The positron-emitting, non-steroidal estrogen (2R*, 3S*)-1-[18F]fluoro-2,3-bis(4-hydroxyphenyl)pentane [( 18F]-fluoronor-hexestrol), has been prepared by fluoride ion displacement on a labile trifluoromethanesulfonate (triflate) derivative of a suitably protected precursor, followed by removal of the aryl triflate groups with lithium aluminum hydride and purification by HPLC. In immature female rats, this compound is taken up selectively by the uterus and is retained for prolonged periods, due to its binding to the estrogen receptor. This compound and related 18F-labeled estrogens thus appear to be promising agents for imaging estrogen receptor-positive breast tumors in humans.  相似文献

Nucleosides, bases, and nucleotides can be separated from one another rapidly (10–15 min) on 1-ml silica cartridges. Samples adjusted to 4 mm ammonium borate, 90% acetonitrile are loaded onto 1-ml columns equilibrated with the same solvent. Bases do not absorb to the silica under these conditions. Nucleosides are eluted with 16 ml of 0.5 m acetic acid in 90% acetonitrile. Nucleotides are then eluted with water. The 1-ml silica columns have performed well with samples up to 10 ml in volume. We have found the procedure to be quantitative and the gels to have high capacity (61 μmol Cyd/ml silica). Acid extracts from a large number of cells (108) have been processed on a single cartridge.  相似文献

Using indirect immunofluorescence we have examined the distribution of the cell surface and extracellular matrix glycoprotein fibronectin at the epithelial-mesenchymal interface and in the mesenchyme of developing chick and duck wing buds. At all stages examined, in both species, staining for fibronectin is greatly enhanced in the basement membrane subjacent to the apical ectodermal ridge (AER), a site of inductive tissue interaction, relative to the epithelial basement membranes in the noninductive dorsal and ventral limb epithelial-mesenchymal interfaces. In stage 23, 25, and 28 chick limb buds, staining for fibronectin is uniform in the least mature distal mesenchyme, retained between more proximal cells undergoing precartilage condensation and lost in those regions undergoing myogenesis, and persistent in all but the most mature cartilage present at the latest stage examined. These results are consistent with a role for fibronectin in AER-induced limb outgrowth, and with a transient role for the glycoprotein in the formation of the skeletal pattern of the limb.  相似文献

Experiments were designed to determine the mechanism by which methylxanthines elevate abalone sperm cAMP concentrations and induce the acrosome reaction (AR). Theophylline or, more effectively, 1-methyl-3-isobutylxanthine (MIX) inhibit the cyclic nucleotide phosphodiesterase activities of abalone sperm homogenates. 45Ca2+ uptake by sperm is also stimulated by theophylline, and more effectively by MIX, and this stimulatory effect is blocked by KCN. Verapamil, a compound known to antagonize Ca2+ conductance, has no effect on the Ca2+ or MIX-induced cAMP elevation at concentrations up to 200 microM. However, verapamil reduces the sperm cAMP elevation caused by the addition of Ca2+ plus MIX. This inhibition is not complete, even at 200 microM verapamil. The AR induced by Ca2+ plus MIX is completely inhibited by 200 microM verapamil. The data suggest that these methylxanthines elevate abalone sperm cyclic nucleotide concentrations by inhibiting cyclic nucleotide phosphodiesterase activities. Furthermore, since sperm cAMP metabolism is modulated by Ca2+ flux, methylxanthines also appear to elevate abalone sperm cAMP concentrations by their effects on Ca2+ transport. The Ca2+-induced cAMP elevation occurs through a verapamil-insensitive mechanism, whereas the potentiation by MIX of the Ca2+ effect to elevate cAMP occurs through both verapamil-insensitive and -sensitive mechanisms. The methylxanthine-induced AR is mediated by a primary effect on Ca2+ transport and occurs through a verapamil-sensitive mechanism. Cyclic AMP may play a role in the methylxanthine-induced AR, but does not appear to act as the primary mediator of this exocytotic event.  相似文献

Nuclear magnetic resonance methods have been developed for assignment of the absolute configuration of (4,4,4-d3)-β-hydroxyisovalerate and (2-d1)-β-hydroxyisovalerate. (E) and (Z)-(4,4,4-d3)-β-methylcrotonate and (2-d)-β-methylcrotonate have been administered to biotin-deficient rats, and the resultant β-hydroxyisovalerate was isolated from their urine. The NMR spectra of derivatives of the biosynthetic products established that the hydration of β-methylcrotonate had proceeded nonstereospecifically.  相似文献

The colour yield from glucose and the anthrone reagent is more than doubled in the presence of hydrochloric and formic acids. By using a suitable heating time, approximately equal colour yields can be obtained from glucose and fructose.  相似文献

The insoluble acrosome granule content of sea urchin sperm consists of a single 30,500 dalton protein named bindin. Bindin mediates species-specific recognition and adhesion of sperm to the egg surface. Bindin from Strongylocentrotus purpuratus (Sp) and Strongylocentrotus franciscanus (Sf) have tyrosine as their single N-terminal amino acid. The pI of Sp bindin is 6.62 and of Sf 6.59. Amino acid analysis reveals almost identical composition between the two species for 16 amino acids. Only two (or three) amino acids, Pro and Asx, show large species differences. Tryptic peptide maps of the two species of bindin show very similar patterns with 24 spots of identical correspondence.  相似文献

A synthetic protein kinase substrate, PRO-LEU-SER-ARG-THR-LEU-SER-VAL-SER-SER-NH2, undergoes calcium-dependent binding by calmodulin. Phosphorylation of the peptide decreases its affinity for calmodulin with the dissociation constant increasing from 2.4 to ca. 7 mM. The results are consistent with the suggestion that calmodulin and the cAMP-dependent protein kinase can act on common recognition sequences.  相似文献

The requirement of direct covalent association of trinitrophenyl (Tnp) groups with cell surface components for functional interactions with anti-Tnp cytotoxic T lymphocytes (CTLs) was analyzed. This question was approached by comparing the ability of two methods of trinitrophenylation to render cells susceptible to lysis by anti-Tnp CTLs. As previously shown, cells modified with trinitrobenzene sulfonic acid were susceptible to H-2-restricted lysis by anti-Tnp CTLs. However, cells incubated with Sendai virus covalently associated with Tnp groups, were not rendered susceptible to lysis by anti-Tnp CTLs. These same target cells, however, were susceptible to H-2-restricted lysis by anti-Sendai virus CTLs. Direct analysis of the number of Tnp groups on cells modified by either method indicates no significant difference in the number of Tnp molecules associated with the different target cells. The results suggest that direct covalent association of Tnp groups with cell surface specific components is a minimal molecular requirement for recognition and lysis by anti-Tnp CTLs.  相似文献

The concentrations of radioactive and nonradioactive purine bases, purine nucleosides, purine mono-, di-, and trinucleotides in acid extracts of fibroblasts were determined by anion-exchange column chromatography. The concentrations of nonradioactive components were determined by computerized integration of the signal from a double-beam uv-detector. The radioactive metabolites were quantitated by high-efficiency, continuous liquid scintillation counting, employing a discrete sample transport system.  相似文献

The effects of Δ9-tetrahydrocannabinol and cannabidiol on the esterase isozymes of rat testicular tissue were examined using acrylamide gel electrophoretic techniques followed by staining for non-specific esterase activity. Both Δ9-tetrahydrocannabinol and cannabidiol caused a specific depression of an esterase isozyme located in the interstitial tissue of the testis.  相似文献

Morphine administration (20 mg/kg) to awake rhesus monkeys which had been chronically implanted with catheters for aspiration of cerebrospinal fluid (CSF) produced a significant elevation in the CSF level of guanosine-3′, 5′-cyclic monophosphate (cGMP). Additionally, biopsies of cerebral and cerebellar cortex were taken from anesthetized monkeys given 20 mg/kg of morphine sulfate. Only cerebellar cGMP levels changed significantly, showing a 35% decrease relative to anesthetized controls. Although the controlling factors of brain tissue and CSF cGMP levels are poorly understood, the possibility of a reciprocal relationship between cGMP levels in certain brain regions and in CSF under some conditions is discussed.  相似文献

The unusual highly phosphorylated nucleotide, guanosine 5′-diphosphate 3′-diphosphate, has been implicated in the control of development of the mouse (Irr, J. D., et al. (1974) Cell3, 249). We have been unable, however, to detect guanosine 5′-diphosphate 3′-diphosphate synthesis either in preimplantation and postimplantation mouse embryos cultured in the presence of [32P]orthophosphate or in assays using ribosomes isolated from 10- to 13-day mouse embryos. Three unidentified phosphorous-containing compounds were detected in blastocyst stage mouse embryos.  相似文献

Exposure of human α-1-proteinase inhibitor to 75 ppm nitrogen dioxide and 1 mM hydrogen peroxide results in a 65% loss in the anti-elastase activity of the protein after one hour. Exposure to either nitrogen dioxide or hydrogen peroxide alone has no effect. The inhibition is partially prevented when mannitol or superoxide dismutase is included during the exposure, implying that superoxide and the hydroxyl radical are formed in reactions between nitrogen dioxide and hydrogen peroxide and play a role in the inhibitory process.  相似文献

Cholesteryl esters, the intracellular storage form and intravascular transport form of cholesterol, can exist in crystal, liquid crystal and liquid states. The physical state of cholesteryl esters at physiologic temperatures may be a determinant of their pathogenicity. This review has surveyed saturated aliphatic cholesteryl esters of chain length 1 to 24 carbons and a series of medium-chained unsaturated cholesteryl esters from chain lengths 14 to 24 carbons. A systematic study of transition temperatures by polarizing microscopy and enthalpies by differential scanning calorimetry has provided unifying concepts concerning the phase behavior as a function of chain length and unsaturation. Neat cholesteryl esters show chain-length dependence of transition temperature and enthalpy of both the crystal and liquid crystal transitions. Double bond position along the fatty acyl chain affected stability of the liquid crystal phases; a smectic phase was not observed for any cholesteryl ester with a double bond more proximal than delta 9. 13C NMR spectroscopy in the isotropic liquid phase has provided evidence suggesting a balance of ring-ring vs. chain-chain interactions as a determinant for isotropic liquid----cholesteric vs. isotropic liquid----smectic transitions. Specifically, anisotropic molecular motions of the steroid ring are greater for cholesteryl esters forming a cholesteric phase than a smectic phase from the melt. Chain-chain interactions apparently predominate in smectic phase formation. The X-ray diffraction patterns of cholesteryl esters as a function of chain length reveal several isostructural series and known single crystal data are presented. A chain length depending on the periodicity of the smectic phase is observed which may be different for saturated vs. unsaturated esters. In summary, the phase behavior of cholesteryl ester molecules is complex and cannot be determined a priori from the phase behavior of component cholesterol and fatty acid. The data presented here should provide insight into the biological behavior of this lipid class.  相似文献

The transport of carnitine by rat kidney cortex slices against a concentration gradient has been demonstrated. Similarities to other transport systems included a linear period of uptake, as well as indications of saturability of the system with increasing concentrations of substrate. The transport of carnitine was inhibited by anoxia, and carbonyl cyanide-m-chloro-phenylhydroxazone (CCC1P), an uncoupler of oxidative phosphorylation. Carnitine uptake was stimulated approximately 50% when kidney slices were treated with dibutyryl cAMP.  相似文献

The four isomers of N-ethylprotoporphyrin IX have been synthesized. The two isomers with the N-ethyl group on pyrrole rings A or B inhibit rat liver ferrochelatase as effectively as the corresponding N-methyl analogues, whereas those with the N-ethyl moiety on rings C or D are 30–100 times less effective. The ability of N-alkyl porphyrins to inhibit ferrochelatase thus depends not only on the size of the N-alkyl group but also on its precise location on the porphyrin face.  相似文献

An analysis has been made of the composition and structure of the two types of sheets assembled from material from dissociated bacteriophage T2 (Poglazov &; Mesyhanzhinov, 1967) and T4 capsids. Serological techniques have been used to show that both types of sheet are assembled from proteolytic fragment of P231, the major capsid constituent. The two types of sheets have been found to interconvert depending on the concentration of Mg2+ ions in the buffer. Computer modelling experiments show that the “hexagonal” and “rectangular” morphologies observed in the negative stain are due to in-register and staggered associations, respectively, of a single basic hexagonal lattice. Analysis by polyacrylamide gel electrophoresis of samples of sheets and dissociated capsids, together with previous results from immune electron microscopy (Kistler et al., 1978), suggest that hexamers of the proteolytic fragment are derived conservatively from capsomers of the phage head.The value of this proteolytic P23 fragment has been twofold: (1) it has proved to be a useful peptide in the ongoing primary sequence determination of P23 and (2) antibodies raised against it have been employed to follow the fate of P23 antigenic sites during various steps of phage capsid maturation (Kistler et al., 1978).  相似文献

We have critically evaluated hydrodynamic data from 21 proteins whose molecular dimensions are known from X-ray crystallography. We present two useful equations relating the molecular weights and sedimentation coefficients of globular proteins. The hydrodynamic data combined with data for small molecules from the literature indicate that failure of the Stokes equation occurs only for molecular weights <850. Calculated hydration values for the 21 proteins have a mean value and standard deviation of 0.53 ± 0.26 g H2O/g protein. Furthermore, statistical arguments indicate that only 5.3% of the variance is due to experimental error. The mean value and especially the dispersion of values are in sharp contrast to the values 0.36 ± 0.04 obtained by others from nmr measurements on frozen protein solutions. Hydration values calculated from nmr measurements are closely correlated with the number of charged and polar amino acid residues. In contrast to this result, our analysis of the amino acid compositions of the four proteins with the lowest hydration and the four monomeric proteins with the highest shows that the range of values we observe cannot be accounted for on the basis of amino acid composition. In fact there appears to be a weak correlation between the number of apolar residues and hydrodynamic hydration. We therefore conclude that the dispersion must result from variations in fine details of the surface structures of individual proteins. We propose a model of hemispherical clathrate cages which if correct, would account for the differences in the data obtained by these two methods.  相似文献
