Variation in respiratory traits was quantified between two populations of the sailfin molly Poecilia latipinna (one from a periodically hypoxic salt marsh, Cedar Key, and one from a chronically normoxic river site, Santa Fe River). Two suites of characters were selected: traits that may show both short‐term acclimation response and interdemic variation in acclimation response (metabolic rate, critical oxygen tension and respiratory behaviour), and those that are not likely to respond to short‐term acclimation but may vary among populations (gill morphometric characters). Sailfin mollies from the salt marsh, acclimated to hypoxia (1 mg l−1, c . 20 mmHg) for 6 weeks, spent less time conducting aquatic surface respiration and had lower gill ventilation rates than hypoxia‐acclimated conspecifics from the well‐oxygenated river site. Poecilia latipinna acclimated to hypoxia exhibited a lower critical oxygen tension ( P c) than fish acclimated to normoxia; however, there was also a significant population effect. Poecilia latipinna from Cedar Key exhibited a lower P c than fish from the Santa Fe River, regardless of acclimation. Cedar Key fish had a 14% higher mean gill surface area relative to fish from the Santa Fe River, a character that could account, at least in part, for their greater tolerance to hypoxia.  相似文献

Compromisation of food intake when confronted with diets deficient in essential amino acids is a common response of fish and other animals, but the underlying physiological factors are poorly understood. We hypothesize that oxygen consumption of fish is a possible physiological factor constraining food intake. To verify, we assessed the food intake and oxygen consumption of rainbow trout fed to satiation with diets which differed in essential amino acid (methionine and lysine) compositions: a balanced vs. an imbalanced amino acid diet. Both diets were tested at two water oxygen levels: hypoxia vs. normoxia. Trout consumed 29% less food under hypoxia compared to normoxia (p<0.001). Under both hypoxia and normoxia trout significantly reduced food intake by 11% and 16% respectively when fed the imbalanced compared to the balanced amino acid diet. Oxygen consumption of the trout per unit body mass remained identical for both diet groups not only under hypoxia but also under normoxia (p>0.05). This difference in food intake between diets under normoxia together with the identical oxygen consumption supports the hypothesis that food intake in fish can be constrained by a set-point value of oxygen consumption, as seen here on a six-week time scale.  相似文献

Escape performance was investigated in the golden grey mullet Liza aurata exposed to various levels of oxygen: >85 ( i.e. normoxia), 50, 20 and 10 % air saturation. Since the golden grey mullet performed aquatic surface respiration when air saturation approached 15–10 %, escape performance was tested at 10 % air saturation with and without access to the surface (10 % S and 10 % C, respectively). Various locomotor and behavioural variables were measured, such as cumulative distance, maximum swimming speed, acceleration, responsiveness (per cent of responding fish), response latency and directionality. Golden grey mullet showed a decrease in responsiveness when the oxygen level was reduced to 10 % air saturation, whether the surface access was obstructed or not. Hypoxia did not have any effect on the response latency. Cumulative distance and maximum swimming speed over a fixed time were significantly different between normoxic conditions and 10 % C, while no differences were found in maximum acceleration. While the fish's 'C‐bend' was mainly directed away from the stimulus in normoxia, the proportion of away and towards 'C‐bend' was random when the oxygen was ≤20 % air saturation. This suggested an impairment of the left‐right discrimination at the initiation of the fast start. Hypoxia affected golden grey mullet escape performance mainly through an impairment of responsiveness and directionality, while locomotor performance was affected only in severe hypoxia when the surface was obstructed. The study showed that, in addition to forcing the fish to the surface as shown by previous studies, hypoxia may also reduce fish elusiveness facing a predator by directly impairing its escape performance.  相似文献

The routine metabolic rate of Pseudocrenilabrus multicolor victoriae , laboratory-reared, F1 offspring of parents collected from a hypoxic swamp, did not differ between sibs raised under normoxia and hypoxia. In the offspring of parents originating from a river where the oxygen levels were generally high but fluctuating, the metabolic rate of fish raised under normoxia was higher than for sibs raised under extreme hypoxia. Although further experiments with a larger random sample of the parental stock will be necessary to validate the patterns, the difference between the experiments may indicate the selective environment of the parents.  相似文献

The armoured catfish, Liposarcus pardalis, tolerates severe hypoxia at high temperatures. Although this species can breathe air, it also has a strong anaerobic metabolism. We assessed tissue to plasma glucose ratios and glycogen and lactate in a number of tissues under "natural" pond hypoxia, and severe aquarium hypoxia without aerial respiration. Armour lactate content and adenosine in brain and heart were also investigated. During normoxia, tissue to plasma glucose ratios in gill, brain, and heart were close to one. Hypoxia increased plasma glucose and decreased tissue to plasma ratios to less than one, suggesting glucose phosphorylation is activated more than uptake. High normoxic white muscle glucose relative to plasma suggests gluconeogenesis or active glucose uptake. Excess muscle glucose may serve as a metabolic reserve since hypoxia decreased muscle to plasma glucose ratios. Mild pond hypoxia changed glucose management in the absence of lactate accumulation. Lactate was elevated in all tissues except armour following aquarium hypoxia; however, confinement in aquaria increased armour lactate, even under normoxia. A stress-associated acidosis may contribute to armour lactate sequestration. High plasma lactate levels were associated with brain adenosine accumulation. An increase in heart adenosine was triggered by confinement in aquaria, although not by hypoxia alone.  相似文献

Fifty‐three one‐sea‐winter Atlantic salmon Salmo salar (45–63 cm L T) were radio‐tagged in the Tana fjord, Barents Sea, in 1995. Thirty‐seven fish (70%) entered the freshwater zone of the River Tana in an average of 3 days after release in the fjord. The migration speeds in the lowest river section below the first riffle area were significantly higher than in the subsequent river section below the second riffle area. Similarly, the observed time spent in the first riffle area was significantly lower than in the next riffle area. The majority of Atlantic salmon entered the river during the hours of high tide and the subsequent ebb tide. In addition, most river entries were recorded around midnight. No effects of river flow on the river entry or migration speed were detected, but the migration speed of Atlantic salmon in both river sections examined was greater at lower temperatures. Twenty‐eight fish (72%) were recaptured in the river, 71% of them with weirs and gillnets, and 29% by rod and line. Over half of the Atlantic salmon (54%) were recaptured within 3 weeks following river entry, and within the first 100 km of the river (56%). The results are discussed in relation to earlier studies on multi‐sea‐winter Atlantic salmon in the River Tana.  相似文献

We present the first data on the effect of hypoxia on the specific dynamic action (SDA) in a teleost fish. Juvenile cod (Gadus morhua) were fed meals of 2.5% and 5% of their wet body mass (BM) in normoxia (19.8 kPa Po(2)) and 5% BM in hypoxia (6.3 kPa Po(2)). Reduced O(2) availability depressed the postprandial peaks of oxygen consumption, and to compensate for this, the total SDA duration lasted 212.0+/-20 h in hypoxia, compared with 95.1+/-25 h in normoxia. The percentage of energy associated with the meal digestion and assimilation (SDA coefficient) was equivalent between the different feeding rations but higher for fish exposed to hypoxia. Comparing peak oxygen consumption during the SDA course with maximum metabolic rates showed that food rations of 2.5% and 5% BM reduced the scope for activity by 40% and 55%, while ingestion of 5% BM in hypoxia occupied 69% of the aerobic scope, leaving little energy for other activities.  相似文献

The problem of availability of storage carbohydrates (fructans and starch) in oxygen‐deficient roots was investigated in wetland species ( Senecio aquaticus Hill., Myosotis palustris [L.] L. Em. Rchb.) and compared with related non‐wetland species ( Senecio jacobaea L., Myosotis arvensis [L.] Hill.) with respect to ethanolic fermentation (PDC activity, ethanol production).
In response to 24 h of hypoxic treatment, the pyruvate decarboxylase (PDC) activity in roots increased 4‐fold in M. arvensis and S. jacobaea , 2‐fold in S. aquaticus and only slightly in M. palustris . The rise in PDC activity was accompanied by an increase in ethanol content in the roots. The increase in ethanolic fermentation in roots of intact plants was associated with a slight increase in fructose and glucose and in a clear rise in sucrose content during the first 24‐48 h after commencement of the hypoxic treatment. Following 24 h of hypoxia, the content of fructans started to increase significantly for the duration of the experiment (9 days) in the four species. Since starch content changed only slightly during this period, the fructan:starch ratio increased under low energy availability. In the roots of flooding‐tolerant Senecio aquaticus , the ratio shifted most clearly from 2:1 in normoxia to 9:1 in hypoxia. For the roots of the two wetland species investigated, the results indicate a stronger accumulation of carbohydrates accompanied by a lower increase in PDC activity under root hypoxia, when compared with the related non‐wetland species respectively.  相似文献

本文旨在探讨线粒体ATP敏感钾(mitochondrial ATP-sensitive K+,MitoKATP)通道对大鼠肺动脉平滑肌细胞低氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)表达和细胞增殖的影响。原代培养大鼠肺动脉平滑肌细胞,分为常氧对照组、常氧+diazoxide(MitoKATP通道的选择性开放剂)组、常氧+5-hydroxydecanoate(5-HD,MitoKATP通道的选择性阻断剂)组、低氧对照组、低氧+diazoxide组、低氧+5-HD组,共6组,分别应用罗丹明123荧光技术检测各组大鼠肺动脉平滑肌细胞的线粒体膜电位,免疫组化检测HIF-1α的表达及酶联免疫检测仪检测细胞增殖的变化。结果显示,常氧+ diazoxide组与常氧对照组比较,罗丹明123荧光、HIF-1α表达及细胞增殖明显增强(P〈0.05);低氧+diazoxide组与低氧对照组比较,罗丹明123荧光、HIF-1α表达及细胞增殖明显增强(P〈0.05):常氧+5-HD组与常氧对照组比较,罗丹明123荧光、HIF-1α表达、细胞增殖没有明显变化(P〉0.05);但低氧+5-HD组与低氧对照组比较,罗丹明123荧光明显减弱、HIF-1α表达及细胞增殖有所减弱(P〈0.05)。结果提示:MitoKATP通道的开放能引起大鼠肺动脉平滑肌细胞线粒体膜去极化,并可以促进HIF-1α的表达及细胞增殖。  相似文献

We examined the energy status, nitrogen metabolism and hepatic glutamate dehydrogenase activity in the African lungfish Protopterus annectens during aestivation in normoxia (air) or hypoxia (2% O(2) in N(2)), with tissues sampled on day 3 (aerial exposure with preparation for aestivation), day 6 (entering into aestivation) or day 12 (undergoing aestivation). There was no accumulation of ammonia in tissues of fish exposed to normoxia or hypoxia throughout the 12-day period. Ammonia toxicity was avoided by increased urea synthesis and/or decreased endogenous N production (as ammonia), but the dependency on these two mechanisms differed between the normoxic and the hypoxic fish. The rate of urea synthesis increased 2.4-fold, with only a 12% decrease in the rate of N production in the normoxic fish. By contrast, the rate of N production in the hypoxic fish decreased by 58%, with no increase in the rate of urea synthesis. Using in vivo (31)P NMR spectroscopy, it was demonstrated that hypoxia led to significantly lower ATP concentration on day 12 and significantly lower creatine phosphate concentration on days 1, 6, 9 and 12 in the anterior region of the fish as compared with normoxia. Additionally, the hypoxic fish had lower creatine phosphate concentration in the middle region than the normoxic fish on day 9. Hence, lowering the dependency on increased urea synthesis to detoxify ammonia, which is energy intensive by reducing N production, would conserve cellular energy during aestivation in hypoxia. Indeed, there were significant increases in glutamate concentrations in tissues of fish aestivating in hypoxia, which indicates decreases in its degradation and/or transamination. Furthermore, there were significant increases in the hepatic glutamate dehydrogenase (GDH) amination activity, the amination/deamination ratio and the dependency of the amination activity on ADP activation in fish on days 6 and 12 in hypoxia, but similar changes occurred only in the normoxic fish on day 12. Therefore, our results indicate for the first time that P. annectens exhibited different adaptive responses during aestivation in normoxia and in hypoxia. They also indicate that reduction in nitrogen metabolism, and probably metabolic rate, did not occur simply in association with aestivation (in normoxia) but responded more effectively to a combined effect of aestivation and hypoxia.  相似文献

The effect of cortisol administration on reproductive performance was investigated in channel catfish Ictalurus punctatus broodfish. Cortisol was added to a commercial catfish feed by dissolving in ethanol and spraying the feed to yield a dietary concentration of 150 mg kg−1 feed. The cortisol diet and the control (no cortisol) diet were offered at a rate of 1% of biomass to three replicate ponds each containing 28 female and 14 male broodfish, respectively, three times per week for 11 weeks. Spawning began 10 days after the start of the experiment, and continued for 10 weeks. In fish fed cortisol, body mass and the hepato‐somatic index were reduced ( P  ≤ 0·02) and concentrations of plasma cortisol and glucose were significantly higher ( P  ≤ 0·0003) compared to those of controls. The relative frequency of spawning was similar between the two treatments; however, cortisol‐fed channel catfish had an average of 47·1% more spawns than the control‐fed fish. On average, there were 25·5 spawns per pond in the treated groups compared to 12·3 spawns per control pond ( P  = 0·10). No effect was observed on egg production, with individual egg mass, fecundity, and hatching success being similar ( P  ≥ 0·27) for both treatments. Despite the observed negative effects of cortisol on somatic and hepatic growth, the increased reproductive output coupled with no observable effects on the eggs or hatching success demonstrates that cortisol does not suppress channel catfish reproduction.  相似文献

The present study evaluates the influence of previous nutritional status, fish fed on diets supplemented with tea and methionine, on acute hypoxia tolerance and subsequent recovery of Sparus aurata juveniles. Four isonitrogenous (45% of protein) and isolipidic (18% lipid) diets were formulated to contain 0.3% methionine, 2.9% white tea dry leaves or 2.9% of white tea dry leaves+0.3% methionine. An unsupplemented diet was used as control. Hepatic key enzymes of intermediary metabolism and antioxidant status, superoxide dismutase isoenzyme profile, glutathione (total, reduced and oxidized) and oxidative damage markers were determined under normoxia, hypoxia challenge and during normoxia recovery. Dietary white tea inclusion decreased plasma glucose levels under normoxia and seemed to induce an increase in anaerobic pathways as showed by enhanced liver lactate dehydrogenase activity. Hypoxia challenge reversed some of the responses induced by diet tea supplementation. Hypoxia decreased plasma glucose levels, increased glucose 6-P-dehydrogeanse activity, decreased superoxide dismutase activity (especially Mn-SOD and CuZn-SOD isoforms) and increased glutathione peroxidase activity in all dietary treatments. Catalase activity during hypoxia varied with dietary treatments and glutathione reductase was not modified. Antioxidant defenses were insufficient to avoid an oxidative stress condition under hypoxia, independently of dietary treatment. In general, pre-challenge values were recovered for almost all parameters within 6 h recovery time.  相似文献

The bimodally respiring catfish Clarias macrocephalus Günther responded to a toxic extract of Croton tiglium (Euphorbiaceae) seeds by increased air breathing under both normoxic (8.1 ± 0.4 mgO2 l−1) and hypoxic (0.7 ± 0.1 mgO2l−1) conditions. Fish in hypoxia survived longer than those in normoxia when surface access was provided. When air breathing was prevented, survival time in toxin was greatly reduced at both levels of dissolved oxygen, and fish in normoxia survived longer than those in hypoxia. Non-toxin controls without surface access survived in normoxia but in hypoxia died at the same time as the fish in toxin. These results suggest that air breathing increases the resistance offish to toxins by permitting a decrease in the rate of gill ventilation and hence the rate at which toxins are absorbed.  相似文献

Fructose1,6diphosphate (FDP) is a glycolytic intermediate which has been theorized to increase the metabolic activity of ischemic tissues. Here we examine the effects of externally applied FDP on cardiomyocyte uptake and metabolism. Adult rat cardiomyocytes were isolated and exposed to varying concentrations (0, 5, 25 and 50 mM) of FDP for either 1, 16 or 24 h of hypoxia (95% N2/5% CO2), each time period followed by a 1 h reoxygenation (95% air/5% CO2). The uptake of FDP by rat cardiomyocytes was more concentrationdependent than timedependent. Furthermore, the uptake of FDP by the cardiomyocytes was similar in the hypoxia and normoxia treated cells. Alamar Blue, a redox indicator that is sensitive to metabolic activity, was used to monitor the effects of the FDP on cardiomyocyte metabolism. In the 1 h hypoxia or normoxia group, the 5, 10 and 25 mM FDP showed a significant increase in metabolism compared to the control cells. When the length of hypoxia was extended to 16 h, all doses of FDP were greater than control. And at the 24 h hypoxia or normoxia time period, only the 10, 25 and 50 mM FDP groups were greater than control. The results indicate a non-linear trend between the external concentration of FDP and the changes noted in metabolism. The findings from this study indicate that a narrow concentration range between 5–10 mM augments cardiomyocyte metabolism, but higher or lower doses may have little additional affect.  相似文献

The effect of chloride cell proliferation on the respiratory function was evaluated by measuring oxygen consumption (VO2) and ventilatory parameters during normoxia and gradual hypoxia in the tropical fish Hoplias malabaricus. Chloride cell proliferation was induced by keeping fish in deionized water, and the effect on the respiratory function was measured on the 1st, 2nd, and 7th day in this water using a flow-through respirometry system. Plasma osmolarity and the partial pressure of arterial oxygen (PaO2) and carbon dioxide (PaCO2) were measured under conditions of normoxia and severe hypoxia. Chloride cell proliferation on the lamellae significantly increased the water-blood diffusion distance on the 2nd and 7th day in deionized water. VO2 was kept constant until the critical oxygen pressure (PcO2) of 21.6+/-0.9 mmHg in both the control and deionized water fish was reached. The ventilatory parameters were higher in deionized water fish in normoxia, and increased during hypoxia, matching decreases in the water's partial O2 pressure. Impairment of the respiratory function was evidenced by the decrease of PaO2 of deionized water fish in normoxic condition. However, despite the changes in the epithelial morphology of gills in fish kept in deionized water, H. malabaricus proved be a hypoxic-tolerant tropical species.  相似文献

The objective of this study was to identify behavioural adjustments leading to avoidance of hypoxia. Using the oxygen-sensitive species rainbow trout Oncorhynchus mykiss as a model, individual fish were recorded while moving freely between two sides of a test arena: one with normoxia and one with stepwise progressive hypoxia [80-30% dissolved oxygen (DO) air saturation]. The results demonstrated a gradual decrease in the total time spent in hypoxia starting at 80% DO air saturation. At this DO level, the avoidance of hypoxia could not be attributed to changes in spontaneous swimming speed, neither in normoxia nor in hypoxia. Reducing the DO level to 60% air saturation resulted in decreased spontaneous swimming speed in normoxia, yet the number of trips to the hypoxic side of the test arena remained unchanged. Moreover, data revealed increased average residence time per trip in normoxia at DO levels ≤60% air saturation and decreased average residence time per trip in hypoxia at DO levels ≤50% air saturation. Finally, the spontaneous swimming speed in hypoxia increased at DO levels ≤40% air saturation and the number of trips to hypoxia decreased at the 30% DO air saturation level. Thus, avoidance of the deepest hypoxia was connected with a reduced number of trips to hypoxia as well as decreased and increased spontaneous swimming speed in normoxia and hypoxia, respectively. Collectively, the data support the conclusions that the mechanistic basis for avoidance of hypoxia may (1) not involve changes in swimming speed during mild hypoxia and (2) depend on the severity of hypoxia.  相似文献

The effect of intrauterine growth retardation (IUGR) on striatal energy metabolites and amino acid concentrations was studied in the fetuses of eight nulliparous rat dams after uterine artery ligation on day 18 of gestation. On day 22 (term = 23), four dams were subjected to normoxia and four to hypoxia (10% oxygen) for 58 min, while monitoring hemodynamics and blood gases. After decapitation of the dam, fetuses were delivered by sectio and decapitated. The measured parameters in the dams were stable under normoxia but exhibited decreased oxygen availability under hypoxia. Striatal energy balance was preserved in IUGRs, both under maternal normoxic and hypoxic conditions, compared to appropriately grown (AGA) littermates. Under maternal normoxia, the striatal concentration of aspartate was reduced (P < 0.01) in IUGRs and the level of alanine was increased (P < 0.01) as compared to AGAs. Under hypoxia, the level of GABA was higher in IUGRs (P < 0.01). Lactate was increased in all fetuses under hypoxia. It is concluded that striatal energy metabolism is preserved in IUGR rat fetuses in late gestation under both maternal normoxia and hypoxia. Amino acid metabolism, however, is disturbed and depends on the degree of growth retardation and on the severity of perinatal stress.  相似文献

Lymnaea stagnalis were exposed to hypoxic and chemical challenges while ventilation, heart rate and metabolism were monitored. Hypoxia increased ventilatory behavior, but this response was eliminated by immersion in 0.75 mM nitric oxide synthase (NOS) inhibitor, 7-nitroindazole (7 NI). 7 NI also suppressed ventilatory behavior under normoxia. 10.0 mM L-arginine (ARG, the NOS substrate) increased ventilatory behavior under normoxia, but dampened the hypoxic response. The heart-rate response to NOS inhibition exhibited dose-dependent contradictory characteristics. Under both normoxia and hypoxia 0.25 mM 7 NI increased heart rate, while 0.75 mM 7 NI suppressed it. The effect of 0.50 mM 7 NI depended on whether normoxia or hypoxia was coincident; under normoxia 0.50 mM 7 NI increased heart rate, while under hypoxia this concentration suppressed heart rate. Exposure to ARG did not elicit dose-dependent contradictory responses. Heart rate increased when treated with 10.0 mM ARG under normoxia and hypoxia, while 1.0 mM ARG increased heart rate only under hypoxia. Metabolic responses to NOS inhibition also exhibited dose-dependent contradictory changes. V.O2 decreased over 60% in response to 0.75 mM 7 NI, and baseline V.O2 was restored when exposure ceased. In contrast, 0.25 mM 7 NI increased V.O2 10%, and the increase continued after exposure ceased. 0.50 mM 7 NI decreased V.O2 40%, but V.O2 increased when exposure ceased. ARG had only the effect of increasing V.O2, and only at 10.0 mM concentration. Based on these results and on NO's known role as a neuromodulator, we conclude that the cardio-respiratory responses to hypoxia are, in part, mediated by NO.  相似文献

Endurance and strength training are established as distinct exercise modalities, increasing either mitochondrial density or myofibrillar units. Recent research, however, suggests that mitochondrial biogenesis is stimulated by both training modalities. To test the training "specificity" hypothesis, mitochondrial respiration was studied in permeabilized muscle fibers from 25 sedentary adults after endurance (ET) or strength training (ST) in normoxia or hypoxia [fraction of inspired oxygen (Fi(O(2))) = 21% or 13.5%]. Biopsies were taken from the musculus vastus lateralis, and cycle-ergometric incremental maximum oxygen uptake (VO(2max)) exercise tests were performed under normoxia, before and after the 10-wk training program. The main finding was a significant increase (P < 0.05) of fatty acid oxidation capacity per muscle mass, after endurance and strength training under normoxia [2.6- and 2.4-fold for endurance training normoxia group (ET(N)) and strength training normoxia group (ST(N)); n = 8 and 3] and hypoxia [2.0-fold for the endurance training hypoxia group (ET(H)) and strength training hypoxia group (ST(H)); n = 7 and 7], and higher coupling control of oxidative phosphorylation. The enhanced lipid oxidative phosphorylation (OXPHOS) capacity was mainly (87%) due to qualitative mitochondrial changes increasing the relative capacity for fatty acid oxidation (P < 0.01). Mitochondrial tissue-density contributed to a smaller extent (13%), reflected by the gain in muscle mass-specific respiratory capacity with a physiological substrate cocktail (glutamate, malate, succinate, and octanoylcarnitine). No significant increase was observed in mitochondrial DNA (mtDNA) content. Physiological OXPHOS capacity increased significantly in ET(N) (P < 0.01), with the same trend in ET(H) and ST(H) (P < 0.1). The limitation of flux by the phosphorylation system was diminished after training. Importantly, key mitochondrial adaptations were similar after endurance and strength training, regardless of normoxic or hypoxic exercise.
