Effect of dexamethasone (2.0 mg/kg) on activity of enzymes hydrolyzing proteinaceous and carbohydrate food components in the intestine of the starlet Acipenser ruthenus is studied. Dexamethasone modifies the activity of proteinases more than of glycosidases. As a rule, the hormone significantly decreases the level of proteolytic and general amylolytic activity of the intestinal mucosa and chyme in comparison with intact fish on the first day into the experiment and increases it on the 7th or 14th day. The dynamics of activity was different in enzymes of different chains (glycosidase and proteinase) and preparations (the mucosa and chyme).  相似文献

Observation of the influence of testosterone (0.7 mg/kg) on the activity of glycosidases and proteinases, which function in the chyme and the mucosa of the sterlet intestine (Acipenser ruthenus), revealed a decrease in enzyme activity of both chains as against that one typical for intact individuals. The activity of the investigated enzymes changes in both the experimental and control groups of fish. Nevertheless, they have unlike dynamics of this activity. In a number of cases, we indicated a significant increase in enzyme activity of both chains in comparison to the control group. We also observed an increase in activity of chyme glycosidases, which was significant on the 21st day as compared to intact fish.  相似文献

The present study was conducted to evaluate the effects of Grobiotic®‐A, a commercial prebiotics, when administered in feed on the growth performance, plasma thyroid hormones and mucosal immunity of great sturgeon (Huso huso). The commercial prebiotic mixture was supplemented in the diets at four different levels (i.e. 0.0% as control, 0.5%, 1% and 2%, in three replicates, 20 fish per replicate) and fed to the fish for an 8‐week period wherein 240 fish were cultured in 1,800‐L fiberglass tanks that formed part of a flow‐through system. Water temperature was maintained at 20.4 ± 1.5°C. Significant changes in growth performance parameters were observed, but only in those groups fed with 1% and 2% prebiotics. Specifically, marked improvements relative to the control group were observed in percentage weight gain, body weight gain, feed conversion ratio and specific growth rate in prebiotic‐fed fish. The levels of plasma thyroid hormones, specifically thyroxine and thyroid stimulating hormones were significantly elevated in the group receiving 2% prebiotics. Activities of lysozyme and alkaline phosphatase in skin mucus were significantly enhanced in prebiotics‐fed groups, particularly at an inclusion level of 1% and higher (2% group compared to the control). Inhibitory activity of the skin mucus against pathogens, particularly Streptococcus iniae and Yersinia ruckeri, was significantly improved following prebiotic feeding. Taken together, dietary inclusion of GroBiotic®‐A promoted growth, modulated thyroid hormones, and enhanced mucosal immunity of H. huso. This prebiotic mixture has the potential for use in improving the growth performance and health status of farmed great sturgeon.  相似文献

The concentration of mercury in muscles of experimental carp Cyprinus carpio whose diet contained high levels of mercury gradually increased and in 6 months became 5.4 times higher than in the control group. This increase was accompanied by a decrease in the activity of glycosidases in the intestinal mucosa, as a rule, and an increase in the activity of proteases. The observed effects were most pronounced in the optimum pH range of the enzymes. The maximum rates of the reactions catalyzed by proteinases in the fish of the experimental group were equal to or slightly higher than in the control throughout the experiment, whereas the Michaelis constant values, conversely, were much lower. The maximum rates of the reaction catalyzed by glycosidases decreased 2–3 months after the beginning of the experiment (by 14–21%), and the Michaelis constant values decreased almost throughout the experiment.  相似文献

Effect of temperature on proteinases activities of enteral microbiota and of intestinal mucosa was studied in five fish species (roach Rutilus rutilus, crucian carp Carassius carassius, common perch Perca fluviatilis, pike-perch Zander lucioperca, and pike Esox lucius) belonging by the nutrition type to different ecological groups. Essential differences of temperature characteristics of proteinases of intestinal mucosa and of enteral microbiota are revealed in fish belonging to different ecological groups. The character of the t-function of proteinases of intestinal mucosa and enteral microbiota for casein and hemoglobin as a rule is different. The values of the apparent E act proteinases of intestinal mucosa for casein in most cases are higher than those of enteral microbiota, while those for hemoglobin, on the contrary, are lower. The lowest values of relative proteinase activities for casein in the zone of low temperatures (38 and 45.3% of the maximal activity) and the Eact value (less than 2.0 kcal/mol) are found at study of proteinases of enteral microbiota in common perch and crucian carp. The latter indicates a significant adaptability of the enteral microbiota proteinases of common perch and crucian carp to functioning at low temperatures.  相似文献

The effect of temperature on activity of proteinases of the stomach mucosa, intestinal mucosa, chime, and the entire body of kilka Cluperonella cultriventris is described. Significant differences are revealed in temperature characteristics (t o-function, Q 10, and E act) of the investigated enzymes. The temperature has a larger influence on the activity of intestinal proteinases than the activity of proteinases of the stomach, chime, and the whole kilka organism. The lowest value of E act within the temperature range of life of this species is characteristic of chime proteinases. Hemoglobinlytic proteinases of various tissues of kilka that substantiate, at pH 3.0 and 5.0 (the action zone of cathespsins), the processes of induced autolysis contribute to efficient alimentation of other fish species. The assumption is made that a high activity level of proteinases of the digestive tract, comparable with that of other fish species of the upper Volga, enables kilka that have assimilated to the Rybinsk Reservoir to feed efficiently within the temperature range characteristic of the water-bodies of this zone.  相似文献

For the first time, a complex investigation of characters of some enzymes effecting the membrane hydrolysis of food in the paddlefish Polyodon spatula is made. The zone of optimum values of temperature of alkaline phosphatase, maltase, and casein-lytic proteinases of the intestinal mucosa is determined within the range from 50 to 60°C, in α-amylase it is shifted towards lower temperatures—10–30°C. A high thermal stability is noted in all investigated enzymes. The maximum level of activity for α-amylase, alkaline phosphatase, and maltase of the intestinal mucosa in the paddlefish Polyodon spatula is within pH 7–9, 9–10, and 6–8, respectively; for casein-lytic proteinases the optimum is at pH 11. In the paddlefish Polyodon spatula, the interaction of nutrients and enzymes of the intestinal mucosa may cause a significant change in the activity level of the enzymes.  相似文献

The chronic effects that polychlorinated biphenyls (PCBs) have on the activities of proteinases and carbohydrates in intestinal mucosa and chyme in juvenile roach Rutilus rutilus (L.) have been studied for the first time. Upon consuming food with PCB 50.8 ng/g wet weight for 218 days, the exposition of fingerlings in aquariums with dirt bottoms (contents PCB 425.6 ng/g dry weight) reduces the proteolytic activity of mucosa to a greater degree than amylolytic activity; the activity of sucrase changes in different directions. In 2-year-old fish receiving PCB only with food (50.8 ng/g wet weight bottom), proteolytic and amylolytic activities of the mucosa are reduced 18 and 35% in comparison with the control (52 days); the sucrose activity is reduced 13% (52 days) and 22% (169 days). In most cases the relation of amylolytic activity to proteolytic activity in fish of the experimental groups is less than in the control. The enzyme activities in chyme change in different directions, depending on the terms and conditions of the experiment.  相似文献

Development of teeth on the pharyngeal bones of roach Rutilus rutilus and the effect of thyroid hormones on this development are investigated. The addition of exogenous triiodothyronine leads to accelerated development of the teeth, but the deficit of triiodothyronine (provoked by the addition of thiourea in the media) stimulates the retardation of this development. Change of developmental rate of the organism leads to change in the definitive state of the pharyngeal teeth formula. Owing to accelerated development, the number of teeth significantly decreases, and the formulas 5–5, 5–4, and 4–4 appear instead of the typical formula 6–5 in the control group and in the fish from natural populations. Retarded development of the organism leads to increased frequency of occurrence of the formula 6–6. The directed asymmetry in the numbers of pharyngeal teeth with the formula (6–5), most likely, is connected with different types of teeth development on the left and right pharyngeal bones.  相似文献

The dependence of the effects of serotonin and cholecystokinin on the activity of peptidases and glycosidases in the intestine of the carp Cyprinus carpio from the content of zinc and copper ions in different photoperiods (6: 18 and 0: 24 h) was studied. It was shown that the activity of enzymes in the intestine of fish under the influence of both hormones in conditions of light deprivation significantly increases, especially under the action of copper. The mechanisms of the influence of these factors on the activity of peptidases and glycosidases of fish intestines are discussed.  相似文献

Proteolytic activities of the intestinal mucosa, chyme, and enteral microbiota have been studied in a wide range of pH values in five fish species from the Cuciurgan reservoir (Moldova). Differences in pH dependence of the intestinal proteinase activity of fish are determined by their feeding type. The maximum activity of proteinases is found in the pumpkinseed Lepomis gibbosus. The minimum activity of proteinases has been demonstrated by the zander Zander lucioperca. The pH optimum of the mucosa and chyme in all fish species (except for the European perch Perca fluviatilis) is 10. The pH optimum of the intestinal microflora varies from 6.0 (in the common carp Cyprinus carpio) to 10 (in the crucian carp Carassius carassius), whereas that in the perch from the Cuciurgan and Rybinsk reservoirs is 7. The majority of fish species, mostly Zander lucioperca and Lepomis gibbosus, are characterized by high proteniase activity of the microbiota, in the pH ranging from 6 to 9. It is assumed that proteinases in the enteral microbiota of fish are able to make up for the relatively low activity of those synthesized by their digestive system in the range of low pH values.  相似文献

The role of thyroid hormones in the development of the locomotor and cardiac muscles of the barnacle goose (Branta leucopsis) was investigated. From 2 weeks of age, goslings were treated with thyroxine, triiodothyronine, or methimazole (a thyroid inhibitor). Birds were killed at 6 weeks (n = 5) or 9 weeks (n = 4) and various locomotor and cardiac muscle masses recorded and tissue samples taken for analysis of citrate synthase activity. The effects of thyroxine and triiodothyronine were not significantly different from each other, except in the case of the iliofibularis at 9 weeks. The mass-specific citrate synthase activity of the iliofibularis, semimembranosus muscles and liver were significantly increased by thyroid hormone treatment. Cardiac muscle showed a significant hypertrophy at 9 weeks of age following treatment with thyroxine. Hypothyroidism induced by treatment with methimazole exhibited the greatest effect on the pectoralis muscle, reducing citrate synthase activity by 33%. Mass-specific citrate synthase activity of the pectoralis and pectoralis mass (% body wt.) were found to be highly correlated (r 2 = 0.74) at 6 weeks of age. It is suggested that thyroid hormones may be involved in controlling the tissue-specific timing of the maturation of locomotor and cardiac muscles. Accepted: 16 September 1996  相似文献

The influence of exogenous triiodothyronine (0.25 ng/mL) on the activity glycosidases (maltase, amylolytic activity) and their sensitivity to Cu, Zn and Pb ions (25 mg/L) has been studied in the whole organism of juvenile blue bream Ballerus ballerus (L.). We have found that the treatment of fish with triiodothyronine resulted in an increase in amylolytic activity. Maltase activity is not affected. In addition, the exposure to exogenous triiodothyronine results in a decrease in the sensitivity of glycosidase to the in vitro action of the ions of biogenic metals Cu and Zn and in an increase in the sensitivity of the maltase to the action of Cu, Zn, and Pb ions.  相似文献

Using radioimmunological method, the level and dynamics of thyroid hormones, thyroxine (T4) and triiodothyronine (T3) were studied in eggs, embryos, and pre-larvae of the Russian sturgeon Acipenser güldenstadti L. The level of hormones in eggs was higher than in the individuals at the stage of hatching. In embryogenesis the content of T4 and T3 hormones changed simultaneously, while in pre-larvae, synchronization was absent. The maximal T4 values in sturgeon pre-larvae were recorded not long before transition to active nutrition. The T4 level was statistically significantly higher in eggs and larvae developed from the eggs with a high degree of fertility. The problem of participation of thyroid hormones in processes of development and formation of adaptive mechanisms in early ontogenesis of sturgeons is discussed.  相似文献

In new-born piglets' thyroid gland culture, thyrocytes were capable of synthesising thyroxine and triiodothyronine for over 12 days. A 48-hour decline of the medium pH to 6.45 did not affect further functional activity of the cells. Decrease in temperature to 26-28 degrees C augmented the thyroid hormones level as opposed to continuous cultivation at 37 degrees C. Degradation of the thyroid gland follicular structure induced a change in the hormones biosynthesis. The thyrocytes not arranged in the follicles mainly produced triiodothyronine in concentrations 5 to 8-fold higher than the T4 contents.  相似文献

Vitamin A is derived from provitamin A carotenoids, mainly beta-carotene, by beta-carotene 15,15'-monooxygenase (CMO1; EC 1.13.11.21). We previously found that enhancement of CMO1 mRNA expression was related to the levels of hormones, such as thyroid hormones, in chick duodenum. We investigated whether CMO1 expression was increased by triiodothyronine (T3), a thyroid hormone, using human intestinal Caco-2 BBe cells. Treatment of 7 days post-confluent Caco-2 BBe cells with T3 significantly enhanced CMO1 mRNA levels in both dose- and time-dependent manners. This T3-inducing effect on CMO1 mRNA level was blocked by actinomycin D. The levels of mRNAs for the thyroid hormone receptors TRalpha1 and TRbeta1 were significantly increased in 7 days post-confluent Caco-2 BBe cells. CMO1 enzyme activity was also significantly increased by T3 treatment in medium supplemented with fetal bovine serum. Furthermore, T3 treatment also increased the level of mRNA for lecithin:retinol acyltransferase (LRAT), but not those for cellular retinol-binding protein, type II (CRBPII) and retinal dehydrogenase 1 (RALDH1), in Caco-2 BBe cells. These results indicate that T3 is an important hormone for the regulation of vitamin A and beta-carotene metabolism-related gene expression in human small intestinal cells.  相似文献

The paper cites evidence on the activity of casein- and hemoglobinlytical proteinases of mucosa of the digestive tract of nine species of freshwater bony fish differing in their pattern of feeding, as well as in the whole organism of their potential prey. It was demonstrated that the ratio of proteinase activity with respect to casein and hemoglobin in the intestine mucosa of consumers and in the whole organism of their food items is different. In mucosa of fish intestine, the activity of caseinlytic proteinases is higher than the activity of hemoglobinlytic proteinases. In the whole organism of juvenile fish and invertebrates, on the other hand, the activity of hemoglobinlytic proteinases is higher.  相似文献
