BackgroundAflatoxins are one of the important environmental factors that pose a risk to living organisms. On the other hand, it has been indicated in research that boron intake has beneficial effects on organisms. In this study, the effect of boron was disclosed in rats exposed to aflatoxin B1 (AFB1), which poses a toxicological risk.MethodsA total of 36 male Sprague Dawley rats were separated into 6 groups and 0.125 mg/kg bw AFB1 and 5, 10, or 20 mg/kg bw doses of boron were given orally for 21 days. End of the experiment, biochemical, molecular, and histopathological analyses were performed.ResultsAFB1 treatment increased liver enzyme activities (AST, ALT, and ALP) and malondialdehyde level; on the other hand, it caused a decrease in glutathione level, superoxide dismutase and catalase activities. In addition, the mRNA expression levels of apoptotic (Bax, Caspase-3, Caspase-8, Caspase-9, and p53) and pro-inflammatory (TNF-α and NFκB) genes increased and the mRNA expression of the anti-apoptotic gene (Bcl-2) decreased in liver tissue. Also, AFB1 treatment increased DNA damage and caused histopathological alterations in the liver tissue. Additionally, boron applications at doses of 5, 10, and 20 mg/kg bw given with AFB1 reversed these negative changes.ConclusionsAs a result, boron exhibited hepatoprotective effect together with antioxidant, anti-inflammatory, and anti-apoptotic effects against AFB1-induced liver damage.  相似文献

The reported beneficial effects of boron on mineralized tissues in animals and humans vary. Thus, a study was performed to assess whether the variability was the result of different forms of boron supplementation, method of supplementation, and increased adiposity of the rabbit experimental model.Thirty-one female New Zealand White rabbits, (aged 8 months, 2–2.5 kg weight) were fed a grain-based high energy diet containing 11.76 MJ/kg (2850 kcal/kg) and 3.88 mg boron/kg. The rabbits were randomly divided into four treatment groups: Control group was not supplemented with boron (n:7; C), and three groups supplemented with 30 mg boron/L in drinking water in the forms of borax decahydrate (Na2O4B7 10H2O, n:10; BD), borax anhydrous (Na2O4B7, n:7; Bah) or boric acid (H2BO3, n:7; BA). Cone beam micro computed tomographic (micro-CT), histological and elemental analysis was used to evaluate the bones/teeth.Results of the experiments demonstrated that boron supplementation had beneficial effects on mineralized tissue but varied with the type of treatment. Mineral density of the femur was increased by the Bah and BA treatments (p < 0.001), but only BA increased mineral density in the tibia (p = 0.015). In incisor teeth, mineral density of dentin was increased by all boron treatments (p < 0.001), and mineral density of enamel was increased by the BD and Bah treatments. Mineral analysis found that all boron treatments increased the boron concentration in tibia and femur. In the tibia, both the BD and Bah treatments decreased the iron concentration, and the BD treatment decreased the magnesium concentration. Sodium and zinc concentrations in the tibia were decreased by the Bah and BA treatments. The boron treatments did not significantly affect the calcium, copper, molybdenum, potassium phosphorus, and sulfur concentrations.The findings show that boron supplementation can have beneficial effects on mineralized tissues in an animal model with increased adiposity, which is a model of increased inflammatory stress. However, this effect varies with the form of boron supplemented, the method of supplementation, and the mineralized tissue examined.  相似文献

IntroductionBoron (B) is thought to play key role in proper bone growth and development as well as have some role in regulation of minerals such as calcium (Ca), phosphorus (P) and magnesium (Mg) which act synergistically with vitamin D.ObjectivePresent study was planned in two phases to assess the effect of optimum and supranutritional levels of (B) in the form of boric acid (BA) supplementation on bone health of growing cross bred calves.MethodDuring Phase-1, twenty four male crossbred calves were blocked into four groups (n = 6) on the basis of their body weight (154.83 ± 8.5 kg), age (7–9 months) and were supplemented with 0 (C), 2.6 (T-1), 5.4 (T-2) and 10.7 (T-3) g BA for appropriate B (0.175 adjustment factor to calculate B form BA) consumption i.e. 0, 100, 200 and 400 ppm in each group respectively, for 90 days. During phase 2, twenty-one male crossbred calves were divided into 3 groups (n = 7) on the basis of their body weight (103.76 ± 4.34 kg) and age (5–8 months). All the groups were on similar dietary regimen with additional supplementation of boric acid as 0 g (control); 3.6 g (200 ppm B; T-1) and 10.8 g (600 ppm B; T-2), respectively for a period of 120 d.ResultsFrom the first experiment it is reported that plasma levels of bovine alkaline phosphatase (BALP), type I collagen cross-linked N-telopeptide (NTx) and Ca were significantly (P < 0.05) affected in T-2 and T-3 groups as compared to T-1 and control groups. Whereas, plasma osteocalcin (OCN) concentration was found to be higher in T-2 and T-3 groups as compared to control group. However, plasma concentrations (ng/mL) of tartrate resistant acid phosphatase (TRAP) remained unaltered due to dietary treatments. Based on the results, another experiment was conducted to validate the above findings and further to determine the effect of still higher i.e supranutritional levels of BA supplementation on bone health of calves. Results revealed that supplementation of BA in T-2 group had no beneficial effect on bone health as the plasma concentration of BALP, OCN, NTx, 25 (OH) vitamin D and Ca as compared to T-1 group in phase 2. Other possible attributes of bone health i.e. plasma concentration of Mg, P, parathyroid hormone (PTH), and calcitonin were not affected by BA supplementation at any levels.ConclusionOverall from present study it can be concluded that supplementation of boric acid 3.6 g/d (equivalent to 200 ppm B) in the diet of growing animals has positive effect on bone health related biomarkers (OCN, NTx and BALP) and supplementation of supranutritional level of BA i.e. 10.8 g (equivalent to 600 ppm B) level had neither additional beneficial nor harmful effect on bone health of calves.  相似文献

Smallmouth bass (Micropterus dolomieu) were collected to quantify the nature and prevalence of biomarker responses, including biochemical indices, toxicopathic lesions and general health indices, among fish collected from polychlorinated biphenyl (PCB)-contaminated and nearby uncontaminated reaches of the Kalamazoo River, Michigan, USA. Blood and tissue samples (gill, liver, spleen, head kidney, trunk kidney, thyroid and gonads) were collected and preserved at necropsy for biochemical and histological analyses. The body condition factor and liver somatic index were significantly lower in fish collected from the downstream, contaminated site. Plasma vitellogenin was not detected in male fish collected from either site. Liver ethoxyresorufin-O-deethylase activity and liver and spleen superoxide dismutase activity were significantly depressed in fish collected from the downstream site. Significant toxicopathic lesions such as glycogen depletion, enhanced macrophage aggregates, hepatic foci of cellular alteration (i.e. preneoplastic lesions) and neoplasia were also detected in the liver of fish collected from the downstream site. This study indicates that many of the biochemical and histopathological biomarker responses were associated with liver and body tissue PCB concentrations. Taken together, the biomarkers of exposure and effect strongly suggest that fish within the downstream site are adversely affected by PCBs and other chemical stressors.  相似文献

A systematic procedure is provided for determination of D-amino acid oxidase (EC 1.4.3.3) activity in fish tissues. Activity was surveyed in crude liver homogenates of twenty-three species of fish. Activity ranged from 0-047 to 3-12 units (ρmol product produced per min) per gram wet weight of tissue. Activity was highest in liver, with significant levels occurring in kidney and pyloric caecae. It was below the limit of detection in brain, heart, spleen, testis, gill filaments, and muscle. Some properties of the enzyme from sockeye salmon, Oncorhynchus nerka (Walbaum), were studied. Knowledge of occurrence and levels of D-amino acid oxidase in fish may be useful to fish aquaculturists contemplating supplementation of feed with racemic mixtures of amino acids.  相似文献

The effects of dietary boric acid on bone strength in rats   总被引:4,自引:0,他引:4  
The effects of dietary boron (B) (from boric acid [BA]) on bone strength were evaluated using male F344 rats. B was administered by dietary admixture of BA to NIH-07 feed at concentrations of 200, 1000, 3000, and 9000 ppm. The latter two levels were found in previous studies to be reproductively toxic to both males and the developing fetus. The first two levels are below and just at, respectively, the levels for producing fetal malformations, and are below the dose required to produce male reproductive toxicity. Resistance to destructive testing was measured on femora, tibiae, and lumbar vertebrae. Although femur and tibia resistance to bending force were not affected by any amount of dietary B, vertebral resistance to a crushing force was increased by ≈10%, at all dose levels (200-9000 ppm). These data show that even levels of BA that are not reproductively toxic can affect the strength of the axial skeleton in rats.  相似文献

An active monitoring (caging experiment) and a passive monitoring (sampling of wild fish) were performed to investigate the effects of effluent from a sewage treatment works (STW) on brown trout (Salmo trutta) by histopathological examinations of the skin, gill, liver and kidney. Histopathological lesions were evaluated according to a standardised assessment tool, which allows calculation of indices for every organ. According to the results of both monitorings, trout exposed to river water supplemented with treated waste water from the STW Lyss showed higher histopathological indices than trout caught upstream of the discharge point of the STW or kept in river water only. These results indicate a negative effect of treated waste water from the STW on the histopathological status of the examined organs of brown trout. Both monitoring approaches revealed the liver to be the most affected organ compared with reference fish. However, data from the two monitoring approaches were not completely consistent: histologically the gills were the most sensitive organ to the effects of treated waste water in the active monitoring, but were not affected in the passive monitoring. The data provide relevant information about both the comparability and the pros and cons of the two monitoring approaches to assess effects of pollution on histopathological alterations in fish.  相似文献

Short-and long-term oral exposures to boric acid (BA) in laboratory animals and birds caused toxic effects. However, the toxicity data on adult poultry skeletal muscles of BA was not documented with metabolic studies. Livability, weight gain, and feed conversion might be adversely affected in broilers as a result of changes in energy metabolism. Therefore, this study was conducted to investigate the influences of acute BA doses on energy metabolism of chick pectoral muscle (PM). Chicks were fed by giving the aqueous solutions supplemented with BA (0, 0.27, 0.54, 1.08, 1.35, 2.25, 3.375 and 4.5 mmol B/kg b.wt.). Breast muscle samples were taken at 24, 48 and 72 h and analyzed histochemically, ultrastructurally and biochemically. Data collected in these analyses indicated that consumption of diets containing up to 2.25 mmol B/kg at 24, 48 and 72h was not detrimental to broiler PM. However, 3.375 mmol B/kg b.wt. (at 24 h) and 4.5 mmol B/kg b.wt. (at 24 and 48 h) caused decreased metabolite concentrations (glucose, glycogen, lactate and ATP) in muscle fibers (Type IIB). Subsarcolemmal (SS) mitochondria and intermyofibrillar (IM) mitochondrial damage (cristae dissolution) were also observed by toxic effect of BA (4.5 mmol B/kg b.wt.). These observations proved that BA at the high doses (3.375 and 4.5 mmol B/kg b.wt.) causes to altered energy metabolism in Type IIB as dependent on time. Based on these results we think that energy protection in muscle against BA toxicity will be the most important study subject. Thus, high BA doses will not have detrimental effects on broiler performance.  相似文献

Boron (B) is a developmental and reproductive toxin. It is also essential for some organisms. Plants use uptake and efflux transport proteins to maintain homeostasis, and in humans, boron has been reported to reduce prostate cancer. Ca2+ signaling is one of the primary mechanisms used by cells to respond to their environment. In this paper, we report that boric acid (BA) inhibits NAD+ and NADP+ as well as mechanically induced release of stored Ca2+ in growing DU-145 prostate cancer cells. Cell proliferation was inhibited by 30% at 100μM, 60% at 250μM, and 97% at 1,000μM BA. NAD+-induced Ca2+ transients were partly inhibited at 250μM BA and completely at 1,000μM BA, whereas both NADP+ and mechanically induced transients were inhibited by 1,000μM BA. Expression of CD38 protein increased in proportion to BA exposure (0–1,000μM). In vitro mass spectrometry analysis showed that BA formed adducts with the CD38 products and Ca2+ channel agonists cyclic adenosine diphosphate ribose (cADPR) and nicotinic acid adenine dinucleotide phosphate (NAADP). Vesicles positive for the Ca2+ fluorophore fluo-3 acetoxymethyl ester accumulated in cells exposed to 250 and 1,000μM BA. The BA analog, methylboronic acid (MBA; 250 and 1,000μM), did not inhibit cell proliferation or NAD+, NADP+, or mechanically stimulated Ca2+ store release. Nor did MBA increase CD38 expression or cause the formation of intracellular vesicles. Thus, mammalian cells can distinguish between BA and its synthetic analog MBA and exhibit graded concentration-dependent responses. Based on these observations, we hypothesize that toxicity of BA stems from the ability of high concentrations to impair Ca2+ signaling.  相似文献

Our study was designed to evaluate effects of an herbicide, pendimethalin on biochemical biomarkers and histopathological indices of the freshwater fish Channa punctata Bloch. Fish were acutely exposed (96 h) to sub-lethal concentrations (0.5 and 0.8 ppb of pendimethalin). Various oxidative stress indicators such as thiobarbituric acid reactive substances levels and protein carbonyl content, as well as antioxidant defenses parameters, such as glutathione-S-transferase (GST), catalase (CAT), reduced glutathione (GSH) and non-protein thiols (NP-SH) levels were studied, using the liver, kidney and gill tissues. Pendimethalin exposure increased lipid peroxidation and protein oxidation processes. There was significant inhibition in levels of GSH and NP-SH. The activity of antioxidant enzymes GST and CAT depleted in all the tissues in a dose dependent manner. The histopathological change in the gill showed necrosis and atrophy of primary and secondary gill lamellae. The tissue damages like degeneration of cytoplasm in hepatocytes, atrophy, formation of vacuoles, are some histopathological changes observed in the liver. The changes in histoarchitechture observed in the kidney included necrosis, cellular hypertrophy and granular cytoplasm. The present study demonstrates the disturbances in antioxidant armamentarium and importance of study in the potential risk assessment of herbicides on fish species.  相似文献

Increased synthesis of heat shock protein 70 (Hsp70) occurs in prokaryotes and eukaryotes in response to physiological, environmental, and chemical exposures, thus allowing the cell survival from fatal conditions. Hsp70 cytoprotective properties may be clarified by its anti-apoptotic function. Boron has been reported to play an essential role in various organ developments and metabolisms. However, it is not known if boron is also able to modulate the Hsp70. In the present study, the actions of boron on ostrich spleen and expression level of Hsp70 were investigated. Thirty healthy ostrich chicks were randomly assigned to six groups: groups I, II, III, IV, V, and VI and fed the basal diet spiked with 0-, 40-, 80-, 160-, 320-, and 640-mg boric acid (BA)/L, respectively, in drinking water. The histomorphological examination in the spleen was done by hematoxylin and eosin (HE) staining. The expression level of Hsp70 was analyzed by immunohistochemistry (IHC) and western blotting, and mRNA expression of Hsp70 was investigated by quantitative real-time PCR (qPCR). In order to investigate apoptosis, TUNEL assay reaction in all treatment groups was analyzed. Our results showed that the histological structure of spleen up to 160 mg/L BA supplementation groups well developed. The Hsp70 expression level first induced at low-dose groups (up to group IV) and then inhibited dramatically in high-dose groups (V and VI) while comparing with the group I (0 mg BA). The TUNEL assay reaction revealed that the cell apoptosis amount was decreased in group IV, but in group V and especially in group VI, it was significantly increased (P < 0.01). Taken altogether, proper dietary boron treatment might stimulate ostrich chick spleen development by promoting the Hsp70 expression level and inhibiting apoptosis, while a high amount of boron supplementation would impair the ostrich spleen structure by inhibiting Hsp70 expression level and promoting cell apoptosis.  相似文献

Silver nanoparticles (Ag-NPs) have wide applications in the medical field; however, the toxicological effects are still poorly studied. The study was aimed to determine the effects of 15.78 nm spherical and amine-coated Ag-NPs on hematology and histology of gills and liver tissues in 28 days treated Labeo rohita (L. rohita). It was found that Ag-NPs induced alterations in the hematological parameters in a dose dependent manner. The Ag-NPs also induced histological alterations in a dose-dependent manner. In gill tissues, it induced fusion of secondary lamellae, separation of gill epithelium, fusion and necrosis of lamellar cells, hyperplasia, deformed cartilaginous skeleton, separation and lifting of epithelium, and curling of lamellae in a dose dependent manner. In the liver, Ag-NPs produced abnormalities in hepatic tissues by reducing the size of hepatocytes and nuclei, and stimulated the production of necrotic and apoptotic bodies. It was concluded that Ag-NPs are toxic to aquatic organisms and induce hematotoxicity and histopathological conditions in exposed fish.  相似文献

BackgroundSelenium (Se) appears in the selenoproteins in the form of selenocysteine (Sec) and is important for the growth and development of vertebrates. The present study characterized seven selenoproteins, consisting of the GPX1, GPX3, GPX4, SELENOW, SELENOP, TXNRD2 and TXNRD3 cDNAs in various tissues of yellow catfish, explored their regulation to dietary Se addition.Methods3′ and 5′ RACE PCR were used to clone full-length cDNA sequences of seven selenoprotein genes (GPX1, GPX3, GPX4, SELENOW, SELENOP, TXNRD2 and TXNRD3). Their molecular characterizations were analyzed, including conservative motifs and the SECIS elements. The phylogenetic trees were generated through neighbor-joining (NJ) method with MEGA 6.0 with 1000 bootstrap replications. Quantitative real-time PCR was used to explore their mRNA tissue distribution in the heart, anterior intestine, dorsal muscle, head kidney, gill, liver, brain, spleen and mesenteric fat. Yellow catfish (mixed sex) were fed diets with dietary Se contents at 0.03 (low Se), 0.25 (adequate Se) and 6.39 (high Se) mg Se/kg, respectively, for 12 weeks, and their spleen, kidney, testis and brain were used for the determination of the mRNA levels of the seven selenoproteins.ResultsThe seven selenoproteins had similar domains to their corresponding members of other vertebrates. They were widely expressed in nine tissues, including heart, liver, brain, spleen, head kidney, dorsal muscle, mesenteric fat, anterior intestine and gill, but showed tissue-dependent expression patterns. Dietary Se addition affected the expression of the seven genes in spleen, kidney, testis and brain tissues of yellow catfish.ConclusionTaken together, our study demonstrated the characterization, expression and regulation of seven selenoproteins, which increased our understanding of the biological functions of Se and selenoproteins in fish.  相似文献

热激蛋白(heat shock protein,HSP)是进化上非常保守的蛋白质家族之一,普遍存在于各种生物体中,在多种生理活动中起到重要作用。该实验克隆了大黄鱼HSP90基因,并分析了温度和病原菌感染对其表达的影响。克隆到的大黄鱼HSP90序列长3 930 nt,含4个外显子和3个内含子,其中编码区2 178 nt,编码725个氨基酸。同源性分析发现,大黄鱼HSP90基因序列和其它鱼类的同源性在90%以上。在不同水温下,HSP90基因在不同组织中的表达量变化不同,心、肠和脑等组织在29 oC时表达量最高,而肌肉、脾和肝等组织在24 oC时表达量最高。用病原菌感染大黄鱼,感染48 h后,鳃、心、脾、肠、肾和脑组织HSP90的表达量明显上升;发病时(感染7天后),受检的9种组织中HSP90的表达量都比未感染时明显增加。  相似文献

In plant species not containing polyols, boron (B) is regarded as practically phloem immobile. This has been explained by the high membrane permeability of boric acid (BA) resulting in a rapid efflux out of the phloem and re-transport into the leaf in the xylem. The present study investigated how the xylem flow rate affects the phloem mobility of foliar-applied BA in Ricinus communis L. cv. Impala. Xylem flow rates were varied by exposure of the canopy to different levels of relative humidity (RH). In seedlings with severed hypocotyls, i.e. without xylem flow, B was highly mobile. In intact seedlings and plants, the degree of mobility and the within-plant distribution of B were strongly RH-dependent. At RH of 70% or above, up to 16–24% of the B was translocated to other plant parts, whereas at lower RH no significant movement of B was detected. Only at an intermediate RH (70–80%), did leaf-applied B accumulate in roots. At 100% RH, B transport in the xylem was significantly increased, suggesting that the build up of root pressure induced the recycling of phloem delivered B from roots to shoots. These results indicate that in R. communis phloem B mobility is not constant, but strongly affected by transpiration rates.  相似文献

Gill, trunk kidney, spleen, and liver of rainbow trout (Oncorhynchus mykiss) were examined after exposure to different sublethal concentrations of carbosulfan (25, 50 and 200 μg L−1), propineb (3, 6 and 24 mg L−1), and benomyl (2, 5 and 20 mg L−1) for 14 days. Lesions were observed in gill, trunk kidney, spleen, and liver of rainbow trout exposed to either concentration of pesticides. The most important lesions were determined in the highest concentrations of pesticides. Lamellar fusion, lamellar hyperplasia, epithelial lifting, vacuolization of epithelial tissue, epithelial necrosis, hypertrophy and sloughing of epithelium were observed on fish exposed to carbosulfan, propineb and benomyl. Fish had cell necrosis, degeneration and oedemas in liver, trunk kidney and spleen. None of these lesions were seen in control fish.  相似文献

黏质沙雷氏菌(Serratia marcescens)是引起人类、动物及植物感染的重要条件致病菌,但其作为鱼类致病菌却鲜有报道。【目的】本研究以从患病牙鲆(Paralichthys olivaceus)病灶处分离的一株黏质沙雷氏菌YP1为研究对象,分析黏质沙雷氏菌对鱼类的致病性及对疾控的影响。【方法】利用形态学、分子生物学及生理生化实验综合鉴定菌株YP1;利用菌株YP1进行人工感染实验、组织病理实验及药敏试验,研究其感染症状、组织病理学、毒力和药物敏感性。【结果】分离自患病牙鲆体表溃疡病灶处的菌株YP1鉴定为黏质沙雷氏菌。感染实验结果显示,牙鲆和斑马鱼的半数致死量(LD50)分别为3.44×107CFU/g和6.28×105CFU/g,除牙鲆外菌株YP1对其他鱼类也具有高致病性;菌株YP1主要导致牙鲆腹水,同时伴有呼吸急促、摄食减弱、脱肛、白便、鳃缺血及多脏器膨大出血等症状,并随着感染时间的延长对脏器损伤呈加重趋势。病理组织切片结果显示,菌株YP1对牙鲆鳃、肠、肝、脾、肾、心均造成损伤。药敏试验结果表明,YP1对左氧氟沙星、诺氟沙星等14种药物敏感;但对氨苄西林、头孢拉定等19种药物具有耐药性。【结论】本研究结果证实了黏质沙雷氏菌是能导致牙鲆腹水病的一种病原菌,同时对其他鱼类也具高致病性,为该菌感染鱼类导致疾病的检测、鉴别和防治提供科学依据。  相似文献

KLF6具有抗细胞增殖的特性,在抑制肿瘤细胞方面起着重要作用.在获得KLF6基因EST序列的基础上,克隆到了KLF6基因,全长1185 bp,其中5′端198 bp,3′端333 bp,开放阅读框654 bp,编码217个氨基酸.与KLF转录因子家族其它成员一样,大黄鱼KLF6的C-端含有3个连续的C2H2型锌指结构域.其氨基酸序列高度保守,与其它鱼类的同源性在90%以上.在检测的大黄鱼骨骼肌、肝脏、眼、脑、脾、心、鳃、肠和肾等9种组织中,KLF6都有不同程度的表达,其中肾、肝脏、鳃、脾脏和脑等组织中的表达量较高.KLF6表达范围的广泛性提示其在多种组织中参与细胞功能活动的调控.  相似文献

A new class of drugs, borylated derivatives of ferrocenium compounds, which show a comparatively facile synthesis is investigated on their boron neutron capture accumulation. Investigations focused on the fast and effective testing of 12 ferrocene derivatives with tetracoordinated boron atoms, which should accumulate in rodent tumors. The macroscopic studies on time-dependent boron distributions and boron concentrations in mice were carried out by inductively coupled plasma-atomic emission spectrometry, inductively coupled plasma-mass spectrometry, and quantitative neutron capture radiography. The determination of boron concentrations after injection of 2b showed high boron contents in spleen, liver, kidneys, less in lung and muscle, and poor in integral blood, blood plasma, tumor, and brain.
