Arundo donax (commonly called Giant Reed) is a perennial rhizomatous grass native to Asia, nowadays diffused all over the world. Due to its high biomass production and great adaptability to marginal land, interest in this species is increasing. In fact A. donax could represent an important and promising energy crop for heat and bioethanol second generation production. The propagation of A. donax is strictly agamic by rhizome fragmentation and cane node germination, strongly limiting the possibility of genetic improvement by breeding. The sterility could be caused by the fact that A. donax is a hybrid with uneven ploidy or a triploid species. It is difficult to propose an explanation for its sterility, because the chromosome number of A. donax is still a matter of debate, due to the high number and small size of the chromosomes; in the bibliography different counts ranging from 40 to 110 are reported. With the aim of establishing the chromosome number of A. donax we selected and counted 17 metaphase plates prepared from root tips obtained by hydroponic cultivation of cane nodes; our counts showed that A. donax most probably has 110 chromosomes. Our results suggested us two possible hypotheses, also based on SSR molecular marker results, concerning the evolutionary processes involved in the origins of A. Donax.  相似文献

Arundo donax L. has a high biomass production and a tendency toward community dominance in many habitats and thereby a tolerance to a wide range of environmental conditions. Therefore, the present study investigated the potentiality of A. donax to accumulate nutrients and trace metals in its biomass. Six main habitats (Nile Bank, Drain Bank, Canal Bank, Field Edges, Railways and Roadsides) were recognized. At each habitat, six quadrats (each 1 m2), distributed equally in two sites, were selected for growth measurements (e.g., density, shoot height, diameter, leaf area and biomass), plant and soil analyses. Plants from Nile, Canal and Drain Banks had the highest values of most growth measurements, while those from Railways and Roadsides had the lowest. Canal Bank plants accumulated the highest concentrations of P, Cu and Pb in their leaves; Zn in the stem; and Mg, Cd and Fe in the rhizome. The bioaccumulation factor (BF) of A. donax, for Cd, Fe, Mn and Zn, was greater than 1, while the translocation factor (TF) of most trace metals was less than unity in most habitats. In conclusion, A. donax showed morphological plasticity in response to habitat heterogeneity, and its growth was most vigorous in the riparian habitats. The high BF, as well as the significant positive correlations between trace metals, especially Cd, in soil and plant, renders A. donax a powerful phytoremediator.  相似文献

Yucca valida is a close relative to Yucca schidigera and a potential alternative source of the steroidal saponins extracted from the tissues of the latter. The exploitation from the wild is not only inefficient but endangers the natural populations and an alternative to these would be their domestication and establishment of commercial plantations. Both species have a long life span cycle and low growth rates affected by environmental conditions that make it difficult to select fast growing, high yield elite individuals for cloning purposes. Here, we report the in vitro establishment and propagation of mature Y. valida plants derived from seed that showed very different growth rates in an experimental field and their propagative and growth characteristics in vitro. No correlation was found between the growth of field cultivated plants and the relative increase of fresh (FW) or dry weight (DW) of in vitro cultured plants.  相似文献

The dependence of vegetative propagation on the production of individual plants was examined inDisporum smilacinum A. Gray on the basis of shading experiments and field surveys. This species typically showed four types of reproductive behavior: sterile plants producing one plantlet (no propagation), sterile plants producing more than two plantlets (vegetative propagation), fertile plants producing one plantlet (sexual reproduction) and fertile plants producing more than two plantlets (both sexual reproduction and vegetative propagation). The propagation ofD. smilacinum was clearly related to the annual net production of each individual plant. The probability of a mother plant producing more than two vegetative propagules (plantlets) increased with net production of the plant in the current year. The number of propagules per plant and runner length increased with net production. It was possible to explain the types of reproductive behavior of this species on the basis of both the initial plant size before sprouting and its net production during the growing season. There was a critical initial plant size for sexual reproduction and a critical level of production for vegetative propagation.  相似文献

The potential of the biocontrol agent Trichoderma harzianum strain T-203 to induce a growth response in cucumber plants was studied in soil and under axenic hydroponic growth conditions. When soil was amended with T. harzianum propagules, a 30% increase in seedling emergence was observed up to 8 days after sowing. On day 28, these plants exhibited a 95 and 75% increase in root area and cumulative root length, respectively, and a significant increase in dry weight (80%), shoot length (45%) and leaf area (80%). Similarly, an increase of 90 and 30% in P and Fe concentration respectively, was observed in T. harzianum inoculated plants. To better characterize the effect of T. harzianum during the early stages of root colonization, experiments were carried out in a gnotobiotic hydroponic system. An increased growth response was apparent as early as 5 days post-inoculation with T. harzianum, resulting in an increase of 25 and 40% in the dry weight of roots and shoots, respectively. Similarly a significant increase in the concentration of Cu, P, Fe, Zn, Mn and Na was observed in inoculated roots. In the shoots of these plants, the concentration of Zn, P and Mn increased by 25, 30 and 70%, respectively. Using the axenic hydroponic system, we showed that the improvement of plant nutritional level may be directly related to a general beneficial growth effect of the root system following T. harzianum inoculation. This phenomenon was evident from 5 days post-inoculation throughout the rest of the growth period, resulting in biomass accumulation in both roots and shoots.  相似文献

Given its high biomass and plasticity, Arundo donax L. is a promising ligno-cellulosic crop for cultivation in marginal lands in temperate climates. In order to test for adaptation to salinity, growth parameters of several A. donax clones were evaluated under two salt regimes in hydroponics. Mild NaCl stress (50 mM NaCl, 5.6 mS cm?1 EC, for 10 days) failed to discriminate between ecotypes, while a more severe NaCl treatment (150 mM, 18.8 mS cm?1 EC, for 21 days) enabled the identification of ecotypes maintaining plant growth under high salinity. Among several biometric parameters, 4th leaf width, and shoot and root DW consistently highlighted differences between ecotypes. Gas-exchange parameters also responded to severe NaCl treatment, while the photosystem efficiency was good, regardless of treatment. The results confirm that A. donax can be considered moderately tolerant to NaCl stress, with variation between ecotypes. Our screening protocol identified ecotypes with higher biomass production under severe NaCl treatment and can be useful for preliminary evaluation of NaCl tolerant clones for increasing productivity under salinity. The detected inter-ecotype variability could also be investigated to identify suitable clones for different environments.  相似文献

Biometric and physiological analyses of salt stress responses were performed in two time-course experiments on giant reed (Arundo donax L). Experiment I evaluated biomass production in plants exposed to 128, 256, 512 mM NaCl for 84 days. For Experiment II, plants grown under 256 mM NaCl were further assessed for chlorophyll a fluorescence, ionic partitioning, and proline content at 14 and 49 days after treatment (DAT). Biomass allocation was affected with all the concentrations of NaCl used from 28 DAT onward. Proline biosynthesis in leaves was more stimulated than that in roots after salt stress. Photosynthetic efficiency of photosystem II (PSII) was not affected by salt stress up to 42 DAT, while 49 DAT plants exhibited a significant reduction of both potential (ΦPSII) and maximal (Fv/Fm) PSII quantum yield. A. donax resulted a moderately sensitive species in response to 256 and 512 mM NaCl, concentrations that are however higher than that commonly found in most marginal lands (such as 128 mM or lower), where the biomass yield is appreciable, especially in short-term cultivation (56 DAT here). Altogether, this study indicates that A. donax can be considered as a promising and valuable energy crop for exploiting the Mediterranean marginal land.  相似文献

The demand for perennial nonfood crops, such as miscanthus, is increasing steadily, as fossil resources are replaced by biomass. However, as the establishment of miscanthus is very expensive, its cultivation area in Europe is still small. The most common propagation method for miscanthus is via rhizomes, the harvesting of which is very labour‐intensive. Seed propagation is promising, but not suitable for sterile genotypes. In this study, a new vegetative propagation method, ‘collar propagation’, was tested in field and controlled environment studies. Collars are built at the junction between rhizome and stem. They can be harvested in a less destructive way than rhizomes by pulling out the stems from winter‐dormant miscanthus plants. One genotype of each of the species M. sacchariflorus, M. × giganteus, M. sinensis in combination with three fragment types (collars, rhizomes, collars + rhizomes) were tested for establishment success and plant performance. The performance (e.g. dry matter yield) of collar‐propagated plants was either better than or not significantly different from rhizome‐propagated plants. Pregrown plantlets transplanted into the field showed no significant differences in establishment success between the fragments within a genotype. When directly planted into the field however, the fragment ‘rhizome+collar’ had a significantly better establishment success than the other two. The winter survival rate of the fragment ‘rhizome+collar’ was 70% for M. sacchariflorus and 75% for M. × giganteus. Emergence success from collar‐derived plants was not affected by harvest date (harvested monthly from November to February). This study showed that miscanthus propagation via collars is feasible and a promising alternative to rhizome propagation, as the multiplication rate of collars is comparable to that of rhizome propagation. Collar propagation is the more suitable method for the tested genotypes of the species M. sachariflorus and M. × giganteus, but not for M. sinensis genotypes, which may be better propagated by seeds.  相似文献

该研究采用盆栽试验法,选取石菖蒲、水生鸢尾、美人蕉、花叶芦竹为试验材料,将石菖蒲、水生鸢尾、美人蕉与花叶芦竹混种,用花叶芦竹单种作为对照,研究了混种对花叶芦竹生物量、根冠比以及铅积累量的影响。结果表明:(1)在不同浓度铅污染处理下,与花叶芦竹单种相比,混种模式增加了花叶芦竹的根冠比,且随着铅污染浓度的增加,表现出先上升后下降的趋势。(2)在单种和混种模式下花叶芦竹体内和土壤内的铅含量均表现出明显的浓度积聚效应;在混种模式下植物体内和土壤内的铅含量明显低于单种模式,且与单种模式之间的差异性显著(P0.05)。(3)在高浓度铅污染处理下,单种和混种模式的花叶芦竹铅转运系数均大于1,而富集系数则表现为在低浓度和高浓度铅污染处理下均大于1;在美人蕉+花叶芦竹的混种模式中,花叶芦竹的地上部分和地下部分的铅富集系数达到最大。因此,我们认为美人蕉+花叶芦竹混种模式,显著提高了花叶芦竹的生物量和根冠比,并对铅污染土壤的净化效果最佳。  相似文献

Arundo donax L., common name giant cane or giant reed, is a plant that grows spontaneously in different kinds of environments and that it is widespread in temperate and hot areas all over the world. Plant adaptability to different kinds of environment, soils and growing conditions, in combination with the high biomass production and the low input required for its cultivation, give to A. donax many advantages when compared to other energy crops. A. donax can be used in the production of biofuels/bioenergy not only by biological fermentation, i.e. biogas and bio-ethanol, but also, by direct biomass combustion. Both its industrial uses and the extraction of chemical compounds are largely proved, so that A. donax can be proposed as the feedstock to develop a bio-refinery.  相似文献

A cost-effective and efficient protocol has been described in the present work for large scale and rapid in vitro propagation of a valuable medicinal herb Bacopa monnieri (L.) by shoot bud proliferation on internodal segments isolated from field grown mature plants. This shooting was achieved on low concentration of cytokinin with BAP 1.0 mg/l and Kn 0.5 mg/l within 3 weeks of inoculation contrary to earlier reports where auxin is also used in combination with cytokinin. Normally internodes gives rise to a clump of shoots (18 shoots). On an average within a period of three subcultures the internode explant generated 324 shoots thereby improving the economics of the cost of the plants produced and time required. For rooting this is a first report of its growth in a liquid culture. The regenerated plants resemble the mother plants in the general habitat without any morphological variation. This paper reports a very simple, reproducible procedure for in vitro propagation of Bacopa monnieri (L.) that can be used to form a foundation stock of elite plant material for large-scale cultivation.  相似文献

The effect of feeding by the armored scale, Rhizaspidiotus donacis (Leonardi, 1920) (Hemiptera: Diaspididae) on the growth of the plant Arundo donax L. (Poaceae) was evaluated under field conditions in its native range. The study was designed to evaluate the impact of R. donacis, a candidate agent for biological control of A. donax which is invasive in arid riparian ecosystems of the Southwestern USA and Mexico. The study was carried out at five A. donax sites in the Province of Alicante, Spain, differing in altitude and climate. At each site, 30 infested lateral shoots were selected and 15 were randomly treated monthly with imidacloprid insecticide. Shoot lengths were measured monthly over a 1-year period in a comparative growth analysis. Shoots infested with R. donacis had an over 2-fold reduced growth rate as compared to treated shoots. Growth of shoots varied by site, and the effect of R. donacis on growth was most pronounced in the late spring, when mature females produced first instar scale crawlers. The impact of R. donacis on A. donax growth under field conditions in the native range, combined with its narrow host specificity, indicate that R. donacis is a promising candidate for biological control of A. donax in North America and other areas invaded by this weed.  相似文献

A 2-year field experiment was performed to assess regeneration, establishment and growth of the invasive polycarpic perennial Brassicaceae Bunias orientalis L. from transplanted juveniles and propagules (seeds, fruits and root fragments). Further treatments were: different planting depths of the propagules, different root fragment sizes and a varied competition intensity by the matrix vegetation. In contrast to a high number of individuals established from planted juveniles (ca. 90 %), only relatively few propagules gave rise to seedlings or regenerating plants (0–16 %). However, upon emergence, all plants showed low mortality. Although the number of regenerated or recruited plants was affected by all treatments, only one treatment level (the use of very small root fragments of 1 cm length) completely failed in establishing plants. Plants experiencing low competition intensity attained high aboveground performance and reproductive output as well as reproductive success within two growth periods. Under high competition intensity recruitment, regeneration and growth were strongly negatively affected. The high morphological plasticity of rosette growth of B. orientalis seems to be advantageous for persistence in such situations. The results show that young B. orientalis plants are robust and can efficiently sequester and translate elevated resources into rapid growth and early reproductive output although the species is a rather long-lived iteroparous perennial. In addition, population foundation or regeneration can be based on both seeds and root fragments, even when buried under a thick soil layer. This seems to be a very efficient adaption to habitats which are subject to anthropogenic soil perturbations.  相似文献

Plant tissue culture technology is being widely used for large-scale, rapid, clonal multiplication and genetic transformation in cassava. The main limitation of this technology is the period of acclimation of the fragile in vitro plants after they have been multiplied or regenerated. Most losses of in vitro plants occur when the plantlets are moved directly from the test tubes to the ex vitro conditions. Our aim was to design a simple, rapid, low-maintenance hydroponic system to improve survival rate of transplanting to the ex vitro conditions through the rapid acclimation process of in vitro plants. In this paper, we have developed a simple hydroponic system to accelerate the cassava acclimation and multiplication process. This system considerably increased the survival percentage of in vitro and/or transgenic lines and reduces the time requirement for multiplication by hydroponic acclimation. In order to assess the effectiveness of the acclimation of seedlings on their establishment, we analyzed plant growth and field survival rate with response to different nitrogen (N) sources using different cassava accessions. Nitrogen sources of NO3 ? and NH4NO3 increased plant growth and root length compared to NH4 + alone, or water treatments. The greenhouse and field survivability of N-hardened plants, including transgenic lines, were significantly different in growth and development. We present a simple NO3 ? hydroponic acclimation system that can be quickly and cheaply constructed and used by the cassava community around the world. The efficiency of our proposed N hydroponic acclimation system is validated in the transgenic development pipeline which will enhance the cassava molecular breeding.  相似文献

Widespread invasion of riparian ecosystems by the large bamboo-like grass Arundo donax L. has altered community structure and ecological function of streams in California. This study evaluated the influence of wildfire on A. donax invasion by investigating its relative rate of reestablishment versus native riparian species after wildfire burned 300 ha of riparian woodlands along the Santa Clara River in southern California in October 2003. Post-fire A. donax growth rates and productivity were compared to those of native woody riparian species in plots established before and after the fire. Arundo donax resprouted within days after the fire and exhibited higher growth rates and productivity compared to native riparian plants. Arundo donax grew 3–4 times faster than native woody riparian plants—up to a mean of 2.62 cm day−1—and reached up to 2.3 m in height less than 3 months after the fire. One year post-fire, A. donax density was nearly 20 times higher and productivity was 14–24 times higher than for native woody species. Three mechanisms—fire-adapted phenology, high growth rate, and growth response to nutrient enrichment—appear to promote the preemption of native woody riparian species by A. donax after fire. This greater dominance of A. donax after wildfire increased the susceptibility of riparian woodlands along the Santa Clara River to subsequent fire, potentially creating an invasive plant-fire regime cycle. Moreover, A. donax infestations appear to have allowed the wildfire to cross the broad bed of the Santa Clara River from the north, allowing thousands of acres of shrubland to the south to burn.  相似文献

Giant reed (Arundo donax L.) is a perennial rhizomatous grass, which has attracted great attention as a potential lignocellulosic feedstock for bioethanol production due to high biomass yield in marginal land areas, high polysaccharide content and low inhibitor levels in microbial fermentations. However, little is known about the trait variation that is available across a broad ecotypic panel of A. donax nor the traits that contribute most significantly to yield and growth in drought prone environments. A collection of 82 ecotypes of A. donax sampled across the Mediterranean basin was planted in a common garden experimental field in Savigliano, Italy. We analysed the collection using 367 clumps representing replicate plantings of 82 ecotypes for variation in 21 traits important for biomass accumulation and to identify the particular set of ecotypes with the most promising potential for biomass production. We measured morpho‐physiological, phenological and biomass traits and analysed causal relationships between traits and productivity characteristics assessed at leaf and canopy levels. The results identified differences among the 82 ecotypes for all studied traits: those showing the highest level of variability included stomatal resistance, stem density (StN), stem dry mass (StDM) and total biomass production (TotDM). Multiple regression analysis revealed that leaf area index, StDM, StN, number of nodes per stem, stem height and diameter were the most significant predictors of TotDM and the most important early selection criteria for bioenergy production from A. donax. These traits were used in a hierarchical cluster analysis to identify groups of similar ecotypes, and a selection was made of promising ecotypes for multiyear and multisite testing for biomass production. Heritability estimates were significant for all traits. The potential of this ecotype collection as a resource for studies of germplasm diversity and for the analysis of traits underpinning high productivity of A. donax is highlighted.  相似文献

The grey mangrove, Avicennia marina, grows in intertidal habitat that is under pressure from expanding human settlement and industry along coastlines in the tropics and subtropics. Inappropriate clearing, and also the dieback of large stands of A. marina associated with pollution, have created an interest in methods for revegetation and also the need for a better understanding of the species physiological tolerances including to herbicide residue. Eco-toxicological studies have so far been exclusively on seedlings because of perceived difficulties cultivating mature plants in controlled environments. Reforestation projects are often associated with low rates of establishment, because of the susceptibility of propagules, seedlings and saplings to wind and wave erosion, as well as flooding and desiccation associated with the natural inter-tidal environment. We report on the successful excavation, and cultivation in containers with an automated irrigation system of eight mature A. marina plants. It is possible to transplant mature mangroves and achieve significant vegetative growth and also flowering and fruiting within the first year. It is difficult to compare the likely costs of transplanting a more limited number of mature plants to many propagules. However, we suggest that in situations where reforestation with young propagules, seedlings or saplings has failed because of excessive wash, drowning or smothering, the transplanting of mature mangroves could be a better option.  相似文献

Worldwide, many medicinal and aromatic plants (MAPs) are still collected from the wild and only a small fraction of them are exclusively sourced from cultivation. This practice when performed non-sustainably threatens species and populations. Micropropagation of MAPs is a powerful tool to conserve rare, threatened, and valuable MAPs, and to massively produce high-value plant material for cultivation without seasonal constraints. In this study, the in vitro propagation protocols of 22 Greek native MAPs assigned with conservation priority were assessed (herbaceous perennials, bulbous, subshrubs, and trees), including 17 range-restricted plants and 5 taxa of Orchidaceae. For the latter, current micropropagation efforts include seed germination, callus induction, and protocorm formation for successful plantlet development; however, these propagation protocols are still fragmentary. For the rest (n = 17), a five-stage detailed procedure is outlined (plant material, establishment, proliferation, rooting, and acclimatization), while materials, treatments, and data per stage are shown comparatively and discussed. Emphasis is given on the selection and preparation of plant material obtained from nature for research, sustainable use, and ex situ conservation actions, and on their effectiveness for conservation purposes or mass production needs. The protocol effectiveness was calculated using a specific equation to estimate the potential number of acclimatized plants raised from a single explant within a year. All protocols can facilitate conservation, and almost half of them could be used for commercialization with high cost (five cases), intermediate cost (eight), or low cost (four), which enables their possible sustainable use.

Studying Arabidopsis thaliana (L.) Heynh. root development in situ at the whole plant level without affecting shoot development has always been a challenge. Such studies are usually carried out on individual plants, neglecting competition of a plant population, using hydroponic systems or Agar-filled Petri dishes. Those both systems, however, present some limitations, such as difficulty to study precisely root morphogenesis or time-limited culture period, respectively. In this paper, we present a method of Arabidopsis thaliana (L.) Heynh. cultivation in soil medium, named “Ara-rhizotron”. It allows the non-destructive study of shoot and root development simultaneously during the entire period of vegetative growth. In this system, roots are grown in 2D conditions, comparable to other soil cultures. Moreover, grouping several Ara-rhizotrons in a box enables the establishment of 3D shoot competition as for plants grown in a population. In comparison to a control culture grown in pots in the same environmental conditions, the Ara-rhizotron resulted in comparable shoot development in terms of dry mass, leaf area, number of leaves and nitrogen content. We used this new culture system to study the effect of irrigation modalities on plant development. We found that irrigation frequency only affected root partitioning in the soil and shoot nitrogen content, but not shoot or root growth. These effects appeared at the end of the vegetative growth period. This experiment highlights the opportunity offered by the Ara-rhizotron to point out tardy effects, affecting simultaneously shoot development and root architecture of plants grown in a population. We discuss its advantages in relation to root development and physiology, as well as its possible applications.  相似文献

Five years post-release of the arundo gall wasp, Tetramesa romana, into the riparian habitats of the lower Rio Grande River, changes in the health of the invasive weed, Arundo donax, or giant reed, have been documented. These changes in plant attributes are fairly consistent along the study area of 558 river miles between Del Rio and Brownsville, TX, and support the hypothesis that the arundo wasp has had a significant impact as a biological control agent. Plant attributes were measured prior to release in 10 quadrats at each of 10 field sites in 2007, and measured again at the same undisturbed sites, 5 years after the release of T. romana, in 2014. Above ground biomass of A. donax decreased on average by 22% across the 10 sites. This decline in biomass was negatively correlated to increased total numbers of T. romana exit holes in main and lateral shoots per site in 2014 compared to 2007. Changes in biomass, live shoot density and shoot lengths, especially the positive effect of galling on main and lateral shoot mortality, appear to be leading to a consistent decline of A. donax. Economically, this reduction in A. donax biomass is estimated to be saving 4.4 million dollars per year in agricultural water. Additional impacts are expected as populations of the wasp increase and as other biological control agents such as the arundo scale, Rhizaspidiotus donacis, become more widespread.  相似文献
