Summary Kinetics of ethanol fermentation at varying sugar concentrations of Jerusalem artichoke tuber extract has been studied using Kluyveromyces marxianus cells immobilized in calcium alginate gel beads. A maximum ethanol concentration of 111 g/l was achieved at an initial sugar concentration of 260 g/l in 20 hours, when the immobilized cell concentration in the calcium alginate beads was 53.3 g dry wt./l bead volume. Ethanol yield remained almost unaffected by initial sugar concentration up to 250 g/l and was found to be about 88% of the theoretical. Maximum rate of ethanol production decreased from 22.5 g ethanol/l/h to 10.5 g ethanol/l/h while the maximum rate of total sugars utilization decreased from 74.9 g sugars/l/h to 28.5 g sugars/l/h as the initial substrate concentration was increased from 100 to 300 g/l. The concentration of free cells in the fermentation broth was low.  相似文献

Escherichia coli JM109(DE3) harboring expression plasmid pkAQNÆC30, which carries the Thermus protease aqualysin I (AQI) gene, was cultivated with glucose as a sole carbon source. The final cell concentration was over 15 g dry weight/l and the amount of AQI produced reached approximately 130 kU/ml broth. Moreover, by using two carbon sources, glucose and glycerol, the production yield was increased to over 200 kU AQI/ml, while suppressing the formation of inhibitory acetic acid.  相似文献

Summary Mass production of Poly(-hydroxybutyrate) (PHB) from Methylobacterium organophilum under potassium-limited condition was carried out using a microcomputeraided automatic fed-batch culture system. The concentration of methanol was kept within the range of 2 – 3 g/l which did not show any inhibitory effect on cell growth. The PHB accumulation was stimulated when potassium concentration in the culture broth fell below 25 mg/l. After 70 hours of cultivation, the concentrations of cell mass and PHB were obtained to be 250 g/l and 130 g/l, respectively, which corresponded to a volumetric productivity of 1.8 – 2.0 g-PHB/1-hr. PHB contents ranged from 52% to 56% of dry cell weight with a yield factor (YP/S) of 0.19 g-PHB/g-methanol.  相似文献

A stable and fast l-tryptophan producer, AGX1757, was isolated from Escherichia coli W3110 trpAE1 trpR tnaA, which carried pSC101-trpI15·14. Cells of AGX1757 did not lose the composite plasmid during fermentation. Whenever a fed-batch culture of AGX1757 attained an l-tryptophan concentration of about 30 g/l, indole began to appear in the broth. The emergence of indole was caused by inhibition of tryptophan synthase due to accumulated l-tryptophan. Hence, the production rate of l-tryptophan sharply decreased. A higher solubility of l-tryptophan in the supernatant of culture broth (about 32 g/l) than that in the initial medium (about 22 g/l) was attributed to some unknown interaction between l-tryptophan and certain macromolecular material(s) coming from the bacterial cells. An addition of non-ionic detergents into the supernatant was effective for decreasing the solubility of l-tryptophan, hence causing crystallization of l-tryptophan. Pluronic L-61 was supplied from outside to an extent of 0.5% in terms of wt% concentration at around 45 h of fermentation when the l-tryptophan accumulated reached about 25 g/l. This addition actually caused crystallization of l-tryptophan and, as a result, the inhibitory effect of tryptophan synthase by l-tryptophan accumulated in the broth could be alleviated. Thus far, further fermentation became possible. l-Tryptophan of more than 50 g/l was finally produced by feeding solutions of both glucose and anthranilic acid. Correspondence to: H. The production of lipase by Candida rugosa in batch cultures was studied. The initial concentration of the carbon source employed, oleic acid, had an important effect on the final lipolytic activity levels. The maximum lipase/substrate yield and specific productivity obtained correspond to an initial oleic acid concentration of 2 g/l. At higher concentrations, up to 8 g/l oleic acid, specific productivity decreased. Lipase production was not observed below 1 g/l oleic acid. Lipase inactivation in culture broth due to surface forces and shear stress at the gas/liquid interface was not observed. There was no shear stress denaturation at stirring rates of 250, 500 and 750 rpm. No temperature inactivation was detected up to 50° C. Two different lipases with a similar molecular weight of 60kDa were purified from culture broth.  相似文献

将变形假单胞菌的精氨酸脱亚胺酶(ADI)编码基因arc A克隆至具有阿拉伯糖启动子的分泌型表达载体pBAD/gⅢB中,经鉴定得到重组质粒pBAD-ADI。将重组质粒转化大肠杆菌TOP10F'后进行诱导表达,分别考察了不同诱导物L-arabinose浓度、诱导温度、诱导时间对重组蛋白表达的影响,最适诱导条件为L-arabinose浓度0.002%(w/v),25℃下诱导5 h,全细胞的酶活为68 mU/mL(指单位发酵液体积,下同)。采用Osmotic Shock法使ADI从胞周质释放出来,经检测分泌到胞周质的重组蛋白活性为53 mU/mL,细胞内的酶活为34 mU/mL。SDS-PAGE分析显示,重组蛋白大小约为46 kD。  相似文献

Summary WhenMortierella alpina ATCC 32222 was incubated in a glucose salts medium at 25°C the biomass (17.5 g/l) contained 9.62% arachidonic acid which amounted to 54% (w/w) of total biomass lipids. When the glucose concentration in the medium was varied from 0 to 150 g/l, the percentage of arachidonic acid in biomass and in lipids was highest at a glucose concentration of 30 g/l, but highest yield of arachidonic acid per litre of culture broth was observed at a glucose concentration of 100 g/l. While production of biomass reached a plateau of 17 g/l after a 3-day incubation at 25°C, the percentage of arachidonic acid in lipids and biomass increased dramatically from 3 to 6 days with a concurrent arachidonic acid yield increase from 0.89 to 1.63 g/l. Optimum initial culture pH for arachidonic acid production was in the range 6.0–6.7. By increasing the concentration of the glucose salts medium three-fold, yields of biomass and arachidonic acid were increased to 35.8 g/l and 3.73 g/l, respectively.  相似文献

Pseudomonas cepacia was cultivated on salicylate as sole carbon and energy source in a fed-batch culture. On-line measurement of salicylate concentration was carried out using a filtration system. Cell-free permeate was passed through a flow-through spectrophotometer. Measurements were carried out at 325 nm. A Proportional-Integral controller was used for regulating the feed rate around a basic dosage scheme to maintain a constant salicylate concentration of 0.2 g/l in the broth. The cell mass concentration increased from less than 1 g dry weight (dw)/l to 12 g dw/l in less than 6 h, and the yield coefficient was 0.4 g dw/g salicylate. Summary High concentration cultivation of Bifidobacterium longum in a fermenter with cross-flow filtration using a ceramic filter is described. Continuous cross-flow filtration allowed complete recycling of the cells to the fermenter and also continuous separation of inhibitory metabolites. The final cell concentration attained in the cultivation was 54.4 g dry wt./l; this was seven times as high as that without cross-flow filtration. The basic parameters were studied influencing the conversion of orbitol to sorbose by Gluconobacter oxydans(industrial strain from FARMAKON Co., Czechoslovakia). The most effective conversion in the stationary phase was reached at pH 5.0, no inhibitory effect of sorbitol in a concentration ranging from 20 to 200 g/l and a minimum inhibitory effect of the sorbose concentration up to 200 g/l were observed. According to the optimum conditions mentioned above the optimized course of the fed-batch cultivation was proposed. The final concentration of sorbose of 410 g/l was reached after 36 hours.  相似文献

Zoogloea ramigera, a gram-negative bacterium found in activated sludge, formed volutin granules when excess orthophosphate was added to a phosphate-starved culture. These volutin granules were stainable by hydrogen sulfide after lead acetate treatment and extractable by N-perchloric acid but were not adsorbed by activated charcoal. They appeared to consist of inorganic polyphosphate. Optimum granule formation in the arginine broth required 10 g of glucose, 3 mg of phosphate, and 1 to 20 mg of magnesium per liter of medium. At an Mg(2+) concentration of 1 mg/liter, very large granules appeared which often appeared to fill the cell. An excess of glucose, orthophosphate, or magnesium reduced granule formation. In the absence of sulfate, moderate granulation occurred in arginine broth before the addition of excess orthophosphate; granulation did not increase after the addition of phosphate.  相似文献

【目的】采用响应面法对戈壁三素链霉菌PW409发酵合成α-糖苷酶抑制剂的培养基进行优化。【方法】采用Plackett-Burman法筛选影响α-糖苷酶抑制剂产生的关键因素,用最陡爬坡试验逼近关键因素的最大响应区域,采用Box-Behnken设计以及响应面分析法,得到各因素的最佳浓度,通过液相色谱-串联质谱法(LC-MS/MS)对发酵液中α-糖苷酶抑制剂进行定量分析。【结果】发酵培养基中可溶性淀粉、KNO3和K2HPO4的浓度对α-糖苷酶抑制剂的产量影响较大。优化后的培养基组成为:可溶性淀粉9.01 g/L,KNO3 11.0 g/L,K2HPO4 0.32 g/L,MgSO4.7H2O 0.50 g/L,FeSO4.7H2O 0.01 g/L,pH 7.5。【结论】在此优化条件下,链霉菌PW409发酵液对麦芽糖苷酶的半数抑制浓度IC50为22 mg/L,抑制活性较优化前提高了近10倍。发酵液中的1-脱氧野尻霉素含量可达7.84 mg/L,较优化前提高了668倍,米格列醇的含量可达0.94 mg/L,较优化前提高了10倍。  相似文献

By immobilizing fungi on a urethane foam carrier (UFC), a novel solid-state fermentation system was developed in order to produce repeatedly industrial useful enzymes. In this study, alkaline protease was produced by growing Aspergillus oryzae 460 (ATCC 20386) in a flask scale. Repeated batch production of alkaline protease was carried out by exchanging a part of the culture broth with fresh medium every 12 hr. The effects of feeding medium composition, and feeding volume were examined. Alkaline protease production stopped in the early phase at high values of soluble starch feeding rate and culture broth dilution rate. The enzyme production continued longer when 10 to 30 g/l polypepton was added to the feeding medium. Using soluble starch solution as feeding medium, a maximum activity of 520,000 U/l-bulk volume alkaline protease was obtained at culture time of 168 hr where the culture conditions of soluble starch concentration and feeding volume were 100 g/l and 0.025 l/l-bulk volume/dose, respectively. Production of the enzyme continued for 300 hr and total aklaline protease activity reached 870,000 U/l-bulk volume by adding 30 g/l polypepton to the fresh medium.  相似文献

Xylitol is a well-known sugar substitute with low-calorie and anti-cariogenic characteristics. An effort of biological production of xylitol from xylose was made in repeated fed-batch and cell-recycle fermentations of recombinant Saccharomyces cerevisiae BJ3505/δXR harboring the xylose reductase gene from Pichia stipitis. Batch fermentation with 20 g/l xylose and 18 g/l glucose resulted in 9.52 g/l dry cell mass, 20.1 g/l xylitol concentration and approximately 100% conversion yield. Repeated fed-batch operation to remove 10% of culture broth and to supplement an equal volume of 200 g/l xylose was designed to improve xylitol production. In spite of a sudden drop of cell concentration, an increase in dry cell mass led to high accumulation of xylitol at 48.7 g/l. To overcome loss of xylitol-producing biocatalysts in repeated fed-batch fermentation, cell-recycle equipment of hollow fiber membrane was implemented into a xylitol production system. Cell-recycle operation maintained concentration of the recombinant cells high inside a bioreactor. Final dry cell mass of 22.0 g/l, 116 g/l xylitol concentration, 2.34 g/l h overall xylitol productivity were obtained in cell-recycle fermentation supplemented with xylose and yeast extract solution, which were equivalent to 2.3-, 5.8- and 3.8-fold increases compared with the corresponding values of batch-type xylitol production parameters.  相似文献

The angiotensin-converting enzyme (ACE) inhibitory effect was tested in the culture broth from submerged mycelial cultures of 20 basidiomycetes. The ACE inhibitory effect of culture broth from Flammulina velutipes strain 414 was the highest (52.8%), followed by Lentinus edodes strains 2 (44.4%) and 16 (41.3%). Nutritional requirements for the production of ACE inhibitory substance from F. velutipes were studied. Sucrose, ammonium acetate, and glutamic acid were chosen for the maximum production of ACE inhibitory substance. The optimal medium composition was (g/l): sucrose 20, ammonium acetate 5, glutamic acid 2, KH2PO4 3, MgSO4·7H2O 0.8, and yeast extract 0.5. The efficiency of bipolar electrodialysis (BED) for the recovery of lactic acid from fermentation broth was evaluated. Three systems of BED (bipolar-anion, bipolar-cation and bipolar-anion-cation) at fixed voltage (20 V) were compared using a model solution of ammonium lactate (100 g l(-1)). Results showed that bipolar-anion (BED-anion) was the most beneficial in terms of lactate flux, current efficiency, energy consumption and recovery ratio. Consequently, BED-anion was used to purify lactic acid from fermentation broth which had been pre-treated with mono-polar electrodialysis (MED). The final lactic acid concentration and lactate flux obtained were 144 g l(-1) and 393 g m(-2) h(-1), respectively. Using the two-step process (MED and BED-anion) the concentration of fermentation broth was increased by 33% and the total energy consumption was 2.76 kW h kg(-1).  相似文献

Serum-free perfusion cultures of hybridoma TO-405 cells were carried out in spinner flasks coupled with zeolite A-3 packed beads. Ammonia was selectively removed from the culture broth by passing cell free permeate from ceramic cross flow filtration, through the zeolite packed bed. Ammonia concentration in the culture broth was effectively maintained between 1 to 4 mmol/l which was below the inhibitory concentration for cell growth. Maximum cell density levels of 107 cells/ml as well as improved percentage cell viability higher than in serum-supplemented cultures were feasible in this system.The possible effects of shear stress, generated by variation of the flow rates of the broth through the ceramic filter module, on the growth of the hybridoma cells were investigated. Backwashing, by reversing the direction of the permeate, was found necessary to prolong the life of the filter. Variation of the flow rates of the broth through the ceramic module between 0.29 m/s to 0.59 m/s did not cause immediate cell damage but growth was repressed at the higher flow rate.This study also showed that glutamine appears to be one of the factors limiting the growth of the hybridoma cells.  相似文献

用二苯代苦味肼基自由基(DPPH)-TLC法和酶标仪法对一株细脚拟青霉RCEF0394发酵液甲醇提取物的清除自由基活性进行了定性和定量测定,发现该提取物具有较强的清除自由基活性,在浓度为5.0mg/mL,于37℃下保温10min时,它对0.4mg/mL的DPPH自由基的清除率可达75.4%。以大鼠肝脏线粒体单胺氧化酶为靶标的体外实验发现供试细脚拟青霉发酵液有较强的抑制单胺氧化酶活性,且其活性和浓度呈量效关系。该发酵液冻干品对单胺氧化酶的半数抑制浓度IC50为118.1μg/mL。其三氯甲烷提取物对单胺氧化酶的抑制活性明显强于发酵液冻干品,表明该抑制剂可能为极性较低的化合物。进一步的分型试验表明该三氯甲烷提取物对A型单胺氧化酶呈混合抑制,对B型呈竞争性抑制,其Km值分别为0.44mg/L,0.34mg/L。  相似文献

目的观察布洛芬对曲霉临床分离株的体外抗真菌活性。方法分别用微量液基稀释法和纸片扩散法,测定布洛芬对10株烟曲霉、黄曲霉和土曲霉的抗菌活性。结果微量液基稀释法显示布洛芬对曲霉的最低抑菌浓度(MIC)范围为1000~2000μg/mL,最低杀菌浓度(MFC)范围为2000~8000μg/mL;纸片扩散法也显示布洛芬有体外抗曲霉活性:48h时,1000μg布洛芬对曲霉产生的抑菌圈直径为(20.1±3.89)mm。结论布洛芬有体外抗曲霉活性。  相似文献
