目的:探讨数据管理系统DM2检验结果自动确认范围设定.方法:统计分析2008年全自动流水线测定项目检验结果.以1%和99%分位作为自动审核上下限,查看2009年3月数据自动审核通过率并分析未通过审核原因.结果:2008年数据统计结果与项目的参考范围有差异,数据自动审核通过率超过75%,未通过审核数据中超出自动审核范围的结果占23%以上.结论:通过对实验室数据的回顾性分析,设定合理的自动审核范围,充分发挥全自动流水线和数据管理系统的效能,提高检测效率,为临床提供快捷、准确的服务.  相似文献

目的:利用生物信息学方法,将高通量基因表达数据与单核酸多肽(SNP)基因型数据进行整合分析,研究并注释前列腺癌风险基因。方法:本文基于EST、SAGE、基因芯片三类功能基因组数据整合的方法研究前列腺癌风险基因。首先,通过三类数据寻找前列腺癌中异常表达和差异表达基因。利用全基因组关联分析得到前列腺癌相关基因的SNPs。定位所得到的前例腺癌差异表达基因与SNPs到染色体,获取与SNPs在同一区段的差异表达基因,并通过各种数据库注释所得基因。结果:通过数据整合分析,最终得到前列腺癌风险基因84个,其中20多个基因已被证实与前列腺癌极其相关。结论:整合前列腺癌高通量表达数据与SNP基因型数据,能够快速有效的获得前列腺癌显著相关基因。此方法可以推广于其它癌症的研究。  相似文献

高通量实验方法的发展导致大量基因组、转录组、代谢组等组学数据的出现,组学数据的整合为全面了解生物学系统提供了条件.但是,由于当前实验技术手段的限制,高通量组学数据大多存在系统偏差,数据类型和可靠程度也各不相同,这给组学数据的整合带来了困难.本文以转录组、蛋白质组和代谢组为重点,综述了近年来组学数据整合方面的研究进展,包括新的数据整合方法和分析平台.虽然现存的数据统计和网络分析的方法有助于发现不同组学数据之间的关联,但是生物学意义上的深层次的数据整合还有待于生物、数学、计算机等各种领域的全面发展.  相似文献

长期以来, 学者普遍认为大多数鸟类的嗅觉能力较弱或丧失。早期实验未能得到统一而清晰的结果, 进一步扩大了这一认知误区。随着研究手段和技术的发展, 解剖学、电生理学、分子生物学和行为生态学等学科提供了鸟类嗅觉存在的证据。目前, 相关研究在鸟类14目33科中发现了嗅觉通讯的证据。与视觉和听觉一样, 这一感觉通路可能在鸟类的觅食、导航、防御、隐蔽、警戒和交流等社会行为中发挥着重要作用。本文着重于嗅觉通讯在鸟类社会行为中的功能, 通过回顾近十年的相关研究, 综述鸟类嗅觉在物种和个体识别、繁殖行为、亲缘识别、配偶选择与竞争等方面发挥的作用。我们也指出: 研究手段的创新将揭示更多鸟类物种在社会行为中对于嗅觉通讯的运用。此外, 当前研究多聚焦于少数物种和单一层面(如生理、生态和环境等), 使用分子生物学、解剖与生理学、行为学和神经生物学手段的整合研究较为缺乏; 而后者更有可能全面地揭示鸟类嗅觉通讯的复杂机理及其在社会行为中发挥的多种功能。  相似文献

众所周知,随着基因组测序工作的蓬勃发展和后基因组时代的到来,生物信息学数据呈指数级增长.生物界在享受着资源共享所带来便利的同时,也随着数据总量和复杂性的不断增加而变得异构化和分布化.目前,各种生物计算软件和数据库资源通常标准不一而且很难兼容.因此,如何在这些异构资源之间实现数据集成与软件共享是有效利用生物信息资源的关键.为解决以上问题,本文提出了一种新型的数据整合架构,该架构通过将web服务与并行计算相结合的方法,轻松地实现了对异地资源数据的访问、提取、转化以及整合.实验证明,本系统在处理异构、海量数据方面有着巨大的计算潜力.  相似文献

In recent years, with the availability of high resolution models of the human body, numerical computations of induced electric fields and currents have been made in more than one laboratory for various exposure conditions. Despite the verification of computational methods, questions are often asked about the reliability of these data. In this paper, computational results from two laboratories that presented data in compatible formats are compared, supplemented with additional data from the third laboratory. Two exposures to uniform fields at 60 Hz are evaluated. The human body models used in the computations are different and so are the computation al methods and codes. There are some differences in the conductivity values used for some of the tissues, as well. The results of the comparison confirm that these data are reliable, as the overall agreement is reasonably good and the differences can be rationally explained. This comparison also underscores the importance of accurate data on the dielectric properties of tissues.  相似文献

Power-frequency electric fields are strongly perturbed in the vicinity of human beings and experimental animals. As a consequence, the extrapolation of biological data from laboratory animals to human-exposure situations cannot use the unperturbed exposure field strength as a common exposure parameter. Rather, comparisons between species must be based on the actual electric fields at the outer surfaces of and inside the bodies of the subjects. Experimental data have been published on surface and internal fields for a few exposure situations, but it is not feasible to characterize experimentally more than a small fraction of the diverse types of exposures which occur in the laboratory and in the field. A predictive numerical model is needed, one whose predictions have been verified in situations where experimental data are available, and one whose results can be used with confidence in new exposure situations. This paper describes a numerical technique which can be used to develop such a model, and it carries out this development for a test case, that of a homogeneous right-circular cylinder resting upright on-end on a ground plane and exposed to a vertical, uniform, 60-Hz electric field. The accuracy of the model is tested by comparing short-circuit currents and induced current densities predicted by it to measured values: Agreement is good.  相似文献

For more than a decade, Midwest Research Institute (MRI) has investigated the effects of exposure to 60 Hz electric and magnetic fields (EMF) on human physiology, performance, and biochemistry. This accumulated experience, new research directions, and limited resources made it important to design more comprehensive and operationally cost-effective exposure facilities. Here we describe the new, integrated laboratory exposure test facilities recently constructed at MRI and present data on relevant characteristics of the exposure systems. Concentric coil systems were developed to generate uniform magnetic fields within the three new exposure rooms, with rapid cancellation of the field to ambient levels in the rest of the laboratory. Control systems are fully automated, computer-based, and independent. These provide the operational flexibility needed to present fields of different magnitudes, frequencies, and polarization. The local geomagnetic field can be modulated and/or canceled, and both AC and DC fields can be presented in various combinations. Capabilities for conducting double-blind experiments with true active-sham exposure conditions were implemented using bifilar windings and applying current flow in the opposite direction for each wire in a pair. The new facilities provide a comprehensive capability for laboratory-based human research on the potential effects of exposure to AC and DC magnetic fields.  相似文献

Efficient analysis of protein 2D NMR spectra using the software packageEASY   总被引:10,自引:0,他引:10  
Summary The programEASY supports the spectral analysis of biomacromolecular two-dimensional (2D) nuclear magnetic resonance (NMR) data. It provides a user-friendly, window-based environment in which to view spectra for interactive interpretation. In addition, it includes a number of automated routines for peakpicking, spin-system identification, sequential resonance assignment in polypeptide chains, and cross peak integration. In this uniform environment, all resulting parameter lists can be recorded on disk, so that the paper plots and handwritten notes which normally accompany manual assignment of spectra can be largely eliminated. For example, in a protein structure determination by 2D1H NMR,EASY accepts the frequency domain datasets as input, and after combined use of the automated and interactive routines it can yield a listing of conformational constraints in the format required as input for the calculation of the 3D structure. The program was extensively tested with current protein structure determinations in our laboratory. In this paper, its main features are illustrated with data on the protein basic pancreatic trypsin inhibitor.  相似文献

We apply a method to evaluate the strength of the evidence for deviations from uniform land change in a coastal area, in the context of Intensity Analysis. The errors in the CORINE maps at 1990 and 2006 can influence the apparent change, but the errors are unknown because error assessment of the 1990 map has never been released, while the error of the 2006 map has been checked for only some countries. The 1990 and the 2006 maps of a coastal watershed in Portugal served as the data to compute the intensities of changes among eight categories. We evaluate the sizes and types of errors that could explain deviations from uniform intensities. Errors in 2.0% of the 2006 map can explain all apparent deviations from uniform gains. Errors in 1.5% of the 1990 map can explain all apparent deviations from uniform losses. Errors in less than 0.7% of the 1990 map can explain all apparent deviations from uniform transitions to each gaining category. We analyse the strength of the evidence for deviations from uniform intensities in light of historical processes of change. Historical processes can explain some transitions that the data show, while the hypothesised errors in the data are the explanation for other transitions that are not consistent with known processes. Inconsistent transitions are an indication of the misclassification errors that could propagate to other land cover change applications, as in the assessment of hydrological processes.  相似文献

A device for cutting brain slices is described as an alternative to cutting angle guides and the “brain macrotome”. With this new device, slices of uniform thickness optimal for assessing morphological detail and photography can be produced. A similar but smaller device for cutting pieces of tissue for paraffin embedding is also presented. These devices should be useful in either the histopathology laboratory or mortuary.  相似文献

Any change in shape of a configuration of landmark points in two or three dimensions includes a uniform component, a component that is a wholly linear (affine) transformation. The formulas for estimating this component have been standardized for two-dimensional data but not for three-dimensional data. We suggest estimating the component by way of the complementarity between the uniform component and the space of partial warps. The component can be estimated by regression in either one space or the other: regression on the partial warps, followed by their removal, or regression on a basis for the uniform component itself. Either of the new methods can be used for both two- and three-dimensional landmark data and thus generalize Bookstein's (1996, pages 153-168 in Advances in morphometrics [L. F. Marcus et al., eds.], Plenum, New York) linearized Procrustes formula for estimating the uniform component in two dimensions.  相似文献

采用5块实地调查样地和30块模拟样地,通过基于Ripley K-函数的L-函数、双相关函数和角尺度方法在林木空间分布格局研究中的比较分析,得出如下结论:双相关函数和角尺度在判断的准确性方面优于L-函数;角尺度在有效性和可行性方面比L-函数和双相关函数更强,并且它能利用角尺度分布图和均值同时作定性和定量分析,从而使其优势更加明显。  相似文献

Internal quality control (IQC) is a critical component of laboratory quality management, and IQC products can determine the reliability of testing results. In China, given the fact that most blood transfusion compatibility laboratories do not employ IQC products or do so minimally, there is a lack of uniform and standardized IQC methods. To explore the reliability of IQC products and methods, we studied 697 results from IQC samples in our laboratory from 2012 to 2014. The results showed that the sensitivity and specificity of the IQCs in anti-B testing were 100% and 99.7%, respectively. The sensitivity and specificity of the IQCs in forward blood typing, anti-A testing, irregular antibody screening, and cross-matching were all 100%. The reliability analysis indicated that 97% of anti-B testing results were at a 99% confidence level, and 99.9% of forward blood typing, anti-A testing, irregular antibody screening, and cross-matching results were at a 99% confidence level. Therefore, our IQC products and methods are highly sensitive, specific, and reliable. Our study paves the way for the establishment of a uniform and standardized IQC method for pre-transfusion compatibility testing in China and other parts of the world.  相似文献

The Functional Genomics Experiment data model (FuGE) has been developed to facilitate convergence of data standards for high-throughput, comprehensive analyses in biology. FuGE models the components of an experimental activity that are common across different technologies, including protocols, samples and data. FuGE provides a foundation for describing entire laboratory workflows and for the development of new data formats. The Microarray Gene Expression Data society and the Proteomics Standards Initiative have committed to using FuGE as the basis for defining their respective standards, and other standards groups, including the Metabolomics Standards Initiative, are evaluating FuGE in their development efforts. Adoption of FuGE by multiple standards bodies will enable uniform reporting of common parts of functional genomics workflows, simplify data-integration efforts and ease the burden on researchers seeking to fulfill multiple minimum reporting requirements. Such advances are important for transparent data management and mining in functional genomics and systems biology.  相似文献
