BackgroundOlive-trees (Olea europaea L.) are the dominant rustic trees cultivated in the Mediterranean agricultural zones. Major and micronutrients play an indispensable role in their plant physiological functions although; the effect of trace elements on metabolic processes has not been sufficiently investigated, especially in olive-trees.MethodsIn the current study, we have used X-ray fluorescence (XRF) spectrometry to determine selected major and trace elements (Br, Cu, Fe, K, Mn, P, Rb and Zn) in the main olive cultivar cultivated in Algeria, cv.‘Sigoise’. Certified reference materials viz. IAEA-336 (Lichen) and NIST-1646a (Estuarine sediment) were evaluated simultaneously with the soil and plant samples for quality control of the analytical method.ResultsThe results show that Fe and Mn concentrations were superior in leaves than fruits. However large amounts of K, Cu and Rb were accumulated in the olive-fruits. The contents of all chemical elements were above the threshold limits for possible plant nutrient deficiencies, except for P whose concentration was in borderline requirement of olive trees. High values of a translocation factor index were found for K, Cu and Rb (TFs > 4). Principal component analysis (PCA) indicated that K was highly related with olives-fruits, suggesting that the fruit was the principal organ of K storage. Furthermore, dietary element intake through consuming olives was also estimated and compared to recommended daily intakes (RDIs) and daily permissible limits (DPLs). The estimations of chemical element intakes were below the DPLs set by WHO/FAO guidelines for human nutrition.ConclusionThe present work indicates that the concentrations of macro- and microelements (Cu, Fe, K, Mn and Zn) were above the threshold limits for possible plant deficiencies except for P, and this cultivar can easily accumulate high amount of K in their organs (predominance in olives). These findings will be used to achieve efficient fertilization for O. europaea orchards.  相似文献

Water relations are a key factor limiting olive production. In this study, effects of plating density on physiological aspects and productivity of ‘Chemlali’ olive trees were analyzed under rain-fed conditions in four planting densities (156, 100, 69 and 51 trees ha−1), in an experimental olive orchard located in the center of Tunisia. Seasonal changes in leaf relative water content (RWC), leaf water potential, stomatal conductance (g s), CO2 assimilation rate and tree production were studied. Accompanying the changes in leaf water status, all the monitored trees reduced leaf stomatal conductance (g s) and photosynthetic rate (A) throughout the summer drought, mirroring the increase in soil moisture deficit and vapor pressure deficit. However, the decrease in gas exchange was much more pronounced in high planting densities than in low ones. Our results confirm that the increase of tree-to-tree water competition with planting density was significant in the dry climate of Tunisia. Thus, planting density is critical when planting new olive orchards in arid regions.  相似文献

Olea europaea L. subsp. laperrinei (Oleaceae) is an endemic taxon of the mountainous regions of central Sahara, consisting of currently fragmented and small relict populations. The tree can propagate vegetatively or by seed, but no recent natural regeneration was observed in the Algerian massive populations, some of which are considered threatened with extinction. Sterile triploid individuals were also identified in some populations showing increasing vigour. As a result of its long persistence and despite its rarity, the Laperrine’s olive is an iconic component of Saharan mountain ecosystems. The aim of this study is to develop an efficient micropropagation protocol for both diploid and triploid Laperrine’s olive to safeguard and preserve this genetic resource. Best shoot propagation was obtained on a modified Murashige and Skoog medium supplemented with 9.2 μM zeatin. Best rooting rate of regenerated shoots was achieved on the same culture medium supplemented with 4.8 μM indole-3-butyric acid. In absence of morphological changes of in vitro regenerated plants acclimatized to the greenhouse, genetic conformity was assessed by simple sequence repeat screening. Our results suggest that in vitro propagation could be a useful tool for conservation of both diploid and triploid threatened Laperrine’s olive.  相似文献

The chlorosis susceptible Vitis vinifera L. cv. Pinot blanc was grafted on two hybrid rootstocks with different iron efficiency, as follows: V. Berlandieri × V. rupestris 140 Ru (iron-efficient) and V. riparia × V. rupestris 101-14 (iron-inefficient). The grafted vines were grown in pots of a calcareous and a non-calcareous soil. The shoot growth was periodically checked and leaves, selected at two different times (at the middle of the annual growing period), were assayed for total chlorophyll, ferrous iron, ash alkalinity, percentage of dry matter and chlorosis score. At the end of the growing cycle the roots were oven-dried and weighed. The most significant findings of the trial were: (a) the soil strongly affected the shoot growth, with canes about twice as long in the non-calcareous soil; (b) the iron-efficient rootstock (140 Ru) did not induce chlorosis when growing on the calcareous soil, while the opposite occurred with the iron-inefficient rootstock (101=14); and (c) a high ash alkalinity occurred in light chlorotic leaves compared to green ones, under the same iron concentration.  相似文献

A set of 14 SCAR markers were developed starting from RAPD, AFLP and SAMPL analysis of several olive germplasm accessions. Eight RAPD, two AFLP and four SAMPL fragments were converted into dominant and codominant SCARs by cloning and sequencing the selected fragments. The markers obtained were evaluated on forty different olive cultivars from different Italian production areas (mainly from Liguria). The combined use of these SCARs made possible to univocally identify 26 cultivars while the remaining 14 will require the development of further markers since most of them are placed in a main group containing six genetically similar cultivars (among which Frantoio and Taggiasca) and four minor groups containing two cultivars each. A total of 31 different haplotypes were identified and the analysis of several individual plants indicated no intra-cultivar variability. Considering the SCAR polymorphism two alleles were scored for each markers with the only exception of markers IGPS3 and IGPS4 showing 4 alleles with 7 recognised groups and 5 alleles with 4 groups, respectively. Though less polymorphic in comparison with other markers like SSRs, the developed SCARs proved useful in genotype identification. The effect of irrigation intervals was studied on physiological, morphological, and antioxidant traits of two marigold (Calendula officinalis L.) cultivars in Karaj, Iran, in a split-plot experiment based on a randomized complete block design with three replications. The experimental treatments included irrigation at three levels of I1 (irrigation interval of 3 days), I2 (irrigation interval of 5 days), and I3 (irrigation interval of 7 days) as the main plot and cultivar at two levels of V1 (cv. ‘sparse petal’) and V2 (cv. ‘compact petal’) as the sub-plot. The results based on the comparison of the means showed that the increase in irrigation interval from 3 to 7 days decreased the leaf area index, crop growth rate, relative growth rate, and net assimilation rate by 73.53, 85.76, 93.47, and 94.81%, respectively. It also decreased the flower yield, plant height, flower number, and leaf number by 71.92, 41.84, 99.31, and 58.67%, respectively. The interaction between irrigation and cultivar revealed that I3V1 had the highest total phenol content (3013.59 g gallic acid 100 g?1 tissue) and antioxidant capacity (60.8%). It can be inferred that the treatment of I1 and cv. ‘compact petal’ give the best results for flower yield and physiological and morphological traits, and the treatment of I3 and both cultivars provide the best results for antioxidant traits in the climatic conditions of the Karaj region.

The effects of humic substances on in vitro culture of Golden Delicious apple are reported. Potassium humate (KH) when used in proliferation showed a negative interaction with BA while it enhanced rooting when IBA was not present in the culture medium. In the presence of IBA, KH increased root number and reduced root growth. The highest concentration tested, 500 mg l-1, caused a drastic reduction in root system development. 50 mg l-1 KH hastened rooting and plants grew more rapidly when transferred to soil.  相似文献

The main goal of the study was to determine the structure of endophytic bacteria inhabiting different parts (endosperm, germ, roots, coleoptiles, and leaves) of two wheat species, Triticum aestivum L. (cv. ‘Hondia’) and Triticum spelta L. (cv. ‘Rokosz’), in order to provide new knowledge about the stability and/or changeability of the core microbiome in different plant organs. The endophytic core microbiome is associated with plants throughout their whole life cycle; however, plant organs can determine the actual endophytic community. Therefore, next generation sequencing with MiSeq Illumina technology was applied to identify the endophytic microbiome of T. aestivum and T. spelta. Bioinformatic analyses were performed with the use of the DADA2(1.8) package and R software (3.5.1).It was demonstrated that wheat, which is an important crop plant, was associated with beneficial endophytic bacteria inside the endosperms, germs, roots, leaves, and coleoptiles. Importantly, for the first time, biodiversity was recognized in the coleoptiles of the investigated wheat species. Flavobacterium, Pseudomonas and Janthinobacterium were shown to be common genera for both tested wheat cultivars. Among them, Pseudomonas was found to be the only endophytic genus accompanying both wheat species from the endosperm stage to the development of the leaf. Paenibacillus was recognized as a core genus for the ‘Hondia’ cv., whereas Pedobacter and Duganella constituted the core microbiome in the ‘Rokosz’ cv. In addition, the first insight into the unique and yet unrecognized endophytic microbiome of T. spelta is presented.  相似文献

Knowledge of the main biological and climate factors influencing final harvest is becoming increasingly necessary in order to obtain reliable crop estimates and, thus, ensure optimised, effective private crop management. This knowledge is also of great value to public agricultural institutions for the planning of government subsidies. Castilla-La Mancha (Central Spain) is the second largest olive-oil-producing region in Spain, the highest olive-oil-producing country in the world. This study sought to identify the main factors influencing olive fruit production in this region, including atmospheric pollen as an index of flowering intensity, and meteorological data over the flowering and fruiting seasons in two main olive-producing provinces of the region: Ciudad Real and Toledo. Statistical analysis indicated that the annual pollen index (PI) was the variable influencing most the final olive crop in both provinces. The maximum temperature in March was the meteorological variable affecting most the annual olive crop. Also, the rainfall registered in October influences the final fruit production. The integration of aerobiological and meteorological data represents an important step forward in the development of future crop forecasting models in the region of Castilla-La Mancha.  相似文献

Stem sections from shoot cultures maintained in vitro were used to produce transgenic plants of the potato, Solanum tuberosum L. cv. Russet Burbank. Stem internode pieces inoculated with Agrobacterium tumefaciens containing coat protein genes from potato virus X and potato virus Y, produced shoots with a frequency of 60% in the absence of selection and 10% on medium containing 100 mg/l kanamycin monosulfate. Regenerated shoots were assayed for kanamycin resistance by placing stem segments on callus induction medium containing an increased level of kanamycin. Of a total 255 regenerated shoots, 47 (18%) were kanamycin resistant. Of the kanamycin resistant shoots, 25 (53%) expressed the PVX or PVY coat protein genes as assayed by enzyme-linked immunosorbent assay or Western immunoblot analysis.  相似文献

Date palm (Phoenix dactylifera L.) is an economically important and widely cultivated palm of the family Arecaceae. We sequenced the complete date palm chloroplast genome (cpDNA) from Pakistani cv. ??Aseel??, using a combination of Sanger-based and next-generation sequencing technologies. Being very similar to a sequence from a Saudi Arabian date palm cultivar ??Khalas?? published recently, the size of the genome was 158,458?bp with a pair of inverted repeat (IR) regions of 27,276?bp that were separated by a large single-copy (LSC) region of 86,195?bp and a small single-copy (SSC) region of 17,711?bp. Genome annotation demonstrated a total of 138 genes, of which 89 were protein coding, 39 were tRNA, and eight were rRNA genes. Comparison of cpDNA sequences of cultivars ??Aseel?? and ??Khalas?? showed following intervarietal variations in the LSC region; (a) two SNPs in intergenic spacers and one SNP in the rpoc1 gene, (b) polymorphism in two mono-nucleotide simple sequence repeats (SSR), and (c) a 4-bp indel in the accD-psaI intergenic spacer. The SSC region has a polymorphic site in the mono-nucleotide SSR located at position 120,710. We also compared cv. ??Aseel?? cpDNA sequence with partial P. dactylifera cpDNA sequence entries deposited in Genbank and identified a number of potentially useful polymorphisms in this species. Analysis of date palm cpDNA sequences revealed a close relationship with Typha latifolia. Occurrence of small numbers of forward and inverted repeats in date palm cpDNA indicated conserved genome arrangement.  相似文献

As part of a large-scale genomics project focused on understanding and improving the Shepody potato, we have increased the regeneration and transformation rates for this cultivar. Using combinations of auxins and trans-zeatin, leaf and stem explants were evaluated for callus induction and shoot formation. Several plant growth regulator combinations resulted in higher plant regeneration rates over a previous method. Using the best combination of auxin and cytokinin in combination with Agrobacterium-mediated transformation, we regenerated independent putative transformants from 59.5% of the total explants plated. We ran PCR on a sample of the plants to confirm transformation and 47.1% were nptII positive; giving a confirmed transformation rate of 28.0%.  相似文献

When excised, light-grown leaves of Hordeum vulgare were fed with (±)-[2-14C]-abscisic acid and stressed until they had lost 12% of their original fresh weight, marked changes in the distribution of radioactivity between abscisic acid and its catabolites were observed. Wilted leaves were less able than their turgid counterparts to transform (±)-[2-14C]-abscisic acid into 2-hydroxymethyl abscisic acid, dihydrophaseic acid and water-soluble conjugates of abscisic acid. Water stress had little effect on the production of phaseic acid although refeeding studies with [14C]-phaseic acid showed that the step from phaseic acid to dihydrophaseic acid was inhibited in wilted leaves. Evidence was obtained which suggested that these changes did not result from dilution of applied, radiolabelled substrate by endogenous abscisic acid. The catabolites of (±)-abscisic acid were identified by capillary gas chromatography-mass spectrometry.  相似文献

Efficient callus formation was achieved from cotyledon, stem, and leaf expiants of the domestic safflower cultivar Centennial on MS salts medium containing 1 mg/L BAP and 1 mg/L NAA. Shoot buds were regenerated from 26% of leaf-derived calli on callus induction medium, although attempts to root regenerated shoots were not successful. Centennial expiants inoculated with Agrobacterium tumefaciens containing NPT II and GUS genes produced kanamycin-resistant calli from which buds were regenerated. Summary A wide range of phenotypic variation occurred among protoplast — derived plants of tetraploid potato cultivar Bintje. The variant plants had alterations in growth and vigour, and in leaf and stem characteristics. The results suggest that the altered morphologies are caused predominantly by changes in ploidy levels. Some alterations could be attributed typically to octoploidy and aneuploidy. The occurrence of mixoploidy indicates that at least part of the observed variation arose during culture stage. The exogeneous cytokinin or auxin level and their combination during in vitro phase influenced the frequency of the variants observed. The origin of variation is discussed.  相似文献

The remains of Olea europaea in archaeological contexts in the southern Iberian Peninsula have been found in the Epipalaeolithic levels of Cueva de Nerja (10860±160 b.p.). The abundant appearance of charcoal and some seed remains from the Copper Age (3rd millennium b.c.) in the coastal zones of the southeast indicate that this species formed part of the vegetation of the Thermo-mediterranean zone and that its fruits were collected during these periods. However, Olea did not appear in the Meso-mediterranean zone until the Roman period, when olive cultivation was introduced there. The presence of charcoal and olive stones from the 1st century a.d. onwards is abundant, together with remains of structures for oil pressing.  相似文献

Callus cultures from cotyledon and hypocotyl explants of a Spanish cultivar of melon (Amarillo Oro) have been tested for their growth and morphogenic capacity on a series of media with different concentrations of indole-3-acetic acid (IAA) and 6-furfurylaminopurine (kinetin). Melon tissues were able to undergo morphogenesis both via organogenesis and embryogenesis, depending on culture conditions and explant source. Shoot buds were obtained at high rates in cotyledon explants. In response to 1.5 mg/l IAA and 6.0 mg/l kinetin, more than 90% of the calli produced well-developed shoots. Hypocotyls failed to form shoots but formed somatic embryos on auxin containing media while cotyledon explants usually gave abundant shoots but only rarely formed embryos. It was possible to maintain organogenic callus lines for at least 12 months under defined conditions. Plants were recovered from adventitious shoots produced both in cotyledon-derived calli and from organogenic cell lines.  相似文献
