The accumulation of phytic acid during development of lily (Lilium longiflorum Thunb.) pollen and its degradation during germination have been studied. A substantial amount of phytic acid accumulates in lily pollen by 5 days before anthesis, and little change occurs during subsequent maturation. Mature lily pollen contains 7 to 8 micrograms phytic acid per milligram pollen. Considerable degradation of phytic acid occurs by 15 minutes of incubation in glucose culture medium, and very little is left by 3 hours. No partially phosphorylated myo-inositol accumulates during germination. The breakdown of phytic acid proceeds at a constant rate during this time period. The rate is calculated to be 0.037 microgram phytic acid/milligram pollen/minute. Two phytases are detected in germinated lily pollen extract using high performance liquid chromatography with an anion exchange column (diethylaminoethyl-5PW). The results suggest that one of the phytases is already present in mature ungerminated lily pollen and the other one is newly synthesized during germination from a long-lived, pre-existing mRNA.  相似文献

The germination of intact, dehusked, and peeled seeds (caryopses) of the japonica rice cultivar Sasanishiki, harvested 30, 40, 47 and 60 days after anthesis, and of the indica rice cultivar Assam IV, harvested 14 and 28 days after anthesis, was examined. Dehusking strongly inhibited germination of Sasanishiki seeds, with the exception that seeds harvested 30 days after anthesis gave minimal germination percentages even when left intact. Peeling (removal of the pericarp and testa) restored or enhanced germination, and 60–100% of seeds germinated after 10 days. By contrast, the rank order of germination of Assam IV seeds was intact, dehusked, and peeled seeds, with peeled seeds yielding germination percentages of 100%. In Sasanishiki, inhibition of germination of peeled seeds was observed at reduced oxygen concentrations (1–4% oxygen). This inhibition might explain the inhibitory effects of dehusking on germination of seeds from the japonica cultivar. It is possible that the testa and pericarp, which cover the embryos of dehusked seeds, acted as a barrier to the diffusion of oxygen to the embryo.  相似文献

The freeze-preservation of pollen is dependent on the interaction of several factors such as freezing rate, thawing rate, freeze-drying temperature and duration, storage temperature and environment and rehydration rates. Changes in any of these variables affects the others directly or indirectly.Rapid freezing of pollen at rates of approximately 200 °C/min maintains the highest degree of viable pollen in combination with rapid thawing rates of 218 °C/min. Rapid cooling and slow rewarming resulted in a substantial loss of pollen viability. This might indicate that intracellular ice crystals formed during rapid cooling perhaps grow into larger ice masses during slow rewarming or storage at temperatures above ?50 °C.The germinability of pollen freeze-dried at temperatures below ?50 °C was also prolonged over that of the controls. Germination values for unfrozen pollen stored for 30 days at 0–5 °C averaged 50% for lily and 20% for corn. Freeze-dried pollen stored for 30 days at the same temperature yielded considerably higher viability percentages for both lily and corn pollen. Drying time is an important factor, perhaps indicating that residual moisture is critical. Freeze-dried pollen can be stored at higher temperatures than frozen and control pollen. Freeze-dried material stored for five months at 0–5 °C, upon slow rehydration yielded intact grains which has average germination percentages of 25 for lily and 15 for corn. The same pollen upon rapid rehydration showed rupturing of 20–40% of the cells and practically no germination.  相似文献

Cotton (Gossypium hirsutum L.) fruiting forms exhibit pronounced changes, with age, in their probability of abscission. Large floral buds rarely abscise, but after anthesis the young fruits (bolls) have a high probability of abscising. Abscission rate reaches a peak about 5 to 6 days after anthesis and then gradually decreases. An experiment was conducted to try to determine the reason for the rapid and pronounced increase in probability of abscission just after anthesis. Cotton was grown in the field and fruiting forms of various ages from 9 days before to 9 days after anthesis were all harvested the same day and subsequently analyzed for ABA and IAA. The concentration of ABA decreased slightly at anthesis and increased gradually thereafter. In contrast, the concentration of IAA was high before anthesis and then decreased at anthesis to about one-fifth the previous concentration. IAA remained low for at least 4 days after anthesis and then increased rapidly between 7 and 9 days after anthesis. The high concentration of IAA in floral buds before anthesis is probably a major factor in their resistance to abscission. Likewise, the low concentration of IAA at anthesis and for about 4 days thereafter may promote fruit abscission during the young boll stage.  相似文献

Seed germination was evaluated for Annona glabra L. and Pachira aquatica Aubl. in the wetland conditions of La Mancha, Veracruz, Mexico. These species have recalcitrant seeds and hydrochoral dispersal. Germination experiments were carried out under varying moisture (high, middle, and low) and light (below canopy and open sky) levels as well as after being stored in contrasting natural conditions. Seeds were stored both floating in water and buried in wetland soil for 15, 30, 60, and 90 days. P. aquatica seeds germinated faster in low and medium moisture, regardless of light intensity. After 45 days, for example, they exhibited 87–73% germination in medium-moisture/canopy and low-moisture/canopy treatments, respectively. In high moisture, seeds reached similar percentages after 60 days (80%). Storage by burial caused the death of seeds regardless the time they spent underground, while those stored in water germinated at a rate of over 90%. A. glabra seeds germinated better (98%) in low-moisture/sunny conditions. As to storage, they responded favorably to both burial and water techniques but germinated more readily in treatments that involved a long storage period. Evaluation of the germination behavior of A. glabra and P. aquatica seeds subject to varying in situ storage and germination conditions illustrates the response capacity of each species during early phases of development.  相似文献

In a succession of seed harvests of carrot, the highest percentage and most rapid germination was obtained from seed harvested 51 days after anthesis (DAA) when dried by conditioning at 25°C and 60% RH for one week and from seeeds harvested 65 or 79 DAA with or without conditioning treatment. Seed from these harvests had reached maximum weight when dried, had embryos of maximum length and were considered mature. The germinaton of seed from these treatments was unaffected by a mixture of the gibberellins A4 and A7 (GA4/7) applied in the incubation medium. Seed harvested 37 DAA also gave maximum percentage germination when it was both conditioned and incubated in GA4/7 solution. Seed harvested earlier than this germinated poorly. Carrot seeds taken from the parent plant were capable of germinating before the stage when maximum seed dry weight was reached but even after this stage, when the seed moisture content had fallen below 20%, improvement in seed germination characteristics continued. In the latter stages of seed growth losses by shedding were 12–20 kg/ha/day. For low density crops (10 plants/m2) the yield of viable seed was at a maximum in crops whose seed was harvested with a moisture content of between 20 and 40% but no consistent relationship could be established for high density crops (80 plants/m2). There were no effects of umbel order or plant density on mean germination time or spread of germination. At any early harvest, percentage germination was highest for primary-umbel-seeds and seeds from low density crops but the differences between the seed origins diminished with later harvests. Drying the seeds on the umbels improved the percentage germination, reduced the mean germination time and the spread of germination particularly at the early harvests compared with seeds removed from the umbels and germinated immediately without drying.  相似文献

Developing pollen of Papaver dubium L. becomes functional and desiccation tolerant at approximately 2 to 1 days prior to anthesis, coincident with degradation of starch and a doubling of the amount of sucrose, the primary soluble carbohydrate present. When anthers were taken from flower buds at 3 days before anthesis, pollen dehisced upon exposure to the ambient air. This dried pollen did not fluoresce with the vital stain fluorescein diacetate, had increased leakage of K+, and did not swell properly in a germination medium. In contrast, pollen became functional and desiccation resistant when these young anthers were first incubated in a water-saturated atmosphere for 30 hours. Phospholipid composition revealed no major differences over the last 3 days of development. When this immature pollen was liberated mechanically and allowed to mature in humid air, starch degraded and sucrose content nearly doubled, and the grains became largely functional and dehydration tolerant. Large unilamellar vesicles were prepared from isolated phospholipids to study dehydration-induced fusion and leakage. When dried in the presence of increasing concentrations of sucrose, vesicle integrity was progressively retained. These data indicate that pollen maturation during the last 3 days of development occurred independently from the parent plant. Sucrose may play an essential role in the acquired tolerance to severe dehydration.  相似文献

The changes of chemical composition and functional properties of derooted sesame (Sesamum indicum L.) seeds (DSS) before, during, and after germination were investigated. Sesame seeds germinated in dark chambers maintained near 100% relative humidity at 35 degrees C without presoaking reached >99% germination rate in 4 days with the final moisture content stayed ca. 2% (w/w), characterizing sesame seeds as orthodox seeds that are suitable for long term storage at low temperature and humidity under defined environment. With noticeable reduction in fat content (23%), germinated DSS were found rich in linolenic acid, P, and Na, increasing from 0.38% (w/w), 445 mg/100 g, and 7.6 mg/100 g before germination to 0.81% (w/w), 472 mg/100 g, and 8.4 mg/100 g after germination, respectively. DSS after germination contained considerable amount of Ca (462 mg/100 g), higher than that of soybean. Germinated DSS presents an excellent source of sesamol (475 mg/100 g), a potent natural antioxidant, and alpha-tocopherol (32 mg/100 g), the most active form of vitamin E.  相似文献

Summary Stylar self-incompatibility barriers in L. peruvianum can be avoided if pollen germination and growth through immature pistils is promoted under specific environmental conditions approximately 2–3 days before the initiation of anthesis. Since immature stigmas lack sufficient exudate for pollen germination, the sandwiching of a thin layer of pollen germination medium between the stigma and a mineral oil layer containing pollen allows precocious pollen germination and some compatible pollen tube growth through the style. This procedure is rapid, inexpensive, applicable in the field, and makes efficient use of pollen. Consistent though low seed yields have been obtained. A high proportion of aborted seed, seedling lethals, and aberrant seedling phenotypes in selfed progeny indicate the presence of strong post-zygotic barriers to such selfing. No evidence for a reduction in the strength of the SI response with increasing pistil age was observed.  相似文献

Abstract Moisture content and germination percentages on agar-solidified medium were determined for pollen of Juglans regia and J. nigra at the time of release from anther and at 24-h intervals until germination fell to zero. Moisture content of fresh J. regia pollen ranged from 4.6 to 12.1%, and from 3.9 to 4.0% for J. nigra pollen; in vitro germination percentages were uniformly high throughout these ranges. Germination declined as pollen moisture fell below 5% in J. regia with regression analyses predicting 50% germination at 3.9–4.4% moisture. After 4 d at ambient laboratory conditions, all pollen had lost the ability to germinate when incubated directly on medium. This pollen was incubated in chambers where humidity was maintained at saturation and 33% saturation, and retested. Pollen up to 22 d old and having moisture contents as low as 2.6% regained the ability to germinate in vitro when it was incubated in a water-saturated environment before being placed on the agar-solidified medium.  相似文献

Background and Aims

Pollinator-limited seed-set in some terrestrial orchids is compensated for by the presence of long-lived flowers. This study tests the hypothesis that pollen from these insect-pollinated orchids should be desiccation tolerant and relatively long lived using four closely related UK terrestrial species; Anacamptis morio, Dactylorhiza fuchsii, D. maculata and Orchis mascula.

Methods

Pollen from the four species was harvested from inflorescences and germinated in vitro, both immediately and also after drying to simulate interflower transit. Their tolerance to desiccation and short-term survival was additionally assessed after 3 d equilibration at a range of relative humidities (RHs), and related to constructed sorption isotherms (RH vs. moisture content, MC). Ageing of D. fuchsii pollen was further tested over 2 months against temperature and RH, and the resultant survival curves were subjected to probit analysis, and the distribution of pollen death in time (σ) was determined. The viability and siring ability, following artificial pollinations, were determined in D. fuchsii pollen following storage for 6 years at –20 °C.

Key Results

The pollen from all four species exhibited systematic increases in germinability and desiccation tolerance as anthesis approached, and pollen from open flowers generally retained high germinability. Short-term storage revealed sensitivity to low RH, whilst optimum survival occurred at comparable RHs in all species. Similarly, estimated pollen life spans (σ) at differing temperatures were longest under the dry conditions. Despite a reduction in germination and seeds per capsule, long-term storage of D. fuchsii pollen did not impact on subsequent seed germination in vitro.

Conclusions

Substantial pollen desiccation tolerance and life span of the four entomophilous orchids reflects a resilient survival strategy in response to unpredictable pollinator visitation, and presents an alternative approach to germplasm conservation.  相似文献

Pollen viability and germination are known to be sensitive to high temperature (HT). However, the mode by which high temperature impairs pollen functioning is not yet clear. In the present study, we investigated the effect of high temperature on changes occurring in carbohydrate of bell pepper (Capsicum annuum L. cv. Mazurka) pollen in order to find possible relations between these changes and pollen germination under heat stress. When pepper plants were maintained under a moderate HT regime (32/26 degrees C, day/night) for 8 days before flowers have reached anthesis, pollen count at anthesis was similar to that found in plants grown under normal temperatures (NT 28/22 degrees C). However, the in vitro germination, carried out at 25 degrees C, of pollen from HT plants was greatly reduced. This effect matched the marked reduction in the number of seeds per fruit in the HT plants. Maintaining the plants at high air CO2 concentration (800 &mgr;mol mol-1 air) in both temperature treatments did not affect the in vitro germination of pollen from NT plants, but restored germination to near the normal level in pollen from HT plants. Under NT conditions, starch, which was negligible in pollen at meiosis (8 days before anthesis, A-8) started to accumulate at A-4 and continued to accumulate until A-2. From that stage until anthesis, starch was rapidly degraded. On the other hand, sucrose concentration rose from stage A-4 until anthesis. Acid invertase (EC 3.2.1.26) activity rose parallel with the increase of sucrose. In pollen from HT plants, sucrose and starch concentrations were significantly higher at A-1 pollen than in that of NT plants. Under high CO2 conditions, the sucrose concentration in the pollen of HT plants was reduced to levels similar to those in NT pollen. In accordance with the higher sucrose concentration in HT pollen, the acid invertase activity in these pollen grains was lower than in NT pollen. The results suggest that the higher concentrations of sucrose and starch in the pollen grains of HT plants may result from reduction in their metabolism under heat stress. Elevated CO2 concentration, presumably by increasing assimilate availability to the pollen grain, may alleviate the inhibition of sucrose and starch metabolism, thereby increasing their utilization for pollen germination under the HT stress. The L-ascorbic acid (AA) content of pollen from three cultivars of Lilium longiflorum Thunb. was 260–280 μg/g fresh wt. of pollen. During germination ascorbic acid content gradually decreased reaching 70% of the original value at 6 h. Pollen germinated in media containing 0.29 M D-glucose (an osmoregulator and carbon source) failed to synthesize ascorbic acid but pollen germinated in 0.29 M pentaerythritol (a non-metabolizable osmoregulator) supplemented with L-galactono-1,4-lactone (L-GalAL) did form ascorbic acid, dependent upon the concentration of the lactone. Lycorine inhibited germination but had negligible effect on the conversion of L-GalAL to AA.  相似文献

Dietary diversification, including consumption of plant tissues such as pollen, can enhance the fecundity of generalist predators, resulting in improved control of pest prey. Supplemental pollen feeding has been observed in many natural enemies, including sheet-web spiders (Araneae: Linyphiidae), which represent a major component of food webs in agroecosystems. Their horizontal, ground-based webs have the potential to intercept pollen grains during anthesis of crop plants, providing the opportunity for consumption of pollen to occur. In laboratory feeding trials, Frontinella communis and Tennesseellum formicum (Araneae: Linyphiidae) readily fed on pollen grains dusted on their webs, with 82 and 92% of spiders consuming pollen within the 210 min trial. These results revealed a strong potential for dietary supplementation with pollen in ground-based sheet-web spiders, indicating that pollen feeding may be an important component of the feeding biology of linyphiids. To measure pollen and prey interception in simulated linyphiid webs, a 20 m × 20 m grid of miniature sticky traps was established within and downwind of a corn agroecosystem. Traps were exposed for 24 h, all intercepted material was transferred to the laboratory for subsequent identification, and replaced with additional traps for 28 consecutive days in July and August 2008, to encompass periods before, during and after anthesis. Over 150,000 corn pollen grains and 5,000 prey items (dominated by Collembola and Hemiptera) were intercepted at simulated web-sites. Dates of peak anthesis resulted in pollen counts as high as 4,000 grains per web-site in the interior of the cornfield. Spatial Analysis by Distance Indices (SADIE) indicated significant temporal and spatial variability in pollen interception within and outside the corn field, but interestingly there was no significant spatial association between pollen and prey. Furthermore, transgenic Bacillus thuringiensis corn expresses insecticidal proteins in pollen, posing an exposure risk to non-target arthropods. Consumption of corn pollen may be a route to transgenic protein exposure in this important taxon of generalist predators.  相似文献

Lasia spinosa seeds were not dormant at maturity in early spring. The most favorable temperatures for germination were between 25 and 30 °C, and final percentage and rate of germination decreased with an increase or decrease in temperature. When L. spinosa seeds were transferred to 25 °C, after 60 days at 10 °C (where none of the seeds germinated), final germination increased from 0% to 78%. Seeds germinated to high percentage both in light and in dark, although dark germination took more than twice as long as in the light. During desiccation of seeds at 15 °C and 45% relatively humidity, moisture loss decreased exponentially from 2.02 to 0.13 g H2O g−1 dry wt within 16 days, and only a few seeds (12%) survived 0.13 g H2O g−1 dry wt moisture content. Seeds stored at 0.58 g H2O g−1 dry wt moisture content at four constant temperatures (4, 10, 15, and −18 °C) for up to 6 months exhibited a well-defined trend of decreasing viability with decreasing temperature. Thus, we concluded that freshly harvested L. spinosa seeds are non-dormant and recalcitrant. Also, the seeds with 0.58 g H2O g−1 dry wt moisture content could be effectively stored for a few months between 10 and 15 °C although the most appropriate temperature for wet storage appears to be 10 °C, as it is close to the minimum temperature for germination and so there will be less pre-sprouting compared to 15 °C.  相似文献

Muskmelon (Cucumis melo L.) seeds are germinable 15 to 20 days before fruit maturity and are held at relatively high water content within the fruit, yet little precocious germination is observed. To investigate two possible factors preventing precocious germination, the inhibitory effects of abscisic acid and osmoticum on muskmelon seed germination were determined throughout development. Seeds were harvested at 5-day intervals from 30 to 65 days after anthesis (DAA) and incubated either fresh or after drying on factorial combinations of 0, 1, 3.3, 10, or 33 micromolar abscisic acid (ABA) and 0, −0.2, −0.4, −0.6, or −0.8 megapascals polyethylene glycol 8000 solutions at 30°C. Radicle emergence was scored at 12-hour intervals for 10 days. In the absence of ABA, the water potential (Ψ) required to inhibit fresh seed germination by 50% decreased from −0.3 to −0.8 megapascals between 30 and 60 DAA. The Ψ inside developing fruits was from 0.4 to 1.4 megapascals lower than that required for germination at all stages of development, indicating that the fruit Ψ is sufficiently low to prevent precocious germination. At 0 megapascal, the ABA concentration required to inhibit germination by 50% was approximately 10 micromolar up to 50 DAA and increased to >33 micromolar thereafter. Dehydration improved subsequent germination of immature seeds in ABA or low Ψ. There was a linear additive interaction between ABA and Ψ such that 10 micromolar ABA or −0.5 megapascal osmotic potential resulted in equivalent, and additive, reductions in germination rate and percentage of mature seeds. Abscisic acid had no effect on embryo solute potential or water content, but increased the apparent minimum turgor required for germination. ABA and osmoticum appear to influence germination rates and percentages by reducing the embryo growth potential (turgor in excess of a minimum threshold turgor) but via different mechanisms. Abscisic acid apparently increases the minimum turgor threshold, while low Ψ reduces turgor by reducing seed water content.  相似文献

以结球甘蓝品种冬升种子为材料,研究了不同成熟度和烘干温度下种子秕粒率、千粒重、发芽率、生理活性情况以及不同烘干温度下种子的含水量.结果表明,结球甘蓝冬升开花后45~55 d采收的种子,发芽率均达到了95%以上;随着种子成熟度提高,其种子质量、发芽活力及其超氧化物歧化酶(SOD)、过氧化物酶(POD)、脱氢酶活性显著上升,而相对电导率显著下降.与对照(自然风干)相比,30~50℃的烘干温度对种子千粒重和秕粒率无显著性影响,也仅在50℃下可使种子的发芽活力显著降低;随着烘干温度的升高,种子的SOD、POD和脱氢酶活性逐渐显著下降,相对电导率则逐渐显著上升;30~50℃烘干6 h种子的含水量由13.3%降至5.4%左右.研究发现,结球甘蓝冬升开花授粉后45 d种子已达到了采收程度,30~50℃烘干6 h种子含水量已达到储藏要求,并且愈接近自然干燥温度(30~40℃)的处理,种子发芽能力愈好;甘蓝种子活力与其SOD、POD和脱氢酶活性呈正相关,而与其相对电导率呈负相关.  相似文献

九里香种子自花后 42~ 77d,含水量和电导率逐渐降低 ,种子干重、发芽率、发芽指数和活力指数逐渐增加。硅胶脱水 1~ 6d后 ,种子含水量下降 1 0 %~ 3 5 % ,发芽率、发芽指数和活力指数均有不同程度的降低 ,不同发育时期九里香种子的脱水耐性有别 ,花后 42~ 70d不断增强 ,77d有所减弱。花后 70d的种子含水量降至 1 0 % ,种子发芽率无明显降低 ;含水量为 9%的种子在 4°C和 2 0°C的低温条件贮存 3 0d和 42d ,多数种子仍能萌发 ,这表明九里香种子是一种正常型种子。光照能促进种子的萌发 ;在 2 0~ 3 0°C、室温和 2 0 /3 0°C变温条件下种子萌发较好 ;光照和温度对种子萌发有单独影响 ,但又相互作用 ,同时光照对萌发的影响还与种子含水量有关。  相似文献

Most angiosperms release bicellular pollen. However, in about one-third of extant angiosperms, the second pollen mitosis occurs before anthesis such that pollen is tricellular upon release. The shift from bicellular to tricellular development has occurred several times independently, but its causes are largely unknown. In this work, we observed the coexistence of both kinds of pollen at anther dehiscence in Annona cherimola, a species that belongs to the basal angiosperm family Annonaceae. Examination of pollen cell number during anther development showed that this coexistence was due to a late mitosis starting shortly before pollen shedding. Both types of pollen germinated equally well over the course of development. Because variable proportions of bicellular and tricellular pollen were observed at different sampling times, we tested the role of temperature by performing field and growth chamber experiments, which showed that higher temperatures near anthesis advanced the time of pollen mitosis II. The results show that selection could favor the production of tricellular pollen under certain environmental circumstances that prime rapid pollen germination and provide evidence of a system in which developmental variation persists, but that can be modified by external factors such as temperature.  相似文献
