The morphology and infraciliature of two hypotrichous ciliates, Urosoma salmastra and U. karinae sinense nov. sspec., were investigated for populations collected from the surface of intertidal gravel in the Huguang Mangrove Forest, Zhanjiang, China and the upper 10 cm layer of soil in the Sangke Grass Land in the southern part of Gansu Province, China, respectively. Urosoma salmastra is characterized by its elongate-elliptical body with no tail-like structure; two macronuclear nodules; cortical granules colourless, less than 1 μm across, and arranged in short rows; adoral zone occupying 25% of body length in vivo; paroral conspicuously short and located in front of endoral. Urosoma karinae sinense nov. sspec. is characterized by its elongate-elliptical body with no tail; 2–4 macronuclear nodules; cortical granules colourless, less than 1 μm across, and arranged in short rows; adoral zone occupying 30% of body length in vivo; paroral shorter than, and located ahead of endoral. Phylogenetic analyses based on SSU rRNA gene sequence data suggest a close relationship between U. salmastra, U. karinae sinense nov. sspec. and Oxytricha granulifera within the Oxytrichinae assemblage.  相似文献

The morphology and morphogenesis of a new saline soil hypotrichous ciliate, Urosoma quadrinucleatum n. sp., collected from northwestern China, were studied based on live observations and protargol stained specimens. The new species is characterized as follows: size in vivo 90–130 × 20–30 μm; body outline elongate-elliptical with both ends broadly rounded; four macronuclear nodules; cortical granules present; paroral in front of endoral; usually 16 frontal-ventral-transverse cirri. Urosoma quadrinucleatum n. sp. has almost the same morphogenetic characteristics as its congeners U. gigantea and U. salmastra, and differs from other three congeners whose morphogenesis is known in the formation of the frontal-ventral-transverse cirral anlagen as well as the development of marginal and dorsal kineties anlagen. The sequence differences among U. quadrinucleatum n. sp. and other Urosoma species further support the validity of the present organism as novel species. Further, U. quadrinucleatum n. sp. clusters with U. salmastra in the phylogenetic analyses based on small subunit ribosomal RNA gene (SSU rDNA) sequence data.  相似文献

The ultrastructure of cells of Acidiphilium rubrum, which is an acidophilic aerobic photosynthetic bacterium containing zinc-complexed bacteriochlorophyll a, was studied by electron microscopy with the rapid substitution technique. Thin-section electron microscopy indicated that any type of internal photosynthetic membranes was not present in this organism despite a relatively high content of the photopigment. The majority of cells had poly-β-hydroxybutyrate granules and electron-dense spherical bodies identified as being polyphosphate granules. When the organism was grown chemotrophically with 0.1% FeSO4, it produced another group of electron-dense granules that were associated with the inner part of the cytoplasmic membrane. An energy-dispersive X-ray analysis showed that these membrane-bound, electron-dense granules contained iron. Received: 24 November 1999 / Accepted: 5 January 2000  相似文献

Spermiogenesis and the ultrastructural organization of the spermatozoon of the digenean Neoapocreadium chabaudi are described by means of transmission electron microscopy.Spermiogenesis follows the usual pattern found in the digeneans. It begins with the formation of a zone of differentiation bordered by cortical microtubules, characterized by the presence of an intercentriolar body composed of seven electron-dense plates situated between two striated rootlets and two centrioles. These centrioles give rise to two free flagella. Later, both flagella undergo a rotation of 90° and fuse with the median cytoplasmic process. Spermiogenesis finishes when the ring of arched membranes constricts. The mature spermatozoon of N. chabaudi is characterized by the presence of 2 axonemes of different lengths presenting the 9 + “1” trepaxonematan pattern, 2 bundles of parallel cortical microtubules, 2 mitochondria, a nucleus, and granules of glycogen. Nevertheless, several characters such as the morphology of sperm extremities and the presence of spinelike bodies allow us to distinguish N. chabaudi from other digenetic trematodes. The present paper provides the first ultrastructural results of a digenean belonging to the family Apocreadiidae that may be useful for the understanding of digenean relationships and phylogenetic studies.  相似文献

The developmental oogenesis of gorgonian coral was investigated at the histological level. The objective of this study was to examine and improve the understanding of Junceella juncea oogenesis using ultrastructural methods, such as histological sectioning and transmission electron microscopy. At least three types of yolk materials were observed in this study: yolk body, lipid granules and cortical alveoli. Some of the complex yolk materials were encompassed by concentric or arched layers of smooth and rough endoplasmic reticulum and the Golgi complex in early stage oocytes. Different types of vesicles were found in both early and late stage oocytes and some granules could be seen inside the empty vesicles. This may be a possible method for elaborating complex yolk materials. Homogeneous yolks from different types of inclusions were abundant and the autosynthesis of yolk may be a major mechanism in J. juncea oocytes. This is the first report of the ultrastructural observation of oogenesis in gorgonian coral species using transmission electron microscopy. Our study obtained relatively detailed information at the ultrastructural level, and it provides an overview of the oocyte ultrastucture of the gorgonian coral J. juncea.  相似文献

By using scanning and transmission electron microscopy, the present study demonstrates a great number of trichocyst-like extrusomes distributed in the cortical cytoplasm of the protozoan Pseudourostyla cristata, a hypotrichous ciliate. Of these, the mature organelles are rod-shaped with a cap consisting of tubular structures, a tip located at the apex of the cap, a body consisting of strateform structures of uneven electron density and an elongated shaft located along the longitudinal central axis of the body. The electron microscopic observations suggest that the extrusive organelles in P. cristata might undergo a morphogenetic process including the following sequential events: the occurrence of the vesicles in the cytoplasm, the condensation of the fibrous substances within the vesicles, the appearance of the electron-dense shaft, and the formation of the cap. In contrast with a large quantity of extrusomes in trophozoit P. cristata, there are no such extrusive organelles in the encysted cells of the ciliate. The phenomena that P. cristata ciliates can readily enter physiological reorganization or encysting phases and discharge a great number of their extrusomes when prepared for SEM and TEM observation suggest that the extrusive process of the extrusomes in P. cristata might have an important influence on the life activity of the ciliate and could be one of the causes leading to the physiological reorganization and the encysting of the ciliate. These reactions of P. cristata might be a protective or defensive response to the environmental changes.  相似文献

This paper describes the ultrastructure of the male gamete of Glossobothrium sp. (Bothriocephalidea: Triaenophoridae). The mature spermatozoon of Glossobothrium sp. is filiform and possesses two axonemes, a single helicoidal crested body, a parallel nucleus, parallel cortical microtubules and granules of glycogen. In Glossobothrium sp. we describe for first time a 200-250 nm thick crest-like body in the Bothriocephalidean. The anterior part of the spermatozoon exhibits a ring of 27 electron-dense cortical microtubules encircling the first axoneme. This structure persists until the appearance of the second axoneme. When the ring of electron-dense cortical microtubules disappears, the spermatozoon exhibits two bundles of thin cortical microtubules. The posterior part of the spermatozoon contains the posterior extremity of the second axoneme, the posterior extremity of the nucleus and few cortical microtubules. Soon nucleus disappears and the axoneme is disorganized. Thus the posterior extremity of the spermatozoon of Glossobothrium sp. exhibits only singlets produced by the disorganization of the doublets of the second axoneme and few cortical microtubules. This type of posterior extremity of the mature spermatozoon has never been described previously in the Triaenophoridae.  相似文献

Two hypotrichous ciliates, Paraurostyla wuhanensis nov. spec. from Wuhan (China) and a new North American population of the Paraurostyla weissei complex, were studied based on live observations and protargol impregnation. Paraurostyla wuhanensis nov. spec. differs from the congeners by the combined features of having six or seven frontal cirri, 2–4 frontoventral cirri, 5–7 ventral rows, and yellow-greenish cortical granules. Ontogenesis proceeds as in the type species, except that fewer frontoventral cirri are formed in the new species. The morphology of the new population of the Paraurostyla weissei complex corresponds well with other American populations. In the phylogenetic trees based on the 18S rRNA gene, Paraurostyla sequences nest in a large clade together with Apoamphisiella and Notohymena. Monophyly of Paraurostyla is rejected by the results of the approximately unbiased test analyses. Morphological, morphogenetic, and phylogenetic analyses show a close relationship between the North American populations of the Paraurostyla weissei complex and Apoamphisiella, indicating that the taxonomic position of the former needs to be reassigned. A new combination, viz. Apoamphisiella polymicronucleata (Merriman, 1937) comb. nov., a reevaluation of the Paraurostyla weissei complex, and emended diagnoses of Paraurostyla and Apoamphisiella, are provided.  相似文献

The morphology, morphogenesis and molecular phylogeny of a hypotrichous ciliate, Lamtostyla granulifera sinensis subsp. nov., isolated from northern China, were investigated. This population appeared highly similar in morphology to L. granulifera Foissner, 1997. However, on detailed investigation some non-overlapping features were identified, i.e., the body shape and the arrangement of the cortical granules. These differences suggested the separation at subspecies level. Furthermore, the morphogenesis of the new subspecies is described, which is characterized by: (1) the posterior part of the parental adoral zone of membranelles is renewed; (2) the amphisiellid median cirral row is formed from two anlagen; and (3) the frontoventral-transverse cirral anlagen II to VI generate one transverse cirrus each. Phylogenetic analyses based on SSU rDNA sequence data show that Lamtostyla species are scattered in different clades. The monophyly of the genus Lamtostyla is also rejected by the AU test in this study.  相似文献

The synthesis of DNA in two hypotrichous ciliates, Styx sp. and an amicronucleated strain of Oxytricha sp., was studied by high voltage (1000 kV) electron microscopy. High voltage EM permits use of thick sections (0.25-0.40 μm), including serial sections; thick sections produce strong autoradiographic images with relatively short exposure times. The autoradiographs show that DNA synthesis occurs in a narrow part of the rear zone of a replication band in the macronucleus. Macronuclear DNA synthesis occupies a substantial part of the interdivision interval, and micronuclear DNA synthesis in Styx sp. takes place in early prophase at a time when macronuclear DNA synthesis is in its terminal phase.  相似文献

The present study focuses on the ultrastructural characteristics of both spermiogenesis and the spermatozoon in the order Trypanorhyncha. New ultrastructural data are presented for two species of the unexplored superfamily Tentacularioidea, Nybelinia queenslandensis, and Kotorella pronosoma. The present study also provides supplementary data on the superfamily Eutetrarhynchoidea, with the analysis of spermiogenesis and spermatozoon of two progrillotiids, Progrillotia dasyatidis and Pro. pastinacae, and new ultrastructural data concerning spermiogenesis in the eutetrarhynchids Dollfusiella spinulifera and Parachristianella trygonis. Spermiogenesis in trypanorhynchs follows the Bâ and Marchand's type I and the ultrastructural organisation of the mature spermatozoon corresponds to the Levron et al.’s type I. The most remarkable characters concerns the number of electron-dense plates constituting the intercentriolar body during spermiogenesis and in the variability of the arc-like row of thick cortical microtubules present in the anterior areas of the spermatozoon because of its variability according to the species.  相似文献

The neurosecretory cells of the supra- and suboesophageal ganglia of young, unmated, adult male midges, Chironomus riparius, have been examined by both light and electron microscopy. The 5 cell types recognized have been placed in three major categories on the basis of their ultrastructural characteristics:—α1 cells, of which there are 8 in each medial neurosecretory cell (MNC) group and 3 in each group of ventral neurosecretory cells (VNC), contain electron-dense granules, 150 to 200 nm in diameter; α2 cells containing irregular, electron-dense granules, 70 to 120 nm in diameter comprise the remaining 3 cells in each VNC group and the 2 or 3 cells in each outer neurosecretory cell (ONC) group; α3 cells, of which there are 1 or 2 on each side of the midline in the ventral cortex of the sub-oesophageal ganglion (SNC2), contain electron-lucent, spherical granules, 70 to 120 nm in diameter. The β cells contain spherical or ellipsoidal, electron-lucent granules, 80 to 100 nm in diameter, and make up the lateral neurosecretory cell (LNC) groups, each of three or four cells. The γ cells contain both spherical and flattened, electron-dense granules, 130 to 160 nm in diameter and 150 to 250 by 70 to 150 nm in size respectively, only 1 cell of this category being found in each half of the suboesophageal ganglion in the dorsal cortex (SNC1). Axons from the MNC and VNC form the nervi corporis cardiaci I (NCCI) and those of the LNC and ONC, the nervi corporis cardiaci II (NCCII). Those of the SNC1 appear to enter the wall of the stomodaeum but axons of the SNC2 could not be traced.  相似文献

ABSTRACT

Spermatological characteristics of the troglotrematid digenean Nephrotrema truncatum, a parasite of the shrew Crocidura russula, have been investigated by means of transmission electron microscopy. The ultrastructural study reveals that the mature spermatozoon of N. truncatum exhibits many ultrastructural characters previously described in most gorgoderoideans. These are two axonemes of the 9+‘1‘ trepaxonematan pattern, four attachment zones, a lateral expansion, an external ornamentation of the plasma membrane associated with spine-like bodies and cortical microtubules, and in the posterior part of the anterior spermatozoon region, two bundles of parallel cortical microtubules with the maximum number located in the anterior part of the spermatozoon, a nucleus, two mitochondria, and granules of glycogen. The obtained results are compared with those of other digeneans, particularly the Gorgoderoidea. The sperm cells gorgoderoideans are of type IV, characterised by a 9+‘1‘ pattern of axonemes, the presence of an external ornamentation associated with cortical microtubules and located in the posterior area of the anterior extremity, the presence of two bundles of cortical microtubules, the maximum number of cortical microtubules located in the anterior region of the spermatozoon, and the presence of generally two mitochondria. However, dicrocoeliids and troglotrematids have spermatozoa with ornamentation of the plasma membrane and lateral expansions.  相似文献

The ultrastructural organization of the spermatozoon of the digenean Hypocreadium caputvadum (Lepocreadioidea: Lepocreadiidae) is described. Live digeneans were collected from Balistes capriscus (Teleostei: Balistidae) from the Gulf of Gabès, Tunisia (Eastern Mediterranean Sea). The mature spermatozoon of H. caputvadum shows several ultrastructural characters such as two axonemes of different lengths exhibiting the classical 9 + “1” trepaxonematan pattern, a nucleus, two mitochondria, granules of glycogen, external ornamentation of the plasma membrane and two bundles of parallel cortical microtubules. Moreover, in the anterior extremity, the second axoneme is partly surrounded by a discontinuous and submembranous layer of electron-dense material. Our study provides new data on the spermatozoon of H. caputvadum in order to improve the understanding of phylogenetic relationships in the Digenea, particularly in the superfamily Lepocreadioidea. In this context, the electron-dense material surrounding one of the axonemes in the anterior spermatozoon extremity constitutes the unique distinguishing ultrastructural character of lepocreadioideans, and it is present in spermatozoa of lepocreadiids, aephnidiogenids and gyliauchenids.  相似文献

Transmission electron microscopy has been used to identify poly-3-hydroxybutyrate (PHB) granules in cyanobacteria for over 40 years. Spherical inclusions inside the cell that are electron-transparent and/or slightly electron-dense and that are found in transmission electron micrographs of cyanobacteria are generally assumed to be PHB granules. The aim of this study was to test this assumption in different strains of the cyanobacterium Synechocystis sp. PCC 6803. Inclusions that resemble PHB granules were present in strains lacking a pair of genes essential for PHB synthesis and in wild-type cells under conditions that no PHB granules could be detected by fluorescence staining of PHB. Indeed, in these cells PHB could not be demonstrated chemically by GC/MS either. Based on the results gathered, it is concluded that not all the slightly electron-dense spherical inclusions are PHB granules in Synechocystis sp. PCC 6803. This result is potentially applicable to other cyanobacteria. Alternate assignments for these inclusions are discussed.  相似文献

Mouse, rat and Mongolian gerbil atrial and ventricular cardiocytes were examined by immunohistochemistry, and the right atrium including the auricle was examined by transmission electron microscopy. In addition the ANP granules of both right atrial and auricular cardiocytes were analyzed by ultrastructural morphometry. ANP immunoreactivity was detected in the atria of all three species, and the most intensely reacting cardiocytes were localized in the right auricular part of the atrium. These reactions were more prominent in the mouse and rat than in the Mongolian gerbil. ANP immunoreactivity was not detected in the ventricular myocardium of any of the three species, but was occasionally seen in the subendocardium of the ventricular septum. Ultrastructurally, the ANP granules in the auricular and atrial cardiocytes were observed to be variable in size and number, and these granules were located principally in the paranuclear region in association with the Golgi apparatus, and found throughout the sarcoplasmic layers in all three species. The ANP granules were classified into two types: A-granules containing a conspicuous electron-dense core possessing a membrane, and B-granules having profiles with a fibrillogranular, less electron-dense core than the A-granules and an indistinct membrane. The features of these granules were similar in all three species. When examined by ultrastructural morphometry, the number of each type granule and the total number of granules in the right auricular and atrial cardiocytes of the mouse and rat were significantly greater than in the Mongolian gerbil. The total number of granules in the right auricular cardiocytes was significantly greater than in the cardiocytes of the right atrium exclusive of the auricle, however, there was no significant difference between the number of A-granules and B-granules in the three species. The diameter of each type of granule in the right auricular and atrial cardiocytes of the mouse and rat was significantly greater than in the Mongolian gerbil, and the diameter of the A-granules was significantly greater than the diameter of the B-granules in all three species.  相似文献
