This work describes the isolation and characterization of twenty compounds from the tubers of Pinellia ternata (Thunb.) Breit, including 17 lignanoids (117) and three alkaloids (1820). All structures were established based on analysis of their spectroscopic data. It is the first report of compounds 3–11 and 13–18 from the genus Pinellia, and the first report of compounds 3–4, 6–9, 11 and 13–18 from the family Araceae. In addition, the biosynthetic pathways of the 17 lignanoids are summarized. A liquid chromatography coupled with electrospray ionization quadrupole time-of-flight mass spectrometry (LC-ESI-QTOFMS/MS) method in both positive and negative ion mode was also used to confirm the identity of these seventeen lignanoids from the tubers of P. ternata. The results from the present work provide further information about the diversity of compounds in the genus Pinellia.  相似文献

目的:鉴别半夏及其混伪品,确立半夏各变异类型间的关系。方法:采用紫外光谱聚类分析。结果:紫外光谱聚类分析可从某溶剂提取成分的数量特征初步判别半夏与其混伪品,反映各类型之间的亲疏远近关系;结论:双生珠芽半夏、多头半夏和4-5裂叶半夏与半夏居群其它变异类型的关系较为复杂,存在居群内遗传分化,但这种遗传分化是否足以导致半夏种系分化值得进一步研究。4-5裂叶半夏是否为普通半夏与掌叶半夏的中间类型有待于研究。  相似文献

In traditional Chinese medicine, multiple herbs are usually used in combination to generate the joint actions of a multiherb remedy. The recent development of LC-hyphenated techniques enables efficient and rapid profiling of the chemical constituent in extracts from multiherb remedies. Xiaochaihu-tang is a seven-herb remedy that has attracted a great deal of attention for reported ability to treat liver dysfunction. Dried tubers of Pinellia ternata (banxia in Chinese) is one of the ingredients, but its chemical contribution to Xiaochaihu-tang remains poorly understood. In the study presented here, LC-UV-MS, LC-MS-MS, and LC-NMR were used in a complementary manner to determine the nature and content of eight water-soluble constituents of banxia and their presence in various tea granules from Xiaochaihu-tang. Among the eight chemicals identified in banxia, cytidine, adenosine, tryptophan, uridine, and adenine are reported for the first time, while tyrosine, guanosine, and phenylalanine were previously described. These chemicals are also present in all of the samples of Xiaochaihu-tang granules, and the amounts of the chemicals ingested due to a daily dose of the multiherb remedies range from 0.008 to 6.3mg.  相似文献

研究Ca2+对半夏叶柄珠芽形成位置的影响.在MS+6-BA 1mg/L+IAA 0.5mg/L+3%蔗糖培养基中添加不同浓度Ca2+,观察正置培养的叶柄形芽形成情况并测定内源生长素(IAA含量).结果表明:对照Ca2+为3mmol/L培养基中,珠芽10.56%在其上端生成,92.22%在下端生成;低浓度和高浓度Ca2+对半夏叶柄珠芽形成位置有影响,Ca2+为1.5 mmoL/L和8 mmol/L时影响最显著,正置叶柄的珠芽大多数在叶柄的上端形成,形成率最高达87.22%.内源IAA测定表明:在Ca2+为3 mmol/L培养基中,叶柄上端的I从浓度高于下端,珠芽在下端生成.在Ca2+为1.5mmol/L和Ca2+为8mmol/L时,叶柄上端的生长素浓度低于叶柄下端,珠芽多在叶柄上端生成.因此,叶柄上下两端的生长素浓度差异影响了半夏珠芽的形成位置.  相似文献

An assessment of the genetic diversity and variation of Pinellia ternata collected from 43 populations in China was undertaken using SRAP + TRAP markers. A total of 13 SRAP primers in addition to 3 TRAP primer combinations yielded 292 bands in total of which 286 were polymorphic (98.0%), with an average of 16 for each. The PIC value ranged from 0.88 to 0.95, with a mean polymorphic information content (PIC) of 0.92 over all the primers. A cluster analysis was conducted based on the unweighted pair group method with arithmetic average (UPGMA), and a principle coordinate analysis (PCA) was performed relying on Nei's genetic distance, both of which showed similar outcomes in that the results of clustering did not correlate to the geography locations but showed an association with morphology. An analysis of molecular variance (AMOVA) was performed to detect the source of variation; the results indicated that the main variation existed within populations (67.2%), whereas there was still 32.8% differentiation existing among the populations.  相似文献

Macroautophagy/autophagy has profound implications for aging. However, the true features of autophagy in the progression of aging remain to be clarified. In the present study, we explored the status of autophagic flux during the development of cell senescence induced by oxidative stress. In this system, although autophagic structures increased, the degradation of SQSTM1/p62 protein, the yellow puncta of mRFP-GFP-LC3 fluorescence and the activity of lysosomal proteolytic enzymes all decreased in senescent cells, indicating impaired autophagic flux with lysosomal dysfunction. The influence of autophagy activity on senescence development was confirmed by both positive and negative autophagy modulators; and MTOR-dependent autophagy activators, rapamycin and PP242, efficiently suppressed cellular senescence through a mechanism relevant to restoring autophagic flux. By time-phased treatment of cells with the antioxidant N-acetylcysteine (NAC), the mitochondria uncoupler carbonyl cyanide m-chlorophenyl hydrazone (CCCP) and ambroxol, a reagent with the effect of enhancing lysosomal enzyme maturation, we found that mitochondrial dysfunction plays an initiating role, while lysosomal dysfunction is more directly responsible for autophagy impairment and senescence. Interestingly, the effect of rapamycin on autophagy flux is linked to its role in functional revitalization of both mitochondrial and lysosomal functions. Together, this study demonstrates that autophagy impairment is crucial for oxidative stress-induced cell senescence, thus restoring autophagy activity could be a promising way to retard senescence.  相似文献

Oxidative stress has been implicated in the induction of programmed cell death in a wide variety of organisms. Acquiring antioxidant capacity is thought to enhance the viability of cells challenged by a subsequent oxidative stress. Counter-intuitively, we show that in two phytoplankton species, Chlamydomonas reinhardtii and Peridinium gatunense , representing the green and red plastid lineages, oxidative stress induced cell death in cultures that already possessed high antioxidant activity but not in cells that exhibited low activity. Cell death of low antioxidant possessing cultures was markedly enhanced by the addition of dehydroascorbate, a product of ascorbate peroxidase (APX), but not of ascorbate or reduced glutathione, and was preceded by increased metacaspase expression and activity. These data suggested that the level of APX and its products, strongly upregulated by oxidative stress, serves as a possible surveillance signal, reporting that the cells already experienced an earlier oxidative stress. Our data presents a novel role of APX in antioxidant activity and response to oxidative stress in photosynthetic microorganisms. Elimination of cysts production by phytoplankton cells that were already damaged by oxidative stress (indicated by the rise in oxidized proteins) as the inoculum for the following year's population may be the evolutionary trigger for this phenomenon.  相似文献

Two marihuana constituents, cannabidiol (1) and cannabidiolic acid (4) were each converted mainly to two metabolites using tissue segments of Pinellia ternata tuber. The structures of the metabolites formed from 1 were determined to be 1-O-D-glucopyranoside and 1-O-D-diglucopyranoside by 1H nmr, 13C nmr and fabms. Those from 4 were determined as 4-O-D-glucopyranoside and 10-hydroxyl 4-O-D-glucopyranoside. In time course experiments, 1 was absorbed rapidly by the tissues and glucosylated. 半夏居群遗传分化的同工酶和可溶性蛋白电泳的聚类分析   总被引:2,自引:0,他引:2  
用聚丙烯酰胺凝胶电泳(PAGE)技术分析了安徽产7个半夏(PinelliaternataBreit)居群的4种同工酶和可溶性蛋白谱带。根据其中的PER、EST和可溶性蛋白谱带的分布特征,用Forguson和Aquado&Avise的公式分别计算其谱带的相似性系数和遗传距离,以类平均法(UPGMA)进行了聚类分析。结果表明:半夏居群间表现出一定的遗传分化;鹞落坪半夏与半夏的黄山、九华山、大别山万佛寺居群间的遗传距离较小(D≤0.214),结合形态变异,笔者认为鹞落坪半夏只能是半夏居群中形态上较为特殊的地方宗,而不是一新种;狭叶半夏与其它6个居群的遗传距离较大(D=0.416),是半夏居群中种系分化较为强烈的一个居群。文中还讨论了半夏的遗传分化与形态变异、生境等的相关性。  相似文献

Somatic cells undergo a permanent cell cycle arrest, called cellular senescence, after a limited number of cell divisions in vitro. Both the tumor suppressor protein p53 and the stress-response protein p66(shc) are suggested to regulate the molecular events associated with senescence. This study was undertaken to investigate the effect of different oxygen tensions and oxidative stress on cell longevity and to establish the role of p53 and p66(shc) in cells undergoing senescence. As a model of cellular senescence, primary fetal bovine fibroblasts were cultured in either 20% O(2) or 5% O(2) atmospheres until senescence was reached. Fibroblasts cultured under 20% O(2) tension underwent senescence after 30 population doublings (PD), whereas fibroblasts cultured under 5% O(2) tension did not exhibit signs of senescence. Oxidative stress, as measured by protein carbonyl content, was significantly elevated in senescent cells compared to their younger counterparts and to fibroblasts cultured under 5% O(2) at the same PD. p53 mRNA gradually decreased in 20% O(2) cultured fibroblasts until senescence was reached, whereas p53 protein levels were significantly increased as well as p53 phosphorylation on serine 20, suggesting that p53 might be stabilized by posttranslational modifications during senescence. Senescence was also associated with high levels of p66(shc) mRNA and protein levels, while the levels remained low and stable in dividing fibroblasts under 5% O(2) atmosphere. Taken together, our results show an effect of oxidative stress on the replicative life span of fetal bovine fibroblasts as well as an involvement of p53, serine 20-p53 phosphorylation and p66(shc) in senescence.  相似文献

Cataracts are the leading cause of blindness worldwide owing to the increasing proportion of elderly individuals in the population. The purpose of this study was to investigate whether metformin could alleviate the occurrence and development of age-related cataract (ARC) and the underlying mechanism. In the present study, we established a senescence model induced by oxidative stress, which was confirmed by measuring β-galactosidase activity, qRT-PCR and Western blotting. In addition, we showed that metformin alleviated the oxidative stress-induced senescence of HLE-B3 cells via the activation of AMPK. Next, we provided evidence that oxidative stress impaired autophagic flux and induced lysosomal dysfunction. Subsequently, we found that metformin restored autophagic flux that had been impaired by oxidative stress by activating AMPK. Additionally, we found that metformin suppressed HLE-B3 cell senescence by improving lysosomal function and inactivating mTOR. Furthermore, the inactivation of AMPK, impairment of autophagic flux and lysosomal dysfunction were observed in the human lens epithelium of ARC. In summary, our data suggest that the activation of AMPK may be a potential strategy for preventing ARC, and metformin may be an emerging candidate to alleviate the formation and development of ARC.  相似文献

比较了灰树花Grifola frondosa、巴氏蘑菇Agaricus blazei、 猴头菌Hericium erinaceus、刺芹侧耳Pleurotus eryngii、毛头鬼伞Coprinus comatus、蛹虫草Cordyceps militaris、灵芝Ganoderma lingzhi、香菇Lentinula edodes、桑黄Sanghuangporus sanghuang和桦褐孔菌Inonotus obliquus 10种食用菌醇提物对前列腺癌LNCaP细胞增殖的影响,其中灵芝醇提物对LNCaP细胞增殖及5ɑ-还原酶活性具有良好的抑制效果,对LNCaP细胞释放PSA的抑制效果也好于其他食用菌。进一步研究灵芝醇提物对LNCaP细胞的抑制机理,结果表明,灵芝醇提物通过诱导细胞凋亡、使Caspase-3、Bax、P53活性升高,PARP、Bcl-2活性下降来实现对LNCap细胞的抑制作用。利用自噬抑制剂氯喹和自噬激活剂雷帕霉素观察细胞自噬在灵芝醇提物抗肿瘤过程中的作用,结果表明,体外激活细胞自噬对LNCaP细胞起到保护,从而减弱了灵芝醇提物的抗肿瘤作用。  相似文献

对半夏进行不同程度遮荫处理,研究了其生长过程中叶组织的光合色素含量、叶绿素a/b值、保护酶体系(SOD、POD、CAT)活性和丙二醛(MDA)含量的变化及其生理特性.结果表明:不同处理的半夏在生长过程中,光合色素的含量呈先升后降的趋势;与全光照相比,遮荫处理的叶绿素含量较高,叶绿素a/b值较低,类胡萝卜素含量在生长发育中早期较低而后期较高;SOD、POD、CAT活性呈由低到高再降低的趋势,MDA含量持续升高;遮荫处理的SOD、POD活性和MDA含量均低于全光照处理,CAT活性则表现为先高于后低于全光照处理.适度的遮荫可以提高半夏叶绿素的含量,降低膜系统受到的伤害,改善半夏的倒苗状况,为半夏叶片的生长提供较好的环境.  相似文献

Ginkgo biloba extract (EGb761) has been shown to be neuroprotective; however, the mechanism by which EGb761 mediates neuroprotection remains unclear. We hypothesized that the neuroprotective effect of EGb761 is mediated by inhibition of cytosolic phospholipase A(2) (cPLA(2)), an enzyme that is known to play a key role in mediating secondary pathogenesis after acute spinal cord injury (SCI). To determine whether EGb761 neuroprotection involves the cPLA(2) pathway, we first investigated the effect of glutamate and hydrogen peroxide on cPLA(2) activation. Results showed that both insults induced an increase in the expression of phosphorylated cPLA(2) (p-cPLA(2)), a marker of cPLA(2) activation, and neuronal death in vitro. Such effects were significantly reversed by EGb761 administration. Additionally, EGb761 significantly decreased prostaglandin E(2) (PGE(2)) release, a downstream metabolite of cPLA(2). Moreover, inhibition of cPLA(2) activity with arachidonyl trifluromethyl ketone improved neuroprotection against glutamate and hydrogen peroxide-induced neuronal death, and reversed Bcl-2/Bax ratio; notably, EGb761 produced greater effects than arachidonyl trifluromethyl ketone. Finally, we showed that the extracellular signal-regulated kinase 1/2 signaling pathway is involved in EGb761's modulation of cPLA(2) phosphorylation. These results collectively suggest that the protective effect of EGb761 is mediated, at least in part, through inhibition of cPLA(2) activation, and that the extracellular signal-regulated kinase 1/2 signaling pathway may play an important role in mediating the EGb761's effect.  相似文献

Analyses of randomly amplified polymorphic DNA (RAPD) and PCR-restriction fragment length polymorphism (PCR-RFLP) were performed in an effort to distinguish between two different origins of Pinellia ternata. To determine whether the origin of Pinellia ternata is in China or Korea. RAPD analysis was carried out using ten 20-mer random primers. Although the coefficients of similarity between the DNA of Chinese and Korean accessions of Pinellia ternata were high, distinguishable band patterns were observed in the reaction performed using primer numbers 3, 6 and 10. Primer 3 produced one (410 bp) and primer 6 produced four (410 bp, 350 bp, 300 bp, 250 bp) Chinese Pinellia-specific fragments. Primer 10 produced one (900 bp) Korean Pinellia rhizome-specific fragment. In addition, using PCR-RFLP analysis, different fingerprints were obtained from Korean and Chinese Pinellia ternata respectively. These results suggest that the analyses with RAPD and PCR-RFLP can be used to authenticate the relevant Chinese and Korean herbal medicines.  相似文献

  • In recent years, natural Pinellia ternata populations of have gradually been exhausted, while the cultivated yield has been limited due to lack of research and uncertain climate condition. Therefore, it is necessary to explore methods of improving yield and quality in P. ternata using brassinolide (BR) treatments and choice of a suitable seed bulb size.
  • This article reports the effects of BR and two seed bulb sizes (diameter: 0.5–1.0 cm and 1.0–1.5 cm) on active and nutrient components and antioxidant activity in P. ternata. The experiment included six levels of BR (0, 0.05, 0.10, 0.50, 1.00 and 2.00 mg l?1).
  • The tuber yield of the two seed bulb sizes and bulbil yield of small seed bulbs increased 5.67%, 22.66% and 69.23% by day 105 after 0.50 mg l?1 BR treatment, compared with the control. On day 105, only 0.05 mg l?1 BR increased scores in principal components analysis (PCA) in tubers of small seed bulbs by 167.29%, and 0.05 and 0.50 mg l?1 BR increased PCA score in bulbils of large seed bulbs by 145.66% and 252.97%, respectively, compared with the control. Significant BR × seed bulb size interactions were found on yield and quality of P. ternata.
  • The results indicate that BR effects on yield and quality of tubers and bulbils of P. ternata are not only related to BR concentration but also to seed bulb size.

In addition to their key role as structural lens proteins, α-crystallins also appear to confer protection against many eye diseases, including cataract, retinitis pigmentosa, and macular degeneration. Exogenous recombinant α-crystallin proteins were examined for their ability to prevent cell death induced by heat or oxidative stress in a human lens epithelial cell line (HLE-B3). Wild type αA- or αB-crystallin (WT-αA and WT-αB) and αA- or αB-crystallins, modified by the addition of a cell penetration peptide (CPP) designed to enhance the uptake of proteins into cells (gC-αB, TAT-αB, gC-αA), were produced by recombinant methods. In vitro chaperone-like assays were used to assay the ability of α-crystallins to protect client proteins from chemical or heat induced aggregation. In vivo viability assays were performed in HLE-B3 to determine whether pre-treatment with α-crystallins reduced death after exposure to oxidative or heat stress. Most of the five recombinant α-crystallin proteins tested conferred some in vitro protection from protein aggregation, with the greatest effect seen with WT-αB and gC-αB. All α-crystallins displayed significant protection to oxidative stress induced cell death, while only the αB-crystallins reduced cell death induced by thermal stress. Our findings indicate that the addition of the gC tag enhanced the protective effect of αB-crystallin against oxidative but not thermally-induced cell death. In conclusion, modifications that increase the uptake of α-crystallin proteins into cells, without destroying their chaperone-like activity and anti-apoptotic functions, create the potential to use these proteins therapeutically.  相似文献

研究叶面喷施外源MeJA对高温胁迫下半夏的超氧化物歧化酶(SOD)、过氧化物酶(POD)、抗坏血酸过氧化物酶(APX)活性的影响;对脯氨酸(Pro)、丙二醛(MDA)、可溶性糖含量及有效成分含量的影响;对小分子热激蛋白(sHSP)等胁迫相关基因表达的影响。将生长状况一致的半夏植株在40℃高温下进行胁迫处理,实验组喷施50μmol·L^-1的外源MeJA溶液,空白组喷施等量的水,分别在处理2、6、12、24、48 h时进行取样,对样品进行指标测定。结果发现,在40℃高温下,喷施一定浓度的外源MeJA可以提高半夏叶片的SOD、POD、APX活性,降低MDA质量分数,增加脯氨酸及可溶性糖含量;对半夏有机酸含量无明显影响。2个细胞质型小分子热激蛋白和GRRBP蛋白,在MeJA处理后表达量显著增加。由此得出结论:喷施外源MeJA可以增强半夏中部分抗氧化酶的活性,保护半夏的细胞膜,增强细胞的渗透调节能力,且外源MeJA可能增加半夏高温胁迫响应基因的表达。  相似文献
