Microbial mobilization of sediment calcium-bound P constitutes an important process of P cycling in aquatic environments. The present study was conducted to identify the bacterial community responsible for inorganic phosphate solubilization in tropical oxbow lakes. Fifty eight phosphate-solubilizing bacteria were isolated from bottom soil, water, and fish gut and examined for solubilization of tricalcium phosphate. Results revealed aquatic PSB to be low-to-moderately capable in P solubilization (mean: 33.5 mg P L?1; range: 6.3–68.8 mg P L?1), and bacteria from wetland sediment and water were more effective than those from fish gut. The PSB were identified to belong to diverse genera, viz. Bacillus, Brevibacillus, Enterobacter, Agrobacterium, Pseudomonas, Acinetobacter, Microbacterium, Curtobacterium, Stenotrophomonas and Novosphingobium. The findings help in understanding the microbial role in inorganic P solubilization and identifying important P solubilizers in freshwater environments.  相似文献

Isolation and characterization of fluorescent pseudomonads with high phosphate-solubilizing ability is reported from the alkaline and calcium-rich soils with low P availability in the cold desert region of Lahaul and Spiti in the trans-Himalayas of India. Of 216 phosphate-solubilizing isolates, 12 exhibiting high solubilization of tricalcium phosphate (TCP) in NBRIP liquid culture were identified as Pseudomonas trivialis, P. poae, P. fluorescens, and Pseudomonas spp. on the basis of phenotypic features, whole-cell fatty acids methyl ester (FAME) profiles, and 16S rDNA sequencing. These isolates also showed relatively high solubilization of North Carolina rock phosphate (NCRP) in comparison to the solubilization of Mussoorie rock phosphate (MRP) and Udaipur rock phosphate (URP). The solubilization of phosphate substrates by P. trivialis and P. poae is reported for the first time.  相似文献

【背景】土壤中大部分磷元素是以难溶性磷酸盐的形式存在,不能被农作物有效利用,而传统化学肥料会带来环境污染等问题。【目的】解决土壤磷缺失现状,开发新型、安全、高效的微生物菌肥。【方法】取武汉科技大学图书馆后土壤为试验材料,筛选出一株高效解磷菌。通过个体形态鉴定、生理生化鉴定、16S rRNA基因序列分析鉴定菌株,以NBRIP为基础培养基进行条件优化,借助高效液相色谱进行细菌解磷机理探究。【结果】所筛选的高效解磷菌株为唐菖蒲伯克霍尔德氏菌(Burkholderia gladioli)。在20种氨基酸中,D-蛋氨酸对菌株的生长和溶磷促进作用最好,促进效果达到19.09%和16.16%,甲酸钠对菌株的生长和溶磷有抑制效果,抑制效果达到39.08%和10.66%。该菌株通过分泌葡萄糖醛酸、D-L-苹果酸等有机酸溶解环境中的磷酸盐,将菌株制作成菌肥对辣椒幼苗有明显的促生长作用。【结论】利用唐菖蒲伯克霍尔德氏菌(Burkholderia gladioli)分泌有机酸溶解土壤中的磷酸盐,可为生物肥料的制备和应用提供一定的理论参考。  相似文献

The effect of phosphate-solubilizing bacteria (PSB) application on phosphorus (P) availability in reclaimed soil in coal mining subsidence region was investigated. Seven treatments were carried out including control, chicken manure (CM), PSB, PSB + tricalcium phosphate (TCP), CM?+?TCP, PSB?+?ground phosphate rock (GPR) and CM?+?GPR. The results showed soil Olsen-P concentration and phosphatase level as well as the yield of pakchoi (Brassica chinensis L.) were significantly higher in PSB application treatments compared to the corresponding CM application treatments. Soil phosphatase, invertase and urease contents were increased most significantly in PSB treatment, 1.18-, 1.31- and 2.32-fold higher than those in the control, respectively. Soil Ca2-P, Ca8-P, Fe-P and Al-P concentrations exhibited the greatest increases in PSB?+?TCP treatment, while occluded-P showed minor changes in different treatments. Application of PSB fertilizer reduced the transformation of Olsen-P to Ca10-P, thus increasing P availability in reclaimed soil of coal mining subsidence area.  相似文献

Biofertilizers offer alternative means to promoting cultivation of medicinal plants less dependent on chemical fertilizers. Present study was aimed at evaluating the potential of phosphate-solubilizing bacteria (PSB) Burkholderia gladioli MTCC 10216, B. gladioli MTCC 10217, Enterobacter aerogenes MTCC 10208 and Serratia marcescens MTCC 10238 for utilizing Mussoorie rock phosphate (MRP) to enhance plant growth, and stevioside (ST) and rebaudioside-A (R-A) contents of Stevia rebaudiana. The solubilization of MRP by PSB strains varied from 1.4 to 15.2 μg ml−1, with the highest solubilization by Enterobacter aerogenes 10208. The PSB treatment increased the growth and ST and R-A contents of plants. Plant growth and stevioside contents were more pronounced with plants treated with a mixture of strains and grown in MRP amended soil compared to the unamended soil. The increment in shoot length (47.8%), root length (17.4%), leaf dry weight (164%), stem dry weight (116%), total shoot biomass (136%) resulted in enhanced productivity of ST (291%) and R-A (575%) in plants inoculated with mixture of PSB as compared to the uninoculated plants. The soils of PSB treated plants contained more available P than the soils of uninoculated plants (increase of 86–576%). PSB inoculated plants also recorded higher P content (64–273% increase) compared to uninoculated plants. The PSB strains differed in the extent of rhizosphere colonization, carbon source utilization pattern and whole cell fatty acids methyl esters composition.  相似文献

解无机磷细菌能够溶解土壤中的难溶性磷酸盐,增加土壤有效磷含量,促进植物生长。以一株杨树根际土壤中筛选得到的解无机磷细菌Mp1-Ha4为研究对象,利用分子生物学的方法对该菌株进行鉴定,测定了其对磷酸钙、磷酸铝和磷酸铁的解磷能力,并对该菌株9 d内的磷酸钙溶解动力学进行了研究。结果表明,解无机磷细菌Mp1-Ha4为一株西地西菌Cedecea sp.,其对磷酸钙的溶解能力远强于对磷酸铝和磷酸铁的溶解能力。在NBRIP液体培养基中,该菌株对磷酸钙的溶解能力达到了497.4 mg/L,在其对磷酸钙解磷过程中,培养基pH及可滴定酸度与解磷量分别呈显著负、正相关。高效液相色谱分析显示,该菌株在解磷过程中分泌了大量有机酸,主要包括α-酮戊二酸,酒石酸和苹果酸。分泌有机酸,降低环境pH可能是解无机磷细菌西地西菌(Cedecea sp.)Mp1-Ha4溶解难溶性磷酸盐的主要机制,同时该菌株对磷酸钙的高效溶解作用使其具有较大的研究和应用前景。  相似文献

A phosphate solubilizing bacterium ZB was isolated from the rhizosphere soil of Araucaria, which falls into the species Pantoea agglomerans. Optimization for phosphate solubilization by strain ZB was performed. At optimum culture conditions, the isolate showed great ability of solubilizing different insoluble inorganic phosphate sources viz. Ca3(PO4)2 (TCP), Hydroxyapatite (HP), CaHPO4, AlPO4, FePO4 along with rock phosphates (RPs). Inoculation with planktonic cells was found to enhance dissolved phosphorous as compared to that achieved by symplasma inoculation. Besides inoculation with different status of cells, pre-incubation could also exert a great effect on phosphate solubilization ability of P. agglomerans. When isolate ZB was cultured with glucose as carbon sources, phosphorous was more efficiently dissolved from HP and RP without pre-incubation in comparison to that obtained with pre-cultivation. Pre-cultivation, however, was more suitable for P solubilization than no pre-cultivation when bacteria were grown with xylose. A positive correlation was detected between the production of organic acids and phosphate solubilization. P. agglomerans ZB possessed many plant growth promotion traits such as N2 fixation and production of indole 3-acetic acid, phytase, alkaline phosphatase. Pot experiment showed inoculation with single isolate ZB or biofertilizer prepared from semi-solid fermentation of isolate ZB with spent mushroom substrate (SMS) compost could enhance plant growth with respect to number of leaves, plant leave area, stem diameter, root length, root dry mass, shoot dry mass and biomass when compared to the abiotic control, revealing strain ZB could be a promising environmental-friendly biofertilizer to apply for agricultural field.

An ecological survey was conducted to characterize 4800 bacterial strains isolated from the root-free soil, rhizosphere, and rhizoplane of Prosopis juliflora growing in alkaline soils. Of the 4800 bacteria, 857 strains were able to solubilize phosphate on plates. The incidence of phosphate-solubilizing bacteria (PSB) in the rhizoplane was highest, followed by rhizosphere and root-free soil. Eighteen bacterial strains out of 857 PSB were able to produce halo at 30°C in a plate assay in the presence of 5% salt (NaCl) and solubilize tricalcium phosphate in National Botanical Research Institute's phosphate growth medium (NBRIP) broth, in the presence of various salts, pHs, and temperatures. Among the various bacteria tested, NBRI4 and NBRI7 did not produced halo in a plate assay at 30°C in the absence of salt. Contrary to indirect measurement of phosphate solubilization by plate assay, the direct measurement of phosphate solubilization in NBRIP broth assay always resulted in reliable results. The phosphate solubilization ability of NBRI4 was higher than in the control in the presence of salts (NaCl, CaCl2, and KCl) at 30°C. Phosphate solubilization further increased in the presence of salts at 37°C as compared with 30°C. At 37°C, CaCl2 reduced phosphate solubilization ability of NBRI4 compared with the control. An efficient phosphate-solubilizing plant growth–promoting Acinetobacter rhizosphaerae strain BIHB 723 exhibited significantly higher solubilization of tricalcium phosphate (TCP) than Udaipur rock phosphate (URP), Mussoorie rock phosphate (MRP) and North Carolina rock phosphate (NCRP). Qualitative and quantitative differences were discerned in the gluconic, oxalic, 2-keto gluconic, lactic, malic and formic acids during the solubilization of various inorganic phosphates by the strain. Gluconic acid was the main organic acid produced during phosphate solubilization. Formic acid production was restricted to TCP solubilization and oxalic acid production to the solubilization of MRP, URP and NCRP. A significant increase in plant height, shoot fresh weight, shoot dry weight, root length, root dry weight, and root, shoot and soil phosphorus (P) contents was recorded with the inoculated treatments over the uninoculated NP0K or NPTCPK treatments. Plant growth promotion as a function of phosphate solubilization suggested that the use of bacterial strain would be a beneficial addition to the agriculture practices in TCP-rich soils in reducing the application of phosphatic fertilizers.  相似文献

Eupenicillium parvum was recorded for first time during isolation of phosphate-solubilizing microorganisms from the tea rhizosphere. The fungus developed a phosphate solubilization zone on modified Pikovskaya agar, supplemented with tricalcium phosphate. Quantitative estimation of phosphate solubilization in Pikovskaya broth showed high solubilization of tricalcium phosphate and aluminium phosphate. The fungus also solubilized North Carolina rock phosphate and Mussoorie rock phosphate, and exhibited high levels of tolerance against desiccation, acidity, salinity, aluminium, and iron. Solubilization of inorganic phosphates by the fungus was also observed under high stress levels of aluminium, iron, and desiccation, though the significant decline in phosphate solubilization was marked in the presence of aluminium than iron. The fungal isolate showed 100 % identity with E. parvum strain NRRL 2095 ITS 1, 5.8S rRNA gene and ITS 2, complete sequence; and 28S rRNA gene, partial sequence.  相似文献

Secretion of organic acids is an important mechanism for phosphate solubilizing bacteria (PSB) to dissolve insoluble phosphorus in soil. However, the composition of organic acids produced by PSB in the presence of different phosphorus compounds is poorly known, and little is known about the ability of PSB to degrade pollutants in sediment. In this study, we isolated a strain Pseudomonas sp. PSB12 from the sediment of the Qihe River. PSB12 had maximum phosphate solubilization index (SI) of 3.86 on Pikovskaya's agar medium. The phosphate solubilizing activity was associated with the release of organic acids produced from glucose, while the composition of organic acids produced by PSB12 was dependent on the phosphorus forms. When initial soluble phosphorus was insufficient (in MP1 and MP2 media), gluconic acid was the predominant organic acid. In contrast, formic acid, butyrate, and propanedioic acid were the main organic acids produced when only soluble phosphorus (MP3) was supplied. RT-PCR indicated that the expression of glucose dehydrogenase gene (gcd) of PSB12 was two- to four-fold higher in MP1 than in MP3. PSB12 also possessed the phenol hydroxylase gene (phe) suggesting that phenol could be used as the carbon source to dissolve insoluble phosphorus. PSB12 is a potential candidate for in situ bioremediation and for promoting plant growth in soil contaminated by phenol with low levels of soluble phosphorus.  相似文献

Phosphate solubilizing bacteria (PSB) play a significant role in plant P nutrition by their effect on soil P dynamics and their subsequent ability to make P available to plants via solubilization and mineralization processes. This study aimed to evaluate the effect of separate and combined use of indigenous PSB, poultry manure (PM) and compost on solubilization and mineralization of rock phosphate (RP) and their subsequent effect on growth and P accumulation of maize (Zea mays L.). A group of fifty seven bacteria were isolated from the rhizosphere/rhizoplane of maize that had been grown in soils collected from varying altitudes (655–2,576 m) of the mountain region of Rawalakot, Azad Jammu and Kashmir, Pakistan. After screening, the capacity of eleven isolates to solubilize mineral phosphate was quantitatively evaluated using insoluble Ca3(PO4)2 in culture medium as a time course study through spectrometer. The growth hormone producing (IAA) capacity of the isolates was also determined. Furthermore, five potential isolates were tested for their ability to increase P release capacity (mineralization) of insoluble RP in an incubation study. The effect of PSB inoculation on maize was determined in a completely randomized greenhouse experiment where root and shoot biomass and P accumulation in plants were assessed. The P solubilization index of selected isolates varied from 1.94 to 3.69, while the P solubilization efficiency ranged between 94.1% and 269.0%. The isolates MRS18 and MRS27 displayed the highest values. The P solubilization in the liquid medium was maximum at 6 and 9 days of incubation ranging between 9.91 and 44.04 µgmL?1 and the isolates MRS27 and MRS34 exhibited the highest solubilization. Six isolates showed additional capability of producing IAA ranging between 2.66 and 28.41 µgmL?1. Results of the incubation study indicated that P release capacity (P mineralization) of RP-amended soil varied between 6.0 and 11.8 µgPg?1 that had been significantly increased to 30.6–36.3 µgPg?1 (maximum value) when PSB were combined with RP. The combined application of PSB and organic amendments (PM, compost) with RP further increased P mineralization by releasing a maximum of 37.7 µgPg?1 compared with separate application of RP (11.8 µgPg?1) and organic amendments (21.5 and 16.5 µgPg?1). The overall effect of PSB (as a group) with RP over RP alone on maize growth showing a relative increase in shoot length 21%, shoot fresh weight 42%, shoot dry weight 24%, root length 11%, root fresh weight 59%, root dry weight 35% and chlorophyll content 32%. This study clearly indicates that use of PSB, and organic amendments with insoluble RP could be a promising management strategy to enhance P availability in soil pool and improve plant growth in intensive cropping systems.  相似文献

Screening soil samples collected from a diverse range of slightly alkaline soil types, we have isolated 22 competent phosphate solubilizing bacteria (PSB). Three isolates identified as Pantoea agglomerans strain P5, Microbacterium laevaniformans strain P7 and Pseudomonas putida strain P13 hydrolyzed inorganic and organic phosphate compounds effectively. Bacterial growth rates and phosphate solubilization activities were measured quantitatively under various environmental conditions. In general, a close association was evident between phosphate solubilizing ability and growth rate which is an indicator of active metabolism. All three PSB were able to withstand temperature as high as 42°C, high concentration of NaCl upto 5% and a wide range of initial pH from 5 to 11 while hydrolyzing phosphate compounds actively. Such criteria make these isolates superior candidates for biofertilizers that are capable of utilizing both organic and mineral phosphate substrates to release absorbable phosphate ion for plants.  相似文献

Native microflora present in the alkaline vertisols and two phosphate-solubilizing bacteria (PSB) isolated from soil using conventional screening media could not release phosphorus from alkaline Indian vertisol soils supplemented with carbon and nitrogen sources. The two PSBs could solubilize both rock phosphate and di-calcium phosphate in unbuffered media but failed to solubilize rock phosphate in buffered media. The organic acids secreted by these PSBs were 20–50 times less than that required to solubilize phosphorus from alkaline soil.  相似文献

In the research, the single-and dual effects of phosphate-solubilizing bacteria (PSB) (B0, Pseudomonas sp. FA1, and Bacillus simplex UT1) and silicon (Si) (0, 150, 300, and 600 mg kg−1 used as silicic acid) on P uptake by sorghum (Sorghum bicolor L.) plant fertilized with soluble or insoluble P (rock phosphate—RP) were studied via a perlite-potted experiment. Moreover, the effects of various treatments on morphological (shoot and root dry weight), nutritional (the uptake of Si and K) and physiological parameters (activity of catalase, superoxide dismutase, and peroxidase enzymes) of this plant were also measured. When grown in RP-fertilized medium compared with those grown in soluble P-fertilized medium, both shoot biomass and root biomass of sorghum plants were noticeably diminished. The PSB strains and Si levels independently improved all the aforementioned parameters. Use of Si and PSB strains to sorghum plants grown in soluble P or insoluble P medium significantly augmented P use efficiency. Silicon not only augmented the uptake of P from sparingly soluble-P source (RP), but also augmented uptake of P from water-soluble P source. Both B. simplex UT1 and Pseudomonas sp. FA1 indicated a meaningful betterment in sorghum plant dry matter and uptake of P (and K and Si) under both soluble and insoluble P fertilization conditions with Pseudomonas sp. FA1 being more efficacious than B. simplex UT1. But, the dual use of the PSB with Si resulted in the greatest increase in sorghum plant P uptake and other measured growth indices. Application of 600 mg Si kg−1 and Pseudomonas sp. FA1 significantly augmented the P shoot concentration of sorghum plant fertilized with RP to an sufficient level (> 0.3%) in the range of P-fertilized sorghum plants. Therefore, in addition to PSB utilization, Si should be considered as soil amendment in agricultural soils inadequate in plant-available Si as a means of sustainable agriculture with respect to possible savings of scarce P resources.

Five phosphate-solubilizing bacteria (PSB) used in this study were isolated based on their ability to solubilize tricalcium phosphate (TCP) in Pikovskaya’s medium. Among the tested bacterial strains Burkholderia sp. strain CBPB-HIM showed the highest solubilization (363 μg of soluble P ml−1) activity at 48 h of incubation. Further, this strain has been selected to assess its shelf life in nutrient-amended and -unamended clay, rice bran and rock phosphate (RP) pellet-based granular formulation. The results showed that the maximum viability of bacterium was observed in clay and rice bran (1:1) + 10% RP pellets than clay-RP pellets, irrespective of tested storage temperatures. Further, clay and rice bran (1:1) + 10% RP pellets amended with 1% glucose supported the higher number of cells compared to glycerol-amended and nutrient-unamended pellets. In this carrier solubilization of Morocco rock phosphate (MRP) by Burkholderia sp. strain CBPB-HIM was also investigated. The maximum of water and bicarbonate extractable P (206 and 245 μg P g−1 of pellet respectively) was recorded in clay and rice bran (1:1) + 10% RP pellets amended with 1% glucose and glycerol respectively on day 5 of incubation. Therefore, this study proved the possibility of developing granular inoculant technology combining clay, rice bran and RP as substrates with phosphate-solubilizing Burkholderia.  相似文献

Background  

Phosphorus deficiency is a major constraint to crop production due to rapid binding of the applied phosphorus into fixed forms not available to the plants. Microbial solubilization of inorganic phosphates has been attributed mainly to the production of organic acids. Phosphate-solubilizing microorganisms enhance plant growth under conditions of poor phosphorus availability by solubilizing insoluble phosphates in the soil. This paper describes the production of organic acids during inorganic phosphate solubilization and influence on plant growth as a function of phosphate solubilization by fluorescent Pseudomonas.  相似文献

The mineral phosphate-solubilizing (MPS) activity of a Pantoea agglomerans strain, namely MMB051, isolated from an iron-rich, acidic soil near Ciudad Piar (Bolívar State, Venezuela), was characterized on a chemically defined medium (NBRIP). Various insoluble inorganic phosphates, including tri-calcium phosphate [Ca3(PO4)2], iron phosphate (FePO4), aluminum phosphate (AlPO4), and Rock Phosphate (RP) were tested as sole sources of P for bacterial growth. Solubilization of Ca3(PO4)2 was very efficient and depended on acidification of the external milieu when MMB051 cells were grown in the presence of glucose. This was also the case when RP was used as the sole P source. On the other hand, the solubilization efficiency toward more insoluble mineral phosphates (FePO4 and AlPO4) was shown to be very low. Even though gluconic acid (GA) was detected on culture supernatants of strain MMB051, a consequence of the direct oxidation pathway of glucose, inorganic-P solubilization seemed also to be related to other processes dependent on active cell growth. Among these, proton release by ammonium (NH4+) fixation appeared to be of paramount importance to explain inorganic-P solubilization mediated by strain MMB051. On the contrary, the presence of nitrate (NO3) salts as the sole N source affected negatively the ability of MMB051 cells to solubilize inorganic P.  相似文献

通过从牡丹根际土壤中分离筛选溶磷放线菌,得到一株具有较强溶磷能力的菌株PSPSA1,根据形态特征、生理生化特性以及16S rDNA序列分析对菌株进行鉴定,并研究其溶磷遗传稳定性及溶磷特性.菌株PSPSA1被鉴定为白网链霉菌,具有较好的溶磷遗传稳定性.在不同磷源培养液中溶磷量依次为磷酸钙(158.5 mg·L-1)>磷酸铝(139.9 mg·L-1)>磷酸铁(127.7 mg·L-1)>卵磷脂(45.6 mg·L-1),在无机磷培养液中的溶磷量均与pH呈现显著负相关性,在有机磷培养液中的溶磷量与pH没有显著相关性.在不同碳源条件下的溶磷量依次为乳糖>葡糖糖>麦芽糖>果糖>蔗糖>淀粉>纤维素,在不同氮源条件下的溶磷量依次为蛋白胨>硝酸铵>硫酸铵>硝酸钾>尿素,以葡萄糖为碳源、蛋白胨为氮源时,菌株的溶磷量最高可达202.6 mg·L-1.土培60 d,单施菌株土壤有效磷含量比对照增加68.2%,菌株与有机肥混施土壤有效磷含量比单施有机肥增加76.7%.表明菌株PSPSA1能够溶解多种难溶磷,在土壤中溶磷效果显著,与有机肥混施其溶磷能力明显提高,有望成为高效生物磷肥的优良菌种.  相似文献

Plant growth-promoting rhizobacteria (PGPR) are known to influence plant growth by various direct or indirect mechanisms. A total of 216 phosphate-solubilizing bacterial isolates were isolated from different rice rhizospheric soil in Northern Thailand. These isolate were screened in vitro for their plant growth-promoting activities such as solubilization of inorganic phosphate, ammonia (NH3), catalase and cell wall-degrading enzyme activity. It was found that 100% solubilized inorganic phosphate, 77.77% produced NH3 and most of the isolates were positive for catalase. In addition, some strains also produced cell wall-degrading enzymes such as protease (7%), chitinase (1%), cellulase (3%) and β-glucanase (3%), as evidenced by phenotypic biochemical test and quantitative assay using spectrophotometry. The isolates could exhibit more than two or three plant growth-promoting (PGP) traits, which may promote plant growth directly or indirectly or synergistically. Part of this study focused on the effect of NaCl, temperature, and pH on a specific the bacterial isolate Acinetobacter CR 1.8. Strain CR 1.8 was able to grow on up to 25% NaCl, between 25 and 55°C, and at pH 5–9. Maximum solubilization of tricalcium phosphate and aluminium phosphate was obtained at neutral pH, and 37°C.
