Two gamma- and UV-radiation resistant, Gram-positive, red- or pink-pigmented, rod-shaped, strictly aerobic, oxidase- and catalase-positive bacterial strains, TDMA-25T and TDMA-uv51T, were isolated from fresh water collected at Misasa, a radioactive site in Japan. Phylogenetic analysis based on 16S rRNA gene sequences placed both in a distinct lineage in the family Deinococcaceae, and the highest degrees of sequence similarity determined belonged to Deinococcus maricopensis LB-34T (88.8–89.3%), Deinococcus pimensis KR-235T (86.4–86.7%) and Deinococcus yavapaiensis KR-236T (86.1%). The DNA G+C content of the strains was 53–58 mol%. The major respiratory quinone was MK-8. The predominant fatty acids were C15:0 iso, C16:0 iso, C13:0 iso, C17:0 iso, C16:0, C13:0 anteiso, C15:0 and C12:0 iso. The strains degraded gelatin, casein, starch and Tween 80. Unique physiological characteristics, differences in their fatty acid profiles, and genotypic and phylogenetic features, differentiated strains TDMA-25T and TDMA-uv51T from closely related Deinococcus species. Hence, the two strains are described as novel species of the genus Deinococcus. The names Deinococcus misasensis sp. nov. (type strain TDMA-25T=JCM 14369=NBRC 102116=CCUG 53610) and Deinococcus roseus sp. nov. (type strain TDMA-uv51T=JCM 14370=NBRC 102117=CCUG 53611) are proposed.  相似文献

Two strains designated strains L-1T and L-9T were isolated from activated sludge of a treatment plant that receives wastewater from the tannery industry contaminated with chromium. Phylogenetic analysis showed that the organisms represented two new species of the genus Leucobacter. Strains L-1T and L-9T could be distinguished from the type strain of L. komagatae and from the type strain of “L. albus” by the B-type peptidoglycan composition, fatty acid composition, several phenotypic and physiological characteristics. The major fatty acids of the organisms were iso- and anteiso-branched C15:0 and C17:0, straight-chain C16:0 was also found in relatively high proportions. The organisms were halotolerant, grew in medium containing 9% NaCl, and all strains, including the type strain of L. komagatae grew in medium containing 5 mM Cr(VI). On the basis of the distinct peptidoglycan composition, 16S ribosomal DNA sequence analysis, percentage of DNA-DNA reassociation values, and phenotypic characteristics we are of the opinion that strain L-1T represents a new species of the genus Leucobacter for which we propose the name Leucobacter chromiireducens and that strain L-9T represents an additional new species of the same genus for which we propose the name Leucobacter aridicollis.  相似文献

Mycobacterium strain FA4T was isolated with fluoranthene as the single carbon source from soil of a former coal gas plant, polluted with polycyclic aromatic hydrocarbons. The physiological properties, fatty acid pattern, and the 16S ribosomal RNA gene sequence indicated membership to the genus Mycobacterium, but were different from all type strains of Mycobacterium species. Based on comparative 16S rRNA gene sequence analyses strain FA4T could be assigned to the Mycobacterium neoaurum taxon showing 98% sequence similarity to M. diernhoferi as its closest neighbour. The occurrence of epoxymycolate in the cell wall differentiates FA4 from all members of this taxon which synthesize wax-ester mycolates in addition to alpha-mycolates. Strain FA4T is able to degrade aflatoxin B1. This biological attribute might be useful in biological detoxification processes of foods and feeds. From the investigated characteristics it is concluded that strain FA4T represents a new species, for which we propose the name Mycobacterium fluoranthenivorans sp. nov. Nine psychrotolerant bacteria were isolated from fast ice in the middle of Geologie Archipelago, Adelie Land, Antarctica and were categorized into two groups, based on their SDS-PAGE profiles. Representatives from each of the two groups, namely strains DD 48T and SJ 14T exhibited phenotypic and chemotaxonomic characteristics confirming to the genus Psychrobacter. The 16S rRNA gene sequence indicated that the two isolates are closely related to each other and to the already reported fifteen species of Psychrobacter. Detailed studies on the phenotypic characteristics, chemotaxonomic properties and phylogenetic analysis of strains DD 48T and SJ 14T indicated that they are distinctly different from each other and the reported species of Psychrobacter. At the DNA-DNA hybridisation level, the two species exhibit less than 70% similarity. Thus, strains DD 48T and SJ 14T are identified as new species of the genus Psychrobacter for which the names Psychrobacter salsus sp. nov. and Psychrobacter adeliensis sp. nov. respectively are proposed.  相似文献

An actinomycete, strain BS2T, was isolated from a sand sample collected from an estuary in Plettenberg Bay, Western Cape Province, South Africa. Based on 16S rRNA gene sequence similarities and chemotaxonomy, strain BS2T was identified as a member of the genus Gordonia. It exhibited weak antibiosis against Mycobacterium aurum A+ and Mycobacterium smegmatis LR222. Phenotypic and phylogenetic results allowed for the differentiation of strain BS2T from other species within the genus Gordonia. DNA–DNA hybridization further differentiated strain BS2T from its nearest phylogenetic neighbour, Gordonia terrae NRRL B-16283T (57.5±4.4% DNA relatedness). Strain BS2T is therefore a novel species within the genus Gordonia, for which the name Gordonia lacunae is proposed, with the type strain being BS2T (=DSM 45085T=NRRL B-24551T).  相似文献

A Gram-negative, strictly aerobic, diplococcoid bacterium (strain D2-3T) was isolated from the biofilter of a recirculating marine aquaculture system. Phylogenetic analysis of the 16S rRNA gene sequence of D2-3T indicated that the new organism occupied a novel lineage within the -1 subclass of Proteobacteria and was related to the genera Rhodothalassium, Azospirillum, Craurococcus, Acidiphilium, and Tistrella. The highest sequence similarity (90.8%) of the 16S rRNA gene sequence of D2-3T was to that of Candidatus “Alysiosphaera europaea”. D2-3T was mesophilic, heterotrophic, required sea salt, and had a pH optimum of 8.0. Growth in the presence of light resulted in the formation of pink colonies, a 25% increased cell yield, and a slightly increased growth rate. D2-3T contained carotenoids and low amounts of bacteriochlorophyll a. Membranes of D2-3T contained b-type cytochromes. The G+C content of the DNA was 60.3±0.1 mol%. Phylogenetic, morphological, physiological, and biochemical analyses demonstrated that D2-3T represented a new aerobic phototrophic genus, for which the name Geminicoccus roseus gen. nov., sp. nov. is proposed for the type species (D2-3T=DSM 18922T=ATCC BAA-1445T).  相似文献

A Gram-positive strain designated as MSL-14T isolated from a soil sample collected from Bigeum Island, Korea, was subjected to polyphasic taxonomy. The isolate was strictly aerobic. Cells were short rods and motile. Optimum growth temperature and pH was 28 degrees and 7.0, respectively. It was characterized chemotaxonomically as having a cell-wall peptidoglycan type based on LL-2,6-diaminopimelic acid and MK-8(H4) as the predominant menaquinone. The major fatty acids were iso-C16:0, C17:1 omega8c, and C18:1 omega9c. The G+C content was 67.6 mol%. Phylogenetic analysis based on the 16S rRNA gene sequence revealed that strain MSL-14T is affiliated to the genus Nocardioides and formed a distinct lineage within the genus. MSL-14T showed highest sequence similarity to Nocardioides aestuarii JCM 12125T, having a similarity of 96.5%. Based on the 16S rRNA gene sequence divergence and phenotypic characteristics, it is proposed that strain MSL-14T should be classified as representing a novel member of the genus Nocardioides, for which we propose the name Nocardioides tritolerans sp. nov. In a project aiming to isolate strains with the ability to produce nonribosomal peptides, a Gram-negative, endospore-forming, rod-shaped strain, designated B5T, was isolated from a soil sample collected from Tianmu Mountain national natural reserve in Hangzhou, China. Strain B5T contained meso -diaminopimelic acid in the cell wall peptidoglycan. The major cellular fatty acids were anteiso-C15:0 and iso-C15:0. The DNA G+C content was 42.5 mol%. The phylogenetic analysis based on 16S rRNA gene sequence indicated that strain B5T fell within the genus Bacillus , with highest sequence similarity values to Bacillus barbaricus DSM 14730T (96.4%) and Bacillus macauensis JCM 13285T (95.5%). The isolate, however, could be distinguished from Bacillus strains with validly published names by low 16S rRNA gene sequence similarity values, distinct phenotypic and chemotaxonomic characteristics. On the basis of these polyphasic evidences, it is demonstrated that the isolate B5T represents a novel species of the genus Bacillus , for which the name Bacillus tianmuensis sp. nov. is proposed. Three strains of sulfate-reducing bacteria (ADR21, ADR26 and ADR28) were isolated from Adour estuary sediments (French South Atlantic coast). Cells of these isolates were rod-shaped, motile and stained Gram-negative. The 16S rRNA and dsrAB genes sequence analyses indicated that these three strains belonged to the genus Desulfomicrobium within the delta Proteobacteria, with Desulfomicrobium escambiense strain DSM10707T as their closest relative. According to phenotypic characteristics, strains ADR21 and ADR28 could be considered as members of the same species. The relatedness values, based on DNA–DNA hybridization studies, between strains ADR21/DSM10707T, ADR26/DSM10707T and ADR21/ADR26 ranged between 30.6–40.8%, 45.2–43.0% and 19.0–26.4%, respectively. Strains ADR21 and ADR28 grew well on lactate, fumarate, malate, formate, ethanol and H2/acetate in the presence of sulfate as an electron acceptor. Thiosulfate, nitrate, fumarate and DMSO were alternative electron acceptors. Malate was well fermented but pyruvate and fumarate only poorly. Strain ADR26 could not grow on ethanol or fumarate and was unable to use DMSO or fumarate as electron acceptors. The three new strains exhibited differences compared to the type strain of D. escambiense, such as temperature optima, substrate utilization and mercury methylation capacities. On the basis of both genetic and phenotypic evidences, strain ADR21 is proposed as the type strain of the species Desulfomicrobium salsuginis sp. nov., and strain ADR26 as the type strain of the species Desulfomicrobium aestuarii sp. nov.  相似文献

Strain AC1100 is well-known for its ability to degrade a variety of recalcitrant xenobiotics, including 2,4,5-trichlorophenoxyacetic acid. We performed a polyphasic-taxonomic study to determine its taxonomic position. The G+C content of strain AC1100 was 62.6 mol%. On the basis of 16S rRNA gene sequence similarity, strain AC1100 belonged to the b-Proteobacteria and was most closely related to Burkholderia fungorum (98.3% similarity). DNA-DNA hybridisations, comparison of protein profiles, cellular fatty acid analysis and biochemical tests allowed genotypic and phenotypic differentiation of strain AC1100 from other Burkholderia species. Our data show that strain AC1100 represents a novel species for which the name Burkholderia phenoliruptrix sp. nov. is proposed. The type strain is AC1100T (= LMG 22037T = CCUG 48558T).  相似文献

A short coccoid-rod-shaped, nonmotile actinobacteria strain MSL-11T was isolated from soil in Bigeum Island, Korea. A polyphasic study was undertaken to establish the taxonomic position of this strain. Phylogenetic analysis based on the 16S rRNA gene sequence revealed that strain MSL-11T forms an evolutionary lineage within the radiation of the genus Nocardioides. The cell wall peptidoglycan of strain MSL-11T contained ll-diaminopimelic acid, indicating wall chemotype I. The predominant menaquinone was MK-8(H4). Strain MSL-11T had a cellular fatty acid profile containing straight-chain, branched, unsaturated, and 10-methyl fatty acids, with iso-C16:0 as a major fatty acid component detected. The DNA G + C content of the strain was 71.8 mol%. Comparative 16S rRNA gene sequencing revealed that the strains constituted a distinct subclade within the genus Nocardioides, displaying a 16S rRNA gene sequence similarity of about 95.68% with Nocardioides jensenii DSM 20641T. On the basis of both phenotypic and phylogenetic evidence, the strain is separated from previously described Nocardioides species and should be assigned to represent a novel species of the genus Nocardioides, for which the name Nocardioides dilutes sp. nov. is proposed. The aerobic degradation of phenol, chlorobenzene and dichlorobenzene as a sole carbon source has been observed in bacterial Gram-positive strain G2PT isolated from a wastewater bioprocessor. Cells display branching mycelia fragmenting into rod and coccoid elements when grown on TSA. Aerial hyphae formation occurs when grown on phenol and chlorinated aromatics as the sole carbon source. Growth was observed at up to 0.75% phenol as a sole carbon source, indicating a strong tolerance for the compound. The 16S rRNA gene sequence shares the greatest similarity with members of the Rhodococcus genus, with the closest shared nucleotide identity of 98% with the aromatic toxin degrading bacteria Rhodococcus zopfii DSM 44108T. Neighbor-joining and parsimony analysis of Corynebacterineae 16S rRNA gene sequences consistently places strain G2PT in a clade shared with R. zopfii within the Rhodococcus rhodochrous subclade. Based on a unique polyphasic profile involving phenotypic, ribosomal DNA sequence analysis, DNA–DNA hybridization, mol% DNA G+C content and fatty acid composition, G2PT is proposed to represent a previously uncharacterized, novel species in the genus Rhodococcus. The name Rhodococcus phenolicus is proposed for the isolate with the type strain G2PT (=DSM 44812) (=NRL B-24343).  相似文献

Two novel strains of the Cytophaga-Flexibacter-Bacteroides (CFB) group, designated Gsoil 219" and Gsoil 2381, were isolated from soil of a ginseng field of Pocheon Province in Korea. Both strains were Gram-negative, aerobic, nonmotile, nonspore-forming, and rod-shaped. Phylogenetic analysis based on 16S rRNA gene sequences indicated that both isolates belong to the genus Chitinophaga but were clearly separated from established species of this genus. The sequence similarities between strain Gsoil 219T and type strains of the established species and between strain Gsoil 238T and type strains of the established species ranged from 91.4 to 94.7% and 91.6 to 94.2%, respectively. Phenotypic and chemotaxonomic data (major menaquinone, MK-7; major fatty acids, iso-C15:0 and C(16:1) omega5c; major hydroxy fatty acid, iso-C(17:0) 3-OH; major polyamine, homospermidine) supported the affiliation of both strains Gsoil 219T and Gsoil 238T to the genus Chitinophaga. Furthermore, the results of physiological and biochemical tests allowed genotypic and phenotypic differentiation of both strains from the other validated Chitinophaga species. Therefore, the two isolates represent two novel species, for which the name Chitinophaga soli sp. nov. (type strain, Gsoil 219T=KCTC 12650T=DSM 18093T) and Chitinophaga terrae sp. nov. (type strain, Gsoil 238T=KCTC 12651T=DSM 18078T) are proposed.  相似文献

A new thermophilic sulphate-reducing bacterium (strain Hbr5T) was enriched and isolated from a terrestrial Tunisian hot spring. It was a non-spore-forming Gram-negative curved or vibrio-shaped bacterium. It appeared singly or in long chains and was actively motile by a polar flagellum. It possessed c-type cytochromes and desulfofuscidin. Growth occurred between 50 and 70 °C, with an optimum of 65 °C at pH 7.1. In the presence of sulphate as a terminal electron acceptor, this strain readily used H2 but formate only poorly. It could use sulphate, thiosulphate, sulphite or arsenate as electron acceptors. Its DNA G+C content was 36.1 mol%. Based on phenotypic, genomic, and phylogenetic characteristics, strain Hbr5T (=DSM 18151T, =JCM 13991T) is proposed to be assigned to a novel species of genus Thermodesulfovibrio, T. hydrogeniphilus sp. nov.  相似文献

A novel actinobacterium designated as MSL-26T was isolated from soil in Bigeum Island Korea. A polyphasic study was undertaken to establish the taxonomic position of isolate MSL-26T. Strain MSL-26T was found to have chemical and morphological characteristics similar to Nocardioides. The strain grew optimally at pH 7·5 and 28°C. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain MSL-26T forms a distinct line of descent within the radiation enclosed by the genus Nocardioides. The cell wall of strain MSL-26T contained LL-2, 6-diaminopimelic acid. The principal menaquinone was MK-8 (H4). The phospholipids detected were diphosphatidylglycerol, phosphatidylglycerol and some unidentified lipids. C18:1 w7c (50.38%) was the major fatty acid. The DNA G + C content of strain MSL-26T was 71.4 mol%. The 16S rRNA gene sequence of strain MSL-26T shares the highest sequence similarity with Nocardioides kribbensis KCTC 19038T (95.78%) and Nocardioides aquaticus DSM 11439T (95.52%). Based on the morphological, physiological, biochemical and chemotaxonomical data presented in this study, strain MSL-26T should be classified as a novel species, for which the name Nocardioides islandiensis sp. nov. is proposed. The taxonomic positions of five Gram-negative, non-spore-forming and non-motile bacterial strains isolated from the rhizosphere of sand dune plants were examined using a polyphasic approach. The analysis of the 16S rRNA gene sequence indicated that all of the isolates fell into four distinct phylogenetic clusters belonging to the genus Chryseobacterium of the family Flavobacteriaceae. The 16S rRNA gene sequence similarities of isolates to mostly related type strains of Chryseobacterium ranged from 97.5% to 98.5%. All strains contained MK-6 as the predominant menaquinone, and iso-C15:0, iso-C17:0 3-OH and a summed feature of iso-C15:0 2-OH and/or C16:1 ω7c as the dominant fatty acids. Combined phenotypic, genotypic and chemotaxonomic data supported that they represented four novel species in the genus Chryseobacterium, for which the names Chryseobacterium hagamense sp. nov. (type strain RHA2-9T=KCTC 22545T=NBRC 105253T), Chryseobacterium elymi sp. nov. (type strain RHA3-1T=KCTC 22547T=NBRC 105251T), Chryseobacterium lathyri sp. nov. (type strain RBA2-6T=KCTC 22544T=NBRC 105250T), and Chryseobacterium rhizosphaerae sp. nov. (type strain RSB3-1T=KCTC 22548T=NBRC 105248T) are proposed.  相似文献

A novel Gram-positive bacterium, designated SYB2T, was isolated from wastewater reservoir sediment, and a polyphasic taxonomic study was conducted based on its morphological, physiological, and biochemical features, as well as the analysis of its 16S rRNA gene sequence. During the phylogenetic analysis of the strain SYB2T, results of a 16S rRNA gene sequence analysis placed this bacterium in the genus Arthrobacter within the family Micrococcaceae. SYB2T and Arthrobacter protophormiae ATCC 19271T, the most closely related species, both exhibited a 16S rRNA gene sequence similarity of 98.99%. The genomic DNA G+C content of the novel strain was found to be 62.0 mol%. The predominant fatty acid composition was anteiso-C15:0, anteiso-C17:0, iso-C16:0, and iso-C15:0. Analysis of 16S rRNA gene sequences and DNA-DNA relatedness, as well as physiological and biochemical tests, showed genotypic and phenotypic differences between strain SYB2T and other Arthrobacter species. The type strain of the novel species was identified as SYB2T (= KCTC 19291T= DSM 19449T).  相似文献

Two Gram-negative, nonmotile, coccobacilli, SW-3T and SW-100T, were isolated from sea water of the Yellow Sea in Korea. Strains SW-3T and SW-100T contained ubiquinone-9 (Q-9) as the predominant respiratory lipoquinone and C18:1 omega9c and C16:0 as the major fatty acids. The DNA G+C contents of strains SW-3T and SW-100T were 44.1 mol% and 41.9 mol%, respectively. A neighbor-joining tree based on 16S rRNA gene sequences showed that the two isolates fell within the evolutionary radiation enclosed by the genus Acinetobacter. Strains SW-3T and SW-100T exhibited a 16S rRNA gene similarity value of 95.7% and a mean DNA-DNA relatedness level of 9.2%. Strain SW-3T exhibited 16S rRNA gene sequence similarity levels of 93.5-96.9% to the validly described Acinetobacter species and fifteen Acinetobacter genomic species. Strain SW-100T exhibited 16S rRNA gene sequence similarity levels of less than 97.0% to the other Acinetobacter species except Acinetobacter towneri DSM 14962T (98.0% similarity). Strains SW-3T and SW-100T exhibited mean levels of DNA-DNA relatedness of 7.3-16.7% to the type strains of some phylogenetically related Acinetobacter species. On the basis of phenotypic, phylogenetic, and genetic data, strains SW-3T and SW-100T were classified in the genus Acinetobacter as two distinct novel species, for which the names Acinetobacter marinus sp. nov. (type strain SW-3T=KCTC 12259T=DSM 16312T) and Acinetobacter seohaensis sp. nov. (type strain SW-100T=KCTC 12260T=DSM 16313T) are proposed, respectively.  相似文献

A novel pink-coloured, non-spore-forming, non-motile, Gram-negative bacterium, designated YIM 48858T, is described by using a polyphasic approach. The strain can grow at pH 6.5–9 (optimum at pH 7) and 25–30°C (optimum at 28°C). NaCl is not required for its growth. Positive for oxidase and catalase. Urease activity, nitrate reduction, starch and Tween 80 tests are negative reaction. 16S rRNA gene sequence similarity studies showed that strain YIM 48858T is a member of the genus Rubellimicrobium, with similarities of 96.3, 95.7 and 95.5% to Rubellimicrobium mesophilum MSL-20T, Rubellimicrobium aerolatum 5715S-9T and Rubellimicrobium thermophilum DSM 16684T, respectively. Q-10 was the predominant respiratory ubiquinone as in the other members of the genus Rubellimicrobium. The major polar lipids were diphosphatidylglycerol, phosphatidylcholine, phosphoglycolipid, glycolipid and the major fatty acids were C18:1 ω7c, C16:0 and C10:0 3-OH, which are very different from the valid published species. The DNA G + C content was 67.7 mol%. Both phylogenetic and chemotaxonomic evidence supports that YIM 48858T is a novel species of the genus Rubellimicrobium, for which the name Rubellimicrobium roseum sp. nov. is proposed. The type strain is YIM 48858T (=CCTCC AA 208029T =KCTC 23202T).  相似文献
