During cytological screening for pollen sterility in a wild population of Haplopappus gracilis (n = 2), several partially sterile plants were found that had good pachytene pairing but varying numbers of univalents. Some plants had chromosome A bivalents or A univalents, while in the same cells chromosome B had only bivalents. In other plants the reverse condition occurred; the B chromosome had B bivalents or B univalents and only A bivalents. This demonstrates a chromosome-specific effect for the desynapsis genes. Hybridization between the two homozygous mutant genotypes produced only normal bivalents; this indicates the two mutants are not alleles and each is recessive. An F2 generation showed independent assortment of the desynaptic mutations. The chromosome A bivalent is the larger of the two and normally has one or two chiasmata; the B bivalent normally has a single chiasma. Chiasmata distribution was tested in the desynaptic mutant A bivalents and showed an acceptable fit to a binomial distribution. This occurs also in heterozygous, asynaptic pairing control gene mutations. Analysis of the NOR bivalent in two hologenomic desynaptic mutations in tomato also showed a good fit to a binomial distribution of chiasmata. This indicates the same methods are applicable to diverse species.  相似文献

Summary Cytological studies in desynaptic plants, isolated at the F6 generation of an intervarietal cross of Corchorus olitorius L., have shown variable numbers of bivalents and univalents in the PMC's at metaphase I, resulting in irregular distribution of chromosomes at anaphase I. The progenies of the desynaptic plants consisted of 9.24 percent of all possible primary trisomies except trisomie 6. The desynaptic condition is controlled by a pair of simple recessive genes.  相似文献

Male meiosis was studied in a population of Acanonicus hahni (Stål), and nine of the sixteen individuals analyzed showed desynapsis. The frequency of univalents varied from one to seven percent in eight of them, while in the ninth the percentage of cells with univalents was higher (12%). The univalents auto-orientate at metaphase I in the center of the ring formed by autosomal bivalents and divide equationally at anaphase I; at metaphase II they show touch-and-go pairing, and lie in the center of the ring of autosomes.A desynaptic origin of the univalents is proposed, and the arrangement of the chromosomes in the first and second metaphase plate in the normal and desynaptic individuals is compared and discussed. The meiotic characteristics of these desynaptic individuals are also compared with those described in other insects with holocentric and monocentric chromosomes. It is suggested that any achiasmatic chromosome, whether a univalent, m or sex chromosome, will induce the formation of a ring and with some or all of them lying in its centre.  相似文献

In most eukaryotes, homologous chromosomes undergo synapsis during the first meiotic prophase. A consequence of mutations that interfere with the fidelity or completeness of synapsis can be failure in the formation or maintenance of bivalents, resulting in univalent formation at diakinesis and production of unbalanced spores or gametes. Such mutations, termed desynaptic mutations, can result in complete or partial sterility. We have examined the effect of the maize desynaptic1-9101 mutation on synapsis, using the nuclear spread technique and electron microscopy to examine microsporocytes ranging from early pachytene until the diplotene stage of prophase I. Throughout the pachytene stage, there was an average of about 10 sites of lateral element divergence (indicating nonhomologous synapsis), and during middle and late pachytene, an average of two and three sites of foldback (intrachromosomal) synapsis, per mutant nucleus, respectively. By the diplotene stage, the number of sites of lateral element divergence had decreased to seven, and there was an average of one foldback synapsis site per nucleus. Lateral element divergence and foldback synapsis were not found in spread pachytene nuclei from normal plants. These results imply that the normal expression of the dsy1 gene is essential for the restriction of chromosome synapsis to homologues. The abundance of nonhomologous synapsis and the persistence of extended stretches of unsynapsed axial elements throughout the pachytene stage of dsy1–9101 meiocytes suggests that this mutation disrupts both the fidelity of homology search and the forward course of the synaptic process. Ultrastructural analysis of B chromosome synapsis in surface-spread (2B) pollen mother cells of the leek, Allium porrum, has clarified their structural organization and shed new light on their origin. In pachytene cells containing two B chromosomes, these chromosomes either formed a pair of univalents showing foldback hairpin loops or synapsed together to form bivalents of several different types. The synaptic configurations of univalents and bivalents indicate that these B chromosomes have a basically isochromosome organization, but this is modified by a slight centric shift giving an arm ratio of 1.1:1. This analysis adds to the growing number of B chromosomes that have been shown to be isochromosomes or isochromosome derivatives. The phenotype of the desynaptic (dy) mutant of maize in microsporocytes at meiotic prophase was compared with normal microsporocytes of a closely related strain and with microsporocytes of a maize inbred line (KYS) assumed to be normal. Strikingly more univalents and open arms of bivalents were found in the mutant cells than in normal cells at diakinesis, and where there was heterozygosity for a distal knob (heterochromatic region), separation was usually equational, indicating the occurrence of normal crossing-over followed by failure of chiasma maintenance in the mutant. Differences found in the mutant by electron microscopy were a statistically significant wider dimension of the synaptonemal complex central region and also less twisting of synapsed configurations at pachytene. It is suggested that these are side-effect symptoms of a defect in the synaptonemal complex (or associated substance), which is expressed later as sporadic loss of chiasma maintenance.  相似文献

Chromosome numbers, karyotypes, meiotic behaviour and pollen analysis are presented for species of Chamaecrista Moench (Leguminosae, Caesalpinioideae, Cassieae ) native to southern Brazil: C. nictitans ssp. patellaria , C. nictitans ssp. disadena , C. repens , C. rotundifolia , C. flexuosa , C. vestita and C. desvauxii . Meiotic behaviour is reported for the first time for all the taxa and was very regular; only bivalents were formed at diakinesis and metaphase I, chromosome disjunction and segregation were regular at anaphases I and II, meiotic indexes were over 99% and pollen fertility was over 92%. Pollen grains were subprolate in C. flexuosa and C. vestita and prolate–spheroidal in the other taxa. Karyotypes were symmetrical in all six species and the data are original, except for C. nictitans ssp. p atellaria. Chromosome number is presented for the first time for C. repens (2 n  = 16) and has been confirmed for the other taxa: 2 n  = 14 for C. desvauxii , 2 n  = 32 for the tetraploid C. nictitans ssp. patellaria and C. nictitans ssp. disadena , and 2 n  = 16 for the other species. Two types of filamentous microfossil are preserved within vessel elements and rays of pyritized and partly carbonized twigs from the Lower Eocene London Clay. The first type, probably a Streptomyces -like actinomycete, is slender (<1 µm) with branches and some regular septation. Wider filaments (>2 µm) are fungal hyphae; no reproductive structures are preserved. These filamentous organisms probably started growing saprophytically after the death of the twigs; the fungi created lysis tracks on cell walls. New findings are given for the Astragalus sect. Incani , which occurs within the area covered by the Flora of Iran. Astragalus wojciechowskianus Ranjbar is described as a new species. Populations of clonal organisms are often represented as being evolutionary inert with persistent genetic fidelity. The advent of molecular methods and the corresponding increased genetic resolution of clonal populations forces a reconsideration of this viewpoint. Webs of theridiid spiders: construction, structure and evolution   总被引:4,自引:0,他引:4  
Understanding the web construction behaviour of theridiid (comb-footed) spiders is fundamental to formulating specific evolutionary hypotheses and predictions regarding the reduction of orb-webs. We describe for the first time in detail the web construction behaviour of Achaearanea tepidariorum , Latrodectus geometricus , Theridion sisyphium and T. varians as well as webs of a range of other theridiids. In our survey we distinguish four major web types. Among webs with gumfooted lines, we distinguish between webs with a central retreat ( Achaearanea -type) and those with a peripheral retreat ( Latrodectus -type). Among webs without gumfooted lines, we distinguish between those which contain viscid silk ( Theridion -type) and those with a sheet-like structure, which do not ( Coleosoma -type). Theridiid gumfoot-webs consist of frame lines that anchor them to surroundings and support threads which possess viscid silk. Building of gumfooted lines constitutes a unique stereotyped behaviour and is most probably homologous for Nesticidae and Theridiidae. Webs remained in place for extended periods and were expanded and repaired, but no regular pattern of replacement was observed. We suggest that the cost of producing and maintaining viscid silk might have led to web reduction, at least in theridiids.  © 2003 The Linnean Society of London. The species of Goniothalamus (Annonaceae) occurring in Peninsular Malaysia and Singapore are revised, updating the previous taxonomic treatment by James Sinclair, published in 1955. A total of 18 species is recognized, including a new species, G. tomentosus . Collections referrable to G. tomentosus were previously determined as ' G. marcanii '; examination of the types of G. marcanii reveals that the two taxa are not conspecific, however, and a new name is accordingly validated here. Goniothalamus tomentosus has closest affinities with the Javan/Sumatran species G. costulatus . Astragalus neo-assadianus (Fabaceae), a new species endemic to Iran, is described and illustrated. This species belongs to A. sect . Alopecuroidei and is distinct and interesting among the Iranian species.
