Recombination occurs in many RNA viruses and can be of major evolutionary significance. However, rates of recombination vary dramatically among RNA viruses, which can range from clonal to highly recombinogenic. Here, we review the factors that might explain this variation in recombination frequency and show that there is little evidence that recombination is favoured by natural selection to create advantageous genotypes or purge deleterious mutations, as predicted if recombination functions as a form of sexual reproduction. Rather, recombination rates seemingly reflect larger-scale patterns of viral genome organization, such that recombination may be a mechanistic by-product of the evolutionary pressures acting on other aspects of virus biology.  相似文献

Recent research, combined with reanalysis of previous results, has revealed that chromium can no longer be considered an essential trace element. Clinical studies are ambiguous at best as to whether Cr has a pharmacological effect in humans. Observed effects of Cr on rodent models of insulin resistance and diabetes are best interpreted in terms of a pharmacological role for Cr. Studies on the effects of Cr on rat models of diabetes are reviewed herein and suggest Cr increases insulin sensitivity in peripheral tissues of the rodent models. The lack of effects in human studies may stem from humans receiving a comparably smaller dose than the rodent models. However, given the different responses to Cr in the rodent models, humans could potentially have different responses to Cr.  相似文献

Cdk2 has been viewed as a key cell cycle regulator that is essential for S phase progression. The recent discovery that Cdk2 is not required for cell proliferation in mice now shows that other factors must be able to replace Cdk2 in stimulating DNA replication. Experiments performed in Xenopus egg extracts identify the mitotic protein kinases Cdk1/Cyclin B and Cdk1/Cyclin A as likely candidates. These observations raise the intriguing possibility that Cdk1 normally participates in genome duplication in wild type cells.  相似文献

GcvB is a non-coding RNA that regulates oppA mRNA in different bacterial species by binding a GcvB GU-rich region named R1 to oppA mRNA. A secondary putative interaction site (PS1) was identified in this study that is able to form a second nearly perfect 10 base-pair duplex between these two RNAs in Escherichia coli. In this work, we have studied whether the formation of a second interaction site could help stabilize the previously reported GcvB/oppA complex. Several mutations and the full deletion of PS1 were engineered. None of these modifications affected the ability of GcvB to control OppA expression. Therefore the second, putative, interaction site appears to be unnecessary for the regulatory function of GcvB with regard to its oppA target mRNA.  相似文献

Biological membranes are complex and well-organized multimolecular assemblies composed of a wide variety of protein and lipid molecular species. If such a diversity in protein and lipid polar headgroup structures may easily be related to a large panel of functions, the wide dispersion in acyl chain length and structure which the lipids display is more difficult to understand. It is not required for maintaining bilayer assembly and fluidity. Direct information on the lateral distribution of these various molecular species, on their potential specificity for interaction between themselves and with proteins and on the functional implications of these interactions is also still lacking. Because hydrophobic interactions play a major role in stabilizing membrane structures, we suggest considering the problem from the point of view of the matching of the hydrophobic surface of proteins by the acyl chains of the lipids. After a brief introduction to the hydrophobic matching principle, we will present experimental results which demonstrate the predictive power of the current theories and then, we will introduce the new and important concept of protein/lipid sorting in membranes. Finally, we will show how the hydrophobic matching condition may play a key role in the membrane organization and function.  相似文献

The conventional view of AD (Alzheimer's disease) is that much of the pathology is driven by an increased load of β-amyloid in the brain of AD patients (the 'Amyloid Hypothesis'). Yet, many therapeutic strategies based on lowering β-amyloid have so far failed in clinical trials. This failure of β-amyloid-lowering agents has caused many to question the Amyloid Hypothesis itself. However, AD is likely to be a complex disease driven by multiple factors. In addition, it is increasingly clear that β-amyloid processing involves many enzymes and signalling pathways that play a role in a diverse array of cellular processes. Thus the clinical failure of β-amyloid-lowering agents does not mean that the hypothesis itself is incorrect; it may simply mean that manipulating β-amyloid directly is an unrealistic strategy for therapeutic intervention, given the complex role of β-amyloid in neuronal physiology. Another possible problem may be that toxic β-amyloid levels have already caused irreversible damage to downstream cellular pathways by the time dementia sets in. We argue in the present review that a more direct (and possibly simpler) approach to AD therapeutics is to rescue synaptic dysfunction directly, by focusing on the mechanisms by which elevated levels of β-amyloid disrupt synaptic physiology.  相似文献

Mathematical models of predator-prey population dynamics are widely used for predicting the effect of predators as biocontrol agents, but the assumptions of the models are more relevant to parasite-host systems. Predator-prey systems, at least in insects, substantially differ from what is assumed by these models. The main differences are: (i) Juveniles and adults have to be considered as two different entities, as the former stay within a patch and do not reproduce, while the latter move between patches of prey and reproduce there. (ii) Because of their high mobility, food availability is likely to be less restrictive for adults than juveniles, which are confined to one patch. Therefore, a functional response to prey abundance may not be important for adults. (iii) Egg and larval cannibalism are common in insect predators. Therefore, the quality of patches of prey for their larvae determines the reproductive strategy of adult predators more than the availability of food for the adults. Here we develop a new model, based on the above considerations, which is suitable for modelling these interactions. We show that selection should favour mechanisms that enable predators to avoid reproducing in patches with insufficient prey and those already occupied by predators.  相似文献

A quasispecies is a well-defined distribution of mutants that is generated by a mutation-selection process. Selection does not act on a single mutant but on the quasispecies as a whole. Experimental systems have been designed to study quasispecies evolution under laboratory conditions. More recently, virus populations have been called quasispecies to indicate their extensive genetic heterogeneity. The most prominent examples are probably the human immunodeficiency viruses HIV-1 and HIV-2. The quasispecies nature of HIV has formed the basis of a model that provides a mechanism for the pathogenesis of acquired immunodeficiency syndrome (AIDS) in humans. This article focuses on the nature of the quasispecies concept and its implications for evolutionary biology and virology.  相似文献

The possibility to derive the analogs of native proteins by the chemical synthesis is considered to be a serious argument for the concept of posttranslational protein folding. The present paper analyzes for the first time chemically synthesized proteins to reveal whether they are relevant to the problem of protein folding. The results enable the following conclusions to be drawn. The acquisition of the peculiar conformations by the chemically synthesized proteins to exhibit the specific functions is conditioned by the highly marked features of the secondary and tertiary structures of the corresponding native proteins. These features will make themselves evident only if favorable conditions are carefully chosen during the experiments for each individual protein. Thus, in our opinion, the possibility to derive a synthetic protein is hardly evidence for the posttranslational folding of proteins.  相似文献
