Transformation and Functional Expression of the rFCA-RRM2 Gene in Rice   总被引:2,自引:0,他引:2  
The primary aim of the present study was to investigate the overexpression of the rice (Oryza sativa L. subsp, japonica var. Zhonghua 11) flowering control gene (rFCA-RRM2) in monocotyledonous model rice. Constitutive expression of rFCA-RRM2 from the Actl-5 rice promoter caused late flowering in transgenic rice and increased grain weight that was more than 50% higher than that of control plants, which is the first demonstration of rFCA-RRM2 being able to increase rice production. Late flowering was accompanied by strong phenotype and some morphological modifications. These observations suggest that rFCA-RRM2 is a useful tool for phenotype improvement and yield enhancement in cereal crops.  相似文献

2-bromoethanesulfonate (BES) is a structural analogue of coenzyme M (Co-M) and potent inhibitor of methanogenesis. Several studies confirmed, BES can inhibit CH4 prodcution in rice soil, but the suppressing effectiveness of BES application on CH4 emission under rice cultivation has not been studied. In this pot experiment, different levels of BES (0, 20, 40 and 80 mg kg-1) were applied to study its effect on CH4 emission and plant growth during rice cultivation. Application of BES effectively suppressed CH4 emission when compared with control soil during rice cultivation. The CH4 emission rates were significantly (P<0.001) decreased by BES application possibly due to significant (P<0.001) reduction of methnaogenic biomarkers like Co-M concentration and mcrA gene copy number (i.e. methanogenic abunadance). BES significantly (P<0.001) reduced methanogen activity, while it did not affect soil dehydrogenase activity during rice cultivation. A rice plant growth and yield parameters were not affected by BES application. The maximum CH4 reduction (49% reduction over control) was found at 80 mg kg-1 BES application during rice cultivation. It is, therefore, concluded that BES could be a suitable soil amendment for reducing CH4 emission without affecting rice plant growth and productivity during rice cultivation.  相似文献

5,6-Cl_2-IAA可以诱导水稻芽鞘产生斜向地性生长。其倾斜角度与外加5,6-Cl_2-IAA的剂量呈线性关系,其反应具有特异性(IAA,6-BA,ABA和BR均不能诱导这种反应)。反应可在2~3h后产生,它可作为5,6-Cl_2-IAA的生物检定依据。切除芽鞘顶尖或外加TIBA对这一反应无明显影响,另外乙烯生物合成抑制剂AVG也不能消除5,6-Cl_2-IAA诱导的斜向地性生长。  相似文献

用转PEPC、PPDK、NADP-ME、PEPC+PPDK酶基因水稻(Oryza sativa L.)及原种为材料 ,研究了光合作用对光照、温度、CO2的响应和光抑制条件下的叶绿素荧光特性,结果如下: 1.转C4光合酶基因水稻的饱和光合速率比原种高,其中转PEPC、PEPC+PPDK双基因水稻的光饱和点比原种高200 μmol*m-2*s-1,饱和光合速率比原种分别高51.6%和 58.5%;转PEPC基因水稻的羧化效率比原种高49.3%,CO2补偿点降低26.2%;在高温(35 ℃)下,转PEPC基因水稻的光合速率比原种高17.5%.2.经光抑制处理8 d后,转PEPC、PEPC +PPDK酶基因水稻的PSⅡ光化学效率(Fv/Fm)和光化学猝灭(qP)下降20%- 30%,非光化学猝灭(qN)增加了约30%;但原种的Fv/Fm和qP下降了5 0%多,qN变化不明显,表明转C4光合基因水稻耐光抑制能力增强.这些结果为用生物技术提高水稻光合效率研究提供了新的依据和途径.  相似文献

Ultrastructural studies were made using plants of five selected rice cultivars infected with rice tungro viruses. In all cultivars, rice tungro bacilliform virus (RTBV) was found in both xylem and phloem, while rice tungro spherical virus (RTSV) was observed only in the phloem. In tolerant cultivars Balimau Putih and Utri Merah, plants infected with RTBV and RTSV or RTBV alone, had fewer cells containing tungro viruses than plants of the sensitive cultivar Taichung Native 1. In RTBV-sensitive cultivars FK 135 and ASD 8 infected with both viruses or RTBV alone, electron-dense cytoplasm was observed in most cells of the phloem. Abundant phloem necrosis was also observed in FK 135. These observations were correlated with the reaction of each rice cultivar to tungro infection.  相似文献

Oryza sativa ) and wild rice (O. rufipogon) tissues. It is detected at every developmental stage, and is transmitted very efficiently to progeny via seeds (more than 98%). The dsRNA is maintained at a constant level (approximately 100 copies/cell) in almost all tissues. However, the number of copies increases about 10-fold when host cells are grown in suspension culture. Complete nucleotide sequences of cultivated rice (temperate japonica rice, cv. Nipponbare, J-dsRNA) and wild rice (W-1714, W-dsRNA) dsRNAs have been determined. Both wild and cultivated rice dsRNAs have a single long open reading frame (ORF) containing the conserved motifs of RNA-dependent RNA polymerase and RNA helicase. The coding strands of both contain a site-specific discontinuity (nick) at nt 1,211 (J-dsRNA) or at nt 1,197 (W-dsRNA) from the 5′ end of their coding strand. Rice dsRNA has several unique properties and can be regarded as a novel RNA replicon. This paper discusses the origin and evolution of the rice dsRNA. Interactions between sulfate reduction (SR) and phototrophic nitrogenase activities were investigated in rice soil slurries mixed with rice straw. Activation of SR by adding exogenous sulfate suppressed acetylene-reducing activity (ARA) of the slurries, which was associated with phototrophic purple bacteria (PB) enumerated to 108-109 MPN g-1 dry weight (dw) soil. Adding 5 mm sodium molybdate, an inhibitor of SR, markedly increased ARA. However, in the slurries receiving both molybdate and exogenous sulfate, the effects declined simultaneously with partial recovery of SR. These results indicate outcompetition of sulfate-reducing bacteria (SRB) with PB in rice soil, when sulfate concentrations are high enough to support SR. The increasing effects of molybdate on ARA continued during the incubation in the sulfate-depleted condition, probably because of absence of SR and toxicity of molybdate to methanogenesis. Accordingly, stopping activities of the competitive microorganisms may be efficient to increase N2 fixation in rice soil.  相似文献

以籼稻ZYQ8与粳稻JX17为亲本的DH群体作为研究材料,考察DH群体及双亲的米粒延伸率相关性状,并使用该群体的分子连锁图谱进行QTL分析.共检测到14个与稻米延伸性有关的QTL,包括2个粒长QTL、7个饭粒长QTL和5个米粒延伸率QTL,分别位于第1、2、3、5、6、7、10、11和12染色体.所有QTL的LOD值介于2.26~9.25,分别解释性状变异的5.31%~17.21%.在第3染色体上的G249~G164、第6染色体上的G30~RZ516和第10染色体上的G1082~GA223区间同时检测到控制饭粒长和米粒延伸率的QTL.米粒延伸性受多基因控制,Wx基因与位于第6染色体上的qCRE-6的G30~RZ516区间相近,对米饭的延伸性具重要影响.  相似文献

It is often claimed that fish stocked in rice fields have a significant impact on the rice pest population. However, there is little experimental evidence for this. We used the data of six experiments to find out whether a polyculture of silver barb Barbodes gonionotus (Bleeker), common carp Cyprinus carpio (L.) and Nile tilapia Oreochromis niloticus (L.) has any effect on the population of rice leaffolders Cnaphalocrocis medinalis (Guenée) and rice caseworms Nymphula depunctalis (Guenée), two common pests in intensively cultured rice fields. Fish had no effect on the rice leaffolder population, but significantly lowered the number of rice caseworm adults and larvae. We concluded that fish have some effect on the arthropod pest population in rice fields, although, most probably, only on sluggish pests living in the water or on the water surface.  相似文献

根据营养液培养试验从水稻Azucena×IR64 发展的一双单倍体(DH) 群体中筛选出抗铁毒与敏感品系。在铁毒害处理后,各品系的抗坏血酸过氧化物酶(AP) 、脱氢抗坏血酸还原酶(DR) 、谷胱甘肽还原酶(GR) 的活性均有明显提高;水稻受铁毒后的生物量递减量与AP、DR、GR 活性呈负相关。说明抗坏血酸过氧化物酶H2O2 清除系统在水稻抗铁毒中起着十分重要的作用。  相似文献

Mitogen-activated protein kinase (MAPK) cascades are evolutionary conserved from unicellular to complex eukaryotic organisms, and constitute one of the major signalling pathways involved in regulating a wide range of cellular activities from growth and development to cell death. MAPKs of rice (Oryza sativa L.), the most important of all food crops and an established monocot plant research model, have seen considerable progress mainly on their identification and characterization during the past one year alone. These studies have provided new information on the response and regulation of rice MAPKs, in particular on their possible role/function in the rice self-defense pathways. It is believed that further work on MAPK cascades in rice will widen our understanding of the MAPK signalling pathways, and may lead to the establishment of a biological model on this critical early signalling event in monocots. In this review, we bring together all the recent developments in rice MAPKs and discuss their significance and future direction in light of the present data and the progress made in dicot model plants.  相似文献

为了研究谷蛋白胚乳特异性表达启动子在我国栽培稻品种中的表达模式,将UidA基因分别置于水稻谷蛋白GluA—2基因750bp和2.3kb上游序列下游,利用农杆菌转化法导人栽培稻品种中花8号并获得转基因植株。Southern blot检测表明,UidA基因已经整合到水稻基因组当中并以单拷贝存在。Northern blot检测表明,开花后13~15d和11~13d,UidA基因和水稻内源的GluA—2基因的表达量分别达到最高,随后逐渐降低。对转基因植株种子的GUS染色表明,UidA基因仅在胚乳中表达,在糊粉层中GUS表达量最高。测定了2.3kb和750bp转基因植株种子的GUS活性,结果表明前者的GUS活性是后者的2~3倍。序列分析表明,位于GluA—2基因转录启始位点上游2170bD的G-box可能是一个与表达量相关的顺式调控元件。  相似文献

水稻Rim2/Hipa是最近鉴定的一个受逆境诱导的转座因子超级家族.研究表明,Rim2的核心序列在不同来源的水稻材料中存在显著的差异,暗示Rim2家族的长期进化历程.基于Rim2因子间的差异性以及该因子的静止状态,开发出一种利用Rim2因子展示的新的分子指纹技术,可以灵敏地区分不同水稻资源以及它们的遗传关系.仅用5对引物就可以清楚地将53个栽培稻和普通野生稻材料鉴定出来,并可将它们分为不同的系统进化组.研究表明不仅在水稻资源而且在野生稻种质间均存在明显的多样性.野生稻可以被单独分组,或者分散在粳稻中间.这种新的指纹技术还可以将水稻的杂交子代和它们的亲本区分出来,并可用于种子纯度的鉴定,在水稻基因组进化研究、水稻育种和种子生产中有很好的应用前景.  相似文献

为将不同启动子用于转基因水稻的研究,从武运粳8号水稻中克隆了Rubisco小亚基基因(rbcS)的5'上游调控区,构建了由rbcS启动子引导的GUS融合基因,并经农杆菌介导导入到水稻中.对转基因水稻植株中GUS活性的定性与定量测定结果表明,rbcS启动子可驱动GUS报告基因在转基因水稻植株叶片和叶鞘内的叶肉细胞中特异性高效表达,而在茎、根和种子等器官中不表达或表达活性极弱,表现出明显的组织与细胞特异性.结果还表明,光诱导处理可明显提高rbcS启动子启动的外源基因的表达量.  相似文献

利用micro array 技术对水稻幼苗在营养胁迫条件下根部基因表达的研究中发现: 一个与豌豆Pra2(小G蛋白)基因有同源性的基因的RNA水平在营养胁迫后再补充营养时, 表达量下降。用RT-PCR和PCR方法分别获得该基因的cDNA克隆—OsPra2和该基因翻译起始位点上游1 kb的启动子序列。OsPra2基因编码的蛋白质具有结合GTP/GDP的4个保守结构域和构成小G蛋白Rab家族的特有的结构域。该基因cDNA与GST蛋白基因融合表达载体在洋葱表皮细胞中的瞬间表达结果显示该蛋白定位在在细胞膜和细胞核上, OsPra2基因启动子与GUS报告基因融合表达转基因水稻显示该基因启动子驱动GUS在胚芽鞘和根中表达, 35S启动子驱动OsPra2基因过表达转基因水稻与野生水稻株型相比明显矮化, 类似BR缺陷型植物株型。本实验还对OsPra2和P450蛋白的相互作用及在BR代谢途径中的可能作用进行了分析。  相似文献

在常温下用不同浓度的外源H2O2(0~20 mmol·L-1)预处理水稻幼苗,再进行12 h 6℃低温胁迫,根据幼苗相对含水量和质膜相对透性筛选最佳外源H2O2处理浓度,并分析最佳外源H2O2浓度下幼苗的渗透调节物质和活性氧相关指标的变化.结果表明:(1)0~8 mmol·L-1 H2O2预处理可以增加水稻幼苗的相对含水量,降低其质膜相对透性,并以4 mmol·L-1 H2O2的效果最佳.(2)低温胁迫后,与对照组相比,4 mmol·L-1外源H2O2预处理降低了水稻幼苗萎蔫程度,并使其总呼吸速率、交替途径容量都有增加,同时还抑制了丙二醛的含量,增加了可溶性糖、可溶性蛋白质和脯氨酸的含量.(3)外源H2O2预处理对水稻幼苗的内源H2O2含量以及O(-)/(·)2产生速率没有显著影响.研究发现,外源H2O2可以通过提高呼吸速率、降低脂质过氧化程度、增加碳氮代谢来有效增强水稻幼苗的抗寒性,它可能以一种独立于内源活性氧系统之外的方式发挥作用.  相似文献
