Summary Intracellular C1, K and Na activities (a Cl i ,a k i anda Na i ) and transmucosal membrane potential (E m) in epithelial cells ofNecturus gallbladder were measured at different external Na concentrations ([Na]o), with liquid ion-exchanger and conventional microelectrodes. Bladders were mounted in a divided chamber at 23°C between identical HCO3-free Ringer solutions containing 5mm K. The pH was 7.2. Tris was substituted for Na. Measurements were made under steady-state conditions as determined by the constancy of the transepithelial potential difference. Both,a Cl i anda Na i increased in a saturable fashion with [Na]o.E m did not change significantly. Average values (±sem) under normal conditions ([Na]o=100mm) fora Cl i ,a Na i andE m were 16.8±0.8mm (n=9), 9.7±0.6mm (n=10) and –52.6±0.6 mV (n=26), respectively. In Na-free mediaa Cl i declined to its equilibrium value.a K i (96±2mm;n=7) did not change when [Na]o was varied between 100 and 10mm but decreased to 80±3mm (n=4) in Na-free media.Transmembrane electrochemical potential differences, , for Cl and Na were calculated at four different [Na]o levels. A highly significant linear relation between and was found, indicating that Cl and Na transport are energetically linked. The results support the view that the energy necessary for intracellular Cl accumulation is derived from the simultaneous dissipation of the chemical potential gradient of Na across the apical membrane and that the coupled entry mechanism is electroneutral.  相似文献

Summary Near-instantaneous current-voltage relationships and shot-noise analysis of amiloride-induced current fluctuations were used to estimate apical membrane permeability to Na (P Na), intraepithelial Na activity (Na c ), single-channel Na currents (i) and the number of open (conducting) apical Na channels (N0), in the urinary bladder of the toad (Bufo marinus). To facilitate voltageclamping of the apical membrane, the serosal plasma membranes were depolarized by substitution of a high KCl (85mm) sucrose (50mm) medium for the conventional Na-Ringer's solution on the serosal side.Aldosterone (5×10–7 m, serosal side only) elicited proportionate increases in the Na-specific current (I Na and inP Na, with no significant change in the dependence ofP Na on mucosal Na (Na o ).P Na and the control ofP Na by aldosterone were substrate-dependent: In substrate-depleted bladders, pretreatment with aldosterone markedly augmented the response to pyruvate (7.5×10–3 m) which evoked coordinate and equivalent increases inI Na andP Na.The aldosterone-dependent increase inP Na was a result of an equivalent increase in the area density of conducting apical Na channels. The computed single-channel current did not change. We propose that, following aldosterone-induced protein synthesis, there is a reversible metabolically-dependent recruitment of preexisting Na channels from a reservoir of electrically undetectable channels. The results do not exclude the possibility of a complementary induction of Na-channel synthesis.  相似文献

Summary The basal-lateral surface of the epithelium of the urinary bladder of the toad (Bufo marinus) was depolarized by exposure of the serosal surface to 85mm KCL and 50mm sucrose. The extent of depolarization appeared to be virtually complete, as evaluated by the invariance in the transepithelial electrical potential difference and conductance on addition of nystatin (a monovalent cation ionophore) to the serosal medium. The Na-specific current (I Na) was defined as the current sensitive to the removal of Na from the mucosal medium or inhibitable by addition of amiloride to this medium. In the presence of the high K-sucrose serosal medium, rapid, serial, stepwise clamping of the transepithelial voltage (V) yielded a curvilinear dependence ofI Na onV; which is taken to represent theI–V curve of the apical Na channels. The constant field equation (Goldman, D.E. 1943;J. Gen. Physiol. 27:37) fits theI–V data points closely, allowing estimates to be made of the permeability to Na of the apical membrane (P Na) and of the intracellular Na activity (Na c ). Exposure of the apical surface to amiloride (5×10–7 m) decreasedP Na in proportion to the decrease inI Na (i.e., 70%) but decreased Na c only 25%. In contrast, an equivalent lent reduction inI Na elicited by exposure of the basallateral surface to ouabain was accompanied by only a 20% decrease inP Na and a sixfold increase in Na c . The effects of amiloride onP Na and ouabain on Na c are consistent with the primary pharmacological actions of these drugs. In addition,P Na appears to be under metabolic control, in that 2-deoxyglucose, a specific inhibitor of glycolysis, decreasedI Na andP Na proportionately, and lowered Na c marginally, effects indistinguishable from those obtained with amiloride.  相似文献

Summary Transepithelial electrogenic Na transport (INa) was investigated in the colon of the frog Xenopus laevis with electrophysiological methods in vitro. The short circuit current (Isc) of the voltage-clamped tissue was 24.2±1.8 A·cm-2 (n=10). About 60% of this current was generated by electrogenic Na transport. Removal of Ca2+ from the mucosal Ringer solution stimulated INa by about 120%. INa was not blockable by amiloride (0.1 mmol·l-1), a specific Na-channel blocker in epithelia, but a fully and reversible inhibition was achieved by mucosal application of 1 mmol·l-1 lanthanum (La3-). No Na-self-inhibition was found, because INa increased linearly with the mucosal Na concentration. A stimulation of INa by antidiuretic hormones was not possible. The analysis of fluctuations in the short circuit current (noise analysis) indicated that Na ions pass the apical cell membrane via a Ca-sensitive ion channel. The results clearly demonstrate that in the colon of Xenopus laevis Na ions are absorbed through Ca-sensitive apical ion channels. Summary Cell K activity,a k, was measured in the short-circuited frog skin by simultaneous cell punctures from the apical surface with open-tip and K-selective microelectrodes. Strict criteria for acceptance of impalements included constancy of the open-tip microelectrode resistance, agreement within 3% of the fractional apical voltage measured with open-tip and K-selective microelectrodes, and constancy of the differential voltage recorded between the open-tip and the K microelectrodes 30–60 sec after application of amiloride or substitution of apical Na. Skins were bathed on the serosal surface with NaCl Ringer and, to reduce paracellular Cl conductance and effects of amiloride on paracellular conductance, with NaNO3 Ringer on the apical surface.Under control conditionsa k r was nearly constant among skins (mean±SD=92±8mM, 14 skins) in spite of a wide range of cellular currents (5 to 70 A/cm2). Cell current (and transcellular Na transport) was inhibited by either apical addition of amiloride or substitution of Na by other cations. Although in some experiments the expected small increase ina k r after inhibition of cell current was observed, on the average the change was not significant (98±11mM after amiloride, 101±12mM after Na substitution), even 30 min after the inhibition of cell current. The membrane potential, which in the control state ranged from –42 to –77 mV, hyperpolarized after inhibition of cell current, initially to –109±5mV, then depolarizing to a stable value (–88±5mV) after 15–25 min. At this time K was above equilibrium (E k=98±2mV), indicating that the active pump mechanism is still operating after inhibition of transcellular Na transport.The measurement ofa k r permitted the calculation of the passive K current and pump current under control conditions. assuming a constant current source with almost all of the basolateral conductance attributable to K. We found a significant correlation between pump current and cell current with a slope of 0.31, indicating that about one-third of the cell current is carried by the pump, i.e., a pump stoichiometry of 3Na/2K.  相似文献

Summary We have studied current (I Str) through the Na, K pump in amphibian oocytes under conditions designed to minimize parallel undesired currents. Specifically,I Str was measured as the strophanthidin-sensitive current in the presence of Ba2–, Cd2+ and gluconate (in place of external Cl). In addition,I Str was studied only after the difference currents from successive applications and washouts of strophanthidin (Str) were reproducible. The dose-response relationship to Str in four oocytes displayed a meanK 0.5 of 0.4 m, with 2–5 m producing 84–93% pump' block. From baseline data with 12 Na+-preloaded oocytes, voltage clamped in the range [–170, +50 mV] with and without 2–5 m Str, the averageI Str depended directly onV m up to a plateau at 0 mV with interpolated zero current at –165 mV. In three oocytes, lowering the external [Na+] markedly decreased the voltage sensitivity ofI p , while producing only a small change in the maximal outwardI Str. In contrast, decreasing the external [K+] from 25 to 2.5mm reducedI Str at 0 mV without substantially affecting its voltage dependence. At K+ concentrations of 1mm, both the absolute value ofI Str at 0 mV and the slope conductance were reduced. In eight oocytes, the activation of the averagedI Str by [K+] o over the voltage interval [–30, +30 mV] was well fit by the Hill equation, with K=1.7±0.4mm andnH (the minimum number of K+ binding sites) =1.7±0.4. The results unequivocally establish that the cardiotonic-sensitive current ofRana oocytes displays only a positive slope conductance for [K+] o >1mm. There is therefore no need to postulate more than one voltage-sensitive step in the cycling of the Na, K pump under physiologic conditions. The effects of varying external Na+ and K+ are consistent with results obtained in other tissues and may reflect an ion-well effect.  相似文献

Summary To characterize the molecular properties conveyed by the isoforms of the subunit of Na,K-ATPase, the two major transepithelial transporting organs in the brine shrimp (Artemia salina), the salt glands and intestines, were isolated in pure form. The isoforms were quantified by ATP-sensitive fluorescein isothiocyanate (FITC) labeling. The salt gland enzyme exhibits only the 1 isoform, whereas the intestinal enzyme exhibits both the 1 and the 2 isoforms. After 32 hours of development, Na,K-ATPase activity [in mol Pi/mg protein/hr (1u)] in whole homogenates was 32±6 in the salt glands and 12±3 in the intestinal preparations (mean±sem). The apparent half-maximal activation constants (K 1/2) of the salt gland enzyme as compared to the intestinal enzyme were 3.7±0.6mm vs. 23.5±4mm (P<0.01) for Na+, 16.6±2.2mm vs. 8.29±1.5mm for K+ (P<0.01), and 0.87±0.8mm vs. 0.79±1.1mm for ATP (NS). The apparentK i's for ouabain inhibition were 1.1×10–4 m vs. 2×10–5 m, respectively. Treatment of whole homogenates with deoxycholic acid (DOC) produced a maximal Na,K-ATPase activation of 46% in the salt gland as compared to 23% in the intestinal enzyme. Similar differences were found with sodium dodecyl sulfate (SDS). The two distinct forms of Na,K-ATPase isolated from the brine shrimp differed markedly in three kinetic parameters as well as in detergent sensitivity. The differences inK 1/2 for Na+ and K+ are more marked than those reported for the mammalian Na,K-ATPase isoforms. Summary The Na conductance of the apical membrane of the toad urinary bladder was measured at different concentrations of Na both in the external medium and in the cell. Bladders were bathed in high K-sucrose medium to reduce basal-lateral resistance and voltage, and the transepithelial currents measured under voltage-clamp conditions. Amiloride was used as a specific blocker of the apical Na channel. At constant external Na, the internal Na concentration was increased by blocking the basallateral Na pump with ouabain. With high Na activity in the mucosal medium (86mm), increases in intracellular Na activity from 10 to over 40mm increased the amiloride-sensitive slope conductance at zero voltage while apical Na permeability, estimated from current-voltage plots using the constant field equation, decreased by less than 20%. Lowering the serosal Ca concentration from 1 to 0.1mm had no effect on the change inP Na with increasing Nac, but increasing serosal Ca to 5mm enhanced the reduction inP Na with increasing Na c , presumably by increasing Ca influx into the cell.P Na was also reduced by serosal vanadate (0.5mm), a putative blocker of ATP-dependent Ca extrusion from the cell, and by acute exposure to CO2, which presumably acidifies the cytoplasm. Current-voltage relationships of the amiloridesensitive transport pathway were also measured in the absence of a Na gradient across the apical membrane. These plots show that outward current passes through the channels somewhat less easily than does inward current. The shape of theI-V relationships was not significantly altered by changes in cellular Na, Ca or H, indicating that the effects of these ions onP Na are voltage independent.  相似文献

Summary Effects of a potent synthetic glucocorticoid, methylprednisolone (MP), on transepithelial Na transport were examined in rabbit descending colon. Current-voltage (I–V) relations of the amiloride-sensitive apical Na entry pathway were measured in colonic tissues of control and MP-treated (40 mg im for 2 days) animals. Tissues were bathed mucosally by solutions of various Na activities, (Na)m, ranging from 6.2 to 75.6mm, and serosally by a high K solution. TheseI–V relations conformed to the constant field flux equation permitting determination of the permeability of the apical membrane to Na,P Na m , and the intracellular Na activity, (Na)c. The following empirical relations were observed for both control and MP-treated tissues: (i) Na transport increases hyperbolically with increasing (Na)m obeying simple Michaelis-Mentin kinetics; (ii)P Na m decreased hyperbolically with increasing (Na)m, but was unrelated to individual variations in (Na)c; (iii) (Na)c increased hyperbolically with (Na)m; (iv) both spontaneous and steroid-stimulated variations in Na entry rate could be attributed entirely to parallel variations inP Na m at each mucosal Na activity. Comparison of these empirical, kinetic relations between control and MP-treated tissues revealed: (i) maximal Na current andP Na m were greater in MP tissues, but the (Na)m's at which current andP Na m were half-maximal were markedly reduced; (ii) (Na)c was significantly increased in MP tissues at each (Na)m while the (Na)m at half-maximal (Na)c was unchanged. These results provide direct evidence that glucocorticoids cause marked stimulation of Na absorption across rabbit colon primarily by increasing the Na permeability of the apical membrane. While the mechanism for the increased permeability remains to be determined, the altered relation betweenP Na m and (Na)m suggests possible differences in the conformation or environment of the Na channel in MP-treated tissues.  相似文献

Summary Na, K-ATPase function was studied in order to evaluate the mechanism of increased colonic Na+ transport during early postnatal development. The maximum Na+-pumping activity that was represented by the equivalent short-circuit current after addition of nystatin (I sc N ) did not change during postnatal life or after adrenalectomy performed in 16-day-old rats.I sc N was entirely inhibited by ouabain; the inhibitory constant was 0.1mm in 10-day-old (young) and 0.4mm in 90-day-old (adult) rats. The affinity of the Na, K pump for Na+ was higher in young (11mm) than in adult animals (19mm). The Na, K-ATPase activity (measured after unmasking of latent activity by treatment with sodium dodecylsulfate) increased during development and was also not influenced by adrenalectomy of 16-day-old rats. The inhibitory constant for ouabain (K I ) was not changed during development (0.1–0.3mm). Specific [3H]ouabain binding to isolated colonocytes increased during development (19 and 82 pmol/mg protein), the dissociation constant (K D ) was 8 and 21 m in young and adult rats, respectively. The Na+ turnover rate per single Na, K pump, which was calculated fromI sc N and estimated density of binding sites per cm2 of tissue was 500 in adult and 6400 Na+/min·site in young rats. These data indicate that the very high Na+ transport during early postnatal life reflects an elevated turnover rate and increased affinity for Na+ of a single isoform of the Na, K pump. The development of Na+ extrusion across the basolateral membrane is not directly regulated by corticosteroids.  相似文献

Summary Single barnacle muscle fibers fromBalanus nubilus were used to study the effect of elevated external potassium concentration, [K] o , on Na efflux, membrane potential, and cyclic nucleotide levels. Elevation of [K] o causes a prompt, transient stimulation of the ouabain-insensitive Na efflux. The minimal effective concentrations is 20mm. The membrane potential of ouabain-treated fibers bathed in 10mm Ca2+ artificial seawater (ASW) or in Ca2+-free ASW decreases approximately linearly with increasing logarithm of [K] o . The slope of the plot is slightly steeper for fibers bathed in Ca2+-free ASW. The magnitude of the stimulatory response of the ouabain-insensitive Na efflux to 100mmK o depends on the external Na+ and Ca2+ concentrations, as well as on external pH, but is independent of external Mg2+ concentration. External application of 10–4 m verapamil virtually abolishes the response of the Na efflux to subsequent K-depolarization. Stabilization of myoplasmic-free Ca2+ by injection of 250mm EGTA before exposure of the fiber to 100mm K o leads to 60% reduction in the magnitude of the stimulation. Pre-injection of a pure inhibitor of cyclic AMP-dependent protein kinase reduces the response of the Na efflux to 100mm K o by 50%. Increasing intracellular ATP, by injection of 0.5m ATP-Na2 before elevation of [K] o , fails to prolong the duration of the stimulation of the Na efflux. Exposure of ouabain-treated, cannulated fibers to 100mm K o for time periods ranging from 30 sec to 10 min causes a small (60%), but significant, increase in the intracellular content of cyclic AMP with little change in the cyclic GMP level. These results are compatible with the view that the stimulatory response of the ouabain-insensitive Na efflux to high K o is largely due to a fall in myoplasmicpCa resulting from activation of voltage-dependent Ca2+ channels and that an accompanying rise in internal cAMP accounts for a portion of this response.  相似文献

Summary Ouabain abolishes the short-circuit current (I sc ) and decreases the transepithelial conductance (G t ) of rabbit colon. In contrast, amphotericin B elicits a maximumI sc and markedly increasesG t . However, inboth instances the amiloride-sensitive Na entry step is completely blocked, presumably due to an increase in cell Na. Conversely, when Na-depleted tissues are suddenly exposed to 140mm Na, the amiloride-sensitiveI sc and the amiloride-sensitive component ofG t ( a G Na ) increase abruptly to their maximum values and the decline to steady-state plateaus with a half time of 6 min; throughout the decline (I sc/a G Na)=E Na is constant at a value of 95 mV. In the presence of amphotericin B, theI sc abruptly rises to the same maximum but does not decline. These findings indicate that in the presence of 140mm Na the conductance of the amiloride-sensitive Na entry step can vary from a maximum value of approximately 1.6 mmhos/cm2 when cell Na is depleted, to zero when cell Na is abnormally elevated (e.g., in the presence of ouabain or amphotericin B). Our findings are consistent with a system in which the pathway responsible for transcellular Na transport parallels another cellular compartment with which it communicates. The Na capacity of the active transport pathway appears to be very small so that this compartment fills rapidly after exposure of Na-depleted cells to 140mm Na, and active transepithelial Na transport is initiated and reaches steady-state levels quickly. The Na capacity of the second compartment is much larger; the Na content of this compartment appears to be responsible for the negative feedback effect on the permeability of the amiloride-sensitive entry step.  相似文献

Summary Intracellular potassium activity (a K i ) was measured in control conditions in mid-cortical rabbit proximal convoluted tubule using two methods: (i) by determination of the K+ equilibrium potential (E K) using Ba2+-induced variations in the basolateral membrane potential (V BL) during transepithelial current injections and (ii) with double-barrel K-selective microelectrodes. Using the first method, the meanV BL was –48.5±3.2 mV (n=16) and the meanE K was –78.4±4.1 mV corresponding to aa K i of 68.7mm. With K-selective microelectrodes,V BL was –36.6±1.1 mV (n=19),E K was –64.0±1.1 mV anda K i averaged 40.6±1.7mm. While these lastE K andV BL values are significantly lower than the corresponding values obtained with the first method (P<0.001 andP<0.01, respectively), the electrochemical driving force for K transport across the basolateral membrane ( K =V BLE K) is not significantly different for both techniques (30.1±3.3 mV for the first technique and 27.6±1.8 mV for ion-selective electrodes). This suggests an adequate functioning of the selective barrel but an underestimation ofV BL by the reference barrel of the double-barrel microelectrode. Such double-barrel microelectrodes were used to measure temporal changes ina K i and K in different experimental conditions where Na reabsorption rate (J Na) was reduced.a K i was shown to increase by 12.2±2.7 (n=5) and 14.1±4.4mm (n=5), respectively, whenJ Na was reduced by omitting in the luminal perfusate: (i) 5.5mm glucose and 6mm alanine and (ii) glucose, alanine, other Na-cotransported solutes and 110mm Na. In terms of the electrochemical driving force for K exit across the basolateral membrane, K, a decrease of 5.4±2.0 mV (P<0.05,n=5) was measured when glucose and alanine were omitted in the luminal perfusate while K remained unchanged whenJ Na was more severely reduced (mean change =–1.7±2.1 mV, NS,n=5). In the latter case, this means that the electrochemical driving force for K efflux across the basolateral membrane has not changed while both the active influx through the Na–K pump and the passive efflux in steady state are certainly reduced. If the main pathway for K transport is through the basolateral K conductance, this implies that this conductance must have decreased in the same proportion as that of the reduction in the Na–K pump activity.  相似文献

Summary We have examined the effect of internal and external pH on Na+ transport across toad bladder membrane vesicles. Vesicles prepared and assayed with a recently modified procedure (Garty & Asher, 1985) exhibit large, rheogenic, amiloridesensitive fluxes. Of the total22Na uptake measured 0.5–2.0 min after introducing tracer, 80±4% (mean±se,n=9) is blocked by the diuretic with aK 1 of 2×10–8 m. Thus, this amiloridesensitive flux is mediated by the apical sodium-selective channels. Varying the internal (cytosolic) pH over the physiologic range 7.0–8.0 had no effect on sodium transport; this result suggests that variation of intracellular pHin vivo has no direct apical effect on modulating sodium uptake. On the other hand,22Na was directly and monotonically dependent on external pH. External acidification also reduced the amiloride-sensitive efflux across the walls of the vesicles. This inhibition of22Na efflux was noted at external Na+ concentrations of both 0.2 m and 53mm.These results are different from those reported with whole toad bladder. A number of possible bases for these differences are considered and discussed. We suggest that the natriferic response induced by mucosal acidification of whole toad urinary bladder appears to operate indirectly through one or more factors, presumably cytosolic, present in whole cells and absent from the vesicles.  相似文献

Summary Na–K–Cl cotransport stoichiometry and affinities for Na, K and Cl were determined in flounder intestine. Measurement of simultaneous NaCl and RbCl influxes resulted in ratios of 2.2 for Cl/Na and 1.8 for Cl/Rb. The effect of Na and Rb on Rb influx showed first order kinetics withK 1/2 values of 5 and 4.5mm and Hill coefficients of 0.9 and 1.2, respectively. The effect of Cl on rubidium influx showed a sigmoidal relationship withK 1/2 of 20mm and a Hill coefficient of 2.0. The effects of variations in Na and Cl concentration on short-circuit current (I sc) were also determined. TheK 1/2 for Na was 7mm with a Hill coefficient of 0.9 and theK 1/2 for Cl was 46mm with a Hill coefficient of 1.9. Based on the simultaneous influx measurements, a cotransport stoichiometry of 1Na1K2Cl is concluded. The Hill coefficients for Cl suggest a high degree of cooperativity between Cl binding sites. Measurements of the ratio of net Na and Cl transepithelial fluxes under short-circuit conditions (using a low Na Ringer solution to minimize the passive Na flux) indicate that the Cl/Na flux ratio is approximately 21. Therefore Na recycling from serosa to mucosa does not significantly contribute to theI sc. Addition of serosal ouabain (100 m) inhibited Rb influx, indicating that Na–K–Cl cotransport is inhibited by ouabain. This finding suggests that a feedback mechanism exists between the Na–K-ATPase on the basolateral membrane and the apical Na–K–2Cl cotransporter.  相似文献

Summary Apical Na+ entry into frog skin epithelium is widely presumed to be electrodiffusive in nature, as for other tight epithelia. However, in contrast to rabbit descending colon andNecturus urinary bladder, the constant field equation has been reported to fit the apical sodium current (N Na)-membrane potential (mc) relationship over only a narrow range of apical membrane potentials or to be inapplicable altogether. We have re-examined this issue by impaling split frog skins across the basolateral membrane and examining the current-voltage relationships at extremely early endpoints in time after initiating pulses of constant transepithelial voltage. In this study, the rapid transient responses in mc were completed within 0.5 to 3.5 msec. Using endpoints to 1 to 25 msec, the Goldman equation provided excellent fits of the data over large ranges in apical potential of 300 to 420 mV, from approximately –200 to about +145 mV (cell relative to mucosa). Split skins were also studied when superfused with high serosal K+ in order to determine whether theI Na-mc relationship could be generated purely by transepithelial measurements. Under these conditions, the basolateral membrane potential was found to be –10±3 mV (cell relative to serosa, mean±se), the basolateral fractional resistance was greater than zero, and the transepithelial current was markedly and reversibly reduced. For these reasons, use of high serosal K+ is considered inadvisable for determining theI Na-mc relationship, at least in those tissues (such as frog skin) where more direct measurements are technically feasible. Analysis of theI Na-mc relationships under baseline conditions provided estimates of intracellular Na+ concentration and of apical Na+ permeability of 9 to 14mm and of 3 × 10–7 cm · sec–1, respectively, in reasonable agreement with estimates obtained by different techniques.  相似文献

Summary The apical membrane of rabbit urinary bladder can be functionally removed by application of nystatin at high concentration if the mucosal surface of the tissue is bathed in a saline which mimics intracellular ion concentrations. Under these conditions, the tissue is as far as the movement of univalent ions no more than a sheet of basolateral membrane with some tight junctional membrane in parallel. In this manner the Na+ concentration at the inner surface of the basolateral membrane can be varied by altering the concentration in the mucosal bulk solution. When this was done both mucosal-to-serosal22Na flux and net change in basolateral current were measured. The flux and the current could be further divided into the components of each that were either blocked by ouabain or insensitive to ouabain. Ouabain-insensitive mucosal-to-serosal Na+ flux was a linear function of mucosal Na+ concentration. Ouabain-sensitive Na+ flux and ouabain-sensitive, Na+-induced current both display a saturating relationship which cannot be accounted for by the presence of unstirred layers. If the interaction of Na+ with the basolateral transport process is assumed to involve the interaction of some number of Na+ ions,n, with a maximal flux,M max, then the data can be fit by assuming 3.2 equivalent sites for interaction and a value forM max of 287.8pm cm–2 sec–1 with an intracellular Na concentration of 2.0mm Na+ at half-maximal saturation. By comparing these values with the ouabain-sensitive, Na+-induced current, we calculate a Na+ to K+ coupling ratio of 1.40±0.07 for the transport process.  相似文献

Summary The Na+ requirement for active, electrogenic Cl absorption byAmphiuma small intestine was studied by tracer techniques and double-barreled Cl-sensitive microelectrodes. Addition of Cl to a Cl-free medium bathingin vitro intestinal segments produced a saturable (K m =5.4mm) increase in shortcircuit current (I sc) which was inhibitable by 1mm SITS. The selectivity sequence for the anion-evoked current was Cl=Br>SCN>NO 3 >F=I. Current evoked by Cl reached a maximum with increasing medium Na concentration (K m =12.4mm). Addition of Na+, as Na gluconate (10mm), to mucosal and serosal Na+-free media stimulated the Cl current and simultaneously increased the absorptive Cl flux (J ms Cl ) and net flux (J net Cl ) without changing the secretory Cl flux (J sm Cl ). Addition of Na+ only to the serosal fluid stimulatedJ ms Cl much more than Na+ addition only to the mucosal fluid in paired tissues. Serosal DIDS (1mm) blocked the stimulation. Serosal 10mm Tris gluconate or choline gluconate failed to stimulateJ ms Cl . Intracellular Cl activity (a Cl i ) in villus epithelial cells was above electrochemical equilibrium indicating active Cl uptake. Ouabain (1mm) eliminated Cl accumulation and reduced the mucosal membrane potential m over 2 to 3 hr. In contrast, SITS had no effect on Cl accumulation and hyperpolarized the mucosal membrane. Replacement of serosal Na+ with choline eliminated Cl accumulation while replacement of mucosal Na+ had no effect. In conclusion by two independent methods active electrogenic Cl absorption depends on serosal rather than mucosal Na+. It is concluded that Cl enters the cell via a primary (rheogenic) transport mechanism. At the serosal membrane the Na+ gradient most likely energizes H+ export and regulates mucosal Cl accumulation perhaps by influencing cell pH or HCO 3 concentration.  相似文献

Summary It was previously shown that ouabain decreases the potential difference across anin vitro preparation of bullfrog retinal pigment epithelium (RPE) when applied to the apical, but not the basal, membrane and that the net basal-to-apical Na+ transport is also inhibited by apical ouabain. This suggested the presence of a Na+–K+ pump on the apical membrane of the RPE. In the present experiments, intracellular recordings from RPE cells show that this pump is electrogenic and contributes approximately –10 mV to the apical membrane potential (V AP). Apical ouabain depolarizedV AP in two phases. The initial, fast phase was due to the removal of the direct, electrogenic component. In the first one minute of the response to ouabain,V AP depolarized at an average rate of 4.4±0.42 mV/min (n=10, mean ±sem), andV AP depolarized an average of 9.6±0.5 mV during the entire fast phase. A slow phase of membrane depolarization, due to ionic gradients running down across both membranes, continued for hours at a much slower rate, 0.4 mV/min. Using a simple diffusion model and K+-specific microelectrodes, it was possible to infer that the onset of the ouabain-induced depolarization coincided with the arrival of ouabain molecules at the apical membrane. This result must occur if ouabain affects an electrogenic pump. Other metabolic inhibitors, such as DNP and cold, also produced a fast depolarization of the apical membrane. For a decrease in temperature of 10°C, the average depolarization of the apical membrane was 7.1±3.4 mV (n=5) and the average decrease in transepithelial potential was 3.9±0.3 mV (n=10). These changes in potential were much larger than could be explained by the effect of temperature on anRT/F electrodiffusion factor. Cooling the tissue inhibited the same mechanism as ouabain, since prior exposure to ouabain greatly reduced the magnitude of the cold effect. Bathing the tissue in 0mm [K+] solution for 2 hr inhibited the electrogenic pump, and subsequent re-introduction of 2mm [K+] solution produced a rapid membrane hyperpolarization. We conclude that the electrogenic nature of this pump is important to retinal function, since its contribution to the apical membrane potential is likely to affect the transport of ions, metabolites, and fluid across the RPE.  相似文献

Summary This paper describes measurements of electrical potentials generated by renal Na/K-ATPase reconstituted into proteoliposomes, utilizing the anionic dye, oxonol VI. Calibration of absorption changes with imposed diffusion potentials allows estimation of absolute values of electrogenic potentials.ATP-dependent Nacyt/Kexc exchange in K-loaded vesicles generates large potentials, up to 250 mV. By comparing initial rates or steady-state potentials with ATP-dependent22Na fluxes in different conditions, it is possible to infer whether coupling ratios are constant or variable. For concentrations of Nacyt (2–50mm) and ATP (1–1000 m) and pH's (6.5–8.5), the classical 3Nacyt/2Kexc coupling ratio is maintained. However, at low Nacyt concentrations (<0.8mm), the coupling ratio is apparently less than 3Nacyt/2Kexc.ATP-dependent Nacyt/congenerexc exchange in vesicles loaded with Rb, Cs, Li and Na is electrogenic. In this mode congeners, including Naexc, act as Kexc surrogates in an electrogenic 3Nacyt/2congenerexc exchange. (ATP+Pi)-dependent Kcyt/Kexc exchange in K-loaded vesicles is electroneutral.ATP-dependent uncoupled Na flux into Na- and K-free vesicles is electroneutral at pH 6.5–7.0 but becomes progressively electrogenic as the pH is raised to 8.5. The22Na flux shows no anion specificity. We propose that uncoupled Na flux is an electroneutral 3Nacyt/3Hexc exchange at pH 6.5–7.0 but at higher pH's the coupling ratio changes progressively, reaching 3Na/no ions at pH 8.5. Slow passive pump-mediated net K uptake into Na- and K-free vesicles is electroneutral, and may also involve Kcyt/Hexc exchange.We propose the general hypothesis that coupling ratios are fixed when cation transport sites are saturated, but at low concentrations of transported cations, e.g., Nacyt in Na/K exchange and Hexc in uncoupled Na flux, coupling ratios may change.  相似文献
