The relative importance of bay habitats, consisting of mangrove creeks and channel, seagrass beds, and mud and sand flats, as feeding grounds for a number of fish species was studied in Chwaka Bay, Zanzibar, Tanzania, using gut content analysis and stable isotope analysis of carbon and nitrogen. Gut content analysis revealed that within fish species almost the same food items were consumed regardless of the different habitats in which they were caught. Crustaceans (mainly copepods, crabs and shrimps) were the preferred food for most zoobenthivores and omnivores, while fishes and algae were the preferred food for piscivores and herbivores, respectively. The mean δ13C values of fishes and food items from the mangrove habitats were significantly depleted to those from the seagrass habitats by 6·9 and 9·7‰ for fishes and food items, respectively, and to those from the mud and sand flats by 3·5 and 5·8‰, respectively. Fishes and food items from the mud and sand flats were significantly depleted as compared to those of the seagrass habitats by 3·4 and 3·9‰, for fishes and food, respectively. Similar to other studies done in different geographical locations, the importance of mangrove and seagrass themselves as a primary source of carbon to higher trophic levels is limited. The different bay habitats were all used as feeding grounds by different fish species. Individuals of the species Gerres filamentosus , Gerres oyena , Lethrinus lentjan , Lutjanus fulviflamma , Pelates quadrilineatus and Siganus sutor appeared to show a connectivity with respect to feeding between different habitats by having δ13C values which were in-between those of food items from two neighbouring habitats. This connectivity could be a result of either daily tidal migrations or recent ontogenetic migration.  相似文献

Hydrolytic enzymes activities are studied in comparison in doubly virulent partial nicotinic acid requiring mutant and non-virulent para-aminobenzoic acid requiring mutant ofFusarium vasinfectum Atk. Protease, cellulase, amylase and lipase were found to be more active in the doubly virulent strain than the parent strain and para-aminobenzoic acid requiring mutant. Nucleases did not show any significant variations. In Vitro studies revealed that the enzyme activities are dependent on the total substrate available. Protease, Cellulase and Amylase are always extracellularly more active. Nucleases appear to be less active in mutants in comparison with parent strain. Here I investigate whether patterns of scarid biomass across the continental shelf of the northern Great Barrier Reef can be explained by species associating with particular characteristics of the reef environment. Despite the widely documented tendency of scarids to graze and browse over exposed calcareous reef surfaces, scarid biomass was not significantly correlated with the availability of feeding substrata for any species investigated. Indeed positive correlations between biomass and substrata variables were rare for the 18 species investigated, indicating that biomass in these taxa was not strongly reliant on the availability of preferred substrata quantified at the spatial scale of sites (1620m2). Rather, species specific biomass was commonly highly variable between sites, suggesting that local aggregation of scarids commonly occurs at this scale. Such spatial patchiness potentially reflects the effects of spatially variable recruitment, fishes associating with unmeasured habitat characteristics or aggregating prior to spawning. Despite variability in the biomass of individual species between replicate sites within exposure regimes, exposure was generally a far more reliable predictor of biomass than the other variables quantified. This study provides little evidence to indicate that adult scarids have strict habitat requirements, rather they appear to be habitat generalists whose biomass is strongly influenced by exposure but weakly related to the cover of particular reef substratum.  相似文献

Six hectares, three in a primary forest and three in a 40 year old secondary forest were inventoried for all trees with Diameter at Breast Height (DNH) of 10 cm or greater in a terra firme forest 200 km north-east of Manaus, central Amazonia in order to compare the difference between structure, species richness and floristic composition. Both species richness and tree density were significantly higher in the upland forest than in the secondary forest. The forest structure pattern analysed (DBH, basal area and estimated dry biomass) did not differ significantly between the two forest types. Similarity indices at species level were only 14%. In the 3 ha of primary forest the number of species varied from 137 to 159, the number of individuals from 639 to 713, total basal area from 32.8 to 40.2 m2 and estimate total of above-ground dry biomass (AGBM) from 405 to 560 tons per ha. In the 3 ha of secondary forest, the number of species varied from 86 to 90, the number of individuals from 611 to 653, total basal area from 28.8 to 39.9 m2 and estimated total AGBM from 340 to 586 tons per ha. Family Importance Value (FIV) is the sum of relative density, dominance and richness of a family. The most important families in relation to FIV were Burseraceae, Chrysobalanaceae, Lecythidaceae, Myristicaceae, Bombacaceae, Fabaceae and Mimosaceae in the 3 ha of primary forest, while Burseraceae, Lecythidaceae, Sapotaceae, Arecaceae and Cecropiaceae were the most important families in the 3 ha of secondary forest. Importance Value Index (IVI) is the sum of relative density, dominance and frequency of a species. Alexa grandiflora (Caesalpiniaceae), Sckronema micranthum (Bombacaceae) and Pourouma guianensis (Cecropiaceae) were the most important species in relation IVI, in the primary forest, while Eschweilera grandiflora (Lecythidaceae), Protium apiculatum (Burseraceae) and Bertholletia excelsa (Lecythidaceae) were the most important species in the secondary forest. We conclude that species richness was significandy different between the two forests, but that forest structure patterns analysed in this study (DBH, basal area and dry biomass) were similar. This demonstrates that 40 years was sufficient time for the secondary forest to recover the original structure of the primary forest, but not the original species richness. The low species similarity between the two forests indicates that the floristic composition was quite distinct and that the mixture of primary forest and disturbed forest has led to an increase in total species diversity.  相似文献

Abstract. A comparative study of two chromosomal races of the winter-active ephemeral Machaeranthera gracilis showed that the seasonal magnitudes of photosynthesis were only slightly greater for a progeny desert race than for an ancestral foothills race. Maximum observed photosynthetic capacity and the seasonal reduction in foliar photosynthesis occurred earlier in the year for the desert race. The relative growth rate was higher in this race up until the time of its peak seasonal biomass. A ratio of harvested net production to estimated gross primary production decreased until anthesis. Photosynthesis contributing to net growth continued into periods with moderate environmental stress. The continuation of growth by the desert race was enhanced by maintenance of a higher root-shoot ratio, as well as greater relative stem growth. During reproduction, foliar CO2 assimilation could not solely provide the measured dry matter accumulation, suggesting the importance of assimilate contribution by photosynthetic stems. Seasonal increases in the enthalpy content of whole plants and plant organs occurred for both races, indicating the absence of significant translocation during reproduction and the potential for stem photosynthesis.  相似文献

Electron microscopic and biochemical studies revealed a salient difference in the response to toxic doses of ouabain by cultured cardiac muscle and non-muscle cells from neonatal rats. Progressive cellular injury in myocytes incubated with 1 · 10?4–1 · 10?3 M ouabain ultimately leads to swelling and necrosis. The morphological damage in myocytes was accompanied by a drastic decrease in 14CO2 formation from 14C-labeled stearate or acetate but not glucose. Neither morphological nor biochemical impairments were observed in non-muscle cells. The interaction between ouabain and the cultured cells, using therapeutic doses of ouabain (i.e., <1 · 10?7 M), was characterized. Two binding sites were described in both classes of cells, one site is a saturable K+-sensitive site whereas the other is non-saturable and K+-insensitive. The complexes formed between the sarcolemma receptor(s) and ouabain, at low concentrations of the drug (e.g., 7.52 · 10?9 M), had Kd values of 8.9 · 10?8 and 2.3 · 10?8 M for muscle and non-muscle cells, respectively. The formation and dissociation of the complexes were affected by temperature and potassium ions.  相似文献

This review describes the effects of the current and emerging lighting technologies on plants, and the plant-mediated effects on herbivorous and beneficial arthropods in high-technology year-round greenhouse production, where light quality, quantity and photoperiod differ from the natural environment. The spectrum provided by the current lighting technology, high-pressure sodium lamp (HPSL), differs considerably from that of solar radiation. The major plant-mediated effects on arthropods were predicted to result from (a) extended photoperiods and lower light integrals, (b) the attenuation of ultraviolet (UV) wavelengths, particularly UV-B, (c) the high red: far-red (R : FR) ratio and lower blue : red (B : R) in comparison with solar radiation and (d) the high proportion of yellow wavelengths during winter months. Of these light factors (a–d) (ceteris paribus), (a) and (b) were hypothesised to result in increased performance of herbivores in winter months, whereas the high R : FR ratio decreased herbivore performance or not affected it, at least when interlights are used. The predictions obtained on the basis of this review are also discussed in relation to the modifying factors prevailing in these production environments: enriched CO2 levels, high nutrient amounts, optimised irrigation and temperatures optimal for plants' needs. Based on the carbon/nitrogen and growth/differentiation balance theories, these modifying factors tend to produce plants that allocate most resources to growth at the expense of defensive secondary metabolism and physicochemical defensive structures. At the end, this review discusses knowledge gaps and future research prospects, in which light-emitting diodes, the emerging lighting technology, play an important role by enabling the targeted manipulation of plant responses to different wavelengths.  相似文献

This experiment studied effects of a mixture of exogenous enzymes (ZADO®) from anaerobic bacteria on ruminal fermentation, feed intake, digestibility, as well as milk production and composition in cows fed total mixed rations (TMRs; 0.7 corn silage and 0.3 of a concentrate mixture). Twenty lactating multiparous Brown Swiss cows (500 ± 12.4 kg live weight) were randomly assigned into two experimental groups of 10 immediately after calving and fed a TMR with or without (CTRL) addition of 40 g/cow/d of enzymes for 12 weeks. Addition of enzymes increased (P<0.05) rumen microbial N synthesis. Intake of dry matter (DM) and organic matter (OM) was positively influenced (P<0.05) by supplementation, and digestibility of all nutrients was higher (P<0.05) in the total tract of supplemented cows, although the magnitude of the improvement varied among nutrients, with the highest improvement in aNDFom and ADFom (418–584 and 401–532 g/kg respectively; P<0.05) than the other nutrients. Supplementation of enzymes also increased (P<0.05) rumen ammonia N and total short chain fatty acid (SCFA) concentrations, and individual SCFA proportions were also altered with an increase in acetate (61.0–64.8 mol/100 mol; P=0.05) before feeding, and acetate and propionate increased 3 h post-feeding (60.0–64.0 and 18.3–20.8 mol/100 mol respectively; P<0.05). Milk and milk protein production was higher (12.8–15.7 and 0.45–0.57 kg/d respectively; P<0.05) for cows fed the ZADO® supplemented diet. This exogenous enzyme product, supplemented daily to the TMR of cows in early lactation, increased milk production due to positive effects on nutrient intake and digestibility, extent of ruminal fermentation and microbial protein synthesis.  相似文献

Irrespective of the constituent protein, all amyloid fibrils show similar morphology in the electron microscope and x-ray diffraction patterns characteristic of a "cross-beta" structure, with extended beta-strands perpendicular to the fibril's long axis. Little is known about the amount or type of this structure. I have measured CD spectra of films formed from a number of amyloid proteins and polypeptides, and estimated their contents of extended secondary structure, by analysis of their g-factor spectra, the ratio of the CD and absorbance signals (P. McPhie, Analytical Biochemistry, 2001, Vol. 293, pp. 109-119). Amyloid films of Abeta-(1-40) peptide, beta-2-microglobulin, insulin, and three homopolypeptides show very intense CD spectra, compatible with the presence of a beta-helix-like structure, arranged in a common framework in the fibrils. The extent of this structure was estimated as 45-80% in the protein fibrils and 30-80% in the polypeptide fibrils.  相似文献

Some plants in arctic and alpine habitats have heliotropic flowers that track the sun. This results in a heating of the flower's interior, which may improve the possibilities for insect pollination and seed production. Here, I examine whether flower heliotropism in an alpine population of the self-incompatible Ranunculus acris L. (Ranunculaceae) enhances pollinator visitation and seed production. Flowers of Ranunculus acris tracked the sun during the day. Tracking accuracy was greatest during the middle of the day. The temperature elevation in flowers was negatively correlated with the flower's angle of deviation from the sun. Despite the increased temperature, insects did not discriminate among flowers on the basis of their angle of deviation from the sun, or tend to stay longer in the flowers aligned closest towards the sun. A tethering experiment was conducted on three groups of plants flowering at different times in the 1993 season and on one group the following season. Manipulation plants were constrained not to track the sun, whereas control plants tracked the sun naturally. Solar tracking had no effect on seed:ovule ratio, seed mass, or abortion rate in any of the groups. There is probably a very narrow range of weather conditions (cold, sunny, and calm) where flower heliotropism may enhance visitation rate to flowers and their seed production.  相似文献

Recently developed computer programs, including secondary structure and epitopic site predictions, have been used to align lysosomal proteins for maximum homology, based on conservative interchanges, and the aligned sequences have been searched for potential sites for posttranslational modification, glycosylation, and binding and catalysis of substrate. The homology and prediction of the posttranslational modification of the alpha- and beta-subunits of hexosaminidase is in good agreement with previous observations, and an explanation of the differing substrate specificities of the two subunits is advanced. We show that the striking homology between alpha-glucosidase and isomaltase is reflected in the apparent conservation of the active site in both enzymes. Nonhomologous regions have been examined in detail in a search for binding sites for glycogen and maltose, and two such sites have been tentatively identified. A highly redundant consensus sequence for the phosphorylation of mannose in lysosomal proteins, YXX(Y, W, or F), is suggested.  相似文献

Summary The water permeability of human red blood cell (RBC) membrane has been monitored by a doping nuclear magnetic resonance (NMR) technique on intact cells and resealed ghosts following exposure to various sulfydryl-reacting (SH) reagents and proteolytic enzymes. The main conclusions are the following: (i) When appropriate conditions for exposure of erythrocytes or ghosts to mercury-containing SH reagents (concentration, temperature and duration of incubation) were found, the maximal inhibition of water diffusion could be obtained with all mercurials (including HgCl2 and mersalyl that failed to show their inhibitory action on RBC water permeability in some investigations). While previous studies claimed that long incubation times are required for the development of maximal inhibition of water diffusion by mercurials, the present results show that it can be induced in a much shorter time (5–15 min at 37°C) if relatively high concentrations of PCMBS (2–4mm) are used and no washings of the inhibitor are performed after incubation. Higher than optimal concentrations of mercurials and/or longer incubation times result in lower values of inhibition, sometimes a loss of inhibition, or can even lead to higher values of permeability compared to control RBCs. (ii) The conditions for inhibition by mercurials are drastically changed by preincubation of erythrocytes with noninhibitory SH reagents (such as NEM or IAM) or by exposure to proteolytic enzymes. If the cells are digested with papain, the duration of incubation with PCMBS should be decreased in order for inhibition to occur. This explains the lack of inhibition reported previously, when a relatively long duration of incubation with PCMBS was used subsequent to papain digestion. (iii) The degree of inhibition of water diffusion induced by mercurials appeared to be dependent upon the temperature of which the water permeability was measured. The values of maximal inhibition ranged from 45–50% at 37°C, increased 10–15% at 20°C and further increased at lower temperatures, reaching values above 75% below 10°C; these results clarify the conflicting reports of various authors. (iv) The inhibition of water diffusion, either reversible, or irreversible, was not accompanied by significant changes in the pattern of RBC membrane polypeptides fractionated by polyacrylamide gel electrophoresis. (v) The mean value of the activation energy of water diffusion (E a,d) obtained on 42 donors was 25.6 kJ/mol. The values ofE a,d increased in parallel with the values of the inhibition of water diffusion induced by PCMBS until the maximal inhibition was reached (whenE a,d=41 kJ/mol) and then both sets of values decreased in parallel.  相似文献

Salinity influences plant growth, seed yield and seed quality even of halophytic crops such as Chenopodium quinoa. Plant growth, total seed yield, number of seeds, fresh weight and dry weight of seeds, were all significantly reduced in the presence of salinity. Only at high salinity did the content of proteins (as well as total N) increase significantly in the seeds whereas the content of total carbohydrates (as well as total C) decrease. Aside from that the capacity for germination was diminished by a reduced seed size and a disproportionate reduction of the volume of the perisperm. However, the reduced capacity seemed to be compensated by an accelerated germination owing to high Na and Cl concentrations leading to a low water potential in the walls of the plant ovary. At high salinity the passage of NaCl to the seed interior was hindered by the seed cover. There was an obvious gradient between potentially toxic (Na and Cl) and essentially needed elements (K, Mg, Ca, P and S) across the seed coat of salt treated plants and also a significant change of the distribution of elements in the embryo. The results indicate a highly protected seed interior leading to a high salinity resistance of quinoa seeds.  相似文献

Diurnal raptor habitat use has conservation implications due to environmental and anthropogenic interactions. Three tree-nesting diurnal raptors, black kites (Milvus milvus), steppe buzzards (Buteo vulpinus) and African fish eagles (Haliaeetus vocifer), were studied from December 2016 to October 2017. The objective was to determine factors influencing diurnal raptor habitat selection and use in and around Chembe Bird Sanctuary of Kalulushi district, Zambia. We surveyed for birds in ten randomly selected 200 × 200 m sample plots in each of the five stratified sampling units (miombo woodland, grasslands, human settlements, Lake Chembe and marsh). Seasons and dietary composition variably influenced the distribution, habitat selection and use by raptors. The three raptors devised a suit of behaviours to curtail challenges associated with seasonal food and water availability, and anthropogenic disturbances. There is a need to promote multi-stakeholder engagement and involvement in raptor conservation.  相似文献

Nandrolone decanoate (Nd) is a highly abused androgenic-anabolic steroid among body builders. Even though it has weak androgenic effects, its prolonged use may have harmful impact on male reproductive system which needs to be evaluated. This study aimed to reinvestigate its possible oxidative stress induced alteration on male rat reproductive system. Twenty-eight male rats were divided into two groups. Nd treated group (n = 18) injected intramuscular with 10 mg/kg body weight once a week for four weeks. While, the control group (n = 10) was injected with physiologic saline by the same route for four weeks. Body weight was recorded for all rats and after animal dissection weight of testes, prostate and seminal vesicles were also recorded. The results showed that the average testicular weight was decreased in treated group compared to the control. The average weights of the prostate and seminal vesicles were increased compared to the control. Morphometric study revealed that in Nd treated group, there was a decrease in the width of seminiferous tubules and the height of spermatogenic cell layer compared to the control. Testicular degeneration was expressed by presence of spermatid giant cells, vacuolation, and degenerated spermatozoa. Tunnel technique showed scattered positive reaction among the spermatogenic cell layers and interstitial cells. Severe alterations of the prostate were expressed by benign prostate hyperplasia and retained secretions. Lipid peroxidation products (malonaldehyde concentration as ng/g of testicular tissue) were increased in treated group compared to the control and suggested the occurrence of oxidative damage. Nd induced severe alterations in the male genital organs were resulted from oxidative stress. It is concluded that the male genital organs are highly sensitive to the anabolic steroids and there is a high extent of reproductive risk associated with the use of AASs.  相似文献

1. Beaver Lake, a large epishelf lake in eastern Antarctica was sampled on two occasions during the austral summer of 2000. Two sites, one 1 km offshore and another 6 km offshore were sampled at intervals to depths of 40 and 110 m, respectively. 2. The lake is an end member of ultra‐oligotrophic lake systems with a very low carbon pool. Dissolved organic carbon concentrations ranged between 95 and 652 μg L–1. Nutrient levels were generally low with soluble reactive phosphorus ranging from undetectable to 8.4 μg L–1, ammonium ranged between 1.8 and 5.0 μg L–1, nitrate from undetectable to 161 μg L–1 and nitrite 1.1–5.3 μg L–1. 3. Chlorophyll a concentrations (0.39–4.38 μg L–1) showed an unusual distribution with the highest levels close to the lake bottom at the offshore site (110 m) where the phototrophic nanoflagellates (PNAN) displayed strong autofluorescence. 4. Bacterial concentrations were low, with a maximum of 7.60 × 107 L–1, as were the concentrations of heterotrophic nanoflagellates that exploit them. 5. Primary production ranged between 19.7 and 25.49 μg C L–1 day–1 and bacterial production from 0.32 to 1.15 μg C L–1 day–1. 6. In common with other continental Antarctic lakes, the system was dominated by a microbial plankton. However, a dwarf variety of the calanoid copepod, Boeckella poppei, occurred below 25 m at concentrations of 3–5 L–1. 7. The data suggest that primary production and bacterial production were not limited by nutrient availability, but by other factors, e.g. in the case of bacterial production by organic carbon concentrations and primary production by low temperatures.  相似文献

The present study optimized ethanol yield using nickel oxide (NiO) nanoparticles (NPs) as a biocatalyst. Additionally, Saccharomyces cerevisiae BY4743 cell growth and the bioethanol production kinetics were assessed. The Response Surface Methodology (RSM) model showed a coefficient of determination (R2) value of 0.93. The optimized process gave a biomass concentration and ethanol yield of 2.04 g/L and 0.26 g/g (1.03 and 1.19-fold increment compared to the control experiment), respectively. The process kinetic data showed that the inclusion of NiO NPs improved the affinity of S. cerevisiae BY4743 to glucose consumption, carbohydrate and protein accumulation. A significant reduction in volatile fatty acid (VFA) was observed in the presence of NiO NPs. The application of nano biocatalyst in simultaneous saccharification and fermentation of potato peel waste, meaningfully enhanced bioethanol production (>65 %). The study provided major insights into the use of NiO NPs to enhance the bioprocess of ethanol production.  相似文献
