In the experiments on different animal species (mice, cats, dogs) lithium hydroxybutyrate has been shown to have antihypoxic and anti-ischemic effects. Lithium hydroxybutyrate improved the functional state of the ischemic myocardium, stimulated the accumulation of macroergic phosphates (ATP) in the heart, protected the ischemic myocardium and delayed the progression of the reversible ischemic damage into the irreversible one. The improvement of the collateral coronary circulation plays an important role in the anti-ischemic action of the drug.  相似文献

Sodium salt of gamma-hydroxybutyric acid (Na-GHB) was administered to 37 drug-free alcoholic men in placebo controlled study. The psychotropic effect was evaluated using psychometric scale of 4 ranges. Changes in plasma levels of epinephrine, norepinephrine, dopamine, serotonin and platelet MAO-B activity were assessed. The psychotropic effect was observed in 27 (76%) patients. The baseline of dopamine concentration was significantly correlated with the expression of psychotropic effect. Na-GHB decreased dopamine concentration in responders. The possible involvement of dopaminergic system in the realization of psychotropic effect of Na-GHB was discussed.  相似文献

The activity of natural killers from the mouse spleen was determined by 51Cr clearance from labeled target cells (YAC-1) at different times after resection of the two-thirds of the spleen. In the early periods after operation, there was a decrease in the natural cytotoxic activity of splenocytes followed by a relatively rapid rise for 8 days until the maximum exceeding the cytotoxicity of intact spleen cells was reached. However, the total lytic potential of the regenerating spleen did not return to normal during 28 days.  相似文献

The experiments on white rats have confirmed that the development of lung edema following pituitrin infusion is characterized by considerable changes in phospholipid and cholesterol lung metabolism and cholesterol blood metabolism. Lithium hydroxybutyrate preinjection at a dose of 400 mg/kg prevented a decrease in cholesterol lung content and an increase in cholesterol blood plasma level which was accompanied by less prominent lung edema.  相似文献

The activities of natural killer (NK) cells and T-killers of mice after transplantation of allogeneic ovaries were studied. It was shown that during first month after transplantation cytotoxic and membrano-toxic activities of these effector cells were increased. Activation of NK and T-killers depends on tension of transplantation immunity and may serve as prognostic criteria of rejection.  相似文献

The data are presented on the comparative evaluation of a poststress decrease in the activity of normal killer cells, determined in the test of the release of 41Cr from target cells YAC-1 in young and old CBA mice after 6-hour immobilization. The use of mouse leukocyte interferon and poly I . poly C acid has been shown to be highly effective for the restoration of the activity of normal killer cells in young animals after its stress-induced suppression, while in old animals these immunomodulators have proved to be considerably less effective in restoring the activity of natural killer cells at the period following the stress. The probable role of disturbances, affecting the functional properties of the normal killer cell population in old animals, in the mechanism of the poststress decrease of natural cell-mediated immunity is discussed.  相似文献

Lithium oxybutyrate microinjections (10 mg/ml) produce a depressant action on spontaneous bioelectrical activity of the cortex and subcortical structure. The drug brings down excitability of the motor cortex, hippocampus, caudate nucleus, thalamus, posterior hypothalamus and mesencephalic reticular formation; it also raises excitability of the tonsils. The depressant effect of lithium oxybutyrate is superior to that of lithium chloride.  相似文献

The influence of sodium oxybutyrate on the adrenergic neurotransmitter content in the nerve fibers and the synaptic vesicles and the uptake of exogenous noradrenaline (NA) by the sympathetic nerves of rat Vas deferens was studied by spectrofluorimetry, fluorescent-histochemistry and cytochemical electron microscopy. Sodium oxybutirate failed to influence the stores of the adrenergic mediator, but was capable of blocking the uptake and accumulation of the exogenous NA.  相似文献

Single administration of lithium hydroxybutyrate (10 mg/kg) to rabbits decreased serotonin and 5-hydroxyindoleacetic acid (5-HIAA) content in the caudate nucleus. The drug administration for 8 days is accompanied by mediator accumulation in the cortex, caudate nucleus, tonsils, hypothalamus, thalamus, and midbrain with parallel reduction in 5-HIAA level in these structures. 15 days of lithium hydroxybutyrate administration lead to the increase of serotonin and 5-HIAA concentration, while 28 days of administration reduced the content of mediator and its metabolite.  相似文献

Using adsorption methods and elimination of Thy-1+ lymphocytes, the null cell fraction was obtained. The electrophoretic mobilities of unfractionated splenocytes and different fractions of lymphoid cells and null fractions (to which the natural killers belong) were measured with "Parmoquant-2" device. Inflammation plays a role in the pathophysiology of depression. Sulforaphane (SFN), an isothiocyanate compound derived from broccoli, is a potent activator of the NF-E2-related factor-2 (Nrf2), which plays a role in inflammation. In this study, we examined whether the prevention effects of SFN in lipopolysaccharide (LPS) induced depression-like behavior in mice. Pretreatment with SFN significantly blocked an increase in the serum tumor necrosis factor-α (TNF-α) level and an increase in microglial activation of brain regions after a single administration of LPS (0.5 mg/kg). Furthermore, SFN significantly potentiated increased serum levels of IL-10 after LPS administration. In the tail-suspension test and forced swimming test, SFN significantly attenuated an increase of the immobility time after LPS administration. In addition, SFN significantly recovered to control levels for LPS-induced alterations in the proteins such as brain-derived neurotrophic factor, postsynaptic density protein 95 and AMPA receptor 1 (GluA1) and dendritic spine density in the brain regions. Finally, dietary intake of 0.1% glucoraphanin (a glucosinolate precursor of SFN) food during the juvenile and adolescence could prevent the onset of LPS-induced depression-like behaviors and dendritic spine changes in the brain regions at adulthood. In conclusion, these findings suggest that dietary intake of SFN-rich broccoli sprout has prophylactic effects on inflammation-related depressive symptoms. Therefore, supplementation of SFN-rich broccoli sprout could be prophylactic vegetable to prevent or minimize the relapse by inflammation in the remission state of depressed patients.  相似文献

Natural killers (NK cells) possessing cytotoxic activity were isolated from the blood, spleen and bone marrow of Syrian hamsters in discontinuous Percoll density gradient. These cells were morphologically identified as granular lymphocytes (GL). The largest amount of GL was isolated from the fraction containing 52-55% of Percoll. Cytotoxic activity of cells in this fraction was the highest, as compared to nonfractionated control or cells in other Percoll fractions.  相似文献

Serotonin (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) levels in rat brain were analysed 24 hours after 7-, 15-, 29- days lithium hydroxybutyrate (LH) injections (10 mg/kg daily). After 7 days the drug reduced 5-HT in hypothalamus and 5-HIAA in the mid brain by 35%. After 15 days LH decreased 5-HT in striatum, hypothalamus by 32 and 17% and 5-HIAA in thalamus, hypothalamus by 28 and 44% respectively. After 29 days LH diminished 5-HT in striatum, hippocampus, amygdala by 24, 29 and 32% and 5-HIAA--in hypothalamus by 42%. The role of adaptative changes and stabilization processes in the central serotoninergic system in mechanism of LH psychotropic effects is discussed.  相似文献

目的观察木豆素于抗抑郁方面的作用,并初步探讨其可能的作用机制。方法采用小鼠慢性不可预见温和应激模型(chronic unpredictable mild stress,CUMS),进行糖水偏好实验评价经木豆素处理的小鼠的快感缺失情况,采用酶联免疫吸附测定法测定小鼠血清皮质酮的含量,采用LC-MS/MS法检测小鼠皮层和海马中多种神经递质的含量。结果木豆素可以逆转CUMS引起的糖水偏爱指数降低和血清皮质酮水平升高。并且与正常组相比,模型组小鼠皮层和海马中相关神经递质的含量发生了显著的改变,而木豆素给药对于CUMS小鼠体内相关神经递质具有明显的调节作用。结论木豆素可能通过降低血清皮质酮水平和调节脑内神经递质来实现抗抑郁作用。  相似文献

The effect of sodium hydroxybutyrate on the blood flow in the aorta, carotid, mesenteric and femoral arteries were studied on cats and dogs. The circulation was assessed by the electromagnetic and resistographic methods, in the anesthetized and nonanesthetized animals. The tonic activity was recorded in the sympathetic nerves and the EEG. Sodium hydroxybutrate was shown to decrease the sympathetic activity, resulting in the increase of the regional circulation and induced the EEG synchronization. The latter effect was more pronounced in the arotid arteries. It can be assumed that sodium hydroxybutyrate affects the nervous control of the blood vessels.  相似文献
