Mature stage IVb Phycomyces sporangiophores show left-hand spiral growth; that is, viewed from above, the sporangium rotates clockwise. It has been shown that mechanical conditioning (strain-hardening) of the cell wall by the Instron technique increases the ratio of rotation to the elongation growth rate compared to nonmechanically conditioned controls. It is reported that the addition of a saturating light stimulus to these sporangiophores causes a decrease in the ratio of rotation to elongation growth rate. This result is in agreement with the fibril slippage model, i.e. the counterclockwise rotation of stage IVa is a result of parallel fibrils lying in a right-handed spiral configuration slipping by one another. It is suggested that a light stimulus added to a mechanically conditioned stage IVb sporangiophore activates one or more cell wall-loosening enzymes which act by decreasing the number of intermolecular bonds between parallel fibrils causing fibril slippage, resulting in counterclockwise rotation. It is precisely this counterclockwise contribution that decreases the rotation to elongation growth ratio of mechanically conditioned and then light-stimulated stage IVb sporangiophores.  相似文献

Studies have been mode of the growth curvatures induced in theroots of Lepidium sativum, the coleoptiles of Avena saliva andthe sporangiophores of Phycomyces blakesleeanus by exposureto an intense magnetic gradient during continuous rotation ona slow klinostat (0.5 rpm). Attempts have been made to determinethe relationships of the curvature responses to the parametersof the magnetic field (H, dH/dx, and H. dH/dx). Analysis ofthe data was by a progressive regression analysis up to thethird order coupled with an analysis of variance to test theimprovement of fit at each stage. Response is characterizedby an initial lag (reaction time, T) followed by a constantrate of curvature down the magnetic gradient for the first 20or 30? of curvature. The reciprocal of T was the most satisfactorymeasure of response (i. e. a measure of the rate of inductionof the response while the organ was in a constant magnetic environment)and in Avena and Phycomyces was best fitted by linear regressionson logedH/dx and on logeH. dH/dx. For Lepidium, with much morevariable data, the best fit was with a linear regression onlogeH. The significance of these results is discussed.  相似文献

Previous views on the physical basis of phototropism in Phycomycessporangiophores are briefly discussed.

Rapid reversal of the curvature followedwhen theillumination was transferred to the opposite edge ofthe sporangiophore.

The iron-protein ferritin has been purified from mycelium, sporangiophores, and spores of the fungus Phycomyces blakesleeanus. It has a protein-to-iron ratio of 5, a sedimentation coefficient of 55S, a buoyant density in CsCl of 1.82 g/cm3, and the characteristic morphology of ferritin in the electron microscope. Apoferritin prepared from Phycomyces ferritin has a sedimentation coefficient of 18S and consists of subunits of molecular weight 25,000. In the cytoplasm of Phycomyces, ferritin is located on the surface of lipid droplets (0.5–2.0 µ in diameter) where it forms crystalline monolayers which are conspicuous in electron micrographs of sporangiophore thin-sections. Ferritin is found in all developmental stages of Phycomyces but is concentrated in spores. The level of ferritin iron is regulated by the iron level in the growth medium, a 50-fold increase occurring on iron-supplemented medium.  相似文献

Glass microelectrodes were inserted into the growing zone of sporangiophores of Phycomyces blakesleeanus that had been submersed in artificial pond water. The membrane potential (inside negative) increased with increasing pH of the bathing solution from an average of ?98 mV at pH 5 up to ?131 mV at pH 7. Removal of Ca2+ from the medium hyperpolarized the membrane potential in the wild type, but caused a significant depolarization in the blue-light-insensitive madC mutant. KCN, diethylstilbestrol, and N,N′-dicyclohexylcarbodiimide depolarized the membrane potential in both the wild type and the madC mutant, while fusicoccin had no effect. Endogenous ion current of up to 2 μA cm?2 was measured in the growing zone of sporangiophores with an extracellular vibrating electrode. The current density and current pattern varied with the pH of the medium. At pH 5 most sporangiophores had weak inward current along the growing zone, whereas at pH 7 most sporangiophores had strong outward current. The response of the membrane potential to specific inhibitors and the presence of an endogenous ion current indicate an electrogenic H+-ATPase in the plasma membrane. The results show a negative correlation between growth rate of sporangiophores growing in buffered aqueous medium and magnitude of membrane potential, as well as density of outward current. They also indicate an important role of protons in controlling the growth of Phycomyces sporangiophores.  相似文献

The initiation of sporangiophores of Phycomyces was analyzed under oxygen-limiting conditions. Mutants lacking -carotene have a higher oxygen threshold than the wild type depending on the residual amount of -carotene. The supersensitivity to low oxygen tension is specific for sporangiophore initiation and can be suppressed by addition of either retinal, retinol or retinol acetate to the medium. It is suggested that retinol is a natural regulator of differentiation in Phycomyces.  相似文献

Dynamic Mechanical Properties of the Cell Walls of Some Green Algae   总被引:2,自引:0,他引:2  
Stress relaxation and constant rate of strain measurements weremade on the cell walls of the green algae Nitella, Acetabularia,and Penicillus. These plants have, as theirmain structural polysaccharide,cellulose, mannan, and xylan respectively. The stress relaxationmeasurements were made in the temperature range 0 to 50 °Cfor the interval 0.02 to 103 s. A limited number of constantrate of strain curves were also obtained for comparison withtheoretical curves deduced from the relaxation data. All three algae exhibit typical fibrous behaviour but Acetabulariacould not be mechanically conditioned to give reproducible results.The time-temperature correspondence of the data and its relevanceto the molecular organization of the cell wall are discussed.Structural changes with temperature, deduced in previous workfrom rigorous matching of the relaxation curves at differenttemperatures, are now thought not to occur. The matching procedureonly appears to work because the curves lack distinctive featureson which to base the superposition. Comparison of stress relaxationand constant rate of strain data show that the cell walls ofthese algae exhibit non-linear viscoelastic behaviour and apparentchanges in the quasi-static modulus with temperature can beexplained satisfactorily in terms of reaction rate theoriesfor viscoelastic processes.  相似文献

 Small segments of Phycomyces sporangiophores regenerate various structures on incubation in a moist chamber. We tested the regeneration capacities of middle sporangiophore segments whose protoplasm had been totally or partially removed and replaced with protoplasm from various segments of a genetically different strain. The structures that were regenerated depended on the source of the injected protoplasm (sporangia of various sizes and segments from middle and basal parts of the sporangiophores), implying a positional differentiation of protoplasm along the sporangiophore axis. Protoplasm from various sources showed a high affinity; that is, they mixed successfully and led, in most cases, to the formation of heterokaryotic regenerating structures. The steady-state phototropic bending of Phycomyces sporangiophores was studied using apparatus designed to keep the growing zone vertical and the angle of illumination constant over long periods of time. The bending speed is quite constant if the intensity and angle of illumination are fixed. A phototropic inversion occurs in response to a sudden change in intensity, either an increase or a decrease. A bending component lateral to the illumination direction is strongly evident at normal incidence. It is shown that this component is due to a rotation between the stimulus and response loci about the axis of the growing zone, which is probably related to the spiral growth of the cell. The steady-state bending speed is at a maximum value for illumination directions ranging from normal incidence to about 45°. From 45 to 14° the bending speed decreases linearly with angle, reaching zero at 14°. Angles less than 14° elicit a weak negative phototropic response. Using an optical model of the growing zone, the intracellular intensity distribution was determined as a function of the angle of illumination. Several hypotheses relating the intensity distribution to the phototropic response are discussed.  相似文献

Two hydroxybenzoic acids, gallic and protocatechuic acids, are abundant in the giant sporangiophores of the zygomycetePhycomyces blakesleeanus,much more so than in the basal mycelium or the culture medium. The actual concentrations vary with illumination, age of the culture, and composition of the medium. We devised a simple screening procedure to isolatehbamutants whose sporangiophores contained less gallic acid than the wild type. The most useful mutant had very low concentrations of hydroxybenzoic acids in the sporangiophores, but about the same as the wild type in the basal mycelium and the medium. The mutant was only slightly different from the wild type in growth and morphology. Mutant and wild-type sporangiophores grew away from ultraviolet C sources (260 nm) equally well. Contrary to previous conjectures, ultraviolet tropism does not depend on the ultraviolet absorption of gallic acid or other free hydroxybenzoic acids in the sporangiophore. Against expectations, phenols did not impair DNA extraction: sporangiophores produced better DNA preparations than basal mycelia and thehbamutant only slightly better than the wild type.  相似文献

Summary The respiration of stage-I,-III, and-IV sporangiophores of Phycomyces was measured. The of stages-I and-IV is low compared to that of actively growing mycelium. The of stage-III sporangiophores is somewhat higher than that of stages-I and-IV. As stage-I sporangiophores elongate their respiration rate increases proportionately. The respiration rate of stage-IV sporangiophores does not change as they elongate. In stage-IV sporangiophores the growing zone and the columella together consume as much oxygen as the rest of the sporangiophore does. The growth rate of the Phycomyces sporangiophore fluctuates under constant environmental conditions. These fluctuations underlie the well-characterized sensory responses to environmental changes. We compared growth fluctuations in sporangiophores of unstimulated wild type and behavioral mutants by use of maximum entropy spectral analysis, a mathematical technique that estimates the frequency and amplitude of oscillations in a time series. The mutants studied are believed to be altered near the input (“night-blind”) or output (“stiff” and “hypertropic”) of the photosensory transduction chain. The maximum entropy spectrum of wild type shows a sharp drop-off in spectral density above 0.3 millihertz, several minor peaks between 0.3 and 10 millihertz, and a broad maximum near 10 millihertz. Similar spectra were obtained for a night-blind mutant and a hypertropic mutant. In contrast, the spectra of three stiff mutants, defective in genes madD, madE, or madG, had distinctive peaks near 1.6 mHz and harmonics of this frequency. A madF stiff mutant, which is less stiff than madD, madE, and madG mutants, had a spectrum intermediate between wild type and the three other stiff mutants. Our results indicate that alterations in one or more steps associated with growth regulation output cause the Phycomyces sporangiophore to express a rhythmic growth rate.  相似文献

Refractive indices of intact sporangiophores of Phycomyces were computed from measurements of focal length and radius of curvature of the cells. For the six cells studied, effective values of n were obtained ranging from 1.35 to 1.40. The average effective n was 1.38. Senn''s determination of refractive indices of other plants cells gave much higher values: n = 1.37 to 1.52. The precision of the method and possible sources of this discrepancy are discussed.  相似文献

The effect of high levels of natural light on leaf photosynthesisin olive trees (Olea europaea L. var. Coratina), grown in potsoutdoors in the summer and subjected to water, stress, was studied.Net photosynthetic rates reached maximum values early in themorning in both control and stressed plants and subsequentlydeclined gradually. This inactivation of photosynthetic activitywas accompanied by changes in the fluorescence characteristicsof the upper intact leaf surface. The maximum fluorescence yield(Fp) and the ratio Fv/Fp decreased at midday especially in water-stressedplants, but the initial fluorescence (Fo) rose to a maximumvalue at midday and declined again in the afternoon. In controlplants the values of maximum fluorescence Fp and the ratio Fv/Fpincreased again in the afternoon and had recovered almost completelyby 8 p.m. as the leaf water potential recovered. In stressedplants this diurnal recovery was not complete, so that the photosyntheticrates and the ratio Fv/Fp declined gradually during the developmentof water stress. These results indicate that in olive treessubjected to severe water stress the non-stomatal componentof photosynthesis was affected and perhaps a light-dependentinactivation of the primary photochemistry associated with photosystemII (PSII) occurred. Four to five days after rewatering severelystressed plants, the predawn leaf water potential, net photosyntheticrates and chlorophyll fluorescence indices recovered only partially. Phycomyces sporangiophores grow away from stationary objects, a phenomenon known as the avoidance response. Evidence is presented suggesting that a growth-stimulating gas is emitted from the sporangiophore and is then swept to the leeward side by air currents resulting in higher gas concentration on that side. The presence of a stationary barrier decreases the passive movement of the gas away from the leeward side. It is proposed that an increase of this gas on one side causes that side to grow faster. Indirect evidence suggests that the gas is water vapor.  相似文献

Five specimens that contained a continuous gradient of wood, from normal to tension wood regions, were collected from an inclined yellow poplar (Liriodendron tulipifera), and the released strain of tensile growth stress was quantified. Ultraviolet (UV) microspectrophotometry was used to examine the relationship between lignin distribution in the cell wall and the intensity of tensile growth stress. The UV absorption of the secondary wall and the cell corner middle lamella decreased with increasing tensile released strain (i.e., tensile growth stress). The UV absorption in the compound middle lamella region remained virtually constant, irrespective of the tensile released strain. The absorption maximum (5max) remained virtually constant in the secondary wall, the cell corner middle lamella, and the compound middle lamella region at 273-274, 277-278, and 275-278 nm respectively, irrespective of the tensile released strain. The ratios of the UV absorbance at 280 to 260 nm and 280 to 273 nm of the secondary wall decreased with increasing tensile released strain. The ratios in the cell corner and compound middle lamella region remained constant, irrespective of the tensile released strain. The lignin content of the secondary wall decreased, while the syringyl/guaiacyl ratio increased with increasing tensile released strain. Gelatinous fibers were not observed in the tension wood regions, but the secondary wall became gelatinous-layer-like, i.e., the lignin content and microfibrillar angle decreased and the cellulose content increased. A definite gelatinous layer seems to be important for generating greater tensile growth stress. It is concluded that a decrease in lignin and an increase in cellulose microfibrils parallel to the fiber axis in the secondary wall are necessary to produce large tensile growth stress.  相似文献

Under the circumstances of experimentation described, the sporangiophores of Phycomyces are found to be most sensitive to stimulation by light in the violet between 400 and 430 mµ. Toward the red, sensitivity falls to nearly zero near 580 mµ, while in the near ultra-violet around 370 mµ, sensitivity is still high. The previous experiments of Blaauw had placed the point of greatest sensitivity some 80 mµ nearer the red end of the spectrum. Because of the known presence in the sporangiophores of Phycomyces of "accessory" pigments, care must be taken in identifying such results with the absorption spectrum of the photosensitive substance.  相似文献

A low-speed centrifuge was used to study the tropic responses of Phycomyces sporangiophores in darkness to the stimulus of combined gravitational and centrifugal forces. If this stimulus is constant the response is a relatively slow tropic reaction, which persists for up to 12 hours. The response is accelerated by increasing the magnitude of the gravitational-centrifugal force. A wholly different tropic response, the transient response, is elicited by an abrupt change in the gravitational-centrifugal stimulus. The transient response has a duration of only about 6 min. but is characterized by a high bending speed (about 5°/min.). An analysis of the distribution of the transient response along the growing zone shows that the active phase of the response has a distribution similar to that of the light sensitivity for the light-growth and phototropic responses. Experiments in which sporangiophores are centrifuged in an inert dense fluid indicate that the sensory mechanism of the transient response is closely related to the physical deformation of the growing zone caused by the action of the gravitational-centrifugal force on the sporangiophore as a whole. However, the response to a steady gravitational-centrifugal force is most likely not connected with this deformation, but is probably triggered by the shifting of regions or particles of differing density relative to one another inside the cell.  相似文献

Summary An efficient method is described for obtaining heterokaryons in Phycomyces by grafting stage I sporangiophores of two strains to one another. The grafts are successful in over 70% of the cases and success may be ascertained after 10 hours. The successful grafts in almost all cases regenerate one or more sporangiophores and sporangia, and about 85% of the regenerates are heterokaryotic.  相似文献

Phototropic reversal of Phycomyces sporangiophores can be elicited by a change to darkness during steady-state phototropism. The reversal lasts 25–30 min under these conditions. Control experiments show that the reversal is not caused by gravitropism. Tropic reversal is also elicited by the removal of a barrier during an avoidance response, showing that the reversal occurs at the output of the sensory transduction chain.  相似文献
