Lucilia sericata Meigen (Diptera: Calliphoridae) larvae are manufactured worldwide for the treatment of chronic wounds. Published research has confirmed that the primary clinical effect of the product, debridement (the degradation of non‐viable wound tissue), is accomplished by a range of enzymes released by larvae during feeding. The quality assessment of larval activity is currently achieved during production using meat‐based assays, which monitor insect growth and/or the reduction in substrate mass. To support this, the present authors developed a complementary radial diffusion enzymatic assay to produce a visual and measureable indication of the activity of larval alimentary products (LAP) collected under standardized conditions, against a gelatin substrate. Using basic laboratory equipment and reagents, the assay is rapid and suited to high throughput. Assay reproducibility is high (standard deviation: 0.06–0.27; coefficient of variation: 0.75–4.31%) and the LAP collection procedure does not adversely affect larval survival (mortality: < 2%). Because it is both cost‐ and time‐effective, this method is suited to both academic and industrial use and supports good manufacturing and laboratory practice as a quality control assay.  相似文献

Development of embryos and larvae in Ancherythroculter nigrocauda Yih et Woo (1964) and effects of delayed first feeding on larvae were observed after artificial fertilization. The fertilized eggs were incubated at an average temperature of 26.5°C (range: 25.7–27) and the larvae reared at temperatures ranging from 21.8 to 28°C. First cleavage was at 50 min, epiboly began at 7 h 5 min, heartbeat reached 72 per min at 24 h 40 min and hatching occurred at 43 h 15 min after insemination. Mean total length of newly hatched larvae was 4.04 ± 0.03 mm (n = 15). A one‐chambered gas bladder was observed at 70 h 50 min, two chambers occurred at 15 days, and scales appeared approximately 30 days after hatching. Larvae began to feed exogenously at day 4 post‐hatch at an average temperature of 24°C. Food deprivation resulted in a progressive atrophy of skeletal muscle fibres, deterioration of the larval digestive system and cessation of organ differentiation. Larval growth under food deprivation was significantly affected by the time of first exogenous feeding. Starved larvae began to shrink, with negative growth from day 6 post‐hatch. The point of no return (PNR) was reached at day 11 after hatching. Mortality of starved larvae increased sharply from day 12 after hatching.  相似文献

The sugarbeet root maggot Tetanops myopaeformis Röder (Diptera: Ulidiidae) can be stored in moist sand at 4–6 °C for up to 5 years and is freeze‐tolerant. The majority of stored larvae survive in a state of post‐diapause quiescence and the remainder are in a multi‐year diapause. The present study aims to determine larval water content and water loss rates in diapausing and low‐temperature stored larvae. Body water content ranges from 57% to 70.1%. Two distinct groupings of larvae are revealed based on dry weights. The first group consists of the diapausing larvae and larvae stored for 1 year. This group has significantly higher dry weights than the second grouping, which consists of the larvae stored for 2 and 3 years. There are no significant differences within each group. Larval water losses follow a first‐order kinetic relationship with time. Larvae stored for 2 years lose water at a significantly higher rate than diapausing larvae. Larvae exhibit no active water uptake at storage temperatures. A freezing event does not induce a significant decrease in wet weights, nor does it increase larval water loss rates. These results indicate that metabolic water and the microclimate during storage are key factors enabling the long‐term survival of T. myopaeformis larvae during low‐temperature storage, and may provide insights for maintaining other insect species under similar conditions.  相似文献

Prior to anthropogenic modifications, the historic Missouri River provided ecological conditions suitable for reproduction, growth, and survival of pallid sturgeon Scaphirhynchus albus. However, little information is available to discern whether altered conditions in the contemporary Missouri River are suitable for feeding, growth and survival of endangered pallid sturgeon during the early life stages. In 2004 and 2007, nearly 600 000 pallid sturgeon free embryos and larvae were released in the upper Missouri River and survivors from these releases were collected during 2004–2010 to quantify natural growth rates and diet composition. Based on genetic analysis and known‐age at release (1–17 days post‐hatch, dph), age at capture (dph, years) could be determined for each survivor. Totals of 23 and 28 survivors from the 2004 and 2007 releases, respectively, were sampled. Growth of pallid sturgeon was rapid (1.91 mm day?1) during the initial 13–48 dph, then slowed as fish approached maximum length (120–140 mm) towards the end of the first growing season. The diet of young‐of‐year pallid sturgeon was comprised of Diptera larvae, Diptera pupae, and Ephemeroptera nymphs. Growth of pallid sturgeon from ages 1–6 years was about 48.0 mm year?1. This study provides the first assessment of natural growth and diet of young pallid sturgeon in the wild. Results depict pallid sturgeon growth trajectories that may be expected for naturally produced wild stocks under contemporary habitat conditions in the Missouri River and Yellowstone River.  相似文献

The effects of delayed first feeding on growth and survival of spotted mandarin fish Siniperca scherzeri larvae were examined under controlled conditions. Morphometric characters [yolk‐sac volume, oil globule volume, head depth (HD), body depth (BD), eye diameter (ED), musculature height (MH), mouth diameter (MD) and total length (LT)], body mass (M), specific growth rate (SGR) and survival were evaluated under different first‐feeding time (2, 3, 4 and 5 days after hatching). Larvae began to feed exogenously at 2 days after hatching (DAH) and the point of no return (PNR) occurred between 5 and 6 DAH at 23° C, range ±1·0° C. The yolk volume of larvae first‐fed at 2 days had a significant difference compared with that of larvae first‐fed at 3, 4 and 5 days on 3 and 4 DAH. The larvae first‐fed at 2 days achieved comparatively better growth performance than that of 3, 4 and 5 days. On 5 DAH, all morphometric characters had significant differences between 2 and 5 days and 2 and 4 days initial feeding, respectively. Total mortality was recorded on 9 DAH for the larvae first‐fed at 5 days. On 12 DAH, significant differences were observed between 2 and 4 days and 3 and 4 days initial feeding for all morphometric characters. From 16 DAH to the end of experiment, all growth variables of the larvae first‐fed at 2 days were significantly higher than those in other treatments. The SGR (2–9 DAH) first‐fed at 2 and 3 days were significantly higher than 4 and 5 day treatments, and the SGR (9–16 DAH) first‐fed at 2 days was significantly higher than 3 and 4 day treatments. There was no significant difference, however, of SGR (16–28 DAH) among treatments. Survival rate was significantly higher at 2 days initial feeding (27·42%) when compared with 3 (15·96%) and 4 days (7·92%) initial feeding at the end of experiment. The present study suggests that the first feeding of S. scherzeri larvae should be initiated at 2 days after hatching for achieving good growth and survival.  相似文献

The growth of river catfish Mystus nemurus (Cuvier & Valenciennes) larvae fed four isocaloric diets (4200 kcal kg?1) with different protein levels during weaning was determined. Diets containing 45, 50, 55, and 60% protein were formulated by linear programming using amino acid profiles based on that of 2‐day‐old river catfish larvae. Artificial diets were fed to the larvae beginning at day 5 after being initially fed Artemia nauplii for 4 days. The larvae thrived solely on artificial diets from day 8 to day 16. On the other hand, the control larvae were fed Artemia nauplii from day 1 to day 16. Results of the feeding trial showed that growth and survival of M. nemurus larvae given the diet containing 60% protein were high and comparable to those of the larvae given only live food (control). Larvae fed the 55% protein diet had significantly lower growth and survival than the larvae on the control and 60% diets but significantly higher growth and survival rates than did larvae fed with 45 and 50% protein diets. Carcass moisture and total lipids after 16 days of feeding did not differ significantly (P > 0.05), but body protein increased with increasing dietary protein. Body protein of the control larvae was similar to that of larvae given the 60% protein diet.  相似文献

This study evaluated factors that influence the regurgitation behaviour of sixth instar spruce budworm, Choristoneura fumiferana Clemens (Lepidoptera: Tortricidae), reared on balsam fir, Abies balsamea (L.) Mill. (Pinaceae), under various experimental conditions in the laboratory. Upon physical disturbance, larvae discharged a median volume of regurgitant of 0.4 μl when fed and 1.6 μl when food‐deprived. Larvae deprived of food for 24 or 48 h disgorged more regurgitant than larvae feeding on balsam fir foliage, and the effect was consistent for laboratory‐reared and field‐collected larvae. The water content of the foliage fed upon by larvae had no immediate impact on the volume of regurgitant; following a 24‐h period of food deprivation, however, larvae that previously fed on fresh foliage discharged >2.5 times more regurgitant than larvae that previously fed on dry foliage. Self‐regulated regurgitation by larvae, measured using the number of regurgitant stains on filter paper, was >10 times higher when larvae had access to balsam fir foliage than when they were starved. The number of larvae confined inside the Petri dish (one or four individuals) had a relatively small effect on regurgitation. Larvae were deterred from feeding when balsam fir needles were entirely covered with regurgitant, but not when only a portion of the foliage was treated. These results suggest that the regurgitant does not serve as resource marking or spacing pheromone. The high level of regurgitation by larvae after contact with ants suggests that the regurgitant has evolved in part as a defence mechanism against natural enemies.  相似文献

This study aimed to elucidate the causes of variability in larval survival and juvenile abundance (recruitment) within and among cohorts of Japanese sea bass (JSB; Lateolabrax japonicus), a winter‐spawning temperate coastal marine fish. Larvae and settled individuals (settlers) belonging to four cohorts were collected from Tango Bay (the Sea of Japan coast) during eight sampling cruises in 2007 and 2008. Larvae were sampled in January and February each year using an ichthyoplankton net, and settlers were collected in February and March each year using a beam trawl. Age of individual larva and settlers was determined and growth history was back‐calculated from otolith microstructure, and the hatch date distribution was computed. Temperature, daily growth rate, size‐at‐age, hatch date, and density data of larvae and settlers allowed elucidating the effects of the timing of spawning and larval quantity and quality (growth rate and body size) on larval survival and recruitment within and among cohorts of JSB. Results showed that cohorts that hatched earlier in the season had higher quantity of larvae, experienced higher mean temperatures and survived better than cohorts hatched later. Recruitment variability among cohorts is determined largely by the initial quantity of larvae, as this explained >97% of the variability in recruitment among cohorts. Within cohorts, larger hatched larvae grew faster than their smaller conspecifics, and the bigger and faster growing larvae survived and settled. Results from this study suggest the following scenarios for recruitment of JSB: (i) earlier spawning in the season promotes larval survival since earlier cohorts are likely to encounter a better temperature and perhaps food conditions, and therefore recruit better than later cohorts; (ii) the initial quantity of larvae appears to be an important determinant of recruitment variability among cohorts; and (iii) the size‐ and growth‐related mechanisms operating during the larval phase appear to start at the time of the hatch.  相似文献

1. Larvae of the smooth newt, Triturus vulgaris, were exposed to four concentrations of ammonium nitrate in artificial pond water (50, 100, 200 and 500 mg 1-?1) under controlled laboratory conditions. 2. Larvae exposed to a 50 mg 1-?1 solution of ammonium nitrate showed no significant difference to control larvae in feeding rate, mass at metamorphosis or time to metamorphosis. However, larvae exposed to 200 or 500 mg 1-?1 ammonium nitrate, for a single period of 24, 48 or 72 h, were significantly smaller than controls at metamorphosis. Larvae exposed to 100 mg 1-?1 had a significantly higher feeding rate than those reared under control conditions or those exposed to 200 or 500 mg 1-?1 ammonium nitrate, but this was not reflected in their size at metamorphosis, which was not significantly different from the controls. Larval survival remained high in all trials. 3. In separate trials, Daphnia, used as a food source for the newt larvae, were exposed to the same series of ammonium nitrate concentrations. Daphnia behaviour was affected by exposure to 200 and 500 mg 1-?1 ammonium nitrate, and in both cases animals were significantly more likely to be found at the top of the water column than those exposed to 50 or 100 mg 1-?1 ammonium nitrate or the control medium.  相似文献

This study investigated the effect of strong magnetic fields as insecticidal activity on Ephestia kuehniella (Zeller) (Lepidoptera: Pyralidae) larvae and eggs at different stages of development and their preference by the egg parasitoid, Trichogramma embryophagum Hartig (Hymenoptera: Trichogrammatidae). Eggs ranging in age from 24-h to 48-h and 72-h-old and larvae (1 to 2 days) were exposed to 1.4 Tesla (T) magnetic fields from a DC power supply at 50 Hz for different time periods (3, 6, 12, 24, 48 and 72 h). Twelve hours of exposure at 1.4 T was toxic to 24-h-old eggs of E. kuehniella. The 72-h-old host eggs treated with 1.4 T for 6–72 h were not significantly preferred by T. embryophagum. The magnetic field was toxic to 24-h-old eggs of E. kuehniella exposed to 1.4 T for 12. The treatment of magnetic fields on the 72-h-old host egg with 1.4 T at 6–72 h was not significantly preferred by T. embryophagum. Magnetization of 24-h-old eggs of E. kuehniella for 3 h could be effectively used with T. embryophagum as sterilised host eggs. These eggs were markedly preferred by T. embryophagum. The LT50 and LT99 values of magnetic fields at different egg stages of E. kuehniella, and larvae were measured. A level of 1.4 T at 72 h completely prevented the development of the larvae. There was no significant effect on larval survival at 1.4 T at 48 and 72 h. Increasing magnetic fields exposure times for eggs that were 24-h, 48-h and 72-h-old prevented larval emergence and increased their mortality rate. Consequently, magnetic fields could be used in controlling stored-product pest eggs and larvae of E. kuehniella.  相似文献

Abstract. The tiger beetle, Phaeoxantha klugii Chaudoir, survives the annual inundation period in Central Amazonian floodplains as a third‐instar larvae submerged in the soil at approximately 29 °C for up to 3.5 months. Previous studies showed an exceptional anoxia resistance in these larvae and this study investigates whether they perform anaerobiosis. Larvae collected in the field were exposed to a pure nitrogen atmosphere for 0–9 days in the laboratory. The content of lactate, alanine, free sugars and glycogen is analysed in surviving larvae. Lactate and alanine contents rise during anoxia from around 1.5 and 7 to 6–14 and 15–22 µmol g?1 fresh mass, respectively, providing evidence for anaerobic metabolism. Both compounds show a steep increase during the first 12 h and a tendency to rise further with increasing duration of anoxic conditions, indicating a significant metabolic depression within the first day. Content of free sugars and glycogen varies greatly between individuals and ranges from 0.08–2.5 and 0.05–2.9 mg g?1 fresh mass, respectively. Whether glycogen is used as metabolic substrate for anaerobiosis could not be verified. The findings for free sugars indicate that larvae apparently maintain the ability to regulate the level of glucose and/or trehalose even after 9 days of anoxia.  相似文献

Aim: To (i) identify chronic wound bacteria and to test their ability to produce acyl‐homoserine‐lactones (AHLs) and autoinducer‐2 (AI‐2) cell–cell signalling molecules and (ii) determine whether chronic wound debridement samples might contain these molecules. Methods and Results: Partial 16S rRNA gene sequencing revealed the identity of 46 chronic wound strains belonging to nine genera. Using bio‐reporter assays, 69·6% of the chronic wound strains were inferred to produce AI‐2, while 19·6% were inferred to produce AHL molecules. At least one strain from every genus, except those belonging to the genera Acinetobacter and Pseudomonas, were indicated to produce AI‐2. Production of AI‐2 in batch cultures was growth‐phase dependent. Cross‐feeding assays demonstrated that AHLs were produced by Acinetobacter spp., Pseudomonas aeruginosa and Serratia marcescens. Independent from studies of the bacterial species isolated from wounds, AHL and/or AI‐2 signalling molecules were detected in 21 of 30 debridement samples of unknown microbial composition. Conclusion: Chronic wound bacteria produce cell–cell signalling molecules. Based on our findings, we hypothesize that resident species generally produce AI‐2 molecules, and aggressive transient species associated with chronic wounds typically produce AHLs. Both these classes of cell–cell signals are indicated to be present in human chronic wounds. Significance and Impact of the Study: Interbacterial cell–cell signalling may be an important factor influencing wound development and if this is the case, the presence of AHLs and AI‐2 could be used as a predictor of wound severity. Manipulation of cell–cell signalling may provide a novel strategy for improving wound healing.  相似文献

Maggot debridement therapy (MDT) refers to the use of blowfly larvae to clean or debride an infected wound. Most commonly, larvae of Lucilia sericata (Meigen) (Diptera: Calliphoridae) are used, and are sterilized prior to use to ensure no further bacterial infections are introduced during treatment. Current methods sterilize eggs from laboratory‐reared blowfly colonies, after which sterile early second instar maggots can be provided to hospitals for use in treatment. Maggots not required for treatment are used for colony regeneration, in which sterility is not maintained. The ability to maintain sterility beyond this would allow further research into fly–bacteria interactions and the effects of different bacteria on the blowfly lifecycle. This study aimed to produce a colony of sterile adults, using current egg sterilization practice, but maintaining sterility through to pupation and emergence. The production of a sterile colony allows further research into the impact of bacteria on fly development and survival. Eggs were placed on a sterile food source within autoclaved plant tissue culture containers to allow growth under sterile conditions. Nutrient agar plating of sterilized and non‐sterilized eggs, larvae and adults (post‐emergence), as well as the pupation medium and feed source in nutrient broth confirmed the aerobic sterility of all samples involved. The lifecycle of L. sericata was successfully completed through pupation to emergence with no effects on lifespan or oviposition by the newly emerged, sterile adult colony.  相似文献

Abstract

Larvae of Inopus rubriceps (Macquart) were reared from eggs originating from spring and autumn adult emergence periods (1975–78), under both insectary and field conditions. Rate of development appeared to depend largely on temperature. Autumn-hatched larvae showed little growth for the initial 6 months, but then grew rapidly until the end of summer. Spring-hatched larvae also grew rapidly over summer, but their development slowed over winter. Year-to-year differences were found in larval survival, weight gain, and rate of development, and in subsequent adult emergence. Pest status appeared to be related to the cumulative yearly heat budget. Overall, larval survival was higher and growth and development were more rapid in the insectary than under field conditions. A male-dominant sex ratio was demonstrated.  相似文献

In recent years, much progress has been made in the rearing of fish larvae fed only artificial diets. A preliminary study was made in an attempt to evaluate the effects of live food and formulated diets on survival, growth and body protein content of first‐feeding larvae of Plelteobagrus fulvidraco. Three test diets varying in protein level were formulated: Feed 1 containing 45% protein, Feed 2 with 50% protein and Feed 3 with 55% protein. Larvae fed live food (newly hatched Artemia, unenriched) were the control. The experiment started 3 days post‐hatch and lasted for 23 days. At the end of the 23‐day trial, survival was best in the control group (65.6%) whereby the final body weight and specific growth rate (SGR) were significantly lower than those in the test feed groups. At the same time, coefficients of variation for SGR and final body weight in the test groups were significantly higher than those in the control. Whole body protein content in all treatments showed a similar tendency during development: significantly higher 3 days post‐hatch, then decreasing significantly, and then increasing unstatistically 10 days post‐hatch. All results suggest that live food is still better for first‐feeding larvae of P. fulvidraco, since live food leads to healthier larvae growth.  相似文献

The present 20‐day trial was performed in order to improve growth of tench Tinca tinca (L.) larvae reared intensively in a controlled environment. Larvae of 4.53 mm initial length, 45 per dm3, were fed each day with live Artemia nauplii for 12, 18 or 24 h at 28°C. Final survival rates were very high in all treatment groups, ranging from 88.9 to 91.8%. Duration of daily feeding significantly affected growth and final condition, expressed by Fulton's coefficient (K) (P ≤ 0.05, Duncan's test). The fastest larval growth (17.6 mm total length, TL; 88.8 mg body weight, BW) and the best condition (K = 1.54) were recorded in the group fed 24 h each day. The second best results were attained with the larvae fed 18 h a day (16.5 mm TL, 67.9 mg BW, K = 1.40) whereas those fed 12 h daily grew the slowest (13.5 mm TL, 31.7 mg BW) and showed the worst condition (K = 1.19). Thus, to maximize growth and condition of tench larvae being reared at the optimal growth temperature, intensive 24 h feeding is necessary.  相似文献

Recent dramatic spread of the grapevine yellows disease Bois Noir (BN) in Germany is above all explained by highly increased abundances of the vector Hyalesthes obsoletus (Hemiptera: Cixiidae) associated to the plant Urtica dioica, the reservoir of the BN pathogen stolbur tuf‐type‐I. The vector acquires BN‐phytoplasma as larvae whilst feeding on the roots of infected U. dioica. To understand the dynamics of the Urtica‐cycle, we tested at what instar larvae become infected and whether infection affects larvae size (i.e. growth) at two sites in the Mosel Valley, Germany. Larvae were tested from infected plants and collected at instar‐stages 3, 4 and 5. Larvae at stage 3 were already infected but infection rates increased significantly between stage 3 and 5, mean infection rates: 0.12–0.62. There was no effect of infection on larval size at any instar stage.  相似文献

Effects of exposure to maize pollen of event Bt176 (cultivar “Navares”) on the larvae of the European common swallowtail (Papilio machaon L.) were studied in the laboratory. First instar larvae were exposed to different pollen densities applied to leaf disks of Pastinaca sativa L. for 48 h. Pollen densities applied in this study were in the range recorded from the field. Larvae which were exposed to higher Bt maize pollen densities consumed more pollen and had a lower survival rate. The LD50 with regard to larvae surviving to adulthood was 13.72 pollen grains consumed by first-instar larva. Uptake of Bt maize pollen led to a reduced plant consumption, to a lower body weight, and to a longer development time of larvae. Effects on pupal weight and duration of the pupal period were present but less pronounced and smaller than effects on larvae. Larvae having consumed Bt-maize pollen as first instars had a lower body weight as adult females and smaller forewings as adult males. We conclude that possible effects of Bt maize on European butterflies and moths must be evaluated more rigorously before Bt maize should be cultivated over large areas.  相似文献

Ground beetles (Coleoptera: Carabidae) are important components of soil ecosystems in temperate zones, but our knowledge regarding the ecology of many species is limited. This study examined larval food habits in the ground beetle Amara (Curtonotus) gigantea (Motschulsky) using laboratory‐rearing experiments. Because this beetle is a member of the tribe Zabrini, which includes several species that exhibit granivory during larval and/or adult stages, three diet types were tested, with starvation as a control: Tenebrio larvae, mixed seeds (Bidens pilosa (Asteraceae), Setaria spp., Digitaria ciliaris (both Poaceae), and Humulus scandens (Moraceae)), and Tenebrio larvae + mixed seeds. Because of high larval mortality during overwintering under laboratory‐rearing conditions, survival and the duration of development through pre‐overwintering stages (first and second instars) were compared. Larvae fed the diet of Tenebrio larvae + mixed seeds showed the highest survival (89%), followed by the diet of Tenebrio larvae (83%), but the difference between the two treatments was not significant; similarly, developmental durations did not differ between these two diets. Larvae fed the mixed‐seeds diet showed markedly lower survival, and no larvae reached the second instar. Therefore, for A. gigantea larvae, animal food results in high larval performance, whereas seeds provide a low value diet; animal food is essential for larval development, but seeds provide no benefit, even as a dietary supplement. Overall, the present results suggest that A. gigantea larvae are omnivorous but with a mainly carnivorous food habit.  相似文献
