Phomopsis and related taxa comprise important endophytic and plant pathogenic species, and are known for the production of a diverse array of secondary metabolites. Species concepts within this group based on morphological characters and assumed host specificity do not reflect phylogenetic affinities. Additional phenotypic characters, such as profiles of secondary metabolites, are needed for practical species recognition. We investigated 36 strains of Phomopsis spp. and Cytospora-like fungi, obtained as endophytes of different host plants in Brazil, using metabolite profiling based on HPLC-UV/liquid chromatography -mass spectrometry (LC-MS) combined with cluster analysis of the results. Strains were also subjected to phylogenetic analyses based on internal transcribed spacer (ITS) rDNA. Six chemotypes were identified. Chemotypes 1-5 contained Phomopsis strains, while Cytospora-like strains formed the chemotype 6. Strains of chemotype 1 typically produced alternariols, altenusin, altenuene, cytosporones, and dothiorelones. Alternariol and seven unknown compounds were consistently produced by strains of chemotype 2. Members of chemotypes 3-5 produced poor metabolite profiles containing few chemical markers. Cytospora-like endophytes (chemotype 6) produced a characteristic set of metabolites including cytosporones and dothiorelones. Bayesian and Maximum Parsimony (MP) trees classified strains of each chemotype into single phylogenetic lineages or closely related groups. Strains of chemotypes 1 and 2 formed a monophyletic group along with Diaporthe neotheicola. The remaining Phomopsis strains formed monophyletic (chemotype 4) or polyphyletic (chemotypes 3 and 5) lineages inside a large and well supported clade. Cytospora-like strains formed a monophyletic lineage located at an intermediary position between Diaporthe/Phomopsis and Valsa/Cytospora clades. The combined results show that the production of secondary metabolites by Phomopsis and related Diaporthales may be species-specific, giving support to the use of metabolite profiling and chemical classification for phenotypic recognition and delimitation of species.  相似文献

The ability of cyanobacteria to produce toxins and other secondary metabolites is patchily distributed in natural populations, enabling the use of cellular oligopeptide compositions as markers to classify strains into ecologically-relevant chemotypical subpopulations. The composition and spatiotemporal distribution of Microcystis chemotypes within and among waterbodies was studied at different time scales by analyzing (i) Microcystis strains isolated between 1998 and 2007 from different Spanish reservoirs and (ii) individual Microcystis aeruginosa colonies collected from pelagic and littoral habitats in Valmayor reservoir (Spain) during a bloom. No agreement between chemotypes and both morphotypes and genotypes (based on cpcBA-IGS, 16S–23S rRNA ITS and mcyB genes) was found, suggesting that oligopeptide profiles in individual strains evolve independently across morphospecies and phylogenetic genotypes, and that the diversity of microcystin variants produced cannot be explained by mcyB gene variations alone. The presence of identical chemotypes in spatially-distant reservoirs with dissimilar trophic state, lithology or depth indicate that waterbody characteristics and geographical boundaries weakly affect chemotype composition and distribution. At smaller spatiotemporal scales (i.e. during bloom), M. aeruginosa populations showed high number of chemotypes, as well as marked differences in chemotype composition and relative abundance among the littoral and pelagic habitats. This indicates that the factors influencing chemotype composition, relative abundance and dynamics operate at short spatial and temporal scales, and supports emerging hypotheses about interactions with antagonistic microorganisms as possible drivers for widespread chemical polymorphisms in cyanobacteria.  相似文献

Bactericidal factors in antisera against various chemotype strains of Salmonella were assayed by means of the spot method. Certain smooth and rough strains were resistant to the killing effect of immune mouse sera when the activity was assayed in an isotonic salt medium and guinea pig complement was used. However, the sensitivity was found to be increased in various degrees by assaying it in a medium containing hypotonic concentrations of NaCl or tris(hydroxymethyl)-aminomethane-HCl (Tris-HCl). Keeping the resistant bacteria in hypotonic solutions before or after treatment with complement increased the titer of antiserum, indicating that the hypotonic solution increases the sensitivity of bacterial cells to the antiserum and/or complement. The optimal salt concentration for the assay of serum-sensitive and -resistant smooth strains was 0.02 M NaCl. With Ra through Rd chemotype strains, 0.1 m NaCl before complement treatment and 0.05 m NaCl after the treatment was the best. Isotonic medium was necessary for the titration of the Re chemotype strain. Specificity of the killing effect which was assayed by the hypotonic spot method was shown by adsorption studies on the immune sera. Use of C4-deficient guinea pig complement resulted in low titers against certain strains of bacteria and high titers were restored by the addition of the C4 component of complement. These results indicate that serologically specific bactericidal factors including antibody can be assayed by the spot method using hypotonic NaCl or Tris-HCl.  相似文献

The Plankthotrix Anagn. et Komárek population in the mesotrophic Lake Steinsfjorden has been intensively studied over several decades. This Planktothrix population produces a number of different classes of oligopetides. However, over the study period, only four main oligopeptide profiles (chemotypes) have been associated with the strains isolated from the lake. The chemotypes show distinct interactions with the environment, demonstrated by shifts in abundance along time series and vertical profiles. Here, we present genetic analysis of nonribosomal peptide synthetase (NRPS) gene regions in strains representing the four Planktothrix chemotypes in Lake Steinsfjorden. On the basis of phylogenetic analyses, we show that the NRPS genes for microcystin (mcy) and cyanopeptolin (oci) display the same clustering as do the chemotypes. Nucleotide diversity in mcy and oci was significantly higher between strains of different chemotypes than between strains of the same chemotype. Ka/Ks (nonsynonymous vs. synonymous mutations) values indicated positive selection in several polymorphic regions of the mcy and oci genes. Notably, incongruence between the phylogenetic trees for different gene segments and split decomposition analyses for segments of oci suggested horizontal gene transfer (HGT) events between strains showing different oligopeptide profiles. The oci HGT region encodes a module responsible for incorporating a variable amino acid in cyanopeptolin and is one of the regions suggested to be under positive selection. Taken together, our data suggest that there are four genetically distinct sympatric subpopulations—displayed as distinct chemotypes—in Lake Steinsfjorden. The diversification process of the chemotypes, and consequently the subpopulations, is driven by HGT and reinforced by positive selection of the corresponding NRPS gene regions.  相似文献

Eighty‐seven Planktothrix (Anagnostidis and Komàrek 1988) strains isolated from 13 lakes in Scandinavia and Finland between 1964 and 2007 were screened for oligopeptides. Forty‐six individual compounds were detected in total, belonging to the structural classes anabaenopeptins (six variants), aeruginosins (six variants), cyanopeptolins (21 variants), microcystins (five variants), microginins (two variants), and microviridins (two variants). Oscillatorin was also found. Three additional compounds could not be assigned to known oligopeptide classes. Thirty oligopeptides have not been described in previous studies. Of these new compounds, five were aeruginosins and 20 cyanopeptolins. The number of oligopeptides per strain ranged from one to 13. No oligopeptide‐free strains were found, suggesting that oligopeptide production is vital for Planktothrix. On the basis of their oligopeptide patterns, the Planktothrix strains of the present study were assigned to 17 chemotypes. Three major chemotypes occurred in up to six lakes. One chemotype occurred in lakes around the city of Oslo (Norway), on the Finnish island Fasta Åland, which is situated in the Baltic Sea, and on the Finnish mainland. This wide distribution suggests that chemotypes can be subjects of recurrent dispersal and/or strong directional selection. Lake size, maximum depth, and nutrient availability appeared to be of minor importance for the ability of some chemotypes to colonize a water body successfully as long as the general requirements of Planktothrix were met. Four chemotypes were reisolated from the Oslo lake district over a period of 33–40 years, suggesting that they have been members of local Planktothrix populations for decades.  相似文献

Experimental studies of the evolutionary biology of lichen fungi have been hampered by massive difficulties of in vitro culture and artificial crosses are still not possible. Gene flow in these organisms is demonstrated here for the first time by the analysis of secondary products in the progeny of individuals from natural populations of mixed chemotypes of the Cladonia chlorophaea complex. All of the chemotypes in this study have been interpreted as distinct sibling species. In the Appalachian Mountains of North Carolina, however, the grayi and merochlorophaea chemotypes are found to belong to a single interbreeding populations that is reproductively isolated from the cryptochlorophaea chemotype. In the Coastal Plain, the cryptochlorophaea chemotype hybridizes with the local endemic perlomera chemotype. This study has major impact for species concept in lichens because consideration of neither morphologial tendencies nor biogenetic relationships of the secondary products could have predicted its result.  相似文献

Populations of the cyanobacterium Planktothrix comprise multiple coexisting oligopeptide chemotypes that can behave differently in nature. We tested whether this population subdivision can, in principle, be driven by parasitic chytrid fungi, which are almost neglected agents of Planktothrix mortality. Two chytrid strains, Chy-Lys2009 and Chy-Kol2008, were isolated from Planktothrix-dominated lakes in Norway. The two strains shared 98.2% and 86.2% of their 28S and internal transcribe spacer rRNA gene sequences, respectively. A phylogenetic analysis placed them in the order Rhizophydiales family Angulomycetaceae. Chy-Lys2009 and Chy-Kol2008 could completely lyse Planktothrix cultures within days, while they failed to infect other filamentous cyanobacteria. The effect on Planktothrix was chemotype dependent, and both chytrid strains showed distinct chemotype preferences. These findings identify chytrid fungi infecting Planktothrix as highly potent and specialized parasites which may exert strong selective pressure on their hosts. According to established hypotheses on host-parasite coevolution, parasitism with the above properties may result in subdivision of Planktothrix populations into coexisting chemotypes and periodic shifts in the relative Planktothrix chemotype composition. These predictions are in agreement with field observations. Moreover, a genetic analysis verified the co-occurrence of Chy-Lys2009 and Chy-Kol2008 or related chytrid strains along with distinct Planktothrix chemotypes in at least one water body. Our findings are consistent with a scenario where chytrid parasitism is one driving force of Planktothrix population subdivision, which in turn leads to polymorphism in parasitic chytrid fungi. Future studies should test the validity of this scenario under field conditions.  相似文献

The bactericidal activity of sera not containing antibodies (sera from precolostral piglets and calves) was tested with strains of gram-negative bacteria with different surfaces. The accuracy of the method of bactericidal test was evaluated statistically the bactericidal unit of complement was defined for comparing the activity of sera of different animals and different species. Various methods used for estimating the character of bacterial surface were compared. It was found that the bactericidal activity of piglet sera is directly dependent on the content of complement in the sera tested and the character of the bacterial surface (in the R-form). In selected strains there is a correlation in all criteria characterizing the surfaces of bacteria, and their susceptibility to bactericidal activity of sera; in a group of 37 strains selected at random, correlation with only one of the surface characteristics (stability in solution after heating to 100°C for 1 hour) was found. In calf sera a component was found which increases the effect of complement to some strains (e.g.Shigella shigae). This component may by absorbed from the serum only in the presence of complement. The O polysaccharide (OPS) of the lipopolysaccharide (LPS) of Pseudomonas syringae pv. atrofaciens IMV 7836 and some other strains that are classified in serogroup O1 was shown to be a novel linear alpha-D-rhamnan with the tetrasaccharide O repeat -->3)-alpha-D-Rhap-(1-->3)-alpha-D-Rhap-(1-->2)-alpha-D-R hap-(1-->2)- alpha-D-Rhap-(1--> (chemotype 1A). The same alpha-D-rhamnan serves as the backbone in branched OPSs with lateral (alpha1-->3)-linked D-Rhap, (beta1-->4)-linked D-GlcpNAc, and (alpha1-->4)-linked D-Fucf residues (chemotypes 1B, 1C, and 1D, respectively). Strains of chemotype 1C demonstrated variations resulting in a decrease of the degree of substitution of the backbone 1A with the lateral D-GlcNAc residue (chemotype 1C-1A), which may be described as branched regular left arrow over right arrow branched irregular --> linear OPS structure alterations (1Cleft arrow over right arrow 1C-1A --> 1A). Based on serological data, chemotype 1D was suggested to undergo a 1D left arrow over right arrow 1D-1A alteration, whereas chemotype 1B showed no alteration. A number of OPS backbone-specific monoclonal antibodies (MAbs), Ps(1-2)a, Ps(1-2)a(1), Ps1a, Ps1a(1), and Ps1a(2), as well as MAbs Ps1b, Ps1c, Ps1c(1), Ps1d, Ps(1-2)d, and Ps(1-2)d(1) specific to epitopes related to the lateral sugar substituents of the OPSs, were produced against P. syringae serogroup O1 strains. By using MAbs, some specific epitopes were inferred, serogroup O1 strains were serotyped in more detail, and thus, the serological classification scheme of P. syringae was improved. Screening with MAbs of about 800 strains representing all 56 known P. syringae pathovars showed that the strains classified in serogroup O1 were found among 15 pathovars and the strains with the linear OPSs of chemotype 1A were found among 9 of the 15 pathovars. A possible role for the LPS of P. syringae and related pseudomonads as a phylogenetic marker is discussed.  相似文献

The chemical composition of the volatile fraction of Ocimum gratissimum concrete (romba) from Madagascar has been determined for the first time by GC/MS and GC-FID. A methyl cinnamate chemotype has been determined for this material, along with a set of compounds typical in essential oils and extracts from plants of the Ocimum genus. Variability was mostly observed on terpenes and terpenoids components. GC-O-MS was also used for a sensory evaluation of this material performed by a master perfumer. The chemical composition of this O. gratissimum extract was then compared with literature data to assess subtle differences between chemotypes of the same species and other species of the same genus within natural variability. A mapping illustrates the occurrence of the cinnamate chemotype in Eastern Africa, India and now Madagascar, while other origins generally present eugenol, thymol, camphor, or linalool chemotypes.  相似文献

Host plant nutritional and non-nutritional variability can have a significant effect on herbivore populations by influencing survival, larval performance, and fecundity. The effect of chemical and physical variation of the leaves of two chemotypes of the weed Melaleuca quinquenervia was determined on the biomass and fecundity of the biological control agent Oxyops vitiosa (Coleoptera: Curculionidae). M. quinquenervia chemotypes were distinguished by the principal terpenoids E-nerolidol and viridiflorol using gas chromatography and mass spectroscopy. Not only were the terpenoid profiles of the two chemotypes different but the viridiflorol leaves had greater toughness (1.2-fold) and reduced nitrogen (0.7-fold). When the larvae and adults were fed leaves of the E-nerolidol chemotype increased adult biomass (1.1-fold) and fecundity were found (2.6- to 4.5-fold) compared with those fed leaves of the viridiflorol chemotype. Regardless of the larval diet, when adults were fed the E-nerolidol chemotype leaves they had greater egg production compared with those adults fed the viridiflorol leaves. Moreover, adult pre-oviposition period was extended (1.5-fold) when individuals were fed the viridiflorol leaves compared with those fed the E-nerolidol leaves. By rearing the O. vitiosa weevil on the more nutritious chemotype plants these results assisted in the mass production and establishment of the M. quinquenervia biological control agent.  相似文献

For Thymus pulegioides L. (Lamiaceae), occurring in almost entire Europe, about 20 different essential oil chemotypes are described in approximately 25 studies. However, only few studies mention chemotype diversity on population level, describing up to five chemotypes growing together. The aim of the study was to investigate the chemotype diversity within one T. pulegioides population of a limestone grassland in Germany. Essential oil compounds from samples of 93 thyme cushions were extracted with solid phase extraction (SPE) and analysed by GC-MS. Cluster analysis and nonmetric multi-dimensional scaling (NMDS) of the samples’ essential oil composition revealed the existence of seven different chemotypes: linalyl acetate-chemotype (56 individuals), geraniol-citral-linalyl acetate-chemotype (5), geraniol-citral-chemotype (7), thymol-chemotype (8), carvacrol-chemotype (2), linalool-chemotype (1), and β-caryophyllene-germacrene D-β-bisabolene-chemotype (14). The pattern of major monoterpenes from all chemotypes could be explained by the inhibition of specific steps in monoterpene biosynthesis. The sesquiterpene-dominant chemotype might be caused by a block in a very early step of monoterpene production or a regulatory/channeling mechanism. While the geraniol-citral-linalyl acetate-chemotype has been found for the first time, the other chemotypes have been found in other regions and the β-caryophyllene-germacrene D-β-bisabolene-type is similar to other sesquiterpene chemotypes rarely found in other areas. The large intra-population chemical polymorphism in this study raises questions about the chemotype diversity of thyme in other regions of Germany and potential correlations between essential oil composition and abiotic factors or biotic interactions.  相似文献

ThreeFusarium species:F. graminearum, F. culmorum andF. cerealis were identified in laboratory cultures and in sporodochia from spikelets of scabby wheat. SCAR (sequence characterized amplified region) primers were used to identifyFusarium species and nivalenol (NIV) and deoxynivalenol (DON) chemotypes within species in laboratory cultures and field collected heads harvested in 2006. Results from PCR analyses confirmed preliminary identifications of species on the basis of examination of macroconidia under a light microscope and identification of cultures on agar media. NIV and DON (3Ac-DON and 15Ac-DON) chemotypes were identified using PCR assay. Among samples and isolates ofF. graminearum, the 15Ac-DON chemotype dominated, and among those whereF. culmorum was identified, the 3Ac-DON chemotype prevailed. Only 5 of the 41 isolates ofF. graminearum tested, displayed the NIV chemotype. An increase in the frequency ofF. graminearum and a decrease in the frequency ofF. culmorum were found during 1998 to 2006.  相似文献

Individual leaves of the three most common chemotypes of Melaleuca alternifolia were examined both quantitatively and qualitatively for volatile constituents from the emergence of the first true leaves, through to 6-week-old tenth leaf set material. The 1,8-cineole and terpinolene chemotypes were investigated and compared with the recently reported commercial terpinen-4-ol chemotype. The 1,8-cineole chemotype was found to accumulate 1,8-cineole and associated p-menthanes limonene, terpinen-4-ol and alpha-terpineol gradually with increasing leaf set number. As with the terpinen-4-ol variety, higher than expected concentrations of the pinenes and terpinolene were found only in the early leaf sets. The terpinolene variety showed two stages of terpinolene accumulation, the first at leaf sets 2-3 similar to the unexpected biosynthesis of terpinolene in the terpinen-4-ol chemotype and the second at leaf sets 8-9 which is characteristic of the terpinolene variety.  相似文献

Essential oil (EO) extracts coming from two representative populations of Mentha suaveolens Ehrh . subesp. suaveolens in Eastern Iberian Peninsula were analyzed by gas chromatography coupled with mass spectrometry and flame ion detector. Plant sampling was carried out in the morning and evening in order to study diurnal variation in EO profiles. Likewise, leaves and inflorescences were analyzed separately. Two chemotypes corresponding to each one of the populations were identified, with piperitenone oxide (35.2 – 74.3%) and piperitone oxide (83.9 – 91.3%), respectively, as major compounds. Once different chemotypes were identified, canonical correspondence analysis was employed to evaluate the effect of the bioclimatic and edaphic factors recorded in each location on the observed differences. Statistical analysis suggested that these chemotypes were closely related to specific environmental factors, mainly the bioclimatic ones. Concretely, piperitenone oxide chemotype can be associated to supramediterranean bioclimatic conditions and soils with major salinity and water field capacity. On the other hand, the most volatile fraction (hydrocarbon monoterpenes) reached its higher level in the morning; specifically, a noticeable amount of limonene was found in morning samples of flowers (4.8 – 10.6%). This fact can be related to ecological role of volatile compounds in order to attract pollinator insects.  相似文献

Fifty-five strains of enterococci isolated from the piglet intestine were characterized in vitro for probiotic activity. Identification of the isolates revealed Enterococcus faecium as the predominant species (84 %). Forty strains (73 %) were found to produce bacteriocin-like substances (only into solid media) with activity almost only toward Gram-positive genera. Thirty-eight % of strains were resistant to tetracycline, 27 % to chloramphenicol, 18 % to erythromycin and 16 % to vancomycin. In addition to control of strain safety, 6 % of isolates were β-hemolytic and 16 % produced gelatinase. Seven strains selected for further probiotic assays exhibited sufficient survival rate at pH 3.0 after 3 h, in the presence of 1 % ox-bile and lysozyme after 1 d (over 107 CFU/mL in all tests). The adhesion of tested strains to porcine and human intestinal mucus was found in a similar range (1.4–14.0 % and 1.4–17.6 %, respectively). In accordance with current research effort to use and/or to combine various health promoting substances, the sensitivity of all isolates toward plant extracts and toward bacteriocins produced by animal and environmental strains was determined. All enterococci were sensitive toward oregano and sage extracts and toward one (E. faecium EF55 — chicken isolate, activity of 25 600 AU/mL) of ten bacteriocin substances. It means that a similar anti-enterococcal potential of some bacteriocin substances may be observed as for certain plant extracts.  相似文献

The pathogenicity of nine Fusarium graminearum isolates belonging to three different trichothecene-producing chemotypes, i.e. NIV + FUS, DON + 3-AcDON, and DON + 15-AcDON was tested on corn, wheat, triticale and rye. Although these compounds are known to have different toxicity, no difference in pathogenicity was observed among the mean values for chemotypes. However, within each chemotype there were isolates with different pathogenicity which was not correlated with the ability to produce trichothecenes in vitro. The pathogenicity of each isolate was almost the same for all tested cereals. The involvement of trichothecenes in pathogenesis was investigated in infected corn stalk tissues. All the expected trichothecenes were detected with the exception of NIV and FUS.  相似文献

Fusarium graminearum clade species are among the main causative agents of Gibberella ear rot (GER) in maize and responsible for the various trichothecene mycotoxins accumulated in contaminated maize grains. In this study, a total of 620 isolates from diseased maize ears collected from 59 districts in 19 provinces throughout China, previously identified morphologically as Fusarium graminearum clade, was genetically characterized at the species level based on SCAR (Sequence Characterized Amplified Region) and for their potential capability of mycotoxin production using the genetic chemotyping assay. The results showed that 359 isolates were F. asiaticum (SCAR 5), which consisted of 97% nivalenol (NIV)‐chemotypes, 0.8% 3‐acetyldeoxynivalenol (3‐ADON)‐producing isolates and 2.2% 15‐acetyldeoxynivalenol (15‐ADON) producers, whereas the remaining 261 isolates were identified as F. graminearum sensu stricto (SCAR 1), all of which produced 15‐ADON mycotoxins. This high proportion of NIV producers present in F. asiaticum is different from the chemotype patterns in F. asiaticum populations isolated from wheat and barley, where DON and its acetylated chemotypes were the predominant mycotoxins. Moreover, the majority of NIV producers (59.1%) and all the 3‐ADON‐producing strains were derived from the warmer regions in southern China, whereas most of the 15‐ADON‐producing strains (78.4%) were isolated from the colder regions in northern China. Our study is the first report of NIV chemotypes of F. asiaticum and 15‐ADON chemotypes of F. graminearum sensu stricto that were associated with the GER of maize in China.  相似文献

The essential oil components and a karyotypic analysis of five Lippia alba (Verbenaceae) accessions from Brazil were performed with the objective of investigating the variation among different populations. The chemistry analysis allowed the grouping of the accessions in two main chemotypes: neral chemotype (LaCat, LaJF and LaRJ) and linalool chemotype (LaGua and LaVC). However, large karyotypic differences, verified by different chromosome banding techniques, were not detected. The results presented the same chromosome number for all accessions (2n = 30) with 10 metacentric chromosomes and 5 submetacentric. The chromosome banding showed great blocks of constitutive heterochromatin (C-bands) around the centromeric region, which was rich in AT bases (DAPI+), while the CMA bands were observed only in terminal regions of six chromosomes. Through Ag-NOR techniques, only two active pairs of NORs were detected on the three pairs of secondary constrictions (the NOR activity is discussed). This work relates the pattern of heterochromatin for Lippia alba for the first time.  相似文献
