Regulation of Auxin Levels in Coleus blumei by Ethylene   总被引:3,自引:9,他引:3       下载免费PDF全文
An investigation of the effects of ethylene pretreatment on several facets of auxin metabolism in Coleus blumei Benth “Scarlet Rainbow” revealed a number of changes presumably induced by the gas. Transport of indoleacetic acid-1-14C in excised segments of the uppermost internode was inhibited by about 50%. Decarboxylation of indoleacetic acid-1-14C by enzyme breis was not affected by the pretreatment. Levels of extractable native auxin in upper leaf and apical bud tissue of the pretreated plants were approximately one-half of those present in untreated plants. The rate of formation of auxin from tryptophan by enzyme breis from pretreated plants was approximately one-half that occurring in incubation mixtures containing the enzyme system from untreated plants. The conjugation of indoleacetic acid-1-14C in a form characterized chromatographically as indoleacetylaspartic acid was increased 2-fold in the upper stem region of plants pretreated with ethylene.  相似文献

Movement of lucifer yellow in leaves of Coleus blumei Benth   总被引:1,自引:3,他引:1  
Abstract Individual spongy mesophyll cells in green areas of variegated Coleus blumei leaves were injected with the symplast-mobile dye lucifer yellow and its movement to other cell types was followed with fluorescence microscopy. In 13 trials, the dye remained in the injected cell twice, moved only to other mesophyll cells five times, and moved up to and along minor veins six times. Where extensive movement of the dye occurred, the tissue was fixed with 4% glutaraldehyde, dehydrated, embedded in plastic, sectioned at 3 μm, and examined again with fluorescence microscopy. The dye was found in abaxial bundle-sheath cells for up to 200 μm or more distant from the site of injection near the minor vein, but no convincing evidence was found for its presence in the vascular tissue itself. It thus appears that superficial whole-mount views of lucifer yellow movement along leaf minor veins cannot be taken as certain evidence for symplastic transport of the dye into and along the vascular tissues.  相似文献

In order to study the accumulation and transport of rosmarinic acid in suspension cells of Coleus blumei we established an efficient method to isolate protoplasts and vacuoles. Protoplasts were disrupted by an osmotic shock in a medium with basic pH containing ethylenediamine tetraacetic acid. The resulting vacuoles were purified on a two-step Ficoll gradient. The comparison of the rosmarinic acid contents of cells, protoplasts and vacuoles showed that the depside is localized in the vacuole. Data concerning the yield and purity of the vacuoles are presented. In addition we show that at the physiological pH of the cytoplasm rosmarinic acid is present almost exclusively as an anion and cannot pass a membrane by simple diffusion. Rosmarinic acid synthase from cell cultures of Coleus blumei Benth. was purified to apparent homogeneity by fractionated ammonium sulfate precipitation (60–80% saturation), hydrophobic interaction chromatography, affinity chromatography and gel filtration. This purification procedure resulted in a 225-fold-enriched specific enzyme activity with a yield of 9%. The protein preparation was apparently pure according to sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and two-dimensional gel electrophoresis. Mature, variegated leaves of Coleus blumei Benth. contained stachyose and other raffinose series sugars in both green, photosynthetic and white, nonphotosynthetic tissues. However, unlike the green tissues, white tissues had no detectable level of galactinol synthase activity and a low level of sucrose phosphate synthase indicating that stachyose and possibly sucrose present in white tissues may have originated in green tissues. Uptake of exogenously supplied [14C]stachyose or [14C]sucrose into either tissue type showed conventional kinetic profiles indicating combined operation of linear first-order and saturable systems. Autoradiographs of white discs showed no detectable minor vein labelling with [14C]stachyose, but some degree of vein labeling with [14C]sucrose. Autoradiographs of green discs showed substantial vein loading with either sugar. In both tissues, p-chloromercuribenzenesulfonic acid had no effect on the linear component of sucrose or stachyose uptake but inhibited the saturable component. Both tissues contained high levels of invertase, sucrose synthase and α-galactosidase and extensively metabolized exogenously supplied 14C-sugars. In green tissues, label from exogenous sugars was recovered as raffinose-series sugars. In white tissues, exogenous sugars were hydrolysed and converted to amino acids and organic acids. The results indicate that variegated Coleus leaves may be useful for studies on both phloem loading and phloem unloading processes in stachyose-transporting species.  相似文献

Excised shoots and potted plants of Coleus blumei Benth, were subjected to different treatments with kinetin solutions. Control treatments were made with water. Free and bound auxin were extracted with ether. The acid fractions were purified by electrophoresis to remove all traces of kinetin, and were then analyzed with the Avena straight-growth test. As compared with the controls, kinetin treatment increases the bound auxin, whereas the corresponding free auxin is unchanged. Also methanol extracts of treated stems contain more acid auxin than corresponding extracts of water-treated stems. This indicates that on methanol extraction not only free auxin but also some bound auxin is obtained. The extracted auxin behaves like indoleacetic acid in paper chromatography with four different solvent systems, as well as in gel filtration through Sephadex. New formation or decreased breakdown of auxin seem the most likely explanations for the observed effects of kinetin.  相似文献

The purpose of this study was to analyse the accumulation of amino acid in source and sink tissues of variegated Coleus blumei Benth. leaves during an extended exposure to salinity stress. The imposed stress resulted in a reduction in shoot height and leaf size, as well as a reduction in total protein and nitrogen content in both the sink and source tissues. At the same time, accumulation of low molecular weight nitrogen-containing compounds in Coleus leaves was observed, which peaked within the first 10 d of exposure to salinity, and then declined, but remained slightly elevated for the remainder of the study. A number of amino acids were found to accumulate in both the sink and source tissues, including arginine, asparagine, and serine. A larger proportion of asparagine and less arginine was observed in the sink tissue than the source tissue of the salinity-stressed plants. This difference may reflect the mobility of these compounds in the phloem. No proline was found to accumulate in either the source or sink tissue during the exposure to salinity. From the pulse-chase labelling of stressed Coleus leaves it can deduced that some of the observed accumulation of amino acids and amides observed is due to de novo synthesis and not simply the result of protein degradation.  相似文献

The nonchlorophyllous (albino) tissue of mature C. blumei leaves is a sink for photoassimilate. Transport from the green to the albino region of the same leaf was inhibited by cold and anoxia. When the green tissue of mature leaves was removed, the remaining albino portion imported labeled translocate from other mature leaves in the phloem. Photoassimilate unloading in the albino region of mature leaves was studied by quantitative autoradiography. The unloading was inhibited by cold but not by anoxia. No labeled photoassimilate could be detected in the free space of mature albino tissue by compartmental efflux analysis as phloem unloading proceeded in a N2 atmosphere, indicating that unloading, may occur by a symplastic pathway as it apparently does in sink leaves of other species. The minor veins of mature albino leaf tissue did not accumulate exogenous [14C]sucrose. Minor veins of green tissue in the same leaves accumulated [14C]sucrose but, in contrast to other species studied to date, this accumulation was insensitive to the inhibitor p-chloromercuribenzensulfonic acid (PCMBS).In its capacity to import and unload photoassimilate, and in the inability, of the minor veins to accumulate exogenous sucrose, the albino region of the mature C. blumei lamina differs from mature albino tobacco leaves and darkened mature leaves of other species. This, together with evidence indicating that phloem loading in C. blumei and other species may occur by different routes and with different sensitivity to PCMBS, indicates that the mechanism of transfer of photoassimilates between veins and surrounding tissues, and the mechanism of the sink-source transition, may not be the same in the leaves of all species. It is speculated that the unusual properties of the C. blumei leaf may be a consequence of the presence, in the minor veins, of intermediary cells, large companion cells connected to the bundle sheath by abundant plasmodesmata.Abbreviation PCMBS p-chloromercuribenzenesulfonic acid  相似文献

A biosynthetic pathway for rosmarinic acid is proposed. This pathway is deduced from studies of the enzymes detectable in preparations from suspension cells of Coleus blumei. Phenylalanine is transformed to 4-coumaroyl-CoA by the enzymes of the general phenylpropanoid pathway: phenylalanine ammonia-lyase (EC 4.3.1.5), cinnamic acid 4-hydroxylase (EC 1.14.13.11) and hydroxycinnamic acid:CoA ligase (EC 6.2.1.12). Tyrosine is metabolized to 4-hydroxyphenyllactate by tyrosine aminotransferase (EC 2.6.1.5) and hydroxyphenylpyruvate reductase. The ester can be formed from 4-coumaroyl-CoA and 4-hydroxyphenyllactate by the catalytic activity of rosmarinic acid synthase with concomitant release of CoA. The photosynthetic tissue of green portions of variegated Coleus blumei leaves consists primarily of palisade and spongy parenchyma cells as well as bundle-sheath cells. The moderate numbers of plasmodesmata connecting these cells may be sufficient to provide a symplastic pathway for assimilates moving toward the minor veins. The minor veins, however, are unusual in having two sets of phloem-loading cells which have little symplastic continuity with one another: one consisting of large, peripherally located intermediary cells, and a second set made up of smaller, usually more internal companion cells, both sets having their associated sieve-tube members. The intermediary cells are connected to vascular-parenchyma and bundle-sheath cells by unique branched plasmodesmata which are particularly abundant at the bundle-sheath interface. In addition, numerous plasmodesmata-pore connections occur between the intermediary cells and their associated sieve-tube members. Neither the intermediary cells nor their sieve-tube members plasmolyze when treated with 1.4 M mannitol, whereas mesophyll and vascular-parenchyma cells plasmolyze at 0.5 M and bundle-sheath cells at 0.6 M mannitol. By contrast, the companion cells and their associated sieve-tube members are symplastically isolated from the bundle-sheath cells and the sieve-tube-intermediary-cell complexes, and share few plasmodesmata with the vascular-parenchyma cells. Moreover, the companion cells plasmolyze at 1.1 M mannitol and their sieve tubes at 1.3 M. A method is described for the estimation of the rate of movementand the quantity transported of the natural growth hormone instandard isolated segments of apple shoots. During controlledstorage diffusible auxin is collected, and later by dividingthe standard length of stem into small sections the locationof the auxin front is determined, from which the rate of transportis deduced. Temperature markedly affects both rate of transportand amount of auxin transported (cf. van der Weij, 1932), amaximum occurring at 27–30° C.; followed by a rapidfall to zero. The total diffusible auxin in a given length ofstem is not affected by storage temperatures below 30° C.but falls to zero at 42° C. The rate of transport and amounttransported are proportional to the oxygen tension over therange 0 to 5 per cent. O2, and there is some evidence for destructionof auxin in tensions below 2 per cent.  相似文献

Transport of indoleacetic acid-114C following application to the buds of intact white ash (Fraxinus americana L.) shoots proceeds at a velocity of about 1.3 centimeters per hour in actively growing seedlings, but only 0.3 centimeter per hour in dormant seedlings. The rapid movement is metabolically controlled, and at 1 C or in a nitrogen environment it is reduced to 0.2 centimeter per hour, suggesting that the slower movement is due to diffusion. The transport profile for growing shoots shows a logarithmic decrease in activity in stems treated for 3 hours. However, over longer treatment intervals, especially after 12 hours, a steady state of recoverable activity occurs in the more basal stem segments. Cold-treated shoots acquire the capacity for rapid transport 7 days after they are placed into favorable growing conditions, at which time dormancy callose disappears from the phloem, respiratory activity of the stem tissue increases, and mitotic reactivation occurs in the bud. Following shoot reactivation, the velocity and amount of exogenously supplied indoleacetic acid transported remained relatively uniform until the onset of the succeeding dormant period. Five per cent, or less, of the applied tracer moves into the shoot, with substantial portions remaining as indoleacetic acid.  相似文献

The hypothesis that auxin (IAA) and gibberellic acid (GA3) control the formation of lignin is confirmed for the primary phloem fibers and for the secondary xylem in the stem of Coleus blumel Benth. Indoleacetic acid alone, or a combination of high IAA/low GA3 (w/w), induced short phloem fibers with thick secondary walls, that contained lignin rich in syringyl units (high ratio of syringyl/guaiacyl). On the other hand, a combination of high GA3/low IAA (w/w), which promoted the differentiation of long phloem fibers with thin walls, decreased the relative content of the syringyl units (low syringyl/guaiacyl ratio). In the secondary xylem, these hormonal treatments yielded only slight changes in the noncondensed monomeric guaiacyl units, confirming the relative stability of the guaiacyl lignification pattern in this tissue. In the xylem, indoleacetic acid alone, or a combination of high IAA/low GA3 induced lignin poor in syringyl units (low syringyl/guaiacyl ratio). A combination of high GA3/low IAA promoted a relatively slight increase in syringyl yield, indicating greater responsiveness of the syringyl lignification pattern to growth regulators. Sugar-synthesis and -transport patterns were analyzed in Coleus blumei Benth. leaves to determine where galactinol, raffinose, and stachyose are made and whether phloem loading includes an apoplastic (extracellular) step or occurs entirely within the symplast (plasmodesmata-connected cytoplasm). To clarify the sequence of steps leading to stachyose synthesis, a pulse (15 s) of 14CO2 was given to attached leaves followed by a 5-s to 20-min chase: sucrose was rapidly labeled while galactinol, raffinose and stachyose were labeled more slowly and, within the first few minutes, to approximately the same degree. Leaf tissue was exposed to either 14CO2 or [14C]glucose to identify the sites of synthesis of the different sugars. A 2-min exposure of peeled leaf tissue to [14C]glucose resulted in preferential labeling of the minor veins, as opposed to the mesophyll; galactinol, raffinose and stachyose were more heavily labeled than sucrose in these preparations. In contrast, when leaf tissue was exposed to 14CO2 for 2 min for preferential labeling of the mesophyll, sucrose was more heavily labeled than galactinol, raffinose or stachyose. We conclude that sucrose is synthesized in mesophyll cells while galactinol, raffinose and stachyose are made in the minorvein phloem. Competition experiments were performed to test the possibility that phloem loading involves monosaccharide uptake from the apoplast. Two saturable monosaccharide carriers were identified, one for glucose, galactose and 3-O-methyl glucose, and the other for fructose. Washing the apoplast of peeled leaf pieces with buffer or saturating levels of 3-O-methyl glucose, after providing a pulse of 14CO2, did not inhibit vein loading or change the composition of labeled sugars, and less than 0.5% of the assimilated label was recovered in the incubation medium. Plant cell culture of Coleus blumei when entrapped in temperature-sensitive hydrogel beads [poly(N-isopropylacrylamide) containing 0.5% alginate], retained viability during the three weeks culture. Treatment with dimethipin (2,3-dihydro-5,6-dimethyl-1,4-dithiin 1,1,4,4 tetroxide) inhibited the increase in cellulase activity and decrease in breakstrength associated with the normal course of abscission in Coleus. Application of the surfactant UBI-1126 (Emery OAL 20 in isopropyl alcohol) increased cellulase activity and accelerated the process of abscission in Coleus expiants within 24 h of application. Cellulase activity was localized histochemically at the electron microscopic level in surfactant-treated tissue. The enzyme activity was localized primarily in the cell wall, middle lamella, and paramural bodies of abscission zone cells.  相似文献

A rapid and highly-effective method for micropropagation from nodal segment and shoot tip explants was established for Coleus blumei Benth. Nodal segments and shoot tips were inoculated on MS medium containing 0.7 % agar, 3 % commercial sugar, and different combinations of 6-benzyladenine (BA) with indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) or α-naphthaleneacetic acid (NAA). Hundred percent shoot induction from both explants was achieved on the medium containing BA (2 mg dm−3) and NAA (1 mg dm−3). Shoot tips were proved to be the better explant in comparison to nodal segments in having high rate of shoot induction and more number of shoots. The same media conditions were found suitable for shoot multiplication. Multiplied shoots rooted best on MS medium supplemented with IBA (2 mg dm−3). Micropropagated plants were successfully established in soil after hardening, with 100 % survival rate.  相似文献
