The Panlong river passes through Kunming City and receives a large quantity of municipal sewage and wastewater from industrial effluent. Along the river, 20 sites were selected to collect water samples to assess the genotoxicity using two Tradescantia assays, the micronucleus (Trad-MCN) and the stamen-hair-mutation (Trad-SHM) assays. The lowest frequencies in the Trad-MCN assay and the Trad-SHM assay are 3.19 MCN/100 tetrads and 1.32 M/1000 stamen hairs, respectively. In the water samples obtained from the Songhua Reservoir, the MCN frequencies and mutation rates are not statistically significantly different from the data found for the negative control (2.49 MCN/100 tetrads and 0.71 M/1000 stamen hairs). Among the other water samples, 19 in Trad-MCN assay and 17 in Trad-SHM assay show significantly higher genotoxicity than the control. The highest genotoxicity was in samples No. 19 for the MCN assay (8.73 MCN/100 cells), over three times higher than the negative control, and in sample No. 11 for the SHM assay (4.30 M/1000), six times higher than the negative control, and were about the same as for the positive control (10.0 mg/l NaN3, 9.28 MCN/100 tetrads and 7.44 SHM/1000 stamen hairs), respectively. The peak frequencies for the Trad-MCN assays were observed in the water samples obtained from the sites that were near industrial and municipal wastewater that ran into the river as effluent. In general, the frequency of MCN and SHM mutations increased where the river passed through Kunming. The Trad-MCN assay seemed more sensitive than that of the Trad-SHM assay in detecting genotoxicity of the river water pollution.  相似文献

Genotoxicity of sulfur dioxide (SO(2)) and its hydrates (bisulfite and sulfite) in human lymphocytes and other mammalian cells have been found earlier in our laboratory. In the present studies, we used Allium stavium and Vicia faba cytogenetic tests, which are the highly sensitive and simple plant bioassays. A mixture of sodium bisulfite and sodium sulfite (1:3), at various concentrations from 1 x 10(-4) to 2 x 10(-3)M was used for the treatment. Genotoxicity was expressed in terms of anaphase aberration (AA) frequencies in the Vicia-AA test and in terms of micronuclei (MCN) frequencies in both Vicia-MCN test and Alllium-MCN test. On average, the results showed a 1.7-3.9-fold increase of AA frequencies and a 3.5-4.5-fold increase of MCN frequencies in Vicia root tips as compared with the negative control. Similarly, results of Allium-MCN test also showed a significant increase in MCN frequencies in the treated samples. In addition, pycnotic cells (PNC) appeared in Allium root tips of treated groups. The frequencies of MCN, AA and PNC increased dose-dependently and the cell cycle delayed at the same time in bisulfite treated samples. Results of the present study suggest that the Vicia and Allium cytogenetic bioassays are efficient, simple and reproducible in genotoxicity studies of bisulfite.  相似文献

Natural lake systems represent important reservoirs for residential water supply, fish production, recreational activities and enjoyment of their natural beauty. Nevertheless, human impacts may affect their health status resulting in degradation and loss of biodiversity. The aim of the present study was to obtain data on the health status of a natural lake located in an indigenous reservation in the Brazilian Amazon, using the phytoplankton community changes along the rainy (June) and dry (November) seasons of 2006. We collected water (temperature, pH, Secchi depth and conductivity) and phytoplankton samples from the subsurface, middle of the water column, and approximately 30 cm above the bottom, over 24-hour sampling periods, from a central station in the lake. Samples taken from biotic and abiotic variables were correlated using canonical correspondence analysis (CCA). Results showed that the lake exhibited high temperatures in both seasons, and showed thermal stratification only during the rainy season. Dissolved oxygen exhibited a clinograde pattern in the rainy season and high oxygen in the hypolimnion in the dry season. In the rainy season, the water near the bottom was acidic, turbid and had a greater concentration of phosphorus. Dissolved oxygen, conductivity, pH, nitrite, total phosphorus and total dissolved phosphorus exhibited diel variations in the rainy season, whereas water temperature, dissolved oxygen, total nitrogen and total dissolved phosphorus exhibited significant differences between hours of the day in the dry season. The phytoplankton was represented by 39 taxa, and Chlorophyta showed the greatest species richness, totaling 25 taxa. Among Chlorophyta, desmids were the most diverse, accounting 52%. Bacillariophyta (nine species) was the second most diverse group. Cyanophyta was represented by three species, including Merismopedia tenuissima, the most abundant taxon. Despite the occurrence of taxa that indicate organic pollution, their biomass and frequency indicate that the system is not currently threatened. Lake Caracaran? is an oligotrophic system, with low algal density and isolated blooming episodes due to its shallow depth.  相似文献

Mitotic root micronucleus (MCN) frequency in Vicia faba as a bioassay, is primarily based on the extent of the sentinel Vicia response in terms of cytogenetic damage quantitatively or qualitatively to indicate the presence of mutagenic contaminants. This paper describes an investigation designed to assess changes in MCN frequencies of Vicia faba from three generation plants obtained, respectively from a reference site (RS) and a metal-contaminated experimental field (EF) in the bioassay of mutagenic Cd(2+) and NaN(3). The background value, dose-response to Cd(2+) and to NaN(3) in three generation (F(1), F(2) and F(3)) plants of the EF and the initial (F(0)) plants were determined in terms of MCN frequencies. With more generations of growing Vicia plants in the EF, a higher background value of MCN frequency, a lower slope value in the regression equation, a smaller ratio of MCN frequency between the control and treatment in the same generation and larger perturbation values were observed. This denotes that the decreased reliability and efficiency are represented in Vicia plants from the EF if the plants are used as sentinels in the bioassay of mutagenic Cd(2+) and NaN(3). It was concluded that the Vicia MCN bioassay should be used as an endpoint biomarker acceptable in biomonitoring environmental mutagens when the sentinel plants were collected from clean areas. Because no place is absolutely without pollutants, it is suggested that several seed stock centers should be established for providing sentinel Vicia if Vicia MCN bioassay is used as a biomarker to identify the global environmental status.  相似文献

目的 利用EFM技术对纳帕海湿地不同季节7个水样样品中的浮游病毒丰度开展了调查,并对影响浮游病毒丰度的环境因子进行了相关性分析。方法 元素检测,叶绿素a检测,荧光显微镜检测浮游细菌及浮游病毒的丰度。结果 从生态分布上来看旱季和雨季浮游病毒丰度的平均值分别为3.63×106/mL和3.71×107/mL,浮游病毒丰度季节性变化不显著。从对影响浮游病毒丰度的环境因子相关性分析来看,旱季浮游病毒与Chl-a具有弱负相关性(r=0.49,P<0.05);而雨季浮游病毒丰度与Chl-a具有显著正相关性(r=0.37,P<0.05),表明雨季纳帕海湿地浮游病毒丰度受Chl-a含量的影响较大。旱季浮游病毒丰度与温度显著负相关,虽然在雨季浮游病毒丰度与温度也呈负相关,但并不显著;在雨季,水体pH变化较明显,而浮游病毒丰度与水体pH呈显著负相关,表明湿地的来水主要在雨季,而离子组成变化大。虽然在雨季和旱季,从多点采样的平均值来看浮游病毒丰度并无明显差异,但从一些局部的采样点来看,雨季的浮游病毒丰度显著高于旱季,说明在纳帕海区域浮游病毒的分布并不平均,因采样点环境的不同会产生明显的区别。结论 对纳帕海高原湿地浮游病毒生态分布的调查,是对该地域生态学研究的补充,使得这一独特地理环境的微生物研究更加系统化。  相似文献

The Vicia root-micronucleus (VR-MCN) bioassay was used to determine the genotoxicity of water samples collected from the Kui River near Xuzhou city, China. Results show that all water samples induced elevated MCN frequencies over background. This indicates that the river water was polluted with genotoxic material. Teressa goat is a unique goat breed in Andaman and Nicobar Islands (ANI) of India. Effects of Flaxseed oil (FSO) supplementation in body weight (BW), scrotal circumference (SC), testicular volume (TV) and testicular weight (TW), endocrinological profiles, sex behavioural profiles (SBPs), oxidative stress markers and semen production and its quality profiles in rainy and dry summer season were studied in Teressa goat. Male goats (n = 12) of 3–4 years old were equally divided into control and treated groups. Treated animals received 25 mL FSO per day. Oral drenching of FSO was done in the morning before feeding the concentrate ration. Body weight, scrotal circumference, TV and TW were measured in bucks of FSO treated and untreated during rainy and dry summer seasons. Blood follicle stimulating hormone (FSH), luteinizing hormone (LH), testosterone, thyroid stimulating hormone (TSH), triiodothyronine (T3), thyroxine (T4), cortisol and prolactin, total antioxidant capacity (TAC), superoxide dismutase (SOD), catalase (CAT), glutathione (GSH) and malondialdehyde (MDA) were measured in bucks of FSO treated and untreated during rainy and dry summer seasons. Libido score (LS), mating ability score (MAS) and sex behavioural score (SBS) were estimated at time of semen collection in bucks of FSO treated and untreated during rainy and dry summer seasons. Semen samples (n = 100; 50 semen samples from each season; each 25 semen samples from control and treatment groups per season) were collected and analysed for semen quality profiles. One-way ANOVA (control rainy, control dry, treated rainy and treated dry) revealed that BW, SC, TV and TW, FSH, LH, testosterone, TSH, T3 and T4 were higher (P < 0.05) and cortisol and prolactin were lower (P < 0.05) in FSO treated bucks of rainy season followed by untreated bucks of rainy season, FSO treated bucks of dry summer season and were lower (P < 0.05) in untreated bucks of dry summer season. Similarly, TAC, CAT, SOD and GSH, LS, MAS and SBS, and volume, pH, sperm concentration, mass activity, total motility (TM), viability, acrosomal integrity (AcI), plasma membrane integrity (PMI) and nuclear integrity (NI) were higher (P < 0.05) and MDA and TSA were lower (P < 0.05) in FSO treated bucks of rainy season followed by FSO treated bucks of dry summer season, untreated bucks of rainy season and were lower (P < 0.05) in untreated bucks of dry summer season. The results of the present study indicated that the breeding bucks suffered physiological stress (higher cortisol), oxidative stress (higher MDA and deficiency of antioxidants), hormonal imbalance (higher prolactin and cortisol and deficiency of gonadotropins, gonadal hormone and thyroid hormones) and infertility due to poor libido and poor semen production and its quality profiles during dry summer season. Thus, dry summer was more stressful season compared to rainy season for the goat bucks. FSO supplementation mitigated these stresses and improved the scrotal and testicular biometrics, libido, antioxidants, hormones and semen quality profiles in Teressa goat bucks. The current study concluded that FSO effectively improved the hormones, libido, antioxidant profiles, and scrotal and testicular biometrics with cascading beneficial effects on semen quality profiles in Teressa goat bucks under humid tropical island ecosystem of Andaman and Nicobar Islands.  相似文献

Drosophila ananassae, a desiccation and cold sensitive species, is abundant along the latitudinal gradient of the Indian subcontinent. Analysis of seasonally varying wild-caught flies showed two independent patterns of melanisation: (1) narrow to broad melanic stripes on three anterior abdominal segments only; (2) a novel body color pattern (dark vs. light background). We investigated the degree to which these two melanisation systems vary; first with latitude and secondly among seasons. There is a shallow latitudinal cline for percent striped melanisation as well as for frequency of body color alleles during the rainy season. The frequencies of body color alleles vary significantly across seasons in the northern populations i.e. the light allele occur abundantly (>0.94) during the rainy season while the frequency of the dark allele increases (0.22–0.35) during the dry season causing steeper clines during the dry season. By contrast, the low variations in abdominal stripes showed non-significant changes and the cline was similar across seasons. Furthermore, both types of melanisation patterns showed no plasticity with respect to temperature. The present study also investigated clines related to desiccation, heat and cold stress in D. ananassae females across seasons (rainy and dry) from nine latitudinal populations. The clines for stress related traits changes to steeper and non-linear during the dry season. Thus, latitudinal populations of D. ananassae differ in slope values of clines for stress related traits across seasons. This study reports seasonal changes in latitudinal clines of stress resistance traits as seen in a changing frequency of body color alleles of D. ananassae in northern locality, while in southern localities it remains constant. This is presumably the result of only minor seasonal changes in humidity and temperature in the South, whereas in the North seasonal climatic variability is much higher.  相似文献

Seasonal variation of the physicochemical conditions of streams plays an important role in aquatic insect diversity and community structure. Asian tropical streams profoundly change between seasons due to the effects of monsoons. However, little is known about how these changes affect aquatic insect diversity and community structure. The objectives of this study are to examine seasonal variations of the physicochemical conditions in tropical streams in Thailand and to assess the effects of these changes on black fly community structure and diversity. Black flies were sampled and physicochemical conditions recorded at eight sites between December 2007 and December 2008. A total of 10 black fly species were found. Comparisons of the streams between seasons revealed that physical conditions related to rainfall rate were significantly different. Canonical correspondence analysis differentiated sampling sites from each season. Streams in the rainy season were faster and deeper, with higher discharge and conductivity than those of the cold and dry seasons. Species richness was significantly higher in the rainy season than in the cold and dry seasons ( F  = 6.23, P  = 0.004). Community structure profoundly changed between the low-flow season (cold and dry) and high-flow season. Black fly species found predominantly in the low-flow season ( Simulium siamense "cytoform A", S. aureohirtum ) decreased dramatically during the high-flow season. In contrast, species found at high frequency during the high-flow season ( S. nakhonense , S. angulistylum ) disappeared in the low-flow season. The study demonstrates the importance of seasonal variation of stream conditions on black fly community structure and diversity.  相似文献

During the dry season, the Nyanza Gulf was strongly dominated by blue‐greens, while diatoms dominated in the open lake. During the rainy season, blue‐greens remained dominant in the Nyanza Gulf although the number of species found was lower than during the dry season; in the open lake, blue‐greens replaced diatoms as the dominant group and there were more species than in the dry season.
  • 4 Canonical correspondence analysis indicated that the phytoplankton species distribution was significantly correlated with turbidity during the dry season and with SiO2 during the rainy season. Chlorophyll a concentrations ranging from 2.0 to 71.5 mg m‐3 in the dry season and 2.0–17.2 mg m‐3 in the rainy season confirm earlier reports of increasing phytoplankton biomass in Lake Victoria since the 1960s.

Here we assessed the efficacy of performing field surveys during both dry and rainy seasons to estimate species richness and composition of phyllostomid bats in 12 sites of the Brazilian Atlantic Forest. Our goal was to elucidate two opposite scenarios. If bat species compositions differ between the dry and rainy seasons, then field surveys in both seasons will be considered complementary for estimating local bat assemblages. In contrast, if bat species compositions in the dry season are a nested subset of the species present in the rainy season, then field surveys in both seasons will be considered redundant. Our results showed that field surveys performed only during the rainy season exhibited the same efficacy in recording bat species as field surveys considering both dry and rainy seasons combined. Consequently, the rare species sampled in the rainy season contributed to the uniqueness of communities in terms of species composition. Considering that time, funding, and the number of field personnel available for work are important factors when determining the sampling effort in highly biodiverse regions like the tropics, our results can be useful in establishing protocols for research projects with limited resources aiming to create a comprehensive list of species.  相似文献

Spring Lake reservoir of Macomb, Illinois, is a typical model of the drinking water supply of some midwestern towns of the United States. Water samples collected periodically in 1980 and 1981 from this lake were tested for mutagenicity using the Tradescantia micronucleus (Trad-MCN) test, a highly sensitive mutagen-detecting bioassay. Water samples from 1981 were also analyzed chemically. The micronucleus (MCN) frequency peaked (12-14 MCN/100 tetrads) in mid-July in both years, as compared with the average frequency (5 MCN/100 tetrads) of the base-line control that was maintained in nutrient solution (prepared with distilled water and pure chemicals). Drinking water from the tap was tested in parallel with lake water, and its mutagenicity tended to fluctuate with the mutagenicity of the lake water.  相似文献

This study assessed the levels of damage by leaf pathogens and their variability in terms of host species, space (four mature forest sites) and season of the year (dry and rainy), and the mechanisms of infection in the understory of the Los Tuxtlas tropical rain forest. Sixty-five percent of the species surveyed in the dry season (N = 49) and 64.9% of those surveyed in the rainy season (N = 57) were damaged by fungi. Leaf area damaged per plant, on average, was <1% (range: 0.25-20.52%). There was considerable variation in the degree of infection among species, but not among sites and seasons. The survey showed that 43% of the leaves were damaged by herbivores and pathogens concurrently, 16% showed damage by insect herbivory alone, and only 1.4% of the sampled leaves showed damage by pathogens alone. Pathogenicity assays experimentally confirmed that the predominant mechanism of fungal establishment was wounding, such as that caused by herbivory (or other similar sources), and only rarely did infection occur through direct contact (without wounds). The results revealed the omnipresence of leaf fungal infection, although with low damage per plant, and the importance of herbivorous insects in the facilitation of fungal infection in tropical understory plants.  相似文献

The diet of the brown four-eyed opossum (Metachirus nudicaudatus) at a riparian savanna in Brazil was analyzed using 22 fecal samples collected monthly from November 2009 to October 2011. A comparison was established between samples from rainy and dry seasons. Diet composition varied seasonally. Arthropods and fruits were consumed in different frequencies of occurrence, in both seasons, but small vertebrates were only found in the dry season. Diet differences among biomes (Atlantic Forest and Cerrado) were also assessed by comparing the frequency of food items in the fecal sample in our study with data from the literature. Invertebrates (100%) were the most frequent food category found, followed by fruits (59%) and vertebrates (13.6%). Small differences in the consumption of invertebrates were found among the biomes. Nevertheless, seed frequency was significantly higher in samples from the Cerrado than in those from the Atlantic Forest. In our study site, Metachirus nudicaudatus is opportunistic and has an omnivorous diet, tending to include more fruits in its natural diet than has been previously described.  相似文献

The freshwater microbial community in a recreational area of Xochimilco, México was investigated and compared based on spatial (three different sites) and temporal (dry and rainy seasons) environmental variables. Many of the 16S- and 18S rRNA gene sequences recovered by DGGE fingerprinting analysis were related to phototrophic microbial phylotypes of known identity. Our genetic and morphological analysis indicated the ubiquitous presence of the microeukaryotic green algae Desmodesmus-Scenedesmus spp. and of the unicellular cyanobacteria Cyanobium spp. as the most representative populations in the samples. While 18S rRNA-DGGE fingerprinting analysis revealed a homogeneous community composition across sites and seasons, the 16S rRNA showed significant differences between localities and seasons. None of the cyanobacteria species with potential to produce toxins were identified across the investigated samples. Correlations between biotic and abiotic variables evidenced an important difference between the dry and the rainy season, with a greater consistency in data from the rainy season. According to Principal Component Analysis (PCA), a strong relation between inorganic nitrogen, species richness, and subaquatic irradiance determines environmental variability in Xochimilco. Complementary and relevant data in results obtained from microscopy, fingerprinting, and statistical analysis applied in ecology indicate that a multifaceted approach to the study of microbial communities is necessary to accomplish a comprehensive scientific framework and to generate proper management strategies.  相似文献

The aim of this study was to analyze the viable fungi in the air of indoor environments of the National Archive of the Republic of Cuba (NARC). The samples were taken in fifteen areas (five points for the three floors) at 2 days (rainy and dry seasons) using a biocolector SAS in 3 h: 10:00 a.m., 1:00 p.m. and 4:00 p.m. The fungal concentration was indicated as colony forming units per cubic meter of air (cfu/m3 of air). It was concluded that indoor air of the NARC was not polluted due to the average fungal concentration that was obtained (59 and 327 cfu/m3 of air in rainy and dry seasons, respectively). In front of the electric transformers, the fungal concentration was significantly higher than those obtained along the corridors of the lower-ground floor. A trend to the increasing of fungal concentration was observed: when the floors were increased, in dry season was higher than rainy season, where also there was biggest fungal diversity. Among the fungal genera isolated, Aspergillus had the highest relative frequency of appearance, and in dry season, it was isolated Actinobacillus sp. (Actinomycetes). Six fungal species were isolated in both samplings: Aspergillus alliaceus, Aspergillus niger, Cladosporium cladosporioides, Fusarium oxysporum, Penicillium chrysogenum and Mucor racemosus. All of fungal species isolated have high biodeteriogenic capacity, and the 61 % showed pathogenic attributes.  相似文献

Faecal samples (n?=?1947) from captive wild impala (Aepyceros melampus melampus) were examined over a period of 14 months to determine quantitative seasonal helminth egg excretion patterns and qualitative protozoan oocyst excretion patterns. Geometric mean monthly faecal egg counts (FECs) ranged from 20 to 575 and coprocultures revealed three parasite genera, namely Trichostrongylus, Haemonchus and Strongyloides. Larvae of the Trichostrongylus spp. were most predominant from faecal cultures. No trematode eggs or lungworms were detected and eggs of the cestode Monezia were only seen in two samples during the entire study period. The nematode FECs showed a marked seasonal variation, being higher during the rainy season, moderate during the cool dry season and low during the hot dry season. The influence of season on the ejaculate characteristics of Zebu, Friesian and their crossbred bulls in an A.I. programme in Nigeria was investigated over a 2-year period.Ejaculate volume, sperm concentration, percent morphologically normal spermatozoa and percent live spermatozoa were significantly higher in the rainy season than in the dry season. Total spermatozoa per ejaculate averaged 3.32 × 109 and 10.10 × 109 for the dry and rainy seasons respectively. Corresponding proportions of total morphologically defective spermatozoa per ejaculate were 14.05% and 6.46%. Percent live spermatozoa were 82.34% and 84.61% while the corresponding sperm concentrations were 0.97 × 109 rmand 1.74 × 109 for the dry and rainy seasons respectively. All differences were statistically significant (P < 0.05).Ejaculate quality was better during the rainy season. Consequently semen collected and frozen during the rainy season may produce higher fertility rates in an A.I. programme.  相似文献

In the present study, we applied Chromosome Aberration (CA) and Micronucleus (MN) tests to Allium cepa root cells, in order to evaluate the water quality of Guaecá river. This river, located in the city of S?o Sebasti?o, SP, Brazil, had been affected by an oil pipeline leak. Chemical analyses of Total Petroleum Hydrocarbons (TPHs) and Polycyclic Aromatic Hydrocarbons (PAHs) were also carried out in water samples, collected in July 2005 (dry season) and February 2006 (rainy season) in 4 different river sites. The largest CA and MN incidence in the meristematic cells of A. cepa was observed after exposure to water sample collected during the dry season, at the spring of the river, where the oil leak has arisen. The F(1) cells from roots exposed to such sample (non-merismatic region) were also analyzed for the incidence of MN, showing a larger frequency of irregularities, indicating a possible development of CA into MN. Lastly, our study reveals a direct correlation between water chemical analyses (contamination by TPHs and PAHs) and both genotoxic and mutagenic effects observed in exposed A. cepa cells.  相似文献

Resource availability can influence animal movement causing changes in home‐range size and arrangement between seasons. We investigated the influence of acuri palm (Attalea phalerata) on the occupancy of agouti Dasyprocta azare during both the dry and rainy seasons, as well as the abandonment probabilities in the transition between seasons in the Brazilian Pantanal. The agoutis occupied a high proportion of the forested areas in the rainy season (0.83), but the occupancy decreased in the dry season (0.39). In the rainy season, occupancy by agoutis was not correlated with acuri palm availability, while in the dry season, it was positively correlated with the palm's availability. The acuri palm availability also drove the agoutis' probability of site abandonment from the rainy to the dry season, with higher abandonment probability in sites with low acuri palm availability. These findings show that this large‐seeded palm can be an important resource for the agoutis' populations during the fruit‐scarcity period. The acuri palm seeds may be particularly important for agoutis using fragmented forests, given that the large‐seeded palms are able to persist even in small fragments, which is untrue for other important resource.  相似文献
