Somatic Embryogenesis from Clonal Leaf Tissues of Cassava   总被引:3,自引:0,他引:3  
Leaf lobes were isolated from palmate leaves of clonal cassava(Manihot esculenta Crantz) material growing in vitro or in glasshouseconditions and subjected to a two-stage culture procedure involvingincubation on Murashige and Skoog (MS2) basal medium supplementedwith 2–12 mg l–1 2,4-D for 20 d (Stage I) beforetransfer to MS2 basal medium supplemented with 0.01 mg l–12,4-D and 0.1 mg l–1 6-benzylamino purine (BAP) (StageII medium). Embryogenetic tissues, foliose structures and somatic embryosdeveloped from leaf lobes at all Stage I 2,4-D concentrations,except on those explants isolated from shoot-tip cultures incubatedon MS2 basal medium supplemented with 0.1 mg l–1 NAA and1.0 mg l–1 BAP. Leaf lobes isolated directly from glasshouse plants showed optimalembryogenetic competence when subjected to a Stage I cultureperiod of 17 d, although foliose structure initiation was optimalwith shorter Stage I durations. Leaf lobes of 2–4 mm lengthand those isolated from phyllotaxic leaf numbers 4 and 5 showedthe greatest embryogenetic competence. Embryogenic cell suspension cultures were established from calliderived from young leaves of sugarcane (Saccharum officinarumL.) by placing them in liquid medium containing 5 per cent coconutwater (CW), 2–3 mg 1–1 2, 4-D and 500 mg 1–1casein hydrolysate (CH). The cultures were maintained by transferring2.5–5.0 ml of the suspension to 35 ml of fresh mediumevery 4–5 days. Organized structures resembling the earlystages of embryogeny were formed when 2, 4-D in the medium waslowered (0.1–1.0 mg 1–1) but these did not developbeyond the globular or early scutellar stages. High levels ofsucrose (6–10 per cent) promoted the formation of proembryoids.Plating of the suspension on MS agar medium supplemented with0.25–2.0 mg 1–1 2, 4-D, 5 per cent CW, 500 mg 1–1CH, with or without activated charcoal, resulted in the formationof embryogenic calli. A large number of embryoids were formedin media containing lower 2, 4-D concentrations. Transfer ofembryoids to half-strength MS medium with 6 per cent sucroseestablished plantlets which were successfully transferred tosoil. Dunaliella tertiolecta grew in a medium that contained MgSO4(MgSO4 medium), while this alga did not grow at all in mediathat contained MgCl2 or Mg(NO3)2. The growth in the MgSO4mediumwas inhibited in comparison with that in media that containedsodium salts, such as NaCl, NaNO3, and Na2SO4. The energy chargeobtained from measurements of levels of adenine nucleotidesby HPLC were almost constant in Na- and Mg-containing media(about 0.87), indicating that the failure of growth in MgCl2medium and Mg(NO3)2 medium was not directly related to.changesin the energy metabolism. K+ and Mg2+ were the dominant intracellularcations not only in Na-containing media (Na-media) but alsoin Mg-cohtaining media (Mg-media). The intracellular concentrationof Ca2+ was lower in Mg-media (1.6 mM) than that in Na-media(6mM). The concentrations of HPO42– in cells incubatedin Mg-media were lower (less than 60 mM) than those in Na-media(greater than 110 mM). By contrast, the intracellular concentrationof SO42– was higher in a MgSO4 medium (26 mM) than thatin a Na2SO4 medium (4 mM) which, at least, compensated by 40%for the decrease in HPO42–. Non-selected and Na2SO-, K2SO4- or KCl-selected callus culturesof Vaccinium corymbosum L. cv. Blue Crop were grown on mediasupplemented with 0, 25 and 50 mM Na2SO4 (non-selected and Na2SO(-selectedonly), 0, 25 and 50mMK2SO4 (non-selected and K2SO4-selectedonly) or 0, 50 and 100 mM KCl (non-selected and KCl-selectedonly). On all media, growth of selected callus (on a fresh-weightor dry-weight basis) was greater than that of non-selected callus,and selected callus grew optimally on the level and type ofsalt on which it was selected. Selected callus was friable andmaintained a higher f. wt:d. wt ratio. Tissue water potentialin selected callus was more negative than in non-selected callus. Flame photometry and chloridometry showed Na+, K+ and Claccumulated in callus to concentrations equal to or greaterthan the initial concentration in the medium. Turbidometry showedthat tissue SO42- concentration was lower than the concentrationin the medium. In most cases selected callus accumulated moreNa+, Ksup, SO42– or Cl than non-selected callus.Vacuolar ion concentration was measured by electronprobe X-raymicroanalysis, and on most media selected callus had highervacuolar ion concentrations than non-selected callus. SO42–and Cl were accumulated in the vacuoles at concentrationshigher than the external medium, but vacuolar Na+ concentrationdid not reach external concentration on Na2SO4 and on potassiumsalts was maintained between 12 and 17 mM. Vacuolar K+ concentration(approx. 142–191 mM on no salt) decreased on Na2SO4 andincreased on K2SO4 and KCl. There was no precise correlation between total or specific ionaccumulation (Na+, K+, SO42– and Cl and fresh-weightyield. Results suggest that selection results in adaptationin response to decreased water potential of the medium. To find whether cytoplasmic streaming in Acetabularia is controlledby Ca2+, a tonoplast-permeabilized cell model was prepared usinga vacuolar perfusion technique. The cytoplasmic streaming remainedalmost normal after perfusion with EGTA medium (10 mM EGTA,40 mM PIPES, 5mM MgCl2 and 800 mM sorbitol, pH 6.9), but stoppedwithin 10 min when saponin medium (EGTA medium plus 50 µg/mlsaponin, 50 µg/ml hexokinase and 5 mM glucose) was perfused.This model system was reactivated with a solution containing0.5 mM ATP and different concentrations of Ca2+ (reactivationmedium). With the reactivation medium at pCa 6–5, theresumed streaming lasted for about 10 min before the cytoplasmaggregated. At pCa 4–3, the streaming was observed onlyfor a few minutes because the cytoplasm aggregated quickly.At pCa 7, no reactivated movement was observed. Reactivationwas not induced in an ATP- or Mg2+-deficient medium even inthe presence of an adequate concentration of Ca2+, and was inhibitedby 50 µg/ml cytochalasin B or 1 mM N-ethylmaleimide. We concluded from these observations that the cytoplasmic streamingin Acetabularia is very likely to be driven by the actomyosinsystem in the presence of Mg-ATP and Ca2+ at pCa 6–5. Callus of sainfoin (Onobrychis viciifolia Scop.) was initiatedfrom stem and root explants which were obtained from seedlingsgrowing in vitro, on Linsmaier Skoog (LS) medium supplementedwith 1 mg l–1 2, 4-D and 1 mg l–1 BA or only 1 mgl–1 BA, and the Vacin and Went medium without hormones.Somatic embryos were formed on LS medium containing 1 m l–1BA. Treatment of S-sufficient or S-deficient Chlorella pyrenoidosacells with NaHSO3, during an 8-h period in the light, significantlydecreased their chlorophyll and dry matter contents when thecells were incubated in the presence or absence of SO42+. Incontrols lacking HSO3, when the starting pH was 7.5,dry matter and chlorophyll contents increased slightly, whereasno significant changes in either occurred at a starting pH of3.0 when the cells and medium contained SO4. In the dark,at both pH 3.0 and 7.5, dry matter and chlorophyll contentsdecreased slightly. Bisulphite treatment in the dark causedlittle decrease of either dry matter or chlorophyll when cellsand medium contained SO42–. However, in its absence, drymatter decreased markedly, but there was little change of chlorophyllcontent in the dark. The interactions between HSO3 asa source of S and as an inhibitor of growth and chlorophyllformation are discussed in the context of the changes inducedby light and alternative sources of S. Overall, the harmfuleffects of HSO3 outweigh any role it has as a sourceof S, since its effects are ameliorated by SO42–.  相似文献

The intracellular concentration of the major ions in Dunaliellasalina cells were determined, following the removal of extracellularions by ion-exchange minicolumns. Log phase cells, grown inmedia containing 1–4 molar NaCl, contained 30–50mM chloride and 200–350 mM magnesium (5 mM in medium).Phosphorus, which is present intracellularly mostly as polyphosphate,was present in amounts of 60–100 fmoles per cell, equivalentto a concentration of 600–1,000 mM (0.2 mM in medium).Previous data indicated that such cells contained 20–40mM Na+, 150–300mM K+, 20mM SO2–4, and very low concentrationsof Ca2+ and charged nitrogenous compounds. Mg2+ and K+ seemto serve as the major counter ions for the intracellular negativecharge present in the massively accumulated polyphosphates.The former accounts for about 2/3 of the required positive charge.This is supported by the observation that limitation in thephosphate or K+ supply in the medium lead to a parallel decreasein the accumulation of intracellular phosphorus, Mg2+ or K+. 1Present address: Department of Vegetables, The Volcani Center,Bet-Dagan 50250, Israel. An efficient medium for multiplication of Prunus cerasifera,J. F. Ehrh. cv. ‘Atropurpurea’, Pissardi plum, consistedof Linsmaier-Skoog basal medium (LS) supplemented with 3% sucrose,10mg 1–1 N4-(2-isopentenyl) adenine (2iP), and 162 mg1–1 phloroglucinol (Phl). Phl in the medium significantlyenhanced growth (P < 0.1 %) over cultures maintained on LSmedium with 10 mg 1–1 2iP and no Phl. Conidial germination in liquid shake culture of Neurospora crassawas affected by the number of conidia in the medium (conidialdensity effect) with the optimum density being around 2?106conidia/ml. The conidial density effect at 2?104 and 2?105 conidia/mlwas eliminated by the addition of auxin, IAA or 2,4-D with theoptimum concentrations of IAA and 2,4-D being around 10–6M. IAA and 2,4-D had no effect on the effect at 2?107 conidia/ml.An active substance(s) for conidial germination which was relatedto the conidial density effect was detected in the cell-freefiltrate of the germination medium, and was found to be non-dialyzableand thermolabile. At the early stage of germination, the concentrationof active substance(s) in the medium increased in proportionto the conidial density and reached a supraoptimum amount forgermination at 2?107 conidia/ml. IAA (10–6M) enhancedthe concentration. Endogenous auxin concentrations in filtratesof the germination media containing 2?105, 2?106 and 2?107 conidia/mlwere 5.8?10–12, 4.6?10–11 and 1.7?10–10M IAAequivalent, respectively. A study was made of the effects of cadmium on the Cyanobacterium(blue-green alga) Anabaena cylindrica Lemm. as part of the paddy-fieldecosystem. A simple culture vessel has been designed, which allows periodicalmeasurement of growth (optical density) and nitrogenase activity(C2H2-C2H4 method). The influence of medium renewal was checked:the renewal of the medium maintained a higher growth rate andhigher nitrogen fixation ability. The cadmium effects were studied using six concentration levelsranging from 0 (control) to 2 parts 10–6 with renewedmedia (10% every day). No significant differences could be seen up to 1 part 10–6for nitrogenase activity and relative percentage of heterocysts(decreasing as a function of time from ±4% to ±1.5%). Inhibition of growth (OD and dry weight) was weak at 1 part10–6 but important at 2 parts 10–6; at this concentrationcadmium induced morphological and physiological effects: chlorosis,cellular malformations and destruction, and increase in heterocystfrequency (up to 7.72% ±0.19). The response of isolated stolons cultured in vitro, to abscisicacid (ABA) has been studied in the presence and absence of kinetin(6-furfurylaminopurine). ABA alone in concentrations from 7.5x 10–4 mM to 7.5 x 10–2 mM, inhibited stolon elongationbut failed to promote tuber initiation. In the presence of kinetin,ABA at concentrations of 3.0 x 10–2 and 7.5 x 10–2mM markedly inhibited kinetin-induced tuber initiation and stolonelongation, but at 7.5 x 10–4 and 7.5 x 10–3 mMABA did not prevent tuber initiation. When stolons were incubated on a medium containing kinetin andlater transferred to one containing ABA with or without kinetin,the inhibitory effect of ABA decreased appreciably as the timeof incubation on kinetin is increased. The results are discussed in relation to the role of ABA inthe inhibition of nucleic acid and protein synthesis and theinteraction with cytokinins and the possible effect of ABA onkinetin uptake, transport and accumulation at the locus of action. Phosphate uptake rates in Synechococcus R-2 in BG-11 media (anitrate-based medium, not phosphate limited) were measured usingcells grown semi-continuously and in continuous culture. Netuptake of phosphate is proportional to external concentration.Growing cells at pHo 10 have a net uptake rate of about 600pmol m–2 s–1 phosphate, but the isotopic flux for32P phosphate was about 4 nmol m–2 s–1. There appearsto be a constitutive over-capacity for phosphate uptake. TheKm and Vmax, of the saturable component were not significantlydifferent at pHo 7.5 and 10, hence the transport system probablyrecognizes both H2PO4and HPO2–4. The intracellularinorganic phosphate concentration is about 3 to 10 mol m–3,but there is an intracellular polyphosphate store of about 400mol m–3. Intracellular inorganic phosphate is 25 to 50kJ mol–1 from electrochemical equilibrium in both thelight and dark and at pHo 7.5 and 10. Phosphate uptake is veryslow in the dark ( 100 pmol m–2 s–1) and is light-activated(pHo 7.51.3 nmol m–2 s–1, pHo 10600 pmol m–2s–1). Uptake has an irreversible requirement for Mg2+in the medium. Uptake in the light is strongly Na+-dependent.Phosphate uptake was negatively electrogenic (net negative chargetaken up when transporting phosphate) at pHo 7.5, but positivelyelectrogenic at pHo 10. This seems to exclude a sodium motiveforce driven mechanism. An ATP-driven phosphate uptake mechanismneeds to have a stoichiometry of one phosphate taken up perATP (1 PO4 in/ATP) to be thermodynamically possible under allthe conditions tested in the present study. The mechanism of the Ca2+-dependent Cl efflux was studiedin tonoplast-free cells, in which the intracellular chemicalcomposition can be freely controlled. Tonoplast-free cells wereprepared by perfusing the cell interior of internodal cellsof Chara corallina with a medium that contained EGTA. The Ca2+-inducedCl efflux was measured together with the membrane potentialduring continuous intracellular perfusion. The dependenciesof Cl efflux and the membrane potential on the intracellularCa2+ or Cl concentrations were analyzed. When perfusionwas started with medium that contained Ca2+ ions, Clefflux and membrane depolarization were induced. The amountof Cl efflux varied considerably among individual cells.The rate of efflux decreased exponentially but a residual effluxremained detectable. The Cl efflux was induced at concentrationsof Ca2+ ions above 1 µM and reached a maximum at 1 mM.By contrast, the membrane depolarization reached a maximum atabout 10 µM Ca2+. The rate of Cl efflux increasedlinearly with logarithmic increases in the intracellular Clconcentrations. These findings suggest that more than two kindsof Ca2+-dependent Cl channel might be present in theplasma membrane. Addition of ATP or its removal from the perfusion medium didnot affect the Ca2+-dependent Cl efflux. The standard synthetic culture medium (Stuart and Street, 1969)has been modified by adjustment of its initial pH to 6.4 andby the addition of gibberellic acid (0.25 mg/l) and of a mixtureof 15 L-amino acids formulated from an analysis of the conditionedmedium. The minimum effective density for the growth of sycamorecell suspensions in the standard medium is 9–15 x 103cells ml–1, for the modified synthetic medium it is 2.0x 103 cells ml–1, and for conditioned medium 1.0–1.25x 103 cells ml–1. Using either conditioned medium (Stuart and Street, 1969) orthe modified synthetic medium it is demonstrated that the growthof cultures initiated at low density is enhanced by a volatilefactor released from actively growing cell suspensions. In presenceof conditioned medium and this volatile factor cultures canbe established from stationary-phase cells at a density of 6x 102 cells ml–1. The volatile factor can be absorbedin 40 per cent w/v KOH but attempts to replace the factor byair containing carbon dioxide at concentrations up to 5 percent have so far been unsuccessful.  相似文献

We have assessed the effect of various medium supplements inpromoting the ability of maize (Zea mays L.) inbred FR27rhmsuspension cultures to grow following a period of 4 °C chillingstress. Following a 4 week exposure to 4 °C in culture mediumwithout proline, no cell growth occurred upon subsequent incubationat 28°C for 2 weeks. This inhibition was reversed when 3to 48 mol m–3 proline or 0.1 mol m–3 putrescineor 0.01 mol m–3 spermidine were present in the mediumduring the chilling stress. On the other hand, suspensions weremade more sensitive to 4°C by blocking polyamine biosynthesiswith 1.0 mol m–3 methylglyoxal bis (guanylhydrazone) (MGBG)or a combination of 1.0 mol m–3 difluoromethylornithine(DFMO) and 1.0 mol m–3 difluoromethylarginine (DFMA).The addition of 10 mol m–3 putrescine to the suspensioncontaining DFMO and DFMA prevented the increased chilling sensitivity.Electrolyte leakage studies conducted to assess membrane integrityafter 4 weeks at 4°C and a 2 week regrowth period showedthat cells treated with no polyamines (control), 0.01 mol m–3spermidine, 1.0 mol m–3 putrescine, or 1.0 mol m–3MGBG lost 43, 32, 14, and 100% of the total electrolyte pool,respectively. These results suggest that proline and polyaminesare beneficial for inducing chilling tolerance in FR27rhm suspension. Key words: Proline, polyamine, chilling stress  相似文献

Callus and suspension cultures of Theobroma cacao L., initiatedfrom immature cotyledons of beans from pods harvested 120–130days after pollination were established. A modified B-5 or Murashige—Skoogagar medium sustained growth of callus without loss of vigourafter each sub-culture. A 15-fold weight increase occurred duringthe 4 week culture periods at 30 ± 1 °C. Coconutwater improved callus growth substantially. The optimum hormonalconcentrations for growth of suspensions were 0.5 mg 1–1of 2, 4-dichlorophenoxyacetic acid and 0.1 mg I–1 of kinetinin a Murashige—Skoog basal medium liquid medium. The optimumtemperature for growth of suspensions was 25–30 °C.The cell number and cell mass of suspensions increased 20-foldin 14 days. No organogenesis or embryogenesis was observed. Internodal cells of Chara australis were subjected to two consecutiveintracellular perfusions with a Ca2+-free EGTA medium whichdisintegrated the tonoplast within about 10 minutes and thenwith a Ca2+-buffered medium. All perfusion media usually contained1 mM ATP. To stop the electrogenic pump, the internode was depletedof intracellular ATP. The excitability of the plasmalemma wasnot significantly influenced by intracellular free Ca2+ concentrationsup to 10–4 M. To trigger action potentials, minimum currentdensities of 1 to 2 µA cm–2 had to be applied atall tested Ca2+ concentrations. Merr.] is inhibited by continuous growth in the presenceof 4· mol m–3 KNO3 The presence of 4·0 molm–3 ‘starter nitrate’ for 3-6 d during noduledevelopment, however, subsequently stimulates nodule dry weightaccumulation and nitrogenase activity. These stimulations occureven though 4· mol m–3 nitrate temporarily delaysnodule development, i.e. the late steps of nodule developmentare reversibly inhibited by a short-term exposure to 4·0mol m–3 nitrate. On the other hand, treatment with 4·0mol m–3 nitrate in excess of 14 d significantly reducesnodule dry weight Thus, extended growth in the presence of 4·0mol m–3 KNO3 seems to block both early and late stepsof nodule development. Nodulation of hydroponically grown soybeansis also inhibited by continuous growth in the presence of 2·0mol m–3 (NH4)2SO4 This inhibition is not caused by acidityof the growth medium. On the other hand, nodule development6 d after inoculation with Rhizoblum japonicum is not delayedby a 7-d exposure to 2·0 mol m–3 (NH4)2SO4 butis partially inhibited by a prolonged exposure to (NH4)2SO4Because repression of nodulation by 4·0 mol m–3KNO3 is more severe than that by 2·0 mol m–3 (NH4)2SO4and because ammonium taken up by the soybean plant is not activelyoxidized to nitrate, it is suggested that there are at leasttwo mechanisms by which nitrate utilization represses noduleformation in soybean. Tobacco RuDP carboxylase is completely soluble in 0.07 M NaClor 0.01 M Na2SO2, but is almost completely insoluble in salt-freesolutions at 40°C; the solubility seeming to depend on ionicstrength. Lowering the temperature increased solubility of theprotein. The solubility in 0.01–0.04 M NaCl at 0°Cwas more than double that at 40°C. RuDP solubilized theprotein even in a salt-free medium. The protein became insolubleagain on the addition of various divalent cations. Effectivenessof the metal ions was Zn++> Ni++>Co++>Mn++>Mg++>Ca++.Although most of the metal ions inhibited (Mg++ activated) enzymeactivity, no direct correlation was found between the degreeof solubility depression and the degree of enzyme inhibition.
