Brassinosteroids (BRs) regulate various physiological processes, such as tolerance to stresses and root growth. Recently, a connection was reported between BRs and nitric oxide (NO) in plant responses to abiotic stress. Here we present evidence supporting NO functions in BR signaling during root growth process. Arabidopsis seedlings treated with BR 24-epibrassinolide (BL) show increased lateral roots (LR) density, inhibition of primary root (PR) elongation and NO accumulation. Similar effects were observed adding the NO donor GSNO to BR-receptor mutant bri1-1. Furthermore, BL-induced responses in the root were abolished by the specific NO scavenger c-PTIO. The activities of nitrate reductase (NR) and nitric oxide synthase (NOS)-like, two NO generating enzymes were involved in BR signaling. These results demonstrate that BR increases the NO concentration in root cells, which is required for BR-induced changes in root architecture.  相似文献

Brassinosteroids (BRs) play crucial roles in plant growth and development. Previous studies have shown that BRs promote cell elongation in vegetative organs in several plant species, but their contribution to meristem homeostasis remains unexplored. Our analyses report that both loss- and gain-of-function BR-related mutants in Arabidopsis thaliana have reduced meristem size, indicating that balanced BR signalling is needed for the optimal root growth. In the BR-insensitive bri1-116 mutant, the expression pattern of the cell division markers CYCB1;1, ICK2/KRP2 and KNOLLE revealed that a decreased mitotic activity accounts for the reduced meristem size; accordingly, this defect could be overcome by the overexpression of CYCD3;1. The activity of the quiescent centre (QC) was low in the short roots of bri1-116, as reported by cell type-specific markers and differentiation phenotypes of distal stem cells. Conversely, plants treated with the most active BR, brassinolide, or mutants with enhanced BR signalling, such as bes1-D, show a premature cell cycle exit that results in early differentiation of meristematic cells, which also negatively influence meristem size and overall root growth. In the stem cell niche, BRs promote the QC renewal and differentiation of distal stem cells. Together, our results provide evidence that BRs play a regulatory role in the control of cell-cycle progression and differentiation in the Arabidopsis root meristem.  相似文献

The dumpy (dpy) mutant of tomato (Lycopersicon esculentum Mill.) exhibits short stature, reduced axillary branching, and altered leaf morphology. Application of brassinolide and castasterone rescued the dpy phenotype, as did C-23-hydroxylated, 6-deoxo intermediates of brassinolide biosynthesis. The brassinolide precursors campesterol, campestanol, and 6-deoxocathasterone failed to rescue, suggesting that dpy may be affected in the conversion of 6-deoxocathasterone to 6-deoxoteasterone, similar to the Arabidopsis constitutive photomorphogenesis and dwarfism (cpd) mutant. Measurements of endogenous brassinosteroid levels by gas chromatography-mass spectrometry were consistent with this hypothesis. To examine brassinosteroid-regulated gene expression in dpy, we performed cDNA subtractive hybridization and isolated a novel xyloglucan endotransglycosylase that is regulated by brassinosteroid treatment. The curl-3 (cu-3) mutant (Lycopersicon pimpinellifolium ?Jusl. Mill.) shows extreme dwarfism, altered leaf morphology, de-etiolation, and reduced fertility, all strikingly similar to the Arabidopsis mutant brassinosteroid insensitive 1 (bri1). Primary root elongation of wild-type L. pimpinellifolium seedlings was strongly inhibited by brassinosteroid application, while cu-3 mutant roots were able to elongate at the same brassinosteroid concentration. Moreover, cu-3 mutants retained sensitivity to indole-3-acetic acid, cytokinins, gibberellin, and abscisic acid while showing hypersensitivity to 2, 4-dichlorophenoxyacetic acid in the root elongation assay. The cu-3 root response to hormones, coupled with its bri1-like phenotype, suggests that cu-3 may also be brassinosteroid insensitive.  相似文献

We have isolated a new recessive dwarf mutant of rice (Oryza sativa L. cv Nipponbare). Under normal growth conditions, the mutant has very short leaf sheaths; has short, curled, and frizzled leaf blades; has few tillers; and is sterile. Longitudinal sections of the leaf sheaths revealed that the cell length along the longitudinal axis is reduced, which explains the short leaf sheaths. Transverse sections of the leaf blades revealed enlargement of the motor cells along the dorsal-ventral axis, which explains the curled and frizzled leaf blades. In addition, the number of crown roots was smaller and the growth of branch roots was weaker than those in the wild-type plant. Because exogenously supplied brassinolide considerably restored the normal phenotypes, we designated the mutant brassinosteroid-dependent 1 (brd1). Further, under darkness, brd1 showed constitutive photomorphogenesis. Quantitative analyses of endogenous sterols and brassinosteroids (BRs) indicated that BR-6-oxidase, a BR biosynthesis enzyme, would be defective. In fact, a 0.2-kb deletion was detected in the genomic region of OsBR6ox (a rice BR-6-oxidase gene) in the brd1 mutant. These results indicate that BRs are involved in many morphological and physiological processes in rice, including the elongation and unrolling of leaves, development of tillers, skotomorphogenesis, root differentiation, and reproductive growth, and that the defect of BR-6-oxidase caused the brd1 phenotype.  相似文献

To withstand ever-changing environmental stresses, plants are equipped with phytohormone-mediated stress resistance mechanisms. Salt stress triggers abscisic acid (ABA) signaling, which enhances stress tolerance at the expense of growth. ABA is thought to inhibit the action of growth-promoting hormones, including brassinosteroids (BRs). However, the regulatory mechanisms that coordinate ABA and BR activity remain to be discovered. We noticed that ABA-treated seedlings exhibited small, round leaves and short roots, a phenotype that is characteristic of the BR signaling mutant, brassinosteroid insensitive1-9 (bri1-9). To identify genes that are antagonistically regulated by ABA and BRs, we examined published Arabidopsis microarray data sets. Of the list of genes identified, those upregulated by ABA but downregulated by BRs were enriched with a BRRE motif in their promoter sequences. After validating the microarray data using quantitative RT-PCR, we focused on RD26, which is induced by salt stress. Histochemical analysis of transgenic Arabidopsis plants expressing RD26pro:GUS revealed that the induction of GUS expression after NaCl treatment was suppressed by co-treatment with BRs, but enhanced by co-treatment with propiconazole, a BR biosynthetic inhibitor. Similarly, treatment with bikinin, an inhibitor of BIN2 kinase, not only inhibited RD26 expression, but also reduced the survival rate of the plant following exposure to salt stress. Our results suggest that ABA and BRs act antagonistically on their target genes at or after the BIN2 step in BR signaling pathways, and suggest a mechanism by which plants fine-tune their growth, particularly when stress responses and growth compete for resources.  相似文献

We examined the physiological effects of brassinosteroids (BRs) on early growth of Arabidopsis. Brassinazole (Brz), a BR biosynthesis inhibitor, was used to elucidate the significance of endogenous BRs. It inhibited growth of roots, hypocotyls, and cotyledonous leaf blades dose-dependently and independent of light conditions. This fact suggests that endogenous BRs are necessary for normal growth of individual organs of Arabidopsis in both photomorphogenetic and skotomorphogenetic programs. Exogenous brassinolide (BL) promoted hypocotyl elongation remarkably in light-grown seedlings. Cytological observation disclosed that BL-induced hypocotyl elongation was achieved through cell enlargement rather than cell division. Furthermore, a serial experiment with hormone inhibitors showed that BL induced hypocotyl elongation not through gibberellin and auxin actions. However, a synergistic relationship of BL with gibberellin A3 (GA3) and indole-3-acetic acid (IAA) was observed on elongation growth in light-grown hypocotyls, even though gibberellins have been reported to be additive to BR action in other plants. Taken together, our results show that BRs play an important role in the juvenile growth of Arabidopsis; moreover, BRs act on light-grown hypocotyl elongation independent of, but cooperatively with, gibberellins and auxin.  相似文献

A recessive mutation affecting both root and shoot developmentwas isolated from transformants of lettuce cv. Diana co-transformedwith maize Ac transposase and Ds. Mutant phenotype in the progenywas co-segregated with the T-DNA containing Ac transposase indicatingthat the mutation was caused by insertional mutagenesis. Mutationin this novel genetic locus, designated as ANORMAL ROOT ANDSHOOT (ARS), showed a dwarf phenotype with short thick roots,short hypocotyl and abnormal filamentous leaves without anyfurther reproductive development. The anatomical analysis revealedthat the ars mutant root phenotype is primarily due to the lackof cell elongation and to the abnormal increase in cell numberin the cortex region in the roots. ars mutants are able to initiatenormal leaf primodia, but, the cell elongation and cellularorganization of the developing leaf primodia is impaired andresults in sequentially abnormal development of the leaf. arsmutants also display photomorphogenic development in darknessby producing open cotyledons, developing vegetative leaves,and short hypocotyls. This suggests that the ARS gene may alsobe involved in the regulation of cell elongation in the hypocotylin the absence of light. Abnormal development in ars mutationscan not be normalized by exogenous application of phytohormonessuch as gibberellin and brassinosteroids, indicating that themutant is not impaired in the biosynthesis of these two hormones. Exogenously applied lysophosphatidylethanolamine (LPE) increased the growth of primary roots and the formation of lateral roots in Arabidopsis thaliana. In the presence of brassinolide, lateral root formation induced by LPE was enhanced, implying that both LPE and brassinosteroids (BR) interact positively in the development of Arabidopsis roots. Co-treatment with LPE and BRs increased the bending activity in the rice lamina inclination assay compared to that when BRs were applied alone, suggesting that LPE seems to exert its activity via BRs activity. RT-PCR revealed that LPE did not alter the expressions of genes involved in the biosynthesis of BRs but did activate the expressions of BR signaling genes in A. thaliana. In a BR-insensitive mutant, bri1, enhanced gravitropic response by LPE in wild-type A. thaliana was diminished. In conclusion, LPE is a positive regulator for the growth and development of Arabidopsis roots, and this process seems to be enhanced by BR signaling rather than by increase in endogenous levels of BRs in A. thaliana.  相似文献

Control of organ size by cell expansion and cell proliferation is a fundamental process during development, but the importance of BIG in this process is still poorly understood. Here, we report the isolation and characterization of a new allele mutant of BIG in Arabidopsis: big-j588. The mutant displayed small aerial organs that were characterized by reduced cell size in the epidermis and short roots with decreased cell numbers. The big-j588 axr1 double and big-j588 arf7 arf19 triple mutants displayed more severe defects in leaf expansion and root elongation than their parents, implying BIG is involved in auxin-dependent organ growth. Genetic analysis suggests that BIG may act synergistically with PIN1 to affect leaf growth. The PIN1 protein level decreased in both the root cells and the tips of leaf pavement cell lobes of big-j588. Further analysis showed that the auxin maxima in the roots and the leaves of big-j588 decreased. Therefore, we concluded that the small leaves and the short roots of big-j588 were associated with reduction of auxin maxima. Overall, our study suggested that BIG is required for Arabidopsis organ growth via auxin action.  相似文献

Our understanding of wood anatomy and radial growth in tree roots remains very incomplete, particularly with respect to how ecological factors affect root growth at a relatively small spatial scale, i.e., within a single root system. Here, we compared root growth with and without trampling exposure on a hiking trail. We conducted a quantitative analysis of radial growth and wood anatomical changes, including compression wood (CW) and blue rings (BRs), of two adjacent Scots pine roots in high resolution. A total of 32 cross sections from two roots sampled every 25 cm at the same distance from the respective stem were compared. The buried root (B) was completely buried and had an unexposed segment on a hiking trail. In contrast, the exposed root (E) had an exposed segment that was trampled. 1706 growth rings were analysed for the common period 1954–2015.We found that the volume of the E root in the trampling zone exceeded ten times the volume of the B root. The root surface area of the exposed sections of the E root was on average 14 times larger than that of the unexposed B root section in the trampling zone. The highest number of missing rings was found in the B root. Root sections sampled at the shortest distance from the stem showed the highest coherence in radial growth pattern, which decreased with increasing distance from the stem.BRs were recognized for the first time in tree roots. In total 25 tree rings contained BRs, and their occurrence was restricted to cross sections of the exposed root. BRs were formed over the course of 25 calendar years, i.e., in 40% of tree rings from the common period 1954–2015. Mean monthly temperatures for the years with and without BRs formation showed that colder November (p = 0. 012) and, albeit only slightly, colder September (p = 0.051) temperatures favoured formation of BRs in Scots pine roots. In addition, mean monthly precipitation in July (p = 0.017) was significantly higher for BR years, suggesting an impact of moisture availability on the formation of BRs in Scots pine roots. The study highlights a high rate of growth discrepancies within a single root system. Further, altered growth of trampled roots with high proportions of BRs opens a new challenge for future dendroecological studies on tree roots.  相似文献

The appearance of leaf mesophyll chloroplasts in angiosperms is characterized by their uniform and static shape, which is molded by symmetric division of the preexisting organelles, involving three prokaryote-derived proteins: the division executor protein, FtsZ, and the division site positioning proteins, MinD and MinE. On the other hand, noncolored plastids in roots, where the involvement of the known chloroplast division factors in plastid morphogenesis is yet unclear, are morphologically heterogeneous and transform dynamically. This is further emphasized by the active formation of long tubular protrusions called stromules from the main body of those plastids. Molecular regulation and physiological significance of such dynamic morphology of root plastids also remain unknown. In this context, we have recently demonstrated that the mitochondrial respiratory inhibitor antimycin A induces rapid and reversible filamentation of root plastids (leucoplasts) in Arabidopsis thaliana. In contrast, the same treatment with antimycin A did not affect the morphology of amyloplasts in the columella cells at the root tip. The alternative oxidase inhibitor salicylhydroxamic acid suppresses the antimycin-induced plastid filamentation, perhaps implying an alternative oxidase-mediated interorganellar signaling between the mitochondria and the leucoplasts in the root cells. The molecular control of cell elongation, one of the basic processes of plant morphogenesis, is still largely not understood. This paper describes a Petunia hybrida mutant of dumpy phenotype, trapu, which identifies tra1, a gene required for cell elongation and mediating responses to applied cytokinin. This mutant displayed an extreme reduction in length, due to a single recessive mutation which was expressed in every part of the plant and during the entire life of the plant, including the mature embryo. The mutant was unable to flower. The mutant roots, as well as the leafy organs, were short and thick, and the root elongation zone, hypocotyl and petioles were absent. The mutant plantlets responded neither to applied auxin nor to gibberellin, indicating that this phenotype was not caused by a deprivation of these phytohormones. However, unlike the wildtype, the mutant growth was stimulated by applied cytokinin, even though its morphology remained abnormal. A histological study revealed the presence of all tissue types in normal positions, including root hairs and vascular bundles. The mutant's cells were rounder in every tissue. Both shoot and root meristems were disorganized, without consistent cell shape and size. The regular cell files, which are typical of a normal root apex organization, were totally absent in the mutant root apex. Indirect immunofluorescence of α-tubulin on root apices showed the cortical microtubules in the mutant cells to be unable to form the parallel arrays in elongating cells and the preprophase band in dividing cells. This default resulted in the prevention of unidirectional cell elongation and formation of regular cell files, thus causing the trapu phenotype. This paper discusses the similarities and differences of trapu to the Arabidopsis mutants, fass and ton, trapu confirming that the establishment of plant body pattern and differentiation can be dissociated from cell elongation.  相似文献

We tested that the hypothesis that root elongation might be controlled by altering the level of ethylene in intact primary roots of maize(Zea mays L.). We measured root elongation in a short period using a computerized root auxanometer. Compounds which regulate ethylene production were applied to intact primary roots in different time periods. Root elongation was stimulated by the treatment with ethylene antagonists such as Co2+, aminoethoxyvinylglycine (AVG) and L-canaline. This result suggested that root elongation was closely related to ethylene level of intact primary roots. Furthermore, IAA- and 1-aminocyclopropane-1-carboxylic acid (ACC)-induced inhibition of root elongation was reversed by treatment with Co2+. The application of ACC to roots which have been exposed to IAA and Co2+ have no significant effect on root elongation. However, the inhibition of root elongation by ACC in roots previously treated with IAA and AVG became manifest when the applied IAA concentrations were lower. These results were consistent with the hypothesis that the level of ethylene in intact roots functions to moderate root elongation, and suggested that auxin-induced inhibition of root elongation results from auxin induced promotion of ethylene production.  相似文献

Brassinosteroids (BRs) are plant steroidal hormones that regulate a wide range of developmental processes. Most BR mutants display impaired growth and responses to developmental and environmental stimuli. Here, we found a BR-deficient mutant det2-1 that displayed exceedingly short roots and agravitropic growth, which were not present in other BR mutants. By back-crossing det2-1 with the wild type, we isolated a secondary mutation named det2-1 phenotype modifier 1 (dpm1) and demonstrated that those aberrant phenotypes in the original det2-1 were independent of the BR deficiency. Phenotypic analysis showed that impaired root growth of dpm1 appeared in BR-deficient condition, but not in a normal condition. In the light condition, the mutant showed enhanced shoot growth which was suppressed in the det2-1 background. Starch granules in the columella cells of the root tip were highly accumulated and expanded in dpm1. Agravitropic roots and the expanded starch granules of dpm1 could not be recovered by BR. Taken together, these results suggest that DPM1 is required for gravitropic growth, and that its functions on root and shoot growth are BR-dependent.  相似文献

Brassinosteroids (BRs) are a group of plant hormones and the bioactive BR, brassinolide (BL), is causally implicated in promoting cell elongation and cell proliferation. In Arabidopsis, the biosynthesis of BL is essential for hypocotyl etiolation in the dark, and application of bioactive BRs can promote both hypocotyl and root elongation, although high concentrations of applied BRs result in inhibition of root elongation. A non-steroidal structure consisting of four stereoisomers was designed to contain subunits bearing key functional groups mimicking those of BL. The bioactivity of each of these individual stereoisomers was tested using the Arabidopsis thaliana det2-1 mutant line, which is deficient in BL, and thus does not etiolate in the dark. Application of BL at each of 0.1, 1.0, and 10.0 µM promotes hypocotyl elongation in dark-grown det2-1 plants while simultaneously inhibiting elongation of their primary root. In contrast, the mimetic structures, when applied to dark-grown det2-1 plants, promote hypocotyl elongation without negatively affecting primary root elongation. In fact, two of the mimetic structures, applied at a 10 µM concentration, significantly promoted both hypocotyl and root elongation. Correlation of this contrasting behavior with the configurations of the hydroxylated stereocenters of the mimetics is described. This is the first example of a non-steroidal BL mimetic where the biological activities of individual stereoisomers were tested and compared.  相似文献
