In 1986, house mice in a small defined area of inner Birmingham were reported as not taking a variety of rodenticides from bait containers, a phenomenon labelled 'behavioural resistance'. This study investigated behavioural resistance by comparing the food preferences of West Midlands behaviourally resistant (WMBR) mice with those of normal (BC) mice. Nine bait boxes each containing one of nine different foods (cheese, chicken, tuna fish, peanut butter, canary seed, Cat stars, wheat, PCD (MOD) pellets and Non-tox) were introduced to 12 WMBR and seven BC sites (Experiment 1). The experiment was repeated in the laboratory with six pens of WMBR and six of BC mice (Experiment 2), and to investigate whether the preferences had a genetic basis the offspring were similarly assayed (Experiment 3). In each experiment the consumption of each food was measured over 7 days and the droppings around the bait boxes were counted to assess mouse activity. Food neophobia was noted in some populations of BC mice. Tested in the wild and in the laboratory WMBR mice showed an aversion to foods containing cereals, as did their offspring. These results, with other lines of evidence, strongly suggest that cereal aversion in WMBR mice has a physiological/genetic basis. Since cereal aversion allows WMBR mice to survive cereal-based rodenticidal baits, we conclude that WMBR mice have genetically based behaviours that allow them to survive poisoning regimes that kill other strains.  相似文献

Nest boxes are frequently used in conservation programs for tree-cavity dependent wildlife. There is growing concern that the poor insulation properties of nest boxes may produce an ecological trap, because species may require microclimates less extreme or less variable than those experienced inside nest boxes. I investigated the fitness consequences of nest box use in a non-flying mammal. Fifty-two of 104 squirrel gliders (Petaurus norfolcensis) trapped over a 3-year period used nest boxes. Population modelling of the capture data revealed that the probability of apparent survival increased with increasing nest box use. There was no difference in breeding frequency between females that used or did not use nest boxes. There was no evidence that offspring development was hindered within nest boxes. These findings may arise because: (1) gliders could access tree hollows during extreme temperatures, (2) ambient temperatures were mild during the study, (3) gliders construct leaf nests which insulate against low temperatures in winter, and (4) gliders breed between autumn and spring when temperatures are relatively benign. The estimate of annual survival of animals using nest boxes (0.60), was equivalent to estimates at locations where squirrel gliders were either reliant on nest boxes (0.54) or on tree cavities (0.55) for shelter. Studies such as this need to be conducted on a range of species across a range of locations to better understand the influence of nest box use on non-flying mammals.  相似文献

The origin of replication of the IncL/M plasmid pMU604 was analyzed to identify sequences important for binding of initiator proteins and origin activity. A thrice repeated sequence motif 5'-NANCYGCAA-3' was identified as the binding site (RepA box) of the initiator protein, RepA. All three copies of the RepA box were required for in vivo activity and binding of RepA to these boxes appeared to be cooperative. A DnaA R box (box 1), located immediately upstream of the RepA boxes, was not required for recruitment of DnaA during initiation of replication by RepA of pMU604 unless a DnaA R box located at the distal end of the origin (box 3) had been inactivated. However, DnaA R box 1 was important for recruitment of DnaA to the origin of replication of pMU604 when the initiator RepA was that from a distantly related plasmid, pMU720. A mutation which scrambled DnaA R boxes 1 and 3 and one which scrambled DnaA R boxes 1, 3 and 4 had much more deleterious effects on initiation by RepA of pMU720 than on initiation by RepA of pMU604. Neither Rep protein could initiate replication from the origin of pMU604 in the absence of DnaA, suggesting that the difference between them might lie in the mechanism of recruitment of DnaA to this origin. DnaA protein enhanced the binding and origin unwinding activities of RepA of pMU604, but appeared unable to bind to a linear DNA fragment bearing the origin of replication of pMU604 in the absence of other proteins.  相似文献

Laboratory and field studies were conducted between July and October 1999 to identify bait preference, biomarker efficacy, and bait acceptance rates for delivering an oral plague vaccine to black-tailed prairie dogs (Cynomys ludovicianus). Twenty juvenile captive prairie dogs were offered alfalfa baits containing either alfalfa, alfalfa and 5% molasses, or alfalfa, 5% molasses and 4% salt. Based on the results of these trials we selected a bait containing alfalfa, 7% molasses, and 1% salt for field trials to determine bait acceptance rates by free-ranging animals. The biomarkers DuPont Blue dye, iophenoxic acid, and tetracycline hydrochloride were orally administered to captive prairie dogs to determine their efficacy. Only tetracycline proved effective as a biomarker. Two field trials were conducted at separate prairie dog colonies located at the Buffalo Gap National Grassland (Pennington County, South Dakota, USA). In Trial 1, three baits containing tetracycline were distributed around each active burrow entrance and an additional bait was placed inside the burrow (1,276 baits total). In Trial 2, baits were distributed at the same density per burrow as Trial 1, but along transects spaced 10 m apart (1,744 baits total). Trapping began 3 days after bait distribution, and 30 prairie dogs then were captured at each site to determine the percentage of animals marked. In Trial 1, 67% of the prairie dogs captured had tetracycline deposits indicative of bait consumption. In Trial 2, 83% of the prairie dogs had ingested a bait. Approximately 15% of the animals in both trials ate more than one bait. Fleas (Opisocrostis hirsutus) were found on 64 of 70 (91%) of the prairie dogs captured during this study.  相似文献

Gel and paste bait formulations containing boric acid or hydramethylnon were evaluated in field and laboratory experiments with the German cockroach, Blattella germanica (L.). Bait formulations contained 14-62% water by weight and lost the most water after 1 d of desiccation. Bait age and water content did not alter the toxicity (LT50) of boric acid-based baits. Toxicity of hydramethylnon (Maxforce) bait, however, increased with bait age. In continuous exposure tests without harborage, boric acid formulations had LT50s of 8.5 and 10.1 d for Blue Diamond and IT WORKS, respectively. Maxforce bait had an LT50 of 4.2 d. LT50s determined with cardboard harborage or in Ebeling choice boxes were 1.5 to 2.2 times lower than those from continuous exposure tests without harborage. Relative repellency, measured as the mean percent of live cockroaches in the light side of the choice box, was greatest for IT WORKS (21.4%) and Maxforce (25.4%) formulations, followed by Blue Diamond (10.9%). A performance index that combined choice box repellency and mortality data indicated that Maxforce had the greatest potential for field effectiveness. Blue Diamond and Maxforce significantly reduced German cockroach populations in the field. Maxforce performed best in field trials when applied at 10 or 20 placements per apartment. Apartments treated with IT WORKS had significantly greater cockroach trap catches. Factors that may affect the predictive power of the performance index are discussed.  相似文献

Bat boxes frequently form part of hollow‐bearing tree offsets; however, their effectiveness is poorly documented. We investigated the effectiveness of a bat box program designed to partially offset tree hollow loss from clearing for a coal mine. During the first year of monitoring, we detected bats in 5% of 1,308 box checks. Only 3 of 13 local tree cavity‐roosting bat species/species groups used boxes and occupancy was not strongly associated with modeled box and site attributes. In the second year, we tested two hypotheses that may explain the relatively low box use: (1) solar exposure of boxes was inadequate for heterothermic bats and (2) available box designs were of low suitability. Relocating boxes to increase solar exposure did not increase use, or enhance the temperature profiles of relocated boxes. Introduction of a new box design led to 11 times higher use compared with existing designs for Nyctophilus spp. (long‐eared bat). Overall, our data suggest that the bat box program was ineffective due to few bat species using boxes, infrequent box use by three species, and rarity of maternity roosting. The knowledge gap of species‐specific box designs and roosting ecology limits the effectiveness of boxes to offset cleared hollow‐bearing trees. Lack of knowledge and the widespread use of bat boxes to offset lost tree hollows highlights the need to (1) rigorously protect hollow‐bearing trees and (2) advance our understanding of species‐specific roost ecology, box design preferences and mechanical hollow creation into trees, before artificial hollows can be considered a meaningful offset measure.  相似文献

The expression of the Escherichia coli torCAD operon, which encodes the anaerobically expressed trimethylamine N-oxide (TMAO) reductase respiratory system, requires the presence of TMAO in the medium. The response regulator, TorR, has recently been identified as the regulatory protein that controls the expression of the torCAD operon in response to TMAO. The torC regulatory region contains four direct repeats of a decameric consensus motif designated the tor boxes. Alteration by base substitutions of any of the four tor boxes in a plasmid containing a torC'-lacZ fusion dramatically reduces TorR-dependent torC expression. In addition, deletion of the distal tor box (box1) abolishes torC induction whereas the presence of a DNA fragment starting three bases upstream from box1 suffices for normal torC expression. Footprinting and gel-retardation experiments unambiguously demonstrated that TorR binds to the torC regulatory region. Three distinct regions are protected by TorR binding. One of approximately 24 nucleotides covers the first two tor boxes (box1 and box2); the second is located upstream from the −35 promoter sequence and includes the third tor box (box3); the last is found downstream from the −35 sequence and corresponds to the fourth tor box (box4). Binding to the upstream tor boxes (box1 and box2) appears to be stronger than binding to the downstream tor boxes (box3 and box4) since only the upstream region is protected at the lower concentration of TorR used in the footprinting experiments.
We propose a model in which multiple binding sites (i.e. the tor boxes) contribute to the formation of a nucleoprotein complex, but only one particular proximal site positions TorR properly so that it interacts with RNA polymerase.  相似文献

Alpha-1-microglobulin and bikunin are two plasma glycoproteins encoded by an alpha-1-microglobulin/bikunin precursor (AMBP) gene. The strict liver-specific expression of the AMBP gene is controlled by a potent enhancer made of six clustered boxes numbered 1-6 that have been reported to be proven or potential binding sites for the hepatocyte-enriched nuclear factors HNF-1, -4, -3, -1, -3, -4, respectively. In the present study, electromobility shift assays of wild-type or mutated probes demonstrated that the boxes 1-5 have a binding capacity for their cognate HNF protein. Box 5 is also a target for another, as yet unidentified, factor. A functional analysis of the wild-type or mutated enhancer, driving its homologous promoter and a reporter CAT gene in the HepG2 hepatoma cell line, demonstrated that all six boxes participate in the enhancer activity, with the primary influence of box 4 (HNF-1) and box 2 (HNF-4). A similar analysis in the HNF-free CHO cell line co-transfected with one or several HNF factors further demonstrated various interplays between boxes: box 3 (HNF-3 alpha and beta) has a negative influence over the major HNF-4 box 2 as well as a positive influence over the major HNF-1 box 4.  相似文献

Finding a high quality territory is essential for many animals to reproduce successfully. Despite its importance for fitness, we know little about the process of territory prospecting in wild birds, and whether individual traits and behaviours, such as personality, co‐vary with territory prospecting. Here, we use long‐term data from a wild, insular house sparrow Passer domesticus population to test three hypotheses about territory fidelity and prospecting: 1) house sparrows show high territory fidelity between years and also during winter. 2) Individuals will prospect for a breeding territory during their first winter whereas older, more experienced individuals will keep a territory from previous years and will, therefore, show no or reduced winter territory prospecting. 3) More active behavioural types will prospect more than less active behavioural types. We use data from four winters from automatically, daily recorded nest‐box visits of 188 birds of known age. The number of nest‐boxes that each individual visited within each winter was used as a proxy of winter territory prospecting. We show that house sparrows visit multiple nest‐boxes during their first winter, whereas older individuals keep territories year‐round and, potentially because of this, indeed show reduced winter territory prospecting. Activity was not associated with the number of nest‐boxes visited. Further research is needed to investigate whether time of territory and mate acquisition differs among individuals and the possible effect on lifetime fitness.  相似文献

The replication initiator protein RepA of the IncB plasmid pMU720 was purified and used in DNase I protection assays in vitro. RepA protected a 68-bp region of the origin of replication of pMU720. This region, which lies immediately downstream of the DnaA box, contains four copies of the sequence motif 5'AANCNGCAA3'. Mutational analyses identified this sequence as the binding site specifically recognized by RepA (the RepA box). Binding of RepA to the RepA boxes was ordered and sequential, with the box closest to the DnaA binding site (box 1) occupied first and the most distant boxes (boxes 3 and 4) occupied last. However, only boxes 1, 2, and 4 were essential for origin activity, with box 3 playing a lesser role. Changing the spacing between box 1 and the other three boxes affected binding of RepA in vitro and origin activity in vivo, indicating that the RepA molecules bound to ori(B) interact with one another.  相似文献

During rainfall events, macropores are generally considered to play a dominant role in infiltration after matrix ponding has occurred. Once ponding has been initiated on the soil matrix, surface runoff may be generated at rainfall intensities less than the saturated hydraulic conductivity of the soil. The amount of runoff will depend on detention storage and how efficiently the surface flow is captured by soil macropores. The efficiency of surface water removal by macropores is diminished if surface vents become clogged sealed by washed-in sediment during the runoff event. Post-event opening of surface vents by the animals that created them can remove evidence of the sealing process and so it is particularly important to examine the temporal stability of the soil surface during rainfall events. In this paper evidence of macropore clogging and post-event clearing of the surface vents is presented. A fine sandy loam passed through a 2 mm diameter sieve was packed into two boxes, each with a surface area of 0.5 m2. The boxes were irrigated at 28 mm h−1 using a low energy rainfall sprinkler. This application rate exceeded the saturated hydraulic conductivity of the soil matrix. After measuring runoff and infiltration from the boxes, one box was held as a control and the second was inoculated with earthworms. After four weeks the inoculated box had a burrow density at the soil surface of 380 m−2, with an average diameter of 5 mm. Macropore sealing occurred immediately after ponding and runoff from the macroporous soil was only 10.7% less than a control with no macropores. Within 24 h after cessation of simulated rainfall the earthworms had cleared washed in material from over 95% of burrow vents. Time to matrix ponding was well predicted using hydraulic parameters characteristic of the soil matrix, indicating that matrix sealing was not significant under the experimental conditions.  相似文献

Captive propagation of parrots for the companion bird trade provides a potential means of reducing the economic incentive to capture these birds from the wild. To test whether environmental and social manipulations might stimulate reproduction in captive, wild-caught orange-winged Amazon parrots (Amazona amazonica), we compared reproductive performance in pairs presented with nest boxes (controls) or nest boxes plus additional environmental and social manipulations (enriched group) consisting of misting, fruit supplementation, nest hole restriction, enlarged nest boxes, and pair separation/reunification. In the first breeding trial, in 1990, enriched pairs were more likely to lay eggs (six of seven enriched pairs vs. one of eight control pairs; P < 0.09). When treatments were reversed the following years (e.g., 1990 controls were exposed to enriched conditions in 1991), reproductive performance was not different between the groups (four of seven enriched pairs laid eggs vs. five of seven control pairs), although three pairs in the 1991 enriched group laid eggs which had not laid eggs before. These results are consistent with the idea that while enriched environments may enhance the probability of a first episode of egg laying in captivity, once pairs have laid eggs in captivity, little stimulation beyond that provided by nest box presentation is required to reinitiate egg laying. The environmental and social manipulations of the enriched conditions could be useful in increasing reproductive performance of captive Amazon parrots for the companion bird trade. © 1995 Wiley-Liss, Inc.  相似文献

Nest boxes have grown in popularity as a habitat management tool in Australia during the last decade. This management use remains contentious because some studies suggest nest boxes are ineffective. There are three recent contentions: (i) nest boxes mostly benefit common species, (ii) exotic species may be dominant users of nest boxes, and (iii) species of conservation concern use nest boxes infrequently. We address these contentions using data from 1865 nest boxes involving eight nest box designs. These nest boxes were installed predominantly <200 m from a road in association with highway duplication and re‐alignment across 16 projects in New South Wales. The Common Brushtail Possum (Trichosurus vulpecula) is the species of most relevance to contention 1. It used 9% of boxes overall including 26% of ‘possum’ designated boxes. The most frequent nest box users were small petaurid gliders (mostly Sugar Gliders, Petaurus breviceps) which used 63% of ‘small glider’ designated boxes. This nest box and another suited to the Sugar Glider comprised 40% of all boxes installed, so it is not surprising that this species might be a common user. Exotic species were uncommon users of the nest boxes enabling contention 2 to be rejected. Active hives of Feral Honeybees (Apis mellifera) occupied just 1% of boxes, and another 1% of boxes were used by introduced rodents and birds. The Squirrel Glider (Petaurus norfolcensis) is the species most relevant to contention 3. It was seen in 80 boxes across 11 projects, representing 7% of the three types most frequently used. These observations are not consistent with the third contention. Nest boxes can provide many important insights about the requirements and interactions of hollow‐dependent fauna. However, they are not intended as an alternative to retaining hollow‐bearing trees.  相似文献

cDNA clones originating from the engrailed gene of Drosophila have been isolated from recombinant phage libraries that were made using poly(A)+ RNA extracted from early embryos. The DNA sequence of one of these clones includes a homeo box, a 180 bp sequence present in several other Drosophila genes important in formation of body pattern during development. The homeo boxes found in the other Drosophila genes, as well as in cognate sequences from a wide range of segmented animals, including higher vertebrates, are highly conserved. By contrast, the homeo box within the engrailed gene diverges substantially and, unlike the other homeo boxes, is interrupted by an intervening sequence. The engrailed homeo box is located near the 3' end of a 1700 bp open reading frame. If translated, this sequence would produce a protein of unusual composition. We also show that a neighboring gene has a large region with strong homology to engrailed, and that it also contains a homeo box.  相似文献

The swi8(+) gene of Schizosaccharomyces pombe appears to be involved in the termination step of copy synthesis during mating-type (MT) switching. Mutations in swi8 confer a general mutator phenotype and, in particular, generate specific mutations in the MT region. Sequencing of the MT cassettes of the h(90) swi8-137 mutant revealed three altered sites. One is situated at the switching (smt) signal adjacent to the H1 homology box of the expression locus mat1:1. It reduces the rate of MT switching. The alteration at the smt signal arose frequently in other h(90) swi8 strains and is probably caused by gene conversion in which the sequence adjacent to the H1 box of mat2:2 is used as template. This change might be generated during the process of MT switching when hybrid DNA formation is anomalously extended into the more heterologous region flanking the H1 homology box. In addition to the gene conversion at mat1:1, two mutations were found in the H3 homology boxes of the silent cassettes mat2:2 and mat3:3.  相似文献

Ethological mechanisms which diminish the aggression and competition among sympatricNeomys fodiens (Pennant, 1771) [FF] andN. anomalus Cabrera, 1907 [AA] were studied in enclosure conditions. The animals were kept and subsequently tested (11 FF and 10 AA) in 1- or 2-species groups, in enclosures measuring 275×135 cm or 135×135 cm. During 240 h of direct observation, social behaviours and patterns of hiding-place usage by water shrews were recorded. Mutual avoidance (individual occupation of the nest boxes, territoriality, and spatial dispersion in the enclosure), active antagonism (threats, combats, chases) and absence of typical social hierarchy were the most characteristic intraspecific relations of FF. A high level of tolerance and group-occupation of the nest boxes were the most characteristic intraspecific relations of AA in socially stabilized communities. It seems that a group model of life without hierarchic relations is appropriate for this species. In mixed groups, FF dominated over AA. Over the course of time, the stabilization of interspecific relations and decrease of interspecific conflicts were observed. This may be the result of the habituation of one species to the presence of another, and of active avoidance of predominant species by submissive species (assemblage in 1 nest box, maintenance of distance), in which the same, or very similar, communication systems could be involved.  相似文献

Vigilance is a commonly studied antipredator phenomenon. However, little research has been conducted on the effects of visual obstruction coupled with a manipulation of the cost of performing vigilance behaviours. We tested the hypothesis that antipredator vigilance decreases as the cost of performing antipredator vigilance increases in free-ranging thirteen-lined ground squirrels. We presented adult ground squirrels with peanut butter within six rectangular Plexiglas boxes (opaque on all sides; clear on top) ranging in length from 15.2 to 91.4 cm. Squirrels repeatedly entered and withdrew from the boxes, to scan visually the surrounding area. As box length increased and, therefore, the cost of the vigilance behaviour increased, the ground squirrels increased their mean time within the box per entry and decreased their alertness per unit of trial time. Squirrels took longer to traverse the longer boxes, but their travel time into the boxes was significantly slower than their travel time out of the boxes. We interpreted this difference in travel time to indicate a behavioural conflict that occurred only when the squirrels entered the box (food versus vigilance) and did not exist once the squirrels had begun to exit from the box. The hypothesis that a decrease in ground squirrel antipredator vigilance would benefit the squirrels by decreasing their time exposed to predators (i.e. decreased trial duration, which was determined by the squirrels' behaviour), was not supported.
