such fatty acids, exclusively occurring in dipolyunsaturated species, are esterified to the sn-1 position of their glycerol backbone, docosahexaenoate being the major fatty acid at sn-2. Chromatographically, such PCs display a weakly polar character relative to other species, ascribable to their exceedingly large number of carbons. After hexane extraction of lyophilized disks, PC is the major component of the fraction of lipids that remains associated with rhodopsin, followed by phosphatidylserine, while a large proportion of the phosphatidylethanolamine is removed. The fatty acid composition of the hexane-removable and protein-bound lipid fractions markedly differs, the latter being enriched in lipid species containing long-chain and very long chain polyenes. This is observed for all lipid classes except free fatty acids. VLCPUFA-containing PCs are the most highly concentrated species in the rhodopsin-associated lipid fraction. The very long chain polyenes these PCs have at sn-1 may account for their resistance to being separated from the protein. It is hypothesized that their unusually long polyenoic fatty acids could be well suited to partially surround alpha-helical segments of rhodopsin.  相似文献

The n-6 tetra- and pentaenoic fatty acids with carbon chain lengths greater than 32 found in normal brain are located predominantly in a separable species of phosphatidylcholine. A similar phospholipid is found in increased amounts in the brain of peroxisome-deficient (Zellweger's syndrome) patients, but the fatty acid composition differs in that penta- and hexaenoic derivatives predominate. Our data strongly suggest that the polyenoic very long chain fatty acids are confined to the sn-1 position of the glycerol moiety, while the sn-2 position is enriched in saturated, monounsaturated and polyunsaturated fatty acids with less than 24 carbon atoms. It is postulated that these unusual molecular species of phosphatidylcholine may play some, as yet undefined, role in brain physiology.  相似文献

Higher steryl ester biosynthetic activities were obtained with Triton X-100-phosphatidylcholine-cholesterol mixed micelles than with Tween 80-phosphatidylcholine-cholesterol mixed micelles when incubated with spinach leaf (Spinacia oleracea L.) acetone powder preparations. The best incorporation of [4-14C]cholesterol into [4-14C]cholesteryl ester was obtained with a Triton X-100-phosphatidylcholine-cholesterol (10:1:1, w/w) mixed micelle system. This mixed micelle system, however, required 1,2-dipalmitin and fatty acid-free bovine albumin for optimal activity. The reaction exhibited a diglyceride specificity since the dipalmitin requirement could be replaced with neither 1-monopalmitin nor tripalmitin. Significant amounts of steryl ester biosynthetic activity were detected in the chloroplast (1,000g pellet), mitochondrial (3,000g pellet), and microsomal (20,000g and 88,000g pellet) fractions. Little activity was detected in the water-soluble (88,000g supernatant) fraction. The highest specific activity occurred in the 88,000g pellet. The 88,000g supernatant contained a heatstable, water-soluble substance that was required for optimal steryl ester biosynthesis in all of the pellet fractions. This factor was not lost during extensive dialysis but was destroyed by ashing, indicating that it was large and organic. Silver nitrate thin layer chromatography indicated that 60% of the biosynthesized steryl esters contained saturated fatty acids in the absence of 1,2-dipalmitin and that 83% contained saturated fatty acids in the presence of 1,2-dipalmitin.  相似文献

The plasma-borne long-chain free fatty acids (FFA) enter skeletal muscle cells. Upon entering they are oxidized or esterified and a fraction remains free (non-esterified). The data on free fatty acids in skeletal muscles remain highly controversial. Furthermore, the composition of individual fatty acids in various lipid fractions including free fatty acids, monoglyceride and diglyceride in muscles has not been characterized. Also data on the composition of fatty acids esterified into muscle triglycerides and phospholipids are incomplete. The present study was undertaken to examine a composition of fatty acids in lipid fractions of different skeletal muscle types. For this purpose, samples of the rat soleus, red and white portions of gastrocnemius were excised, trimmed of visible fat and fascias and immediately frozen in liquid nitrogen. Samples were then pulverized and, lipids were extracted and fractionated by thin-layer chromatography. Individual long-chain fatty acids in different fractions were identified, characterized and quantitated by gas-liquid chromatography. FFA composition in the plasma was also determined. The total FFA content in the soleus, red and white gastrocnemius was 69.1 ± 10.8, 49.0 ± 13.6 and 22.7 ± 8.6 nmol/g, respectively. Palmitic and oleic acids were the major fatty acids in the muscles FFA fraction. Monoglyceride fraction of each muscle contained palmitic, stearic and linoleic acid as the major fatty acids, Diglyceride fraction contained mostly palmitic and oleic acid whereas triglyceride fraction mostly palmitic and linoleic acid.. The fraction of phospholipids was composed mostly of palmitic and linoleic acid but contained also considerable percentage of archidonic acid. Total plasma FFA/muscle FFA ratio depended on a muscle type and was: 2.4 in the soleus, 3.5 in the red and 7.4 in the white gastrocnemius. This assured transport of FFA to the myocytes. However, there were great differences in the ratio between particular FFA within the same muscle as well between the muscles. It indicates that individual FFA are either selectively transported from the plasma to the muscles or selectively used within the myocytes or both.  相似文献

The multiplicity of phosphatidylcholines is caused by the presence of different pairs of fatty acids in their individual molecular species and at least 27 miscellaneous fatty acids were identified in phosphatidylcholines in the serum of healthy individuals by combined gas–liquid chromatography and mass spectrometry in our present experiments. A method is described for the separation and quantitation of molecular species of phosphatidylcholine in human serum. Total phosphatidylcholine is isolated from lipids extracted from the serum with chloroform–methanol (2:1) by reversed-phase liquid–liquid extraction and subjected to reversed-phase high-performance liquid chromatography with a discontinuous descending gradient of water. Separation is monitored by fluorometry (340/460 nm) and absorption at 205 nm, if required. Up to 25 different molecular species of phosphatidylcholine may be quantified with a satisfactory reproducibility (±5–8%). Data on the distribution of individual molecular species in phosphatidylcholine of 53 normal serums are presented. The method may be used for quantitation of these phospholipids also in other biological materials (cell lines, leukemic cells from patients), and on a micropreparative scale to isolate individual compounds. The speed of separation as well as a satisfactory reproducibility are its principal advantages.  相似文献

The total amount of fatty acids in the mono- (MGDG) and diglycosyl diglyceride (DGDG) and more polar lipid fractions of frozen Ceratodon purpureus shoots was 4.6, 3.4 and 4.0 mg/g dry weight, respectively. The respective values for the tops of frozen Pleurozium schreberi were 2.6, 3.3 and 3.8 mg/g dry weight. The molar ratios MGDG/DGDG and MGDG + DGDG/chlorophyll were 1.3 and 3.7, respectively, for C. purpureus and 0.8 and 3.5 for P. schreberi. In C. purpureus the main fatty acids in the MGDG fraction were C 18:3ω3 (44% of the total fatty acids) and C 16:3ω3 (26%); in the DGDG fraction C 18:3ω3 (70%); and in the more polar lipid fraction C 18: 3ω3 (26%) and C 16:0 (25%). The proportion of C 20 polyunsaturated fatty acids was 15, 12 and 19% of the total fatty acids found in the MGDG, DGDG and more polar lipid fractions, respectively. In P. schreberi the proportion of C 20 polyunsaturated fatty acids was high in all polar lipid fractions (47, 42 and 25% in MGDG, DGDG and more polar lipid fractions, respectively). In addition, MGDG and DGDG fractions contained abundantly C 18:3ω3 (32 and 45%, respectively), and the more polar lipid fraction both C 18: 3ω3 (24%) and C 16:0 (27%).  相似文献

Fatty acid composition, especially the distribution of eicosapolyenoic acids in several species of Gracilaria, was analyzed in relation to their taxonomy. The species have been grouped into two types based on distribution of these polyenoic acids: Type 1, which contains palmitic, oleic and arachidonic acids as the major components, and Type II, which contains eicosapentaenoic acid in addition to Type I fatty acids. Octadecapolyenoic acids were detected only in trace amounts in each Type. A similar remarkable difference also was observed in the fatty acid composition of lipid classes. The major component of eicosapolyenoic acids in Type I was arachidonic acid in all lipid classes. In Type II, eicosapentaenoic acid was the major component in monogalactosyl diacylglycerol, digalactosyl diacylglycerol, sulfoquinovosyl diacylglycerol and phosphatidylglycerol. Arachidonic and eicosapentaenoic acids were contained in large amounts in Type II phosphatidylcholine. Grouping of Gracilaria species into Type I and Type II is not entirely consistent with morphological and taxonomic features, but the difference in fatty acid composition is likely due to genetic rather than to environmental factors.  相似文献

Synthesis of fatty acids has been studied in aged potato slices. Formation of the very long chain fatty acids was inhibited by the presence of fluoride or by high incubation temperatures. Arsenite caused an increase in the percentage incorporation of radioactivity from acetate-[14C] into palmitic acid, apparently by inhibiting further elongation. The results indicate that the aged potato contains at least three enzymes responsible for saturated fatty acid synthesis, At short incubation times, the newly formed fatty acids were mainly unesterified but later become incorporated into phospholipids. Phosphatidylcholine contained the greatest proportion of radioactive fatty acids. Newly formed polyenoic fatty acids were principally transacylated into phosphatidylcholine and phosphatidylethanolamines. The very long chain fatty acids, on the other hand, were mainly located in the wax ester and unesterified fatty acid fractions, from which they can easily be converted into suberin components.  相似文献

Dried fruiting bodies of the basidiomyceteOudemansiella mucida contain 29.1% total lipids. Their qualitative analysis revealed the presence of mono-, di-, triglycerides, sterols, free fatty acids and sterolesters. Quantitatively most significant were triglycerides (37.9%) and free fatty acids (29.7%). The phospholipid fraction contained phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine and phosphatidic acid. Gas chromatography showed the presence of a broad spectrum of fatty acids. The ratio between the neutral and polar fractions was 6: 1, both having linoleic acid as the main component.  相似文献

Chloroplasts isolated from tobacco leaves in 0.5 M sucrose solution (the 1000 g pellet) contained 83% of the total cellular monogalactosyl diglyceride, 88% of the digalactosyl diglyceride, 76% of the sulfolipid, and 74% of the phosphatidyl glycerol. Phosphatidyl inositol was concentrated in the 15,000 g pellet. Phosphatidyl choline and phosphatidyl ethanolamine were concentrated in the 15,000 g supernatant fraction. Chloroplasts isolated from tobacco leaves by a nonaqueous technique in hexane-carbon tetrachloride show a glycerolipid composition similar to that found in chloroplasts isolated in the aqueous system, even though some lipid, particularly monogalactosyl diglyceride, is extracted by the organic solvent during the process.  相似文献

Casein hydrolysat, lactose and lipids (100 mg of fatty acids) were introduced in the stomach of rats by a gastric tube: either pure tri-oleoylglycerol, or phospholipids, or phosphatidylcholines, or the mixture 9/1 to fatty acid weight of tri-oleoylglycerol-phospholipids or phosphatidylcholines. The rats were killed 2 h later. The intraluminal intestinal lipids of the oil and micellar phases were separated after microfiltration (Millipore filters) in preference to the filtration by gel chromatography on polyacrylamide agarose, as an hydrolysis of intraluminal phospholipid occurred after the column elution. 1. After a quantitative recovery of the intestinal lipids (no separation of the oil and micellar phases), a strong hydrolysis of the tri-oleoyglycerol was observed; in opposition, large amounts of intact phospholipids appeared. 2. After isolation of the micellar phases, no triglycerides were recovered, but fatty acids and partial glycerides from the hydrolysed tri-oleoylglycerol and dietary phosphatidylcholines and small quantities of lyso-phosphatidylcholines (hydrolysed forms) were present. 3. After ingestion of the tri-oleoylglycerol as lipid dietary source, the intestinal micellar phases contained endogenous phosphatidylcholines and a few amounts of lysophosphatidylcholines, which had mainly bile origin, since the fatty acid composition of these micellar phosphatidylcholines approached the bile phosphatidylcholine fatty acid composition. The micellar lysophosphatidylcholine masses represented one-fourth of the micellar phosphatidylcholine masses. 4. In these experiments the phosphatidylcholine lysophosphatidylcholine ratio was always high: this means that small quantities of exogenous and endogenous lysophosphatidylcholines appeared in the micellar phases.  相似文献

With the aid of column chromatography, thin-layer chromatography, and gas chromatography, analyses were made of the fatty acid composition of mushroom glycerophospholipids: phosphatidylcholine (PC), phosphatidylethanolamine (PE), lysophosphatidylcholine (LPC), and lysophosphatidylethanolamine (LPE). Other glycerophospholipids were minor components. Twelve species of Basidiomycetes were analyzed. In some species PC and PE were not detected. Differences in qualitative composition of the fatty acids were not significant. In contrast, there were significant quantitative differences. The percentage composition of the unsaturated fatty acids varied from 0 to 74% in PC, and from 8 to 76% in PE, in various species of mushrooms. Fatty acids with 16 and 18 carbon atoms predominated, whereas the amount of those containing more than 18 carbon atoms was negligible. Hydroxy fatty acids were absent.  相似文献

The distribution of carbon tetrachloride-induced alterations of membrane lipids in various fractions of liver microsomal lipids was studied. The chromatographic spot (referred to as the “D” spot in the previous paper [1]) which has been shown to contain the compounds responsible for the diene conjugation absorption [1], was found in the fatty acid methyl esters prepared from the fraction containing phosphatidylethanolamine (PE) and also in those obtained from the fraction containing phosphatidylserine (PS) and phosphatidylinositol (PI). The absorption of conjugated dienes was very marked in PE and less intense in PS and PI. The fatty acid methyl esters prepared from the fraction containing phosphatidylcholine (PC) showed no presence of the “D” spot and minimal absorption of conjugated dienes.A decrease in arachidonic acid content was found in the fraction containing PE, while no change in content of this fatty acid was found in the fraction containing PC. Results similar to those observed for PC were also found for neutral lipids (NL).Analysis of the fatty acid methyl esters of the various lipid fractions by gas-liquid chromatography (GLC) with an electron capture detector (ECD) gave a qualitative index of the free radical attack by CCl4 metabolites. Quantitative estimation was attained by study of the irreversible binding of 14C from 14CCl4 to the various lipid fractions. It was found that the fraction containing PS had the highest specific activity, while the fraction containing PC had the lowest specific activity of all the phospholipids. Thin layer chromatography (TLC) of the fraction containing PS revealed that only 11% of the radioactivity was associated with the pure PS moiety, while the remainder was associated with uncharacterized lipids (probably oxidation products).The possible relevance of the alterations induced by carbon tetrachloride in the various phospholipid fractions of liver microsomes to functional changes is discussed.  相似文献

Liquid chromatographic class separations of common cellular phospholipids combined with plasma spray ionization of the effluents were investigated. Comparison with true thermospray ionization involving ammonium acetate buffering revealed a gain in total ionization in the plasma spray of a factor of approximately 10 using a cation-exchange column and a solvent mixture consisting of acetonitrile-methanol-water (400:100:15, v/v). Plasma spray ionization studies of bovine brain polyphosphoinositides interrelated by the phosphate content in the inositol moeity showed almost identical monoglyceride and diglyceride ion clusters, indicating possibilities of studying the biochemical turnover of such phospholipids. Plasma spray ionization liquid chromatography-mass spectrometry of bacterial membrane phospholipids (Pseudomonas fluorescens) revealed possibilities of obtaining indications of individual fatty acid compositions from the spectra of the phosphatidylinositol and phosphatidylethanolamine fractions present. Conventional gas chromatographic fatty acid analysis agreed with the direct mass spectrometric structure elucidations. Interestingly, the two phospholipid classes had different relative fatty acid compositions with a significantly higher degree of cyclic fatty acids in the phosphatidyl ethanolamines. Plasma spray ionization yielded linear dose-response curves for both the monoglyceride and diglyceride fragment signals in the selected-ion monitoring mode. The detection limit for the monoglyceride and diglyceride species of phosphatidylcholine under the chromatographic and mass spectrometric conditions used was found to be in the picogram range.  相似文献
