Biosynthesis of the aliphatic components of suberin was studied in suberizing potato (Solanum tuberosum) slices with [1-14C]oleic acid and [1-14C]acetate as precursors. In 4-day aged tissue, [1-14C]oleic acid was incorporated into an insoluble residue, which, upon hydrogenolysis (LiA1H4), released the label into chloroform-soluble products. Radio thin layer and gas chromatographic analyses of these products showed that 14C was contained exclusively in octadecenol and octadecene-1, 18-diol. OsO4 treatment and periodate cleavage of the resulting tetraol showed that the labeled diol was octadec-9-ene-1, 18-diol, the product expected from the two major components of suberin, namely 18-hydroxyoleic acid and the corresponding dicarboxylic acid. Aged potato slices also incorporated [1-14C]acetate into an insoluble material. Hydrogenolysis followed by radio chromatographic analyses of the products showed that 14C was contained in alkanols and alkane-α,ω-diols. In the former fraction, a substantial proportion of the label was contained in aliphatic chains longer than C20, which are known to be common constituents of suberin. In the labeled diol fraction, the major component was octadec-9-ene-1,18-diol, with smaller quantities of saturated C16, C18, C20, C22, and C24-α,ω-diols. Soluble lipids derived from [1-14C]acetate in the aged tissue also contained labeled very long acids from C20 to C28, as well as C22 and C24 alcohols, but no labeled ω-hydroxy acids or dicarboxylic acids were detected. Label was also found in n-alkanes isolated from the soluble lipids, and the distribution of label among them was consistent with the composition of n-alkanes found in the wound periderm of this tissue; C21 and C23 were the major components with lesser amounts of C19 and C25. The amount of 14C incorporated into these bifunctional monomers in 0-, 2-, 4-, 6-, and 8-day aged tissue were 0, 1.5, 2.5, 0.8, and 0.3% of the applied [1-14C]oleic acid, respectively. Incorporation of [1-14C]acetate into the insoluble residue was low up to the 3rd day of aging, rapid during the next 4 days of aging, and subsequently the rate decreased. These changes in the rates of incorporation of exogenous oleic acid and acetate reflected the development of diffusion resistance of the tissue surface to water vapor. As the tissue aged, increasing amounts of the [1-14C]acetate were incorporated into longer aliphatic chains of the residue and the soluble lipids, but no changes in the distribution of radioactivity among the α-ω-diols were obvious. The above results demonstrated that aging potato slices constitute a convenient system with which to study the biochemistry of suberization.  相似文献

Nicotiana tabacum is the only plant known to synthesise large quantities of anteiso- (3-methyl) alkanes and iso- (2-methyl) alkanes. We investigated the carbon isotope ratios of individual long-chain n-alkanes, anteiso- and iso-alkanes (in the C29-C33 carbon number range) extracted from tobacco grown in chambers under controlled conditions to confirm the pathway used by the tobacco plant to synthesise these particular lipids and to examine whether environmental data are recorded in these compounds. Tobacco was grown under differing temperatures, water availabilities and light intensities in order to control its stable carbon isotope ratios and evaluate isotopic fractionations associated with the synthesis of these particular lipids. The anteiso-alkanes were found to have a predominant even-carbon number distribution (maximising at C32), whereas the iso-alkanes exhibit an odd-carbon number distribution (maximising at C31). Iso-alkanes were relatively more abundant than the anteiso-alkanes and only two anteiso-alkanes (C30 and C32) were observed.The anteiso-alkanes and iso-alkanes were found to be enriched in 13C by 2.8-4.3‰ and 0-1.8‰ compared to the n-alkanes, respectively, consistent with different biosynthetic precursors. The assumed precursor for the odd-carbon-numbered iso-alkanes is iso-butyryl-CoA (a C4 unit derived from valine) followed by subsequent elongation of C2 units and then decarboxylation. The assumed precursor for even-carbon-numbered anteiso-alkanes is α-methylbutyryl-CoA (a C5 unit derived from isoleucine) and subsequent elongation by C2 units followed by decarboxylation. The ratio of carbon atoms derived from α-methylbutyryl-CoA and subsequent C2 units (from malonyl-CoA) is 1:5 for the biosynthesis of a C30anteiso-alkane. The ratio of carbon atoms derived from iso-butyryl-CoA and subsequent C2 units (from malonyl-CoA) is 4:25 for the synthesis of a C29iso-alkane. An order of 13C depletion n-alkanes > iso-alkanes > anteiso-alkanes is evident from compound specific isotope data. This trend can probably be attributed to the ratio of the two different sources of carbon atoms in the final wax components.Higher water availability generally results in more depleted stable carbon isotope ratios due to maximised discrimination during carboxylation, associated with less diffusional limitation. This was confirmed in the present study by compound specific isotope analyses of iso-alkanes, anteiso-alkanes and n-alkane lipids extracted from the tobacco leaves. Likewise, light intensity has been shown to influence plant bulk δ13C in previous studies. The carbon isotope ratios of n-alkanes in tobacco grown under low-light conditions were about 2‰ more depleted in 13C than those of lipids extracted from tobacco grown under elevated light conditions. A similar order of difference is observed for the iso-alkanes and anteiso-alkanes (1.8‰ and 1.9‰, respectively). A negligible depletion in carbon isotope ratios was observed for the iso-alkanes and anteiso-alkanes extracted from tobacco grown under elevated temperatures. These results are consistent with the work of Farquhar [Farquhar, G.D., 1980. Carbon isotope discrimination by plants: effects of carbon dioxide concentration and temperature via the ratio of intercellular and atmospheric CO2 concentrations. In: Pearman, G.I. [1-14C]Octadecyl glyceryl ether did not label alkanes in the leaves of Brassica oleracea and Pisum sativum while [1-14C]octadecanol and [1-14C]octadecanoic acid readily labeled the alkanes. About 40% of the exogenous-labeled glyceryl ether was incorporated intact into choline phosphatide while 10–20% was converted into fatty acids and alcohols. [1-14C]octadecanol was not converted into alkyl glyceryl ether, but it was oxidized to the corresponding acid and then incorporated into alkanes. These results show that alkyl ether is not an intermediate in alkane biosynthesis. When [1-14C-1-3H]-octadecanol was fed to the leaves of B. oleracea and P. sativum, only the 14C and no 3H was incorporated into alkanes, ketones, and secondary alcohols. These results show that fatty alcohols are first oxidized to the acid before being incorporated into alkanes, ruling out fatty alcohol, alkyl ether, and alk-1-enyl ether as intermediates in alkane biosynthesis. The exogenous alcohols were also readily esterified into wax esters in both tissues.  相似文献

The leaf alkanes of Parthenium argentatum (guayule), P. tomentosum var. stramonium, P. fruticosum var. trilobatum, and the first filial (F1) generations obtained from crosses with guayule were investigated by GC and mass spectrometry and shown to be useful in chemotaxonomic studies. The identified n-alkanes ranged from C19 to C40 with either n-C29 or n-C31 as the main component. The alkane chemistry of guayule with n-C31 being the main component predominated in most of the F1 hybrids. The presence of iso-branched alkanes (C27, C29, C31) in P. tomentosum and its hybrids could be detected by GC/MS. These preliminary investigations indicate that epicuticular wax alkanes can be useful in inheritance studies of guayule and its hybrids.  相似文献

Epicuticular hydrocarbons of Drosophila pseudoobscura were analyzed by gas chromatography and mass spectrometry. Methyl-branched alkanes and alkadienes were the predominant hydrocarbons, with lesser amounts of monoenes (14%) and trienes (9%) also present. Alkanes (49%) were mostly odd carbon number 2-methylalkanes (C25–C31). Alkadienes (27%) were odd carbon number components (C25–C33), with the (Z,Z)-5,9-isomer predominating. Monounsaturated hydrocarbons were a mixture of 5-, 7-, 9-, 11-, 13-, 14- and 15-isomers containing 25–33 carbon atoms. The major alkatriene components contained 29–31 carbon atoms and were either 5,19,17- or 5,9,19-isomers. Sodium[1-14C]acetate was incorporated into each class of hydrocarbon and into each of the major alkadienes.  相似文献

Material from the testa of decorticated barley grains contained hydrocarbons, esters, triglycerides, free sterols, 5-n-alkylresorcinols, and traces of free alcohols, carbonyl compounds, and various polar, acidic materials. The hydrocarbon fraction was mainly a series of n-alkanes, extending at least from C11 to C36, in which the C29 and C31 components were prominent. Two minor series of alkanes were also present. Sometimes a trace of an unsaturated hydrocarbon was detected. The ester fraction contained sterols and alkanols esterified by fatty acids, which differed in relative amounts from the fatty acids found in the triglycerides. The triglycerides were thought to have leached from within the grain. At least five free sterols were present, including sitosterol and campesterol. The 5-n-alkylresorcinols were at least twelve members of a homologous series, of which four, C25, C27, C29, and C31, made 98% of the total. Members of the series with even numbers of carbon atoms were also present. It is suggested that they are partly responsible for excluding microorganisms from the interior of the grain. The testa membrane, with the associated pigment strand, contained an estolide of fatty acids and various hydroxyacids, a polysaccharide component, and uncharacterized material.  相似文献

Surface alkanes were extracted from thalli of populations ofXanthoria parietinagrowing in two different Piedmont (Italy) valleys: Susa and Vermenagna. The mainn-alkanes detected in the Susa Valley were C27, C28, C29and C31, whereas C25, C27and C29were the most abundant in Vermenagna Valley. The results indicate that then-alkane qualitative composition ofX. parietinawas affected both by elevation and climatic characteristics typical of the two valleys considered.  相似文献

Sodium [1-14C]acetate and [1-14C]stearic acid were readily incorporated into hydrocarbons, secondary alcohols, wax esters, aldehydes, primary alcohols, and fatty acids in young pea leaves (Pisum sativum). Dithioerythritol, dithiothreitol, and mercaptoethanol (but not glutathione and cysteine) severely inhibited the incorporation of labeled acetate into alkanes and secondary alcohols with accumulation of label in wax ester and aldehyde fractions. Detailed radio gas-chromatographic analyses of the fatty acids of both the surface lipid components and internal lipids showed that dithioerythritol and mercaptoethanol specifically inhibited n-hentriacontane (C31) synthesis and caused accumulation of C32 aldehyde, suggesting that the inhibition was at or near the terminal step in alkane biosynthesis, presumably decarboxylation. Trichloroacetate, at a concentration that inhibited C31 alkane synthesis but not the synthesis of alcohols (C26 and C28) specifically inhibited the formation of C32 aldehyde but not that of the C26 or C28 aldehyde. From these results, it is concluded that the C32 aldehyde is derived from the C32 acyl derivative which is the precursor of C31 alkane.  相似文献

The biosynthetic relations between protoberberine-, benzo[C]phenanthridine- and B-secoprotoberberine type alkaloids were demonstrated by use of (±)-tetrahydrocoptisine-[8,14-3H HCl, (±)-tetrahydrocorysamine-[8,14-3H]HCl and corynoline-[6-3H]HCl in Corydalis incisa, and the following results were presented. (±)-Tetrahydrocoptisine was converted to corynoline, corydalic acid methyl ester and corydamine hydrochloride. (±)-Tetrahydrocorysamine was converted to corynoline and corydalic acid methyl ester. Evidence that N-methyl-3-[6′-(3′,4′-methylenedioxyphenethylalcohol)]-4-methyl-7,8-methylenedioxy-1,2,3,4-tetrahydroisoquinoline-[α-3H] HCl was incorporated into corynoline-[11-3H] indicates the occurrence of the ring fission at C6-N followed by linking ofthe C6 and C13 positions in (±)-tetrahydrocoptisine and (±)-tetrahydrocorysamine, and suggests the participation of one of two possible intermediates in the biosynthesis of these alkaloids.  相似文献

In isolated tobacco leaves l-valine-U-14C gave rise to labeled even-numbered isobranched fatty acids containing 16 to 26 carbon atoms and iso C29, iso C31, and iso C33 paraffins. l-Isoleucine-U-14C on the other hand produced labeled odd-numbered anteiso C17 to C27 fatty acids and anteiso C30 and C32 paraffins. Trichloroacetic acid inhibited the incorporation of isobutyrate into C20 and higher fatty acids and paraffins without affecting the synthesis of the C16 and C18 fatty acids. Thus the very long branched fatty acids are biosynthetically related to the paraffins. In Senecio odoris leaves acetate-1-14C was incorporated into the paraffins (mainly n-C31) only in the epidermis although acetate was readily incorporated into fatty acids in the mesophyll tissue. Similarly only the epidermal tissue incorporated acetate into fatty acids longer than C18 suggesting that the epidermis is the site of synthesis of both paraffins and the very long fatty acids. In broccoli leaves n-C12 acid labeled with 14C in the carboxyl carbon and 3H in the methylene carbons was incorporated into C29 paraffin without the loss of 14C relative to 3H. Since n-C18 acid is known to be incorporated into the paraffin without loss of carboxyl carbon these results suggest that the condensation of C12 acid with C18 acid is not responsible for n-C29 paraffin synthesis in this tissue. Thus all the experimental evidence thus far obtained strongly suggests that elongation of fatty acids followed by decarboxylation is the most likely pathway for paraffin biosynthesis in leaves.  相似文献

Cutin, the structural component of plant cuticle, is a polymer of C16 and C18 hydroxy fatty acids. Previous results have suggested that oleic acid undergoes ω-hydroxylation, epoxidation of the double bond, and, finally, hydration of the epoxide to give rise to the three major components of the C18 family of cutin acids. 18-Hydroxy [18-3H]oleic acid and 18-hydroxy-9,10-epoxy[18-3H]stfaric acid have been synthesized and, with these synthetic substrates, the conversion of 18-hydroxyoleic acid to 18-hydroxy-9,10-epoxystearic acid and the hydrolysis of 18-hydroxy-9,10-epoxystearic acid to 9,10,18-trihydroxystearic acid were directly demonstrated in apple fruit skin and in the leaves of apple and Senecio odoris. Trichloropropene oxide, an inhibitor of microsomal epoxide hydrases of animals, specifically inhibited the conversion of [1-14C]oleic acid into 18-hydroxy-9,10-epoxystearic acid and 9,10,18-trihydroxystearic acid, while it had no effect on the conversion of [1-14C]palmitic acid into hydroxylated palmitic acid, a process which does not involve epoxy acid intermediates. Therefore, it appears that this inhibitor affects epoxidation and or epoxide hydration steps involved in cutin biosynthesis.  相似文献

Stem cutin from P. radiata seedlings grown under winter and summer environmental conditions comprised n-alkanoic, (C10–C26), α, ω-alkanedioic (C14–C22), ω-hydroxyalkanoic (C12–C24), hydroxy-α, ω-alkanedioic and polyhydroxyalkanoic acids. 9-Hydroxyheptadecane-1, 17-dioic, 9-hydroxyoctadecene-1, 18-dioic, 9-hydroxynonadecane-1, 19-dioic, and 10, 17-dihydroxyheptadecanoic acids are newly-identified constituents of gymnosperm cutin. Cutin grown under winter temperatures and photoperiod contained twice the amount of 9, 16-dihydroxyhexadecanoic acid than that in summer-grown cutin, suggesting that the winter-grown cutin was formed from a highly cross-linked polymer, and that summer-grown cutin contained more linear polyester portions in the polymer.  相似文献

The hexane extract of Wyethia mollis contains the n-alkanes C15-C18, C20-C25, C27 and C29. Linoleic acid was the only detectable acidic component. A mass spectral analysis of the wax ester fraction indicated that it was a mixture of homologues, the saturated even-carbon acids n-C16-C30 esterfield with the saturated even-carbon alcohols n-C18-C26. The chloroform extract yielded the known isoflavones santal and 3′-O-methylorobol along with a new lanostane-type triterpene, 22,25-epoxy-lanosta-7:9(11)-dien-3-one. The wide distribution of n-alkanes and the decreased odd-even carbon ratio are consistent with the proposed primitive nature of this plant.  相似文献

The hydrocarbon constituents of the leaf waxes of eight species of Khaya were analysed for taxonomic purposes using GLC. The leaf waxes contained neither isoalkanes nor alkanes and the bulk of the n-alkanes were in the range of C25 to C33, odd-carbon number compounds predominating. It was found that the percentage composition of the n-alkane constituents of the leaf waxes varied with the age of leaves, young leaves having n-C29 as the most abundant alkane, whereas older leaves had n- C31.  相似文献

Sodium [1-14C]acetate, sodium [1-14C]propionate, sodium [2-14C]propionate, sodium [3-14C]propionate and sodium [methyl-14C]methylmalonate were readily incorporated into the cuticular hydrocarbons of nymphal stages of the cockroach Periplaneta fuliginosa both in vivo and in vitro, whereas no incorporation of [methyl-14C]methionine was observed. The alkanes of the nymphal stages of this insect are 25+% n-alkanes, 14% 3-methylalkanes, and 59+% internally branched monomethylalkanes, principally 13-methylpentacosane. Sodium [1-14C]acetate was incorporated into each class of alkane at about its percentage composition. In contrast, labeled sodium propionate and sodium methylmalonate were preferentially incorporated into the branched fractions. Radio-gas-liquid chromatography showed that sodium [1-14C]propionate was incorporated almost exclusively into 3-methyltricosane and 13-methylpentacosane, whereas sodium [1-14C]acetate was incorporated into each glc peak at about its percentage composition. These data suggest that propionate, incorporated during chain elongation, serves as the branching methyl group donor for both the 3-methyl and the internally branched monomethylalkanes in insects. The location of hydrocarbon synthesis in P. fuliginosa was studied using an in vitro tissue slice system. Excised cuticle slices, with adhering fat body tissue removed, gave good incorporation of labeled substrates into the hydrocarbon fraction. No hydrocarbon synthesis was observed in fat body preparations.  相似文献

Gas chromatography-mass spectrometry and nuclear magnetic resonance spectroscopy were used to study the metabolism of deuterated n-alkanes (C6 to C12) and 1-13C-labeled n-hexane by a highly enriched sulfate-reducing bacterial culture. All substrates were activated via fumarate addition to form the corresponding alkylsuccinic acid derivatives as transient metabolites. Formation of d14-hexylsuccinic acid in cell extracts from exogenously added, fully deuterated n-hexane confirmed that this reaction was the initial step in anaerobic alkane metabolism. Analysis of resting cell suspensions amended with 1-13C-labeled n-hexane confirmed that addition of the fumarate occurred at the C-2 carbon of the parent substrate. Subsequent metabolism of hexylsuccinic acid resulted in the formation of 4-methyloctanoic acid, and 3-hydroxy-4-methyloctanoic acid was tentatively identified. We also found that 13C nuclei from 1-13C-labeled n-hexane became incorporated into the succinyl portion of the initial metabolite in a manner that indicated that 13C-labeled fumarate was formed and recycled during alkane metabolism. Collectively, the findings obtained with a sulfate-reducing culture using isotopically labeled alkanes augment and support the previously proposed pathway (H. Wilkes, R. Rabus, T. Fischer, A. Armstroff, A. Behrends, and F. Widdel, Arch. Microbiol. 177:235-243, 2002) for metabolism of deuterated n-hexane by a denitrifying bacterium.  相似文献

The cutins of fruits and leaves of four apple cultivars have been analysed using TLC, GLC and GC-MS. They are similarly composed of saturated, monounsaturated and diunsaturated fatty, hydroxy-fatty and epoxyhydroxy-fatty acids. The most abundant monomers are 18-hydroxyoctadeca-9,12-dienoic, 10,16-dihydroxyhexadecanoic, 9,10-epoxy-18-hydroxyoctadec-12-enoic, 9,10-epoxy-18-hydroxyoctadecanoic and 9,10,18-trihydroxyoctadecanoic acids. The fruit cutins have high contents of epoxides (35–40%) and unsaturated components ( > 40%) and C18 compounds predominate over C16. The leaf cutins contain smaller amounts of unsaturated components than the fruits and higher proportions of C16 compounds. The adaxial leaf cutin differs in composition from the abaxial. 10,16-Dihydroxyhexadecanoic and 9,10-epoxy-18-hydroxoctadecanoic acids are the major constituents (each ca. 30%) of the adaxial leaf cutin and 10,16-dihydroxyhexadecanoic acid (55–65%) predominates in the abaxial.  相似文献

The composition of the n-alkanes in the leaf waxes of over 80 clones of Saccharum officinarum, S. edule, S. robustum, S. spontaneum and from a number of related species have been compared by GLC. The waxes contain predominantly odd alkanes, C27–C35, the major components being C29 and C31. In a number of clones, particularly of S. edule, a homologous series of alkenes was also present. No chemotaxonomic relationship could be derived from the compositions as the intraspecific variation was greater than the interspecific variations.  相似文献

Rainbow trout leucocytes contain high levels of neutral lipid (about 70% of total lipid on a wt% basis) consisting of mostly triacylglycerol, free sterols and sterol esters (25%, 15% and 52% of neutral lipid, respectively). The phospholipids, separated by thin-layer chromatography, consisted predominantly of phosphatidylcholine, phosphatidylethanolamine and phosphatidylserine, each present at about 30% of the total phospholipid. Radiolabelling of the leucocytes for 1 h with 1 μCi (approx. 6 μM) [1−14C]20:4(n−6), [1−14C]20:5(n−3) or [1−14C]22:6(n−3) each gave similar uptake values (approx. 1 · 105 cpm/107 leucocytes). The incorporation into total phospholipids was highest for 22:6(n−3) and lowest for 20:4(n−6). A higher percentage of radiolabel from [1−14C]22:6(n − 3) was found incorporated into phosphatidylcholine and phosphatidylethanolamine as compared to that from [1−14C]20:4(n − 6) and [1−14C]20:5(n−3), while the reverse situation was found with phosphatidylinositol and phosphatidylserine. The relative rates of incorporation into the different phospholipid classes for all three fatty acids were in the order phosphatidylinositol > sphingomyelin > diphosphatidylglycerol > phosphatidylcholine > phosphatidylethanolamine > phosphatidylserine. Calcium ionophore-challenge did not significantly alter the pattern of phospholipid radiolabel. Ionophore-challenge released large amounts of radiolabel, much of which was recovered after high-performance liquid chromatographic separation as free fatty acid/monohydroxy fatty acids, although only approx. 0.3% was recovered in leukotriene B4 and leukotriene B5 for the [1−14C]20:4(n−6) and [1−14C]20:5(n−3) labelled leucocytes, respectively. Other lipoxygenase products were also radiolabelled and tentatively identified as 20-carboxy-LTB4, 20-hydroxy-LTB4, 6-trans-LTB4, 6-trans-12-epi-LTB4, 6-trans-8-cis-12-epi-LTB4 and the corresponding LTB5 structures. No ‘6-series’ leukotrienes were produced from [1−14C]22:6(n−3), nor was there any evidence for the synthesis of ‘5-series’ leukotrienes via retroconversion of 22:6(n−3) to 20:5(n−3). This latter finding shows that, despite the preponderance of 22:6(n−3) in the membranes of trout leucocytes, this fatty acid is not a substrate for leukotriene generation.  相似文献

We describe the use of carbon stable isotope (13C) labeled n-alkanes as a potential internal tracer to assess passage kinetics of ingested nutrients in ruminants. Plant cuticular n-alkanes originating from intrinsically 13C labeled ryegrass plants were pulse dosed intraruminally in four rumen-cannulated lactating dairy cows receiving four contrasting ryegrass silage treatments that differed in nitrogen fertilization level (45 or 90 kg nitrogen ha−1) and maturity (early or late). Passage kinetics through the gastrointestinal tract were derived from the δ13C (i.e. the ratio 13C:12C) in apparently undigested fecal material. Isotopic enrichment was observed in a wide range of long-chain n-alkanes (C27–C36) and passage kinetics were determined for the most abundant C29, C31 and C33 n-alkanes, for which a sufficiently high response signal was detected by combustion isotope ratio mass spectrometry. Basal diet treatment and carbon chain length of n-alkanes did not affect fractional passage rates from the rumen (K 1) among individual n-alkanes (3.71–3.95%/h). Peak concentration time and transit time showed a quantitatively small, significant (p≤0.002) increase with carbon chain length. K 1 estimates were comparable to those of the 13C labeled digestible dry matter fraction (3.38%/h; r = 0.61 to 0.71; p≤0.012). A literature review has shown that n-alkanes are not fermented by microorganisms in the rumen and affirms no preferential depletion of 13C versus 12C. Our results suggest that 13C labeled n-alkanes can be used as nutrient passage tracers and support the reliability of the δ13C signature of digestible feed nutrients as a tool to measure nutrient-specific passage kinetics.  相似文献
