Hydrogen sulfide (H2S), a gaseous transmitter, has long been considered as a phytotoxin, but nowadays as a small molecule with multiple functions fulfilled at low concentrations. H2S has many positive effects on plant growth, development, and the acquisition of plant stress tolerance. The focus of this review is to summarize the generation and properties of hydrogen sulfide and its potential physiological functions, including mediating stomatal movements; mediating the responses to abiotic stressors, such as heavy metals, salt, drought, and heating; involving in organogenesis and growth; regulating senescence; priming seed germination; and enhancing photosynthesis. Gases such as ethylene, hydrogen peroxide (H2O2), nitric oxide (NO), carbon monoxide (CO) and hydrogen sulfide (H2S) have been recognized as vital signaling molecules in plants and animals. Of these gasotransmitters, NO and H2S have recently gained momentum mainly because of their involvement in numerous cellular processes. It is therefore important to study their various attributes including their biosynthetic and signaling pathways. The present review provides an insight into various routes for the biosynthesis of NO and H2S as well as their signaling role in plant cells under different conditions, more particularly under heavy metal stress. Their beneficial roles in the plant's protection against abiotic and biotic stresses as well as their adverse effects have been addressed. This review describes how H2S and NO, being very small-sized molecules, can quickly pass through the cell membranes and trigger a multitude of responses to various factors, notably to various stress conditions such as drought, heat, osmotic, heavy metal and multiple biotic stresses. The versatile interactions between H2S and NO involved in the different molecular pathways have been discussed. In addition to the signaling role of H2S and NO, their direct role in posttranslational modifications is also considered. The information provided here will be helpful to better understand the multifaceted roles of H2S and NO in plants, particularly under stress conditions.  相似文献

A newly isolated autotrophic bacterium, Thiobacillus thioparus DW44, which is capable of degrading sulfur-containing gases, was inoculated into a pilot-scale peat biofilter to treat the exhaust gas from a night soil treatment plant. Hydrogen sulfide (H2S), methanethiol (MT), dimethyl sulfide (DMS) and dimethyl disulfide (DMDS) in the exhaust gas were efficiently removed for six months. Average removal ratios were 99.8% for H2S, 99.0% for MT, 89.5% for DMS and 98.1% for DMDS at a space velocity of 46 h−1 during the period of operation. No acclimation period was needed to reach such a high efficiency in the removal of the gases, indicating that the ability of this bacterium to remove these gases was occurred immediately after its inoculation to the peat. Ammonia (NH3) in the exhaust gas was neutralized with SO42−, which is the final product of the oxidation of H2S, MT, DMS and DMDS by the bacterium. No remarkable decline of pH, which often causes a deterioration in bacterial activity, was observed, mainly because of the reaction of SO42− with NH3. This study is the first report on the application of an isolated microorganism to a practical deodorizing system. The inoculation of T. thioparus DW44 into the pilot-scale peat biofilter could overcome such disadvantages of the conventional peat biofilter as a long acclimation period to reach a constant gas removability and the low removability of DMS, and resulted in enhanced removal efficiency of malodorous gases.  相似文献

Hydrogen sulfide (H2S) was once principally considered the perpetrator of plant growth cessation and cell death. However, this has become an antiquated view, with cumulative evidence showing that the H2S serves as a biological signaling molecule notably involved in abiotic stress response and adaptation, such as defense by phytohormone activation, stomatal movement, gene reprogramming, and plant growth modulation. Reactive oxygen species (ROS)-dependent oxidative stress is involved in these responses. Remarkably, an ever-growing body of evidence indicates that H2S can directly interact with ROS processing systems in a redox-dependent manner, while it has been gradually recognized that H2S-based posttranslational modifications of key protein cysteine residues determine stress responses. Furthermore, the reciprocal interplay between H2S and nitric oxide (NO) in regulating oxidative stress has significant importance. The interaction of H2S with NO and ROS during acclimation to abiotic stress may vary from synergism to antagonism. However, the molecular pathways and factors involved remain to be identified. This review not only aims to provide updated information on H2S action in regulating ROS-dependent redox homeostasis and signaling, but also discusses the mechanisms of H2S-dependent regulation in the context of oxidative stress elicited by environmental cues.  相似文献

Salinity stress is one of the most significant global issues that negatively affect plant growth and development. Modern agricultural practices have expanded the destructive effects of salinity stress, affecting plants through immediate osmotic stress, followed by a slow onset of ionic or hyper-osmotic stress. Plants alteration and resistance to salinity stress involve complex physiological, biochemical, and molecular systems to maintain homeostasis. As of late, the investigation of gaseous molecules in plants has attained much consideration, particularly for abiotic stress. Abiotic stresses generally initiate gasotransmitter (GT) generation in plants. In the interim, these GTs enhance the accumulation and activities of few antioxidant molecules, check the destructiveness of reactive oxygen species (ROS), and improve plant resilience under different stress conditions. The current review presented the role of gaseous molecules in plants under salinity stress, which include nitric oxide (·NO), hydrogen sulfide (H2S), hydrogen gas (H2), carbon monoxide (CO), methane (CH4), and the only gaseous phytohormone ethylene. Further, we highlighted the underlying molecular mechanisms of the gasotransmitter signaling and cross-talks in salinity stress. Also, we presented a general update on the inclusion of GT in salt stress response, including the research gaps and its applications in the advancement of salinity-resistant plants.

The oscillation of intracellular calcium (Ca2+) concentration is a primary event in numerous biological processes in plants, including stress response. Hydrogen sulfide (H2S), an emerging gasotransmitter, was found to have positive effects in plants responding to chromium (Cr6+) stress through interacting with Ca2+ signaling. While Ca2+ resemblances H2S in mediating biotic and abiotic stresses, crosstalk between the two pathways remains unclear. In this study, Ca2+ signaling interacted with H2S to produce a complex physiological response, which enhanced the Cr6+ tolerance in foxtail millet (Setaria italica). Results indicate that Cr6+ stress activated endogenous H2S synthesis as well as Ca2+ signaling. Moreover, toxic symptoms caused by Cr6+ stress were strongly moderated by 50 μM H2S and 20 mM Ca2+. Conversely, treatments with H2S synthesis inhibitor and Ca2+ chelators prior to Cr6+-exposure aggravated these toxic symptoms. Interestingly, Ca2+ upregulated expression of two important factors in metal metabolism, MT3A and PCS, which participated in the biosynthesis of heavy metal chelators, in a H2S-dependent manner to cope with Cr6+ stress. These findings also suggest that the H2S dependent pathway is a component of the Ca2+ activating antioxidant system and H2S partially contributes Ca2+-activating antioxidant system.  相似文献

In the recent times, plants are facing certain types of environmental stresses, which give rise to formation of reactive oxygen species (ROS) such as hydroxyl radicals, hydrogen peroxides, superoxide anions and so on. These are required by the plants at low concentrations for signal transduction and at high concentrations, they repress plant root growth. Apart from the ROS activities, hydrogen sulfide (H2S) and nitric oxide (NO) have major contributions in regulating growth and developmental processes in plants, as they also play key roles as signaling molecules and act as chief plant immune defense mechanisms against various biotic as well as abiotic stresses. H2S and NO are the two pivotal gaseous messengers involved in growth, germination and improved tolerance in plants under stressed and non-stress conditions. H2S and NO mediate cell signaling in plants as a response to several abiotic stresses like temperature, heavy metal exposure, water and salinity. They alter gene expression levels to induce the synthesis of antioxidant enzymes, osmolytes and also trigger their interactions with each other. However, research has been limited to only cross adaptations and signal transductions. Understanding the change and mechanism of H2S and NO mediated cell signaling will broaden our knowledge on the various biochemical changes that occur in plant cells related to different stresses. A clear understanding of these molecules in various environmental stresses would help to confer biotechnological applications to protect plants against abiotic stresses and to improve crop productivity.  相似文献

Plants often face a variety of abiotic stresses, which affects them negatively and lead to yield loss. The antioxidant system efficiently removes excessive reactive oxygen species and maintains redox homeostasis in plants. With better understanding of these protective mechanisms, recently the concept of hydrogen sulfide (H2S) and its role in cell signaling has become the center of attention. H2S has been recognized as a third gasotransmitter and a potent regulator of growth and development processes such as germination, maturation, senescence and defense mechanism in plants. Because of its gaseous nature, H2S can diffuse to different part of the cells and balance the antioxidant pools by supplying sulfur to cells. H2S showed tolerance against a plethora of adverse environmental conditions like drought, salt, high temperature, cold, heavy metals and flood via changing in level of osmolytes, malonaldialdehyde, Na+/K+ uptake, activities of H2S biosynthesis and antioxidative enzymes. It also promotes cross adaptation through persulfidation. H2S along with calcium, methylglyoxal and nitric oxide, and their cross talk induces the expression of mitogen activated protein kinases as well as other genes in response to stress. Therefore, it is sensible to evaluate and explore the stress responsive genes involved in H2S regulated homeostasis and stress tolerance. The current article is aimed to summarize the recent updates on H2S-mediated gene regulation in special reference to abiotic stress tolerance mechanism, and cross adaptation in plants. Gaseous molecules, such as hydrogen sulfide(H_2S)and nitric oxide(NO), are crucial players in cellular and(patho)physiological processes in biological systems. The biological functions of these gaseous molecules, which were first discovered and identified as gasotransmitters in animals, have received unprecedented attention from plant scientists in recent decades. Researchers have arrived at the consensus that H_2S is synthesized endogenously and serves as a signaling molecule throughout the plant life cycle.However, the mechanisms of H_2S action in redox biology is still largely unexplored. This review highlights what we currently know about the characteristics and biosynthesis of H_2S in plants. Additionally,we summarize the role of H_2S in plant resistance to abiotic stress. The potential of plant nutrients (such as sulphur, S) and phytohormones (such as salicylic acid, SA) has been explored in isolated studies by researchers in controlling the impact of abiotic stresses such as salinity in plants. However, information is scanty on the major mechanisms underlying the role of S and/or SA in modulation of enzymes involved in nitrogen (N) assimilation, GOGAT cycle, and antioxidant defence system; the cellular status of N-containing osmolyte proline, glucose, S-containing compounds; and their cumulative role in photosynthesis functions and growth in crop plants. The present study aimed to assess the role of cumulative effect of SA and S (SO42−) mediated induction of N assimilatory enzymes, GOGAT cycle, N-osmolyte proline and its metabolizing enzymes, glyoxylase enzymes, and antioxidant capacity in mungbean (Vigna radiata L.) exposed to NaCl with or without SO42− and SA. Salt-exposed V. radiate showed differential elevations in damage (O.2, H2O2, lipid peroxidation; glucose) and defence (ascorbate peroxidase, APX; glutathione reductase, GR; superoxide dismutase, SOD; reduced GSH; proline) and inhibitions in the activities of NR and NiR; N content, photosynthesis, photosynthetic N-use-efficiency (NUE), and growth. The separate supplementation of SA and SO42− to 50 mM NaCl almost equally strengthened the antioxidant machinery and diminished NaCl-accrued damages. However, combined supply of SA and SO42− to NaCl-exposed cultivars led to significant improvements in NR and NiR activities, the accumulation of N, GSH, proline, enhanced activity of APX, GR, and reduced activity of SOD, and also decreases in O.2, H2O2, lipid peroxidation and glucose. These observations were corroborated with SA, SO42− and NaCl-mediated changes in the traits of photosynthesis and growth, stomatal behaviour, and the polypeptide patterns of Rubisco in V. radiata. Overall, in V. radiata, SA-mediated higher enhancements in the activity of N assimilatory enzymes (NR, NiR, and GS), increase in the N and proline, and GSH; and decreases in the contents of Na+ and Cl ions, and glucose (a photosynthesis repressor); maintenance of a fine tuning among SOD, APX, and GR enzymes; and higher minimization of ROS (O.2, H2O2) and lipid peroxidation finally led to a higher promotion in photosynthesis and growth.

Most of the terrestrial deep subsurfaces are oligotrophic environments in which some gases, mainly H2, CH4 and CO2, play an important role as energy and/or carbon sources. In this work, we assessed their biotic and abiotic origin in samples from subsurface hard-rock cores of the Iberian Pyrite Belt (IPB) at three different depths (414, 497 and 520 m). One set of samples was sterilized (abiotic control) and all samples were incubated under anaerobic conditions. Our results showed that H2, CH4 and CO2 remained low and constant in the sterilized controls while their levels were 4, 4.1 and 2.5 times higher respectively, in the unsterilized samples compared to the abiotic controls. The δ13CCH4-values measured in the samples (range −31.2 to −43.0 ‰) reveals carbon isotopic signatures that are within the range for biological methane production. Possible microorganisms responsible for the biotic production of the gases were assessed by CARD-FISH. The analysis of sequenced genomes of detected microorganisms within the subsurface of the IPB allowed to identify possible metabolic activities involved in H2 (Rhodoplanes, Shewanella and Desulfosporosinus), CH4 (Methanobacteriales) and CO2 production. The obtained results suggest that part of the H2, CH4 and CO2 detected in the deep subsurface has a biological origin.  相似文献

Nitric oxide (NO) is a bioactive gaseous, multifunctional molecule playing a central role and mediating a variety of physiological processes and responses to biotic and abiotic stresses including heavy metals. The present study investigated whether NO applied exogenously as sodium nitroprusside (SNP) has a protective role against arsenic (As) toxicity (applied as sodium arsenate) in Vigna radiata (mung bean) germinating seeds. Treatment with 75???M SNP significantly improved mung bean seed germination, growth, and decreased the As-accumulation. Furthermore, As-induced oxidative stress measured in terms of malondialdehyde and H2O2 contents was lesser upon supplementation of SNP indicating a reactive oxygen species scavenging activity of NO. In addition, supplementation of SNP markedly decreased the activity of superoxide dismutase and stimulated catalase, ??-amylase, protease and slightly changed the H+-ATPase activity.  相似文献

Plants are exposed to a plethora of abiotic stresses such as drought, salinity, heavy metal and temperature stresses at different stages of their life cycle, from germination to seedling till the reproductive phase. As protective mechanisms, plants release signaling molecules that initiate a cascade of stress-signaling events, leading either to programmed cell death or plant acclimation. Hydrogen sulfide (H2S) and nitric oxide (NO) are considered as new ‘gasotransmitter’ molecules that play key roles in regulating gene expression, posttranslational modification (PTM), as well as cross-talk with other hormones. Although the exact role of NO in plants remains unclear and is species dependent, various studies have suggested a positive correlation between NO accumulation and environmental stress in plants. These molecules are also involved in a large array of stress responses and act synergistically or antagonistically as signaling components, depending on their respective concentration. This study provides a comprehensive update on the signaling interplay between H2S and NO in the regulation of various physiological processes under multiple abiotic stresses, modes of action and effects of exogenous application of these two molecules under drought, salt, heat and heavy metal stresses. However, the complete picture of the signaling cascades mediated by H2S and NO is still elusive. Recent researches indicate that during certain plant processes, such as stomatal closure, H2S could act upstream of NO signaling or downstream of NO in response to abiotic stresses by improving antioxidant activity in most plant species. In addition, PTMs of antioxidative pathways by these two molecules are also discussed.  相似文献

Hydrogen sulfide (H2S) has been recognized as a toxic gas and environment pollutant. So, it is seldom regarded as a therapeutic gas. H2S has been recognized recently as a novel gaseous messenger and serves as an important neuromodulator in the central nervous system. Many researches have been focused on the protective role of H2S in treatment of several diseases. Like nitric oxide (NO) and carbon monoxide (CO), which are considered as two gaseous transmitters, H2S has been regarded as the third one. Recent studies provided evidence that H2S exerted antioxidant and anti-apoptotic effects, which protected neurons, cardiomyocytes, pancreatic β-cells and vascular smooth muscle cells against oxidative stress by scavenging reactive oxygen species (ROS) and reactive nitrogen species (RNS). It has been known that multiple factors, including oxidative stress, free radicals and neuronal nitric oxide syntheses as well as abnormal inflammatory responses are involved in the mechanism underlying the brain injury after acute CO poisoning. Studies have shown that free radical scavengers can display neuroprotective properties. Therefore, we hypothesize that H2S might be an interesting potential strategy for curing acute CO poisoning.  相似文献

Shewanella oneidensis is a model species for aquatic ecosystems and plays an important role in bioremediation, biofuel cell manufacturing and biogeochemical cycling. S. oneidensis MR-1 is able to generate hydrogen sulfide from various sulfur species; however, its catalytic kinetics have not been determined. In this study, five in-frame deletion mutants of S. oneidensis were constructed and their H2S-producing activities were analyzed. SirA and PsrA were the two major contributors to H2S generation under anoxic cultivation, and the optimum SO32− concentration for sulfite respiration was approximately 0.8 mM, while the optimum S2O32− concentration for thiosulfate respiration was approximately 0.4 mM. Sulfite and thiosulfate were observed to interfere with each other during respiration, and a high concentration of sulfite or thiosulfate chelated extracellular free-iron but did not repress the expression of sirA or psrA. Nitrite and nitrate were two preferred electron acceptors during anaerobic respiration; however, under energy-insufficient conditions, S. oneidensis could utilize multiple electron acceptors simultaneously. Elucidiating the stoichiometry of H2S production in S. oneidensis would be helpful for the application of this species in bioremediation and biofuel cell manufacturing, and would help to characterize the ecophysiology of sulfur cycling.  相似文献

Although GR2(SO4 2-) can be easily formed by abiotic synthesis, the biotic formation of hydroxysulphate as a single iron(II-III) mineral in microbial culture and its characterization was not achieved. This study was carried out to investigate the sole formation of GR2(SO4 2-) during the reduction of γ-FeOOH by a dissimilatory iron-respiring bacterium, Shewanella putrefaciens CIP 8040T. Reduction experiments were performed in a non-buffered medium devoid of organic compounds, with 25 mM of sulphate and with a range of lepidocrocite concentrations with H2 as the electron donor under nongrowth conditions. The resulting biogenic solids, after iron-respiring activity, were characterized by X-ray diffraction (XRD), transmission Mössbauer spectroscopy (TMS) and electron microscopy (SEM and TEM). The sulphate has been identified as the intercalated anion by diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS). In addition, the structure of this sulphate anion was discussed. Our experimental study demonstrated that, under H2 atmosphere, the biogenic solid was a GR2(SO4 2-), as the sole iron(II-III) bearing mineral, whatever the initial lepidocrocite concentration. The crystals of the biotically formed GR2(SO4 2-) are significantly larger than those observed for GR2(SO4 2-) obtained through abiotic preparation, < 15 μ m diameter as against 0.5–4 μm, respectively.  相似文献

Industrial emissions of SO2 and NOx, resulting in the formation and deposition of sulfuric and nitric acids, affect the health of both terrestrial and aquatic ecosystems. Since the mid-late 20th century, legislation to control acid rain precursors in both Europe and the US has led to significant declines in both SO4–S and H+ in precipitation and streams. However, several authors noted that declines in streamwater SO4–S did not result in stoichiometric reductions in stream H+, and suggested that observed reductions in base cation inputs in precipitation could lessen the effect of air pollution control on improving stream pH. We examined long-term precipitation chemistry (1978–2010) from nearly 30 sites in the US and Europe that are variably affected by acid deposition and that have a variety of industrial and land-use histories to (1) quantify trends in SO4–S, H+, NH4–N, Ca, and NO3–N, (2) assess stoichiometry between H+ and SO4–S before and after 1990, and (3) examine regional synchrony of trends. We expected that although the overall efforts of developed countries to reduce air pollution and acid rain by the mid-late 20th century would tend to synchronize precipitation chemistry among regions, geographically varied patterns of fossil fuel use and pollution control measures would produce important asynchronies among European countries and the United States. We also expected that control of particulate versus gaseous emission, along with trends in NH3 emissions, would be the two most significant factors affecting the stoichiometry between SO4–S and H+. Relationships among H+, SO4–S, NH4–N, and cations differed markedly between the US and Europe. Controlling for SO4–S levels, H+ in precipitation was significantly lower in Europe than in the US, because (1) alkaline dust loading from the Sahara/Sahel was greater in Europe than the US, and (2) emission of NH3, which neutralizes acidity upon conversion to NH4 +, is generally significantly higher in Europe than in the US. Trends in SO4–S and H+ in precipitation were close to stoichometric in the US throughout the period of record, but not in Europe, especially eastern Europe. Ca in precipitation declined significantly before, but not after 1990 in most of the US, but Ca declined in eastern Europe even after 1990. SO4–S in precipitation was only weakly related to fossil fuel consumption. The stoichiometry of SO4–S and H+ may be explained in part by emission controls, which varied over time and among regions. Control of particulate emissions reduces alkaline particles that neutralize acid precursors as well as S-containing particulates, reducing SO4–S and Ca more steeply than H+, consistent with trends in the northeastern US and Europe before 1990. In contrast, control of gaseous SO2 emissions results in a stoichiometric relationship between SO4–S and H+, consistent with trends in the US and many western European countries, especially after 1991. However, in many European countries, declining NH3 emissions contributed to the lack of stoichiometry between SO4–S and H+.Recent reductions in NOx emissions have also contributed to declines in H+ in precipitation. Future changes in precipitation acidity are likely to depend on multiple factors including trends in NOx and NH3 emission controls, naturally occurring dust, and fossil fuel use, with significant implications for the health of both terrestrial and aquatic ecosystems.  相似文献

Reactive oxygen species (ROS) act as signaling molecules for regulating plant responses to abiotic and biotic stress and there exist source- and kind-specific pathways for ROS signaling. Recently, we created a novel system for producing H2O2 in Arabidopsis chloroplasts by chemical-dependent thylakoid membrane-bound ascorbate peroxidase (tAPX) silencing using an estrogen-inducible RNAi method. Microarray analysis revealed that the expression of a large set of genes was altered in response to tAPX silencing, some of which are known to be involved in pathogen response/resistance. Furthermore, we found that tAPX silencing enhances the levels of salicylic acid (SA) and the response to SA, a central regulator for biotic stress response. In this addendum, we describe the relationship between chloroplastic H2O2 and SA in stress response, and discuss the function of the kind- and source-specific ROS signaling in SA-mediated stress response.  相似文献
