Shoot branching is essential in ornamental chrysanthemum production and determines final plant shape and quality. Auxin is associated with apical dominance to indirectly inhibit bud outgrowth. Two non-mutually exclusive models exist for indirect auxin inhibition. Basipetal auxin transport inhibits axillary bud outgrowth by limiting auxin export from buds to stem (canalization model) or by increasing strigolactone levels (second messenger model). Here we analyzed bud outgrowth in treatments with auxin (IAA), strigolactone (GR24) and auxin transport inhibitor (NPA) using a split-plate bioassay with isolated chrysanthemum stem segments. Besides measuring bud length, dividing cell percentage was measured with flow cytometry and RT-qPCR was used to monitor expression levels of genes involved in auxin transport (CmPIN1) and signaling (CmAXR2), bud dormancy (CmBRC1, CmDRM1) and strigolactone biosynthesis (CmMAX1, CmMAX3). Treatments over a 5-day period showed bud outgrowth in the control and inhibition with IAA and IAA?+?GR24. Bud outgrowth in the control coincided with high dividing cell percentage, decreased expression of CmBRC1 and CmDRM1 and increased CmPIN1 expression. Inhibition by IAA and IAA?+?GR24 coincided with low dividing cell percentage and unchanged or increased expressions of CmBRC1, CmDRM1 and CmPIN1. Treatment with GR24 showed restricted bud outgrowth that was counteracted by NPA. This restricted bud outgrowth was still concomitant with a high dividing cell percentage and coincided with decreased expression of dormancy genes. These results indicate incomplete inhibition of bud outgrowth by GR24 treatment and suggest involvement of auxin transport in the mechanism of bud inhibition by strigolactones, supporting the canalization model.  相似文献

Flower induction in apple (Malus domestica Borkh.) trees plays an important life cycle role, but young trees produce fewer and inferior quality flower buds. Therefore, shoot bending has become an important cultural practice, significantly promoting the capacity to develop more flower buds during the growing seasons. Additionally, microRNAs (miRNAs) play essential roles in plant growth, flower induction and stress responses. In this study, we identified miRNAs potentially involved in the regulation of bud growth, and flower induction and development, as well as in the response to shoot bending. Of the 195 miRNAs identified, 137 were novel miRNAs. The miRNA expression profiles revealed that the expression levels of 68 and 27 known miRNAs were down‐regulated and up‐regulated, respectively, in response to shoot bending, and that the 31 differentially expressed novel miRNAs between them formed five major clusters. Additionally, a complex regulatory network associated with auxin, cytokinin, abscisic acid (ABA) and gibberellic acid (GA) plays important roles in cell division, bud growth and flower induction, in which related miRNAs and targets mediated regulation. Among them, miR396, 160, 393, and their targets associated with AUX, miR159, 319, 164, and their targets associated with ABA and GA, and flowering‐related miRNAs and genes, regulate bud growth and flower bud formation in response to shoot bending. Meanwhile, the flowering genes had significantly higher expression levels during shoot bending, suggesting that they are involved in this regulatory process. This study provides a framework for the future analysis of miRNAs associated with multiple hormones and their roles in the regulation of bud growth, and flower induction and formation in response to shoot bending in apple trees.  相似文献

Experiments have been done to confirm the previously reported effect of indoleacetic acid (IAA) on the rate of CO2 assimilation in bean leaves. It was shown that spraying the leaves of a variety of plants caused an increase in the rate of CO2 assimilation from 30% to 100% during the half-hour to 1 hour period following spraying. The only plant tested which did not show such an effect was corn.

The breaking of dormancy of axial buds in the bean plant was correlated with an increase in the rate of CO2 assimilation in adjacent leaves for a brief period of time. It has been shown that IAA solution sprayed on 1 leaflet of a leaf can cause an increase in the rate of CO2 assimilation in the other leaflets, and that IAA applied to the cut stem of a leaflet or a developing bud can be transported to adjacent leaves and cause an increase in the CO2 assimilation rate. The reaction caused by IAA is very similar to that caused by the breaking of dormancy of a bud. This indicates that the bud break response in CO2 assimilation in leaves is caused by auxin synthesized in a bud as it begins to grow, and exported into adjacent leaves.

Massive flowering of tropical Phalaenopsis orchids is coordinated by the cold-induced release of reproductive bud dormancy. Light and temperature are the two key factors integrated by the dormancy mechanism to both stop and reactivate the meristem development of many other angiosperm species, including fruit trees and ornamental plants. It is well established that leaves and roots play a major role in inducing flower development; however, currently, knowledge of molecular events associated with reproductive bud dormancy release in organs other than the bud is limited. Using differential gene expression, we have shown that the leaves of a hybrid of Phalaenopsis species, Doritaenopsis ‘Tinny Tender’, undergo major metabolic modifications. These changes result in the production of sucrose and amino acids, both of which can sustain bud outgrowth, and auxin and ethylene, which may play important roles in awaking the dormant buds. Intake of abscisic acid and synthesis of the hormone jasmonate may also explain the inhibition of vegetative growth that coincides with bud growth. Interestingly, many genes that were upregulated by cold treatment are homologous for genes involved in flower induction and vernalization in Arabidopsis, indicating that processes regulating flowering induction and those regulating reproductive bud dormancy release may use similar pathways and effector molecules.  相似文献

The technique of isopiestic thermocouple psychrometry was used for the analysis of bud transition from dormancy to growth and back in 8-18-day-old pea (Pisum sativum L.) seedlings. We monitored changes in the water (ψw) and osmotic (ψs + m) potentials and also turgor pressure (ψp) in dormant buds and threshold turgor (Y) in growing buds, the latter being one of the cell-wall rheological characteristics. Seedling decapitation resulted in a decrease of Y in the bud, which coincided with the start of its outgrowth. The replacement of terminal shoot with exogenous auxin (IAA or NAA) retarded bud outgrowth and maintained the high level of Y, which argues for the auxin control of this parameter. When growth of the first axillary bud was inhibited by the second one, positioned higher and remained on the plant, the beginning of Y increase preceded visible correlative growth suppression; this makes this rheological index an early marker of bud transition from growth to dormancy. The effects of the terminal shoot part and auxin application on the bud osmotic status differed substantially. In fact, bud transition to dormancy in the presence of the terminal shoot, the main or developing from the second axillary bud, was accompanied by the rise in ψs + m, whereas, in the case of the replacement of the second bud with exogenous auxin, the first bud growth suppression occurred with the decrease in ψs + m. The low value of the bud ψs + m is a factor for creating a considerable gradient of the water potential between the stem and bud supporting water transport to the bud, which was much more active than in plants with a terminal shoot. It seems likely that this is the reason for the absence of complete growth suppression observed by us and other researchers even after application of high auxin concentrations. Immediately after seedling decapitation, ψs + m in the buds reduced; however, this was not the result of trophic metabolite redistribution due to the loss of their active sink because ψs + m reduced also in experiments with complete isolation of the bud releasing from dormancy in the chamber at 100% humidity. Auxin application to the cut surface of decapitated seedlings did not affect the ψs + m decrease. Like decapitation, cotyledon removal resulted in the increase in the bud turgor pressure. However, in this case, water stress did not change the bud osmotic status. Thus, the induction of osmotica accumulation in the bud after the removal of the terminal shoot is evidently related to neither trophic, nor auxin, nor hydraulic signal. The data obtained allowed us to conclude that both components of the bud water potential—ψs + m and Y—play an important role in the control of bud growth at apical dominance. Auxin produced in the shoot apex is involved in the control of Y, whereas the nature of the signal controlling the ψs + m level is unclear.  相似文献

Tea (Camellia sinensis [L.] O. Kuntze.) is an important cash crop, which mainly uses tender shoots and young leaves for manufacturing. Due to the marketing characteristic that earlier made tea has higher price, the time of the breaking of winter dormancy buds in spring is extremely important in tea industry. Strigolactones are a group of carotenoids-derived metabolites which regulates bud outgrowth, shoot branching, tiller angle and environmental stress responses. The role of strigolactones in tea plant was briefly summarized in the current review, with an emphasis of the association of strigolactones on bud ecodormancy and shoot branching. The involvement of strigolactones on the biosynthesis of the tea characteristic metabolites flavonoids, caffeine and theanine were also discussed. Moreover, recent advances on the biosynthesis of strigolactones and its regulation by microRNAs and environmental stresses were also presented. This review provides a basis for future investigations underlying the mechanisms of strigolactones on bud winter dormancy and tea secondary metabolism.  相似文献

Hormones play an important role in regulating the growth of rice tiller buds. However, little is known about the hormonal changes that occur during tiller bud growth and the mechanism of hormonal regulation of tiller bud growth. Here, two rice cultivars, Yangdao 6 (Indica) and Nanjing 44 (Japonica), were used to investigate the changes in plant hormones during tiller bud growth and the mechanism that underlies the hormonal regulation of tiller bud growth. In the present study, panicles were removed after heading to stimulate the growth of dormant tiller buds located at the elongated upper internodes. At the same time, external abscisic acid (ABA), gibberellic acid (GA3) and α-naphthalene acetic acid (NAA) were applied. The results demonstrated that auxin and cytokinin (CTK) play important and different roles in the regulation of tiller bud growth. Auxin in the nodes inhibits tiller bud growth, while CTK is transferred to the tiller buds to promote growth. The inhibitory effects of GA3 and NAA on tiller bud growth are mainly due to the control of the indole-3-acetic acid (IAA) or CTK contents in plants. As opposed to auxin and CTK, the ABA contents in nodes and tiller buds remained unchanged before tiller bud growth after panicle removal. Meanwhile, external ABA application only slightly slowed the growth of the tiller buds, suggesting that ABA may not be a key regulator of tiller bud growth. These results indicate that auxin, CTK and ABA together likely play roles in the regulation of tiller bud growth.  相似文献

Many processes have been described in the control of shoot branching. Apical dominance is defined as the control exerted by the shoot tip on the outgrowth of axillary buds, whereas correlative inhibition includes the suppression of growth by other growing buds or shoots. The level, signaling, and/or flow of the plant hormone auxin in stems and buds is thought to be involved in these processes. In addition, RAMOSUS (RMS) branching genes in pea (Pisum sativum) control the synthesis and perception of a long-distance inhibitory branching signal produced in the stem and roots, a strigolactone or product. Auxin treatment affects the expression of RMS genes, but it is unclear whether the RMS network can regulate branching independently of auxin. Here, we explore whether apical dominance and correlative inhibition show independent or additive effects in rms mutant plants. Bud outgrowth and branch lengths are enhanced in decapitated and stem-girdled rms mutants compared with intact control plants. This may relate to an RMS-independent induction of axillary bud outgrowth by these treatments. Correlative inhibition was also apparent in rms mutant plants, again indicating an RMS-independent component. Treatments giving reductions in RMS1 and RMS5 gene expression, auxin transport, and auxin level in the main stem were not always sufficient to promote bud outgrowth. We suggest that this may relate to a failure to induce the expression of cytokinin biosynthesis genes, which always correlated with bud outgrowth in our treatments. We present a new model that accounts for apical dominance, correlative inhibition, RMS gene action, and auxin and cytokinin and their interactions in controlling the progression of buds through different control points from dormancy to sustained growth.  相似文献

Auxin plays a pivotal role in many facets of plant development. It acts by inducing the interaction between auxin‐responsive [auxin (AUX)/indole‐3‐acetic acid (IAA)] proteins and the ubiquitin protein ligase SCFTIR to promote the degradation of the AUX/IAA proteins. Other cofactors and chaperones that participate in auxin signaling remain to be identified. Here, we characterized rice (Oryza sativa) plants with mutations in a cyclophilin gene (OsCYP2). cyp2 mutants showed defects in auxin responses and exhibited a variety of auxin‐related growth defects in the root. In cyp2 mutants, lateral root initiation was blocked after nuclear migration but before the first anticlinal division of the pericycle cell. Yeast two‐hybrid and in vitro pull‐down results revealed an association between OsCYP2 and the co‐chaperone Suppressor of G2 allele of skp1 (OsSGT1). Luciferase complementation imaging assays further supported this interaction. Similar to previous findings in an Arabidopsis thaliana SGT1 mutant (atsgt1b), degradation of AUX/IAA proteins was retarded in cyp2 mutants treated with exogenous 1‐naphthylacetic acid. Our results suggest that OsCYP2 participates in auxin signal transduction by interacting with OsSGT1.  相似文献

Although the co-ordination of promotive root-sourced cytokinin (CK) and inhibitory shoot apex-sourced auxin (IAA) is central to all current models on lateral bud dormancy release, control by those hormones alone has appeared inadequate in many studies. Thus it was hypothesized that the IAA : CK model is the central control but that it must be considered within the relevant timeframe leading to lateral bud release and against a backdrop of interactions with other hormone groups. Therefore, IAA and a wide survey of cytokinins (CKs), were examined along with abscisic acid (ABA) and polyamines (PAs) in released buds, tissue surrounding buds and xylem sap at 1 and 4 h after apex removal, when lateral buds of chickpea are known to break dormancy. Three potential lateral bud growth inhibitors, IAA, ABA and cis -zeatin 9-riboside (ZR), declined sharply in the released buds and xylem following decapitation. This is in contrast to potential dormancy breaking CKs like trans -ZR and trans -zeantin 9-riboside 5'phosphate (ZRMP), which represented the strongest correlative changes by increasing 3.5-fold in xylem sap and 22-fold in buds. PAs had not changed significantly in buds or other tissues after 4 h, so they were not directly involved in the breaking of bud dormancy. Results from the xylem and surrounding tissues indicated that bud CK increases resulted from a combination synthesis in the bud and selective loading of CK nucleotides into the xylem from the root.  相似文献

Previously we identified aminooxy compounds as auxin biosynthesis inhibitors. One of the compounds, aminooxyacetic acid (AOA) inhibited indole-3-acetic acid (IAA) biosynthesis in rice and tomato. Here, we found that AOA induced auxin over-accumulation in Arabidopsis. The results suggest that auxin-related metabolic pathways are divergent among these plant species.  相似文献

In plants, auxin is a key regulator of development and is unique among plant hormones in that its function requires polarized transport between neighboring cells to form concentration gradients across various plant tissues. Although putative auxin-influx and -efflux transporters have been identified by using molecular genetic approaches, a detailed functional understanding for many of these transporters remains undetermined. Here we present the functional characterization of the auxin-influx carrier AUX1. Upon expression of AUX1 in Xenopus oocytes, saturable, pH-dependent uptake of 3H-IAA was measured. Mutations in AUX1 that abrogate physiological responses to IAA in planta resulted in loss or reduction of 3H-IAA uptake in AUX1-expressing oocytes. AUX1-mediated uptake of 3H-IAA was reduced by the IAA analogs 2,4-D and 1-NOA, but not by other auxin analogs. The measured Km for AUX1-mediated uptake of 3H-IAA was at concentrations at which physiological responses are observed for exogenously added IAA and 2,4-D. This is the first report demonstrating detailed functional characteristics of a plant auxin-influx transporter. This biochemical characterization provides new insights and a novel tool for studying auxin entry into cells and its pivotal roles in plant growth and development.  相似文献

Heterologous expression systems based on tobacco BY‐2 cells, Arabidopsis cell cultures, Xenopus oocytes, Saccharomyces cerevisiae, and human HeLa cells have been used to express and characterize PIN, ABCB (PGP), and AUX/LAX auxin transporters from Arabidopsis. However, no single system has been identified that can be used for effective comparative analyses of these proteins. We have developed an accessible Schizosaccharomyces pombe system for comparative studies of plant transport proteins. The system includes knockout mutants in all ABC and putative auxin transport genes and Gateway®‐compatible expression vectors for functional analysis and subcellular localization of recombinant proteins. We expressed Arabidopsis ABCB1 and ABCB19 in mam1pdr1 host lines under the inducible nmt41 promoter. ABCB19 showed a higher 3H‐IAA export activity than ABCB1. Arabidopsis PIN proteins were expressed in a mutant lacking the auxin effluxer like 1 (AEL1) gene. PIN1 showed higher activity than PIN2 with similar protein expression levels. Expression of AUX1 in a permease‐deficient vat3 mutant resulted in increased net auxin uptake activity. Finally, ABCB4 expressed in mam1pdr1 displayed a concentration‐dependent reversal of 3H‐IAA transport that is consistent with its observed activity in planta. Structural modelling suggests that ABCB4 has three substrate interaction sites rather than the two found in ABCB19, thus providing a rationale for the observed substrate activation. Taken together, these results suggest that the S. pombe system described here can be employed for comparative analyses and subsequent structural characterizations of plant transport proteins.  相似文献

Shoot branching and growth are controlled by phytohormones such as auxin and other components in Arabidopsis. We identified a mutant (igi1) showing decreased height and bunchy branching patterns. The phenotypes reverted to the wild type in response to RNA interference with the IGI1 gene. Histochemical analysis by GUS assay revealed tissue-specific gene expression in the anther and showed that the expression levels of the IGI1 gene in apical parts, including flowers, were higher than in other parts of the plants. The auxin biosynthesis component gene, CYP79B2, was up-regulated in igi1 mutants and the IGI1 gene was down-regulated by IAA treatment. These results indicated that there is an interplay regulation between IGI1 and phytohormone auxin. Moreover, the expression of the auxin-related shoot branching regulation genes, MAX3 and MAX4, was down-regulated in igi1 mutants. Taken together, these results indicate that the overexpression of the IGI1 influenced MAX pathway in the shoot branching regulation.  相似文献
