Summary Freshwater rainbow trout,Salmo gairdneri, were injected with different doses of estradiol-17 in order to induce the synthesis of a protein, regarded as identical to vitellogenin. The plasma levels of free and protein-bound calcium, magnesium and inorganic phosphate were studied in control and estradiol-17 treated fish, using an ultrafiltration method. Estradiol-17 caused a dose-dependent increase in plasma vitellogenin levels, which strongly correlated to protein-bound levels of calcium and magnesium in plasma. Calcium and magnesium were bound to vitellogenin in a ratio of 9:1, which was considerably higer than the protein-binding ratio of these ions in normal plasma (5.2:1). The dose-dependent increase in total plasma levels of calcium, magnesium and inorganic phosphate during estradiol-17 treatment was solely due to an increase in the protein-bound fraction of these ions. It is concluded that the physiologically important plasma levels of free calcium, magnesium and inorganic phosphate are effectively regulated at normal levels during vitellogenin synthesis.  相似文献

-1,3-Glucanase (Lu) was isolated from unfertilized eggs of the sea urchin Strongylocentrotus intermedius. A comparative study of some properties of -1,3-glucanase Lu and -1,3-glucanases with different action types—endo--1,3-glucanase from crystalline style of the marine mollusk Spisula sachalinensis (LIV) and exo--1,3-glucanase from the terrestrial snail Eulota maakii (LII)—was performed. It was found that -1,3-glucanase Lu hydrolyzes laminaran with a high yield of glucose in the reaction products. The enzyme hydrolyzes substrates with retention of the glycosidic bond configuration, is able to cleave modified substrates, and exhibits transglycosylating activity. All properties of -1,3-glucanase from S. intermedius were more similar to those of the endo--1,3-glucanase from the marine mollusk (LIV) than exo--1,3-glucanase LII from the terrestrial snail. The differences in the effect of LIV and Lu on laminaran are probably related to the functions of -1,3-glucanase Lu from sea urchin eggs (which, in contrast to LIV, is not a digestive enzyme).  相似文献

An efficient and reliable micropropagation system for Persian clover (Trifolium resupinatum L.) was developed using different explants and media. Node, hypocotyl and cotyledonary node explants were cultured on Murashige and Skoog (MS) medium supplemented with combinations of either 6-benzyladenine (BA) and indole-3-butyric acid (IBA) or BA, Kinetin (KIN) and IBA. Direct multiple shoots developed within 6weeks in all explants in most media tested. The best shoot multiplication capacity was obtained from cotyledonary node explants on MS medium containing 7.1M BA and 1M IBA or 14.1M BA and 1M IBA. Elongated shoots were rooted on either MS medium alone or combination with different concentrations of indole-3-butyric acid (IBA), indole-3-acetic acid (IAA) and -naphthaleneacetic acid (NAA). High rooting was achieved in half strength MS medium containing 8M IBA.  相似文献

An in frame gene fusion containing the coding region for mature -lactamase and the 3-end of hylA encoding the haemolysin secretion signal, was constructed under the control of a lac promoter. The resulting 53 kDa hybrid protein was specifically secreted to the external medium in the presence of the haemolysin translocator proteins, HlyB and HlyD. The specific activity of the -lactamase portion of the secreted protein (measured by the hydrolysis of penicillin G), approximately 1 U/g protein, was close to that of authentic, purified TEM--lactamase. This is an important example of a hybrid protein that is enzymatically active, and secreted via the haemolysin pathway. Previous studies have indicated that haemolysin is secreted directly into the medium, bypassing the periplasm, to which -lactamase is normally targeted. This study indicated, therefore, that normal folding of an active -lactamase, can occur, at least when fused to the HlyA C-terminus, without the necessity of entering the periplasm. Despite the secretion of approximately 5 g/ml levels of the active -lactamase fusion into the medium, there was maximally only a 50% detectable increase in the LD50 for resistance to ampicillin at the individual cell level. This result suggests that, normally, resistance to ampicillin requires a high concentration of the enzyme close to killing targets, i.e. in the periplasm, in order to achieve significant levels of protection.These authors made an equal contribution to this work  相似文献

Bacteria, yeasts, and molds which grew in a medium containing a synthetic lignin — a dehydrogenation polymer (DHP) of coniferyl alcohol — as a sole carbon source, were isolated from soil. One fungus, Fusarium solani M-13-1, was found to degrade the DHP most vigorously among the isolated organisms. It was shake-cultured in a medium containing dehydrodiconiferyl alcohol (DHCA) (I), an important lignin model compound, and the following six metabolic products were isolated and identified: 1) Phenylcoumaran--aldehydic (II) and -carboxylic compounds, 2) phenylcoumaran--aldehydic compound (IV), formed by release of a 2-carbon fragment from the phenylcoumaran--carboxylic compound, 3) 5-acetylvanillyl alcohol (V), formed by cleavage of the coumaran ring and reduction of the -aldehyde group, 4) 5-carboxyvanillyl alcohol (VI), formed by subsequent oxidation of the acetyl group, and 5) the -ether of DHCA (VII), considered to be a by-product. Pulse sequences are presented for the measurement of3JCC and3JNC scalar couplings for allC containing residues in15N,13C uniformly labeled proteins. The methodsdescribed are based on quantitative J correlation spectroscopy pioneered byBax and co-workers [Bax et al. (1994) Methods Enzymol., 239, 79–105].The combination of 3JCC and3JNC scalar coupling constants allows theassignment of discrete rotameric states about the 1 torsion angle in cases where such states exist or, alternatively,facilitates the establishment of noncanonical 1conformations or the presence of rotameric averaging. Summary Fathead minnows, Pimephales promelas, and yellow perch, Perca flavescens, were transferred from moderately soft Lake Superior water (hardness 45mg/l as CaCO3) to very soft diluted Lake Superior water (hardness 4.5mg/l). Sulfuric acid was added in some treatments by means of a multichannel diluter. In very soft water, chloride cells proliferated in the gills, especially in the epithelium of the secondary lamellae. When exposed to acid, chloride cells were damaged and less abundant in the secondary lamellae, and blood osmolality was reduced at pH 5.0 (x = 188 mOsm/kg, 9 days exposure; normal 280 mOsm/kg) for the minnows and pH 4.1 (x = 218 mOsm/kg, 58 days exposure; normal 329 mOsm/kg) for the perch. Certain chloride cells which form gland-like clusters in the primary lamellae of perch gills showed little damage even at pH 4.1. The present study supports the view that chloride cells proliferate in very soft fresh water to help maintain ionic balances, and that damage to these cells in acidified soft water may be related to diminished ionoregulatory capacity. The greater acid tolerance of chloride cells of, and the higher blood osmolality maintained by, perch could help to explain the greater tolerance of this species to low pH. In some cases, a species' ability to acclimate to very soft water and acidified soft water may depend upon the number, distribution, and physiology of its chloride cells.  相似文献

The effect of increased concentrations of calcium (Ca) (3–24 mM) and boron (B) (100–800 M) in the medium was studied on the occurrence of shoot tip necrosis (STN) in cultures of Pistacia vera L. STN was significantly reduced by application of Ca or B, however media with more than 200 M boron had reduced shoot multiplication. Ca (12–24 mM) supplied as calcium chloride reduced STN without any adverse effect on shoot multiplication or elongation, whereas calcium acetate reduced elongation. It is concluded that STN is a physiological mineral disorder associated with Ca and/or B deficiency in the meristematic regions of actively growing shoots. Application of Ca (up to 24 mM) as calcium chloride to the medium was the best treatment for the control of STN. Summary Sodium and water balance ofDytiscus verticalis in fresh water were investigated under three feeding regimes: unfed, and fed a diet either low or high in sodium chloride. Unfed sodium influx was 0.13 and sodium efflux was 0.74 moles/100 gwm·h. These values are low in comparison with most freshwater animals. The electrical potential difference across the integument in artificial soft water (ASW) was about 150 mV smaller than the potential necessary to maintain sodium balance in the absence of active transport. However, sodium influx did not show saturation kinetics over an external concentration range of 91 to 1725 M. Unfed beetles failed to arrest net sodium loss to baths that were initially distilled water or ASW, even when bath sodium concentrations reached 75–298 M. The long-term rate of net sodium loss ranged from 0.61 to 4.4 moles/100 gwm·h for four sets of animals. Beetles decreased sodium efflux during a period of fasting. During subsequent feeding, beetles fed a high sodium diet (HSD) increased sodium efflux while beetles fed a low sodium diet (LSD) maintained low rates of sodium efflux. HSD fed beetles increased body sodium and hemolymph sodium concentration, and expanded extracellular fluid, relative to LSD fed beetles. Summary As 15% of band 3 protein, the assumed chloride channel, is associated with spectrin, the major peripheral protein of a lattice located at the red cell membrane-cytosol interface, the present study was undertaken to evaluate whether a rearrangement of the lattice modifies the functional property of band 3 protein. Such a rearrangement was modulated by depletion of cell ATP and/or by accumulation of Ca2+ ions within the cell.ATP depletion induces an inhibition of the electroneutral one-for-one chloride exchanges. Neither the modification of red cell morphology due to ATP depletion (discocyte-echinocyte transformation) nor a direct effect of the decrease in internal ATP level can account for this inhibition. On the other hand, it seems reasonable to consider that inhibition is related to the changes in membrane protein organization (formation of heteropolymers) induced by the decrease in ATP level. But it does not appear that the degree of inhibition is modified when this altered assembly of membrane protein is stabilized by disulfide linkages.Accumulation of Ca2+ ions in the cell at a relatively low concentration (10m range) inhibits chloride exchange without apparent modification of the assembly of membrane proteins. This effect of calcium on chloride exchanges is speculatively denoted as a direct effect of calcium.Calcium loading of fresh red cells at higher concentrations (500 to 1000 m) obtained by use of the ionophore A23187 induces a very strong inhibition of chloride exchanges. In this case, inhibition can be reasonably accounted for by two simultaneous effects of calcium: a direct effect which explains half of the inhibition and an indirect effect due to the formation of membrane protein complexes stabilized by covalent crosslinkages (activation by Ca2+ ions of a transglutaminase).It is interesting to note that intracellular calcium, whatever the level, inhibits electroneutral exchanges of chloride but increases net chloride movements.  相似文献

Cultured human and rat endothelial cells were used to study cellular toxicity and Ca2+ signalling upon exposure to reactive oxygen species. Superoxide and hydrogen peroxide (O2·–/H2O2) were produced by the hypoxanthine/xanthine oxidase system (HX/XO) and caused intracellular Ca2+ concentration ([Ca2+]i) to rise steadily when activities above 2 mU/ml were used. These Ca2+ increases were also measured when the glucose/glucose oxidase (G/GO) system above 5 mU/ml was used to produce hydrogen peroxide (H2O2). Gross morphological changes appeared to parallel elevated [Ca2+]i levels preceding cell death. However, when HX/XO or G/GO were used at non toxic doses rapid and transient changes in [Ca2+]i were measured. These treatments did not alter subsequent receptor mediated Ca2+ signalling induced by ATP (10 M) or histamine (100 M). Superoxide dismutase (50 U/ml), which dismutates O2·minus; into H2O2 al ient [Ca2+]i responses. H2O2 added directly was able to induce similar Ca2+ transients when concentrations of at least 500 M were used. Buffering trace amounts of iron (o-phenanthroline; 200 M) in order to inhibit úOH radical formation was not effective to alter Ca2+ changes. Experiments performed in Ca2+-free buffer showed a similar rise in [Ca2+]i and readdition of Ca2+ to the extracellular medium indicated the activation of store operated Ca2+ entry. Blocking Ca2+-ATPases of the endoplasmatic reticulum with thapsigargin (1 M) inhibited ROS induced transient increases and cells preincubated with pertussis toxin (200 nM) showed unchanged Ca2+ transients after exposure to both enzyme systems. Phospholipase C inhibitor U73122 (2 M) effectively reduced hydrogen peroxide induced emptying of intracellular stores. Taken together, we demonstrate that enzymatically produced non-toxic H2O2 rather than O· ndash; or · OH causes calcium signalling from thapsigargin sensitive stores, and activates store operated Ca2+ entry at least partially by activating phospholipase C. These changes clearly differ from pathological oxidative stress associated with a progressive increase in [Ca2+]i.  相似文献

Summary Cytological changes in the pars intermedia of the goldfish were investigated after adding calcium to deionized water (DW). In fish maintained in DW, the PAS-positive cells are highly stimulated in comparison to cells of fish kept in fresh water (FW). In DW supplemented with calcium at the same concentration as in FW (2 mM/l), the hyperactivity of the PAS-positive cells is prevented. When calcium ions are added 60 h before the animals are sacrificed, the PAS positive cells start to show signs of regression and their granules are stored: the release of the granular material appears to be suppressed by calcium. In the goldfish, the PAS-positive cells, homologous to a similar cell type in the eel, react only very weakly with the PAS technique. The name calcium-sensitive cells appears to be more appropriate in the goldfish for this particular cell type, secreting an unknown factor. This factor, different from the prolactin produced in the rostral pars distalis of the hypophysis, might be an equivalent of a hypercalcin.  相似文献

Zusammenfassung Zweijährige Selektion auf Frostresistenz bei einer Kreuzung der argentinischen Tomatensorte Platense mit der nordamerikanischen Pearl Harbour unter Freilandbedingungen erwies sich bei Überprüfung in der Kühlkammer im Vergleich zu frostempfindlichen Kultursorten als ergebnislos. Es wird daher der Schluß gezogen, daß auch bei Tomaten eine erfolgreiche Züchtung auf Frostresistenz nur durch Einkreuzung mit frostresistenten Wildarten möglich ist, die auch vorhanden sind. Synopsis Arsenic persists in Chautauqua Lake, New York waters 13 years after cessation of herbicide (sodium arsenite) application and continues to cycle within the lake. Arsenic concentrations in lake water ranged from 22.4–114.81 g l–1, = 49.0 ag l–1. Well water samples generally contained less than 10 g l–1 arsenic. Arsenic concentrations in lake water exceeded U.S. Public Health Service recommended maximum concentrations (10 g l–1) and many samples exceeded the maximum permissible limit (50 g l–1). Fish accumulated arsenic from water but did not magnify it. Fish to water arsenic ratios ranged from 0.4–41.6. Black crappie (Pomoxis nigromaculatus) contained the highest arsenic concentrations (0.14–2.04 g g–1 ), X = 0.7 g g–1) while perch (Perca flavescens), muskellunge (Esox masquinongy) and largemouth bass (Micropterus salmoides) contained the lowest concentrations (0.02–0.13 g g–1). Arsenic concentrations in fish do not appear to pose a health hazard for human consumers.  相似文献

The specific uptake rate of -glucose by yeast (S. cerevisiae) and recombinant yeast is higher than that of -glucose. On the other hand, the specific uptake rate of -glucose byE.coli and recombinantE.coli is less than that of -glucose. These results imply that the unequal specific uptake rates of optical isomers of glucose should be included in modeling and optimizing high-cell density fermentations, fed batch fermentations, and immobilized cell systems, where cell density is maintained high and glucose concentration is low.  相似文献

Summary Hen coprodeum absorbs sodium electrogenically and, when stimulated by theophylline, secretes chloride. In this study the vibrating microprobe technique was used to localize the transport of these ions to intestinal villi/folds and crypts. With the isolated, stretched epithelium, controlled by light microscopy and scanning electron microscopy, in open circuit, currents were inward, 40±7 A/cm2, 50 m vertically above villi, and outward, 36±7 A/cm2 above crypts. The currents decayed exponentially to near zero at 300 m with the same length constant. A physical model simulating the observed loci of current sources and sinks predicts potential profiles consistent with our data. Extrapolation of the currents gives a surface potential of 45 V, negative on villi and positive above crypts. Short circuiting increased villus current to 86±27 A/cm2 at 50 m, and amiloride treatment reduced it to –8 A/cm2; in both cases crypt currents were abolished. The inward currents are compatible with sodium absorption. Induction of chloride secretion after amiloride treatment, resulted in current circuits similar to those induced by sodium absorption, with villus currents of 23±7 A/cm2. This is in accord with chloride secretion at the villi. Quantitative estimates of crypt number (860/cm2) and opening diameter (15 m), in conjunction with isotopic measurements of active and electrical potential-driven ion fluxes demonstrate, however, that only 4% of the potential-driven co-ion transport occurs through the crypts. This indicates that nearly all chloride secretion comes from the sodium-absorbing villar area. Were the chloride secretion to occur solely from the crypts, the current should have been in the opposite direction and 10,000-fold larger.  相似文献

Gills of typical yellow and silver ells, Anguilla anguilla L., were examined by light and electron microscopy. In both eel types, mitochondria-rich cells were located in the epithelium covering the primary lamellae and consisted ofchloride cells and accessory cells. As compared to yellow eels, the primary gill epithelium of silver eels was thicker and contained larger and more numerous chloride cells with enlarged mitochondria. The accessory cells also increased in number but did not show significant modifications in their size or ultrastructural features. These observations indicate that, as far as mitochondria-rich cells are concerned, the silvering process in eels would be equivalent to smoltification in salmonids. It corresponds to a preparation for seawater life and is probably controlled by hormonal factors.  相似文献

Summary From crosses of the mutant strains 402, 404 or 444xwild-type of Sphaerocarpos donnellii about 104000 tetrads with 4 sporelings, and about 20 000 tetrads with 3 sporelings were analyzed. No tetrad was found in which non-reciprocal segregation was induced by conversion. Therefore the frequency of conversion appears to be at least 10 times less in Sphaerocarpos than in ascomycetes. This conclusion is not contradicted by many tetrads (up to 30%), which segregated non-reciprocally from crosses of the mutant strain 429xwild-type, because these non-reciprocal segregations are completely in favor of the wild-type. In addition, plants of the phenotype wild originated in crosses of the mutant strain 429 with itself about twice as often as when crossed with the wild-type. Therefore, these findings cannot be accepted as a result of conversion in the sense of hybrid-DNA-models of crossing over. Summary Holthuisana transversa maintains sodium balance at very low external concentrations (7–25 M). This is achieved by the high affinity of the uptake mechanism (K m=0.18 mM), a low rate of sodium loss (0.17 mol g–1h–1) and a large increase in the rate of sodium uptake after depletion. The site of sodium absorption is the gills and extraction of sodium from respired water by sodium-depleted crabs can be as high as 28%. Crabs tolerate direct transfer to 80% sea-water and, following acclimation, the sodium and calcium levels of the haemolymph are maintained above those of the medium while the level of magnesium is significantly lower.  相似文献
