荧光适配体作为一种无需标记的荧光探针,具有许多潜在的优势,并被应用于多种靶物质(如ATP、RNA)的检测,是目前适配体研究领域的热点。孔雀石绿适配体(malachite green aptamer,MGA)属于荧光适配体,其能通过配体诱导折叠形成结合口袋,进而促进孔雀石绿(malachite green,MG)的发光。目前,已经筛选得到的MGA的种类较少,主要介绍了已知的MG RNA适配体及其变构体和MG DNA适配体的特性,以及影响MG-MGA复合物荧光强度的因素。同时,还对主要的MG衍生物和共聚物进行了总结。最后,综述了MGA在生物传感、荧光成像等方面的应用,并对MGA的发展方向进行了展望,以期为MGA在生物检测、生物成像等方面的应用提供指导。  相似文献

After briefly reviewing recent advances in the knowledge of various molecular species of interferon, some prospects of the basic research in this field are discussed: 1) interferon as a physiological hormone endowed with antiviral and immunomodulatory functions and possibly interacting with the neuroendocrine system; 2) interferon as an inducible protein, a model which allows a refined molecular analysis using the probes made available by cloning; 3) the various mechanisms of interferon action at the biochemical level. In the last section, the past and future of interferon as a therapeutic agent are examined. For its therapeutic evaluation, interferon needs to be compared with the best chemotherapeutic agents now available against viruses and tumors. Large scale clinical trials require a large production of interferons. Hence, a dilemma will be encountered: either pursue trials with natural preparations which are always a mixture of several types and subclasses of interferons in no defined proportions; wait for availability of the interferons produced by genetic recombinants which are cloned species, and try these various species, each one alone and in combination, or even more use hybrid molecular species. The availability of accurate markers for the action of interferon in man will be of great usefulness to study its therapeutic action and furthermore to define its indications. Finally, the possibility is raised that some non toxic interferon inducers, such as Poly A, Poly U, could also be used as well as interferon itself.  相似文献

We investigate the time evolution of disease spread on a network and present an analytical framework using the concept of disease generation time. Assuming a susceptible–infected–recovered epidemic process, this network-based framework enables us to calculate in detail the number of links (edges) within the network that are capable of producing new infectious nodes (individuals), the number of links that are not transmitting the infection further (non-transmitting links), as well as the number of contacts that individuals have with their neighbours (also known as degree distribution) within each epidemiological class, for each generation period. Using several examples, we demonstrate very good agreement between our analytical calculations and the results of computer simulations.  相似文献

Sleep is a physiological recuperative state susceptible to be disturbed or reduced in length under external stimuli, such as noise, one of the most frequent ambient factors. Sleep disturbance is often accompanied by a reduced quality of the following waking period. 基于微生物同化作用的D-丙氨酸生产工艺研究   总被引:1,自引:0,他引:1  
以L-丙氨酸为唯一碳氮源,从采集的若干土壤中初筛出能够降解L-丙氨酸的菌株;再以D-丙氨酸为唯一碳氮源,复筛出降解L-丙氨酸而不降解D-丙氨酸的菌株。依据菌种对DL-丙氨酸的不对称降解活性,筛选出具有最高的L-丙氨酸降解活性的菌株,并对菌株同化L-丙氨酸的反应条件进行了研究。结果表明:编号为ALA-D82的菌株具有最高的降解L-丙氨酸的能力,经鉴定为酵母菌属。在30℃,控制pH6.0,通气比1:1(V/V)和转速900 r.min-1的条件下,L-丙氨酸降解的速度最大。在最适条件下,1500 g DL-丙氨酸分两部分添加入7 L的反应液中。反应72 h后溶液中的L-丙氨酸被完全降解,提取得到D-丙氨酸晶体,产率和光学纯度分别达到92.13%和99%。  相似文献

以小偃-22和Amigo(美国)为对照对麦长管蚜在3个新引进小麦品种Astron、Batis和Xanthos(德国)上的抗蚜性进行了测定。在室内控制条件下,麦长管蚜在5个品种上的发育历期(DD)、体重差(dW)、相对日均体重增长率(MRGR)和生殖力(F)的测定结果表明,Astron、Xanthos与Amigo、小偃-22的抗性水平相当,均优于Batis;在大田自然条件下,通过对不同品种在不同生育期的感蚜指数以及感蚜量动态曲线的监测分析表明,Astron、Xanthos在我国的抗蚜水平与抗性品种Amigo的抗蚜性水平相当,优于Batis和小偃-22。可初步说明Astron、Xanthos在我国也具有较好的抗蚜性,可作为我国抗蚜育种材料。  相似文献

以自制的壳聚糖作为载体,用戊二醛作交联剂,优化了固定化条件,研制成壳聚糖固定化木瓜蛋白酶。其活性回收率达到42—53%,操作半衰期达到一个月以上,对热、乙醇以及尿素的稳定性有很大的提高,Km值为0.67×10~2mg/mL,最适温度65—70℃,最适pH8.0,能使啤酒中的蛋白质浓度从56.5mg/L减少到2.7mg/L,可以消除啤酒的低温混浊现象。  相似文献

Three additional phyletic group types, “periphyletic,” “epiphyletic”, and “anaphyletic” (in addition to Hennigian mono‐, para‐, and polyphyletic) are defined in terms of trees and phylogenetic networks (trees with directed reticulate edges) via a generalization of the algorithmic definitions of Farris. These designations concern groups defined as monophyletic on trees, but with additional gains or losses of members from network edges. These distinctions should be useful in discussion of systems with non‐vertical inheritance such as recombination between viruses, horizontal exchange between bacteria, hybridization in plants and animals, as well as human linguistic evolution. Maximum Reactive Distance (RDmax) and the area of the Reactive Perceptual Field (RPF) for the fish Macropodus cupanus were determined, using the fourth instar mosquito larva (Culex fatigans) as the prey organism. RDmax and the area of RPF increased with increasing body weight of the fish as well as increasing food deprivation time. Satiation time (St) as a function of the weight of the fish and aquarium temperature were also determined. In larger W classes (570 and 270 mg) St was not influenced by temperature. To get satiated the smallest W class (80 mg) required feeding periods of 40, 30 and 20 minutes at 22 or 27, 32 and 37°C, respectively.  相似文献

Summary Experiments with T. denitrificans have shown that this bacterium cannot develop in media devoid of ammonium salts, and that iron is required for growth. Pure cultures have been isolated by means of a modified medium which permits rapid growth in serial transfers. The organism has been characterized as an obligatory chemoautotroph which can oxidize a number of inorganic sulfur compounds (elementary sulfur, thiosulfate, tetrathionate) either aerobically with O2, or anaerobically with nitrate as oxidant.Nitrite was found to be highly toxic to the nitrate-reducing enzyme system; concentrations as low as 3.5 · 10-4 m inhibit denitrification in the presence of sulfur about 40%. In the presence of thiosulfate, nitrite is rapidly decomposed to NO; this gas can subsequently be used as an oxidant, and is reduced to N2. The formation of NO depends on the presence of both thiosulfate and nitrite.With resting cell suspensions CO2 assimilation has been established; the assimilation products are not merely carboxylation products, as demonstrated in experiments with C14O2.  相似文献

In a medium containing high concentration of yeast extract was observed aerobic formation of a large amount of ethanol by various yeasts of non-fermenting or extremely poor fermenting type such as Torulopsis famata, Candida polymorpha and Pichia membranaefaciens etc. Acetaldehyde formed as a metabolic intermediate leading to ethanol formation was trapped by the addition of sulfite and was identified as 2,4-dinitrophenyl-hydrazone.  相似文献

右旋糖酐蔗糖酶是一种以蔗糖为唯一底物,将蔗糖分子中D-葡萄糖基催化转移到受体分子上的葡萄糖基转移酶。利用右旋糖酐蔗糖酶的转糖基作用,以蔗糖为葡萄糖糖基供体,槲皮素为糖基受体,对槲皮素糖苷的酶法合成进行了探索。通过对该酶催化反应体系、催化反应条件及产物分析的研究,结果表明:在25℃下,右旋糖酐蔗糖酶能够在30%DMSO-70%乙酸-乙酸钙(0.02 mol/L,pH值5.4)的反应体系中催化合成一种槲皮素葡萄糖苷,在这个反应体系下,以10%的蔗糖作为糖基供体,槲皮素为糖基受体,右旋糖酐蔗糖酶活力为40 U/mL,转速为150 r/min,槲皮素糖苷的转化率最高,可达39.5%。通过质谱分析确定是一种槲皮素单糖苷,分子量为464。该研究结果为黄酮类物质的糖基化修饰奠定了基础。  相似文献

The effect of an additional carbon source, lactate, on Penicillium camembertii growth on glutamate as both carbon and nitrogen sources was examined. Glutamate (and lactate) was present in excess in both media. Throughout the whole culture, similar growth time-courses were recorded on both media, indicating the absence of a lactate effect on growth. During the first part of growth, corresponding to an increasing amount of viable biomass, the rate of glutamate consumption remained high, as well as the related ammonium production, indicating its use as a carbon source in addition to being nitrogen source. The low growth rates recorded during the last part of growth resulted in low glutamate consumption, while lactate consumption continued mainly by a maintenance mechanism for the energy supply. A clear differentiation appeared therefore between the carbon source and the energy source: glutamate was mainly used as C source (and N source) for biosynthesis, while lactate was mainly assimilated for energy supply. Carbon and nitrogen yield examinations confirmed this result. Indeed, the C/N ratio found for P. camembertii cellular material (8.14) was about twice that of glutamate (4.29). From this, about half of the available nitrogen was used for biomass formation during growth on glutamate-lactate based medium, as experimentally confirmed (constant yield nitrogen from biomass on nitrogen from glutamate was found (0.49), while the excess nitrogen was released as ammonium). The constant and close to unit (0.99) yield carbon from CO2 on carbon from lactate, also recorded during growth on glutamate-lactate based medium, confirmed that lactate was mainly used as an energy source.  相似文献

Vinblastine and colchicine induce the anucleate form of Trypanosoma gambiense. Light microscopic studies indicated that the anucleate form was not always produced as a result of inhibition of nuclear duplication, but was formed as a result of delay or inhibition of separation of the two nuclei after completion of nuclear division. Studies showed that vinblastine and colchicine caused disorder in arrangement of axonemal microtubules of the extracellular flagella and increased formation of both protofilaments and the axoneme composed of protofilaments in trypanosomes. Moreover, treatment with colchicine resulted in disintegration of previously existing pellicular microtubules and formation of cytoplasmic projections that appeared as protrusions from a small part of the surface membrane.  相似文献

Skeletal muscle is one of the few adult tissues that possesses the capacity for regeneration (restoration of lost functional tissue) as opposed to repair. This capacity is due to the presence of 'muscle stem cells' known as satellite cells. Detailed investigation of these cells over the past 50 years has revealed that both these and other cells within the skeletal muscle complex are capable of regenerating both muscle and other cell types as well. Here, we review this information, and suggest that skeletal muscle is an exciting reservoir of cells for regenerating skeletal muscle itself, as well as other cell types.  相似文献

受全球变暖和快速城市化的影响,城市可持续发展面临重大挑战。近10年,基于自然的解决方案(Nature-based Solutions,NbS)受到广泛关注与认可,为解决城市所面临的生态环境问题、提升城市韧性和可持续性提供了新的思路和途径。本文首先回顾了NbS的提出背景与发展历程,剖析了其核心内涵与框架准则,进而以城市中日益突出的内涝问题为例,介绍了NbS在应对和解决城市典型生态环境问题中的应用;通过汇总现有实施案例、整合多方观点,对比分析了NbS与国内外城市内涝管理的常用方法在概念框架、理论内涵和应用实践等方面的区别与联系;综合论述了基于NbS开展城市内涝管理所采用的技术手段、计量方法、评价模型的发展现状;最后,对NbS在我国的应用进行了展望,认为其在适应气候变化、提高城市韧性等方面具有巨大的潜力,但大范围的推广也面临理论认知尚不深入、实践过程存在障碍、专业人才与本土研究不足等多项挑战。  相似文献
