The spatial distribution of leaves is related to the exponent of the self-thinning relationship in plant populations. In this study, we evaluated the fractal dimension of rosette leaves of wild-type (WT)Arabidopsis thaliana and of an abscisic acid (ABA) -insensitive mutant (abi2-1) to test a model of the spatial distribution of leaf form in anArabidopsis population based on subdivision of a cube surrounding the leaf into uniform boxes and to investigate ABA’s affect on this model of the leaf. The values of the self-thinning exponent were -1.31 and -1.45 for WT andabi2-1. The mean dimensions of the box used to model the spatial distribution of leaf form, estimated using our model, were 2.08 and 2.03, respectively. By assuming that the box dimension equals the fractal dimension within the populations, the predicted self-thinning exponent equaled -1.40 for WT and -1.49 forabi2-1. When exogenous ABA was applied to both genotypes, the self-thinning exponent became -1.26 and -1.43 for WT andabi2-1, and the exponents predicted using the dimensions of the box were -1.37 and -1.46, respectively. The empirically predicted exponent equaled that predicted using the dimensions of the box (95% confidence interval). Empirical prediction of the spatial pattern using the two genotypes with and without ABA showed that ABA influenced the spatial form of the rosette leaves. Therefore, sensitivity to ABA can affect self-thinning through genetically determined changes in leaf form and its spatial distribution.  相似文献

The chlorinal-1 (ch1-1) mutant of Arabidopsis thaliana lacks the light-harvesting complexes in photosystem II (LHCII) due to deficiency of ability to synthesize chlorophyll (Chl) b. To investigate if a lack of LHCII affects plant growth and water loss, the Chl content, Chl fluorescence, glutathione (GSH) content, plant growth, water loss and stomatal aperture were measured using wild-type (WT) and ch1-1 mutant plants. The leaves of ch1-1 mutants accumulated significantly lower Chl content, Chl fluorescence and GSH content than WT plants. Plant growth and the leaf area of ch1-1 plants were also lower when compared to WT plants. The ch1-1 plant showed delayed flowering and higher a number of rosette leaves compared to the WT plants. The treatment of N-acetyl-cysteine increased Chl content and Chl fluorescence in leaves of both plants. Stomatal aperture was significantly lower in guard cells of the ch1-1 mutant than that of WT plants. Dark treatment increased stomatal closure which was corrected followed by the light treatment. Abscisic acid (ABA)-induced stomatal aperture was significantly lower in ch1-1 mutant than WT plants. Water loss through stomatal opening in ch1-1 plants was significantly lower than WT plants regardless of ABA treatment. This study suggests that a lack of LHCII might control plant growth and water loss in ch1-1 mutant of Arabidopsis thaliana.  相似文献

We have isolated an Arabidopsis mutant impaired in light- and brassinosteroid (BR) induced responses, as well as in sugar signalling. The bls1 (brassinosteroid, light and sugar1) mutant displays short hypocotyl, expanded cotyledons, and de-repression of light-regulated genes in young seedlings, and leaf differentiation and silique formation on prolonged growth in dark. In light, the bls1 mutant is dwarf and develops a short root, compact rosette, with reduced trichome number, and exhibits delayed bolting. The activity of the BR inducible TCH4 and auxin inducible SAUR promoters, fused with GUS gene, is also altered in seedlings harbouring bls1 mutant background. In addition, the bls1 mutant is hypersensitive to metabolizable sugars. The short hypocotyl phenotype in dark, short root phenotype in light and sugar hypersensitivity could be rescued with BR application. Moreover, the bls1 mutant also showed higher expression of a BR biosynthetic pathway gene CPD, which is known to be feedback-regulated by BR. Using a genome-wide AFLP mapping strategy, the bls1 mutant has been mapped to a 1.4Mb region of chromosome 5. Since no other mutant with essentially a similar phenotype has been assigned to this region, we suggest that the bls1 mutant defines a novel locus involved in regulating endogenous BR levels, with possible ramifications in integrating light, hormone and sugar signalling.  相似文献

Beta vulgaris is a facultative perennial species which exhibits large intraspecific variation in vernalization requirement and includes cultivated biennial forms such as the sugar beet. Vernalization requirement is under the genetic control of the bolting locus B on chromosome II. Previously, ethyl methanesulfonate (EMS) mutagenesis of an annual accession had yielded several mutants which require vernalization to bolt and behave as biennials. Here, five F2 populations derived from crosses between biennial mutants and annual beets were tested for co-segregation of bolting phenotypes with genotypic markers located at the B locus. One mutant appears to be mutated at the B locus, suggesting that an EMS-induced mutation of B can be sufficient to abolish annual bolting. Co-segregation analysis in four populations indicates that the genetic control of bolting also involves previously unknown major loci not linked to B, one of which also affects bolting time and was genetically mapped to chromosome IX.  相似文献

The transition from the vegetative rosette stage to the reproductive growth stage (bolting) in the rosette plant Eustoma grandiflorum has a strict requirement for vernalization, a treatment that causes oxidative stress. Since we have shown that reduced glutathione (GSH) and its biosynthesis are associated with bolting in another rosette plant Arabidopsis thaliana, we here investigated whether a similar mechanism governs the vernalization-induced bolting of E. grandiflorum. Addition of GSH or its precursor cysteine, instead of vernalization, induced bolting but other thiols, dithiothreitol and 2-mercaptoethanol, did not. The inductive effect of vernalization on bolting was nullified by addition of buthionine sulfoximine (BSO), an inhibitor of GSH synthesis, without decreasing the plant growth rate. BSO-mediated inhibition of bolting was reversed by addition of GSH but not by cysteine. These indicate that vernalization-induced bolting involves GSH biosynthesis and is specifically regulated by GSH. Plant GSH increased during the early vernalization period along with the activity of gamma-glutamylcysteine synthetase that catalyzes the first step of GSH biosynthesis, although there was little change in amounts of GSH precursor thiols, cysteine and gamma-glutamylcysteine. These findings strongly suggest that vernalization stimulates GSH synthesis and synthesized GSH specifically determines the bolting time of E. grandiflorum.  相似文献

Environmental stresses that perturb plant water relations influence abscisic acid (ABA) concentrations, but it is unclear whether long‐distance ABA transport contributes to changes in local ABA levels. To determine the physiological relevance of ABA transport, we made reciprocal‐ and self‐grafts of ABA‐deficient flacca mutant and wild‐type (WT) tomato plants, in which low phosphorus (P) conditions decreased ABA concentrations while salinity increased ABA concentrations. Whereas foliar ABA concentrations in the WT scions were rootstock independent under conditions, salinity resulted in long‐distance transport of ABA: flacca scions had approximately twice as much ABA when grafted on WT rootstocks compared to flacca rootstocks. Root ABA concentrations were scion dependent: both WT and flacca rootstocks had less ABA with the flacca mutant scion than with the WT scion under conditions. In WT scions, whereas rootstock genotype had limited effects on stomatal conductance under conditions, a flacca rootstock decreased leaf area of stressed plants, presumably due to attenuated root‐to‐shoot ABA transport. In flacca scions, a WT rootstock decreased stomatal conductance but increased leaf area of stressed plants, likely due to enhanced root‐to‐shoot ABA transport. Thus, long‐distance ABA transport can affect responses in distal tissues by changing local ABA concentrations.  相似文献

A barley (Hordeum vulgare L.) mutant (cool) with leaf transpiration unaffected by the application of 1 mM abscisic acid (ABA) was isolated from the population of M2 seedlings using thermography (electronic visualization, and quantitation of the temperature profiles on the surface of the leaves). Stomata of the mutant plants were insensitive to exogenously applied ABA, darkness, and such desiccation treatments as leaf excision and drought stress. The evaporative cooling of the leaves of the cool barley was always higher than that of the wild-type barley, even without ABA application, indicating that the diffusive resistance of the mutant leaves to water loss was always lower. Guard-cell morphology and stomatal density as well as ABA level and metabolism were seemingly unaltered in the mutant plants. Protein complex patterns of cloves and esterase isoenzyme patterns of apical buds of cloves were studied with Czechoslovak virus-free cultivars ofAllium sativum L. and the wild speciesA. longicuspis Regel, Similarly, four clones-regenerants obtained using explant culture techniques from A.sativum L. cv. Bzenecky paličák (two somaclones and two clones derived from plants regenerated from meristem cultures treatedin vitro with colchicine) differing in their ploidy, morphology, and yields were studied. Immunophoreograms of protein complexes of theA. sativum L. cultivars under investigation differed from one another in the number and mobility of protein fractions in both the cathodic and anodic regions and thus these cultivars can be distinguished. On the basis of esterase isoenzyme patterns, the Czechoslovak cultivars of A. sativum L. can be arranged into three groups - bolting winter cultivars with broad leaves, non-bolting winter cultivars with broad leaves, and non-bolting spring cultivars with narrow leaves. The responses of tobacco plants over-expressing trans-zeatin O-glucosyltransferase gene under constitutive or senescence-inducible promoter (35S:ZOG1 and SAG12:ZOG1) and of wild type (WT) plants to water stress and subsequent rehydration were compared. In plants sufficiently supplied with water, both transgenics have higher net photosynthetic rate (PN) in upper and middle leaves and higher stomatal conductance (gs) in middle leaves than WT. Water use efficiency (WUE = PN/E) was higher in both transgenics than in WT. During prolonged water stress, both PN and E declined to a similar extent in both transgenics and WT plants. However, 7 d after rehydration PN in SAG:ZOG (upper and middle leaves) and 35S:ZOG (upper leaves) was higher than that in WT plants. Increased content of endogenous CKs in 35S:ZOG plants did not prevent their response to ABA application and the results obtained did not support concept of CK antagonism of ABA-induced stomatal closure. The chlorophyll (Chl) a+b content was mostly higher in both transgenics than in WT. During water stress and subsequent rehydration it remained unchanged in upper leaves, decreased slightly in middle leaves only of WT, while rapidly in lower leaves. Total degradation of Chl, carotenoids and xanthophyll cycle pigments (XCP) was found under severe water stress in lower leaves. Carotenoid and XCP contents in middle and upper leaves mostly increased during development of water stress and decreased after rehydration. While β-carotene content was mostly higher in WT, neoxanthin content was higher in transgenics especially in 35S:ZOG under severe stress and after rehydration. The higher content of XCP and degree of their deepoxidation were usually found in upper and middle leaves than in lower leaves with exception of SAG:ZOG plants during mild water stress.  相似文献

Brassinosteroids (BR) are steroid phytohormones that are involved in the growth and stress response in plants, but the precise mechanisms of their action are still being discovered. In our study we have used BR-deficient barley mutants 522DK and BW084 (which carry missense mutations in the HvDWARF and HvCPD genes, respectively). We have also used a BR-signalling mutant that harbors missense substitutions in the HvBRI1 gene. Our aim was (1) to find out if the content of phytohormones in the mutants grown at 20 °C is different than in the wild types and whether/how the content of phytohormones changes after plant acclimation at temperatures of 5 °C and 27 °C?, (2) to characterise the effectiveness of the light reactions of photosynthesis of the barley mutants in comparison to wild types at various temperatures, and (3) to verify the impact of mutations on the tolerance of barley to high and low temperatures. Hormonal characteristics of the BR mutants of barley show the complexity of the interactions between BR and other plant hormones that are additionally modified by temperature and possibly by other factors. The results suggest the participation of BR in auxin catabolism. Further, BR appears to play a role in maintaining the ABA–ABAGlc balance. As for the gibberellin content in plants at a temperature of 20 °C, more in-depth studies will be required to explain the contradictory effects regarding the accumulation of GA3, GA4 and GA5, which appears to be dependent on the type of mutation and connected to the BR level. A fast-kinetic chlorophyll a fluorescence analysis has revealed that the mutants had lower values of energy absorption than the wild types, but the values of the energy transferred via the electron-transport chain was maintained at the wild-type level. We presumed that BR are involved in regulating plant acclimation to extreme (low/high) temperatures, thus the BR-deficient and BR-signalling mutants should be less tolerant to low/high temperatures when compared to the wild types. Unexpectedly, all of the mutants showed a higher tolerance to high temperatures than the wild types. The BW084 and BW312 mutants were less tolerant to frost than the wild type, but 522DK had a similar frost tolerance as the reference wild-type cultivar.

In this study, we examined the influence of UV-B radiation (280–320 nm) on ABA accumulation in 14-day-old Arabidopsis thaliana (L.) Heynh plants of wild type (WT), ethylene receptor mutant (etr1-1), and mutant with a constitutively active ethylene signal transduction pathway (ctr1-1). ABA content in nonirradiated WT plants was twice higher than in each mutant. UV-B irradiation caused dose-dependent ABA accumulation in WT plants. In the etr1-1 mutant, the amount of accumulated ABA was significantly less. In the ctr1-1 mutant, ABA content didn’t increase after UV-B irradiation. These data suggest that start of stress-induced ABA formation requires the adjustable ethylene signal pathway. In the ctr1-1 mutant, a constitutively active (nonadjustable) ethylene signal pathway blocks stress-induced ABA accumulation.  相似文献

To uncover the potential antioxidative role of anthocyanins in vivo in protecting photosynthetic tissues from photoinhibition, the effects of high irradiance [HI, 1300 μmol(photon) m−2 s−1] were studied using detached leaves derived from Arabidopsis wild-type (WT) and the mutant deficient in anthocyanin biosynthesis (tt3tt4). HI stress caused decreased chlorophyll content and photochemical efficiency, but increased cell-membrane leakage and contents of hydrogen peroxide and superoxide radical in the leaves of both Arabidopsis phenotypes, but the WT plants showed better HI tolerance than tt3tt4 mutant. HI caused a significant increase in the 1,1-diphenyl-2-picrylhydrazyl scavenging capacity in WT but not in the tt3tt4 mutant. The anthocyanins could not contribute substantially to light-shielding during the periods of HI stress, because the anthocyanin content in WT was very low and the colour of leaves was the same as in the tt3tt4 mutant. Thus, it was assumed that the better HI tolerance in WT was mostly related to the potential antioxidative role of anthocyanins.  相似文献

Bolting tendency in the crop species Beta vulgaris, which includes sugar beet, is a complex trait governed by various environmental cues, including prolonged periods of cold temperatures over winter (vernalization) and photoperiod, and multiple genetic factors. Two loci which promote bolting in the absence of vernalization are known in beet, the major bolting locus B on chromosome II and the B2 locus on chromosome IX. Here, genetic linkage and quantitative trait locus analyses in two populations derived from a cross between a biennial genotype, which was identified in a phenotypic screen for EMS-induced bolting mutants and requires vernalization to bolt, and an annual wild beet accession revealed the presence of a novel major bolting locus B4 which is linked to the B locus but promotes annual bolting independently of B. The genetic distance between B and B4 on chromosome II is 11 cM. A sequence-based marker was identified which co-segregates with bolting behavior and co-localizes with the B4 locus.  相似文献

To investigate the role of plant mitochondria in drought tolerance, the response to water deprivation was compared between Nicotiana sylvestris wild type (WT) plants and the CMSII respiratory complex I mutant, which has low-efficient respiration and photosynthesis, high levels of amino acids and pyridine nucleotides, and increased antioxidant capacity. We show that the delayed decrease in relative water content after water withholding in CMSII, as compared to WT leaves, is due to a lower stomatal conductance. The stomatal index and the abscisic acid (ABA) content were unaffected in well-watered mutant leaves, but the ABA/stomatal conductance relation was altered during drought, indicating that specific factors interact with ABA signalling. Leaf hydraulic conductance was lower in mutant leaves when compared to WT leaves and the role of oxidative aquaporin gating in attaining a maximum stomatal conductance is discussed. In addition, differences in leaf metabolic status between the mutant and the WT might contribute to the low stomatal conductance, as reported for TCA cycle-deficient plants. After withholding watering, TCA cycle derived organic acids declined more in CMSII leaves than in the WT, and ATP content decreased only in the CMSII. Moreover, in contrast to the WT, total free amino acid levels declined whilst soluble protein content increased in CMSII leaves, suggesting an accelerated amino acid remobilisation. We propose that oxidative and metabolic disturbances resulting from remodelled respiration in the absence of Complex I activity could be involved in bringing about the lower stomatal and hydraulic conductances.  相似文献

Using wild-type (WT) leaves and those from anore9 delayed-senescenceArabidopsis mutant, we investigated the delaying and accelerating effects of benzyladenine (BA) and abscisic acid (ABA), respectively, on the degradation process of the photosynthetic apparatus during dark-induced senescence (DIS). In the mutant, delays were seen for both the breakdown of chlorophyll (Chl) and the decrease in photochemical efficiency of photosystem II (Fv/Fm). Moreover, each step was prolonged in the disassembly process of the Chl-protein complexes. In the presence of BA, Chl degradation was retarded to a similar extent for both the mutant and the WT, but the decrease in Fv/Fm was not. However, in the presence of ABA, the two processes were accelerated in both genotypes. Therefore, although theore9 mutation causes this functional delayed-senescence, it may not be related to the non-functional delay that happens afterwards.
