Selenium (Se) essentiality to plants has not been demonstrated although evidence indicates that it plays a significant role as antioxidant in higher plants. Research concerning to the uptake and allocation of Se in plant tissues is reported in numerous works. However, the effect of sulfur (S) on both the distribution and the antioxidant ability of Se in selenite-treated plant remains unclear. In this work the effect of S application (0–100 mg S kg−1 soil) on shoot Se concentration of Lolium perenne cv. Aries was studied. Se distribution into different fractions of plants supplied with selenite (2 mg Se kg−1 soil) and the state of the antioxidative system were determined. Results showed that shoot Se concentration decreased at least 33% by S application. Plants supplied with S registered the lowest GSH-Px activity and the highest lipid peroxidation. Most of Se was incorporated into the organic fraction of the plant tissue irrespective of the S treatment. However, a significant decrease of both the soluble protein and the amino acid fraction occurred, and the residual Se fraction seemed to increase at expense of the organic-Se soluble fraction. Although no essential selenoproteins have been clearly identified in vascular plants, the decrease of the soluble protein fraction and the different pattern of protein synthesized (SDS-PAGE analysis) may explain the observed reduction of the GSH-Px activity.  相似文献

Selenium is an essential element for antioxidation reactions in human and animals. In order to study its biological role in higher plants, ryegrass (Lolium perenne) was cultivated in a soil without Se or amended with increasing dosages of H2SeO4 (0.1, 1.0, 10.0 and 30.0 mg Se kg−1). Ryegrass was harvested twice and the yields were analyzed for antioxidative systems and growth parameters. Selenium exerted dual effects: At low concentrations it acted as an antioxidant, inhibiting lipid peroxidation, whereas at higher concentrations, it was a pro-oxidant, enhancing the accumulation of lipid peroxidation products. The antioxidative effect was associated with an increase in glutathione peroxidase (GSH-Px) activity, but not with superoxide dismutase (SOD) and αα-tocopherol, which was the only tocopherol detected. In the second yield, the diminished lipid peroxidation due to a proper Se addition coincided with promoted plant growth. The oxidative stress found at the Se addition level ≥ 10 mg kg−1 resulted in drastic yield losses. This result indicates that the toxicity of Se can be attributed, in addition to metabolic disturbances, to its pro-oxidative effects. Neither the growth-promoting nor the toxic effect of Se could be explained by the changes in the total chlorophyll concentration. Selenium (Se) is an essential micronutrient for animal and human nutrition, but whether it is essential to plants remains controversial. However, there are increasing experimental evidences that indicate a protective role of Se against the oxidative stress in higher plants through Se-dependent glutathione peroxidase (GSH-Px) activity. The effects of the Se chemical forms, selenite and selenate, the rate of their application on shoot Se concentration and their influence on the antioxidative system of ryegrass (Lolium perenne cv. Aries), through the measurement of GSH-Px activity and lipid peroxidation, were evaluated in an Andisol of Southern Chile. Moreover, a soil–plant relationship for Se was determined and a simple method to extract available Se from acid soils is proposed. In a 55-day experiment ryegrass seeds were sown in pots and soil was treated with sodium selenite or sodium selenate (0–10 mg Se kg−1). The results showed that the Se concentration in shoots increased with the application of both selenite and selenate. However, the highest shoot Se concentrations were obtained in selenate-treated plants. For both sources of Se, there was a significant positive correlation between the shoot Se concentration and the GSH-Px activity; and the Se-dependence of this enzymatic activity was related especially with the chemical form of applied Se rather than the Se concentration in plant tissues. Furthermore, the lipid peroxidation, as measured by Thiobarbituric Acid Reactive Substances (TBARS), decreased at low levels of shoot Se concentration, reaching the lowest level at approximately 20 mg Se kg−1 in plants and then increased steadily above this level. In addition, the acid extraction method used to evaluate available Se in soil showed a positive good correlation between soil Se and shoot Se concentrations irrespective of chemical form of Se applied.  相似文献

Summary Applications of chlorsulfuron (11.25 g ai ha−1) were made to wheat, flax, canola and lentils at spray volumes of 48, 108 and 2171 ha−1, and with metsulfuron methyl (6.00 g ai ha−1) at spray volumes of 48 and 2171 ha−1. Applications were made to the shoot only, the soil only and to the plant plus soil. Spectrofluorometric analysis was used to determine spray partitioning within the plant-soil system and foliar retention was related to efficacy. Fresh and dry weights of shoot material were determined 3 weeks after treatment. Flax and wheat were more tolerant of restricted-foliar applications than those made to the soil, the converse being true to canola and lentils. Applications made to the plant and soil were always the most deleterious. Foliar retention and efficacy did not correlate directly. Applications in 2171 ha−1 were generally more efficacious than those at 481 ha−1.  相似文献

Biological volatilization of selenium (Se) in contaminated areas represents an environmentally friendly phytoremediation approach. Implementation of phytovolatilization technology for the remediation of Se-contaminated soils or sediments is oftentimes limited by its low remediation efficiency under field conditions. This greenhouse study determined the feasibility of manipulating soil organic content and hydraulic conditions in a soil–pickleweed (Salicornia bigelovii) system for the enhancement of Se volatilization. Based on annual shoot biomass production rate under field conditions (approximately 1.5 kg m−2), the addition of pickleweed shoot tissues to the soil surface resulted in 2.2-fold more biogenic volatile Se than the control, up to 251.6 ± 140.5 μg m−2 d−1. Selenium volatilization was significantly reduced at a soil water potential of −25 kPa, but substantially increased after re-irrigation to 0 kPa. In a 42-day experiment, the rate of Se volatilization was significantly correlated with soil water potential (P < 0.0001). Findings from this study demonstrate that Se volatilization be substantially enhanced by amending soil with pickleweed residues and by creating wetting and drying cycles that can be monitored with soil water potential probes in the field.  相似文献

Changes in contents of reactive oxygen species (O2 and H2O2) and non-enzymatic antioxidants, activities of antioxidant enzymes and lipid peroxidation were investigated during senescence of detached cucumber cotyledons dipped in water (control) and 20 mg dm−3 triadimefon (TDM). O2 and H2O2 accumulation and lipid peroxidation were observed during senescence of cucumber cotyledons, which coincided with a drop in the contents of carotenoids (Car) and ascorbic acid (AsA), and the activities of superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX), and an increase in the activity of peroxidase (POD). However, TDM could significantly inhibit the accumulation of O2 and H2O2, and lipid peroxidation by preventing the decrease of CAT, APX, Car and AsA and the increase of POD, while TDM had little effect on SOD activity during the senescence. Therefore we can draw a conclusion that TDM protects the membrane system and retards the senescence of detached cucumber cotyledons. A field study was carried out near Zürich (Switzerland) to determine the yield of symbiotically fixed nitrogen (15N dilution) from white clover (Trifolium repens L.) grown with perennial ryegrass (Lolium perenne L) and from red clover (Trifolium pratense L.) grown with Italian ryegrass (Lolium multiflorum Lam.). A zero N fertilizer treatment was compared to a 30 kg N/ha per cut regime (90 to 150 kg ha−1 annually). The annual yield of clover N derived from symbiosis averaged 131 kg ha−1 (49 to 227 kg) without N fertilization and 83 kg ha−1 (21 to 173 kg) with 30 kg of fertilizer N ha−1 per cut in the seeding year. Values for the first production year were 308 kg ha−1 (268 to 373 kg) without N fertilization and 232 kg ha−1 (165 to 305 kg) with 30 kg fertilizer N ha−1 per cut. The variation between years was associated mainly with the proportion of clover in the mixtures. Apparent clover-to-grass transfer of fixed N contributed up to 52 kg N ha−1 per year (17 kg N ha−1 on average) to the N yield of the mixtures. Percentage N derived from symbiosis averaged 75% for white and 86% for red clover. These percentages were affected only slightly by supplemental nitrogen, but declined markedly during late summer for white clover. It is concluded that the annual yield of symbiotically fixed N from clover/grass mixtures can be very high, provided that the proportion of clover in the mixtures exceeds 50% of total dry mass yield.  相似文献

Senescence is a developmentally regulated and highly ordered sequence of events. Senescence leads to abscission of plant organs and eventually leads to death of a plant or part of it. Present study revealed that Phalaenopsis flower undergo senescence due to over activation of O2 ·−generating xanthine oxidase (XO), which consequently increases the concentrations of O2 ·− leading to enhanced oxidative damage and disturbed cellular redox environment as indicated by increased lipid peroxidation and DHA/AsA + DHA ratio, respectively. While activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), and non-specific peroxidase (POD) were enhanced in sepals and petals of old flower, activities of catalase (CAT) and glutathione reductase (GR) were decreased. Exogenous application of nitric oxide (NO) retarded H2O2-induced senescence of Phalaenopsis flower by downregulating activity of XO and concentrations of O2 ·−, H2O2 and malondialdehyde (MDA, an index of lipid peroxidation). Exogenous application of NO also downregulated SOD activity and upregulated antioxidant enzymes involved in the detoxification of H2O2 (CAT and APX), and in the regulation of redox couples viz, monodehydroascorbate reductase (MDHAR) and GR, together with the modulation in non-protein thiol status and DHA/AsA + DHA ratio.  相似文献

The essential role of 6-benzylaminopurine (BA) in plant tissue culture has been widely known; however, physiological and biochemical mechanisms behind BA requirement have not been fully understood yet. BA may have an important role on callus growth by regulating antioxidant enzyme activities and acting as an effective free radical scavenger. To test this hypothesis, the impact of exogenous BA concentrations on antioxidative system in Vitis vinifera L. cv. ‘Bogazkere’ callus was investigated under in vitro conditions. Relative fresh weight growth (RFWG) of calli, total phenolics (TP) content, endogenous hydrogen peroxide (H2O2), malondialdehyde (MDA), proline concentrations, percentage of electrolyte leakage (EL), and some of the antioxidant enzyme activities; such as superoxide dismutase (SOD), and guaiacol peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX) were measured. Inhibitory effect of high concentrations of BA on antioxidant enzyme activities and RFWG was found. In the presence of BA at 0.1 mg L−1, SOD, POD, and APX activities decreased, while CAT activity increased in comparison with the controls. Exogenous BA treatments higher than 0.1 mg L−1 resulted in an increase in cellular TP, H2O2, MDA, proline contents, and percentage of EL, while RFWG of calli decreased. Based on the findings, it may be concluded that only 0.1 mg L−1 BA concentration combined with NAA could play a direct role in reducing the level of free radicals and phenolic production associated with proliferation capacity of grape cells under in vitro conditions. Furthermore, cytokinin was effective in the antioxidative enzyme system, lipid peroxidation, and electrolyte leakage.  相似文献

Dinitrogen fixation in white clover (Trifolium repens L.) grown in pure stand and mixture with perennial ryegrass (Lolium perenne L.) was determined in the field using 15N isotope dilution and harvest of the shoots. The apparent transfer of clover N to perennial ryegrass was simultaneously assessed. The soil was labelled either by immobilizing 15N in organic matter prior to establishment of the sward or by using the conventional labelling procedure in which 15N fertilizer is added after sward establishment. Immobilization of 15N in the soil organic matter has not previously been used in studies of N2 fixation in grass/clover pastures. However, this approach was a successful means of labelling, since the 15N enrichment only declined at a very slow rate during the experiment. After the second production year only 10–16% of the applied 15N was recovered in the harvested herbage. The two labelling methods gave, nonetheless, a similar estimate of the percentage of clover N derived from N2 fixation. In pure stand clover, 75–94% of the N was derived from N2 fixation and in the mixture 85–97%. The dry matter yield of the clover in mixture as percentage of total dry matter yield was relatively high and increased from 59% in the first to 65% in the second production year. The average daily N2 fixation rate in the mixture-grown clover varied from less than 0.5 kg N ha−1 day−1 in autumn to more than 2.6 kg N ha−1 day−1 in June. For clover in pure stand the average N2 fixation rate was greater and varied between 0.5 and 3.3 kg N ha−1 day−1, but with the same seasonal pattern as for clover in mixture. The amount of N fixed in the mixture was 23, 187 and 177 kg N ha−1 in the seeding, first and second production year, respectively, whereas pure stand clover fixed 28, 262 and 211 kg N ha−1 in the three years. The apparent transfer of clover N to grass was negligible in the seeding year, but clover N deposited in the rhizosphere or released by turnover of stolons, roots and nodules, contributed 19 and 28 kg N ha−1 to the grass in the first and second production year, respectively. The paper reports the effects of selenium (Se) supply on growth and antioxidant traits of wheat (Triticum aestivum L. cv Han NO.7086) seedlings exposed to enhanced ultraviolet-B (UV-B) stress. Antioxidant responses of seedlings were different depending on the Se concentration. Compared with the control, the lower amount used (0.5 mg Se kg−1 soil) had no significant effect on biomass accumulation. The treatments with 1.0, 2.0, and 3.0 mg Se kg−1 promoted biomass accumulation of wheat seedlings, and the increased amount in biomass was the most at 1.0 mg Se kg−1 treatment. Se treatments with 1.0, 2.0, and 3.0 mg kg−1 also significantly increased activities of peroxidase (POD) and superoxide dismutase (SOD) and reduced the rate of superoxide radical (O2) production and malondialdehyde (MDA) content of wheat seedlings. In addition, anthocyanins and phenolic compounds content in wheat seedlings evidently increased by the treatments with 1.0 and 2.0 mg Se kg−1. The lower Se treatment had no significant effect on MDA content, although it increased activities of antioxidant enzymes (POD, SOD, and catalase activities) and reduced the rate of O2 production in wheat seedlings. These results suggest that optimal Se supply is favorable for the growth of wheat seedlings and that optimal Se supply can reduce oxidative stress of seedlings under enhanced UV-B radiation.  相似文献

Thermotolerance and related antioxidant enzyme activities induced by both heat acclimation and exogenous salicylic acid (SA) application were studied in grapevine (Vitis vinifera L. cv. Jingxiu). Heat acclimation and exogenous SA application induced comparable changes in thermotolerance, ascorbic acid (AsA), glutathione (GSH), and hydrogen peroxide (H2O2) concentrations, and in activities of the antioxidant enzymes superoxide dismutase (SOD), peroxidase (POD), glutathione reductase (GR), ascorbic peroxidase (APX) and catalase (CAT) in grape leaves. Within 1 h at 38 °C, free SA concentration in leaves rose from 3.1 μg g−1 FW to 19.1 μg g−1 FW, then sharply declined. SA application and heat acclimation induced thermotolerance were related to changes of antioxidant enzyme activities and antioxidant concentration, indicating a role for endogenous SA in heat acclimation in grape leaves.  相似文献

Selenium (Se) is beneficial for some plants and is able to increase resistance and antioxidant capacity of plants subjected to stressful environment. In this work, the effects of enhanced ultraviolet-B (UV-B) radiation, Se supply, and their combination on growth and physiological traits of wheat (Triticum aestivum L., cv. Han NO.7086) seedlings were studied. The objective was to elucidate whether Se could alleviate the expected adverse effects of UV-B stress on seedlings. UV-B treatment caused a marked decline in growth parameters and total chlorophyll content and changed biomass allocation between aboveground and underground parts, which led to an increase in the root/shoot ratio. UV-B treatment also increased MDA content and the rate of superoxide radical (O2·−) production, although it increased some antioxidant (proline, phenolic compounds, and flavonoids) content and activity of antioxidant enzymes (peroxidase, superoxide dimutase, catalase (CAT)). Se treatment only increased total chlorophyll content and CAT activity. Compared with UV-B treatment alone, the combined treatment with UV-B and Se induced a significant increase in the biomass, total chlorophyll content, antioxidant content, and activity of antioxidant enzymes, and an evident decrease in MDA content and the rate of O2·− production. The results of this study demonstrated that Se alleviated the damage caused by UV-B to wheat seedlings to some extent by increasing antioxidant enzyme activity and antioxidant content.  相似文献

A hydroponic experiment was carried out in a growth chamber to investigate the impact of Selenium (Se) levels on physiological and biochemical characteristics of a barley cultivar. Membrane lipid peroxidation (LPO), proline accumulation and antioxidant activities of some enzymes of barley seedlings under Se toxicity were investigated. Significant increase in thiobarbituric acid reactive substance (TBARS) content, and a stimulation of catalase (CAT, 1.11.1.6), ascorbate peroxidase (APX, 1.11.1.11), glutathione reductase (GR, 1.6.4.2), and glutathione S-transferase (GST, 2.5.1.18) activities were recorded in barley seedlings subjected to 2, 4, 8, 16 ppm Se. Superoxide dismutase (SOD, EC 1.15.1.1) activity was not altered significantly. Plant height and chlorophyll content of the seedlings were also affected significantly in a dose dependent manner by Se treatment. Considerable amount of proline accumulation was also observed in response to Se treatment. The results indicated that increases in the activities of the antioxidant enzymes were not sufficient to protect cell membrane against Se toxicity.  相似文献

The paper reports the effects of selenium (Se) supply on growth and some physiological traits of wheat (Triticum aestivum L. cv Shijiazhuang NO. 8) seedlings exposed to drought stress. The growth and physiological responses of seedlings were different depending on the Se concentration. The higher (3.0 mg Se kg−1) and lower amount used (0.5 mg Se kg−1) did not significantly affect on biomass accumulation. Treatments with 1.0 and 2.0 mg Se kg−1 promoted biomass accumulation of wheat seedlings. Treatments at 1.0, 2.0, and 3.0 mg Se kg−1 significantly increased root activity, proline content, peroxidase (POD), and catalase (CAT) activities, carotenoids (Car) content, chlorophyll content, and reduced malondialdehyde (MDA) content of wheat seedlings. Lower Se treatment did not significantly effect on chlorophyll content and MDA content, although it also increased some antioxidant index (proline and Car content, POD and CAT activities) in wheat seedlings. These results suggest that optimal Se supply is favorable for growth of wheat seedlings during drought condition.  相似文献

Alfalfa (Medicago sativa L.) was grown in greenhouse sand culture to examine the effect of salinity composition and concentration on Se accumulation by plants. In a 2×2×4 factorial experiment, salinity was added as either C1 or SO 4 2− salts to the irrigating solution to achieve an electrical conductivity of 0.5, 1.5–3.0, or 6.0 dS m−1. Selenium was added to the nutrient solution at a concentration of 0.25 or 1.0 mg Se(VI)I−1. Following the third cutting, the roots were washed and all plant material analyzed for dry weight and Se. Plant biomass production decreased with additions of either Se or salinity, regardless of composition. In the presence of Se, the yield reduction was greater with Cl salinity than with SO 4 2− salinity. Plant Se accumulation was reduced from 948 mg Se kg−1 to 6 mg Se kg−1 in the presence of SO 4 2− salts (0.5 mmol SO 4 2− l−1 vs. 40 mmol SO 4 2− l−1) due to an apparent Se(VI) −SO 4 2− antagonism. This Se−SO 4 2− antagonism prevented accumulation of Se and reduced Se-induced toxicity. A lesser antagonistic effect on Se accumulation was observed between Cl, and Se. A synergistic interaction between SO 4 2− and Se(VI) increased plant S concentrations in the presence of the relatively low basal SO 4 2− concentrations but not at the higher solution SO 4 2− concentrations. In many areas, soil and water containing high Se concentrations also contain large amounts of SO 4 2− . The occurrence of SO 4 2− with Se reduces plant accumulation of Se(VI) and may lower the risk of Se overexposure to animals feeding on forage material grown in high Se−SO 4 2− regions.  相似文献

Shoot growth, root growth and macro-nutrient uptake by a high-yielding (5t/ha grain) winter oilseed rape crop have been measured. Maximum rooting density in the top 20cm of soil was 9.4 cm cm−3 and roots reached a depth of at least 1.8 m. Maximum nutrient uptakes were 364 kg ha−1 for N, 43 kg ha−1 for P, 308 kg ha−1 for K, 287 kg ha−1 for Ca and 16 kg ha−1 for Mg. A 30-day drought coincided with the flowering period and root and shoot growth, as well as nutrient uptake rates, were reduced. Nutrient concentrations in the soil solution necessary to sustain the nutrient fluxes into the root system by diffusive supply have been calculated. Peak values were in the range 10 μM for P to 87 μM for N, lower than the observed concentrations, and it was concluded that nutrient transport to roots was not a limitation to uptake by this rape crop.  相似文献

A field experiment was conducted for 2 years on an alkaline calcareous seleniferous soil to study the effect of different levels of gypsum (0.2 – 3.2 t ha−1) applied to wheat only in the first year on Se accumulation by wheat (Triticum aestivum L.) – rice (Oryza sativa L.) cropping sequence. With gypsum application, grain yield of both rice and wheat crops increased by 0.4 – 0.5 t ha−1; the increase in straw yield was 0.4 – 1.1 t ha−1. Significant reduction in Se accumulation by wheat was observed with gypsum application up to 0.8 t ha−1 and its residual effect was evident on the following crops for 2 years. Reduction in Se accumulation varied from 53 to 64% in wheat grain, 46 to 49% in wheat straw, 35 to 63% in rice grain and 36 to 51% in rice straw with an application of gypsum at 0.8 t ha−1. A corresponding increase in S concentration was observed. In the gypsum-treated plots, the ratio of S:Se increased by 6 – 8 times in wheat and 3 – 6 times in rice. Reduction in Se accumulation by crop plants through gypsum application may help in lowering the risk of Se over-exposure of animals and humans that depend on diet materials grown on high selenium soils. Experimental sites were established at two locations in north-eastern Victoria to define factors limiting the establishment and growth of Trifolium subterranean L. (subterranean clover). Liming the soil, seed inoculation and fungicide application were used in renovating subterranean clover pasture on two acidic soils (Longwood: brown/grey sandy loam DY 3.14 and Seymour: grey brown light clay DY 3.22, Northcote classification) with mean annual rainfall of 650 mm and 600 mm respectively. Soil acidity, low available soil phosphorus and plant disease were identified as factors limiting clover yield on these soils. Significant yield responses to lime (35–140%) were obtained with subterranean clover at both sites, with corresponding decreases in Al in the 0–10 cm soil horizon. Liming the soil, when combined with seed inoculation, increased the number and effectiveness of root nodules at both sites. Soil P available for plant growth was low at both sites (6.1 and 8.4 μg g−1) resulting in sub-optimal P concentrations in the clover herbage (45 mmol kg−1 at Longwood). Levels of root disease were low but Aphanomyces euteiches and Phytophthora clandestina (causal agents of lateral and tap root rot) were detected frequently on roots. Application of fungicide resulted in higher dry matter yields (p=0.05) at both sites. An assessment of the relative contributions of these limiting factors and the benefits to be obtained from better management would provide a clearer picture of the profitability and sustainability of this farming system.  相似文献

In the present study, it was aimed to investigate the influence of exogenous mammalian sex hormones (MSH) (progesterone, β-estradiol and androsterone) on the morphological (root and shoot growth) and biochemical parameters (protein and sugar content, antioxidant enzyme activities, and lipid peroxidation and H2O2 levels) of chickpea (Cicer arietinum L.) plants growing under control conditions. The solutions of hormones prepared at different concentrations (10−4, 10−6, 10−9, 10−12 and 10−15 M) were sprayed once on the leaves of 7-day plants. The plants were harvested on 18 days after the hormone treatment. Although all of the hormones at the tested concentrations significantly increased plant growth, soluble protein and sugar contents, and antioxidant enzyme activities [superoxide dismutase (SOD), peroxidase (POX) and catalase (CAT)], they decreased H2O2 content and lipid peroxidation level when compared with control plants. The activities of SOD, POX and CAT reached to the highest levels at 10−6 M for progesterone, and 10−9 M for β-estradiol and androsterone, which maximum growth, protein and sugar contents were determined. The same concentrations also resulted in the lowest levels for H2O2 content and lipid peroxidation. It can be interpreted that the MSH improve plant growth and development by affecting some biochemical parameters including antioxidative system.  相似文献
