Estradiol has been shown to increase the level of thioredoxin mRNA in the uterus of the ovariectomized (ovx) rat. In this study the influence of progesterone, androgens, the anti-estrogen ICI 182780 and the anti-androgen Flutamid on thioredoxin expression, has been studied in the rat uterus. Thioredoxin mRNA concentrations were determined by solution hybridization. Ovx rats treated with progesterone alone showed no effect on thioredoxin expression. Combined treatment of ICI 182780 and estradiol attenuated the estradiol-induced increase in thioredoxin mRNA. When ovx rats were treated with a testosterone depot, the amount of thioredoxin mRNA was increased five-fold after 48 h and remained at that level during the rest of the 168 h monitored. A similar increase in thioredoxin mRNA could be seen after 5-dihydrotestosterone treatment, indicating a true androgenic effect. In addition, the anti-androgen Flutamid attenuated the thioredoxin mRNA increase seen after 5-dihydrotestosterone treatment alone.
It is concluded that thioredoxin mRNA is regulated by growth promoting gonadal steroids in the rat uterus. The attenuation of the estrogen and androgen-induced increases of the thioredoxin mRNA with ICI 182780 and Flutamid, indicate that the effect is mediated via the estrogen receptor and androgen receptor respectively. None of these hormones affected the hepatic thioredoxin mRNA level in the same animals. 相似文献

Gonadal steroids influence the morphology and function of neurons in the adult spinal cord through cellular and molecular mechanisms that are largely unknown. The cadherins are cell adhesion molecules that participate in the formation and organization of the CNS during embryonic development, and recent evidence suggests that the cadherins continue to regulate neural structure and function in adulthood. Using degenerate oligonucleotides coding conserved regions of the catenin-binding domain of classical cadherins in a RT-PCR cloning strategy, we identified several cadherin subtypes, the most frequently cloned being N-, E-, and R-cadherin, suggesting that these are the major classical cadherin subtypes present in the adult male rat lumbosacral spinal cord. We then examined cadherin expression levels of these cadherin subtypes under steroid conditions known to induce plastic changes in spinal motoneurons. Semiquantitative PCR revealed that mRNA levels of N-cadherin, but not E-cadherin or R-cadherin, are elevated in castrated rats treated with testosterone, 17 beta-estradiol, or dihydrotestosterone relative to castrate rats not treated with steroids. Immunolocalization of N-cadherin revealed that steroid treatment increased N-cadherin expression levels in functionally related neural populations whose morphology and function are regulated by steroids. These results suggest a role for N-cadherin in steroid-induced neuroplastic change in the adult lumbar spinal cord. 相似文献

Effects of gonadal steroids on conspecific odor preference for either (1) sexually active male or active female, (2) sexually active or gonadectomized (gdx) males, (3) sexually active or gdx females, and (4) gdx males or gdx females were determined in male and female rats in a three-chamber apparatus. For the first test, gdx females were made sexually active by treatments with estradiol benzoate (EB) and progesterone (P), and sexually active males were selected by prior screening. Sexually active males and females preferred opposite-sex odor over same-sex odor. Odor of sexually active opposite-sex conspecifics was preferred over that of inactive ones. Immediately after the completion of the first test, sexually active males were gdx and females were left without hormonal treatment. Second and third tests were carried out 2 and 5 weeks after the first test. In the second test, gdx males preferred odor of sexually active males rather than that of receptive females (male-directed preference); in the third test, both males and females showed no preference when tested with four stimulus pairs. The final tests were carried out in gdx males with EB and P, and gdx females with 2-week exposure to testosterone (T). Males with EB and P showed a male-directed preference again, whereas T-treated females kept their own female preference. Injection of EB alone to gdx males did not induce any preference. The present study clearly demonstrated sex difference in conspecific odor preference. Although both male and female preferences depend on their circulating sex steroids, the direction of male preference is more susceptible to their hormonal states, compared to that of females. 相似文献

The synthesis of polar aldosterone metabolites by rat liver microsomes at physiological concentrations of aldosterone (21.5 nM), was markedly inhibited by progesterone, testosterone, corticosterone, K+-canrenoate and estradiol-17 beta. In contrast, corticosterone and estradiol-17 beta significantly increased the synthesis of reduced aldosterone metabolites by 8- and 15-fold respectively, the majority of which were 5 alpha-reduced products of aldosterone. In experiments at higher substrate (aldosterone) concentrations (20-200 microM) the synthesis of ring A-reduced aldosterone metabolites by liver microsomes followed Michaelis-Menten kinetics with a Km[app] for aldosterone of 160 microM and Vmax[app] of 12.2 nmoles/mg protein/5 min. In these experiments progesterone, testosterone and K+-canrenoate all competitively inhibited the synthesis of reduced metabolites with inhibition constants (Ki [app]) of 70, 85 and 55 microM respectively; however, corticosterone did not. In contrast, estradiol-17 beta increased the rate of synthesis of reduced products by 40%, lowering the Km[app] to 83 microM. 相似文献

Evidence for sex-dependent anabolic response to androgenic steroids mediated by muscle glucocorticoid receptors in the rat 总被引:1,自引:0,他引:1
The muscle anabolic/anti-catabolic activity of the androgenic steroids testosterone and trenbolone was studied in rats to investigate whether such steroids act as agonists via muscle androgen receptors, or as antagonists that oppose the catabolic effects of endogenous glucocorticoids via their interaction with muscle glucocorticoid receptors. For comparison, the effects of the potent glucocorticoid antagonist RU486 were also examined. The parameters measured included growth rate, muscle weight, serum growth hormone and corticosterone levels, and receptor binding parameters in muscle cytosol. Females responded better than males to anabolic treatment with the androgenic steroids. Ovariectomy or adrenalectomy abolished this response. Neither the sex difference nor the requirement for ovaries or adrenals could be explained in terms of muscle receptor parameters or serum growth hormone levels. The muscle anabolic activity of androgenic steroids was restored when castrated males were treated with oestradiol and when adrenalectomized females were treated with corticosterone. RU486 also prevented the catabolic/anti-anabolic activity of exogenous corticosterone in adrenalectomized rats. Testosterone and RU486 behaved as anti-glucocorticoids in vivo since they inhibited glucocorticoid-induced liver tyrosine aminotransferase activity. The results suggest that anabolic steroids can act via muscle glucocorticoid receptors, thereby antagonizing the catabolic activity of endogenous glucocorticoids, rather than via muscle androgen receptors. 相似文献

The effects of various psychoactive drugs were studied in rats exposed for 45 min in a circularly polarized, pulsed microwave field (2450 MHz; SAR 0.6 W/kg; 2-microseconds pulses, 500 pps). Apomorphine-induced hypothermia and stereotypy were enhanced by irradiation. Amphetamine-induced hyperthermia was attenuated while stereotypy was unaffected. Morphine-induced catalepsy and lethality were enhanced by irradiation at certain dosages of the drug. Since these drugs have different modes of action on central neural mechanisms and the effects of microwaves depend on the particular drug studied, these results show the complex nature of the effect of microwave irradiation on brain functions. 相似文献

The effect of adrenalectomy on the basal cardiac c-AMP level and the elevated c-AMP level induced by intravenous administration of isoprenaline (10 mcg/kg) were determined by radio-protein assay. The basal cardiac c-AMP level was not altered by adrenalectomy. But the cardiac c-AMP responses to isoprenaline was significantly less in adrenalectomized rats, maintained with 1% sodium chloride solution for one week; this effect, however, disappeared when animals were maintained for 2 or 4 weeks. This could not be restored by acute administration of dexamethasone (100 mcg/rat). The possible reasons for these effects of adrenalectomy are discussed. 相似文献

The mechanism underlying the sexually dimorphic pattern of growth hormone (GH) secretion in the rat has not been clearly elucidated. In the present study, we assayed the possible direct effect of gonadal steroids on both somatostatin (SS) and growth hormone-releasing factor (GRF) in fetal rat hypothalamic cells in culture. Hypothalamic cells, obtained by mechanical dispersion, were maintained as monolayer cultures in serum-supplemented medium. After 20 days in culture, cells were incubated with serum free medium containing testosterone (T, 10, 20, 40 ng/dl) or estradiol (E, 0.1, 1, 10 ng/dl) for 48 h. At the end of the experiments, immunoreactive SS (IR-SS) and immunoreactive GRF (IR-GRF) were measured by specific radioimmunoassays (RIAs) in media and cell extracts. After 48 h of incubation with testosterone, somatostatin in both media and cells was significantly reduced. On the contrary, this treatment lead to a dose-dependent increase in media and cell GRF content. When cells were incubated with estradiol for 48 h, a significant inhibition in medium SS release was observed, whereas intracellular SS slightly increased at the highest concentration of 10 ng/dl. Estradiol treatment resulted in an inconsistent decrease in media and cells IR-GRF. Our results indicate that both SS and GRF are under the influence of testosterone and estradiol acting at the hypothalamic level, and furthermore suggest that at this stage of brain development, gonadal steroids may regulate GH secretion through their ability to modulate hypothalamic SS and GRF. 相似文献

Studies have provided inconsistent evidence regarding the effects on behaviour of nucleotide polymorphism at the DRD4 gene. For instance, the DRD4 gene affects exploratory behaviour in some bird species and populations but not in others. Our earlier experiment with a wild Great Tit Parus major population revealed an important impact of the DRD4 gene on behavioural variation during the breeding season. The aim of this study was to clarify these results by measuring individual behavioural differences (exploration and anxiety) in Great Tits under controlled conditions before the beginning of egg‐laying in order to exclude the potential impacts of breeding (egg‐laying, motivation to feed nestlings, the potential effect of a partner) and the natural environment (e.g. the presence of predators). Birds were caught from their natural habitat and their behaviour was tested in a trial room the following morning. Individuals with the CT heterozygous genotype (SNP830) performed more actively in different contexts (visiting a novel object in the trial room and vocal behaviour during handling) compared with homozygotes, and the relationships were consistent in males and females. In light of the results from our previous study (heterozygous males, but not females, had a shorter latency to feed nestlings than CC homozygotes), we conclude that the same DRD4 gene variants affect behavioural responses in a controlled environment and wild contexts in the same directions, but that these relationships are not sex‐specific under standardized conditions. 相似文献

Administration of gonadal steroids to neonatal rats has a profound effect on the function of the neuroendocrine system in the adult animal. Considering that gonadal steroids modulate hypothalamic and pituitary levels of beta-endorphin (BE) in adult male and female rats, the effects of neonatal gonadal steroid treatment on BE levels in the adult animal were investigated. Neonatal male rats were administered testosterone and neonatal female rats were treated with estrogen. Matched control littermates received vehicle. All animals were sacrificed at 90 days of age. Neonatal gonadal steroid treatment did not affect the level of immunoreactive beta-endorphin (IR-BE) in the anterior pituitary (AP) of male rats but did result in a significant increase in IR-BE in the AP of female rats. Neonatal administration of gonadal steroids produced a significant decrease in IR-BE in the neurointermediate lobe of the pituitary (NIL) of both male and female rats, with the magnitude of the decrease being greater in the NIL of the female rats. IR-BE levels in the hypothalamus of male or female rats were not altered by the treatments. Column chromatography indicated that the increase in IR-BE in the AP represented a proportional increase in BE and beta-lipotropin, while the reduction in IR-BE in the NIL of the treated rats represented a reduction in BE. These findings suggest that gonadal steroids may influence the development of the neurotransmitter systems which regulate BE levels in the adult pituitary, the development of the biosynthetic mechanisms of the adult pituitary, or both. 相似文献

We explored the effects of chronic alpha-linolenic acid dietary deficiency on serotoninergic neurotransmission. In vivo synaptic serotonin (5-HT) levels were studied in basal and pharmacologically stimulated conditions using intracerebral microdialysis in the hippocampus of awake 2-month-old rats. We also studied the effects of reversion of the deficient diet on fatty acid composition and serotoninergic neurotransmission. A balanced (control) diet was supplied to deficient rats at different stages of development, i.e. from birth, 7, 14 or 21 days of age. We demonstrated that chronic n-3 polyunsaturated fatty acid dietary deficiency induced changes in the synaptic levels of 5-HT both in basal conditions and after pharmacological stimulation with fenfluramine. Higher levels of basal 5-HT release and lower levels of 5-HT-stimulated release were found in deficient than in control rats. These neurochemical modifications were reversed by supply of the balanced diet provided at birth or during the first 2 weeks of life through the maternal milk, whereas they persisted if the balanced diet was given from weaning (at 3 weeks of age). This suggests that provision of essential fatty acids is durably able to affect brain function and that this is related to the developmental stage during which the deficiency occurs. 相似文献

Sex-change is an intriguing phenomenon that is common among certain groups of teleost fishes. The process itself has a number of independent origins, although in each case it is initiated and (or) regulated by gonadal steroids. Despite the commercial importance of sex-change technology to fish culturists, our understanding of the relationship between steroids and sex-change is, at best, rudimentary. In this paper I review the current state of knowledge concerning (a) which steroids are involved, (b) how such steroids mediate sex-change, and (c) how steroidogenesis is regulated during gonadal transition. I conclude that the steroidal endocrinology of sex-change is multifarious and species specific – a result which challenges the relative stability of vertebrate endocrine axes, but one which probably reflects the independent evolution of this adaptation. 相似文献

Superovulatory response in anestrous ewes is affected by the presence of a large follicle 总被引:3,自引:0,他引:3
Superovulatory response to conventional treatment with eCG (1200 IU) and progestagen sponges (MAP, n = 9; FGA, n = 9; or controls without sponge, n = 6) was studied in Corriedale anestrous ewes. The follicular population just before the administration of eCG and the total ovarian response (large anovulatory follicles plus normal CL and prematurely regressing CL) to treatment were determined after laparotomy. Pretreatment with progestagen did not modify the number or class of follicles greater than 1 mm observed on the ovarian surface at the time of eCG administration (19 +/- 2.2 follicles vs 19 +/- 2.9 follicles, for pooled progestagen-treated groups and control groups, respectively; mean +/- SEM) but significantly decreased the number of large anovulatory follicles (4.7 +/- 1.0 vs 10.2 +/- 2.6; P < or = 0.01) observed following treatment. Progestagen-treated animals were classified according to the presence (n = 13) or absence (n = 5) of a large follicle (LF: > or = 4 mm diameter) on the ovarian surface at the time of eCG treatment; a qualitatively better superovulatory response was observed in ewes without large follicle (large anovulatory follicles: 1.6 +/- 0.7 vs 5.8 +/- 1.3, P < or = 0.05; normal CL: 7.0 +/- 1.4 vs 3.8 +/- 1.0, P < or = 0.1; normal CL/total ovarian response: 78.7 +/- 10.1 % vs 34.9 +/- 8.2 %, P < or = 0.01; for ewes without LF and ewes with 1 to 2 LF respectively). No differences were observed in the individual ovulatory response when comparing ovaries ipsilateral or contralateral to LF in a same animal, indicating that the effect of LF on the superovulatory response would be fundamentally systemic. This work shows that, similar to what occurs in cows, the presence of a large follicle at the time of gonadotropin administration decreases the superovulatory response in anestrous ewes. 相似文献
