Gross structure and dimensions of the gills have been examined in a hill-stream sisorid catfish,Glyptothorax pectinopterus, which remains adhered to rocks by means of an adhesive organ developed on the ventral side of the thorax. The fish shows a greater weight-specific gill area and greater length of the gill filaments by comparison with other hill-stream fishes. Adaptation for life in a hill-stream habitat is shown by the presence of additional filaments on the gills and patches of specialised cells on the filament epithelium.  相似文献

The phloem is a well-known target of sucking and piercing insects that utilize the transported fluid as their major nutrient source. In addition to small molecules like sugars and amino acids, phloem sap of higher land plants contains proteins that can accumulate up to high concentrations. Although the knowledge about the identities of these phloem sap proteins is increasing, the functions of most of them are still poorly understood. Since many phloem sap proteins have predicted roles in wound and defence responses, they constitute a class of compounds that can potentially influence plant-insect interactions. However, there are as yet no studies published that have examined direct effects of phloem sap proteins on insect feeding or vice versa. This review summarizes the current knowledge about the identities of phloem sap proteins, focused on polypeptides with probable functions in wound and defence reactions, and their potential impact on plant-insect interactions is discussed.  相似文献

Pure phloem sap was collected from insects feeding on rice (Oryza sativa L.) leaves by a laser technique similar to the aphid stylet technique. Rapid circulation of nitrogen in the sieve tubes was demonstrated directly using 15N as a tracer. Application to the roots of the metabolic inhibitors of amino acids, aminooxyacetate and methioninesulfoximine, changed the amino acid composition in the sieve tubes. Feeding methionine to leaf tips resulted in its bulk transfer into the sieve tubes. In vitro experiments confirmed the existence of protein kinases in the pure rice phloem sap. The phosphorylation status of the sieve tube sap proteins was affected by the light regime. The possibility that changes in chemical composition or protein modification such as phosphorylation in the sieve tubes might affect plant growth are discussed.Analysis of pure phloem sap collected from rice plants by insect laser technique has shown dynamic changes in the chemical composition and the quality of proteins in the sap.  相似文献

Lipids have been observed attached to lumen-facing surfaces of mature xylem conduits of several plant species, but there has been little research on their functions or effects on water transport, and only one lipidomic study of the xylem apoplast. Therefore, we conducted lipidomic analyses of xylem sap from woody stems of seven plants representing six major angiosperm clades, including basal magnoliids, monocots and eudicots, to characterize and quantify phospholipids, galactolipids and sulfolipids in sap using mass spectrometry. Locations of lipids in vessels of Laurus nobilis were imaged using transmission electron microscopy and confocal microscopy. Xylem sap contained the galactolipids di- and monogalactosyldiacylglycerol, as well as all common plant phospholipids, but only traces of sulfolipids, with total lipid concentrations in extracted sap ranging from 0.18 to 0.63 nmol ml−1 across all seven species. Contamination of extracted sap from lipids in cut living cells was found to be negligible. Lipid composition of sap was compared with wood in two species and was largely similar, suggesting that sap lipids, including galactolipids, originate from cell content of living vessels. Seasonal changes in lipid composition of sap were observed for one species. Lipid layers coated all lumen-facing vessel surfaces of L. nobilis, and lipids were highly concentrated in inter-vessel pits. The findings suggest that apoplastic, amphiphilic xylem lipids are a universal feature of angiosperms. The findings require a reinterpretation of the cohesion-tension theory of water transport to account for the effects of apoplastic lipids on dynamic surface tension and hydraulic conductance in xylem.  相似文献

Norway spruce is a conifer storing large amounts of terpenoids in resin ducts of various tissues. Parts of the terpenoids stored in needles can be emitted together with de novo synthesized terpenoids. Since previous studies provided hints on xylem transported terpenoids as a third emission source, we tested if terpenoids are transported in xylem sap of Norway spruce. We further aimed at understanding if they might contribute to terpenoid emission from needles. We determined terpenoid content and composition in xylem sap, needles, bark, wood and roots of field grown trees, as well as terpenoid emissions from needles. We found considerable amounts of terpenoids—mainly oxygenated compounds—in xylem sap. The terpenoid concentration in xylem sap was relatively low compared with the content in other tissues, where terpenoids are stored in resin ducts. Importantly, the terpenoid composition in the xylem sap greatly differed from the composition in wood, bark or roots, suggesting that an internal transport of terpenoids takes place at the sites of xylem loading. Four terpenoids were identified in xylem sap and emissions, but not within needle tissue, suggesting that these compounds are likely derived from xylem sap. Our work gives hints that plant internal transport of terpenoids exists within conifers; studies on their functions should be a focus of future research.  相似文献

Abstract. This paper describes studies on trees of Pisonia grandis , bushes of Argusia argentea , and the perennial herb Melanthera biflora , growing on One Tree Island, a coral cay of the Great Barrier Reef with 'soil' of coarse coral rubble. Water potential (Ψb, measured on small shoots with a pressure chamber), sap flow, stomatal conductance, vapour pressure deficit and photon flux density were monitored over day/night cycles. Sap flow and Ψb responded to changes in light and humidity. From these experiments good linear correlations were found between sap flow in a shoot and Ψb of similar adjacent shoots. The linearity suggests that the resistance to sap flow is constant as Ψb varies. The correlation, however, does not indicate a causal relationship between Ψb of an individual shoot on the plant and its sap flow. Ψb was only slightly different in shaded shoots from those in sunshine, although sap flow would be expected to differ between them. Enclosing shoots and so reducing their transpiration and sap flow to very low rates resulted in only small changes in Ψb of the enclosed shoots; Tb of such enclosed shoots should closely approximate that of the xylem at the point of shoot attachment. From these results it is suggested that the resistance to water flow in shoot and leaf xylem is small compared to the resistance further down the plant, in the root or at the root/soil interface. Shoot xylem water potential would be similar for all parts of the plant, and in such plants the water potential of shoots in the shade would be determined by the overall water use of the plant.  相似文献

Diurnal courses of nutrient transport in the xylem and their response to external availability of nutrients were studied. In soil culture, maximal concentrations in all analysed substances were observed during night‐time. Over experimental periods of up to 20 d, concentrations of some ions increased, most by accumulation in the soil. Stringent nutrient conditions were established in a novel pressure chamber. An aeroponic nutrient delivery system inside allows the sampling of xylem sap from intact plants under full control of the nutrient conditions at the root. Analysis of xylem transport under these highly defined conditions established that (1) diurnal variations in concentrations and fluxes in the xylem are dominated by plant‐internal processes; (2) concentrations of nutrients in the xylem sap are highly but specifically correlated with each other; (3) nitrate uptake and nitrate flux to the shoot are largely uncoupled; and (4) in continuous light, diurnal variations of xylem sap concentrations vanish. Step changes in nitrate concentrations of the nutrient solution established that (5) the concomitant increase in nitrate concentration and flux in the xylem is delayed by 2–3 h and is only transient. Diurnal variations of xylem sap composition and use of the new technique to elucidate xylem‐transport mechanisms are discussed.  相似文献

How nutritionally imbalanced is phloem sap for aphids?   总被引:8,自引:0,他引:8  
Aphids harbour intracellular symbionts (Buchnera) that provide their host with amino acids present in low amounts in their diet, phloem sap. To find out the extent to which aphids depend on their symbionts for synthesis of individual essential amino acids, we have evaluated how well phloem sap amino acid composition matches the aphids' needs. Amino acid compositions of the ingested phloem sap were compared to amino acids in aphid body proteins and also to available information about optimal diet composition for other plant feeding insects. Phloem sap data from severed stylets of two aphid species, Rhopalosiphum padi (L.) (Homoptera: Aphididae) feeding on wheat, and Uroleucon sonchi (L.) (Homoptera: Aphididae) feeding on Sonchus oleraceus (L.), together with published information on phloem sap compositions from other plant species were used.Phloem sap has in general only around 20% essential amino acids, with a range from 15–48%. Aphid body proteins and optimal diets for two other plant feeding insects have around 50%. The phloem sap of early flowering S. oleraceus ingested by U. sonchi contained 48%, which seems to be exceptional. Aphids feeding on different plants appear to be very differently dependent on their symbionts for their overall essential amino acid synthesis, due to the large variation in proportion of essential amino acids in phloem sap from different plants.The profile of the essential amino acids in phloem sap from different plant species corresponds rather well to profiles of both aphid body proteins and optimal diets determined for plant feeding insects. However, methionine and leucine in phloem sap are in general low in these comparisons, suggesting a higher dependence on the symbiont for synthesis of these amino acids. Concentrations of several essential amino acids in phloem from different plant species seem to vary together, suggesting that levels of symbiont provisioning of different amino acids are adjusted in parallel.  相似文献

All trematodes which cause infections and diseases in humans (zoonoses) require snails for their first intermediate host. Some have additional intermediate hosts such as crustaceans, fishes and frogs. In this paper I have discussed the use of various procedures for controlling snail populations thereby reducing the population of trematodes whose cercariae penetrate the skin of man, or which are acquired by eating raw aquatic vegetation such as watercress, or which cause pathology in fish. Biological, chemical, and physical snail control techniques are discussed. The most promising molluscucides are copper compounds, Bayluscide, biocidal rubber and molluscicidal bait.  相似文献

This investigation shows diurnal variations in the xylem sap composition of poplar (Populus tremula x P. alba). All major macronutrients reached a maximum concentration in the first half of the light period and decreased to the middle of the night. The relative abundance of the nutrients did not change during the day. The sap flow, which responded very fast to the environmental changes (2.2-fold increase within 10-20 min of illumination), reached a maximum value in the second half of the light period. Transpiration (and photosynthesis) was constant throughout the light phase. The calculated translocation rates displayed a maximum in the first half of the light period and therefore did not fit the time course of sap flow. During the night, translocation rates were 63-69% lower than the maximum. The regulation of nutrient translocation is discussed taking the active xylem loading into account. The axial distribution located the nitrate assimilation in younger leaves and storage of nitrate (and other macronutrients) in older leaves. Nitrate and potassium concentrations in the xylem sap did not change along the plant axis. However, the sap flow was greater in younger shoot sections than in older sections. We assume that the greater demand for nitrate in the younger shoot section was satisfied via an increased volume flow rather an increased nitrate concentration.  相似文献

Cytokinins from the roots may be involved in regulating rose ( Rosa hybrida ) shoot growth and development. The objective of this study was to estimate the export of cytokinins from the roots and their degradation rate in the shoot, which were expected to be correlated with plant development. Hence, the total cytokinin content of the shoot, the concentration of zeatin riboside (ZR) in bleeding sap, and the transpiration rates in three stages of development were determined. The estimations performed are based on the assumption that the cytokinin concentration in bleeding sap is representative for the cytokinin concentration in xylem sap in situ. This was verified by comparing the ZR concentration in bleeding sap and in sap obtaíned after pressurizing the root system to a level equivalent to the leaf water potential; no significant differences could be found. The import of cytokinins could not be correlated with plant development, as it increased linearly with time. The estimated relative degradation rate of cytokinins in the shoot decreased as the plants matured. The half-life of cytokinins in the shoot was found to be approximately 1 day, indicating that cytokinins are rapidly metabolized in the shoot.  相似文献

There are 1...2 corpuscles of Stannius in the Indian hill-stream teleost, Pseudecheneis sulcatus. They are encapsulated bodies, measuring about 400 micrometer in length and 200 micrometer in width. The secretory cells of CS have fine granules in their cytoplasm which are AF, PTAH, CAH and silverpositive. The cells are arranged along blood sinusoids in which their secretions are probably discharged. Individual neurons and other neural structures are found scattered among the secretory cells.  相似文献

本研究建立和改进了检测番木瓜和西葫芦组织粗汁液里的番木瓜环斑病毒(PRV)的DAC-ELISA法和Dot-ELISA法。用不同的ELISA方法来检测不同寄主植物粗汁液里的PRV,其所用的合适的制备粗汁液的缓冲液是不同的。用DAC-ELISA法检测西葫芦粗汁液时,以0.5mol/L磷酸盐缓冲液(pH7.5,内含0.1mol/L乙二胺四乙酸二钠)为宜;而检测番木瓜粗汁液时,则还要加入0.25mol/L脲。用Dot-ELISA法检测时,在上述磷酸盐缓冲液中加入2%聚乙烯吡咯烷铜能提高对西葫芦粗汁液的检测效果。应用合适的制备粗汁液的缓冲液,DAC-ELISA法和Dot-ELISA法的灵敏度分别提高到1/4096和1/1024(稀释度)。本研究还表明,影响DAC-ELISA法的定过测定的主要因素是粗汁液的稀释度和包被液(0.05mol/L碳酸盐缓冲液,pH9.6)的用过。在较高粗汁液稀释度和包被液的用量相同时,粗汁液里的病毒含量与DAC-ELISA法的OD492nm值呈真实的线性关系。  相似文献

The effects of 2,4-dinitrophenol, of changes in the temperatureand concentration of the ambient solution and of variationsin salt status on the electrical potential difference betweenthe xylem exudate of maize roots and the ambient solution havebeen examined. The results are discussed in the light of someof the factors which could give rise to a potential differencebetween the sap and the solution. The rapid response of thepotential difference to dinitrophenol and to changes in temperaturesuggest that, at least in part, it arises directly from metabolicprocesses. Rapid changes in the potential difference broughtabout by addition of salts may be attributed to differentialrates of movement of anions and cations in the initial uptakeprocess. Over longer periods the potential difference appearsto be dependent on the concentration, but not the compositionof the ambient solution, and on the salt status of the roots.The salt status influences the relative rates at which anionsand cations are transported to the xylem sap, and a correlationhas been found between the potential difference and the ratioof the rates of movement of chloride or sulphate to potassiumto the sap. The implications of these findings on the elucidationof the pathways whereby ions are transported to the sap arediscussed.  相似文献

The data available in the literature concerning fish thrombocytes, lymphocytes, monocytes, macrophages and granulocytes is discussed and the dearth of reliable information concerning the functional identity of most fish leucocytes is underlined. The methods of applying nomenclatures to fish leucocytes encountered in the literature is discussed in some detail and found to be unsatisfactory. An attempt is made to simplify the confused and conflicting reports by applying the concepts which have recently arisen in the field of mammalian immunology. Nomenclatures are assigned to cells on a functional and morphological basis where sufficient data exists.
Thrombocytes are responsible for clot formation and are considered to be distinct, separate and unrelated to lymphocytes. Lymphocytes are recognised as immuno-competent cells though as yet little information exists on their role in immune mechanisms. Recent work is furnishing interesting results in this direction, especially in the origin of lymphocyte populations and their co-operative responses in immunity. Previous references to monocytes and macrophages in fish are confused and the literature is discussed in the light of the new classification of the Mononuclear Phagocyte System.
Very few experimental data exist concerning granulocytic leucocytes in fish, indeed the presence of such cells with granulocytic functions still requires proof. Cells with morphological similarity to mammalian granulocytes do exist in fish but information essential for an understanding of their role in defence mechanisms is lacking. The state of the literature concerning the eosinophil, basophil and mast cell in fish is so confused that an entirely new approach to a study of these cells is warranted. It is hoped that this review will supply guidelines for future research.  相似文献

The effect of different light intensities on the nitrate accumulationvis-à-vis the concentration of other solutes in plantsap expressed from lettuce leaves was studied. After growinglettuce plants under constant environmental conditions for 52d, they were transferred to different light intensities andharvested periodically. A quantitative analysis of componentsin solution in the expressed plant sap showed a decrease innitrate concentration and an increase in the organic acids (mainlymalate) and sugars (mainly glucose) with increasing light intensity.The light intensity only slightly increased the osmolarity ofthe expressed plant sap. The measured osmolarity correspondedvery well with the value estimated from the quantitative analysesimplying that all osmotically active compounds had been accountedfor. The decrease in nitrate concentration in the expressedplant sap was fully compensated for by an increase in the dissociatedorganic acids that partly dissociate twofold to sustain electroneutralityand by an increase in both organic acids and sugars to maintainthe osmolarity. The suggestion is supported that nitrate mayserve as osmoticum at low light conditions to compensate forthe shortage of carbohydrates resulting from suboptimal photosynthesis. Root-produced organic compounds in xylem sap, such as hormones and amino acids, are known to be important in plant development. Recently, biochemical approaches have revealed the identities of several xylem sap proteins, but the biological functions and the regulation of the production of these proteins are not fully understood. XYLEM SAP PROTEIN 30 kD (XSP30), which is specifically expressed in the roots of cucumber (Cucumis sativus), encodes a lectin and is hypothesized as affecting the development of above-ground organs. In this report, we demonstrate that XSP30 gene expression and the level of XSP30 protein fluctuate in a diurnal rhythm in cucumber roots. The rhythmic gene expression continues for at least two or three cycles, even under continuous light or dark conditions, demonstrating that the expression of this gene is controlled by a circadian clock. Removal of mature leaves or treatment of shoots with uniconazole-P, an inhibitor of gibberellic acid (GA) biosynthesis, dampens the amplitude of the rhythmic expression; the application of GA negates these effects. These results suggest that light signals perceived by above-ground organs, as well as GA that is produced, possibly, in mature leaves, are important for the rhythmic expression of XSP30 in roots. This is the first demonstration of the regulation of the expression of a clock-controlled gene by GA.  相似文献

Xylem plays a major role in plant development and is considered part of the apoplast. Here, we studied the proteome of Brassica oleracea cv Bartolo and compared it to the plant cell wall proteome of another Brassicaceae, the model plant Arabidopsis thaliana. B. oleracea was chosen because it is technically difficult to harvest enough A. thaliana xylem sap for proteomic analysis. We studied the whole proteome and an N-glycoproteome obtained after Concanavalin A affinity chromatography. Altogether, 189 proteins were identified by LC-MS/MS using Brassica EST and cDNA sequences. A predicted signal peptide was found in 164 proteins suggesting that most proteins of the xylem sap are secreted. Eighty-one proteins were identified in the N-glycoproteome, with 25 of them specific of this fraction, suggesting that they were concentrated during the chromatography step. All the protein families identified in this study were found in the cell wall proteomes. However, proteases and oxido-reductases were more numerous in the xylem sap proteome, whereas enzyme inhibitors were rare. The origin of xylem sap proteins is discussed. All the experimental data including the MS/MS data were made available in the WallProtDB cell wall proteomic database.  相似文献
