Non-alcoholic fatty liver disease (NAFLD) is characterized by hepatic fat accumulation and is presently the most common chronic liver disease. However, the mechanisms underlying the development of steatosis remain unclear. MicroRNAs (miRNAs) are small non-coding RNAs that modulate a variety of biological functions. We have investigated the role of miRNA in the development of steatosis. We found that miR-467b expression is significantly downregulated in liver tissues of high-fat diet fed mice and in steatosis-induced hepatocytes. The downregulation of miR-467b resulted in the upregulation of hepatic lipoprotein lipase (LPL), the direct target of miR-467b. Moreover, the interaction between miR-467b and LPL was associated with insulin resistance, a major cause of NAFLD. These results suggest that downregulation of miR-467b is involved in the development of hepatic steatosis by modulating the expression of its target, LPL.  相似文献

Immobilized Microcystis aeruginosa in a flow-through sorption column was evaluated for the potential to remove Pb2+, Cd2+ and Hg2+ from aqueous solutions. M. aeruginosa showed high affinity for the three heavy metals with removal efficiency of 90% for Cd2+ and Hg2+, and 80% for Pb2+ at saturation conditions. Competitive sorption experiments conducted in paired and ternary systems indicated that Pb2+ was sequestered preferentially over Cd2+ and Hg2+. The presence of Cd2+ interfered only slightly with the uptake of Hg2+, as Pb2+ and Hg2+ did with Cd2+. In contrast, Hg2+ sorption was affected by Pb2+ to a great extent. Desorption with 1 M HCl was completed within 25 min with high efficiency and effectiveness for the three metals. The results of this study indicate that M. aeruginosa is to be a potential biosorbent material except when Hg2+ and Pb2+ are in the same contaminated solution.  相似文献

Due to industrial development, environmental contamination with metals increases which leads to higher human exposure via air, water and food. In order to evaluate the level of the present exposition, the concentrations of metals can be measured in such biological materials as human blood. In this study, we assessed the concentrations of cadmium (Cd), mercury (Hg) and lead (Pb) in blood samples from male blood donors from southern Poland (Europe) born in 1994 (n = 30) and between 1947 and 1955 (n = 30). Higher levels of Pb were seen in the group of older men (4.48 vs 2.48 μg/L), whereas the Hg levels were lower (1.78 vs 4.28 μg/L). Cd concentrations did not differ between age groups (0.56 μg/L). The levels of Cd and Pb in older donors were significantly correlated (Spearman R 0.5135). We also observed a positive correlation between the number of red blood cells (RBC) and Hg concentrations in the older group (Spearman R 0.4271). Additionally, we noted numerous correlations among morphological parameters. Based on our results, we can state that metals influence the blood morphology and their concentrations in blood vary among age groups.  相似文献

Liver has evolved complex enzymatic mechanisms to detoxify a wide array of xenobiotic substances, ranging from dietary components to environmental toxins to pharmaceuticals. Activities of many steroid‐metabolizing enzymes in adult rat liver microsomes are sexually differentiated. Toxic effects of lead and cadmium on hepatic tissue have been well established in our earlier studies. We thus monitored the effects of gestational and lactational coexposure to lead and cadmium on hepatic phase I and phase II xenobiotic‐ and steroid‐metabolizing enzyme activities in both male and female F1 generation postnatal day (PND) 56 rats. Adult pregnant female rats were treated subcutaneously [0.05 mg/(kg body wt. day)] with sodium acetate (control group), lead acetate, and cadmium acetate separately and in combination throughout the gestational and lactational period. Hepatic phase I xenobiotic‐metabolizing enzymes (NADPH‐ and NADH‐cytochrome c reductase) activities significantly decreased significantly in all the metal‐treated groups in both PND 56 male and female rats as compared with the control group. Hepatic phase II enzymes (uridine diphosphate‐glucuronosyl transferase, γ‐glutamyl transpeptidase, glutathione‐S‐transferase, 17‐β‐hydroxysteroid oxidoreductase) were also highly susceptible to all the metal‐treated groups. The observed alterations in the oxidative stress and biochemical parameters in the liver of F1 generation male and female rats resulted from an independent effect of lead and/or cadmium and also from their interaction. Results suggest that early developmental exposure to lead and cadmium both alone and in combination can suppress the hepatic xenobiotic‐metabolizing enzyme activities in the liver of F1 generation male and female rats in a sex‐dependent manner. © 2009 Wiley Periodicals, Inc. Adult virgin 4-day cycling synchronized Charles foster females were treated subcutaneously (0.05 mg/kg body wt/day) with sodium acetate (control), lead acetate or cadmium acetate alone, or both during gestational period, with pretreatment of 5 days prior to mating. There were no alterations in reproductive performance in all metal-treated groups. Implantation enzymes, cathepsin-D and alkaline phosphatase, activity were altered, but no change in the reproductive performance was observed. The key steroidogenic enzymes of ovary and placenta (3beta-HSD and 17beta-HSD), along with gonadal steroids, were affected the most in cadmium and combined treated animals whereas lead-treated animals showed a minimum change compared to the control group. Maximum displacement of zinc bound to metallothionein was more in cadmium and combined treated rats when compared to other metal-treated groups. Biomolecules such as glycogen, protein, RNA, DNA, and protein content were affected in all metal-treated groups, whereas cadmium-treated animals showed greater effect. General parameters of toxicity such as alkaline phosphatase, serum glutamate pyruvate transaminase, and creatinine were altered but were within the normal range. Biochemical effects are correlated with metals accumulated in blood, reproductive tissue such as placenta and ovary.  相似文献

Results on the effect of sub-lethal concentrations of zinc chloride (ZnCl2), cadmium chloride (CdCl2), and mercuric chloride (HgCl2) on Euglena are presented. During the growth cycle respiratory oxygen uptake and photosynthetic oxygen evolution in the light are initially strongly inhibited by Zn, Cd and Hg. The effects of the three metals on photosynthesis, using oxygen evolution as a criterion was confirmed by carbon fixation techniques.Photosystem I (PSI) associated electron transport 2,6-dichlorophenol indophenol (DCPIP)red. methyl viologen (MV) O2, in contrast to total photosynthetic capacity, was only slightly inhibited by Zn, Cd and Hg, whereas the levels of activity of NADP-oxidoreductase in cells untreated or treated with heavy metals showed development like total photosynthesis. Metals strongly inhibited this enzyme which means that the supply of NADPH is lowered due to the action of Zn, Cd and Hg. Photosystem II (PSII) associated electron transport (H2O dibromothymoquinone/2,3-dimethyl-5,6-methylenedioxy-D-benzoquinone O2), however, was severely inhibited in a way similar to total photosynthesis. Evidence relating dietary patterns to obesity and related disorders such as non-alcoholic fatty liver disease (NAFLD) is limited in pediatric age. Aim of this study was to analyze the association between dietary patterns, obesity and development of severe steatosis and the metabolic syndrome in a series of children and adolescents referred for suspected NAFLD, and the interaction with the rs738409 I148M PNPLA3 polymorphism. Two hundred patients (112 females) had completed a food frequency and demographic questionnaire. Nearly 58 % were obese, and 32 % were overweight. Mild, moderate, and severe fatty liver was present in 60 (30 %), 87 (44 %), and 51 (26 %) participants, respectively. A great proportion of overweight/obese children and adolescents reported a correct dietary pattern. At multivariate ordinal regression analysis considering demographic, anthropometric, genetic, and behavioral determinants, the major determinant of steatosis severity was PNPLA3 I148M genotype (p < 0.0001), followed by older age (p = 0.017), higher waist circumference (p = 0.016), and less time spent practising physical exercise (p = 0.034). Furthermore, there was a significant interaction between PNPLA3 I148M and intake of sweetened beverages (p = 0.033) and of vegetables (p = 0.038). In conclusion, although dietary pattern was reportedly correct in at-risk overweight adolescents with NAFLD, we report a novel interaction between PNPLA3 I148M and dietary components with the severity of steatosis.  相似文献

In an attempt to know whether highly consumed food might contribute to metal exposure, we analyzed cadmium, lead, and mercury in 27 rice grain samples commonly consumed in Saudi Arabia by atomic absorption spectrometry after acid digestion. The mean concentrations and ranges of cadmium, lead, and mercury in tested rice samples were 20.261 (range <DL-178.026 μg/kg), 134.819 (range 23.1–1529.0 μg/kg), and 3.186 (range <DL-43.573 μg/kg), respectively. The results showed high concentrations of metals and in some cases exceeded the Provisional Tolerance Weekly Intake (PTWI) recommended by FAO/WHO. It was also noted that different rice grain samples had varying concentrations of these metals. Because the bulk of literature warns against the cumulative effects of prolonged heavy metal exposure, regular consumption of rice by local populations might pose potential health problems.  相似文献

Obesity is associated with chronic diseases such as fatty liver, type 2 diabetes, cardiovascular disease, and severe metabolic syndrome. Obesity causes metabolic impairment including excessive lipid accumulation and fibrosis in the hepatic tissue as well as the increase in oxidative stress. In order to investigate the effect of mulberry leaf (Morus alba L.) extract (MLE) on obesity-induced oxidative stress, lipogenesis, and fibrosis in liver, MLE has been gavaged for 12 weeks in high-fat diet (HFD)-induced obese mice. MLE treatment significantly ameliorated LXRα-mediated lipogenesis and hepatic fibrosis markers such as α-smooth muscle actin, while MLE up-regulated lipolysis-associated markers such as lipoprotein lipase in the HFD-fed mice. Moreover, MLE normalized the activities of antioxidant enzymes including heme oxygenase-1 and glutathione peroxidase in accordance with protein levels of 4-hydroxynonenal in the HFD-fed mice. MLE has beneficial effects on obesity-related fatty liver disease by regulation of hepatic lipid metabolism, fibrosis, and antioxidant defense system. MLE supplementation might be a potential therapeutic approach for obesity-related disease including non-alcoholic fatty liver disease.  相似文献

BackgroundLead exposure all over the world has gradually declined. As fetuses are more prone to lead exposure, even to low levels of lead exposure, it is important to monitor blood lead levels (BLLs) in pregnancy.MethodsWe obtained data on BLLs in the third trimester of pregnancy from medical records and measured cord BLLs obtained from 121 mother-child pairs in Shenyang, China from September 2019 to February 2020. We also estimated relationships between socio-demographic, lifestyle and dietary factors during pregnancy as well as cord BLLs to identify the source of lead exposure during pregnancy. BLLs was estimated by atomic absorption spectrometry through graphite furnace ionization techniques. The data which obtained by questionnaires during pregnancy included maternal sociodemographic, lifestyle, dietary factors. We have established three multivariate logistic regression models in which the dichotomous BLLs was used as the dependent variable (cord BLLs ≥20 μg/L vs <20 μg/L).ResultsThe median and geometric mean of cord BLLs were 22.90 μg/L, 21.88 μg/L and BLLs in the third trimester of pregnancy were 25.29 μg/L, 24.66 μg/L, respectively. BLLs showed significant correlations between cord and the third trimester of pregnancy (r = 0.277, P = 0.012). Pregnant women who had not been exposed to passive smoking had lower OR (95 %) [0.43(0.19–0.94)] for cord BLLs ≥20 μg/L than pregnant women who had. Intake of docosahexaenoic acid (DHA) during third trimester of pregnancy presented an OR (95 %) [0.23(0.08–0.61)] for cord BLLs ≥20 μg/L. Consuming more whole grains (>3 times/week) and beverage (≥1 times/week) showed an OR (95%CI) for cord BLLs ≥20 μg/L of 0.09(0.02–0.53) and 0.19(0.06–0.69), respectively.ConclusionThis study showed the cord BLLs of Chinese are still higher than most developed countries. Passive smoking is a risk factor for cord BLLs ≥20 μg/L and supplement of DHA, whole grains and beverage consumption during pregnancy may act as a beneficial factor against having cord BLLs ≥20 μg/L.  相似文献

BackgroundNonalcoholic fatty liver disease (NAFLD) is one of the most common causes of chronic liver disease and is associated with various co-morbidities. Transient elastography (FibroScan®) is a non-invasive method to detect NAFLD using the controlled attenuation parameter (CAP). We aimed to evaluate the association of the lipid panel and aminotransferases concentrations with the presence or absence of steatosis and fibrosis.MethodsOne hundred and five patients with NAFLD were included. Hepatic steatosis was quantified by CAP (dB/m) and liver stiffness by Kilopascals (kPa), these values were then analyzed against patient lipid panel and serum concentrations of the liver enzymes aspartate aminotransferase (AST) and alanine aminotransferase (ALT). A correlation and multiple regression were used. Mann-Whitney U test was used as non-parametric analysis.ResultsWe observed an association between hepatic steatosis and total cholesterol (B = 0.021, p = 0.038, Exp (B) = 1.021, I.C = 1.001-1.041) as well as serum triglycerides (B = 0.017, p = 0.006, Exp (B) = 1.018 and I.C = 1.005-1.030). Similarly, we found an association between significant hepatic fibrosis and lower concentrations of total cholesterol (B = -0.019, p = 0.005, Exp (B) = 0.982 I.C = 0.969-0.995) and elevated AST (B = 0.042, p = 3.25 × 10-4, Exp (B) = 1.043 I.C = 1.019-1.068) independent of age, gender and BMI.ConclusionsOur results suggest that, total cholesterol and triglyceride concentrations positively correlate with hepatic steatosis while significant hepatic fibrosis is associated with lower total cholesterol and higher AST concentrations.  相似文献

Summary A 24 factorial experiment was conducted under greenhouse conditions. Factors and levels in the experiment were soil pH at 4.5 and 6.4, and cadmium, lead, and nickel added to the soil to provide soil concentrations of 50, 250, and 50 ppm, respectively, above background levels. Two species were grown in succession in the same experimental pots. Ryegrass (Lolium hybridum Hausskn. cv. Tetrelite) was harvested three times and then one crop of oats (Avena sativa L. cv. Garry) was grown and harvested. Plant tissue concentrations of cadmium, lead, and nickel were monitored throughout the experiment.Addition of cadmium to the soil lowered the dry matter yields in all three ryegrass harvests and also reduced the yield of oat grain. The application of lead nitrate to the soil enhanced the yield of ryegrass obtained at the first harvest and also increased the yield of oat grain. Nickel, added to the soil at 50 ppm, was relatively innocuous to ryegrass and oats.The presence of added cadmium, lead, or nickel to the soil resulted in enhanced tissue concentrations of these metals in both ryegrass and oats. This effect was particularly enhanced by a soil pH of 4.5. A single exception to this observation was that lead was not detected, under any of the conditions of this experiment, in oat grain.The presence of lead in the soil enhanced cadmium concentrations in ryegrass tissues. The presence of cadmium in the soil decreased lead tissue concentrations in ryegrass and oat straw. Soil pH interacted with both nickel and lead in the second ryegrass harvest with subsequent changes in tissue cadmium concentrations. At a soil pH of 4.5, the presence of added lead or nickel to the soil significantly increased the ryegrass tissue cadmium concentration beyond that observed at a soil pH of 6.4 with or without enhanced nickel or lead concentrations.  相似文献

BackgroundEnvironmental lead (Pb) and cadmium (Cd) pollution has been considered a risk factor in the etiology of kidney stones. However, the association between Pb and Cd exposure and kidney stone incidence has yet to be determined.ObjectivesThis study aimed to determine a possible the association between kidney stones with Pb and Cd exposure (alone or combined) in a non-occupational population.MethodsPb and Cd contaminations in soil-plant system were determined by flame atomic absorption spectrophotometry. Health risk assessment of dietary Pb or Cd intake from rice and vegetables were calculated. Kidney stones were diagnosed with urinary tract ultrasonography. Urinary cadmium (UCd) and blood lead (BPb) levels were determined by graphite-furnace atomic absorption spectrometry. Multivariate logistic regression models were constructed.ResultsThe hazard indexes (HI) of Pb and Cd were 7.91 and 7.31. The odds ratio (OR) was 2.83 (95 %CI:1.38−5.77) in males with high BPb (BPb ≥ 100 μg/L), compared with those with low BPb (BPb<100 μg/L). Compared to those with low BPb and low UCd (BPb<100 μg/L and UCd<2 μg/g creatinine), the ORs were 2.58 (95 % CI:1.17−5.70) and 3.43 (95 % CI:1.21−9.16) in females and males with high BPb and high UCd (BPb ≥100 μg/L and UCd ≥2 μg/g creatinine), respectively. The OR was 3.16 (95 % CI:1.26−7.88) in males with high BPb and low UCd (BPb ≥ 100 μg/L and UCd <2 μg/g creatinine), compared to those with low BPb and low UCd.ConclusionsKidney stones incidence was increased by high Pb exposure in males, and by Pb and Cd co-exposure in males and females.  相似文献

We examined the transfer of cadmium (Cd), inorganic mercury [Hg(II)], methylmercury (MeHg), and zinc (Zn) in an intertidal rocky shore food chain, namely from marine phytoplankton to suspension-feeding rock oysters (Saccostrea cucullata) and finally to predatory whelks Thais clavigera. The uptake of metals from the dissolved phase was also concurrently quantified in the oysters and the whelks. Metal uptake by the oysters was not directly proportional, whereas metal uptake by the whelks was directly proportional to metal concentration in the water. The order of uptake was MeHg>Hg(II)>Zn>Cd, and was much higher in the oysters than in the whelks. The relative uptake of Zn and Cd was comparable between oysters and whelks, whereas MeHg and Hg(II) showed disproportionally higher uptake in oysters than in whelks as compared to Zn and Cd. The assimilation efficiencies (AEs) were in the order of MeHg>Zn>Cd=Hg(II) in oysters, whereas the AEs were highest for MeHg and comparable for Zn, Cd, and Hg(II) in the whelks. Pre-exposure of the oysters to different dissolved concentrations of Cd significantly elevated the AEs of Cd and Hg(II) but not of Zn, in association with the induction of metallothioneins in the oysters. The whelks significantly assimilated Cd and Zn from various prey (barnacles, oysters, mussels, and snails) with contrasting strageties of metal sequestration and storage. There was no significant relationship between the metal AE and the metal partitioning in the soluble fraction (including metallothionein-like proteins, heat stable protein, and organelles). The insoluble fraction of metals was also available for metal assimilation. Our calculations show that the dietary uptake of metals can be dominant in the overall bioaccumulation in the oysters and whelks, and the trophic transfer factor was >1 for all metals. Thus, the four metals have a high potential of being biomagnified in the intertidal rocky shore food chain. MeHg possessed the highest and Hg(II) and Cd the lowest potential of trophic transfer among the four metals considered.  相似文献

BackgroundLead (Pb) and Cadmium (Cd) are important environmental contaminants. There is no biological monitoring of exposure to these heavy metals and their potential effect on dental caries in children in Tehran, Iran, a polluted megacity. Therefore, the present study investigated the potential association between Pb and Cd levels in primary teeth and saliva and dental caries.MethodsIn a cross-sectional design, 211 children aged 6–11 years referred to Tehran University of Medical Sciences, School of Dentistry and residing in Tehran were examined. Pb and Cd levels of exfoliated primary teeth and stimulated saliva were determined by atomic absorption spectrophotometry (AAS). Dental caries prevalence was evaluated according to WHO criteria. Socioeconomic status, oral hygiene behavior, snacking frequency and salivary pH data were acquired as confounding factors. Frequency and percentages were reported for categorical variables, mean and standard deviation (SD) for continuous variables, and geometric mean for skewed continuous variables. A simple linear regression and Pearson correlation tests were used for statistical analyses. P-values < 0.05 were considered as significant.ResultThe mean (95 % confidence interval) Pb and Cd levels in teeth were 213.26 ppb (164.29–274.84) and 23.75 ppb (20.86–27.05), respectively. The mean Pb and Cd levels in saliva were 11.83 ppb (10.71–13.06) and 3.18 ppb (2.69–3.75), respectively. Furthermore, Pb and Cd in primary teeth and saliva were not associated (p > 0.05) with socioeconomic status, oral hygiene behavior and snacking frequencyConclusionThis study showed no association between Pb and Cd concentrations in primary teeth and in saliva with dental caries prevalence.  相似文献

Summary Alfalfa (Medicago sativa L.), cv. Iroquois, was grown in the greenhouse in soils amended with additions of either lead, cadmium, or nickel. Metals, at rates varying from 0–250 ppm, were not uniformly mixed but were placed close to the soil surface so as to simulate surface deposition. In one series of experiments the sulphate salt of each metal and two soils were used. In a second series of experiments the nitrate salts and one soil were used. Neither salt of lead significantly depressed alfalfa yields. Both salts of either cadmium or nickel significantly depressed yields. Additions of all metals to the soil resulted in both increased metal uptake and concentrations in alfalfa tissue, particularly for cadmium and nickel. The highest tissue concentrations of cadmium and nickel were associated with plant stunting and necrosis. However, at rates of 125 ppm and less, substantial increases in cadmium and nickel concentrations were obtained frequently without serious yield reductions. Generally, metal concentrations were greatest in the first harvest following metal application. Concentration and uptake of lead and cadmium were greater when the metal was applied to the soil as nitrate than when applied as the sulphate salt.  相似文献

目的:探讨莪术醇(CC)对非酒精性脂肪性肝(NAFLD)大鼠模型肝功能和肝纤维化的影响及机制。方法:采用高脂饮食构建非酒精脂肪肝炎(NASH)伴肝纤维化的大鼠模型,将60只SD大鼠随机分为:空白对照组、模型组(NASH)、NASH+复方鳖甲软肝片(CBT)组(阳性对照组)、NASH+CC组(25、50、100 mg/kg),每组10只。测量大鼠肝脏占体重的百分比,测量大鼠高密度脂蛋白(HDL)、甘油三酯(TG)、谷丙转氨酶(ALT)、谷草转氨酶(AST)水平,HE染色观察肝纤维化情况,免疫组化检测鼠肝组织α-平滑肌肌动蛋白(α-SMA)表达及肝组织核因子κB p65(NF-κB p65)的阳性染色情况,蛋白印迹(Western blot)检测α-SMA、基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶抑制剂-1(TIMP-1)蛋白表达及Toll样受体-4(TLR4)、转化生长因子激活激酶-1(TAK1)、NF-κB p65、血管细胞粘附分子-1(VCAM-1)蛋白的表达情况,酶联免疫吸附法(ELISA)检测肝组织中白介素(IL-6、IL-10、IL-1β)、肿瘤坏死因子-α(TNF-α)的表达。结果:与空白对照组相比,模型组大鼠HDL、 IL-10含量、MMP-1蛋白表达量显著降低(P<0.05),TG、ALT、AST、肝组织P65阳性率,α-SMA、TIMP-1、TLR4、TAK1、NF-κB p65、VCAM-1表达、IL-6、TNF-α及IL-1β含量显著升高(P<0.05)。与模型组相比,CBT和CC处理后大鼠HDL、 IL-10含量、MMP-1蛋白表达量显著升高(P<0.05),TG、ALT、AST、肝组织P65阳性率,α-SMA、TIMP-1、TLR4、TAK1、NF-κB p65、VCAM-1表达、IL-6、TNF-α及IL-1β含量显著降低(P<0.05),其中模型+CC组以高浓度组改善最显著(P<0.05),但各剂量改善幅度均低于模型+CBT组(P<0.05)。结论:莪术醇通过调节TLR4、TAK1、NF-κB p65信号通路,减轻炎症反应,改善肝功能,从而缓解非酒精性脂肪肝肝肝纤维化,且在一定范围内呈浓度依赖性。  相似文献

BackgroundExposure of toxic metals from e-cigarette use is a cause for public health concern because youth, young adults, and non-smokers are the target population rapidly adopting e-cigarette use. The purpose of this research is to determine the association of the body burden of heavy metals with e-cigarette use using NHANES (U.S.) 2015–2016 data.MethodsBlood lead (N = 1899) and urinary cadmium, barium, and antimony (N = 1302) data were extracted from NHANES, 2015–2016; geometric means were calculated and bivariate and multivariable linear regression analyses were conducted. Participants were categorized as having neither e-cigarette nor cigarette use; smoking history (including dual use with e-cigarettes); and only e-cigarette (current or former).ResultsIn multivariable analyses adjusted for sex, race/ethnicity, age, and poverty levels, current or former e-cigarette use failed to reach a statistical significance in the association with metals. However, participants with a smoking history were more likely to have higher blood lead and urinary cadmium than participants who neither used e-cigarettes nor cigarettes.ConclusionBlood lead levels, and urinary cadmium, barium, and antimony levels were similar between participants who used e-cigarettes and participants who did not.  相似文献
