The thermal dependence of enzyme kinetic parameters has beenpresented as an indicator of species’ thermal optima andtolerance limits. Previous studies suggest the relationshipbetween temperature and the apparent Michaelis–Mentenconstant (Km) of an enzyme system can be used to predict wholeplant success at specific temperatures. The apparent Kmfor glutathionereductase (EC 1.6.4.2; GR) (oxidized glutathione as substrate)extracted from leaves of American sloughgrass (Beckmannia syzigachneSteud.), tufted hairgrass (Deschampsia caespitosa L.), tallfescue (Festuca arundinaceae Schreb. ‘Titan’), andmaize (Zea mays L.), was determined over a range of temperatures(1–40 °C). For all species, minimum apparent KmforGR was observed at 1 °C, and Kmvalues increased as temperatureincreased. The apparent Kmvalues differed among all speciesat the lower temperatures (1–15 °C), but were similarat higher temperatures. The enzyme from tufted hairgrass hadthe lowest apparent Kmat low temperatures (<15 °C), followedin increasing order by American sloughgrass, tall fescue andmaize. Our experimental system failed to reproduce thermal kineticwindow profiles similar to those reported elsewhere. With respectto the enzyme systems reported here, results suggest that thesecool-season grasses can be ranked as more to less eurythermicwithin the temperature range from 1 to 15 °C. The characteristics of glutathione reductase from a number ofmaize cultivars with contrasting thermotolerance have been investigated.The apparent Km (Michaelis constant) for oxidised glutathione(GSSG) was measured between 10 and 45°C at constant pH.The enzyme from highland cultivars adapted to cool environmentshad a slightly lower apparent Km for GSSG than that from lowlandtropical cultivars at low assay temperatures. Similarly theenzyme from lowland tropical cultivars had a lower apparentKm for GSSG at high assay temperatures. However these effectswere small and regression lines plotted through the data werenot significantly different in slope or intercept. There wasa strong correlation (r = 0·939) between apparent Kmand Vmax (maximum initial velocity) as assay temperature wasvaried. The interpretation of apparent Km/temperature relationshipsis discussed with hypothetical examples of the effects of temperatureon enzyme activity/substrate concentration plots. It is demonstratedthat an increase in apparent Km at higher assay temperaturesneed not necessarily reflect any temperature-dependent impairmentof enzyme function. The apparent Km for GSSG of glutathionereductase from maize increased over four-fold when the temperaturewas raised from 10 to 45°C, but it is concluded that invivo rates of reaction are likely to be increased rather thandecreased by this same change in temperature. Glutathione reductasewould thus appear to be equally well adapted to function atall these temperatures. The lacustrine blue-green alga (= Cyanobacterium) Oscillatoriarubescens D.C. was cultured in chemostats with temperature andlight intensity as the only limiting factors. The experimentswere carried out under combinations of three temperatures 10,20 and 30°C and three irradiances 6, 18 and 30 µEm–2 s–1. Dilution rate, photosynthesis rate anda large number of abiotic and cellular factors were regularlymeasured to determine steady-state periods and the level ofthe associated variables. Various mathematical models were fittedwith series of data by a non-linear regression method derivedfrom Marquardt's method. Four models for calculation of specificgrowth rate and photosynthesis rate are presented. The lastone computes growth rate from calculated photosynthesis: µ= µmax pmaxI/[KpKi + I(Kp + Pmax)], with Pmax =100 + 257Tand K1 = 300/chl a. Values of adjusted parameters are discussed.One conclusion confirms that values of maximal growth ratesand half-saturation constants frequently considered in the literatureas absolute species characteristics should always be consideredas relative to associated prevalent growth conditions.  相似文献

Growth (assessed from intermolt period and molt increment) andmetabolism (oxygen consumption) of the post-larva of Euphausiapacifica from the southern Japan Sea were determined at sevengraded temperatures ranging from 1 to 25°C. The intermoltperiod shortened progressively as temperature increased from1 to 20°C, but an effect of temperature on molt-to-moltgrowth increment was not seen. Oxygen consumption rates wereaccelerated by the increase in temperature up to 20°C. Beyond20°C, E.pacifica exhibited reduced oxygen consumption anddied within 1 day without molting. After removing the effectof body size, the relationships between growth rate and temperature,and between oxygen consumption rate and temperature, were established.The carbon budget was calculated as a function of temperature.Because of differential effects of temperature on growth andmetabolism, the net growth efficiency [K2 growthx100/(growth+metabolism)]changed with temperature. The optimum temperature at which E.pacificaattained the maximum K2 was 11.4°C, which was derived fromcalculation of cumulative carbon invested in growth and metabolismin this animal. In an alternative method, the optimum temperaturewas obtained mathematically by solving a set of differentialequations. The biological and ecological significance of theoptimum temperature which leads to the maximum K2 is discussed.  相似文献

Seeds of two lettuce cultivars (Lactuca sativa L., cv. Meikoninginand cv. Grand Rapids) were hermetically stored with constantmoisture contents ranging between 3.6 and 17.9 per cent (freshweight basis) at constant temperatures ranging between 5 and75 °C. The decline with time in percentage germination andpercentage normal seedlings was determined for each storagetreatment. The data were fitted to an equation which containsthe constants: K1, the probit of the initial percentage germinationor normal seedlings; KE, a species constant; CW, the constantof a logarithmic moisture term; CH, the constant of a lineartemperature term and CQ, the constant of a quadratic temperatureterm. Regression analysis of data from storage periods up to5.5 years at temperatures of 5–75 °C and seed moisturecontents of 3.6–13.6 per cent yielded the following values:KE= 8.218, CW=4.797±0.163, CH=0.0489±0.0050 andCQ=0.000365±0.000056. Although this equation consistentlyprovided a better fit, simplified equations, assuming eithera log-linear relationship between seed longevity and temperature,or a log-linear relationship between seed longevity and bothmoisture content and temperature, accounted for more than 94per cent of the variation at the restricted temperature rangeof 5–40 °C. Longevity of the same seed lots at sub-zero temperatures (–5,–10 and –20 °C) was studied in separate tests.Freezing damage, resulting in abnormal seedlings in the germinationtest, occurred at –20 °C when the moisture contentof the seeds exceeded 12 per cent. No decline in percentagenormal seedlings was observed after a storage period of 18 monthsor longer at –20 °C, provided the seed moisture contentdid not exceed 9.5 per cent. For seeds stored at –5 and–10 °C with 9.6–12.5 per cent moisture content,the observed rate of decline of percentage normal seedlingswas adequately predicted by the viability equation, using theabove values for the constants. This suggests that for low moisturecontents the viability equation can be applied to estimate longevityat sub-zero temperatures. Barley (Hordeum vulgare L.) varieties differed in their raponseto [K+]0, in terms of their utilization efficiencies (UE = freshweight. concentration of [K+]1–1). At low [K+]0, Compana,an efficient-non-responder demonstrated superior utilizationof absorbed K+. On the other hand, at high [K+]0, Fergus (anefficient responder) and BT 334 (an inefficient responder) hadhigher UE values for K+ than Compana which performed poorlyat this [K+]0. Kinetic parameters for K+ activation of the enzyme pyruvatekinase from 12 barley varieties, representing a range of UEvalues, were determined. Varieties showed substantial differencesin their Vmax values (P<0·01). Compana, an efficientvariety, had the highest Vmax (31 µmol g–1 freshwt. h–1) which was about 50% higher than that of Mingo,an inefficient variety. By contrast, Km values for the enzymeswere not significantly different among varieties The mean valuesfor all varieties (3·9±0·15 mol m–3K+) is far below the estimated cytoplasmic [K+] (100-200 molm–3). It is, therefore, unlikely that differences in theutilization of K+ by these varieties can be explained on thebasis of differential requirements for (K+) activation of theseenzymes. Changes in seed quality (assessed by potential longevity, i.e.the value of the seed lot constant K1 of the seed viabilityequation) in three contrasting cultivars of rice (Oryza sativaL.) were monitored during seed development and maturation intwo temperature regimes, viz 28/20°C and 32/24°C (12/12h), provided by controlled environments. Mass maturity (definedas the end of the seed-filling phase) varied only between 18and 20 d after 50% anthesis. In five of the six treatment combinationsmaximum potential longevity was not achieved until 12-19 d aftermass maturity. In contrast, the maximum potential longevityof seeds of a japonica rice cultivar produced in the warmerregime was obtained in the first harvest after mass maturity.After mass maturity, the potential longevity of the japonicarice seed lots produced in the warmer environment was much lessthan that for the cooler environments. Maximum potential longevitywas also consistently greater in the cooler than the warmerregime for the two indica cultivars, although the differencein K1 was small (0·3-0·5). The deleterious effectof increase in temperature on seed quality development was notdetected until after mass maturity. Maximum potential longevityin the cooler regime was greatest in the glutinous indica (K1= 3·9) and least in the japonica cultivar (K1 = 3·1).It is concluded that the japonica cultivar is not as well adaptedto warm seed production regimes as the indica cultivars. Dissolved organic carbon (DOC) decay was measured for samplesfrom cultures of the diatoms Chaetoceros gracilis and Phaeodactylumtricornutum, the flagellate Isochrysis galbana, the dinoflagellateAlexandrium tamarense, and a natural algal assemblage from theNorthwest Arm, Nova Scotia, Canada, by a high-temperature catalyticoxidation (HTCO) method. Decay rate constants were determinedusing first-order reaction kinetics in the multi-G model ofBerner (In Early Diagenesis, a Theoretical Approach, PrincetonUniversity Press, 1980). Decay rates as high as 0.37 day–1wereobtained, which demonstrated that DOC released by phytoplanktonmight be highly labile to bacterial utilization and could bedegraded significantly within hours. Decay rates for most speciestested followed much the same pattern, with K01 values around0.3–0.4, K02 values around 0.03, and K03 and K04 valuesaround 10–3 day–1. DOC released by the senescentcells of A.tamarense was found to be essentially bacteria resistant,in contrast to that of the other species tested. The decay ofDOC was directly temperature dependent over the 10–20°Crange. Six methods for DOC preservation were tested. Acidificationwith HCl and refrigerated storage was demonstrated to be themost convenient and practical method. This method can be usedfor both short- and long-term preservation of DOC samples containinghighly labile organic compounds.  相似文献

The longevity and desiccation tolerance of samples of seedsof a japonica rice (Oryza sativa L.) harvested serially duringdevelopment from plants grown in two temperature regimes, viz28/20 °C and 32/24 °C (12/12 h) were determined. Massmaturity (defined as the end of the seed-filling phase) occurred19·7 and 18·3 d after 50% anthesis, respectively.Longevity (determined at 40 °C with 15% moisture contentand quantified by the value of the constant Ki of the seed viabilityequation) improved during seed development and maturation until17 and 14 d after mass maturity in the cooler and warmer regimes,respectively, but declined thereafter. Changes in Ki with timewere similar in the two environments until mass maturity, butthe increase in Ki values after mass maturity was much greaterin the cooler regime. Tolerance of desiccation to low (4%) moisturecontents improved until 22 and 14 d after mass maturity in thecooler and warmer regimes, respectively, when maturation dryinghad reduced seed moisture contents naturally to 24 and 32% moisturecontent, respectively. Further delays to seed harvest reduceddesiccation tolerance, particularly in the warmer environment.Comparison among 15 samples of seeds harvested at differenttimes in the two environments showed a strong correlation (r= 0·947, P < 0·01) between longevity (Ki) anddesiccation tolerance (to 4% moisture content). The kinetic properties of phosphoenolpyruvate (PEP) carboxylasehave been studied among several Flaveria species: the C3 speciesF. cronquistii, the C3–C4 species F. pubescens and F.linearis, and the C4 species F. trinervia. At either pH 7 or8, the maximum activities (in µmol.mg Chl–1.h–1)for F. pubescens and linearis (187–513) were intermediateto those of the C3 species (12–19) and the C4 species(2,182–2,627). The response curves of velocity versusPEP concentration were hyperbolic for the C3 and C3–C4species at either pH 7 or 8 while they were sigmoidal for theC4 species at pH 7 and hyperbolic at pH 8. The Km values forPEP determined from reciprocal plots were lowest in the C3 species,and of intermediate value in the C3–C4 species comparedto the K' values of the C4 species determined from Hill plotsat either pH 7 or 8. Glucose-6-phosphate (G6P) decreased theKm values for PEP at both pH 7 and 8 in the C3 and C3–C4species. In the C4 species, G6P decreased the K' values at pH8 but increased the K' values at pH 7. In all cases, G6P hadits effect by influencing the activity at limiting PEP concentrationswith little or no effect on the maximum activity. At pH 8 andlimiting concentrations of PEP the degree of stimulation ofthe activity by G6P was greatest in the C4 species, intermediatein F. linearis, a C3–C4 species, and lowest in the C3species. Potential carbon-specific growth rates of phytoplankton wereestimated from a series of measurements of photosynthetic radio-carbonuptake over 4- and 24-h exposure periods in the light fieldsof three large limnetic enclosures (‘Lund Tubes’),each providing different limnological and trophic conditions.Photosynthetic behaviour and short-term, chlorophyll-specificcarbon-fixation rates conformed to well-established criteriabut, over 24 h, the net retention represented 23–82% ofthe carbon fixed during the daylight hours. Potential mean growthrates (k'p, of the photo-autotrophic community were calculatedas the net exponential rates of daily carbon-accumulation relativeto derived, instantaneous estimates of the cell carbon-content.Apparent actual community growth rates (k'D were calculatedas the sum of the exponential rates of change of each of themajor species present, corrected for probable rates of in situgrazing and sinking, and expressed relative to the fractionof total biomass for which they accounted. The correspondingvalues were only occasionally similar, k'p generally exceedingK'D by a factor of between 1 and 30 or 1 and 14, depending uponthe carbon:chlorophyll ratio used. The ratio, K'p/K'D was foundto vary inversely both to k'D and to kn, the net rate of changein phytoplankton biomass, suggesting that measured carbon fixationrates merely represent a capacity for cellular increase which,owing to other likely limitations upon growth, is seldom realized.Apparent rates of loss of whole cells do not account for theloss of carbon; that the ‘unaccounted’ loss rates(K'pK'D varied in direct proportion to K'p (i.e., losseswere least when chlorophyll-specific photosynthetic productivitywas itself limited) is best explained by physiological voidingof excess carbon (for instance, by respiration, photorespiration,excretion) prior to the formation of new cells.  相似文献

Cucumber (Cucumis sativus L.) seedlings were grafted onto cucumber-(CG) or figleaf gourd- (FG, Cucurbita ficifolia Bouché)seedlings in order to determine the effect of solution temperature(12, 22, and 32°C) on the mineral composition of xylem sapand the plasma membrane K+-Mg++-ATPase activities of the roots.Low solution temperature (12°C) lowered the concentrationof NO3 and H2PO4 in xylem sap of CG plants butnot of FG plants. Concentrations of K+, Ca++ and Mg++ in xylemsap were less affected than anions by solution temperature.The plasma membrane of FG plants grown in 12°C solutiontemperature showed the highest K+- Mg++-ATPase activity at allATP concentrations up to 3 mM and at low reaction temperatureup to 12°C, indicating resistance of figleaf gourd to lowroot temperature. In situ light measurements were used to obtain information oninherent and apparent optical properties. The average verticalattenuation coefficient Kd(ave) varied from 1.1 to 4.6 In unitsm–1 During three periods the variation in Kd(ave) correlatedwith changes in chlorophyll a concentration and specific attenuationcoefficients Ks, of 0.013, 0.014 and 0.022 m2 mg Chl a–1were calculated. Chlorophyll-specific diffuse absorption coefficients(A,) for these periods were 0.012. 0.013 and 0.017 m2 mg Chla–1 and only varied significantly from estimates of Ksin the period when scattering was intense. Absorption coefficientsa(zmid) and scattering coefficients b(zmid) calculated for themid-point of the euphotic zone ranged between 0.45 and 2.9 mand 3.5–52.0 m respectively. Chlorophyll-specific absorptioncoefficients Ka, of 0.005, 0.006 and 0.007 m2 mg Chl a–1and scattering coefficients Kb of 0.05. 0.09 and 0.191 m2 mgChl a–1 were measured during the three periods. The highKb value occurred when gas-vacuolate cyanobactena were dominant.Algal photosynthesis and light absorption were related throughthe maximum quantum yield m which varied between 0.019 and 0.11mol C Einstein–1 while average quantum yields a, variedbetween 0.006 and 0.024 with a mean of 0.013 mol C Einstein–1A comparison of changes in the mean irradiance of the mixedzone and chlorophyll concentration indicated that growth waslight limited below 0.04–0.05 Einsteins absorbed mg Chla–1 day–1.  相似文献

The cellular mechanism for Cl and K+ secretion in the colonic epithelium requires K+ channels in the basolateral and apical membranes. Colonic mucosa from guinea pig and rat were fixed, sectioned, and then probed with antibodies to the K+ channel proteins KVLQT1 (Kcnq1) and minK-related peptide 2 (MiRP2, Kcne3). Immunofluorescence labeling for Kcnq1 was most prominent in the lateral membrane of crypt cells in rat colon. The guinea pig distal colon had distinct lateral membrane immunoreactivity for Kcnq1 in crypt and surface cells. In addition, Kcne3, an auxiliary subunit for Kcnq1, was detected in the lateral membrane of crypt and surface cells in guinea pig distal colon. Transepithelial short-circuit current (Isc) and transepithelial conductance (Gt) were measured for colonic mucosa during secretory activation by epinephrine (EPI), prostaglandin E2 (PGE2), and carbachol (CCh). HMR1556 (10 µM), an inhibitor of Kcnq1 channels (Gerlach U, Brendel J, Lang HJ, Paulus EF, Weidmann K, Brüggemann A, Busch A, Suessbrich H, Bleich M, and Greger R. J Med Chem 44: 3831–3837, 2001), partially (50%) inhibited Cl secretory Isc and Gt activated by PGE2 and CCh in rat colon with an IC50 of 55 nM, but in guinea pig distal colon Cl secretory Isc and Gt were unaltered. EPI-activated K+-secretory Isc and Gt also were essentially unaltered by HMR1556 in both rat and guinea pig colon. Although immunofluorescence labeling with a Kcnq1 antibody supported the basolateral membrane presence in colonic epithelium of the guinea pig as well as the rat, the Kcnq1 K+ channel is not an essential component for producing Cl secretion. Other K+ channels present in the basolateral membrane presumably must also contribute directly to the K+ conductance necessary for K+ exit during activation of Cl secretion in the colonic mucosa. Secretion of Cl and K+ in the colonic epithelium operates through a cellular mechanism requiring K+ channels in the basolateral and apical membranes. Transepithelial current [short-circuit current (Isc)] and conductance (Gt) were measured for isolated distal colonic mucosa during secretory activation by epinephrine (Epi) or PGE2 and synergistically by PGE2 and carbachol (PGE2 + CCh). TRAM-34 at 0.5 µM, an inhibitor of KCa3.1 (IK, Kcnn4) K+ channels (H. Wulff, M. J. Miller, W. Hänsel, S. Grissmer, M. D. Cahalan, and K. G. Chandy. Proc Natl Acad Sci USA 97: 8151–8156, 2000), did not alter secretory Isc or Gt in guinea pig or rat colon. The presence of KCa3.1 in the mucosa was confirmed by immunoblot and immunofluorescence detection. At 100 µM, TRAM-34 inhibited Isc and Gt activated by Epi (4%), PGE2 (30%) and PGE2 + CCh (60%). The IC50 of 4.0 µM implicated involvement of K+ channels other than KCa3.1. The secretory responses augmented by the K+ channel opener 1-EBIO were inhibited only at a high concentration of TRAM-34, suggesting further that KCa3.1 was not involved. Sensitivity of the synergistic response (PGE2 + CCh) to a high concentration TRAM-34 supported a requirement for multiple K+ conductive pathways in secretion. Clofilium (100 µM), a quaternary ammonium, inhibited Cl secretory Isc and Gt activated by PGE2 (20%) but not K+ secretion activated by Epi. Diurnal temperature fluctuations induced change in soya bean-pod[Glycine max (L.) Merr.] carbon exchange rate (CER, where positiveCER represents CO2 evolution). CER appeared to depend linearlyon temperature. Linear regressions of CER on temperature interceptedthe temperature axis at 5°C (i.e. zero CER at 5°C).Slopes of these regressions (i.e. temperature sensitivity) changedover the season. In bathing solutions of pH = 5?0, progressively increased from 20?5to 430?60 nmol m–2 s–1 and Gm increased from 0?36?0?02to 3?8?0?8 S m–2 when K? was increased from 0?1 to 10mol m–3. The equivalentconductance attributable to the electrogenic transport of K+ ions was calculated from assuming passive, independent diffusionof K+ ions and the ratio was found to be typically close to one. It was also found that themagnitudes of and Gm measuredsimultaneously for each individual cell were also well correlatedfor K? 1?0 mol m–3, and that the slope of the line ofbest fit was close to one. For each K? it was found that theconductance not attributable to K+ translocation and presumablyassociated primarily with the transport of protons or theirequivalents was typically 0?2–0?5 Sm–2. For K? >1?0 mol m–3 the results indicated that the transport ofK+ ions was essentially independent, i.e. there was no evidencefor flux interactions. The results also indicated that the equivalentconductance derived from the measured 42K influx could usefullyindicate the fraction of the electrical conductance attributableto the translocation of K+ ions. Potassium-Ammonium Uptake Interactions in Tobacco Seedlings   总被引:6,自引:0,他引:6  
Short-term (< 12 h) uptake experiments were conducted with6–7-week-old tobacco (Nicotiana tabacum L. cv. Ky 14)seedlings to determine absorption interactions between K+ andNH4+. At equal solution concentrations (0.5 mol m–3) netK+ uptake was inhibited 30–35% by NH4+ and NH4+ uptakewas decreased 9–24%. Removal of NH4+ resulted in completerecovery in K+ uptake rate, but NH4+ uptake rate did not recoverwhen K+ was removed. In both cases, inhibition of the uptakerate of one cation saturated as the concentration of the othercation was increased up to 0.5 mol m–3. The relative effectof K+-NH4+ interactions was not altered when Cl- was replacedwith SO42–, but the magnitudes of the uptake rates wereless in the absence of Cl-. The Vmax for NH4+ uptake was reducedfrom 128 to 105 µmol g–1 dry wt. h–1 in thepresence of 0.5 mol m–3 K+ and the Km for NH4+ doubledfrom 12 to 27 mmol m–3 in the presence of K+. The resultsof these K+-NH4+ experiments are interpreted as mixed-noncompetitiveinteractions. However, an enhanced efflux of K+ coupled to NH4+influx via an antiporter cannot be ruled out as contributingto the decrease in net K+ uptake.
