A mosaic disease of cowpea ( Vigna sinensis Endl.), prevalent in the bean-growing regions of China, is described.
The characteristic symptoms of the disease are conspicuous mosaic patterns, deformation and distortion of leaves and stunting of the plant.
The cowpea mosaic virus is transmitted by Aphis rumicis L., Macrosiphum pisi Kalt. and Aphis gossypii Glov. It is also transmitted by artificial juice inoculation. The virus is seed-bome. It withstands ageing m vitro for 3 days at 220 C. Its inactivation temperature is approximately 62° C. its tolerance to dilution is about 1: 3000.
Cowpea mosaic virus infects lima bean and Adzuki bean in addition to cowpea.
The relation of this virus to cowpea mosaic virus described by McLean and asparagus-bean mosaic virus described by Snyder is discussed.  相似文献

目前能够用于绿豆(Vigna radiate)种质资源遗传多样性分析的PCR引物极其有限。通过12份农艺性状差异较大的绿豆种质对绿豆以及小豆(Vigna angularis)、豇豆(Vigna unguiculata)、菜豆(Phaseolus vulgaris)等近缘食用豆中的PCR引物进行筛选,结果表明41对绿豆SSR引物中能够有效扩增的有35对,6对有多态性;28对绿豆STS引物中有23对能够有效扩增,2对有多态性;8对小豆SSR引物能够有效扩增的有6对,但均无多态性;27对豇豆SSR引物能够有效扩增的有17对,1对有多态性;24对菜豆SSR引物能够有效扩增的有9对,1对有多态性。这些多态性引物的获得将有助于中国绿豆种质资源的遗传多样性分析。  相似文献

Intrinsic resistance to low concentrations of antibiotics was used to characterise 83 isolates from nodules of cowpea (Vigna unguiculata) and field bean (Phaseolus vulgaris). Characterisation and differentiation of isolates from cowpea was made difficult by associated fast-growing bacteria inside the nodule tissue. Thus, reliable pure culture was difficult to secure without repeated isolation and even via nodulation of the appropriate homologous host. Although the technique may be satisfactory for differentiation and identification of fast-growing rhizobia, it is rated inferior to serology on aspects of facility, time and accuracy where rhizobia from cowpea nodules are concerned. Fingerprint patterns of isolates revealed considerable heterogeneity amongst the populations even where there was commonality of location and/or host plant. Pure cultures of slow-growing rhizobia from V. unguiculata nodules were generally more resistant to the concentrations of antibiotics used than fast-growing nodule bacteria from P. vulgaris.  相似文献

Scab disease of cowpea (Vigna unguiculuta) was shown to be caused by Sphaceloma sp. It affects all above ground parts of the plant. The first symptom of the disease, appearing within 3 to 6 days of inoculation, is puckering of the lamina. Spots on mature leaves are white with or without brown margins. Typical scab lesions on petiole, stem, peduncle and pod are white turning dark brown when chlamydospores are formed and are oblong-elongate except for pod lesions that are ovoid. The most destructive phase is scab of the flowering axis which causes flower and, or, pod abortion or completely prevents flower formation. Inoculation of asparagus pea (V. sesquipedalis) with a cowpea isolate of Sphaceloma sp. produced symptoms similar to those on cowpea. Inoculated hyacinth bean (Lablab niger) produced atypical mild lesions. The following legumes were not affected when artificially inoculated with the fungus: black gram (Phaseolus mungo), green gram (P, aureus), French bean (P. vulgaris), Lima bean (P. lunatus), groundnut (Arachis hypogaea), and soyabean (Glycine max). The pantropical genus Vigna (Leguminosae) comprises 7 cultivated species that are adapted to a wide range of extreme agroclimatic conditions. Few data are available on the relationships among these cultivated species or on their importance as sources of resistance against biotic and abiotic stresses. Therefore, we optimized DNA amplification fingerprinting (DAF) to estimate the genetic diversity within, and genetic relationships among, a representative core collection of cowpea, as compared with 16 accessions representing cultivars from 6 Vigna species. A set of 26 primers was selected from 262 tested random primers and used for the characterization of 85 Vigna accessions (6 V. angularis, 4 each of V. mungo and V. radiata, 2 V. umbellata, 1 V. aconitifolia, and 68 V. unguiculata), with Phaseolus vulgaris subsp. vulgaris as outgroup. A total of 212 polymorphic bands were used for maximum parsimony analysis. Our results clearly distinguished Brazilian from African V. unguiculata genotypes. At the species level, V. angularis was the most related and V. radiata the most divergent species relative to V. unguiculata. DAF markers were also informative at the intraspecific level, detecting a large diversity between cowpea cultivars. The implications of the presented results for cowpea breeding programs are discussed.  相似文献

Zabrotes subfasciatus larvae possess three alpha-amylase isoforms as determined by in gel assays following SDS-PAGE. The two minor isoforms present lower electrophoretic mobility than the major form, and seem to occur as a heterodimer. When developed inside Vigna unguiculata (cowpea) seeds, fourth instar larvae have minor quantities of the slow-migrating forms, but when reared on seeds of Phaseolus vulgaris (common bean) or Phaseolus lunatus, the two slow-migrating forms are expressed in higher amounts, while activity of the major form was independent of the host seed. Larvae developing inside cowpea seeds at the beginning of the fourth instar were fed on flour from cotyledons of cowpea or common bean. Larvae fed on the common bean flour started to express the dimer in higher amounts when compared with the control larvae fed on cowpea flour. In an attempt to correlate differences between starch granules and the induction of alpha-amylases, a detailed study on the digestive process of the granules was conducted. Incorporation of purified starch granules into artificial diets did not induce the two minor alpha-amylases. The in vitro hydrolysis rates of purified granules and the pattern of dextrins liberated by the different alpha-amylases were similar for the two legume species. The starch granules enter the midgut extensively damaged, which may facilitate the access to the more susceptible parts of the granules to enzymatic attack.  相似文献

Four cultivars of cowpea (Vigna unguiculata [L]. Walp.) were infected with cowpea aphid-borne mosaic virus (CABMV) by natural infection in field plots. Seeds taken from these plants were tested for the presence of the virus by ELISA and symptom observation on the plantlets grown from the seeds. A biotin/ streptavidin ELISA technique was used and found to be more sensitive than a standard ELISA protocol for detecting CABMV infection in seed. There was a good correlation between the ELISA detection of CABMV in tissue taken from single cowpea seeds and subsequent development of infected plants grown from the same seeds. The ELISA technique is reliable for selecting CABMV-free stocks of cowpea seeds.  相似文献

Cowpea (Vigna unguiculata (L.) Walp.) is an important food legume in the tropics. It belongs to the Phaseoleae (L.) tribe (Fabaceae family), it is diploid and its chromosome number is 22. Its gene pool includes the cultivated cowpea and its wild relatives, which are connected with Vigna subgenus, Catiang section. Cowpea has a great potential in increasing food legume production. The cowpea varieties, however, are susceptible to a number of insect pests, especially the pod borer Maruca testulalis and a pod sucking-bug complex (e.g.: Clavigralla tomentosicollis, Anoplocnemis curvipes and Riptortus dentipes), which cause severe damage. The crossing programme presented here exploits the variability existing in the wild African germplasm of V. unguiculata and cultivated cowpea. To incorporate the insect pest resistance into the cultivated cowpea economically, reciprocal crosses between wild forms and cowpea varieties were performed, using the stigmatic pollination methods at anthesis. Some barriers were found in these intraspecific crosses. In the majority of reciprocal crosses, the growth of the pollen tubes was arrested in the stigmatic tissue. Only 16.01% of the ovules were fertilised. In these ovules, embryo development was normal at about 20-25 days after pollination. The failure of the intraspecific crosses in about 80.7% of the cases is thus the result of the lack of fertilisation and the unfertilised ovules. There seems to exist considerable incompatibility within the primary cowpea gene pool. The breeding programme carried out under controlled conditions has proved to be less successful in developed cowpea intraspecific F1 hybrids. Further studies should concentrate on germplasm from Africa with documented resistance to major insect pests. In addition, the application of techniques for bypassing barriers to hybridisation of parent genotypes should enable these embryos to grow to plants.  相似文献

The cowpea strain of tobacco mosaic virus was isolated from a range of leguminous hosts at Ibadan, but was rare in cultivated crops. Systemic symptoms in species infected experimentally are described.
A new virus of cowpea was also found in Nigeria. The physical properties (thermal inactivation point 56° C., dilution end-point 1/50,000 and longevity in vitro 4 days at 25° C.) differ from those for cowpea viruses reported elsewhere and the name cowpea yellow mosaic virus is proposed. This virus produces local lesions in French bean ( Phaseolus vulgaris L.) and local and systemic lesions in Bengal bean ( Mucuna aterrima Holland), but does not infect other leguminous hosts. The virus was purified and an antiserum prepared against it.
Both viruses are transmitted by a beetle ( Ootheca mutabilis Sahlb.) which loses infectivity within 48 hr. of leaving plants infected with either or both viruses.  相似文献

Five stored unprocessed cowpea (Vigna spp) and four groundnut (Arachis hypogeae) varieties available in Borno State were examined for the mould flora. The degree of infestation of the grains ranged from 31% to 100% and 68% to 86% for surface sterilized cowpea and groundnut respectively. The mould flora commonly encountered were species of the genera Aspergillus. Penicillium species, Scopulariopsis species and Trichoderma species were also found. The flora of the cowpea was dominated by Aspergillus niger while Aspergillus flavus was the dominant mould on groundnut.  相似文献

Summary In a cross-inoculation experiment using crushed nodules from cowpea (Vigna unguiculata), groundnut (Arachis hypogea), bambara groundnut (Voandzeia subterranea), lima bean (Phaseolus lunatus) and soybean (Glycine max.), it was found that soybean did not nodulate with Rhizobia from any of the other species whilst its Rhizobia nodulated with all species. Cowpea and lima bean, on the other hand, nodulated with Rhizobia from all species, but their Rhizobia nodulated only with each other. Groundnut and bambara groundnut nodulated with Rhizobia from all species except cowpea and lima bean, and their Rhizobia also nodulated with all species except soybean.  相似文献

Genetic diversity and phylogenetic relationships among 22 local cowpea (Vigna unguiculata) varieties and inbred lines collected throughout Senegal were evaluated using simple sequence repeat molecular markers. A set of 49 primer combinations were developed from cowpea genomic/expressed sequence tags and evaluated for their ability to detect polymorphisms among the various cowpea genotypes. Forty-four primer combinations detected polymorphisms, with the remaining five primer sets failing to yield PCR amplification products. From one to 16 alleles were found among the informative primer combinations; their frequencies ranged from 0.60 to 0.95 (mean = 0.79). The genetic diversity of the sample varied from 0.08 to 0.42 (mean = 0.28). The polymorphic information content ranged from 0.08 to 0.33 (mean = 0.23). The local varieties clustered in the same group, except 53-3, 58-53, and 58-57; while Ndoute yellow pods, Ndoute violet pods and Baye Ngagne were in the second group. The photosensitive varieties (Ndoute yellow pods and Ndoute violet pods) were closely clustered in the second group and so were inbred line Mouride and local cultivar 58-57, which is also one of the parents for inbred line Mouride. These molecular markers could be used for selection and identification of elite varieties for cowpea improvement and germplasm management in Senegal.  相似文献

We have prepared a rabbit antiserum specifically directed against the genome-linked protein (VPg) of cowpea mosaic virus by injecting an hydrolysate of purified virion RNA. Using this antiserum as a probe in combination with “Western” (protein) blots of subcellular fractions of cowpea mosaic virus-infected cowpea (Vigna unguiculata) cells, we have detected a bottom component RNA-encoded, 60,000-dalton polypeptide which is membrane bound and presumably represents the immediate precursor of VPg.  相似文献

The capability of cowpea mosaic virus to attach to and infect protoplasts of immune, hypersensitive, and susceptible cowpea (Vigna unguiculata) lines was examined by inoculating protoplasts with either purified virus or radioiodinated purified virus 125I-CPMV. Systems were used in which plants were immune and protoplasts susceptible, plants were immune and protoplasts resistant, and plants and protoplasts were susceptible to CPMV. No differences were observed in the attachment of 125I-CPMV to resistant and susceptible protoplasts. Polycations, proteins, or virus particles were added to the inoculation medium to neutralize potential nonspecific interactions between cells and virus particles. The parasitoid Apanteles taragamae Viereck (Hymenoptera: Braconidae) has been introduced into Benin by the International Institute of Tropical Agriculture (IITA-Benin station) from Taiwan in an attempt to control the cowpea pod borer, Maruca vitrata Fabricius (Lepidoptera: Crambidae), a serious pest of flowers and pods of cowpea Vigna unguiculata Walp. (Leguminosae: Fabaceae). Both the pest and its parasitoid are currently mass-reared using a cowpea and maize flour-based meridic diet. In this study, the suitability of germinating seeds from three cowpea varieties, Kpodji-guèguè, Tawa and TVX3236, as substrates, was tested for rearing A. taragamae under laboratory conditions. The seeds that were soaked for 48 h were the most suitable for M. vitrata larval rearing with up to 87 % survival. The highest parasitism rate of M. vitrata larvae by A. taragamae was observed on the local variety Tawa. Compared to meridic diet, the development cycle of the parasitoid was shorter on Tawa sprouts which also lead to the highest lifetime fecundity. The parasitoid sex ratio varied significantly between feeding substrates with a higher proportion of female wasps on the improved varieties TVX2336 and local variety Kpodji-guèguè. Rearing M. vitrata larvae on germinating cowpea grains is 4.38 times cheaper than on meridic diet. The data are discussed with emphasis on the effects of feeding substrates quality on life history parameters of insect pests and their parasitoids.  相似文献

Xanthine dehydrogenase (XDH, EC 1.2.1.37) was immunopurified from root nodules of three legume species, soybean [ Glycine max (L.) Merr. cv. Pella], cowpea [ Vigna unguiculata (L.) Walp. cv. California Black Eye], and lima bean [ Phaseolus lunatus L. Henderson]. Polyclonal antibodies raised against each enzyme and monoclonal antibodies raised against soybean XDH were used to compare the three enzymes serologically. Double diffusion and enzyme-linked immunosorbent assays with polyclonal and monoclonal antibodies showed that the cowpea and lima bean enzymes are very similar immunologically but both differ measurably from soybean. Amino acid compositions of the legume nodule XDHs are presented as well. Although relatedness between these enzymes can be detected by immunological crossreactivity, each XDH has unique epitopes that can be used to distinguish the three proteins.  相似文献

Summary In a soil pot culture experiment conducted under greenhouse conditions, two varieties of cowpea (Vigna unguiculata (L) Walp) were found to be differentially susceptible to phosphorus-induced zinc deficiency. Although phosphate application, in the absence of applied zinc, caused growth disorder in both the varieties, the symptoms were more virulent, appeared early and were induced with both 25 and 75 ppm phosphorus in variety HFC-42-1, as against FOS-1, in which case zinc deficiency symptoms appeared under P75Zn0 treatment only. Contrary to the expectation the more susceptible variety was found to have higher zinc concentration in its shoots and thus the differential susceptibility of these varieties of cowpea could not be related to their zinc absorption characteristics. High phosphate application caused efflux of zinc during 16–32 day growth in both the varieties. However, higher sensitivity to phosphorus-induced zinc disorder in variety HFC-42-1, appeared to be due to nearly 2 times higher P concentration in this variety as against FOS-1. The results indicate that variety HFC-42-1 has a greater need for zinc because of its inherent capacity to accumulate more phosphorus. In order to investigate the possible correlation between the induction of alpha-amylases and the ingestion of the common bean alpha-amylase inhibitor 1 (alphaAI-1), this inhibitor was incorporated into artificial diet. It was observed that larvae fed on diet containing chronic doses of alphaAI-1 during their development, produced the two slow-migrating forms in higher amounts than control larvae, however, fourth-instar larvae fed on the same diet presented less amylase activity than control larvae. The data suggested that alphaAI-1 is involved in amylase induction and that it has inhibitory activity against the constitutive amylase, when starch granules are used as substrate.  相似文献

A globulin fraction prepared from the meal of Callosobruchus maculatus-resistant cowpea (Vigna wiguiculata) seeds was shown to be detrimental to this bruchid when incorporated in artificial seeds.2. The performance of C. maculatus was also shown to be strongly hindered by vicilins from resistant seeds when these storage proteins were incorporated in artificial seeds at the level of 2%.3. The purified vicilins from seeds of both resistant and susceptible cowpea varieties were shown to have the same SDS-PAGE pattern but different mobilities in non-denaturing polyacrylamide gel electrophoresis.4. These results and previous ones obtained by us (Silva and Xavier-Filho, 1991; Sales et al., 1992) strongly suggest that the resistance of cowpea seeds from the cultivar TVu 2027 and from others bred from it is associated with the presence of vicilin molecules which are refractory to digestion by bruchid midgut proteinases.  相似文献
