Summary. We used qualitative and quantitative fluorescence microscopy of the fluorescein isothiocyanate-conjugated lectins Concanavalin A, phytohaemagglutinin-erythroagglutinin, pokeweed mitogen, and peanut agglutinin to examine sugar composition on the cell surface and cell-associated mucilage (where present) in a number of cultured and environmental algae. Lectin-binding activity was markedly different between laboratory-cultured and environmental samples of the same species. Sugar composition of the cyanobacterium Anabaena cylindrica varied with growth cycle, although no clear pattern of change was observed. Akinetes typically showed lectin-binding activity higher than that of the vegetative cells or heterocysts throughout the growth cycle. Algae with mucilage showed greater lectin binding, indicating that mucilage contained more surface sugars accessible to the lectin probe compared with the cell wall surface. A low level of galactose and N-acetyl galactosamine (detected by peanut agglutinin) was associated with the surface mucilage of most algal species. Relatively high amounts of mannose, glucose, and N-acetyl glucosamine (detected by Concanavalin A, phytohaemagglutinin, and pokeweed mitogen) were also present. Lectin binding was shown to be a highly specific and sensitive approach to the examination of cell surface chemistry of both cultured and environmental algae and to the study of biodiversity in phytoplankton. Young gametophytes of Botrychium dissectum produce a mucopolysaccharide coating on the external surface of the proximal cell. Scanning electron microscopy reveals deposition of the mucilage, which initially has a patchy distribution, in the shape of a thick triangular ring. Young gametophytes in the hydrated condition have a thin coating of mucilage over the other areas of the proximal cell wall. Histochemical staining indicates that the mucilage contains sugars with vicinal hydroxyl groups, carboxylated sugars, and small amounts of sulfated sugars. Protein and phenolic materials are also found in the mucilage. Lipids, β, 1-3 glucans and β,1-4 glucans are not present. Fluorescein labelled lectin binding shows the presence of terminal galactose and terminal fucose units. Germinating spores in the presence of the microtubule inhibitors CIPC (3-chloro-N-phenyl isporopyl carbamate) and griesiofulvin cause abnormal mucilage secretion. The inhibitor CIPC prevents mucilage secretion whereas griesiofulvin disrupts the pattern of deposition. Several functions of the mucilage are postulated.  相似文献

BACKGROUND AND AIMS: The impedance to root growth imposed by soil can be decreased by both mucilage secretion and the sloughing of border cells from the root cap. The aim of this study is to quantify the contribution of these two factors for maize root growth in compact soil. METHODS: These effects were evaluated by assessing growth after removing both mucilage (treatment I -- intact) and the root cap (treatment D -- decapped) from the root tip, and then by adding back 2 micro L of mucilage to both intact (treatment IM -- intact plus mucilage) and decapped (treatment DM -- decapped plus mucilage) roots. Roots were grown in either loose (0.9 Mg m(-3)) or compact (1.5 Mg m(-3)) loamy sand soils. Also examined were the effects of decapping on root penetration resistance at three soil bulk densities (1.3, 1.4 and 1.5 Mg m(-3)). KEY RESULTS: In treatment I, mucilage was visible 12 h after transplanting to the compact soil. The decapping and mucilage treatments affected neither the root elongation nor the root widening rates when the plants were grown in loose soil for 12 h. Root growth pressures of seminal axes in D, DM, I and IM treatments were 0.328, 0.288, 0.272 and 0.222 MPa, respectively, when the roots were grown in compact soil (1.5 Mg m(-3) density; 1.59 MPa penetrometer resistance). CONCLUSIONS: The contributions of mucilage and presence of the intact root cap without mucilage to the lubricating effect of root cap (percentage decrease in root penetration resistance caused by decapping) were 43 % and 58 %, respectively. The lubricating effect of the root cap was about 30 % and unaffected by the degree of soil compaction (for penetrometer resistances of 0.52, 1.20 and 1.59 MPa).  相似文献

The mucilage of Opuntia ficus-indica and a partially degraded mucilage have been investigated by methylation analysis and periodate oxidation. The results show that the mucilage is composed of 1,4-α-D-galactopyranosyluronic acid and 1,2-β-L- rhamnopyranosyl residues to which are attached short chains of 1,6-β-D-galacto-pyranosyl residues at position 4 of all of the rhamnopyranosyl residues. Most of the galactosyl residues in the side chains carry branches at O–3, while some are also branched at O–4. The branches are mainly composed of 1,5-linked arabinofuranosyl, end-group arabinofuranosyl, and end-group xylopyranosyl residues.  相似文献

The mucilage phenomenon, a sporadic but massive accumulation of gelatinous material, can cause serious damage to the tourism and fishing industries along the Adriatic coast. Mucilage is presently thought to be the result of the aggregation of dissolved organic matter (DOM) into particulate organic matter (POM). Three principal classes of compounds have been identified in organic matter by spectrometric determination: carbohydrates, proteins and lipids. Carbohydrates are suspected to play a role in the first steps of DOM aggregation. Despite its importance in understanding the processes leading to mucilage formation, our present knowledge of the composition of the mucilage carbohydrate fraction is incomplete. Due to its high sensitivity and specificity, liquid chromatography coupled with electrospray-ionization tandem mass spectrometry (LC-ESIMS/MS) is gaining an increasing importance as a powerful technique for carbohydrate purification and characterization in complex samples. In this work, LC-ESIMS/MS is proposed as a useful method for the investigation of the oligosaccharide content in mucilage samples. The approach was applied using 3-7 unit maltooligosaccharides as reference compounds. The composition of the investigated mucilage sample was further investigated combining LC-ESIMS/MS with classic approaches, such as spectroscopic techniques and liquid chromatography coupled with the refractory index LC-RI.  相似文献

The Arabidopsis thaliana accession Shahdara was identified as a rare naturally occurring mutant that does not liberate seed mucilage on imbibition. The defective locus was found to be allelic to the mum2-1 and mum2-2 mutants. Map-based cloning showed that MUCILAGE-MODIFIED2 (MUM2) encodes the putative beta-D-galactosidase BGAL6. Activity assays demonstrated that one of four major beta-D-galactosidase activities present in developing siliques is absent in mum2 mutants. No difference was observed in seed coat epidermal cell structure between wild-type and mutant seed; however, weakening of the outer tangential cell wall by chemical treatment resulted in the release of mucilage from mum2 seed coat epidermal cells, and the mum2 mucilage only increased slightly in volume, relative to the wild type. Consistent with the absence of beta-D-galactosidase activity in the mutant, the inner layer of mucilage contained more Gal. The allocation of polysaccharides between the inner and outer mucilage layers was also modified in mum2. Mass spectrometry showed that rhamnogalacturonan I in mutant mucilage had more branching between rhamnose and hexose residues relative to the wild type. We conclude that the MUM2/BGAL6 beta-D-galactosidase is required for maturation of rhamnogalacturonan I in seed mucilage by the removal of galactose/galactan branches, resulting in increased swelling and extrusion of the mucilage on seed hydration.  相似文献

Abstract The dry weight (0.1%) and water potential -7 kPa) of root-cap mucilage from 3-d-old axenically grown maize seedlings have been determined. The results suggest strong gelling properties and weak water-holding capacity for the mucilage. Root tips from seedlings grown under low or high water stress were fixed by freeze-substitution. Micrographs showed that in both conditions, mucilage was secreted into the periplasmic space and extruded through the cell wall, though in dry conditions, the mucilage was tightly pressed against the root-cap surface. Histochemical and structural evidence is presented indicating chemical changes in the composition of the mucilage upon extrusion and a sharp increase in its hydration at increasing distance from the secretory cells. The possible functions of the root-cap mucilage in the rhizosphere are examined in light of these findings.  相似文献

Summary Extracellular polysaccharide/proteoglycan (EPS) mucilages play a crucial role in maintaining the structure of the extensive algal sheets that appear along the undersurface of nearshore Antarctic sea ice during the austral spring. In this study we have determined the composition and ultrastructural location of a family of novel sulphated polysaccharides/proteoglycans from the pennate ice diatomStauroneis amphioxys Gregory. They occur as soluble EPS in the culture supernatant, as an intercellular mucilage sheet, and as components of a distinct organic layer (diatotepum) underlying the silicious cell wall. The ultrastructural location and quantitative extraction of the mucilage EPS and the major diatotepum polysaccharides with hot water and alkali, respectively, was monitored by light and electron microscopy. The EPS and wall components were purified by Ultrafiltration, anion exchange and gel filtration chromatographies, and their monosaccharide composition was determined by gas-chro-matography mass spectrometry. The soluble and mucilage EPS, and major diatotepum polysaccharides/proteoglycans had an apparent molecular mass greater than 2 × 106 Da on gel. They contained a similar complex monosaccharide composition that includes glucuronic acid and galactose as the major sugars and significant levels of rhamnose, fucose, arabinose, xylose, mannose, glucose and the mono-O-methylated monosaccharides 3-O-methylrhamnose, 3-O-methylfucose, 3-O- and 4-O-methylxylose. The ratios of Gal to GlcA, which together account for 45% of the monosaccharides, varied from 0.8 (in the soluble EPS) to 2.3 (in diatotepum polysaccharides). The level of sulphation also varied from 5–15% (w/w), with the mucilage EPS being the most highly sulphated. The soluble EPS also contains a small amount of protein (ca. 5%, v/w) which cochromatographs with the polysaccharide during gel filtration and anion exchange chromatographies suggesting that it may be a sulphated proteoglycan. They are clearly distinct from a sulphated glucuronomannan that remained in the alkali-insoluble fraction and may be tightly associated with the silica wall components. The amount of mucilage EPS increased during logarithmic growth but decreased during stationary phase, when most of the EPS was found in the soluble pool. These changes correlate with the breakdown of the mucilage sheet and dispersal of diatom colonies during stationary growth. Interestingly, the soluble EPS from stationary-growth cultures was indistinguishable from the mucilage EPS of logarithmic- or stationary-phase cells, suggesting that the dissolution of the intercellular mucilage was not due to a change in EPS composition. The possibility that cell motility may be required for mucilage formation and the significance of these polysaccharides in the under-ice community is discussed.  相似文献

以新疆荒漠植物抱茎独行菜为材料,运用光镜与扫描电镜观察以及紫外吸收光谱法、化学反应及种子萌发实验等方法,对粘液质的形态和结构,物理化学特性,粘液质对种子萌发及萌发后的影响进行了研究.结果显示:(1)完整干种子表面覆盖着一层膜状物质(完全脱水的粘液质),并呈同一走向的山脊状突出的网状结构,遇水后粘液物质呈射线状向外发射出来,化学反应实验结果表明,粘液质的组成可能是某种多糖,如β-葡聚糖.(2)粘液质约占干种子重量的1/4,有很强的吸水能力,完全浸润10 min后,种子重量增加约30~40倍,种子长度、宽度、厚度的增加分别多于1倍、2倍、4倍;完全润湿的种子能够粘附相当于其干种子重量68倍的沙粒.(3)种皮粘液质对于不同土壤基质中的种子萌发有重要作用,但是对萌发后幼苗的生长没有作用.  相似文献

The main component of the mucilage in the bulbs of Suisen (Narcissus tazetta L., var. chinensis Roem) has been shown to be a glucomannan composed of D-glucose and D-mannose in the ratio of 2:3 and having a relatively low degree of branching. Acetolysis of the polysaccharide led to the isolation of β-(1→4)-β-(1→3)-linked oligosaccharides composed of D-mannose and/or D-glucose residues. Background

Marine snow (small amorphous aggregates with colloidal properties) is present in all oceans of the world. Surface water warming and the consequent increase of water column stability can favour the coalescence of marine snow into marine mucilage, large marine aggregates representing an ephemeral and extreme habitat. Marine mucilage characterize aquatic systems with altered environmental conditions.

Methodology/Principal Findings

We investigated, by means of molecular techniques, viruses and prokaryotes within the mucilage and in surrounding seawater to examine the potential of mucilage to host new microbial diversity and/or spread marine diseases. We found that marine mucilage contained a large and unexpectedly exclusive microbial biodiversity and hosted pathogenic species that were absent in surrounding seawater. We also investigated the relationship between climate change and the frequency of mucilage in the Mediterranean Sea over the last 200 years and found that the number of mucilage outbreaks increased almost exponentially in the last 20 years. The increasing frequency of mucilage outbreaks is closely associated with the temperature anomalies.

Conclusions/Significance

We conclude that the spreading of mucilage in the Mediterranean Sea is linked to climate-driven sea surface warming. The mucilage can act as a controlling factor of microbial diversity across wide oceanic regions and could have the potential to act as a carrier of specific microorganisms, thereby increasing the spread of pathogenic bacteria.  相似文献

Dendritic cells are believed to play an essential role in regulating the balance between immunogenic and tolerogenic responses to mucosal Ags by controlling T cell differentiation and activation via costimulatory and coinhibitory signals. The CD28/CTLA-4-CD80/CD86 signaling pathway appears to be one of the most important regulators of T cell responses but its exact role in responses to orally administered proteins remains to be elucidated. In the present study, the involvement of the CD28/CTLA-4-CD80/CD86 costimulatory pathway in the induction of allergic sensitization and oral tolerance to peanut proteins was investigated. In both an established C3H/HeOuJ mouse model of peanut hypersensitivity and an oral tolerance model to peanut, CD28/CTLA-4-CD80/CD86 interactions were blocked using the fusion protein CTLA-4Ig. To examine the relative contribution of CD80- and CD86-mediated costimulation in these models, anti-CD80 and anti-CD86 blocking Abs were used. In the hypersensitivity model, CTLA-4Ig treatment prevented the development of peanut extract-induced cytokine responses, peanut extract-specific IgG1, IgG2a, and IgE production and peanut extract-induced challenge responses. Blocking of CD80 reduced, whereas anti-CD86 treatment completely inhibited, the induction of peanut extract-specific IgE. Normal tolerance induction to peanut extract was found following CTLA-4Ig, anti-CD86, or anti-CD80 plus anti-CD86 treatment, whereas blockade of CD80 impaired the induction of oral tolerance. We show that CD28/CTLA-4-CD80/CD86 signaling is essential for the development of allergic responses to peanut and that CD86 interaction is most important in inducing peanut extract-specific IgE responses. Additionally, our data suggest that CD80 but not CD86 interaction with CTLA-4 is crucial for the induction of low dose tolerance to peanut.  相似文献

Aims

Seeds of Henophyton deserti (Brassicaceae), an endemic saharan shrub in south Tunisia, produce a pectinaceous mucilage layer that can imbibe a large amount of water when wetted. The aim of this study was to explore the role of mucilage in seed germination of this shrub under heterogeneous stressful environments.

Methods

Germination of both intact and demucilaged seeds was tested over wide ranges of temperature, and in iso-osmotic solutions of NaCl and PEG. Recovery of germination after NaCl and Polyethylene Glycol (PEG)-6000 treatment was also tested. The effect of mucilage on water uptake was measured and the structure of the seed investigated.

Results

A considerable proportion of seed mass (30 %) is made up of mucilage, which is extremely hydrophilic and able to increase seed mass by 550 % over dry seeds. Mucilage water uptake appears to be unaffected by salt concentration, while higher concentrations of PEG inhibit mucilage hydration. Mucilage decreases germination specifically at 10 °C and this effect can be interpreted in relation to oxygen uptake. High concentrations of NaCl and PEG decrease both germination percentage and rate, with some greater tolerance at 15 °C and 20 °C versus 25 °C. Recovery was higher from higher concentrations of NaCl and PEG and lower temperatures, with a clear inhibitory effect of mucilage.

Conclusions

The study has shown that the mucilage of H. deserti may act as a physical barrier for regulating diffusion of water and oxygen to the inner tissue of the seed and thereby prevent germination under unsuitable conditions.  相似文献

Background

Diagnosing peanut allergy properly is important and can be achieved by combining clinical history with various diagnostic methods such as IgE-antibody (IgE-ab) measurements, skin-prick test, basophil allergen threshold sensitivity (CD-sens) and food challenge. We aimed to evaluate CD-sens to peanut, Ara h 8 and Gly m 4 in relation to an oral peanut challenge in children IgE-sensitized to birch, peanut and Ara h 8 avoiding peanuts.

Methods

Twenty children IgE-sensitized to birch pollen and Ara h 8, but not to Ara h 1, Ara h 2 or Ara h 3 were challenged orally with roasted peanuts. Blood samples were drawn for IgE-ab and CD-sens analysis. To measure CD-sens, basophils were stimulated in vitro with decreasing doses of allergens until threshold sensitivity was reached.

Results

All children passed challenge without objective symptoms, but mild oral allergy syndrome (OAS) symptoms were reported in 6/20 children. Nineteen of twenty children were negative in CD-sens to peanut but 17/20 were positive to rAra h 8. Eleven of twenty children were positive in CD-sens to rGly m 4.

Conclusion

Positive CD-sens to rAra h 8 show that the Ara h 8 IgE-ab sensitized basophils can be activated by a rAra h 8 allergen and initiate an allergic inflammation despite a negative challenge. Hence, children sensitized to Ara h 8 but not to peanut storage proteins may be at risk for systemic allergic reaction when eating larger amounts of peanuts but most likely don’t have to fear smaller amounts.

Background and Aims

Although maize roots have been extensively studied, there is limited information on the effect of root exudates on the hydraulic properties of maize rhizosphere. Recent experiments suggested that the mucilaginous fraction of root exudates may cause water repellency of the rhizosphere. Our objectives were: 1) to investigate whether maize rhizosphere turns hydrophobic after drying and subsequent rewetting; 2) to test whether maize mucilage is hydrophobic; and 3) to find a quantitative relation between rhizosphere rewetting, particle size, soil matric potential and mucilage concentration.

Methods

Maize plants were grown in aluminum containers filled with a sandy soil. When the plants were 3-weeks-old, the soil was let dry and then it was irrigated. The soil water content during irrigation was imaged using neutron radiography. In a parallel experiment, ten maize plants were grown in sandy soil for 5 weeks. Mucilage was collected from young brace roots growing above the soil. Mucilage was placed on glass slides and let dry. The contact angle was measured with the sessile drop method for varying mucilage concentration. Additionally, capillary rise experiments were performed in soils of varying particle size mixed with maize mucilage. We then used a pore-network model in which mucilage was randomly distributed in a cubic lattice. The general idea was that rewetting of a pore is impeded when the concentration of mucilage on the pore surface (g cm?2) is higher than a given threshold value. The threshold value depended on soil matric potential, pore radius and contract angle. Then, we randomly distributed mucilage in the pore network and we calculated the percolation of water across a cubic lattice for varying soil particle size, mucilage concentration and matric potential.

Results

Our results showed that: 1) the rhizosphere of maize stayed temporarily dry after irrigation; 2) mucilage became water repellent after drying. Mucilage contact angle increased with mucilage surface concentration (gram of dry mucilage per surface area); 3) Water could easily cross the rhizosphere when the mucilage concentration was below a given threshold. In contrast, above a critical mucilage concentration water could not flow through the rhizosphere. The critical mucilage concentration decreased with increasing particle size and decreasing matric potential.

Conclusions

These results show the importance of mucilage exudation for the water fluxes across the root-soil interface. Our percolation model predicts at what mucilage concentration the rhizosphere turns hydrophobic depending on soil texture and matric potential. Further studies are needed to extend these results to varying soil conditions and to upscale them to the entire root system.

Background and Aims

Myxospermy is a term which describes the ability of a seed to produce mucilage upon hydration. The mucilage is mainly comprised of plant cell-wall polysaccharides which are deposited during development of those cells that comprise the seed coat (testa). Myxospermy is more prevalent among those plant species adapted to surviving on arid sandy soils, though its significance in determining the ecological fitness of plants is unclear. In this study, the first mathematical model of myxospermous seed mucilage expansion is presented based on seeds of the model plant species Capsella bursa-pastoris (shepherd''s purse).

Methods

The structures underpinning the expansion process were described using light, electron and time-lapse confocal micrographs. The data and experimental observations were used to create a mathematical model of myxospermous seed mucilage expansion based on diffusion equations.

Key Results

The mucilage expansion was rapid, taking 5 s, during which the cell mucilage volume increased 75-fold. At the level of the seed, this represented a 6-fold increase in seed volume and a 2·5-fold increase in seed surface area. These increases were shown to be a function of water uptake (16 g water g−1 mucilage dry weight), and relaxation of the polymers which comprised the mucilage. In addition, the osmotic pressure of the seed mucilage, estimated by assessing the mucilage expansion of seeds hydrated in solutions of varying osmotic pressure, was –0·54 MPa (equivalent to 0·11 m or 6·6 g L−1 NaCl).

Conclusions

The results showed that the mucilage may be characterized as hydrogel and seed-mucilage expansion may be modelled using the diffusion equation described. The potential of myxospermous seeds to affect the ecological services provided by soil is discussed briefly.  相似文献

Placoderm desmids (Conjugates, Chlorophyta) such as Closterium exhibit a gliding locomotory behavior. This results from the forceful extrusion of an acidic polysaccharide from one pole of the cell causing the cell to glide in the opposite direction. A biochemical and cytological analysis of gliding behavior was performed. The mucilage is a high molecular weight polysaccharide rich in glucuronic acid and fucose. Under normal growth conditions, 3 μg of mucilage is produced per cell in 30 days. Mucilage production increased 3–4 fold in cells challenged with low phosphate or nitrate conditions. A polyclonal antibody was raised against the mucilage and used in immunofluorescence studies. These results show that upon contact with another object Closterium aligns itself parallel to that object by a “jack-knife” motion. Subsequently, large amounts of mucilage are released to form elongate tubes enmeshing the cell with that object. In post-cytokinetic phases of the cell cycle, mucilage is extruded only through the pole of the developing semi-cell. Chlorotetracyclene-labeling of mucilage-secreting cells shows a correlation between calcium-rich loci on the cell surface and sites of mucilage release.  相似文献
